Hydrogenases are a family of enzyme that catalyses the bidirectional interconversion of H<sup>+</sup> and H<sub>2</sub>. There are two major classes of hydrogenases: the [NiFe(Se)]- and [FeFe]-hydrogenases. Both of these benefit from characteristics which would be advantageous to their use in technological devices for H<sub>2</sub> evolution and the generation of energy. These features are explored in detail in this thesis, with a particular emphasis placed on defining the conditions that limit the activity of hydrogenases when reducing H<sup>+</sup> to produce H<sub>2</sub>. Electrochemical Impedance Spectroscopy (EIS) was used to develop a novel methodology for studying ionic interaction with lipids arranged in a lipid cubic phase (LCP). Studying different types of ions, both cations and anions, validated the method. Shewanella oneidensis MR-1 is capable of respiring on an extraordinarily large and diverse array of terminal electron acceptors, including extracellular insoluble metal oxides and electrodes. The ability to perform extracellular electron transfer (EET) has sparked great interest in MR-1 which has become both a model organism for fundamental research into EET and a candidate microbe for microbial electrochemical systems. Abstract:
This dissertation relates to the application of density functional theory to the design of novel nanoelectrocatalysts for various electrochemical reduction reactions such as carbon dioxide reduction reactions, carbon monoxide reduction reactions and nitrogen reduction reactions. Many electrocatalysts with high activity, excellent selectivity and stability were designed and engineered using first principle calculations. These findings could potentially guide the experimentalists for creating clean and sustainable energy resources.

<p>Different sample preparation steps prior to a detection method are often essential in analytical chemistry. In this thesis, both static extractions and on-line coupled solid-phase extractions have been studied in combination with different detection techniques. Aspects of performing sample preparations in miniaturized analytical devices and the development of poly(dimethylsiloxane) (PDMS) microchips are discussed.
