Full textAtherosclerosis is an inflammatory disease of thé arterial wall where both innate and adaptative Th1-driven immuno-inflammatory responses contribute to disease development. Th2-related immunological responses hâve been shown to be either protective or pathogenic. Using different mouse models, we have shown that a novel subset of T cells, called regulatory T cells, prevents atherosclerosis initiation and reduces plaque progression. In vivo expansion of regulatory T cell could be a novel therapeutic tool to combat atherosclerosis. We also develop a therapeutic approach in a mouse model of atherosclerosis through T cell modulation. We have shown that repetitive administration of a nucleoprotein (NP) extracted from Measles virus to mice susceptible to atherosclerosis induces a regulatory immune response and inhibits both lesion development and progression. NP administration drives CD4+ T cells and CD11c+ dendritic cells toward a Tr-1 like anti-inflammatory profile. At least, we have shown for the first time, using LDLr/MFGE8"7" chimeric mice, that alteration of phagocytic clearance of apoptotic cells induces a decrease of regulatory T cell suppressive functions. The consequences are expansion of Th-1 pathogenic T cells and atherosclerosis acceleration

Full textComplicated abdominal aortic aneurysm (AAA) is a major cause of mortality in elderly men. Ang II-dependent TGF-P (Transforming Growth Factor-beta) activity promotes aortic aneurysm progression in experimental Marfan syndrome. However, the role of TGF-p in experimental models of AAA has not been comprehensively assessed. Here, we show that systemic neutralization of TGF-P activity breaks the resistance of normocholesterolemic C57BL/6 mice to Ang II-induced AAA formation and markedly increases their susceptibiliry to the disease. These aneurysms displayed a large spectrum of complications on echography, including fissuration, double channel formation, and rupture, leading to death from aneurysm complications. The disease was refractory to inhibition of IFN-y, IL-4, IL-6, or TNF-a signaling. Genetic deletion of T and B cells or inhibition of the CX3CR1 pathway resulted in partial protection. Interestingly, neutralization of TGF-p activity enhanced monocyte invasiveness, and monocyte depletion markedly inhibited aneurysm progression and complications. Finally, TGF-p neutralization increased MMP-12 activity, and MMP-12 deficiency prevented aneurysm rupture. These results clearly identify a critical role for TGF-P in the taming of the innate immune response and the preservation of vessel integrity in C57BL/6 mice, which contrasts with its reported pathogenic role in Marfan syndrome

Full textWe bave previously described that polyclonal anti-lymphocytic globulins (GALP) may contribute to prolonged CD4 Tcell lymphopenia in some renal transplant recipients, associated with some biological abnormalities, such as a chronic inflammatory syndrome. Our work focuses on the factors involved in the induction of tbese abnormalities and susceptible to increase the incidence of infections, cardiovascular diseases and deatb, comparable to the incidence observed in the elderly. We showed that GALP are implicated in the decrease in thymie output and the expansion ofT cells at an advanced stage of differentiation. Both are hallmarks of premature immune senescence. Moreover, the dysfunction of the gut epithelial barrier is responsible for gut bacterial translocation (GBT) and the activation of chronic inflammation observed in chronic kidney disease as well as in renal transplant recipients. The composition of the intestinal microbiota may play a role in the initiation, maintenance and severity of GBT and systernic inflammation. We reported the existence of a dysbiosis a.fier transplantation. The mechanisms involved remain to be elucidated, yet, this work contributes to the understanding of the potential factors involved. in the progression of immune senescence and the persistence of chronic inflammation after kidney transplantation paving the way to new fields of therapeutic research in transplantation
