Abstract:

Tarih boyunca insanlar, çok farklı nedenlerle (savaş, doğal afet, ekonomik vb.) bireysel ya da kitlesel olarak farklı coğrafyalara göç etmişlerdir. Göç sonucunda farklı kültürler temasa geçtiğinde, bireyler ve topluluklar kültürleşme adı verilen bir süreçle birbirlerinin kültürlerini etkilemekte hem göçmenlerin hem de ev sahibi bireylerin yaşamları göç öncesine göre farklılaşmaktadır. Bu çalışmanın amacı Declan T. Barry (2001) tarafından geliştirilen EAAM (the East Asian Acculturation Measure) adlı ölçeğin Türkçe’ye uyarlanarak geçerlik ve güvenirliğini tespit etmektir. Barry’nin kültürleşme ölçeği asimilasyon, bütünleşme, ayrışma ve marjinalleşme olmak üzere dört boyut, altı başlıktan (evlilik, kültürel gelenekler, dil, sosyal etkinlikler, arkadaşlar ve müzik) oluşmaktadır. Yapı geçerliğine yönelik gerçekleştirilen açıklayıcı faktör analizi ve doğrulayıcı faktör analizi sonuçları, özgün ölçekte bulunan dört boyutlu modelin geçerliğini desteklemiştir. Kültürleşme ölçeği ve alt boyutları arasında yapılan korelasyon analizi sonucunda istatistiksel olarak anlamlı ilişkiler bulunmuştur. Kültürleşme ölçeği ve alt boyutları arasında anlamlı bir korelasyon katsayısı bulunması ölçeğin alt boyutlarının benzer yapıları ölçtüğünü göstermektedir. Analiz sonuçları, ölçeğin dört boyutlu özgün yapısının göçmenlerin kültürleşme düzeyini belirleme konusunda geçerli ve güvenilir olduğunu göstermektedir.

Abstract:

The article deals with the study of linguo-stylistic means of the comic category representation at the lexico-semantic level. Modern English prose is characterized by a specific comic modality, which is a characteristic feature of the author’s individual style. The leading means of the lexico-semantic level, that contribute to the realization of the comic effect in belles-letters texts, have been analyzed. The material of research is the English-language prose works of small forms by the most prominent writers, such as: K. Barry, K. Vonnegut, R. Carver and R. Dahl. The aforementioned authors use hyperbolization, which is introduced by means of similes, indefinite pronouns, nouns formed from numerals (in the process of numeralization), temporal and spatial adverbs, a set of lexical units with negative colouring, adjectives in the superlative degree of comparison, modal verbs in the negative form, emphatic constructions.

Abstract:

This paper asserts that we accept or reject humorous texts discursively on the basis of what we perceive as authorial agendas. This “authorial agenda spotting” is activated by discursive “triggers”, which identify, filter, reject, endorse, or otherwise subjectively interpret the discourse of a textual author. This study was prompted by observing the negative reception of a humorous text by a predominantly Muslim postgraduate student cohort who signalled cultural identity and social cohesion by rejecting a text which subverted gender performance according to their discursive expectations. The study sought to compare this triggered effect with the reception of the same text by a distinctly pre-disposed audience comprised of same-sex-attracted bloggers. This reception in turn was contrasted with the reception of the text by mainstream media reviewers. The text itself seems to spark these discursive triggers in all three audiences. It is taken from “The Dame Edna Treatment” (2007), a TV-media entertainment programme, which features the celebrity guests k. d. lang and Ivana Trump being “interviewed” by the Australian comedian Barry Humphries in character as “Dame Edna”.

Abstract:

Dietary potassium (K) deficiency is accompanied by phosphaturia and decreased renal brush border membrane (BBM) vesicle sodium (Na)-dependent phosphate (Pi) transport activity. Our laboratory previously showed that K deficiency in rats leads to increased abundance in the proximal tubule BBM of the apical Na-Pi cotransporter NaPi-IIa, but that the activity, diffusion, and clustering of NaPi-IIa could be modulated by the altered lipid composition of the K-deficient BBM (Zajicek HK, Wang H, Puttaparthi K, Halaihel N, Markovich D, Shayman J, Beliveau R, Wilson P, Rogers T, Levi M. Kidney Int 60: 694–704, 2001; Inoue M, Digman MA, Cheng M, Breusegem SY, Halaihel N, Sorribas V, Mantulin WW, Gratton E, Barry NP, Levi M. J Biol Chem 279: 49160–49171, 2004). Here we investigated the role of the renal Na-Pi cotransporters NaPi-IIc and PiT-2 in K deficiency. Using Western blotting, immunofluorescence, and quantitative real-time PCR, we found that, in rats and in mice, K deficiency is associated with a dramatic decrease in the NaPi-IIc protein abundance in proximal tubular BBM and in NaPi-IIc mRNA. In addition, we documented the presence of a third Na-coupled Pi transporter in the renal BBM, PiT-2, whose abundance is also decreased by dietary K deficiency in rats and in mice. Finally, electron microscopy showed subcellular redistribution of NaPi-IIc in K deficiency: in control rats, NaPi-IIc immunolabel was primarily in BBM microvilli, whereas, in K-deficient rats, NaPi-IIc BBM label was reduced, and immunolabel was prevalent in cytoplasmic vesicles. In summary, our results demonstrate that decreases in BBM abundance of the phosphate transporter NaPi-IIc and also PiT-2 might contribute to the phosphaturia of dietary K deficiency, and that the three renal BBM phosphate transporters characterized so far can be differentially regulated by dietary perturbations.

ABSTRACT Many viruses inhibit or retard apoptosis, a strategy that subverts one of the most ancient antiviral mechanisms. M11L, a myxoma virus-encoded antiapoptotic protein, has been previously shown to localize to mitochondria and block apoptosis of virus-infected cells (H. Everett, M. Barry, S. F. Lee, X. J. Sun, K. Graham, J. Stone, R. C. Bleackley, and G. McFadden, J. Exp. Med. 191:1487-1498, 2000; H. Everett, M. Barry, X. Sun, S. F. Lee, C. Frantz, L. G. Berthiaume, G. McFadden, and R. C. Bleackley, J. Exp. Med. 196:1127-1139, 2002; and G. Wang, J. W. Barrett, S. H. Nazarian, H. Everett, X. Gao, C. Bleackley, K. Colwill, M. F. Moran, and G. McFadden, J. Virol. 78:7097-7111, 2004). This protection from apoptosis involves constitutive-forming inhibitory complexes with the peripheral benzodiazepine receptor and Bak on the outer mitochondrial membrane. Here, we extend the study to investigate the interference of M11L with Bax activation during the process of apoptosis. Myxoma virus infection triggers an early apoptotic signal that induces rapid Bax translocation from cytoplasm to mitochondria, despite the existence of various viral antiapoptotic proteins. However, in the presence of M11L, the structural activation of Bax at the mitochondrial membrane, which is characterized by the occurrence of a Bax conformational change, is blocked in both M11L-expressing myxoma-infected cells and M11L-transfected cells under apoptotic stimulation. In addition, inducible binding of M11L to the mitochondrially localized Bax is detected in myxoma virus-infected cells and in M11L/Bax-cotransfected cells as measured by immunoprecipitation and tandem affinity purification analysis, respectively. Importantly, this inducible Bax/M11L interaction is independent of Bak, demonstrated by the complete block of Bax-mediated apoptosis in myxoma-infected cells that lack Bak expression. Our findings reveal that myxoma M11L modulates apoptosis by multiple independent strategies which all contribute to the blockade of apoptosis at the mitochondrial checkpoint.

A nonselective cation channel specifically activated by intracellular Na+ was identified in cell-free patches taken from cultured lobster olfactory projection neurons. Na+ reversibly activates the channel in a concentration-dependent manner, with a “half-effect” Na+ concentration of 76.4 mM at −60 mV. The conductance of the channel is 32 pS. The channel is permeable to both alkali metal (Li+ > Na+ > K+ > Rb+ > Cs+) and divalent (Ca2+ > Mn2+ > Sr2+ > Mg2+ > Ba2+ > Na+) cations. The presence of a channel with the ability to generate plateau potentials suggests that the channel may potentially contribute to oscillatory behavior in these olfactory interneurons.

The article deals with the investigation of the communicative and linguistic features of the comic discourse on the basis of the English-language fictional works by M. Spark, R. Dahl, K. Vonnegut Jr, I. Shaw, R. Carver and K. Barry. The author represents the description of a comic discourse in terms of Linguistic Pragmatics. Extra-linguistic components of the literary text creating and actualizing, which include pragmatic characteristics, social and cultural factors, psychological peculiarities, cognitive component and paralinguistic supplement, are described. The article also provides a detailed classification of comic discourse linguistic components on the material of belles-letters communication in the works of aforementioned writers. All linguistic characteristics within five groups, according to the violation/lack/incompleteness of the discourse certain communicative qualities disclosure, are distinguished. Besides, there is a thorough analysis of the presupposition as a constituent part in the comic discourse definition. The article dwells on the interlocutors’ micro and macro presuppositions introducing, the absence of which leads to non-cooperative communication with, predominantly, a comic effect. The results of the experiment show that the participants’ (readers’ particularly) background knowledge helps them interpret a literary discourse. The article continues the study of special special means and techniques of comic effect producing that provides a further understanding of the authors’ individual styles.

Adapun jenis tes yang digunakan adalah: (1) koordinasi mata-tangan dengan tes memantulkan bola ke tembok dari Mulyono B, (1988:78), (2) kekuatan otot perut dengan tes Sit-Up dari Barry L. Jhonson & Jack K, Nelson, (1986:133), (3) Tes Persepsi Kinestetik bidang vertical untuk mengukur persepsi kinestetik (Barry L. Johnson and Jack K. Nelson, 1986: 189-190), (4) Tes passing atas yang digunakan dalam penelitian ini menggunakan tes passing atas dari Suharno HP,( 1984 : 71 ). Berdasarkan analisis data dan pengujian hipotesis yang telah dilakukan, maka simpulan yang dapat diperoleh adalah: (1) Ada hubungan yang signifikan antara Koordinasi mata tangan dengan Passing atas, rhitung = 0.410 > rtabel 5% = 0,361. (2) Ada hubungan yang signifikan antara Kekuatan otot perut dengan Passing atas, rhitung = 0.402 > rtabel 5% = 0,361. Thirteen porcelains assigned to Sèvres factory productions and a few references to the other contemporary factories (Chantilly, Limoges, and Venice) have been studied on-site with a portable X-ray fluorescence (pXRF) spectrometer in order to control the provenance attribution. Characteristic XRF signals of major elements (Si, Ca, K, Pb) and minor/trace (Au, Bi, As, Ti, Co, Cu, Zn, Ni, Y, Zr, Rb, and Sr) elements are compared for the paste, blue mark, various glazed (colored) areas, and gilding. The comparison of peak intensities clearly distinguishes different types of hard- and soft-paste porcelain, made from either similar or distinct raw materials. The analysis of transition elements associated with cobalt identifies three types of cobalt blue and reveals that du Barry-style decoration on certain artifacts was typical of 19th-century production. On-site comprehensive studies of the two famous Etruscan-style breast bowls from Rambouillet Castle dairy, using pXRF and Raman spectroscopy, confirm the use of soft-paste porcelain for the cup and hard-paste for its support, providing detailed information on the use of gold nanoparticles in the burgundy-colored decoration.

En el año 2022 se otorgó el premio Nobel de química a Carolyn R. Bertozzi, Morten Meldal y K. Barry Sharpless por sus aportaciones en el desarrollo de la química clic y la química bioortogonal. La trascendencia de este tipo de reacciones radica en que, por su sencillez, selectividad, eficiencia y tolerancia a diferentes medios, incluido el acuoso, ha impactado no sólo en la química sintética de moléculas complejas y materiales, sino también en el área farmacéutica, ya que, aplicando este tipo de reacciones es posible formar conjugados con biomoléculas como proteínas, abriendo la puerta a la exploración y comprensión de diversos procesos biológicos en los organismos vivos para acceder así a terapias dirigidas más eficientes. En el Instituto de Investigaciones en Materiales de la UNAM hemos aplicado esta reacción de manera exitosa para modificar directamente algunas moléculas bioactivas como la curcumina (antioxidante natural), mejorando sus propiedades fisicoquímicas como solubilidad y estabilidad química. Asimismo, aplicando reacciones clic se sintetizaron derivados de β-ciclodextrina como nuevos acarreadores de fármacos, incorporando sustituyentes altamente ramificados llamados dendrones que confieren un adecuado balance hidrofílico/hidrofóbico. Estas modificaciones nos permitieron optimizar su paso a través de las membranas celulares por difusión, y su capacidad para encapsular fármacos hidrofóbicos.

Abstract Ovarian cancer is the second leading cause of cancer-related mortality among gynaecological cancers in the Western world. CAR-T cell therapy is an adoptive immunotherapy used to treat some blood cancers. This study has investigated whether leucine-rich repeat-containing G protein-coupled receptor 5 (LGR5) is a suitable target for CAR-T cell therapy for ovarian cancer. We assessed the cytotoxic effects of LGR5-targeting CAR-T cells on a range of ovarian cancer cell lines and primary serous ovarian cancer cells derived from patient ascites (at diagnosis and following relapse with chemotherapy resistant disease) in monolayer culture, 3D spheroid assays and patient-derived explant assays. OVCAR3 and a chemoresistant patient-derived xenograft (PDX) models have been used to assess whether LGR5-targeting CAR-T cells can inhibit growth and metastasis in vivo. We found that LGR5 is highly expressed in high-grade serous ovarian carcinoma (HGSOC) cancer cell lines and LGR5 expression was increased in chemotherapy resistant primary ovarian cancer cells. We demonstrated that LGR5-targeting CAR-T cells were cytotoxic and significantly inhibited the survival of ovarian cancer cell lines and chemotherapy-resistant primary ovarian cancer cells with high LGR5 expression levels in both monolayer culture and 3D spheroids compared to un-transduced CD3+ T cells. In patient-derived explant assays, we observed increased apoptosis in HGSOC tissues expressing high LGR5 following treatment with LGR5-targeting CAR-T cells. In vivo, LGR5-targeting CAR-T cell treatment decreased the tumour burden in mice with OVCAR3 xenografts and a chemotherapy-resistant PDX model with high LGR5 expression, compared to CD3+ T cells. Two methods of assessing the risk of species introduction by ballast water are discussed, species-specific and environmental similarity assessments, each for alignment with four proposed principles of risk-based resource management: (i) society accepts that low risk scenarios exist; (ii) risk assessment is capable of identifying low risk scenarios; (iii) risk mitigation strategies exist; and (iv) mitigation costs are less than the cost of performing risk assessment. All four principles were met in some circumstances for both methods. Species-specific ballast water risk assessment is best suited to situations where the assessment can be restricted to a limited set of harmful species on journeys within bioregions where ballast water is a small component of natural genetic exchange. Environmental similarity risk assessment is appropriate for journeys that start and end in locations which have very little or no natural genetic exchange, such as journeys between non-contiguous bioregions. Because a large number of species are not assessed individually, environmental match assessments necessarily will be restricted to fundamental variables such as temperature and salinity. A number of bioregion classifications have been identified in the world’s oceans, some of which at a scale that may be appropriate for ballast water management. The suitability of any particular classification, however, needs further examination.
