Abstract:
Потребление заменителей молока на растительной основе быстро распространилось по всему миру благодаря их многочисленным положительным воздействиям на организм человека. Эти напитки предпочитают люди с аллергией на коровье молоко, непереносимостью лактозы и гипохолестеринемией. Востребованность в таких продуктах связана также со стремлением людей вести здоровый образ жизни, с религиозными воззрениями, гуманным отношением к животным. В работе представлен краткий анализ отечественного рынка альтернативных продуктов. Рассмотрена возможность их ферментации молочнокислыми бактериями для получения новых видов продуктов. Были изготовлены образцы на овсяной, гречневой и рисовой основе с применением моно- и комплексных заквасок. Выявлена разная степень адаптации молочнокислых бактерий в зависимости от состава заквасок и типа субстрата. Брожение протекало более активно при использовании поликомпонентных заквасок и овсяной основы жирностью 3,2 %. Комплексный анализ органолептических, структурно-механических свойств и стабильности образцов показал необходимость использования пектина в качестве стабилизатора структуры. Результаты исследования влагоудерживающей способности и склонности к расслоению продукта также показали положительное влияние полисахарида. Всестороннее изучение образцов позволило установить, что продукт на овсяной основе с м.д.ж. 3,2 % является наиболее предпочтительным. The consumption of plant-based milk substitutes has spread rapidly around the world due to their many positive effects on the human body. These drinks are preferred by people with cow's milk allergies, lactose intolerance and hypocholesterolemia. Demand for such products is also associated with the desire of people to lead a healthy lifestyle, religious beliefs, humane treatment of animals. The paper presents a brief analysis of the domestic market of alternative products. The possibility of their fermentation by lactic acid bacteria to obtain new types of products is considered. Samples on oat, buckwheat and rice bases with the use of mono- and complex starter were made. Different degree of adaptation of lactic acid bacteria depending on the composition of starter and type of substrate was revealed. Fermentation was more active when using polycomponent starter and oat base with fat content of 3.2 %. Complex analysis of organoleptic, structural and mechanical properties and stability of the samples showed the necessity of using pectin as a stabilizer of the structure. The results of the study of moisture-holding capacity and tendency to stratification of the product also showed the positive effect of polysaccharide. Comprehensive study of the samples revealed that the oat-based product with a fat content of 3.2 % was the most preferred.

AbstractWith potential storage of several hundred years underground in a geological disposal facility (GDF) before closure, there is a requirement for radioactive waste packages to perform adequately. Over the past 25 years, cementitous grouts based on blast furnace slag (BFS) and ordinary Portland cement (OPC), have been used in the UK to immobilize intermediate-level waste (ILW), Magnox swarf, and currently such wasteforms are in surface storage awaiting geological disposal. Magnox fuel cladding will slowly corrode when encapsulated in alkaline cementitious grouts to produce hydrogen gas and an expansive corrosion product. Expansive corrosion products may lead to degradation of the wasteform and, if extensive, could affect the container. This study investigated the acute and chronic rates of corrosion of unirradiated Magnox swarf encapsulated in a BFS/OPC grout over a range of temperatures (25, 40, 60, 75 and 90°C) for curing times of up to 2½ years. Structural product degradation starts to develop as the tensile strength of the grout is approached by the expansive forces generated. Deformation of some experimental containers was also noted. An estimate of the time taken for a grouted product to fracture from this study due to the corrosion solely of Magnox is 350 years for storage at 25°C. Although not fully described in this paper, the main cement phases were calcium silicate hydrate, portlandite and gehlenite; the main product of Magnox corrosion was brucite. The curing temperature did not affect the compositions of the Magnox metal, corrosion product or grout.

Abstract:
Traditionally, biological methods are used to leaven dough semi-finished products in the production of bakery products. However, today consumers are increasingly including yeast-free bakery products in their diet. Given the growing demand for yeastfree bakery products, it is advisable to expand the range of products in this category. To ensure the formation of a developed crumb in yeast-free bakery products, chemical leavening agents are used in their production. Yeast-free bakery products were made using various fermented dairy products: ayran (2,0% fat), kefir (2,5% fat), bifidoyogurt (1,5% fat), and ryazhenka (4,0% fat). To determine the most effective fermented dairy product for the production of high-quality yeast-free bakery products, trial baking was conducted. Experimental dough samples were kneaded using two types of chemical leavening agents - sodium bicarbonate and ammonium carbonate. The dough was prepared using the straight dough method. The duration of dough resting after kneading was 20-30 minutes. The dough pieces were shaped into "okrayets" (small loaves): the dough was rolled to a thickness of 1,5 cm and the dough pieces were cut out with a cutter, placed on a sheet, and baked in a rack oven at 200-240°C for 16-18 minutes. The research results established that it is advisable to use ryazhenka in the production of yeast-free bakery products using fermented dairy products. At the same time, sodium bicarbonate should be included in the recipe to leaven the crumb. Such a product had the highest comprehensive quality index. Based on organoleptic and microbiological indicators, the product was found to have a shelf life of no more than 48 hours. To improve the taste and aromatic properties of the product, 3% malted barley extract and 4% white crystalline sugar were included in its composition. The developed yeast-free okrayets with ryazhenka was characterized by a higher content of bisulfitebinding substances, indicating an improved aroma of the product and a darker color of the crust and crumb. The developed product meets the body's needs for proteins by 31,5%, fats by 10,5%, carbohydrates by 49,0%, and dietary fiber by 18,3%.

The quark(gluon)–hadron duality constitutes a basis for the theoretical treatment of a wide range of inclusive processes — from hadronic τ decays and Re+e- to semileptonic and nonleptonic decay rates of heavy flavor hadrons. A theoretical analysis of these processes is carried out by using the operator product expansion in the Euclidean domain, with subsequent analytic continuation to the Minkowski domain. We formulate the notion of the quark(gluon)–hadron duality in quantitative terms, then classify various contributions leading to violations of duality. A prominent role in the violations of duality seems to belong to the so-called exponential terms which, conceptually, may represent the (truncated) tail of the power series. A qualitative model, relying on an instanton background field, is developed, allowing one to get an estimate of the exponential terms. We then discuss a number of applications, mostly from heavy quark physics.

Global climate change is resulting in a wide range of biotic responses, including changes in diel activity and seasonal phenology patterns, range shifts polewards in each hemisphere and/or to higher elevations, and altered intensity and frequency of interactions between species in ecosystems. Oak (Thaumetopoea processionea) and pine (T. pityocampa) processionary moths (hereafter OPM and PPM, respectively) are thermophilic species that are native to central and southern Europe. The larvae of both species are gregarious and produce large silken ‘nests’ that they use to congregate when not feeding. During outbreaks, processionary caterpillars are capable of stripping foliage from their food plants (oak and pine trees), generating considerable economic damage. Moreover, the third to last instar caterpillars of both species produce copious hairs as a means of defence against natural enemies, including both vertebrate and invertebrate predators, and parasitoids. These hairs contain the toxin thaumetopoein that causes strong allergic reactions when it comes into contact with human skin or other membranes. In response to a warming climate, PPM is expanding its range northwards, while OPM outbreaks are increasing in frequency and intensity, particularly in northern Germany, the Netherlands, and southern U.K., where it was either absent or rare previously. Here, we discuss how warming and escape from co-evolved natural enemies has benefitted both species, and suggest possible strategies for biological control.

In this article, we examine the impact of banking expansion on income growth in India. The banking expansion indices have been calculated across the region and states/Union Territories, providing the insight that all the regions, excluding the western region, are exhibiting banking expansion indices in the low range. The state-wise analysis indicates that all states exhibit a low-range index, excluding the state of Maharashtra and the UTs of Delhi and Chandigarh. Further, for the examination of the linkage between banking expansion and income growth, a panel data set was prepared for the 23 states/UTs over the period from 1990 to 2015. The panel data regression analysis approach was applied for the estimation of the regression model. It is apparent from the results that the banking expansion has significant and positive effects on credit disbursement. The results indicate that a one crore increase in deposit mobility causes 0.81 crores of increase in credit disbursement. Moreover, credit disbursement and deposit mobilization have a substantial and positive effect on the Net State Domestic Product. Moreover, a 1 percent increase in credit causes a 0.46 percent increase in NSDP, and a 1 percent increase in deposits causes a 0.57 percent increase in NSDP. Further, Net State Domestic Product has a significant and positive effect on the income of individuals. It is evident that a 1 percent increase in NSDP causes a 0.54 percent increase in per capita NSDP, while a 1 percent increase in capital expenditure causes a 0.13 percent increase in per capita NSDP.

3023 Background: Axi-cel (formerly KTE-C19) is an autologous anti-CD19 chimeric antigen receptor (CAR) T cell therapy. ZUMA-1 is a multicenter, registrational trial of axi-cel in patients (pts) with refractory aggressive non-Hodgkin lymphoma. In a prespecified interim analysis, ZUMA-1 met its primary endpoint, with a 76% objective response rate and a 47% complete response rate ( Blood 2016;128:LBA-6). Post-treatment CAR T cell blood levels were associated with objective response. Here, we describe novel associations between product characteristics and CAR T cell levels in pts. Methods: CAR T cell characteristics in axi-cel produced from 62 pts were analyzed by flow cytometry and modeled against CAR T cell levels. In vivo CAR T cell levels were measured by qPCR. T cell expansion during production (fold expansion/total days in culture) was compared with CAR T cell blood levels, using a partition analysis with expansion rates of ≥1 vs < 1. Wilcoxon 2-sample test and linear regression were used. Results: Axi-cel contained CCR7+ T cells (median, 42%; range, 15–73%), with naïve (CD45RA+/CCR7+; median, 12%; range, 1–57%), central memory (CD45RA−/CCR7+; median, 29%; range, 12–49%) phenotypes, and more differentiated CCR7− effector memory and effector T cells. On infusion, CAR T cells expanded rapidly, reaching peak levels within 2 weeks (median, 43 cells/μL; range, 1–1513), and were also measurable in all pts at 1 month (median, 2 cells/μL; range, 0.03–89). The CCR7+/CCR7− T cell ratio in axi-cel associated positively with peak ( P =0.001) and cumulative ( P =0.003) CAR T cell levels through 1 month. Axi-cel lots that expanded more rapidly during production (≥1.0-fold/d; n = 18/62) associated with higher cumulative levels of CAR T cells ( P =0.03). Other product characteristics, eg, CD4/CD8 ratio or number of infused T cells, were not significantly associated with CAR T cell blood levels. Conclusions: An association was observed between CAR T cell expansion in vivo and both the T cell growth rate during production and product cell phenotype pretreatment. A key attribute of axi-cel product was the presence of CCR7+ naïve/central memory T cells, without upfront T cell subset selection. Clinical trial information: NCT02348216.

This study examined the effect of product line strategy on the performance of NASCO Plc, located in Jos, Plateau State. The research employed a survey research design with primary data collected through questionnaires administered to employees of NASCO Plc. The total population was 1,131, and the Taro Yamane method was used to determine a sample size of 295. Out of the 295 distributed questionnaires, 250 were returned, representing an 85% response rate. To test the study's hypotheses, Least Squares regression analysis was employed. The findings revealed that line extension had a positive impact on the support for NASCO Plc. Similarly, line modernization significantly enhanced the demand for the company's products. Additionally, product line strategies were found to substantially influence the business expansion and profitability of NASCO Plc. Specifically, the alignment of the product line ("line fit") played a critical role in improving the company's profitability. The study concluded that product line strategies, including line extension, modernization, trimming, and product fit, positively influence the growth, sustainability, product demand, business expansion, and profitability of NASCO Plc. It was recommended that the management of NASCO Plc invest in extending and enhancing their product line to drive further growth. This can be achieved by adopting modern technologies to improve product quality. Additionally, the company should continue to optimize processes such as packaging and labeling to expand its product range.

This study examined the extrusion cooking of pearl millet and maize blends, focusing changes in physical properties and the effects of incorporating spinach and beetroot powders. Single screw extruder powered by 5.0 kW motor and a rotational speed of 1450 rpm was used to produce extrudates. Pearl millet and maize grits having 13% moisture content and mixed in the ratio of 70:30, 50:50, and 30:70 on weight basis were used for extrusion. The study adopted simplex-lattice mixture design and augmented mixture design for blend formulations. The effect of blend proportions was studied on physical properties of extrudates such as expansion ratio, bulk density, volume mass anddiameter. Extrudates expansion ratio decreased with higher pearl millet proportions, while maize improved expansion ratio. The expansion ratio was highest for 100% maize (4.64), decreasing with pearl millet addition. Spinach and beetroot powders mixed in the range of 1- 4% also reduced expansion and increased bulk density, attributed to fibre content which disrupt starch gelatinization. Optimal powder levels (3%) resulted in desirable physical properties. Bulk density peakedat 0.057 g cm-³ and 0.071 g cm-³ for spinach and beetroot powders, respectively, at 4%. Findings highlighted trade-offs between density and expansion when incorporating vegetablepowders into extruded products. The study underscores the potential of optimized formulations for nutritious, shelf-stable, and texturally appealing snacks, addressing health-conscious consumer demands while ensuring product quality.

Huntington's disease (HD) is an autosomal dominant neurodegenerative disorder caused by a CAG trinucleotide repeat expansion in the HD gene. The expanded repeats are translated into an abnormally long polyglutamine tract close to the N-terminus of the HD gene product (‘huntingtin’). Studies in humans and mouse models suggest that the mutation is associated with a deleterious gain-of-function. Several studies have suggested that the large huntingtin protein is cleaved to produce a shorter N-terminal fragment containing the polyglutamine expansion, and that the polyglutamine expansion causes the protein fragment to misfold and form aggregates (inclusions) in the nuclei and processes of neurons. It is likely that neurotoxicity is caused by the misfolded protein in its soluble form, and/or in aggregates, and/or in the process of aggregation. A wide range of potential mechanisms for neurotoxicity have been proposed, including caspase activation, dysregulation of transcriptional pathways, increased production of reactive oxygen species, and inhibition of proteasome activity. In this review we consider the current status of research in the field and possible mechanisms whereby the HD mutation might result in neurodegeneration.

The purpose of this article is to analyze the indicators of the assortment of juice products in the stores of three retail chains, namely in the Auchan, Globus, and Perekrestok stores, as well as to identify the directions of their assortment policy in this subcategory. It has been established that the assortment of the subcategory of juice products is represented by 12 types, 44 trademarks, and 301 trademark modifi cations, which makes it possible to classify it as comprehensive. 32% of the branded assortment consists of brands. His research analyzes the halal branding strategy of PT Indofood CBP Sukses Makmur Tbk in supporting global expansion in the international Muslim market. The main focus of the research is on the use of product innovation, halal certification, and local culture-based promotion as strategies to increase competitiveness and loyalty of Muslim consumers. By integrating sharia principles and Islamic business practices, Indofood has successfully expanded its product range to more than 80 countries, including the Middle East, Africa, and Asia. The results show that globally recognized halal certification and product customization to local preferences are the main factors for the success of this strategy. However, challenges such as variations in halal regulations between countries and increasing global competition are still issues that need to be addressed. This study provides recommendations for other multinational companies in optimizing halal branding as a strategic tool to expand the international Muslim market. Abstract Introduction Omidubicel-onlv is a nicotinamide (NAM) -modified allogeneic hematopoietic progenitor cell therapy derived from cord blood. NAM expansion increases the number of stem and progenitor cells, inhibits differentiation, and potentiates migration, bone marrow homing, and engraftment efficiency. A global phase 3 trial demonstrated faster hematopoietic recovery, reduced rates of infection, and shorter hospitalizations for patients randomized to omidubicel-onlv compared to standard umbilical cord blood (UCB). Omidubicel-onlv is manufactured for each patient from a single, appropriately matched cord blood unit (CBU). The inherent donor variability as well as the different processing and testing methods used by different Cord Blood Banks (CBBs) present challenges in maintaining a consistent product. Objectives This extensive study was conducted to confirm the robustness of the manufacturing process and the proven acceptable range for the critical and key process parameters. Methods Seven process parameters were evaluated in total. Of these, three parameters were related to the CD34+ expansion phase: CD34+ seeding concentration, incubation temperature, and CO2 levels. Four additional parameters were related to final product formulation and cryopreservation: hold times during harvest and formulation, cell concentration in the drug product, DMSO concentration, and storage temperature. The impact of each process parameter on product performance was assessed by a side-by-side comparison of its target value (control) versus the tested range. Each parameter was evaluated using at least three batches derived from three CBUs. Statistical analysis of the coefficient of variation or recovery against the control was applied to the data sets. Results Statistical analyses demonstrated an overall variability of ≤30% in critical attributes including fold expansion, viability, phenotyping, and recovery of cells post-cryopreservation and stability. Discussion The omidubicel-onlv manufacturing process is robust, overcoming the variability of batches derived from different donors, different processing and testing methods at CBBs, and manufacturing process parameters.

This research seeks to determine the formulation of yellow pumpkin bread. This research used a Completely Randomized Design (CRD) with six different compositions of yellow pumpkin flour, which were 0% (P0), 5% (P1), 10% (P2), 15% (P3), 20% (P4), and 25% (P5). The data obtained was then analyzed using One-way ANOVA and continued with the Duncan Multiple Range Test (DMRT). The research shows that yellow pumpkin flour affects physical properties (specific volume, texture, bread expansion, and bread color intensity) and sensory properties (hedonics and hedonic quality). The chosen formulation of yellow pumpkin bread was P1 (95% wheat flour: 5% pumpkin flour). It has bread expansion at 73.15%, specific volume at 3.46%, texture at 87.44℅, Color (L) at 67.4%, (a) 2.10%, (b) 40.36%, with hedonic quality had a yellowish-white color, tastes somewhat typical of pumpkin, smells somewhat typical of pumpkin, has a soft texture, and the uniform pores.

BackgroundEx-vivo expanded tumour infiltrating lymphocytes (TIL) show promise in delivering durable responses among several solid tumour indications. However, characterising, quantifying and tracking the active component of TIL therapy remains challenging as the expansion process does not distinguish between tumour reactive and bystander T-cells. Achilles Therapeutics has developed ATL001, a patient-specific TIL-based product, manufactured using the VELOS™ process that specifically targets clonal neoantigens present in all tumour cells within a patient. Two Phase I/IIa clinical trials of ATL001 are ongoing in patients with advanced Non-Small Cell Lung Cancer, CHIRON (NCT04032847), and metastatic or recurrent melanoma, THETIS (NCT03997474). Extensive product characterisation and immune-monitoring are performed through Achilles’ manufacturing and translational science programme. This enables precise quantification and characterisation of the active component of this therapy – Clonal Neoantigen T cells (cNeT) – during manufacture and following patient administration, offering unique insight into the mechanism of action of ATL001 and aiding the development of next generation processes.MethodsATL001 was manufactured using procured tumour and matched whole blood from 8 patients enrolled in the THETIS (n=5) and CHIRON (n=3) clinical trials. Following administration of ATL001, peripheral blood samples were collected up to week 6. The active component of the product was detected via re-stimulation with clonal neoantigen peptide pools and evaluation of IFN-γ and/or TNF-α production. Deconvolution of individual reactivities was achieved via ELISPOT assays. Immune reconstitution was evaluated by flow cytometry. cNeT expansion was evaluated by restimulation of isolated PBMCs with peptide pools and individual peptide reactivities (ELISPOT).ResultsThe median age was 57 (range 30 – 71) and 6/8 patients were male. The median number of previous lines of systemic anti-cancer treatment at the time of ATL001 dosing was 2.5 (range 1 – 5). Proportion of cNeT in manufactured products ranged from 0.20% - 77.43% (mean 26.78%) and unique single peptide reactivities were observed in 7 of 8 products (range 0 – 28, mean 8.6). Post-dosing, cNeTs were detected in 5/8 patients and cNeT expansion was observed in 3/5 patients.ConclusionsThese data underscore our ability to sensitively detect, quantify and track the patient-specific cNeT component of ATL001 – during manufacture and post dosing. The paper presents the results of research of the influence of product range parameters on the indicators of development of the design capacities of precast construction plants. The quantity and concrete consumption of products in terms of 1 m2 of total area, as well as the circulation of products, have been taken as the analyzed parameters of product range. It is noted that calculations of production capacity and basic technical and economic indicators of production at the stage of technological design are based on the product range of a typical block section of a house series. At the same time, architectural individuality, flexibility of planning and design solutions of prefabricated houses buildings in industrial design require a variety of modifications of block sections, which is inextricably linked with the growth of the range of manufactured products. Expansion of the range of products for modifications of block sections in the production program of plants is an objective regularity that should be taken into account in the technological design of new or reconstruction of existing precast constructions plants and planning production programs for housing construction. Significant fluctuations in the parameters of the product range in the production programs of plants have been recorded. A regression analysis of the parameters of the range products of production programs of precast construction plants has been carried out. The dependence of product range on the share of dotted houses in housing construction programs has been established. The main parameters of houses determining fluctuations in the range of products have been identified, which include the number of storeys and the number of sections in the house configuration. Taking into account the fact of the significant influence of the product range on the production capacity of plants, the dependence of the indicator of compliance of the concrete capacity of products with 1 m2 of total area on the production capacity indicators has been established. The reliability of the obtained mathematical models is confirmed by the results of statistical processing of research results and verification of models in calculations of the current production capacity of plants.

One apheresis product from each patient was prepared using the Isolex 300 Magnetic Cell Separation System (Baxter Immunotherapy, Irvine, CA) to obtain CD34+ cells. These cells were then cultured in gas permeable bags containing serum-free X-VIVO 10 (BioWhittaker, Walkersville, MD) medium supplemented with 1% human serum albumin and 100 ng/mL PIXY321. At day 12 of culture the mean fold expansion was 26x with a range of 6 to 64x. One patient's cells did not expand because of a technical difficulty. The final cell product contained an average of 29.3% CD15+ neutrophil precursors with a range of 18.5% to 48.1%. The patients underwent high-dose chemotherapy with cyclophosphamide, carboplatin, and thiotepa. On day 0, the cryopreserved PBPCs were reinfused and on day +1 the 12-day cultured cells were washed, resuspended, and reinfused into eight of nine patients. One patient was not infused with cultured cells. The mean number of cultured cells reinfused was 44.6 x 10(6) cells/kg with a range of 0.8 to 156.6 x 10(6) cells/kg. No toxicity was observed after reinfusion. The eight patients have recovered absolute neutrophil counts > 500/microL on a median of 8 days (range 8 to 10 days); the median platelet transfusion independence occurred on day 10 (range 8 to 12 days) and platelet counts > 50,000/microL were achieved by day 12 (range 9 to 14) for the seven patients whose platelet counts could be determined. The analysis of the chemical composition of wheat and chickpea flour, as well as the glycemic index of all components of the recipe of the developed flour culinary product is presented. The possibility of partial and complete replacement of wheat flour with chickpea flour with the addition of caraway, lemon peel and potato starch is studied. It is found that the inclusion of lemon peel in the composition of chickpea flour pancakes improves not only the organoleptic characteristics of products, but also reduces its total energy content, while expanding the range of products included in a gluten-free diet.

Maintaining a stable demand for reduced goods is an obligatory condition for strategizing activities, a guarantee of development. The presented research reflects the theoretical foundations of enterprise management, in terms of building a strategy; the assortment features for both commodity and trade ones are disclosed; the authors highlighted the specifics of agricultural production through strategic management and its goal setting. In the main part of the work, there is a block focused on the study of the assortment within the framework of the agricultural enterprise management, based on the product range optimization. This is supported by the conclusions that this approach is a source of diversification of an agricultural enterprise production activity and takes into account both the existing market needs and the capabilities of the enterprise itself when creating the necessary conditions for its development, ensuring competitiveness not only in local markets, but also includes a possibility to expand its market zone. The assortment of goods was studied in a separate block on the example of a specific agricultural enterprise - the greenhouse complex JSC “Sukhovskiy”, a member of the Siberian agricultural complex of the brand “Em Rodnoe”. The assessment of the main economic indicators of the activities of JSC “Sukhovskiy” was carried out. For the purpose of practical testing of the developed directions of strategizing the product range of JSC “Sukhovskiy”, there was proposed a variant of its expansion by introducing roses into the “line” of production as the most popular and demanded flowers in high demand among the population. Human adipose-derived stem cells (hADSCs) are a promising source of autologous stem cells for personalized cell-based therapies. Culture expansion of ADSCs provides an attractive opportunity for liver cirrhosis patients. However, safety and stability issues can pose big challenges for personalized autologous stem cell products. In the present study, we addressed whether the commercial production program could provide a consistent product for liver cirrhosis therapy. We collected adipose tissue from three human donors by lipoaspirate and isolated ADSCs, which were expanded in culture to reach 1 × 10 8 cells (an approximately 1,000-fold expansion) within four passages. We then examined their morphology, chromosome stability, surface markers, and differentiation ability after culture. Next, we explored their therapeutic potential using a rat model of thioacetamide-induced liver cirrhosis. Culture-expanded ADSCs were injected intrahepatically, and their biodistribution was tracked by immunohistochemistry using an antibody against human mitochondria. Finally, we tested for tumor development by subcutaneously injecting a 100-fold dose range of cultured ADSCs into immunocompromised mice. Taken together, we find that culture expansion of autologous ADSCs is a potentially suitable stem cell product for personalized cell-based therapy for patients with liver cirrhosis.

In this paper, the regioselective reactions of kaolinite and methoxy-modified kaolinite (MeO-Kaol), methanol-expanded kaolinite, with octadecyltrimethylammonium salts are compared. This study mainly concerns the reactions of kaolinite or MeO-Kaol with octadecyltrimethylammonium chloride (C18TAC) in methanol and the subsequent exhaustive washing of the resultant products with ethanol. X-ray diffraction patterns of the products reveal no intercalation of C18TAC between pristine kaolinite layers. Additionally, intercalation and subsequent deintercalation of C18TAC proceed in the product using MeO-Kaol. In the Fourier-transform infrared spectra, the intensities of CH2 stretching bands of the product prepared using MeO-Kaol drastically increase compared to those using kaolinite. In addition, CH2 stretching bands of the product using kaolinite are hardly observed without enlarging the spectrum. The product using MeO-Kaol also displays mass loss in the range of 200–300 °C in the thermogravimetric curve and a nitrogen content with 0.15 mass% estimated using the CHN analysis. These results therefore demonstrate an increase in the available reactive edges in the layered crystal material following an expansion of the stacked layers.

Background: Recent clinical data has demonstrated that the use of 3rd party donor granulocyte transfusions was associated with donor CD8+ T cell expansion and remission induction in patients with relapsed acute myeloid leukemia (AML) undergoing cord blood transplant (CBT) (Hiwarker P, 2020; Borrill R, 2023). Further, accessibility to CBT will be improved by strategies that eliminate the need for a second CB unit without increasing risk to patients. Dilanubicel, a cryopreserved non-HLA-matched expanded progenitor cell product has been shown to be safe, to contribute to early myelomonocytic recovery, and is also known to contain antigen presenting cells. Thus, the addition of dilanubicel, as an adjuvant, 3rd party donor transient graft, was explored to obviate the need for a second CB unit and to enhance graft-vs-leukemia. Methods: From March ‘22 to June ‘24, 15 patients with hematologic malignancies were enrolled in a single center Phase II trial to assess safety of dilanubicel to augment conventional single CBT. Patients received conditioning with either FLU 75mg/m2, TBI 13.2 Gy, CY120 mg/kg (n=8) or with FLU 150mg/m2, TBI 4Gy, CY 50 mg/kg and Thiotepa 10 mg/kg (n=7). Per accepted criteria, CB units had to be at least 4/6 HLA-matched to the patient while dilanubicel was used off-the-shelf with no HLA matching required. The CB unit selected for transplant had to have a minimum of 2.5 x 107 TNC/kg and 1.7 x 105 CD34/kg. The single unmanipulated CB unit was infused first followed 4 hours later by infusion of dilanubicel which contained a total of 800x106 CD34+ cells derived from the expansion of CD34+ cells pooled from 6-8 CB units. Donor/host chimerism studies were performed weekly from day 7 to 28, then at days 80, 180 and 1-year post-HCT on peripheral blood flow sorted into myeloid and lymphoid fractions. Graft versus host disease (GVHD) prophylaxis consisted of Cyclosporine and Mycophenolate Mofetil. Results: With a median follow-up of 12 months (range, 6-26), all patients but one received dilanubicel. Eight patients (53%) were male and 6 were (40%) ethnic minorities. The median age was 33 years (range, 10-63) and weight 73 kg (range, 46-115). All patients had acute leukemias (AML=8, ALL=6, MPAL=1) and 4 (26%) had minimal residual disease pre-HCT. Most (n=11) received a 4/6 HLA-matched CB unit. The median infused TNCx107 and CD34x105 per Kg was 3.38 (range, 2.5-5.8) and 2.43 (range, 1.7-4.3), respectively. All patients engrafted neutrophils (ANC) at a median of 18 days (range, 14-27) and platelets (>20 x 10^9/L) at 31 days (range, 26-42). Early (day 7) myelomonocytic (CD33 and CD14) recovery was entirely derived from dilanubicel and no longer present by day 14. Of note, early lymphocyte cell expansion was observed in all patients starting on day +8 or +9 (median 880 with a range of 100 to 1316 per microliter), which was both transient and preceding ANC recovery. Concurrent chimerism analysis showed that the lymphocyte burst derived exclusively from the unmanipulated CB unit. To date, all patients are alive and in remission. No cases of relapse or transplant-related mortality were observed. Infusion of a non-HLA matched pooled donor derived product was not associated with increased GVHD with no cases of grade III-IV GVHD and chronic GVHD observed. Conclusion: The addition of dilanubicel in the setting of single CBT was safe and resulted in excellent clinical outcomes. As an allogeneic and off-the-shelf cell therapy that does not require donor-recipient matching, dilanubicel is readily accessible on demand and easily adopted into standard CBT. With a significant follow-up, the observation of no relapses may be related to the infusion of 3rd party donor APCs for presentation of MHC and activation of donor T cell responses. These promising results warrant further biological investigations and evaluation in a larger study.

The article considers the effect of vacuum extrusion processing on the physical and physicochemical properties of the extruded feed mixture. The influence of the variable factors of the extrusion process on the feed mixture obtained by processing the mixture of rye and fenugreek using thermal vacuum and conventional extrusion was studied. The barrel temperature, screw rotation speed, raw material moisture content and the vacuum level in the chamber at the outlet of the extruder matrix were selected as the studied factors. The analysis of their influence on the thermal vacuum extrusion process was performed using the response surface meth-odology. For the experiment, a modernized EK-40 extruder equipped with a working screw with a diameter of 40 mm, with a screw length to diameter ratio of 4:1 and a temperature regime of 70-100 °C was used. The mixtures for extrusion were prepared from rye (2 parts) and fenugreek (1 part), and the moisture content in them varied within the range from 25 to 35%. The screw speed was varied from 200 to 350 rpm, and the vacuum in the vacuum chamber was in the range of 0-400 mmHg. Using the analysis of vari-ance and response surface methodology, the influence of barrel temperature, screw speed, feedstock moisture content, and vacuum level on the water absorption index, bulk density, specific mechanical energy, total phenol content, and expansion coefficient of the extrudates was analyzed. The following optimal extrusion parameters were obtained during the study: barrel temperature of 77,5 °C, mixture moisture of 32,5%, screw speed of 350 rpm, and vacuum level of 300 mmHg. The results of the study allow us to con-clude that thermal vacuum extrusion has a more noticeable effect on the expansion coefficient, bulk density, and water absorption index of the extrudates compared to conventional extrusion. The resulting extrudate contained a significant amount of anthocyanins C3G, P3G and other useful substances for the animal organism, which are usually destroyed by high temperatures. This is ex-plained by the fact that the reduced pressure in the vacuum chamber promotes the expansion of the extruded mixture at lower tem-peratures than with "classic" hot extrusion, which helps preserve the bioactive compounds of the processed raw materials.

The title compound Fe2Mo3O12 is prepared using a wet chemical method from aqueous solutions of ferrous nitrate and ammonium molybdate. The various process parameters were optimized to get a suitable precursor, which upon calcination at 700°C for 2 h gave a well crystalline product. The product was characterized by powder XRD to have a monoclinic unit cell of Fe2Mo3O12. Its thermal expansion property was investigated by thermomechanical analyzer and high-temperature XRD. The typical monoclinic to orthorhombic phase transition happened at about 500°C, and Fe2Mo3O12 shows great NTE (macro -13.43×10-6°C-1 and micro -21.77×10-6°C-1) property during the temperature range from 500°C to 800°C.

The result of the study shows that the main criteria for weighting the featured products are competitive advantage, stakeholders' acceptance and societal benefits. Background: CD19-directed CAR T-cell therapies have revolutionized the treatment of B-cell lymphomas, though many patients do not respond or relapse. IMPT-314 (rondecabtagene autoleucel) is a CD19/CD20 autologous bispecific CAR T-cell product uniquely manufactured using CD62L selection to yield a product enriched for naive and central memory T-cells. IMPT-314 was designed to enhance cell persistence, reduce CAR T exhaustion, and limit CD19 or CD20 antigen escape. IMPT-314 incorporates the same CAR construct and similar manufacturing process as CART19/20, which has reported durable responses in a single-institution Phase 1 trial (NCT04007029) (Larson et al., 2023; Puliafito et al., 2023). Here we present the first results with a focus on safety, early anti-lymphoma activity, product characteristics and pharmacokinetics in relapsed or refractory (R/R), CAR T-naive LBCL patients in the dose escalation and expansion Phase 1/2 multi-center trial (NCT05826535). Methods: Duali-T-1 is a multi-cohort, open-label, single-arm trial. Patients enrolled in the CAR T-naive cohort required 2 or more prior lines of therapy, measurable disease at baseline (re-confirmed after optional allowed bridging therapy), and no prior treatment with an approved or investigational CAR T-cell product. After standard lymphodepletion (Flu/Cy 30/500 mg/m2 daily for three days) patients received a single dose of IMPT-314 at dose level 1 (DL1) of 100 million CAR+ cells or dose level 2 (DL2) of 300 million CAR+ cells. The primary objectives for Phase 1 are the evaluation of key treatment emergent adverse events (TEAEs) and rate of investigator assessed response, to inform the recommended Phase 2 dose. Results: As of May 31, 2024, 12 patients with R/R, CAR T-naive LBCL received IMPT-314 (DL1 n=10, DL2 n=2). The median age was 65 years (range 21-83), 67% (8/12) had advanced stage disease, and 58% (7/12) had elevated lactate dehydrogenase (LDH) at baseline. Patients received a median of 2.5 prior lines of therapy (range 2-6), and 41.7% (5/12) of patients received bridging therapy. The efficacy evaluable population consisted of 11 patients. One patient received IMPT-314 less than 28 days prior to the data cutoff date, continues on study, and is included only in the safety-evaluable population. Product characteristics post manufacturing confirmed the intended product profile, with a predominantly naive/central memory phenotype (median total CD62L+ cells 90.0%; range 64.1-98.8%). Responses were rapid with 100% (11/11) objective response rate at Day 28, and 73% (8/11) of patients obtaining complete response by three months. With median follow up of 5.6 months (range 1.1-8.8), 8 of 11 (73%) patients were in response at last follow up. Among related TEAEs following treatment with IMPT-314 through day 84 assessment there were no observed infections. Low grade (Grade 1-2) cytokine release syndrome (CRS) was observed in 75% (9/12) of patients, and no high-grade CRS was observed. Immune effector cell-associated neurotoxicity syndrome (ICANS) was reported in two patients including one case of Grade 3 ICANS in a patient treated at DL2, which resolved promptly with corticosteroids. There were no fatal adverse events. In 11 patients evaluable for pharmacokinetic analysis, median time of maximal expansion was 13 days (range 7-29), median Cmax was 96,633 copies/µg gDNA (range 14,365 - 1,724,520) and median AUC(0-28day) was 993,926 days x copies/µg gDNA (range 83,457 - 8,537,457). Transgene was detectable in 91% (10/11) patients at the last assessable timepoint. Enrollment is ongoing, including in a recently added second-line LBCL cohort. Conclusions: Initial results of IMPT-314 suggest a manageable tolerability profile, robust expansion, cellular persistence, and high rates of complete response in R/R, CAR T-naive LBCL. BackgroundPatients with RCC may achieve remission with immune-checkpoint inhibitors (ICI); however, most patients will progress. Adoptive cell therapy with autologous TIL allows for expansion of T-cells from tumor tissue leading to a polyclonal T-cell product with a diverse T-cell receptor repertoire capable of recognizing an array of tumor antigens. TIL therapy with centrally manufactured lifileucel demonstrated a 36% overall response rate in patients with ICI-refractory melanoma.1 We present our preclinical experience of TIL production in RCC.MethodsThis study was approved by the DF/HCC Institutional Review Board. Fresh tumor samples (≥1.5-cm) were harvested from consented patients undergoing resection for RCC. TIL were manufactured using pre-rapid expansion (1/10th scale) and rapid expansion (1/100th scale) protocol for 22 days. Characterization (total viable cells [TVC],% viability, identity, and potency) was performed on the final TIL product. TIL purity, differentiation, memory, activation, and exhaustion status were characterized using multi-color flow cytometry.ResultsBaseline characteristics of 11 recruited patients are shown in table 1. Clear cell was the most common histology (73%). Two patients had previously treated metastatic disease with samples harvested from the lung and adrenal gland; one patient had prior cryoablation with adjacent local recurrence. The remainder were treatment-naïve primary nephrectomy samples. Eight products (73%) showed acceptable TIL product attributes (table 2). Median (range) TVC, viability, and identity (CD45+CD3+%) for the final TIL product were 74×109 (18×109–133×109), 95% (86%–97%), and 98% (94%–99%), respectively. Median (range) percentage of CD4+ cells was 69% (21%–97%) and CD8+ cells was 27% (2%–72%). The non–T-cell population, including B cells, monocytes, and NK cells, was <7%. The final TIL product was functional, and responded to polyclonal bead stimulation; the median (range) IFNg and granzyme B were 8834 (3319–12,957) pg/mL and 34,329 (15,565–65,521) pg/mL, respectively. Acceptable TIL product was generated from both metastatic lesions and primary tumor samples. Introduction. WHO draws attention to the fact that a medical mask does not completely save a person from a viral infection, but it allows one to contain the danger of viral infection during household or medical contact, providing a limited amount of protection. In order to ensure maximum safety for employees of various organizations and the population in the context of a viral epidemic, it is important to analyze the range of medical masks, and, consequently, manage the expansion of the range of medical masks. The relevance is due to the need to orient employees of various organizations and the population in a wide range of PPE. Goal. The research carried out covers models and methods of assortment management and analysis of the medical masks market. The purpose of the study is to study the medical mask market with the intention of expanding the existing range of medical masks. Materials and methods. In this work, the following research methods were used: information-analytical method, analytical-calculation method, comparative analysis method, generalization method, comparison method. The theoretical basis was various literary sources; information presented on the website of various ministries and departments of the Russian Federation; statistical information data. Results and discussion. The key to managing the expansion of the product range, including medical masks, is the formation of an assortment that corresponds to new goals and opportunities, as well as a complex of individual aspects and tasks associated with it. Conclusion. To summarize, we can conclude that today a mask acts not only as a symbol of a pandemic, fear of an invisible disease, but also as an element of clothing, an attribute that helps to stand out in society and attract attention, even in such a difficult period. Also, the introduction of mask restrictions in many countries was perceived aggressively by many people. For some, this is a manifestation of an unconscious fear of infection, for others it is a desire to contrast the personal with the mass, public. A kind of opposition to the system and regime. It is important to say that medical masks during the COVID-19 epidemic and the period after it are becoming increasingly important and becoming a social good.

Sponge cake is a type of pastry product that is widely consumed because of its soft and light texture, and sponge-like shape. The basic ingredient for making sponge cake is wheat flour, but wheat imports are high and not everyone can tolerate gluten. Mocaf can be used as a flour substitute however mocaf-based sponge cake have a low protein levels. Therefore, it is necessary to add other ingredients such as adlay flour to increase the protein content in the sponge cake. The aim of this study was to determine the effect different mocaf and adlay flour ratios have on sponge cake characteristics as well as to determine the ideal ratio to produce sponge cake with the most favorable attributes. The study used completely randomized design (CRD), with the treatment of the ratio of mocaf to adlay flour 100:0, 90:10, 80:20, 70:30, 60:40, and 50:50 repeated 3 times which resulted in 18 experimental units. Data were analyzed using Analysis of Varians (ANOVA) and further analyzed using Duncan Multiple Range Test if the treatment had a significant effect. The results showed that different ratios produced significant effect (P <0.05) on moisture, protein, fat, carbohydrate and crude fiber contents, texture and baking expansion, and product sensory properties, but had no significant effect (P>0.05) on product ash content. The best sponge cake was produced when mocaf and adlay flour were combined in a 60:40 ratio, which had a moisture content of 36.89%, ash of 0.89%, protein of 7.07%, fat of 27.45%, carbohydrate of 27.7%, texture of 1.2 N, baking expansion of 39.81%, with color liked, aroma somewhat liked, taste liked, texture liked, and overall acceptance liked by panelists.

Expansion of soils has been found to produce significant negative economic and environmental impact on various civil engineering infrastructure. This impact is more deleterious in soils containing sulphates, when treated with calcium-based stabilizers such as Lime and/or Portland cement (PC). The reported study investigated the strength and swell characteristics of Kaolinite clay artificially induced with high levels of Gypsum (sulphate) contents after stabilization with CEM I (PC), which is a calcium-based stabilizer. An optimum stabilizer content/Gypsum dosage, aimed at investigating the maximum magnitude of expansion possible using high levels of 10, 15 and 20% Gypsum contents (4.7, 7 and 9.3 wt.% sulphate) stabilized with calcium-based content of 7, 8, 9 and 10 wt.%. This was expected to provide further understanding on the mechanisms behind high sulphate-bearing clay soils, and the impact of sulphate and calcium content on strength and swell characteristics. The research outcomes showed that the introduction of sulphate to a Kaolinite clay soil reduces the compressive strength of the stabilised product by a factor range of 6–47% at 28 days curing age, while the swell behaviour is mainly dependent on both the sulphate content and curing age. Furthermore, the observed result suggests an 8 wt.% binder content to produce maximum magnitude of expansion (swell) with a high Gypsum content of 10% by weight. This finding is of economic importance, as it is expected to serve as a benchmark for further research on the stabilized clay systems, at high sulphate levels using sustainable binder materials.

Abstract Ex vivo expansion of cord blood products (CB) has been proposed as an approach to increase the number of cells available from a single CB unit. We and others have reported the requirement of CD34 selection for optimal expansion of CB products, however, the selection of frozen CB products results in significant losses of CD34+ cells with a median recovery of 43% (range 6 to 203%, N=40) and low purities resulting in decreased expansion. Therefore we explored approaches to expand CB without prior selection and have described the use of co-culture of CB mononuclear cells (MNC) on mesenchymal stem cells (MSC). In the present study we have evaluated the expansion of clinical CB products (provided by Duke University CB Bank CB). MNC were obtained after ficol separation of RBCs and 10% of the CB product was cultured on preformed layers of MSC in T150 flasks containing 50ml of defined media (Sigma Aldrich) plus 100 ng/ml each of rhSCF, rhG-CSF and rhTpo. After 6 days of culture, the non adherent cells were transferred to a Teflon bag and a further 50 ml of media and GFs added to the flask. Again at day 10, non adherent cells were transferred to the Teflon bag and media and growth factors replaced. At day 12 to 13 of incubation the cells were harvested, washed and total nucleated cell (TNC) counts and progenitor assays performed. In three separate experiments we have achieved greater than 20 fold expansion of TNC with a median of 22, and a median expansion of GM-CFC of 37 fold. Morphologic analysis demonstrated the expanded cells contained high levels of mature neutrophils and neutrophil precursors. In vivo studies in NOD/SCID mice also demonstrated that the expanded cells maintained in vivo engraftment potential. Clinical studies are being designed to evaluate the in vivo potential of CB MNC products expanded on MSC.

Abstract Thermal environment responses to terrain and urbanization of Beijing, a metropolis of China, were simulated by the Weather Research and Forecasting model (WRF) with the single-layer Urban Canopy model (UCM). Four numerical experiments were designed: (1) the control experiment (Control exp), (2) no-terrain sensitive experiment (No-Terrain exp), (3) no-urban surface sensitive experiment (No-Urban exp), and (4) real land surface sensitive experiment (LU exp). Results indicate that better near-surface air temperature simulations were obtained after using MODIS land surface type product data MCD12Q1. The foehn effect due to northwestern mountains in Beijing were simulated to intensify the high temperature intensity in the plain area. Urban heat island effect existed in this area day and night, and the heat island intensity in the nighttime was obviously greater than that in the daytime. As a result of urbanization, the nighttime temperature had risen by 1.5∼3.5 °C and the expansion range of nighttime high temperature areas was basically consistent with the urban expansion range.

Abstract The efficiency of the use of protein-carbohydrate compositions in the production of baked meat product is substantiated. The prospects of using vegetable composites of a protein-carbohydrate composition based on flour from sprouted chickpea seeds, jerusalem artichoke and sodium caseinate dietary fibers, a puree–like semi-finished product from ginger rhizome to improve the functional and technological properties of the stuffing system and obtain a meat product with high technological yield and improved quality characteristics are considered. The optimal dosage of the introduction of additives has been established: protein-carbohydrate complex (15%) to the mass of minced meat and mashed ginger rhizome at a dosage of 5.0% to the mass of minced meat. The use of a complex of additives contributed to the expansion of the range of products produced by the meat industry on the basis of the integrated use of raw materials of animal and vegetable origin.

Abstract Introduction: ZUMA-3 and -4 (NCT02614066 and NCT02625480) are ongoing phase 1-2 multicenter trials evaluating KTE-C19, a CD28/CD3ζ anti-CD19 chimeric antigen receptor (CAR) T cell therapy, in adult and pediatric patients respectively with R/R ALL. The manufacturing process developed to support the clinical trials aimed to accommodate variability in the incoming apheresis material, maximize manufacturing success rate, and minimize timing between leukapheresis collection and KTE-C19 administration at bedside. Methods: Once eligibility criteria were satisfied, patients underwent leukapheresis to process 12-15 L of blood (2-5 blood volumes for pediatric patients) to target collection of 10 × 109 mononuclear cells (MNCs). After collection, leukapheresis material was shipped to the central manufacturing site and processed to enrich T cells using an automated and closed system. The T cell enrichment step was implemented to remove cellular impurities such as leukemic blasts and myeloid cells that may negatively impact T cell activation and expansion. Enriched T cells were activated using an antigen presenting cell (APC)-free and bead-free method and cultured in serum-free medium containing 300 IU/mL of interleukin-2. Activated T cells were transduced with a gamma retroviral vector that encodes the CAR gene and further expanded to achieve target dose of 2 × 106 CAR-positive T cells/kg body weight (minimum of 1 × 106). Overall target duration of the manufacturing process was 8 days with an allowed range for harvest set between process days 6 and 10. Final KTE-C19 product was washed, formulated, cryopreserved and tested for identity, potency, and adventitious agents. After acceptance criteria were met, KTE-C19 was shipped to the clinical site using a validated cryoshipper. Results: As of July 2016, 5 of the 6 patients enrolled in the clinical trials received the target dose of 2 × 106 CAR-positive cells/kg body weight. The assigned dose could not be generated for one patient enrolled in the ZUMA-4 trial. In this patient, analysis performed on the leukapheresis material revealed >99% leukemic blasts. For the other 5 patients, the target dose was achieved within 6 - 7 process days notwithstanding a wide range in T cell composition of the incoming leukapheresis material (range, 4.7% to 81%), and wide variation in percent leukemic blasts (range, 11% to 92%). Median frequency of CAR expression in the KTE-C19 final product was consistent across product lots. The manufacturing process led to final products highly enriched for T cells with a median CD3+ cell content of >99%. Expansion of T cells was consistent among the 5 product lots with a mean 7-fold expansion from transduction to harvest. Median time from leukapheresis collection to final product release was 14 days. Conclusions: Reproducible manufacturing of high-quality, clinical-grade autologous CAR T cell products may be challenged by the inherent variability of starting material in patients with high content of leukemic blasts in peripheral blood. The bead- and serum-free manufacturing process developed to support the ZUMA-3 and -4 clinical trials was able to generate products within 6 -7 days with a low failure rate (including from patients with high percentages of leukemic blasts), and most importantly, a short duration from leukapheresis collection to shipment of KTE-C19 final product back to the clinical center for patient administration. Abstract Background: Myeloablative cord blood transplant (CBT) recipients experience delayed hematopoietic recovery and an increased risk of transplant related mortality (TRM). With the primary goal of generating increased numbers of hematopoietic stem and progenitor cells (HSPC) to supplement a conventional cord blood (CB) graft and thereby enhance engraftment and reduce early TRM, we have developed methods to ex vivo expand CB-derived HSPC in the presence of Notch ligand for 14 days. The product is then harvested and cryopreserved for future clinical use. We have previously reported that infusion of non-HLA matched, off-the-shelf ex vivo expanded CB progenitor cells in patients undergoing myeloablative CBT is safe and associated with faster neutrophil (ANC) recovery. We now present updated outcomes data with a median follow-up of 3.2 years (range 1.9-3.7). Methods: Between September 2010 and August 2012, 15 patients with hematologic malignancies were enrolled in this single center Phase I trial to assess the safety of infusing a non-HLA matched expanded CB HSPC product to augment conventional CBT. Conditioning consisted of Fludarabine (75mg/m2), Cytoxan (120mg/kg) and TBI (13.2 Gy). On the day of transplant, 1 or 2 unmanipulated CB units were infused first followed 4 hours later by infusion of the expanded CB product. Donor(s)/host chimerism studies were performed weekly from day 7 to 28, then at days 80, 180 and 1 year on peripheral blood flow sorted into myeloid and lymphoid fractions. Graft versus host disease (GVHD) prophylaxis consisted of cyclosporine and MMF beginning on day -3. ANC and platelet engraftment were calculated using cumulative incidence rates to accommodate competing risks. Disease-free survival (DFS) was analyzed using Kaplan-Meier estimates. Results: The median age was 21 years (range, 5-45) and weight 59 kg (range, 23-89). Other patient characteristics and median infused cell doses are shown in Table 1. Four patients (26%) received only 1 unmanipulated CB unit along with the expanded graft. All patients engrafted at a median of 19 days (range, 9-31 days), significantly faster than the 25 days (range, 14-45) observed in 41 recipients of two unmanipulated units otherwise treated identically (p=0.01) (Figure 1A).Similar to our initial expansion trial using partially HLA-matched expanded CB cells, early (day 7) myelomonocytic (CD33 and CD14) recovery was almost entirely (98–100%) due to cells arising from the expansion product. Cells derived from the expansion product were no longer detected at day 14 in all but 2 patients. The median time to platelet recovery (>20 x 10^9/L) was 35 days (range 21–45).The cumulative incidence of platelet recovery by day 100 was significantly higher than that observed in our concurrent control group 92% (CI 95%: 59-99) vs. 69% (95%: 51-81), respectively (p=0.005) (Figure 1B). The probability of 3-year DFS was 86% (95% CI, 57-97). No TRM was observed throughout the study period (Figure 1C), although 2 patients relapsed at days 53 and 219 posttransplant and subsequently died after further therapy. All patients presented with grade II acute GVHD at a median time of 32 days (14-86), with no grade III-IV aGVHD observed (Figure 1D). The skin was the most commonly affected organ (n=12). Eight (53%) patients were treated for pre-engraftment syndrome at a median time of 6 days (range 4-9) and five (33%) patients had GVHD after day 100. At 2 years only 2 patients are still on immunosuppressive therapy for mild chronic GVHD. Conclusions: The results of this Phase I safety study demonstrated that infusion of an off-the-shelf non-HLA matched expanded CB HSPC product to augment a CB graft in the myeloablative setting was safe and led to more rapid neutrophil and platelet recovery compared to our concurrent control group. Importantly, no TRM was observed among the 15 patients on this study, with only 2 deaths due to relapsed disease, leading to an excellent OS of 86% at a median follow-up of 3.2 years. Quite interesting is the observation of no grade 3-4 acute GVHD, even with infusion of a non-HLA matched expanded cell product that may induce increased alloreactivity of the unmanipulated CB unit(s). However, in this preliminary pilot study, it appeared that infusion of the non HLA-matched product was associated with less severe acute GVHD. There are suggested hypothesis and models, by which there are offered measures for improving the systems of development institutions. The basis for accepting an investment solution inside any developmental institution should be a confirmed commitment of the end consumer of the technology, product, innovation enterprise. It is necessary to accept the system of regulatory measures to stimulate the coordination between developmental institutions, provision of a transparent support for projects within the whole chain of innovative development. The solution for issues with the deficiency of investment resources at the stage of creation and expansion of the prototype (Stage B+), specialised expertise, lays in new field foundations, created with the state capital.
