Pemahaman seperti ini akan cenderung membatasi perilaku untuk mengasihi orang lain di luar satu ikatan hubungan tertentu. Bahkan praktek hidup primordial seperti ini dapat merusak kesKata kunci kurang atuan dan persatuan bangsa. Karena itu, diperlukan suatu kajian eksegetis tentang konsep “sesamaku manusia” dalam Lukas 10: 25-37, supaya mendapatkan konsep yang benar tentang sesama manusia. Penulis injil Lukas menunjukkan, konsep “sesama manusia” yang harus dikasihi tidak dibatasi oleh ikatan apapun, bahkan musuh atau yang memusuhipun harus dikasihi, seperti mengasihi diri sendiri. Dengan demikian, diharapkan setiap orang untuk mengasihi sesamanya manusia, seperti dirinya sendiri. Hal ini akan berdampak terhadap harmonisasi hubungan dengan siapa saja, sehingga dapat menjaga keutuhan, kesatuan dan persatuan, seperti satu bangsa.

The story of the Good Samaritan reminds us of one of Martin Luther King, Jr.’s most pointed statements on racial reconciliation: “The ultimate measure of a man is not where he stands in moments of comfort and convenience, but where he stands at times of challenge and controversy.” Most of us are familiar with the challenge to “love your neighbor” as emphasized by Jesus in the telling of the story, but we seldom think about loving the perpetrator. In this exposition of Luke 10:25–37, Burris moves us from a call to love the challenging neighbor to a love for all humanity that is deep enough and great enough to enable one to participate in night walks on the streets of Richmond, CA—a city riddled with gun violence.

Abstract:
Low levels of the vitamin D-regulated antimicrobial peptide cathelicidin (LL-37) may negatively impact the immune status of human immunodeficiency virus-1 (HIV-1) infected individuals (HIV+). We compared plasma LL-37 levels in healthy controls (HIV−) and HIV+ individuals on or off antiretroviral therapies (ARTs) (ART+ and ART−, respectively), and evaluated the relationship between vitamin D and LL-37 levels. In this cross-sectional study, levels of LL-37, 25-hydroxycholecalciferol [25(OH)D3] and 1,25-dihydroxycholecalciferol [1,25(OH)2D3] were measured from an initial cohort of 18 healthy controls and 10 HIV+/ART− individuals. Because this cohort lacked HIV+/ART+ subjects, LL-37 was also quantified from a second cohort of 10 HIV+/ART− and 13 HIV+/ART+ individuals. LL-37 levels were significantly lower in the HIV+/ART− group compared to the healthy controls (P = 0.01). A direct relationship was observed between LL-37 and both 25(OH)D3 and 1,25(OH)2D3. The level of 25(OH)D3 was predictive of higher LL-37 (P = 0.04) and for any given level of 25(OH)D3, HIV+/ART− subjects averaged 20 % lower LL-37 compared to the healthy controls (P = 0.045). For any given level of 1,25(OH)2D3, HIV+/ART− subjects averaged 25 % lower LL-37 compared to the healthy controls (P = 0.018), although 1,25(OH)2D3 was not predictive of higher LL-37 (P = 0.28). Finally, LL-37 levels were significantly lower in the HIV+/ART− group compared to the HIV+/ART+ group from the second cohort (P = 0.045). Untreated HIV infection may contribute to lower LL-37 levels, independent of vitamin D levels. ART treatment may potentially mitigate this decrease in LL-37 levels.

Abstract : Hospitality in the new normal era has brought new problems because the actions of hospitality towards other people are at risk of corona virus. Therefore the construction of hospitality needs to be reformulated amid a changing context. This was done through a study of the text of Luke 10: 25-37 with the historical critical method. From the results of the study is found several theological points. First, hospitality must be based on compassion which will enable a person to empathize for strangers although faced with a difficult situation. Second, acts of hospitality need to be done collaboratively across groups, ethnicity and religions facing of an increase in the communities affected by Covid-19. Third, the construction of hospitality need to be done with a digital technology media approach that functions intertwined with human interaction and is beneficial in presenting the hospitality of God to anyone penetrating geographical boundaries.Keywords: hospitality, compassion, collaborative, digital technology. Abstrak: Hospitalitas di era new normal telah membawa persoalan baru karena tindakan hospitalitas terhadap orang lain menjadi beresiko pada penularan virus corona. Karenanya konstruksi hospitalitas perlu dirumuskan kembali di tengah konteks yang sudah berubah. Hal itu dilakukan melalui penelitian teks Lukas 10:25-37 dengan metode historis kritis. Dari hasil penelitian ditemukan beberapa pokok teologis. Pertama, hospitalitas harus berpijak pada belas kasih yang akan memampukan seseorang untuk berempati terhadap orang asing walau diperhadapkan pada situasi yang sulit. Kedua, tindakan hospitalitas perlu dilakukan secara kolaboratif lintas kelompok, etnik, dan agama dalam menghadapi meningkatnya masyarakat yang terdampak Covid-19. Ketiga, konstruksi hospitalitas perlu dilakukan dengan pendekatan media tehnologi digital yang berfungsi terjalinnya interaksi antar manusia dan bermanfaat dalam menghadirkan keramahtamahan Allah pada siapapun menembus batas-batas geografis.Kata kunci: hospitalitas, belas kasih, kolaboratif, tehnologi digital.

Background: Narrowband ultraviolet B (NB-UVB) has been widely used in the treatment of psoriasis. It has been shown that vitamin D is a major regulator of the expression of human antimicrobial peptide cathelicidin LL-37, which has a critical role in inflammatory cascade in psoriasis. Objective: To evaluate the effect of NB-UVB therapy on serum levels of cathelicidin LL-37 and 25-hydroxyvitamin D [25(OH)D] in psoriasis patients. Methods: Ninety-three psoriasis patients and 50 controls were included in the study. For psoriasis patients, serum levels of 25(OH)D and cathelicidin LL-37 were estimated before and after NB-UVB therapy. Results: Before treatment, serum 25(OH)D levels were significantly lower in psoriasis patients (31.5 ± 14.41 nmol/L) compared to controls (53.5 ± 19.6 nmol/L), p = .015. In contrast, serum LL-37 was significantly higher in psoriasis patients (13.24 ± 3.2 ng/mL) than in controls (7.92 ± 5.33 ng/mL), p < .001. After NB-UVB treatment, there was a highly significant elevation of serum 25(OH)D to reach 56.85 ± 5.2 nmol/L ( p < .001) and further elevation of serum LL-37 to reach 29.4 ± 4.2 ( p = .02). Conclusions: The elevation of serum 25(OH)D and cathelicidin LL-37 could be an additional possible mechanism of action of NB-UVB therapy in the treatment of psoriasis.

Constant cross talk betweenCandida albicansyeast cells and their human host determines the outcome of fungal colonization and, eventually, the progress of infectious disease (candidiasis). An effective weapon used byC. albicansto cope with the host defense system is the release of 10 distinct secreted aspartic proteases (SAPs). Here, we validate a hypothesis that neutrophils and epithelial cells use the antimicrobial peptide LL-37 to inactivateC. albicansat sites of candidal infection and thatC. albicansuses SAPs to effectively degrade LL-37. LL-37 is cleaved into multiple products by SAP1 to -4, SAP8, and SAP9, and this proteolytic processing is correlated with the gradual decrease in the antifungal activity of LL-37. Moreover, a major intermediate of LL-37 cleavage—the LL-25 peptide—is antifungal but devoid of the immunomodulatory properties of LL-37. In contrast to LL-37, LL-25 did not affect the generation of reactive oxygen species by neutrophils upon treatment with phorbol esters. Stimulating neutrophils with LL-25 (rather than LL-37) significantly decreased calcium flux and interleukin-8 production, resulting in lower chemotactic activity of the peptide against neutrophils, which may decrease the recruitment of neutrophils to infection foci. LL-25 also lost the function of LL-37 as an inhibitor of neutrophil apoptosis, thereby reducing the life span of these defense cells. This study indicates thatC. albicanscan effectively use aspartic proteases to destroy the antimicrobial and immunomodulatory properties of LL-37, thus enabling the pathogen to survive and propagate.

The effects of fermentation at temperatures of 15°C, 25*C, 30°C and 37 C on the physico chemical and sensory qualities of 'nono', fermented milk products were determined. Of the four temperatures used, milk fermented at 25°C and 30*C produced the most suitable and acceptable 'nono'. While diacetyl, a flavor enhancer was 1.04g/ml and 3.2 wyn in 'nono' fermented at 25°C and 30'C respectively, it was zero ug/ml at 15°C and 6.3 ugel at 37°C temperatures of fermentation. The på readings at 24 hours of milk fermented at 25°C and 30°C temperatures of fermentation were 4.9 and 4.8 respectivdy, while at 15°C and 37°C temperature of fermentation, pH, readings were 5.1 and 4.3 respectively. The taste panellists assessment of 'nono' favoured 25°C and 30°C temperature of fermentation more than 15°C and 37°C.

Abstract:
Introduction: Most children with serious infection diseases suffer from malnutrition. Vitamin D participates in the immune response through endogenous antimicrobial peptides (AMPs) regulation. The aim of this study is to investigate the expression of 25-hydroxyvitamin D3 [25(OH)D3], AMPs [LL-37 and human β-defensin 2 (HBD-2)] in the children with pertussis. Methodology: Serum levels of 25(OH)D3, LL-37, and HBD-2 were detected in 116 children with pertussis aged at 1–12 months (67 males and 49 females). Fifty healthy infants at similar age were employed as normal controls. Results: The serum 25(OH)D3 levels in the children with mild (27.30 ± 5.98 ng/ml) and severe (24.40 ± 6.27 ng/ml) pertussis were significantly lower than that in the healthy group (30.16 ± 5.13 ng/ml; p <0.01). The vitamin D deficiency rates in children with mild (55.9%) and severe (78.12%) pertussis were significantly higher than that in the control group (34%; p < 0.01). The serum levels of LL-37 and HBD-2 were significantly higher in pertussis patients. Spearman rank correlation analysis did not show any correlation of 25-(OH)D3 with LL-37 or HBD-2. Conclusions: Most children with pertussis had vitamin D deficiency accompanied by elevated serum LL-37 and HBD-2 levels. However, the average level of 25(OH)D3 at 26.50 ng/ml in the infants with pertussis may not affect the immuno-regulatory ability; thus, the infants with pertussis still maintained a higher level of AMPs (LL-37 and HBD-2) against pertussis infection.

The purpose of this study was to assess the stability of various antibiotics in PD-bags when stored at different temperatures over a 14-day period.Methods7.5% icodextrin PD-bags were dosed with gentamicin 20 mg/L ( n = 9), vancomycin 1,000 mg/L ( n = 9), cefazolin 500 mg/L ( n = 9) and ceftazidime 500 mg/L ( n = 9) as for intermittent dosing. Combinations of gentamicin/vancomycin ( n = 9), cefazolin/ceftazidime ( n = 9), and cefazolin/gentamicin ( n = 9) were also tested. Nine drug-free bags were used as controls. Bags were stored in triplicate at 37°C, room-temperature (25°C), and refrigeration (4°C). Antibiotic concentrations were quantified at various time intervals using validated chromatography. Storage duration was considered unstable if the concentration of the antibiotic dropped ≤ 90% of the initial value.ResultsGentamicin was stable for 14 days at all temperatures. Vancomycin was stable for 4 days at 37°C and for 14 days at both 25°C and 4°C. The gentamicin and vancomycin combination was stable for 4 days at 37°C and for 14 days at 25°C and 4°C. Cefazolin alone was stable for 24 hours at 37°C, 7 days at 25°C, and 14 days at 4°C. Ceftazidime alone was stable for only 6 hours at 37°C, 2 days at 25°C, and 14 days at 4°C. The cefazolin and ceftazidime combination was stable for 24 hours at 37°C, 2 days at 25°C, and 14 days at 4°C. The cefazolin and gentamicin combination was stable for 1 day at 37°C, 4 days at 25°C, and 14 days at 4°C.ConclusionsAntibiotics premixed in icodextrin PD-bags have varying stabilities with stability generally least at 37°C and best at 4oC, permitting storage for 14 days when refrigerated and pre-warming to body temperature prior to administration. Further research confirming the sterility of these antibiotic-containing bags is recommended.

Free-living infective larvae Strongyloides venezuelensis were cultured in Dulbecco's modified Eagle's medium at 25 and 37 °C, and development to the parasitic stage was evaluated using morphological, protein and antigenicity criteria. Few larvae cultured at 25 °C showed development whereas, in most of the larvae cultured at 37 °C, there appeared characteristic changes such as a bulb-like head and droplets under the cuticle with an increase of body width of the larvae. The results obtained from two-dimensional polyacrylamide gel electrophoresis(2D–PAGE) revealed that the protein spot patterns of the larvae cultured at 25 and 37 °C were differentiated by 17 specific spots. In addition, Western blot analysis combined with 2D–PAGE for reaction with serum obtained from an infected rat revealed that protein spots showing immunodominant antigen at 37 °C were almost the same as those of the larvae recovered from the rats rather than those of the larvae at 25 °C. These results strongly suggested that a temperature shift from 25 to 37 °C has an important role in the development of free-living infective larvae to the parasitic stage of S. venezuelensis. The culture system established in the present study was useful for biological and biochemical studies in the development from/of the free-living to the parasitic stage of Strongyloides species.

Virulence, as determined in a mouse model, and virulence factor activities of catalase (CA), superoxide dismutase (SOD) and listeriolysin O (LLO), was examined in Listeria monocytogenes 10403S. Cells were propagated in media containing various concentrations of sodium chloride (NaCl) at 4, 25 and 37°C. Strain 10403S exhibited significant increases in CA activity and LLO when grown in media containing 428 mM of NaCl at 37°C. The CA activities at 4 and 25°C were significantly less, and the cells exhibited similar increases and decreases as cells grown at 37°C. When comparing the growth temperatures, the CA activity decreased as the growth temperature decreased. The SOD activity was significantly increased only when cells were propagated in media containing either 428 or 1,112 mM of NaCl. The SOD activity increased as the growth temperature decreased. No LLO activity was detected when cells were grown at 4 and 25°C. The production of these enzymes appeared to be thermoregulated. In addition, approximate lethal dose (ALD50) values were determined after intragastric (i.g.) and intraperitoneal (i.p.) infection. Each method of infection indicated that LLO was required for virulence, while growth in salt containing media, growth at 4°C, or the production of higher levels of CA, SOD and LLO did not appear to influence the virulence of L. monocytogenes.

ABSTRACT Fifty strains representing 38 species of the genus Legionella were examined for biofilm formation on glass, polystyrene, and polypropylene surfaces in static cultures at 25°C, 37°C, and 42°C. Strains of Legionella pneumophila, the most common causative agent of Legionnaires' disease, were found to have the highest ability to form biofilms among the test strains. The quantity, rate of formation, and adherence stability of L. pneumophila biofilms showed considerable dependence on both temperature and surface material. Glass and polystyrene surfaces gave between two- to sevenfold-higher yields of biofilms at 37°C or 42°C than at 25°C; conversely, polypropylene surface had between 2 to 16 times higher yields at 25°C than at 37°C or 42°C. On glass surfaces, the biofilms were formed faster but attached less stably at 37°C or 42°C than at 25°C. Both scanning electron microscopy and confocal laser scanning microscopy revealed that biofilms formed at 37°C or 42°C were mycelial mat like and were composed of filamentous cells, while at 25°C, cells were rod shaped. Planktonic cells outside of biofilms or in shaken liquid cultures were rod shaped. Notably, the filamentous cells were found to be multinucleate and lacking septa, but a recA null mutant of L. pneumophila was unaffected in its temperature-regulated filamentation within biofilms. Our data also showed that filamentous cells were able to rapidly give rise to a large number of short rods in a fresh liquid culture at 37°C. The possibility of this biofilm to represent a novel strategy by L. pneumophila to compete for proliferation among the environmental microbiota is discussed.

Abstract The story about the “Good Samaritan” in the gospel of Luke appears in the midst of a halakhic discussion between Jesus and a Judaean “lawyer” over who constitutes a “neighbor” (Luke 10:25-37). While scholars have often interpreted this pericope as a call for social inclusivity, the ways that Luke relies on and perpetuates prejudicial Judaean stereotypes about Samaritans have seldom been analyzed. This study draws on social-scientific and critical theory on ethnicity and the plethora of recent scholarship on Samaritan-Judaean interactions in order to explore the ways in which Luke’s text conveys prevalent ethnic stereotypes about Samaritans. It argues that Luke, like earlier and contemporaneous Judaean sources, appropriates an ethnographic representation of Samaritans as “proximate others” as part of a process of identity formation.

The aim of this study was to investigate the effects of beta-stimulation in deep (25 degrees C) hypothermia. Cardiac catheterization was performed on seven anesthetized beagle dogs. They were cooled between ice bags down to 25 degrees C and received isoproterenol administered intravenously three times: at the normal body temperature (37 degrees C) before cooling, after cooling at 25 degrees C, and after rewarming at 37 degrees C. Circulatory function was measured for every 1 degree C of temperature change. Isoproterenol infusion at 37 degrees C induced cardiac acceleration, including the increases of heart rate, cardiac output, and peak first derivative of the left ventricular pressure curve. Systemic vascular and mean outflow resistances and mean aortic pressure decreased. During cooling, shivering thermogenesis continued, even down to 25 degrees C. At 25 degrees C, cardiac acceleration after isoproterenol infusion did not exist but relaxation rate increased slightly. Systemic vascular and mean outflow resistances decreased, but left ventricular end-diastolic and filling pressures increased. beta-Stimulation at normal body temperature increases shivering thermogenesis during cooling. The venous return to the left ventricle at 25 degrees C increased after isoproterenol infusion while systemic vascular resistance decreased, indicating systemic vasodilatation. This increase in preload is probably due to vasoconstriction in pulmonary vessels, which may be mediated by prejunctional beta-adrenoceptors. For cardiac inotrophy, the isoproterenol had no physiologically significant effects at 25 degrees C. After rewarming at 37 degrees C, the effects of isoproterenol were physiologically similar to the effects at the same temperature before cooling.

Tomato hot pot sauce (THPS) at different storage temperatures (0, 25, and 37°C) and with two kinds of packaging for 120 days was investigated in this study. High performance liquid chromatography was employed for detecting lycopene and 5-hydroxymethylfurfural (HMF). The changes of lycopene and HMF during storage were regressed with kinetic equation of both zero-order and first-order models, and the latter fitted better. The kinetic equation constant (k value) of lycopene or HMF at 37°C was higher than that at 25°C. The k value of lycopene of PET/PE (P1) packaged THPS was 1.60 times of that of PET/Al/EAA/PE (P2) packaged at 37°C, while it was 2.12 times at 25°C. The k value of HMF of P1 packaged THPS was 1.69 times of that of P2 packaged at 37°C, while it was 1.01 times at 25°C. Significant correlations between color index of L⁎, a⁎, and a⁎/b⁎ and lycopene or HMF were found at storage temperature. Browning color was attributed to both Maillard reaction and degradation of lycopene. In conclusion, lower storage temperature and stronger oxygen barrier property of package could maintain color stability and extend shelf life.

Moisture sorption isotherms of oat flakes were determined at temperatures of 5, 25 and 37°C, using a gravimetric technique in an aw range of between 0.107 and 0.855. These curves were modeled using six equations commonly applied in food. The quality of the fit was assessed with the regression coefficient (r2) and the mean relative percentage error (MRPE). The best fit were obtained with the Caurie model with r2 of 0.996, 0.901 and 0.870, and MRPE of 7.190, 17.878 and 16.206, at 5, 25 and 37°C, respectively. The equilibrium moisture presented a dependence on temperature in the studied aw range, as did the security moisture (XS). These results suggest that the recommended storage conditions of oat flakes include: a relative air humidity of 50% between 5 and 25°C and of 38% up to 37°C.

The aim of the present study is to investigate the influence of various experimental conditions, i.e., two different concentrations of D-fructose as a rapidly glycating substance and three incubation temperatures, on the glycation reaction of alanine aminotransferase (ALT, EC 2.6.1.2) and aspartate aminotransferase (AST, EC 2.6.1.1) expressed by decreasing catalytic activities of the enzymes during a 56 day in vitro incubation period. D-fructose in the concentration of 50 mmol/l did not inhibit the catalytic activity of either enzyme at 4 °C, partially inhibited AST activity in samples incubated at 25 °C (to 40% of the initial activity), and completely inhibited this enzyme at 37 °C at the end of the incubation period. In the presence of the same concentration of D-fructose, ALT showed no catalytic activity after 35 days at 25 °C or after 10 days at 37 °C. In 500 mmol/l D-fructose, complete AST inhibition was observed after 35 days (25 °C) or 20 days (37 °C), and no ALT activity was found on day 20 at either 25 °C or 37 °C. Taking into account the highest possible stability of enzymes, we suppose that a three-week observation of their residual catalytic activity in the presence of 50 mmol/l D-fructose at the temperature of 25 °C seems to be the most prospective experimental design for future glycation studies with aminotransferases under the influences of drugs.

OBJECTIVE: To examine the stability of ceftazidime, vancomycin, and heparin, alone and in combination, in dialysis solution over six days at three temperatures. DESIGN: Nine 250-mL Dianeal PD-2 dextrose 1.5% bags were prepared with ceftazidime, vancomycin, and heparin alone and in combination at set concentrations of 100 μg/mL, 50 μg/mL, and 1 unit/mL, respectively. Three bags of each mixture were stored at 4, 25, and 37°C. Duplicate samples for analysis were removed from each bag at the following time points: premix, 0, 12, 24, 48, 72, 96, 120, and 144 hours. MAIN OURCOME MEASURES: Each sample was examined visually for signs of cloudiness and precipitation. Each sample was analyzed by stability-indicating HPLC assay for ceftazidime and vancomycin, with stability defined as less than 10 percent degradation of drug overtime. RESULTS: No color change or precipitation was observed in any bag. Vancomycin with or without heparin was stable for 5–6 days at 4, 25, and 37°C. Ceftazidime with and without heparin was stable for 6 days at 4°C, 4 days at 25°C, and less than 12 hours at 37 °C. Vancomycin plus ceftazidime with and without heparin was stable for 6 days at 4 °C and 25°C, and 4–5 days at 37 °C, Ceftazidime plus vancomycin with or without heparin was stable for 6 days at 4°C, 2–3 days at 25°C, and 12 hours at 37 °C. CONCLUSIONS: Bulk preparations of ceftazidime and vancomycin, alone and in combination and with or without heparin in Dianeal PD dextrose 1.5% solution, are sufficiently stable for use up to 6 days under refrigeration or 48 hours at room temperature.
