Catalytic nanoreactors, prepared by the Layer-by-Layer encapsulation of zeolite H-Beta, were used in the dynamic kinetic resolution (DKR) of 1-phenylethanol and 1-indanol. Yields for both alcohols were clearly above 50%, with ee’s over 85%, indicating successful protection of the pH-sensitive enzyme, candida antarctica lipase B, from the acidic zeolite. The first volume of work (Chapter 2) describes the gold-catalysed double alkylation of indole by enynones. An array of different cyclopropanes have been synthesised, including the structurally simple 1-phenylcyclopropanol. These were synthesised in yields upwards of 60%, using the well published Kulinkovich reaction. From 1-phenylcyclopropanol, variations of the cyclopropane core structure were synthesised, creating species ideal for palladium cross coupling reactions, such as 1-phenylcyclopropyl methanesulfonate and 1-phenylcyclopropyl 4-methylbenzenesulfonate. Amine transaminases are enzymes that catalyze the mild and selective formation of primary amines, which are useful building blocks for biologically active compounds and natural products. In order to make the production of these kinds of compounds more efficient from both a practical and an environmental point of view, amine transaminases were incorporated into multi-step one-pot reactions. With this kind of methodology there is no need for isolation of intermediates, and thus unnecessary work-up steps can be omitted and formation of waste is prevented. Synthesis and flash vacuum pyrolysis (FVP) of stabilised phosphorus ylides containing an o-amino functionalised benzene ring has been examined for the first time. Model studies using N-methyl-N-tosyl and N-mesyl-N-methyl ylides showed that the ylides could be prepared, although yields were variable, and had the expected spectroscopic properties. Upon FVP, however, the expected loss of Ph₃PO and the sulfonyl group was accompanied by unexpected transfer of the reactive site from nitrogen to carbon giving 3- substituted quinolines rather than the expected indole products.
