Full textParticle code that extends the capabilities of the MCNP code, developed at the Los Alamos National Laboratory in New Mexico, was used. The study considers how various parameters such as (1) coincidence summing, (2) volume, (3) atomic number (Z) and (4) density, affects the absolute photopeak efficiency of the ERL&rsquo
s HPGe detector in a close geometry (Marinelli beaker) for soil, sand, KCl and liquid samples. The results from these simulations are presented here, together with an intercomparison exercise of two MC codes (MCNPX and a C++ program developed for this study) that determine the energy deposition of a point source in germanium spheres of radii 1 cm and 5 cm.
A sensitivity analysis on the effect of the detector dimensions (dead layer and core of detector crystal) on the photopeak detection efficiency in a liquid sample and the effect of moisture content on the photopeak detection efficiency in sand and soil samples, was also carried out. This study has shown evidence that the dead layer of the ERL HPGe detector may be larger than stated by the manufacturer, possibly due to warming up of the detector crystal. This would result in a decrease in the photopeak efficiency of up to 8 % if the dead layer of the crystal were doubled from its original size of 0.05 cm. This study shows the need for coincidence summing correction factors for the gamma lines (911.1 keV and 968.1 keV) in the 232Th series for determining accurate activity concentrations in environmental samples. For the liquid source the gamma lines, 121.8 keV, 244.7 keV, 444.1 keV and 1085.5 keV of the 152Eu series, together with the 1173.2 keV and 1332.5 keV gamma lines of the 60Co, are particularly prone to coincidence summing. In the investigation into the effects of density and volume on the photopeak efficiency for the KCl samples, it has been found that the simulated results are in good agreement with experimental data. For the range of sample densities that are dealt with by the ERL it has been found that the drop in photopeak efficiency is less than 5 %. This study shows that the uncertainty of the KCl sample activity measurement due to the effect of different filling volumes in a Marinelli beaker is estimated in the range of 0.6 % per mm and is not expected to vary appreciably with photon energy. In the case of the effect of filling height on the efficiency for the soil sample, it was found that there is a large discrepancy in the trends of the simulated and experimental curves. This discrepancy could be a result of the use of only one sand sample in this study and therefore the homogeneity of the sample has to be investigated. The effect of atomic number has been found to be negligible for the soil and sand compositions for energies above 400 keV, however if the composition of the heavy elements is not properly considered when simulating soil and sand samples, the effect of atomic number on the absolute photopeak efficiency in the low energy (<
400 keV) region can make a 14 % difference.

Full textSignal detection, localization, and separation problems date back to the beginning of the twentieth century. Nowadays, this subject is still a hot topic receiving more and more attention, notably with the rapid growth of wireless communication systems that arose in the last two decades and it turns out that many challenging aspects remain poorly addressed by the available literature relative to this subject. This thesis deals with signal detection, localization using temporal or directional measurements, and separation of dependent source signals. The main objective is to make use of some available priors about the source signals such as sparsity, cyclo-stationarity, non-circularity, constant modulus, autoregressive structure or training sequences in a cooperative framework. The first part is devoted to the analysis of (i) signal’s time-of-arrival estimation using a new minimum error rate based detector, (ii) noise power estimation using an improved order-statistics estimator and (iii) side information impact on direction-of-arrival estimation accuracy and resolution. In the second part, the source separation problem is investigated at the light of different priors about the original sources. Three kinds of prior have been considered : (i) separation of constant modulus communication signals, (ii) separation of dependent source signals knowing their dependency structure and (iii) separation of dependent autoregressive sources knowing their autoregressive structure

In the FSA method, the full energy spectrum is considered and the measured spectrum is described as the sum of the three standard spectra (associated with U, Th and K, respectively), each multiplied by an unknown concentration. The concentrations are determined from the FSA and correspond to the activity concentrations of U, Th and K in the soil. The standard spectra derived from separate calibration measurements using the HPGe detector, represents the response of the HPGe to a Marinelli sample beaker containing an activity concentration of 1 Bq/kg.

Full textIn this study we report on the effect of the deposition parameters on the morphology and structural properties of CNTs, synthesized by means of the hot-wire chemical vapour deposition technique. SEM, Raman and XRD results show that the optimum deposition conditions for the HWCVD synthesis of aligned MWCNTs, with diameters between 50 and 150 nm and lengths in the micrometer range are: Furnace temperature of 500 º
C, deposition pressure between 150 and 200 Torr, methane/hydrogen dilution of 0.67 and a substrateto- filament distance of 10 cm.

Full textChronic pain is an incapacitating and long lasting pathology mainly characterized by threesymptoms: allodynia (a non painful stimulus is perceived as painful), hyperalgesia (a painfulstimulus is perceived as more painful) and ambulatory pains. When chronic pain is due to alesion or dysfunction of nervous system it is called neuropathic pain. In both patients and animalmodels of neuropathic pain, researchers found that thalamic neurons are hyperexcitable. Glialcells, astrocytes and microglia, are strong synaptic partners involved in synaptic transmissionand plasticity and therefore could be involved in this phenomenon. Indeed, these cells canmodify their phenotype when nervous system is damaged. They become reactive: theirmorphology is hypertrophied, mRNA and protein expression of iba-1 (ionized binding-adaptormolecule 1) and CD11b/c (cluster of differentiation 11b/c) for microglia and GFAP (glialfibrillary acidic protein) and S100β (S100 calcium binding protein β) for astrocytes is increased.They could also release pro-inflammatory molecules. All of these could contribute to generate oramplify the thalamic neuronal hyperexcitability.In my PhD work I studied thalamic astrocytes and microglia in a rat neuropathic pain model ofL5-L6 spinal nerves ligation (SNL). Mechanical allodynia and hyperalgesia were characterizedwith von Frey filament test and ambulatory pain with dynamic weight bearing apparatus. mRNAexpression of glial markers were studied with qRT-PCR technique on thalamic punches andlaser-microdissected nuclei. Neurochemical expressions of iba-1, CD11b/c, cathepsin S, GFAPand S100β markers were quantified using an immunohistofluorescence approach to count thenumber of immunopositive cells and surface stained by these markers. All these experimentswere done at D14 and D28 after surgery.At D14, SNL animals develop mechanical allodynia and hyperalgesia as well as ambulatory pain..For these animals, thalamic microglial cells showed signs of reactivity with the increase mRNAexpression of CTSS and CX3CR1, fractalkine receptor, well known markers involved in spinalneuronal hyperexcitability under neuropathic pain conditions. In addition, the number ofimmunopositive cells for the glial markers is decreased in SNL animals. At D28, the neuropathicpain symptoms are still present. Furthermore, thalamic microglial reactivity found at D14 withqRT-PCRm method is still present with the increased mRNA expression of fractalkine (CX3CL1),partner of CTSS/CX3CR1/CX3CL1 pathway. The decreased neurochemical expression of glialmarkers found at D14 was transient as I didn’t find this result at D28. However, thalamicastrocytic reactivity was found at D28 in SNL animals.So, this work reveal a new glial process at thalamic level in this SNL model of neuropathic pain :an early decreased expression of glial markers and then a later thalamic astrocytic reactivityconcomitant with signs of thalamic microglial reactivity. Numerous studies are required toexplore the role of such novel ambivalent glial alterations in the context of neuropathic pain

Full textGamma ray spectroscopy has provided enormous amounts of information on the behaviour and structure of atomic nuclei [SHA88, BEA92, EBE08]. Most of the major discoveries in experimental nuclear physics over the last five decades are strongly associated with improvements in detector technologies. Inorganic scintilators led to the discovery in 1963 of the first excited states of a rotational band based on the ground state of 162Dy. Improvements in peak-to-background ratios and detector resolutions obtained with germanium led to the first evidence of backbending which is associated with a two quasi-particle excitation in 162Dy [SHA88]. More recently the development of composite and highly-segmented Ge detectors has significantly increased the performance and power of detection systems. The Clover detector is such a detector system and is in use at iThemba LABS. This study concerns the evaluation of the particle transport code MCNPX 2.5.0 as a tool to model complex composite detectors such as the Clover. Lanthanum silicate (LSO) and Lead tungstate (PbWO) are also evaluated as possible suppressor shield materials. It is shown that reasonable agreement between experiment and simulation is found when the experiment is accurately reproduced. However, when complex detection modes are implemented in the detector based on the number of elements that fire, MCNPX cannot be used to model the detector performance exactly. Differences between simulated and experimental results are found in suppressed add-back mode. It is proposed that the discrepancies are due to limitations in implementation of the pulse-height and special anti-coincidence tally in MCNPX. LSO and PbWO are compared to BGO as suppressor shield materials. It is found that LSO is not an ideal material for a suppression shield. PbWO is shown to give performance values similar to that of BGO. The back-plug is shown to have no effect on the Peak-to-Total ratio but is effective at reducing the background at lower energies.

Full textAlthough the voluntary intake of drugs of abuse is a behavior largely preserved throughout phylogeny, it is currently unclear whether pathological drug use ("addiction") can be observed in species other than humans. Here, we report that behaviors that resemble three of the essential diagnostic criteria for addiction appear over time in rats trained to self-administer cocaine. As in humans, this addiction-like behavior is present only in a small proportion of subjects using cocaine and is highly predictive of relapse after withdrawal. This addiction-like behaviour seems to be associated with a compulsive seeking of the drug but neither with a differential intake of the drug nor with a disinhibited behaviour or a different anxiety level. This addiction-like behaviour is predicted by behavioural factors relative to anxiety and novelty seeking and is associated with transcriptionnal regulations mainly in the prefrontal cortex

Full textEndothelial progenitor cells (EPCs) seem to be a promising option to treat patients with ischemic diseases. Here, we investigated the effects of late EPCs or Endothelial Colony-Forming Cells (ECFCs), a recently defined homogeneous subtype of EPCs, in a rat model of transient middle cerebral artery occlusion (MCAO). Either vehicle or 4.106 ECFCs, isolated from human cord blood, were intravenously injected 24h after 1 hour of MCAO in rats assigned to control and transplanted groups respectively. 111In-oxine-labeled ECFCs specifically homed to ischemic hemisphere and CM-Dil prelabeled ECFCs preferentially settled in the inner boundary of the core area of transplanted animals. The presence of human cells in rat brain sections was detected by immunohistochemical staining (MAB1281, CD31). We demonstrated that ECFCs injected 24h after MCAO improved functional recovery and survival rate. Beneficial effect was associated with an increase in growth factors expression in homogenates from ischemic area (VEGF, IGF-1, proBDNF) and may be related to the secretion by ECFCs of soluble factors that could affect apoptosis, vascular growth and neurogenesis. These findings raise perspectives for the use of ECFCs as a well-characterized cell therapy product for optimal therapeutic outcome after stroke

Full textSeveral studies indicate that β3-adrenoceptors (β3-AR) responsiveness is generally decreased both in human hypertension and in vanous animal models of hypertension. However, β3-AR dysfunction has not yet been thoroughly established. We have shown that functional β3-AR-mediated relaxation was unaffected in spontaneously hypertensive rats (SHR) at 12 weeks old in spite of the upregulation of the β3-AR. . . . Our findings suggest that induction of cyclo-oxygenases by endotoxin plays a major role in this alteration. . In SHR rats at 12 weeks old, the maintenance of functional β3-AR-mediated relaxation in hypertensive rats suggests a potential beneficial role of β3-AR in the treatment of hypertension. However, impairment of atypical β3-AR-mediated relaxation may be involved in the pathogenesis of the hypertension. In equine laminitis, endotoxin-induced impairrnent of β3-AR-induced relaxation in equine digital veins, suggests that β3-AR could play a role in vascular disorders associated with the action of endotoxin in equine acute laminitis

Full textAdolescent diet often contains an excessive amount of palatable food with high caloric and sugar contents. Clinical and epidemiological data show that overconsumption of these large foods and sugar beverage consumption may have consequences in adolescence during which the brain is still maturing.Several studies have shown that sugar overconsumption during adolescence have long lasting effect of disease incidence such as obesity, diabetes … However, little is known in the long lasting effect of behavior disorder related to sugar overconsumption early in life. Therefore we set up an animal model to investigate the long lasting effect of sugar overconsumption during adolescence (PND 30 - 46) in neurobiological and behavioral alterations at adulthood. Our results reveal that chronic free access to sugar during adolescence decreases motivation and induce a vertical downshift of dose response curve for natural sweet and no sweet solution at adulthood with no behaviour consequences for pharmacological reward such as cocaine. These behavioral alterations are specific: 1) to the 5% sucrose concentration which is the more rewarding dose in our rats; 2) to a long access to 5% sucrose because only these alterations of behavior are observed after 12h et 24h/day access but not after 2h/day access; 3) to the sweet taste of the solution exposure during adolescence but not to its caloric effect since saccharine (a no caloric and sweet solution) induces these behavioral alterations; 4) to the adolescent period because we show no effect when adult rats have access to a sucrose solution; 5) is not dependent on the rearing conditions during adolescence since singly housed or group housed rats show the same behavioural disorder. We also show that these long lasting behavioral alterations to palatable foods are associated to anxiogenic-like behaviors and a depressive-like state evaluated in the Novelty suppressed feeding test and the forced swimming test respectively. Chronic imipramine treatment from the end of adolescence onwards (PND 47) prevents this behavioral disorder. Immunohistochemical studies show that sucrose exposure during adolescence decreases neurogenesis in the dentate gyrus of the hippocampus and imipramine treatment restores this deficit. These results suggest the importance of diet during adolescence in the development of neuropsychiatric disorders at adulthood. Full textThe aim of this study was to compare the water quality of raw wastewater and treated sewage effluents from three different sewage treatment plants in the Western Cape, South Africa. The treatment plants investigated are on the same river system. Sewage treatment plant 1 and 2 use older technologies, while sewage treatment plant 3 has been upgraded and new technologies (membrane bioreactor) were incorporated in the treatment processes. The first objective was to determine the occurrence of total coliforms, Escherichia coli (E. coli) and fluoroquinolone and sulfamethoxazole antibiotic residues in raw wastewater and treated sewage effluents. Bacteria in treated sewage effluents can result in diseases such as dysentery, gastroenteritis, and typhoid upon exposure. A chromogenic test was used to screen for coliforms and E. coli. Enzyme linked Immunosorbent Assays (ELISA) were used to quantitate antibiotic residues (fluoroquinolones and sulfamethoxazole) in raw wastewater and treated sewage effluents. This study showed that bacteria are present in raw wastewater and residual bacteria are released with treated sewage effluents from sewage treatment plants.

Full textWith the development of wireless technologies, electromagnetic fields became an important environmental constraint. However, some people attribute symptoms to electromagnetic fields exposure. The results of experimental studies remain controversial due to contradictory results. The aim of this work is to evaluate the effects of a co-exposure between electromagnetic fields and noise. The hypothesis is that co-exposure led to an exacerbation of the electromagnetic fields effects. This work focused on different physiological functions in a juvenile population: sleep, immune system, food intake, respiratory parameters and behavior. The results showed a higher anxious behavior, a decrease of locomotor parameters and an increase in the weight of the animals, associated with variations in the food intake pattern. Sleep is slightly altered in animals exposed to electromagnetic fields. The immune system of animals exposed to electromagnetic fields exhibits alterations in the adaptive immune system by a redistribution of lymphocyte subpopulations in favor of cell activation and humoral immunity, but without variation of the innate immune system. The alteration of immune system was observed during the co-exposure with a different way than noise exposure. This work was the first study to show a variation of immune parameters in juveniles chronically exposed to electromagnetic fields
