Microencapsulation of the enzyme phenylalanine ammonia-lyase (PAL) was developed for in vivo depletion of systemic phenylalanine (PHE) in Phenylketonuric (PKU) rats. Phenylalanine ammonia-lyase was successfully microencapsulated within artificial cells. The immobilized enzyme had an assayed activity of 20% $ pm$ 4% (Mean $ pm$ S.D.) of the free enzyme in solution. The immobilized enzyme maintained a higher enzyme activity at very low pH compared to the free enzyme in solution. The pH optimum of the free and immobilized enzyme was 8.5. The phenylhydrazine-induced erythropoietic mouse spleen is used as a model system to demonstrate the relationship between tissue growth and polyamine metabolism. Phenylhydrazine produced significant changes in spleen weights, hematocrits and reticulocyte counts in Swiss-Webster mice. The average spleen weight went up from a control of 155 mg to 875 mg at 96 hours after phenylhydrazine administration, while a 49% reduction in the value of hematocrit was observed at 72 hours. Apres plus de 10 ans de travaux évaluant les potentialités de l'insuline nasale dans le traitement du diabète sucre, le constat est plutôt négatif : la biodisponibilité est insuffisante et les résultats métaboliques obtenus chez l'homme ne sont pas en rapport avec les espérances des cliniciens. Nous avons repris et complète les travaux de la littérature pour tenter d'améliorer l'efficacité et la tolérance de la formulation nasale et acquérir des données cliniques suffisantes pour conclure sur l'intérêt de l'insuline nasale. The present study investigated pharmacological properties of Capsicum frutescens-derived capsaicin, including its analgesic, anti-inflammatory and coagulatory properties. The effects of capsaicin on gastrointestinal and myocardial muscles, as well as on myocardial ischaemic-reperfusion, were also investigated.
