Abstract:

This thesis concerns the estimation of extreme quantiles on a dose-response curve. It focuses on the Robbins-Monro and up-and-down procedures. Simulation studies run in search of the LD15 and LD30 using a variety of methods suggest that the Robbins-Monro procedure is optimal in terms of Monte Carlo MSE and bias. The up-and-down procedure's performance differs in many cases only slightly from that of the Robbins-Monro process, therefore indicating its value as a practical alternative to the Robbins-Monro process for extreme-quantile estimation.

Longevity is increasing and retirees face the risk of outliving their income resources, perceived by the (compulsory) second pillar pension scheme. The sequence of 1360 base pairs of the P22 PL operon was determined, linking a continuous sequence from PL through abc2. P22 mutants bearing deletions in the sequenced region were constructed and tested for their phenotypes. Plasmids were constructed to express PL operon genes singly and in combinations from PlacUV5. Substance P (SP) is an undecapeptide whose functions are as varied as its locations. In the nervous system, it is thought to act as a neurotransmitter. In the peripheral vasculature, it has hypotensive effects and it causes contraction in the smooth muscle of the gut. In salivary gland, it is a potent secretagogue and it is how this effect is transduced that is the subject of this dissertation.
