Devant le déclenchement des hostilités entre le FPR et les FAR en 1990, l'organisation des Nations Unies à la demande des deux parties, choisit de s'impliquer directement. Mais l'intervention de l'ONU et l'envoi des casques bleus qui survient après la signature des Accords d'Arusha de 1993 afin d'accompagner lesdits accords, n'arrête cependant pas la montée des violences et de l'extrémisme Hutu. Les différentes résolutions du Conseil de sécurité, loin d'accorder plus de pouvoir aux casques bleus, créent au contraire une situation d'immobilisme général, favorisant au printemps 1994 la reprise des combats et le génocide des Tutsi et des Hutu modérés. Le drame humanitaire que connaît le Rwanda pendant et après les affrontements et les massacres, nous interpelle sur le véritable rôle des Nations Unies dans la gestion de ce conflit, et plus généralement dans le monde de l'après guerre froide<br>Before the outbreak of hostilities between the RPF and the FAR in 1990, the United Nations became directly involved after both parties asked it to arbitrate the conflict. But the intervention of the UN and the deployment of peacekeepers that came after the agreement of Arusha in 1993 in order to accompany the agreements did not stop violence and the rise of Hutu extremism yet. The various resolutions of the UN Security Council, far from granting more power to peacekeepers, instead created a situation of stagnation favoring the resumption of fights in 1994 and the genocide of Tutsi and moderate Hutu. The humanitarian tragedy facing Rwanda during and after the clashes and massacres appeals us on the proper role of the United Nations in the management of this conflict in particular, and in the post-cold war world in general

Magíster en Gestión y Dirección de Empresas<br>El presente trabajo tiene como objetivo el desarrollar una estrategia de crecimiento para una consultora de servicios de ingeniería en gestión de mantenimiento, FPR Ingenieros Consultores, cuyo actual nicho es la industria minera. El objetivo principal es que dicha estrategia le permita obtener rendimientos sobre el promedio de la industria en forma sustentable, teniendo claridad sobre las acciones y medidas pertinentes que harán cumplir los objetivos planeados. Chile es un país rico en recursos mineros, lo que lo hace un foco atractivo para capitales extranjeros. Lo anterior trae consigo una serie de desarrollos económicos adyacentes a esta industria, lo cual ha generado polos de negocios regionales centralizados, atrayendo inversión extranjera, lo que hace al país cada vez más competitivo. Según estadísticas de Sonami, emitidas en la publicación Perspectivas de la minería latinoamericana y chilena nuestro país proyecta una inversión de US$ 4.000 millones para el período 2010 2017 en servicios de ingeniería y consultoría para la industria de la minería. Para cumplir con el objetivo, se utilizó la metodología de un proceso formal de administración estratégica. La metodología considera el análisis del entorno e industria, estructura interna de la empresa, formulación y definición de misión y visión, análisis y evaluación de estrategias, implementación y control de proyectos, todo bajo un análisis económico de valor actual neto (VAN), tasa interna de retorno (TIR) y periodo de recuperación de la inversión (PRI). Los resultados esperados se encuentran descritos por las variables analizadas dentro de un plan de crecimiento, destacando el aumento de la facturación, del número de clientes y su fidelización, y el posicionamiento en lugares estratégicos en relación a la industria minera. Luego del análisis, creación e implementación de las soluciones propuestas, se generó un escenario con un crecimiento sostenido en la facturación de un 40% anual, y un incremento de 30% en el número de clientes para el mismo período. Al realizar la evaluación se obtiene un VAN de $ 690 millones, una TIR de 154% y un PRI de 0.13 años, resultados muy auspiciosos para la decisión de implementación. Se puede concluir que existe un enorme potencial en el desarrollo de negocios, esencialmente por el bajo ítem de inversiones versus el alto grado de retornos, cuyo principal eje radica en la correcta estrategia comercial utilizada para penetrar en los mercados de la industria minera, manejo y conocimiento de la industria de consultoría, y expertise en los procesos.

Les cellules phagocytaires constituent la première ligne de défense contre les pathogènes. Leur migration dirigée vers le site infectieux et leurs fonctions microbicides sont l'aboutissement de voies de signalisation intracellulaires sollicitées par la stimulation de récepteurs couplés aux protéines G, les récepteurs de chimioattractants. Après fixation du ligand et transmission du signal par la protéine G, les récepteurs sont phosphorylés et interagissent avec les b-arrestines, protéines d'échafaudage concourrant à l'internalisation des récepteurs. Plusieurs exemples récents suggèrent que les b-arrestines pourraient également participer à la signalisation. Le travail présenté dans ce mémoire concerne les récepteurs de la famille FPR (Formyl Peptide Receptor) et plus spécialement le récepteur FPRL1 (FPR-like 1), pour lesquels de nouveaux agonistes dérivant de protéines bactériennes ou mitochondriales humaines ont été identifiés. La phosphorylation du récepteur FPRL1 a été caractérisée. Il a été montré que les -arrestines interagissent avec celui-ci et qu'elles sont indispensables à son internalisation. Diverses approches ont conclu que l'activation rapide des MAP kinases ERK1/2, enclenchée par la stimulation du récepteur FPRL1, est majoritairement dépendante de la protéine G héterotrimérique et qu'il n'y pas de signalisation transmise par les b-arrestines. Enfin, une analyse protéomique des complexes multi-protéiques bâtis autour du couple FPRL1/b-arrestine a été menée par la méthode TAP (Tandem Affinity Purification). Their directed migration to infection sites and their microbicidal functions result from intracellular signaling cascades elicited by specific G protein-coupled receptors, the chemoattractant receptors. After ligand binding and G protein-mediated signaling events, receptors are phosphorylated and interact with the b-arrestins, which are scaffolding proteins participating to receptor internalization. Recent examples suggest that b-arrestins could also participate to signaling events. The work presented here is focused on the FPR family (Formyl Peptide Receptor) and specially the FPRL1 receptor (FPR-like 1), for which new agonists from bacterial origin or derived from human mitochondrial proteins have been identified. The phosphorylation of the FPRL1 receptor has been characterised and it has been shown that b-arrestins interact with this receptor and are essential to its internalization. Various approaches have concluded that the FPRL1-elicited rapid and transient activation of the MAP kinases ERK1/2 is mainly dependant on G protein and that there is no signaling events mediated by the b-arrestins. Finally, a proteomic study of the multiprotein complexes surrounded the couple FPRL1/b-arrestin has been conducted by the TAP method (Tandem Affinity Purification). The adaptor complex AP3, an homolog of AP2, has been identified as partner of b-arrestins after FPRL1 stimulation

Abstract:
Les leucocytes peuvent etre actives par des agents chimioattractants tels que des peptides bacteriens (fmlp) ou la fraction c5a du complement. Ces agents chimioattractants induisent la migration des leucocytes, leur degranulation et l'activation du complexe nadph oxydase qui produit les ions superoxyde impliques dans la destruction des pathogenes. Ces reponses cellulaires sont declenchees par la fixation de ces agents sur des recepteurs specifiques qui appartiennent a la superfamille des recepteurs couples aux proteines g heterotrimeriques. Malgre la presence continue d'agonistes, les reponses cellulaires sont transitoires et les cellules deviennent refractaires a une autre stimulation (desensibilisation homologue). Selon le concept actuel, le recepteur est phosphoryle, ce qui permet son interaction avec la proteine adaptatrice -arrestine. Cette interaction inhibe le couplage recepteur/proteine g et induit l'internalisation du recepteur par les puits recouverts de clathrine. Apres addition de c5a, le recepteur du c5a (rc5a) est phosphoryle sur six serines de la queue c-terminale intracellulaire. En utilisant des recepteurs dont certaines serines ont ete mutees, nous avons montre que : (1) cette phosphorylation suit un processus hierarchique, debutant par la phosphorylation des serines s334 et s332. (2) la phosphorylation de la serine s338 est necessaire au decouplage recepteur/proteine g. (3) le decouplage et l'internalisation de rc5a sont deux processus independants. (4) dans les cellules promyelocytaires hl-60, rc5a peut etre internalise par un mecanisme independant de la phosphorylation. Rc5a peut egalement etre internalise par la voie classique dependante de la phosphorylation. Ces deux mecanismes sont sensibles au dominant negatif de la dynamine, une gtpase qui intervient au niveau de la membrane dans la fermeture des vesicules d'endocytose, suggerant l'implication des puits recouverts de clathrine. D'autre part, nous avons identifie le peptide synthetique wkymvm comme un agoniste des recepteurs fpr, fprl1 et fprl2 ; wkymvm est specifique de fprl1 et fprl2. Un peptide antibacterien produit par helicobacter pylori (hp(2-20)) est egalement un agoniste de fprl1 et fprl2.

Abstract:
Les maladies cardiovasculaires (MCV) sont des affections immuno-inflammatoires chroniques qui sont liées à de nombreux facteurs de risque. Les défensines et les cathélicidines sont des peptides antimicrobiens considérés comme des effecteurs clés de l'immunité innée, mais aussi des réponses inflammatoires. Il a été postulé que ces peptides sont impliqués dans le développement de l'athérosclérose. Afin de vérifier cette hypothèse, nous avons étudié l'expression des gènes de la cathélicidine LL-37 et des défensines-a DEFA1-3 dans les cellules mononuclées du sang périphérique (PBMCs) dans la cohorte Stanislas. Nous avons démontré que l'expression des ARNm de ces gènes est significativement corrélée avec les indicateurs de risque des MCV, à savoir l'obésité, la pression artérielle, les taux circulants des triglycérides et du glucose, l'hypo-HDL-C et les leucocytes. De plus, des taux élevés des ARNm de ces gènes sont associés au syndrome métabolique. Nous avons aussi démontré que l'expression des gènes des DEFA1-3 et de LL-37 est corrélée avec celle du récepteur chimio-attractant FPR et que le génotype TT du polymorphisme FPR1 c.32C&gt;T est associé à une diminution des taux circulants de la E-sélectine, suggérant que ces peptides pourraient agir via FPR et que le polymorphisme c.32C&gt;T affecte la fonction endothéliale. Enfin, nos études in vitro ont montré que l'expression des gènes des DEFA1-3 et de CRAMP, cathélicidine de la souris, est modulée par le glucose et l'insuline, respectivement dans les neutrophiles HL-60 et les adipocytes 3T3-L1. L'ensemble de ces études suggère que les DEFA1-3 et LL-37 pourraient constituer un lien entre le système immunitaire inné et le risque des MCV et du syndrome métabolique<br>Multiple risk factors for atherosclerosis and cardiovascular diseases (CVD) act in a synergistic way through inflammatory pathways. Most of CVD risk factors stimulate the release of inflammatory mediators. Defensins and cathelicidins are antimicrobial peptides (AMPs) produced mainly by inflammatory cells. Beside their role in host defense, AMPs are also considered as effectors of inflammatory responses. They have been suggested to play a role in atherosclerosis. To verify this hypothesis, we studied a-defensins DEFA1-3 and cathelicidin LL-37 in a sample of the STANISLAS cohort. We demonstrated that mRNA levels of LL-37 and DEFA1-3 genes in peripheral blood mononuclear cells (PBMCs) of studied subjects are significantly correlated with indicators of obesity, blood pressure, circulating triglycerides and fasting glucose levels, hypo-HDL-C, and leukocytes counts suggesting a role of these genes in CVD. Further analysis revealed that high expression of these genes might be associated with metabolic syndrome. We also showed that expression of LL-37 and DEFA1-3 genes was positively associated with that of FPR receptor gene and that the TT genotype of FPR1 c.32C&gt;T/I11T polymorphism was significantly associated with decreased levels of soluble E-selectin suggesting that these peptides may act through this receptor and such a polymorphism may has an impact on endothelial cells function. In an in vitro model, we found that glucose and insulin modulate the expression of DEFA1-3 and CRAMP cathelicidin genes in human HL-60 neutrophils and mouse 3T3-L1 adipocytes cell lines, respectively. Together, our studies demonstrated that DEFA1-3 and LL-37 could be a potential link between innate immunity and CVD and metabolic syndrome

Abstract:
Les Cyclosporines (Cs) représentent une classe de peptides cycliques neutres composés de 11 acides aminés aliphatiques, produits par différents champignons imparfaits et quelques Ascomycètes. Les activités biologiques des Cs sont nombreuses, bien que seule l'activité immunosuppressive soit généralement connue. En séquestrant dans le cytosol la calcineurine dans un complexe moléculaire avec la cyclophiline, certaines Cs exercent une puissante activité immunosuppressive en supprimant spécifiquement la transcription du gène de l'interleukine-2 (IL-2), cytokine d'activation et de croissance des lymphocytes T. Indépendamment de cette propriété mettant en jeu l'interaction cyclophiline-Cs, les Cs peuvent exercer une ou plusieurs activités biologiques résultant de leur interaction avec des glycoprotéines membranaires. Ainsi, plusieurs Cs sont connues pour leurs propriétés inhibitrices sur la P-glycoprotéine MDR1 humaine (Pgp), une flippase responsable de la résistance de nombreuses tumeurs à la chimiothérapie anticancéreuse, mais exprimée également par certaines cellules immunocompétentes, ainsi que par des cellules épithéliales normales intervenant dans la protection de l'organisme contre des produits toxiques endogènes et exogènes et donc aussi dans le contrôle de la biodisponibilité de médicaments in vivo. Quelques Cs peuvent également inhiber le récepteur humain pour peptides N-formylés d'origine bactérienne ou mitochondriale (FPR), un récepteur chimiotactique à 7 pièces transmembranaires couplé à une protéine G (7TM-GPCR), récepteur dont l'absence est connue pour affecter l'immunité anti-bactérienne, mais dont les rôles physiologiques, pas encore clairement élucidés, semblent être plus nombreux. En plus de ces activités biologiques, les Cs présentent l'avantage galénique, qu'après administration orale, leur dégradation dans le tractus gastro-intestinal est retardée, et qu'une fraction substantielle passe de manière non-transformée dans la circulation générale. En effet, différentes raisons pour cette 'résistance' à la dégradation existent : leur taille relativement importante (leur poids moléculaire (PM) est largement supérieur à 500) implique une élimination par voie biliaire plutôt que par voie rénale (qui concerne surtout les composés avec un PM<300). Si une structure cyclique présente déjà l'avantage sur une structure linéaire d'être moins susceptible aux enzymes de dégradation, la métabolisation d'un peptide cyclique N-méthylé semble se révéler particulièrement difficile (la plupart des Cs présentent 5 ou 6 acides aminés sur 11 N-méthylés). Ces molécules sont ainsi très lentement transformées en métabolites qui maintiennent une structure cyclique. Outre cette stabilité, c'est le caractère hydrophobe de ces molécules qui leur permet une interaction facile avec la membrane plasmique et/ou le passage vers le milieu cytoplasmique. Mes travaux de thèse consistaient en l'étude et la comparaison d'un nombre important de Cs naturelles, synthétiques et semi-synthétiques, ainsi que d'une famille de cyclopeptolides non-apparentés aux Cs, les Auréobasidines, pour leur capacité de moduler (voire inhiber) le fonctionnement de la Pgp pour restaurer ainsi la sensibilité de cellules tumorales résistantes à la chimiothérapie. En parallèle, nous avons étudié l'action de ces molécules sur le FPR. Nos résultats et sur l'inhibition de la Pgp et sur celle du FPR ont été intégrés ensuite sous forme de relations-structure activité, afin d'apporter des précisions sur les modifications chimiques et, par conséquent, sur les propriétés structurales requises pour une interférence optimale avec le fonctionnement de ces deux récepteurs appartenant à des superfamilles structuralement et fonctionnellement très différentes. Enfin, nous avons dans un cadre pharmacologique d'étude générale des processus inflammatoires mettant en jeu des interactions ligand-récepteur spécifiques, étendu nos recherches sur les 7TM-GPCRs à un récepteur homologue du FPR, le FPRL-1, pour lequel la lipoxine A4 et la sérum amyloi͏̈de A ont été décrites comme ligands physiologiques. Dans ce contexte, nous avons comparé un certain nombre d'agonistes présumés ou non de FPR et FPRL-1 et étudié l'inhibition par quelques Cs naturelles de ces deux récepteurs homologues en vue d'apporter des informations supplémentaires sur leur spécificité agoniste/antagoniste et sur leurs rôles physiologiques respectifs.

O câncer de endométrio é um tumor maligno frequente e sua incidência vem aumentando nas últimas décadas. Enquanto, o tumor uterino benigno mais comum, o leiomioma, acomete cerca de 40% das mulheres na idade reprodutiva, sendo relacionado à menorragia, dismenorreia e infertilidade. Investigações indicam que o microambiente tumoral é crucial para o avanço do câncer, sendo caracterizado, principalmente, pela composição alterada da matriz extracelular, alta densidade de microvasos e abundância de células inflamatórias, como mastócitos (MCs). MCs desgranulados liberam fatores quimiotáticos e proteases, como triptase e quimase, para o meio extracelular, contribuindo na degradação da matriz extracelular, promoção da angiogênese, propiciando ambiente favorável para invasão tumoral e remodelação tecidual por meio de proteólises seletivas na matriz e ativação de metaloproteinases. Outro aspecto importante no crescimento tumoral é a proteína anti-inflamatória Anexina A1 (ANXA1), relacionada à regulação dos processos de crescimento e migração/invasão celular, sendo seus efeitos mediados por receptores para peptídeos formilados (FPRs), especialmente FPR1 e FPR2. Diante da importância dos MCs e da ANXA1/FPR no desenvolvimento tumoral, o objetivo desta investigação foi analisar a heterogeneidade dos MCs e a expressão das proteínas ANXA1, FPR1 e FPR2 em biópsias humanas das variáveis clínico-patológicas de útero normal: hiperplasia endometrial simples (HES), adenomiose, leiomiomas e adenocarcinoma (ADC) endometrial de graus I e II. Os MCs foram quantificados de acordo com seu estado de ativação e expressão das proteases triptase e quimase. A expressão da ANXA1 e seus receptores FPR1 e FPR2 nos MCs e tecidos uterinos foram analisadas nas diferentes biópsias estudadas. Nossos resultados mostraram MCs intactos e desgranulados, no endométrio e miométrio normais e aumentados na HES, margens tumorais nos leiomiomas, adenomiose e ADC endometrial de graus I e II, e diminuídos significantemente na região tumoral do leiomioma. Com relação à heterogeneidade, os MCs triptase-positivos foram observados em maior quantidade. As expressões endógenas da ANXA1 e do FPR1 foram observadas nos tecidos uterinos e MCs, com ausência para o FPR2. Endometrial cancer is a frequent malignant tumor and its incidence has been increasing in the last decades. Besides, the most common benign uterine tumor, leiomyoma, affects about 40% of women at reproductive age, being related to menorrhagia, dysmenorrhea and infertility. Investigations indicate that the tumor microenvironment is crucial for the advancement of cancer, being characterized mainly by the altered composition of the extracellular matrix, high microvessel density and abundance of inflammatory cells, such as mast cells (MCs). Degranulated MCs release chemotactic and protease factors, such as tryptase and chymase, to the extracellular medium, contributing to the degradation of the extracellular matrix, promoting angiogenesis, providing a favorable environment for tumor invasion and tissue remodeling through selective proteolysis in the matrix and activation of metalloproteinases. Another important aspect of tumor growth is the anti-inflammatory protein Annexin A1 (ANXA1), related to the regulation of growth and migration / invasion processes, and its effects mediated by receptors for formylated peptides (FPRs), especially FPR1 and FPR2. The objective of this investigation was to analyze the heterogeneity of the MCs and the expression of the ANXA1, FPR1 and FPR2 proteins in human biopsies of clinical-pathological variables of normal uterus: simple endometrial hyperplasia (HES), adenomyosis, leiomyomas and endometrial adenocarcinoma (ADC) of grades I and II. MCs were quantified according to their state of activation and expression of tryptase and chymase proteases. Expression of ANXA1 and its FPR1 and FPR2 receptors in the MCs and uterine tissues were analyzed in the different biopsies studied. Our results showed intact and degranulated MCs in the normal endometrium and myometrium and increased MCs in HES, tumor margins in leiomyomas, adenomyosis and endometrial ADC of grades I and II, but significantly decreased in the leiomyoma tumor region. In relation to the heterogeneity, it was observed that the tryptase-positive MCs were observed in greater quantity. Endogenous expressions of ANXA1 and FPR1 were observed in uterine tissues and MCs, but absent for FPR2. Modulations of MCs and ANXA1 protein expression and the specificity of FPR1 receptor immunolabeling in the clinical-pathological variables of the investigated uterine lesions indicate the involvement of these cells and the interaction ANXA1/FPR1 in the development of inflammation and uterine neoplasia.

This research is geared towards investigating possible means of minimizing the problem of food and energy shortages through the use of the local farmers by introducing basic but essential skills in introductory mechanization and basic agronomic methods and other techniques in their farm sites. This is to produce rice-staple food and bio-fuel (Pure Vegetable Oil; PVO) from jatropha seeds that can be used locally by the people themselves. The research was carried out on the cultivation of rice which is the staple food of Sierra Leone through the use of the traditional method compared with the modern method of the use of machinery such as the tractor to do minimal tillage in the farming site of the farmers. The results of the research showed that the use of introductory mechanization together with some basic modern techniques in rice cultivation resulted to differences in yields with 2.20 t.ha-1 for traditional method and 2.34 tha-1 for modern method at 14% moisture content. However, this might seem to be negligible in terms of yield but there was also some time saved (40 h.ha-1) through the introduction of mechanization. It was also observed that with further use of minimal level of mechanization in rice planting there may be an additional gain of 192 h.ha-1; which is a good time for the farmers to rest, improve their health status and carry out other economic activities. The production of jatropha was seen to be accepted by the farmers by intercropping it with an edible crop. The use of ground nut as intercrop in the cultivation of jatropha was observed to have both agronomic and economic benefits. On the agronomic scale, the intercropped ground nut was observed to improve the soil fertility through the addition of nutrients such as nitrogen and organic matter into the soil. It also helped to suppress weeds, hence reducing the labour cost of weeding and allowing better growth of the jatropha. Economically, the ground nut was able to provide financial benefit of 3,594,500 Le.ha-1. The use of a small electric generator of 6 kW machine in the rural community for rural electrification was observed to be feasible since the farmers can production adequate PVO through the cultivation of 17.1 ha of jatropha which could produce 25500 kgyr-1 of PVO to support the above machine in the community without interrupting their normal food production chain. The research has used the On-Farm Research (OFR) approach where the farmers were also allowed to participate fully in all the activities. The involvement of the farmers in this research (Farmers Participatory Research (FPR) is observed to have some positive results at the end of the exercise both by the farmers and the researcher as learning and teaching processes. Some basic farm techniques and skills used during the research were a positive strategy in which the farm sites were seen as suitable field laboratories for not only the researcher but the farmers as well.

Le propos de cette thèse, est de discuter les rapports entre les dynamiques de transformation sociale de communautés d’expérience et de subjectivation d’acteurs émergeant dans différentes circonstances et qui tentent d’infléchir leur contexte d’action. Comment l’action de l’acteur engagé dans un parcours de subjectivation à la rencontre de l’autre fait-elle retour sur son monde ou milieu, et aussi sur celui de l’autre ? Tout ici est question de frontières et de limites, d’histoire et de mémoire en acte. C’est à l’exploration du travail des gens du bord, passeurs de mémoire vives, de frontières matérielles et symboliques que se consacre cette thèse à partir des terrains et de situations variés.Trois types d’expérience à fort enjeux socio-historiques sont mis en intrigue : celle des générations héritières de l’immigration maghrébine des quartiers populaires; celle des militants anti-guerre dans l’ex-Yougoslavie ; celle des migrants rroms en France et celle d’un mouvement activiste rrom roumain. <p>In this master thesis, the possibilities of customizing a low-cost microprocessor with the purpose of replacing an existing microprocessor solution are investigated. A brief survey of suitable processors is carried out wherein a replacement is chosen. The replacement processor is then analyzed and extended with accelerators in order to match set requirements.</p><p>The result is a port of the processor Lattice Mico32 for the FPGA curcuit Xilinx Virtex-5 which replaces an earlier solution using Xilinx MicroBlaze. To reach the set requirements, accelerators for floating point arithmetics and FIR filtering have been developed. The toolchain for the processor has been modified to support the addition of accelerated floating point arithmetics.</p><p>A final evaluation of the presented solution shows that it fulfills the set requirements and constitutes a functional replacement for the previous solution.</p>

L’analyse des relations entre le gouvernement intérimaire rwandais et la communauté internationale durant le génocide de 1994 met en évidence la primauté du facteur politique voire géopolitique sur la prise en compte de considérations strictement juridiques et humanitaires de la part de l’ONU et des États tiers les plus impliqués dans cette crise (Belgique, France, États-Unis). La marginalisation progressive du gouvernement intérimaire sur la scène internationale, au moment où il sollicite une aide extérieure pour mettre fin aux massacres, ne s’est pas accompagnée d’un renforcement des effectifs et du mandat de la Mission des Nations Unies pour l’Assistance au Rwanda (MINUAR), qui constituait pourtant la principale demande des autorités rwandaises au Conseil de sécurité. Ce désengagement de la communauté internationale s’est effectué au détriment de la sécurité et de la protection des populations civiles menacées. Dans le même temps, la criminalisation de ce gouvernement a contribué à faire entériner l’acceptation d’une issue militaire au conflit souhaitée par la rébellion du Front Patriotique Rwandais (FPR), au mépris des accords de paix et de partage du pouvoir d’Arusha signés en 1993. La présomption de culpabilité à l’égard du camp gouvernemental a par la suite fortement impacté la justice internationale, le Tribunal Pénal International pour le Rwanda (TPIR) ayant échoué dans sa mission consistant à juger tous les auteurs de crimes commis en 1994 et à favoriser la réconciliation nationale, du fait de son manque d’impartialité et d’indépendance tant au niveau des poursuites et de l’instruction que du rendu de ses jugements et de leur pleine application<br>An analysis of relations between the interim government of Rwanda and the international community during the genocide of 1994 demonstrates how political, even geopolitical, factors were given priority over strictly judicial and humanitarian considerations by the UN and the third-party states most implicated in this crisis (Belgium, France, the USA). The progressive marginalisation of the interim government on the international scene, at the time when it was soliciting external help to put a stop to the massacres, did not see a reinforcement of staff or of the mandate of the United Nations Assistance Mission for Rwanda (UNAMIR), which nonetheless constituted the principal demand of the Rwandan authorities to the Security Council.This disengagement on the part of the international community happened to the detriment of the security and protection of the civil population under threat. At the same time the criminalisation of this government contributed to the endorsement of a military outcome to the conflict ; the outcome desired by the rebellion of the Rwandan Patriotic Front (RPF), contravening the Arusha Accords signed in 1993. The presumption of guilt attached to the government camp subsequently had a strong impact on international justice, as the International Criminal Tribunal for Rwanda (ICTR) had failed in its mission of judging all the perpetrators of crimes committed in 1994, and of favorising national reconciliation, due to its lack of impartiality and independence as much at the level of investigation and prosecution as at the level of the delivery of the judgments and their enactment

Esta bactéria pode levar a uma inflamação persistente através da produção de citocinas pró-inflamatórias e de espécies reativas de oxigênio e nitrogênio, estimulando a proliferação celular bem como outros processos envolvidos na carcinogênese. Ainda envolvidos nestes processos tem sido observada a participação de microRNAs, que exercem papel importante na regulação pós-transcricional, influenciando processos fisiológicos normais da célula bem como aqueles ligados às doenças, como por exemplo o câncer gástrico. Alguns miRNAs podem atuar como oncogenes, genes supressores de tumor e biomarcadores para diversas patologias, podendo alvejar genes relacionados com inflamação e câncer como o gene ANXA1 (Anexina-A1). A Anexina-A1 é uma proteína anti-inflamatória e com ação anti-proliferativa, que se liga à receptores do tipo formil peptídeo como por exemplo FPR1 e FPR2, ambos sabidamente relacionados com a progressão de doenças como o câncer. Desta forma o presente trabalho teve como objetivos avaliar a expressão da proteína Anexina A1 e seus receptores FPR1 e FPR2, bem como avaliar a expressão do RNAm da ANXA1 e de miRNAs que possam modular a expressão desse gene (hsa-mir-27a, hsa-mir-196a e hsa-mir-222) em adenocarcinoma gástrico e correlacionar estes resultados com os aspectos clínico-patológicos. Foram avaliadas 31 amostras de adenocarcinoma gástrico, assim como as regiões metaplásica ou normal adjacentes ao tumor. A quantificação relativa (RQ) do RNAm da ANXA1 e miRNAs foi realizada por PCR quantitativo em tempo real utilizando ensaio TaqMan, e a expressão proteica da AnxA1, FPR1 e FPR2 por imuno-histoquímica e análise densitométrica. Em relação à expressão gênica relativa foram observados o aumento da expressão da ANXA1 nos tumores (RQ=1,374; p<0,001), assim como do miR-196a que apresentou aumento de expressão tanto no tecido metaplásico (RQ= 4,784; p=0,0016) e tumoral (RQ=16,99; p< 0,001) em relação ao tecido normal. O miR-27a não se apresentou diferencialmente expresso nos diferentes tecidos e houve diminuição da expressão do miR-222 (RQ=0,687; p=0,01) no tecido tumoral em relação ao tecido normal. Apenas o miR-196a e a ANXA1 apresentaram correlação significantemente inversa (r= -0,55; p=0,003), e este miRNA foi o único que apresentou associação com o sexo feminino, devido aumento de expressão em mulheres. Quando se comparou a expressão relativa aos parâmetros clínico-patológicos e tumorais não foram encontradas diferenças significativas. Quanto à expressão proteica o FPR2 não apresentou marcação no tecido normal, metaplásico e tumoral. Contudo a AnxA1 e FPR1 apresentaram imunomarcação positiva amplamente distribuída no tecido tumoral e positivamente correlacionados tanto no epitélio (r=0,87; p<0,0001) como estroma (r=0,62; p=0,004). Portanto, nossos resultados sugerem que a ANXA1 é modulada pelo miR-196a em câncer gástrico, e evidenciam que tanto a AnxA1 como o FPR1 também estão envolvidos no processo de carcinogênese gástrica. Tais achados podem abrir possibilidades para futuros estudos sobre novos alvos terapêuticos já que AnxA1 e FPR1 são possíveis alvos farmacológicos, e as terapias baseadas em microRNAs tem sido amplamente pesquisadas.<br>Gastric cancer still ranks third in cause of cancer death worldwide, despite the decline in its incidence, being Helicobacter pylori the main risk factor for this disease. This bacterium can lead to persistent inflammation via the production of proinflammatory cytokines and reactive oxygen and nitrogen species, stimulating cell proliferation and other processes involved in carcinogenesis. Still involved in this process, it has been observed the participation of miRNAs, which play an important role in post- transcriptional regulation, influencing normal physiological processes of the cell as well as those linked to diseases such as gastric cancer. Some miRNAs can act as oncogenes, tumor suppressor genes and biomarkers for various diseases, can targetting genes linked to inflammation and cancer such as ANXA1 gene (Annexin A1). Annexin A1 is an anti-inflammatory protein with anti-proliferative action that binds to formyl peptide receptors such as, FPR1 and FPR2, both known to be related to the progression of diseases such as cancer. Thus, the present study aimed to evaluate the expression of Annexin A1, FPR1 and FPR2 receptors, and the expression of mRNA ANXA1 and miRNAs (hsa-mir-27a, hsa-mir-196a and hsa-miR 222) in gastric adenocarcinoma, also correlate these results to the clinicopathological features. 31 adenocarcinoma samples were evaluated, as well as normal or metaplastic region adjacent to the tumor. Relative quantification (RQ) of miRNA and mRNA ANXA1 was evaluated by TaqMan assay and protein expression AnxA1, FPR1 and FPR2 by immunohistochemistry and densitometry analysis. Regarding the relative expression the following results were observed, increased expression in tumors of ANXA1 (RQ = 1.374; p < 0.001) and miR- 196a that showed increased expression it he metaplastic tissue (RQ = 4.784; p = 0.0016) and tumor (RQ = 16.99; p < 0.001) compared to normal tissue. The miR- 27a did not appear differentially expressed in different tissues and there was a decrease of miR -222 expression (RQ = 0.687; p = 0.01) in tumor tissue compared to normal tissue. Only the miR-196a and ANXA1 presented a significant inverse correlation (r = -0.55; p = 0.003), and this miRNA was the only one that showed association to female gender, due to increased expression in women when comparing the relative expression to clinicopathological parameters and tumor features. The FPR2 was not expressed in normal, metaplasic and tumor tissue. However, the AnxA1 and FPR1 were widely distributed positive immunostaining in tumor tissue and positively correlated both in the epithelium (r = 0.87 ; p < 0.0001) and stromal (r = 0.62 ; p = 0.004). Thus, our results suggest that ANXA1 is modulated by miR-196a in gastric cancer, and evidencing that both AnxA1 and FPR1 are also involved in gastric carcinogenesis process. Pharmacometrics is widely used in drug development. Models are developed to describe pharmacological measurements with data gathered from a clinical trial. The information can then be applied to, for instance, safely establish dose-response relationships of a substance. Glycated hemoglobin (HbA1c) is a common biomarker used by models within antihyperglycemic drug development, as it reflects the average plasma glucose level over the previous 8-12 weeks. There are five different nonlinear mixed-effects models that describes HbA1c-formation. They use different biomarkers such as mean plasma glucose (MPG), fasting plasma glucose (FPG), fasting plasma insulin (FPI) or a combination of those. The aim of this study was to compare their performances on a population and an individual level using model averaging (MA) and to explore if reduced trial durations and different treatment could affect the outcome. Multiple weighting methods were applied to the MA workflow, such as the Akaike information criterion (AIC), cross-validation (CV) and a bootstrap model averaging method. Results show that in general, models that use MPG to describe HbA1c-formation on a population level could potentially outperform models using other biomarkers, however, models have shown similar performance on individual level. Further studies on the relationship between biomarkers and model performances must be conducted, since it could potentially lay the ground for better individual HbA1c-predictions. It can then be applied in antihyperglycemic drug development and to possibly reduce sample sizes in a clinical trial. With this project, we have illustrated how to perform MA on the aforementioned models, using different biomarkers as well as the difference between model weights on a population and individual level.

Esta infecção está distribuída mundialmente persistindo como grave problema de saúde pública no Brasil. As incapacidades e deformidades duradouras decorrentes dos quadros reacionais podem ocorrer antes, durante ou após a instituição do tratamento específico. Os episódios reacionais inflamatórios são decorrentes da exacerbação abrupta da resposta imune mediada por célula contra antígenos do M. leprae. O episódio reacional tipo 1 ou reação reversa é uma ocorrência inflamatória aguda que acomete a pele e nervos periféricos, sendo causa comum de incapacidade física. A reação tipo 2 ou eritema nodoso hansênico acomete somente pacientes virchowianos (LL) e borderline-virchowianos (BL) com presença de muitos bacilos e pouca ou nenhuma imunidade celular ao M. leprae. O objetivo principal deste estudo foi avaliar a moléculas MARCO Scavenger, Receptor peptídeo formil 1(FPR1) e Receptor peptídeo N-formil 2 (FPR2) em monócitos de pacientes com hanseníase em associação ao desenvolvimento de reações hansênicas, para alcançar este objetivo células mononucleares do sangue periférico foram obtidas de pacientes com diferentes formas clínicas da hanseníase. A marcação de superfície celular foi realizada por citometria de fluxo e os dados analisados com FlowJo em 35 pacientes com hanseníase, divididos em 19 paucibacilar e 16 multibacilar. Os nossos resultados mostraram que os pacientes com a forma paucibacilar apresentam a frequência de monócitos intermediários aumentada. Não houve diferença estatística na média de intensidade de fluorescência (MFI) de CD80, CD86 e MHC II entre monócitos de pacientes com a forma paucibacilar e multibacilar da hanseníase. No entanto, monócitos intermediários de pacientes com a forma multibacilar apresentaram média de intensidade de fluorescência maior em CD86 e MCHII. Os nossos dados sugerem que o receptor FPR1 pode ter um papel relevante para detecção na amplificação da resposta inflamatória de indivíduos com hanseníase quando comparado com as outras moléculas aplicadas neste estudo.<br>Leprosy is a chronic infectious disease caused by Mycobacterium leprae, characterized by skin lesions and extensive clinical presentation which depends mainly on the immune front of the host response to infection by Mycobacterium leprae. This infection is distributed worldwide persist as a serious public health problem in Brazil. Disability and lasting deformities resulting from reaction symptoms can occur before, during or after the institution of specific treatment. Reactional inflammatory episodes are due to the abrupt exacerbation of immune cell-mediated response against M. leprae antigens. The episode reaction type 1 or reverse reaction is an acute inflammatory occurrence that affects the skin and peripheral nerves, and common cause of disability. Reaction type 2 or ENL only affects lepromatous patients (LL) and borderline-lepromatous (BL) with the presence of many bacilli and little or no cellular immunity to M. leprae. To assess the association of biomarkers MARCO Scavenger, Formyl peptide receptor (FPR1) and N-formyl peptide receptor 2 (FPR2) in monocytes of patients with leprosy associated with the development of leprosy reactions, mononuclear cells from peripheral blood were obtained from patients with different clinical forms of leprosy. To achieve this goal peripheral blood mononuclear cells were obtained from patients with different clinical forms of leprosy. The cell surface stain was performed by flow cytometry and the data analyzed with FlowJo in 35 patients with leprosy, divided into 19 paucibacillary and 16 multibacillary patients. Our results showed that patients with the paucibacillary form exhibit increased monocyte frequency. There was no statistical difference in the mean fluorescence intensity (MFI) of CD80, CD86 and MHC II between monocytes of paucibacillary and multibacillary leprosy patients. However, intermediate monocytes from patients with the multibacillary form had higher mean fluorescence intensity in CD86 and MCHII. Our data suggest that the FPR1 receptor may play a relevant role in detecting the amplification of the inflammatory response of individuals with leprosy when compared with the other molecules applied in this study.

We provide a comprehensive study of out-of-sample forecasts for the EUR/USD exchange rate based on multivariate macroeconomic models and forecast combinations. We use profit maximization measures based on directional accuracy and trading strategies in addition to standard loss minimization measures. When comparing predictive accuracy and profit measures, data snooping bias free tests are used. The results indicate that forecast combinations, in particular those based on principal components of forecasts, help to improve over benchmark trading strategies, although the excess return per unit of deviation is limited.

We compare the predictive ability of Bayesian methods which deal simultaneously with model uncertainty and correlated regressors in the framework of cross-country growth regressions. In particular, we assess methods with spike and slab priors combined with different prior specifications for the slope parameters in the slab. Our results indicate that moving away from Gaussian g-priors towards Bayesian ridge, LASSO or elastic net specifications has clear advantages for prediction when dealing with datasets of (potentially highly) correlated regressors, a pervasive characteristic of the data used hitherto in the econometric literature.

We examine the potential gains of using exchange rate forecast models and forecast combination methods in the management of currency portfolios for three exchange rates: the euro versus the US dollar, the British pound, and the Japanese yen. We use a battery of econometric specifications to evaluate whether optimal currency portfolios implied by trading strategies based on exchange rate forecasts outperform single currencies and the equally weighted portfolio. We assess the differences in profitability of optimal currency portfolios for different types of investor preferences, two trading strategies, mean squared error-based composite forecasts, and different forecast horizons. Our results indicate that there are clear benefits of integrating exchange rate forecasts from state-of-the-art econometric models in currency portfolios. These benefits vary across investor preferences and prediction horizons but are rather similar across trading strategies.

In this paper we forecast daily returns of crypto-currencies using a wide variety of different econometric models. To capture salient features commonly observed in financial time series like rapid changes in the conditional variance, non-normality of the measurement errors and sharply increasing trends, we develop a time-varying parameter VAR with t-distributed measurement errors and stochastic volatility. To control for overparameterization, we rely on the Bayesian literature on shrinkage priors that enables us to shrink coefficients associated with irrelevant predictors and/or perform model specification in a flexible manner. Using around one year of daily data we perform a real-time forecasting exercise and investigate whether any of the proposed models is able to outperform the naive random walk benchmark. To assess the economic relevance of the forecasting gains produced by the proposed models we moreover run a simple trading exercise.

Dans ce mémoire, nous avons étudié dans un premier temps des couches minces de Fe43Co43B14/SiO2 en faisant varier l’angle d’incidence (α = 0°, 25° , 45° et 75°) à épaisseur fixe, puis nous avons effectué des mesures sur une série d’échantillons de différentes épaisseurs pour chacun des angles d’incidences de dépôt. Les propriétés magnétiques statiques et dynamiques ont été caractérisées. Le rôle de la dispersion d’anisotropie a notamment été étudié. Ensuite, les propriétés magnétiques de ces échantillons ont été utilisées pour étudier l’intérêt de coupler ces couches minces à des surfaces absorbantes, constituées de différents motifs, notamment périodiques. Les résultats majeurs de cette thèse, indiquent que le FeCoB déposé à 45° est un bon candidat pour les applications hyperfréquences avec des fréquences de résonance élevées (fr ≅ 6 GHz), une valeur de la perméabilité proche de 100 et un champ d’anisotropie de l’ordre de 200 Oe à forte épaisseur<br>In this thesis, we have initially studied thin films of Fe43Co43B14/SiO2 by varying the incidence angle (α = 0°, 25°, 45° and 75°) at a fixed thickness, then we have performed measurements on a series of samples of different thicknesses for each of the deposited incidence angles. Este estudio se basa en la premisa de que la interpretación es una actividad social y que por tanto necesita describirse y explicarse en referencia al contexto cultural, político y social del escenario en que el intérprete opera. Se estudian los aspectos sociopolíticos de la interpretación en el Tribunal Militar Internacional para el Lejano Oriente (TMILO, 1946-1948) mediante una investigación archivística e histórica de la organización de la interpretación y un estudio de caso del comportamiento de los lingüistas (especialistas del idioma) que trabajaron en el proceso de interpretación.<br/> Tras exponer resumidamente la organización de la interpretación, este estudio analiza las características propias de interpretar en el TMILO. En primer lugar, investiga por qué el tribunal organizó la interpretación de forma que tres grupos diferentes social y étnicamente se encargaban de tres funciones diferentes: los ciudadanos japoneses actuaban de intérpretes en el procedimiento, los nisei (Japoneses- americanos de segunda generación) monitorizaban la actuación de los intérpretes, y oficiales militares caucasianos, en calidad de peritos lingüísticos, decidían las discrepancias en las traducciones e interpretaciones. Este estudio desvela que la carencia de lingüistas americanos competentes condujo al uso de ciudadanos japoneses como intérpretes; y que dicha estructura jerárquica funcionaba como una muestra de autoridad y para impedir la "mala fe" que, supuestamente, albergaban los intérpretes japoneses y los monitores nisei.<br/> En segundo lugar, se aplica el concepto de "normas negociadas" para examinar cómo se desarrollaron los procesos de interpretación durante el primer año del TMILO en que ninguno de los lingüistas había recibido formación como intérprete profesional y los usuarios de la interpretación no estaban familiarizados con su funcionamiento. Este examen subraya el aspecto interactivo de la creación de normas y las limitaciones cognitivas del intérprete como un factor de dicho proceso.<br/> Por último, se examina la ambigua y compleja posición de los monitores nisei. Fueron contratados por un gobierno que les había tratado injustamente como "enemigos ajenos", en un juicio contra los antiguos líderes de la patria de sus padres; y usaban habilidades arraigadas en su herencia para trabajar como monitores.<br/>Se estudia el comportamiento de los lingüistas durante los testimonios de Hideki Tojo y otros testigos japoneses centrándonos en la naturaleza de las interjecciones de los monitores y de los intérpretes y las interacciones entre los lingüistas y otros participantes del juicio. Algunos de los resultados apoyan la hipótesis que conecta el comportamiento de los lingüistas con su posición provisional en la constelación de poder de este escenario.<br/>En último lugar de la jerarquía, los intérpretes japoneses hablan en contadas ocasiones por cuenta propia y casi nunca ponen objeciones a las interjecciones aparentemente innecesarias o incluso erróneas de los monitores. Dada la situación intermedia de los monitores en el sistema de interpretación, sus aparentemente excesivas interjecciones en japonés pueden explicarse como una muestra de autoridad hacia los intérpretes japoneses y como un mensaje al tribunal (que no entendía japonés) de que estaban trabajando con eficacia corrigiendo errores de interpretación. La menor frecuencia de interjecciones en inglés por parte de estos monitores puede ser debida a la consideración por la preocupación de sus patrones por la falta de tiempo. El perito lingüístico no se involucraba en ninguna actividad espontánea, pero su presencia al lado de la fiscalía y sus anuncios de resoluciones de disputas lingüísticas en el tribunal debe haber reforzado la imagen de que el ejército estadounidense estaba al mando.<br/>Los resultados de este estudio refuerzan la idea de que la interpretación es una práctica social. La influencia de los aspectos sociopolíticos del escenario se hace evidente en la organización de la interpretación. Además, la información disponible sugiere que el comportamiento de los lingüistas era consecuente con su posición relativa en la jerarquía. El autor espera que algunos de los temas tratados en este estudio, como la confianza, la ética, las relaciones de poder y las normas negociadas, sean revisados para una mayor comprensión de los temas relacionados con el idioma en nuestra sociedad actual.<br>This study is based on the premise that interpreting is a social activity, which therefore needs to be described and explained with reference to the social, political and cultural context of the setting in which the interpreter operates. Sociopolitical aspects of interpreting at the International Military Tribunal for the Far East (IMTFE, 1946-1948) are studied through historical and archival research of the interpreting arrangements and a case study on the behavior of linguists (language specialists) who worked in the interpreting process during the testimonies of Hideki Tojo and other Japanese witnesses. Three sets of concepts are applied to analyze three salient features of interpreting at the IMTFE. Based on the notions of "trust, power and control", the historical and political context of the IMTFE and the social and cultural backgrounds of each linguist group are examined to explore why the tribunal devised the interpreting arrangements in which three ethnically and socially different groups of linguists engaged in three different functions: interpreter, monitor and language arbiter. The concept of "negotiated norms" is applied to discuss the interactional aspect of how the interpreting procedures developed over the initial stage of the trial, with the interpreters' cognitive constraints as a factor in that process. Cronin's notion (2002) of "autonomous and heteronomous interpreters" is drawn on to discuss the complex position of the Japanese American linguists who worked as monitors. The nature of interjections by the monitors and interpreters and the interactions between the court and each linguist group during the interpreted testimonies of Japanese witnesses are examined. Findings of this analysis support the hypothesis which links interpreters' choices, strategies and behavior to their awareness of where they stand in the power constellation of the interpreted event.

The systematic implementation of intermittent energy production sources has made the energy system more volatile and unpredictable than ever before. This development increases the importance of balancing services, in particular the primary control. However, the current research that has been conducted on the primary regulation products is limited. Specifically, the factors that drive the variation in availability and price of the primary control are unknown and the procurement is in some aspects based on perception rather than quantitative analysis. This study has investigated which these factors are and their relative significance on the availability and price of the primary control product called Frequency Containment Reserve for Normal Operation – FCR-N. The study was conducted according to methods with both qualitative and quantitative aspects to cover the complex nature of the problem from different views. The study was done in collaboration with Fortum POT at their office to gain insight and support from a market actor. Given the research questions, the result from this study is threefold. Firstly, the factors that affect the FCR-N availability have been identified and other factors have been categorised as insignificant. Secondly, the factors’ relative significances are stated to show their dependency with the FCR-N product. Lastly, an outlier case study showed how extreme situations changes the conditions for the FCR-N procurement. It has been concluded that the FCR-N price for normal operation on the Nordic electricity market can be generalised successfully through the identified parameters. It is also shown that deviations from normal operations have the possibility to create deviations in the FCR-N availability, indicating that these hours of extreme values need additional analysis in order to fully understand the available capacity. However, the significance of the researched parameters will be an indication in analysis of both the normal case and during deviations as these are the most important measures for the FCR-N availability and price.

I takt med att Sveriges elsystem byggs ut och allt större andel elenergi kommer från förnybar och väderberoende elproduktion såsom vind, så ökar även behovet av stödtjänster som kan balansera frekvensen i elsystemet. Frekvensen i elsystemet fungerar som en indikator på om elsystemet är i balans och för att elsystemet ska vara i balans krävs att elproduktionen hela tiden motsvarar elanvändningen. Examensarbetet har utrett möjligheterna att leverera stödtjänster från en redan befintlig vindkraftpark och har även undersökt hur möjligheterna ser ut i framtiden i och med att marknaderna för stödtjänster utvecklas. I dagsläget finns fem olika frekvensreglerande stödtjänster i Sverige. Dessa är Fast frequency Reserve (FFR), Frequency Containment Reserve-Normal (FCR-N), Frequency Containment Reserve-Disturbance (FCR-D), automatic Frequency Restoration Reserve (aFRR) och manual Frequency Restoration Reserve (FRR). Dessa stödtjänster upphandlas av Svenska kraftnät för att upprätthålla balans mellan produktion och konsumtion i elsystemet. För varje stödtjänst finns olika krav och specifikationer som bland annat vilken frekvens som de aktiveras vid, hur lång tid aktiveringen får ta samt hur långt i förväg de upphandlas etcetera. I kontakt med sakkunniga och i litteratur så framgår det att det är möjligt att leverera någon typ av frekvenstjänst från de allra flesta vindparker. För att börja leverera stödtjänster krävs att vindturbinen har en effektomriktare, vilket de flesta vindkraftverk har i dagsläget. Därefter handlar det främst om program och mjukvaruinställningar i effektomriktaren. Annars fungerar det i stort sett som vanlig pitch-reglering av turbinerna. Inom detta projekt så har även driftdata från en verklig vindpark i Blaiken studerats, där tre olika testkörningar genomförts. I testkörningarna har man testat att styra produktionen från turbinerna genom att göra nedregleringar och uppregleringar för att dokumentera hur parken reagerar. Utifrån data från testkörningarna som jämförs med krav för olika stödtjänster kan slutsatser dras om att stödtjänsterna mFRR, aFRR samt FCR-N bör kunna levereras från Blaiken. De är stödtjänsterna med de mest generösa aktiveringstiderna. Resultatet visar även på möjligheter att jobba med parkstyrningssystemet för att förbättra parkens reglerförmåga och i bästa fall skulle detta kunna medföra att parken även klarar kraven för aktiveringstid av FCR-D.

The Far-Infrared Outgoing Radiation Understanding and Monitoring (FORUM) mission has recently been selected by the European Satellite Agency (ESA) as 9 th Earth Explorer mission. FORUM mission aims at studying the water vapor and clouds by filling the long-standing gap in Far-Infrared (FIR) spectral observations from space. In the framework of the FORUM mission, this thesis analyses FIR measurements to characterize the spectral signatures of radiance in presence of ice clouds. At this purpose, a cloud identification and classification code (named CIC) is implemented. CIC is an innovative machine learning algorithm, based on principal component analysis, able to perform cloud detection and scene multi-class classification. CIC is easily adaptable to different datasets and type of spectral sensors. It is firstly tested against a synthetic dataset comprising simulated measurements of the FORUM mission. Subsequently, CIC is applied to airborne interferometric data and finally it is used for the analysis of measured downwelling radiances collected in very dry conditions on the Antarctic Plateau. Provided the excellent performances of the algorithm, especially in the identification of thin cirrus clouds, CIC is adopted as the classificator in the official ESA FORUM End-to-End simulator (FE2ES). The FE2ES is a complex chain of codes used to simulate the entire FORUM mission from satellite orbit and geometry to level 2 product analysis. An extensive use of CIC is performed on ground-based radiances collected in Antarctica. The dataset is exploited to test and to optimize the CIC algorithm and for the developing of punctual statistic of cloud occurrence in the Antarctic Plateau. Meteorological conditions from this region are also analysed and correlated with the presence of clouds. The studies presented in this work showed the potentiality and the importance of the exploitation of spectral radiance measurements in the FIR for cloud identification and classification.

Within this species there are three currently recognized varieties: C. humilis var. humilis in Northern California, C. humilis var. venusta in Southern California, and C. humilis var. jonesii in Utah and Northern Arizona. Some populations occur geographically in areas between the typical distribution of each variety and the presently accepted taxonomy inadequately addresses these populations. Using five nDNA regions, we seek to clarify relationships between current varieties and assess the pattern of variation throughout the species. Analyses including K-means clustering, principle component analysis, fields for recombination, AMOVA, and ecological niche modeling were applied. Results indicate significant genetic structure between varieties and supports recognition of C. jonesii at the species level as distinct from C. humilis. Well defined intraspecific groupings are evident in the data, with evidence supporting the recognition of an additional variety in C. humilis, and two varieties in C. jonesii. Haplotype diversity and relationships between metapopulation clusters inform conservation efforts regarding diversity within Cycladenia and offer insights into the historical demography of this genus.

Abstract:
Improvements in the medical field have given many cancer patients and survivors better odds of long-term survival. As more patients become survivors, the demand for psychological treatment becomes greater. The most prevalent concern of survivors is getting help with a psychosocial condition known as fear of recurrence (FOR). Prior to this study, few researchers had explored how having a more aggressive cancer influences the development of FOR. The purpose of this quantitative study was to determine whether cancer stage and type (a measurement of severity) are predictive of FOR development in the high-risk cancer groups lung and bronchus and female breast. The theoretical framework guiding this research was based on Mishel's theory of uncertainty in illness, which states that uncertainties about illness recurrence can cause survivors to experience breakdown in their lives (whether psychological and/or physical). The fear of cancer recurrence inventory (FCRI) survey was administered to 97 lung and bronchus and female breast cancer survivors; the survivors were asked to rate their level of discomfort about the possibility of a cancer recurrence. Data were analyzed using multiple linear regression. The results indicated that cancer type and severity both impacted the development and severity of FOR in lung and bronchus and female breast cancer survivors. Furthermore, regardless of the cancer type, stage of cancer, age of the survivor, or years in remission, survivors reported clinical levels of FOR in all areas of concern. Practitioners can use the current findings to work towards developing better intervention and treatment programs that promote quality survivorship and reduce the risk and rate of FOR in high risk cancer populations.

Fibre-reinforced polymers (FRP) are building materials that permit both the improvement of long-term building performance and the simplification of the construction process, thanks to their high specific strength, low thermal conductivity, good environmental resistance, and ability to be formed into complex shapes. FRP materials are well-suited to fulfilling many building functions. By integrating traditionally separate building systems and layers into single function-integrated components, and by industrially fabricating those components, the amount of on site labour can be greatly reduced and overall quality can be improved. The FRP materials used in the construction industry include glass fibre reinforced polymer (GFRP) and carbon fibre reinforced polymer (CFRP). Most GFRP based buildings are lacking in integration and function and only benefit for the small span, with deep beams or slabs. The CFRP based construction component has higher strength and stiffness. However, the investigation into CFRP based buildings has been lacking. This research aims to investigate the CFRP floor panel, as a primary component in the floor system, to replace traditional concrete floor slab in large buildings. The objectives of this project include the design of CFRP floor panel system in buildings using European design codes, analysing proposed CFRP floor panel by FEA modelling, and experimentally validating design and FEA models using scaled CFRP floor samples. A scale effect of test specimen was investigated in conducting design strength check of full proposed CFRP floor panel. This project supplied design curves with dimensional parameters for practical design of CFRP floor panels, to fit the design specifications required by different buildings with varied dimensions. Design curves present the measurements of deflection and critical stresses against the variation of the proposed CFRP floor panel with different dimensions. The proposed CFRP floor panel was designed as a pultruded beam with an open cross-section. The design was carried out using the Eurocodes and supported by the finite element analysis (FEA). Modelling results indicated that the proposed floor panel passed the tall design check, recommended by Eurocode, and the safety checks on both deflection and material strength, which are important for producing CFRP floor panel products that meet the dimensional requirements in design of buildings with different design specifications. Experimental results of scaled CFRP floor panel samples are also presented in this thesis, which successfully validates the design and modelling analysis. The conducted scale effect was amended by a reduction factor of 0.625 for the material strength of the full CFRP panel, which passed the Hashin criteria check. This project also studied the shear effects on bending behaviour of proposed CFRP panel with open crosssection consisting of thin-walled plates. An important load-deflection correction factor was proposed, which plays an important role together with geometrical shape factor in the calculation of shear related deflection. This novel CFRP floor panel can be easily installed in buildings because of its lightweight feature, and easily integrated with the suspending ceiling, ventilation and lighting system because of its designed shape. This investigation also provided plenty of information concerning the use of this potential building component in the low carbon construction industry, which could save up to 50% heating energy and reduce CO2 emissions by 40% compared to the traditional construction industry.

La connaissance des environnements passés représente un aspect fondamental dans la compréhension de l'histoire évolutive des êtres vivants. Ainsi, l'étude du(des) paléoenvironnement(s) associé(s) aux premiers hominidés permet de définir les contraintes environnementales dans lesquelles l'émergence de ce groupe s'est effectuée. L'objectif de ce travail était de reconstruire le(s) paléoenvironnement(s) associé(s) aux hominidés anciens : Sahelanthropus tchadensis (7 Ma, Toros-Menalla, Tchad), Orrorin tugenensis (6 Ma, Lukeino, Kenya) et Ardipithecus kadabba (5,77-5,54 Ma, Asa Koma, Ethiopie). Pour ce faire, l'étude de la structure faunique (structures taxinomique et écologique) des assemblages fossiles de mammifères associés à ces hominidés et d'autres assemblages africains du Miocène supérieur a été entreprise. Au préalable, il était indispensable de s'intéresser à la représentativité de ces assemblages fossiles par rapport à la/les communauté(s) dont ils sont issus. Ainsi, l'étude de la structure faunique des assemblages de Toros-Menalla a révélé qu'ils provenaient d'une même paléocommunauté, les variations fauniques observées d'un assemblage à l'autre étant probablement liées à des différences d'environnement de dépôt. Un modèle a également été proposé afin d'expliquer, par une sous-représentation des petites espèces, les différences observées entre les structures écologiques des faunes fossiles et celles des faunes modernes. Ces prérequis obtenus, l'analyse de la structure faunique a ensuite permis de reconstruire, à Lukeino et à Toros-Menalla, un mélange de zones arbustives et de zones boisées plus ouvertes. Asa Koma présenterait un environnement plus fermé, étant principalement constitué de formations arbustives. Cette dernière composante végétale semblait donc constituer un élément indispensable à la présence des hominidés anciens. Ces derniers étaient par ailleurs systématiquement associés à une combinaison de taxons (Orycteropus, Giraffa, Hystrix, Aepyceros et Deinotherium), absente des autres gisements africains contemporains.

L'hémochromatose génétique liée au gène HFE (HG) se caractérise par une augmentation de la saturation de la transferrine plasmatique, qui entraîne une surcharge en fer. Ces anomalies sont dues à une sécrétion basse d'hepcidine, le régulateur principal du métabolisme du fer, et définissent le phénotype d'hepcidino déficience. De nouvelles formes de surcharges en fer ayant un phénotype similaire ont été identifiées. Les mécanismes moléculaires en sont parfois mal compris. L'objectif général de ce travail a été de caractériser sur le plan clinique, fonctionnel, et biologique, des surcharges en fer rares d'origine génétique ayant pour point commun un phénotype d'hepcidino-déficience. Nous avons d'abord analysé une cohorte de patients porteurs d'une HG, forme type de l'hepcidino-déficience, et montré que la transplantation hépatique normalisait le métabolisme du fer, montrant ainsi le rôle majeur du foie dans le phénotype d'hepcidino-déficience de l'HG. Nous avons ensuite caractérisé des surcharges en fer à saturation de la transferrine élevée liées à des anomalies de transport du fer : i) nous avons rapporté un cinquième cas de mutations du gène DMT1 et montré le caractère pathogène de la mutation p.Asn491Ser ; ii) nous décrivons un groupe de 12 patients qui présentent une mutation hétérozygote du gène TF, l'augmentation de la saturation de la transferrine ne semblant pas être en rapport avec une hepcidino-déficience, et l'existence de cofacteurs pouvant faciliter la surcharge en fer. Nous avons ensuite décrit l'impact de mutations du gène TFR2, qui sont responsables d'une hepcidino déficience dont l'expression clinique est hétérogène mais qui peut être très précoce. Nous avons alors, pour préciser les mécanismes liant les mutations du gène TFR2 à une diminution anormale de l'hepcidine, montré in vitro que les mutations p.Asn12Ile et p.Gly430Arg altèrent l'adressage cellulaire de TFR2 tandis que la mutation p.Arg768Pro altère son interaction avec la transferrine. N'ayant pu parvenir à induire l'expression d'hepcidine sous l'effet de la transferrine, au contraire de certaines équipes mais en accord avec d'autres, nous n'avons pu analyser l'impact des mutations sur la transduction du signal liant TFR2 et hepcidine. Nos résultats contribuent à préciser les mécanismes impliqués dans l'apparition de surcharges en fer rares à saturation élevée de la transferrine<br>HFE related hemochromatosis (HH) is characterized by an increased plasma transferrin saturation level, which causes iron overload. These anomalies are due to low hepcidin secretion, the key regulator of iron metabolism, and define the hepcidin deficiency phenotype. New forms of iron overload with similar phenotype were identified, but their molecular mechanisms remain unclear. The main objective of this work was to characterize the clinical, functional, and biological aspects of rare genetic iron overload with an hepcidin deficiency phenotype. We firstly analyzed a cohort of HH patients, archetype of hepcidin-deficiency and showed that liver transplantation cured the disease, demonstrating the major role of the liver in the phenotype. We then characterized iron overloads with high transferrin saturation related to abnormalities of iron transport: i) we reported a fifth case of patient with DMT1 gene mutations and demonstrated the pathogenicity of the mutation p.Asn491Ser; ii) we described 12 patients with heterozygous mutation of the TF gene, leading to serum transferrin decrease. The increase of transferrin saturation associated to the disease does not seem to be related to a hepcidin-deficiency, and the presence of cofactors may facilitate iron overload. We then described the impact of mutations in the TFR2 gene, which induce hepcidin-deficiency whose expression is heterogeneous but that can occur in young peoples. Then we tried to clarify in vitro the mechanisms linking mutations in the TFR2 gene to an abnormal decrease of hepcidin and showed that the p.Asn12Ile and p.Gly430Arg mutations alter the TFR2 protein intracellular trafficking while the p.Arg768Pro mutation alters its interaction with transferrin. Being unable to induce expression of hepcidin in response to transferrin, unlike some authors, but in agreement with others, we were not able to analyze the impact of mutations on signal transduction toward hepcidin. The transition from old-growth to second-growth British Columbia coastal Douglas-fir has resulted in reduction of log size and increased proportion of juvenile (core or crown-formed) wood. Determination of the zone of transition from juvenile to mature wood is critical to the definition of wood quality and timber value. Thirteen unpruned, two pruned second-growth, and two unpruned plantation-grown coastal Douglas-fir trees were sampled to analyze the hypothesis that the transition in relative density from juvenile to mature wood occurs at the base of the live crown. X-ray densftometric techniques were utilized to determine yearly pith to bark relative density data of five cross-sectional discs from each tree. Segmented linear regression techniques were utilized to estimate the juvenile - mature wood transition age from the data. The average number of growth increments from the pith at which juvenile - mature wood transition occurred on sections sampled at breast height, 20 percent and 40 percent of total height was 22.18. When the hypothesis was tested on unpruned trees, before and after harvest, the juvenile - mature wood transition occurred below the base of the live crown. When the hypothesis was tested on pruned trees, the transition occurred at the base of the live crown, which represented the upper limit of pruning height. This information may provide a greater insight into juvenile - mature wood transition. It will likely assist in the determination of wood quality and economic value of forest products manufactured from second-growth coastal Douglas-fir.<br>Forestry, Faculty of<br>Graduate

Therefore, how to search for the required OLAM cube from such a massive amount fo OLAM cubes will affects the OMARS system efficiency. The aim of this thesis is the construct a multidimensional index structure in order to search all OLAM cubes and can effectively retrieve the qualified OLAM cube. To this end, we propose a three-layer index structure.Experimental results show this schema can retrieve the required OLAM cube efficiently.
