Abstract:
Several proteins with high proportions of S-containing essential amino acids were incubated in sheep rumen fluid in vitro and their rate of digestion was examined by sodium dodecyl sulphate-polyacrylamide-gel electrophoresis. The S-rich proteins rice prolamin (10 kDa), maize zein (10 kDa) and the 3·2 kDa pumpkin (Cucurbita maxima L.) trypsin inhibitor-1 (CMTI-1) were highly resistant to rumen fluid degradation, relative to control proteins of known degradation rate (casein, bovine serum albumin (BSA) and pea (Pisum sativum) albumin-1 (PA1)). Comparison of PA1 and a recombinant N-terminal epitope-tagged PA1 indicated that addition of the epitope caused a slight increase in resistance to rumen degradation. The proteins were also incubated with a mixture of trypsin (EC 3·4·21·4) and chymotrypsin (EC 3·4·21.1). PA1, BSA and casein were hydrolysed less rapidly than rice prolamin, maize zein and CMTI-1. Digestion by these intestinal proteases appeared to be complete. Thus, the prolamin, zein and CMTI-1 proteins are suitable candidates for expression as foreign proteins in pasture plants to increase throughput and uptake of essential amino acids in sheep.

Abstract:
e12572 Background: The aim of this study was to identify markers for the early diagnosis of brain metastasis by metabolomic methods in breast cancer patients. Methods: A total of 88 breast cancer patients with distant metastases were included the study. The patients were divided into two groups according to their metastasis status as patients with brain metastases and patients with distant metastases without any brain metastases. For metabolomic analyses, liquid chromatography quadrupole time-of-flight mass spectrometry (LC-qTOF-MS) and gas chromatography mass spectrometry (GC-MS) analysis methods were used. Results: 88 patients were included the study. 33 of the 88 patients had brain metastasis group and 55 patients had distant metastases without brain metastasis. A total of 79 metabolites were identified by the GC-MS analysis. Of these, 11 of them (alanine, sphingosine, fructose, fumaric acid, glycine, lactic acid, phenylalanine, pyroglutamic acid, serine, threonine, and valine) were found at statistically significantly higher levels in the patient group with brain metastases (p < 0.05). In LC-qTOF-MS analysis 47 metabolites were identified with statistically significant results between the two groups. Predictive accuriecies for idendification of the brain metastasis with 5 and 10 metobolites models were 94.6% and 95.2%, respectively.Amino acyl tRNA biosynthesis, arginine and proline metabolism, nitrogen metabolism, cyanaminoacid metabolism, nicontic and nicotinicamide metabolism, glycine, serine and threonine metabolism pathways have been involved more significantly in patients with brain metastasis (p < 0.05). Conclusions: Although these results need to be confirmatory prospective studies, these data promising for early detection of the brain metastasis by markers in sera.

Abstract:
Une variation du nombre de copies des gènes du chromosome 21 humain (HSA21) entraine des anomalies morphologiques et physiologiques d'un grand nombre d'organes chez les patients. La Trisomie 21, ou syndrome de Down et les monosomies partielles du HSA21 conduisent à des phénotypes complexes et variables. Afin d'identifier les gènes du HSA21 sensibles aux effets de dose, nous avons développé le modèle de monosomie Ms1Yah, pour la région Prmt2-Col6a1 du chromosome murin 10 (MMU10). L'analyse de ce modèle a montré que les souris Ms1Yah développent une altération des réponses inflammatoire et pulmonaire suite à une instillation de lipopolysaccharide (LPS). L'analyse, par Q-PCR, de l'expression des gènes de la région a mis en évidence deux gènes candidats : Prmt2 et S100B. Les lignées déficientes pour ces gènes ont été utilisées pour tester l'implication de Prmt2 et S100B dans la modification de la réponse inflammatoire et pulmonaire en réponse au LPS. Ces travaux ont montré d'une part que Prmt2 intervient dans la régulation de l'expression des cytokines pro-inflammatoires TNF-α et IL-6 de façon dose dépendante, via son rôle inhibiteur de la signalisation NF-κB. D'autre part, S100B ne semble pas intervenir dans la réponse inflammatoire induite par le LPS, alors que son inactivation entraîne une diminution de l'hyper réponse des voies aériennes (HRA) de façon dose dépendante. La réponse inflammatoire serait la résultante du croisement de deux voies de signalisation : LPS-TLR4-NF-κB-Prmt2 et S100B-RAGE-NFκB. Il faudrait que la voie Prmt2 soit en défaut pour observée un effet de S100B dans l'inflammation. L'injection d'un anticorps anti-S100B chez des souris B6 favorise une diminution de l'HRA induite par l'instillation de LPS. L'analyse préliminaire de l'utilisation de cet anticorps dans un modèle d'asthme montre là encore une capacité à diminuer l'HRA pour les doses les plus fortes de métacholine, ce qui laisse entrevoir le potentiel thérapeutique d'une telle molécule.
