This thesis investigates the effects of substance P on a spinal nociceptive reflex in the rat. Intrathecal administration of substance P and other analogous peptides to the L5 vertebral level produced a dose-dependent decrease in reaction time to tail withdrawal from a noxious radiant heat source. Similar administration of VIP, galanin, CGRP, angiotensin II, somatostatin and TRH failed to mimic these effects of substance P. As substance P was found to equally decrease the reaction time in spinal transected rats, it is concluded that this facilitation occurs via a spinal mechanism alone.<br>Adm

Previous studies suggest that IL-12, the potent Th1-inducing cytokine, IFN-gamma, TNF-alpha and nitric oxide (NO) contribute to resistance against blood-stage malaria. Early Th1 responses correlate with resistance, whereas predominantly Th2 responses are associated with susceptibility. In the present studies, the requirements for endogenous IL-12 and its role in host defense against blood-stage P. chabaudi AS malaria were investigated. Our results reveal, for the first time, significant differences in the kinetics of endogenous IL-12 p70 synthesis and splenic IL-12R beta1 and beta2 mRNA expres

p-Cumate-2,3-dioxygenase (CDO) from Pseudomonas putida F1 converts p-cumate to cis-2,3-dihydroxy-2,3-dihydro-p-cumate, a potential useful acid cis-diol. To enhance the activity of CDO for the production of this acid cis-diol, the recombinant CDO expressed in Escherichia coli Top10 strain was evolved by directed evolution using the error-prone polymerase chain reaction (EP-PCR) method. The mutant library was screened in a 96-well format based on the enzyme's ability to transform colorless indole-2-carboxylate to the water-insoluble blue dye indigo. Anthelmintics, antiparasitic agents, have been developed as a main weapon to control parasitic nematodes of domestic ruminants. Unfortunately, the intensive use of anthelmintics leads to the development of drug resistance in parasite populations. Anthelmintic resistance has compromised the control of nematode parasites and has become a major problem in many countries of the world. Resistance to the newest anthelmintics such as ivermectin (IVM) and related anthelmintics in Haemonchus contortus in sheep has been developing rapidly in recent years. Resistance to the avermectin/milbemycin class of anthelmintics in nematodes has become a serious problem worldwide due to their unrestricted usage. Resistance to these compounds is attributed to the over-expression of the transport protein, P-glycoprotein (P-gp). Anthelmintic treatment of livestock is an important aspect of the control of gastrointestinal parasites. Resistance to anthelmintics is common, and an understanding of resistance requires knowledge of an anthelmintic's mode(s) of action and mechanism(s) of resistance. The parasitic nematode, Haemonchus contortus, has developed resistance to benzimidazoles and avermectins/milbemycins. Proposed mechanisms of resistance are here supported by genetic changes observed in genes whose protein products are believed to interact with these anthelmintics. This study examines Christian experientialist epistemology as articulated of late by Alvin Plantinga especially and also William Alston. It situates their approach to the epistemic status of Christian belief claims within the overall outlook they have respectively developed on what features generally legitimate beliefs as being rationally responsible or even qualify some true beliefs as constituting knowledge. First to be taken up is Plantinga's journey from considering the deontological justification for basic belief in God to making his own externalist proposal for warranted belief at large.
