Abstract:
Dental infections are one of the most common oral health issues in the world. Plaque-forming bacteria found in the oral cavity, such as Myces, Actinobacillus, Streptococcus and Candida species, cause them. Despite the widespread use of tooth brushes and tooth pastes, chewing sticks have remained a popular and accepted teeth cleaning tool in many parts of the world, particularly in developing countries. Furthermore, approximately 72 percent of the country's rural population uses a range of plant species as a tooth brush. An attempt was made to document all of the information found in ancient and modern literature, as well as information gathered during ethnobotanical surveys in various tribal and rural areas of the country. Present review highlights some of the plant species used as chewing-stick practices of specific Gujarat state. Keywords: Chewing-stick, plants, Gujarat state.

Abstract:
This paper presents the results of a biodiversity study of microfungi in ornithogenic soils from Beaufort Island (Ross Sea, continental Antarctic). During the 2004/05 austral sum-mer, we sampled a wide range of soil habitats from an abandoned penguin rookeries to examine the biodiversity of soil microfungi. Beaufort Island is predominantly ice and snow covered, isolated, difficult to access and known to have been visited only infre-quently. Warcup's soil plating method was used for fungal cultivation. A total of ten fungal taxa were isolated, consisting of seven ascomycetes, two anamorphic fungi and one yeast. In terms of their thermal classes, a total of four psychrophilic, five psychro-tolerant and 1 mesophilic fungi were isolated. Thelebolus microsporus, Geomyces sp. and Thelebolus sp. were the most common isolated fungi. Internal Transcribed Spacer (ITS) and 18S rDNA sequences were obtained from 17 fungal isolates, confirming their identification as Thelebolus microsporus, Thelebolus sp., Phoma herbarum and Geo-myces sp.

Abstract:
In the mid-1950s, two distinguished Italian researchers, the mycologist Raffaele Ciferri (1895–1965) and the phytosociologist Ruggero Tomaselli (1920–1982), decided that a separate system of naming lichen mycobionts independent of the intact thalli was necessary. In this they were stimulated by the pioneering studies of Thomas (1939) in particular, which showed how different cultured isolated mycobionts appeared from whole lichen thalli. Thomas had already introduced separate names, ending in the suffix “-myces”, and they determined to introduce a comprehensive suite of generic names for lichen mycobionts in a series of papers (Tomaselli & Ciferri 1952; Ciferri & Tomaselli 1953, 1954, 1957). The numbers of generic names coined and validly published with Latin diagnoses were substantial: 15 in 1952, 203 in 1953, and a further nine in 1954. They made proposals to change the provisions of the Code, which then already ruled that names given to lichens were to be regarded as applying to their fungal components, but these were defeated and never adopted; this rule remains in the Code in Art. 13.1 (Greuter et al. 2000).

Abstract:
Mycorrhizae are typical structures found in a plant's root system found symbiotic mutualism between fungi (myces) and roots (rhiza). Mycorrhizae have an essential role for plants because they can increase water and nutrient uptake, especially phosphorus absorption. The purpose of this study was to determine the mycorrhizal genera on the roots of parring bamboo plants (Gigantochloa atter) in Sabantang Hamlet, Toddopulia Village, Tanralili District, Maros Regency because there is no scientific information regarding this issue. This research was conducted with the wet filter method or sieving. The isolation of spores from the rhizosphere of Gigantochloa atter samples was carried out by referring to the pouring method and wet sieving using a stratified filter set. Staining techniques were used to observe the colonization of arbuscular mycorrhizal structures in the roots of the sample plants. The results showed that 27 spores were found, consisting of 15 spores of the Glomus genera, three spores of the Gigaspora genera, and nine Acaulospora genera. Observation of the root structure has not shown the presence of vesicular and arbuscular. However, only hyphae and spore structures were found because the mycorrhizal hyphae in Gigantochloa atter have not yet reached the infection stage to form arbuscular or vesicular structures

Abstract:
Response of Growth and Yield of Sweet Corn to Dosage of Spores of Indigenous Endomycorrhizae. Endomycorrhizae is a form of mutualism association between fungi (myces) and roots (rhiza) of higher plants. The symbiotic relationship that occurs at the roots is mutually beneficial. Plants can provide carbon organic compounds for fungal growth, whereas fungi benefit plants by increasing nutrient uptake, water, producing enzymes, antibiotics and other compounds given to host plants. The study was conducted to know the effect of dosage of spores of endomycorrhizae indigenous to growth and yield of sweet corn. The experiments were carried out in laboratories and greenhouses. The design used was a completely randomized design with one factor namely the dose of spores of endomycorrhizae consisting of three levels, i.e. D0 = without inoculation of endomycorrhizae, D1 = 75 spores of endomycorrhizae/plant, and D2 = 150 spores of endomycorrhizae/plants. The results showed that dose of spores of endomycorrhizae treatment had a significant effect on growth and yield of sweet corn. The highest fresh weight of cob was obtained at a dose of 150 spores/plant, namely 181.33 g / plant. Based on the results of this study, it is necessary to conduct research in the field using a dose of 150 spores of endomycorrhizae per plant.

Abstract:
Our intention was to trace the origins of names that every microbiologist comes to pronounce or write down in her/his daily practice and, particularly, to elucidate the etymological debt of this medical field to the two great languages of classical antiquity, Greek and Latin. According to the system of binomial nomenclature established by Linnaeus in the 1750s to give a unique name to each species, consisting of two terms, the name of the genus and the name of the species, combinations were based on Greek and Latin. The two major cocci consist of a literally classical example. Streptococci, named so by Bilroth from the Greek word “στρεπτόν—strepton” (necklace), and staphylococci, baptised by Ogston from “staphyli—σταφύλι” (grape), were both so named due to their appearance under microscopy. Two well-known species of these genera bear Latin names denoting colour, greenish for S. viridans and golden for S. aureus. Other bacteria display the discovering scientist like, E. coli for Escherich, Brucella from Bruce, or Pasteurella from Louis Pasteur. Similar pathways also exist for viruses, even back to Hippocrates, like Herpesviruses from “έρπης—herpes” (creeping) or Arenaviruses from “arena—sand”, and fungi, as mycology itself originates from “μύκης—myces”, Greek for fungus, and Aspergillus from the verb “aspergo” (to sprinkle).

Abstract:
Increasing evidence supports an important biological role for Myc in the downregulation of specific gene transcription. Recent studies suggest that c-Myc may suppress promoter activity through proteins of the basal transcription machinery. We have previously reported that Myc protein, in combination with additional cellular factors, suppresses transcription initiation from the c-myc promoter. To characterize the cis components of this Myc negative autoregulation pathway, fragments of the human c-myc promoter were inserted upstream of luciferase reporter genes and assayed for responsiveness to inducible MycER activation in Rat-1 fibroblasts. We found four- to fivefold suppression of a c-myc P2 minimal promoter fragment upon induction of wild-type MycER protein activity, while induction of a mutant MycER protein lacking amino acids 106 to 143 required for Myc autosuppression failed to elicit this response. This assay is physiologically significant, as it reflects Myc autosuppression of the endogenous c-myc gene with regard to kinetics, dose dependency, cell type specificity, and c-Myc functional domains. Analysis of mutations within the P2 minimal promoter indicated that the cis components of Myc autosuppression could not be ascribed to any known protein-binding motifs. In addition, to address the trans factors required for Myc negative autoregulation, we expressed MycEG and MaxEG leucine zipper dimerization mutants in Rat-1 cells and found that Myc-Max heterodimerization is obligatory for Myc autosuppression. Two models for the Myc autosuppression mechanism are discussed.

Abstract:
Chitinases are glycosyl hydrolases, that cleave the ?-1,4 linkage between N-acetyl glucosamines present in chitin chains. Chitin is the second most abundant polysaccharide on Earth after cellulose, and it is produced in the exoskeleton of crustaceans and insects, and in some parts of the cell walls of fungi. Enzymatic development and the extraction of superior derivatives from chitin wastes - such as chitooligosaccharides with vast importance in the medical and biofuels industry - lead to the necessity of creating chitinases using different strains of organisms. In this paper, the chiA gene from the Bacillus licheniformis DSM8785 encoding chitinase A (ChiA) with C-terminal hexahistidine tag was cloned and expressed in the extracellular expression system pYES2 from Saccharomyces cerevisiae INVSc1 as a hyperglycosylated enzyme. The production of recombinant ChiA was successfully confirmed by dot blotting, using anti-His antibodies. The optimal time of expression was identified to be 24 h when galactose was added only at the beginning of fermentation, the chitinase activity starting to decrease after this threshold. Nevertheless, in another experiment, when galactose was added every 24 h for 72 h, the expression continued for the entire period. The purified enzyme was detected, using sodium dodecyl sulphate-polyacrylamide gel electrophoresis (SDS-PAGE), as a heterogeneous diffuse band between 80 and 180 kDa. The molecular mass of the same ChiA enzyme expressed in Pichia pastoris KM71H and Escherichia coli BL21 (DE3) was compared using SDS-PAGE with ChiA expressed in Saccharo-myces cerevisiae INVSc1. The activity of ChiA was determined using the fluorogenic substrate, 4-methylumbelliferyl ?-D-N,N,N-triacetylchitotrioside (4MUTC). Using a bioinformatics simulation, the number of the glycolsylation sites of the chiA gene sequence and the proximity of these sites to the alpha factor sequence were hypothesized to be a possible reason for which ChiA enzyme was internally expressed.

Abstract:
Abstract The proto-oncogene c-MYC encodes a bHLH leucine-zipper transcription factor that acts as a potent differentiation inhibitor and as a promoter of cell cycle progression. Additionally, c-MYC and mTOR are widely acknowledged as major regulators of cell growth, the process by which cells accumulate biomass. Both c-MYC overexpression and constitutive activation of the PI 3-kinase/mTOR pathway are frequent events in acute myeloid leukaemia; a haemopoietic malignancy characterised by failure to undergo terminal differentiation. In this study we used the MPRO system, an in vitro murine myeloid terminal differentiation model, to study the relationship between cell growth effectors and differentiation in more detail. Enforced expression of c-MYC by the addition of 4-hydoxytamoxifen (4-OHT) to MPRO cells carrying the inducible mycER fusion protein prevents downregulation of growth and impairs retinoid-induced terminal myeloid differentiation. Because downregulation of mTOR limits growth we hypothesised that inhibition of mTOR may reverse the phenotypic effects of c-MYC. Strikingly, pharmacological inhibition of mTOR by rapamycin, but not growth limitation by other means such as amino acid deprivation or cycloheximide treatment, restores differentiation in granulocytes with enforced c-MYC expression (74.0±2.0% mature granulocytes in the rapamycin+4-OHT-treated mycER MPROs vs 4.5±1.0% in the cells treated with 4-OHT alone and 92.9±1.1% in vehicle-treated controls). Rapamycin restores cell growth parameters to levels equivalent to those of vehicle-treated control cells undergoing physiological granulocytic differentiation (2–3 fold reduction in 35S methionine incorporation in rapamycin+4-OHT-treated and vehicle-treated controls compared to levels in 4-OHT-treated cells) and facilitates cell cycle exit (26.0±7.14% of rapamycin+4-OHT-treated cells and 10.7±2.48% of vehicle-treated control cells in S phase vs 51.9±1.72% of tamoxifen-treated cells in S phase as determined by BrdU incorporation). Interestingly, rapamycin attenuates c-MYC’s ability to activate transcription of its target genes without impairing activation of RARα target genes. c-mycER protein levels are reduced while levels of retrovirus-driven c-mycER mRNA do not fall in rapamycin-treated MPRO mycER cells. Furthermore, c-mycER protein levels decrease in rapamycin-treated MPRO cells expressing the ER fusion of the mycT58A point mutation known to increase protein stability. Overall, this data suggests that rapamycin restores the differentiation potential of granulocytes in the MPRO mycER system by regulating c-MYC at the level of protein translation, rather than at the level of gene transcription or protein degradation. These findings suggest that mTOR could be targeted to influence terminal differentiation regulated by MYC in haemopoietic malignancies.

Abstract:
Chemical synthesis of vitamin D2, an important nutrient for the human body, is dif ficult due to the asymmetric structure of the molecule of this vitamin. For this reason, the pro spects for the development of biotechnological approaches for the synthesis of ergosterol as a pre cursor of this vitamin are enormous, which is the basis of this work. The use of yeast Saccharo myces cerevisiae was proposed as a producer. These microorganisms are the most well studied, widely used in the food industry, are safe, easy enough to cultivate. YPD medium containing yeast extract, peptone and dextrose was used as nutrient medium. The study investigated the possibility of using ultrasound as a factor of intensification of the process of ergosterol biosynthesis by yeast cell. Ultrasound, on the one hand, was considered as a factor intensifying the dissolved oxygen content in the nutrient medium, on the other hand, as a factor of stress effect on the yeast cells themselves. Cultivation and evaluation of the content of the desired metabolite were carried out in comparison with the control nutrient medium. The physiological state of yeast was evaluated by microscopy and the content of ergosterol in yeast biomass at the beginning and end of cultivation in two types of nutrient media (YPD and YPD-US). The results obtained showed the viability of the proposed approach for culturing Saccharomyces cerevisiae yeast using ultrasound treatment. It was found that ultrasound treatment leads to the accumulation of dissolved oxygen, the optimal modes of exposure were established - power 122 W and time 2 min. The increment of ergosterol content in yeast biomass cultured in nutrient medium after ultrasound treatment amounted to 13.2 mg %, which is 1.21 times more than in yeast biomass cultured in control nutrient medium. At the same time, the understanding of the mechanism of ultrasonic stimulation of ergosterol synthesis by yeast remains to be studied.

Abstract:

Abstract:
The following paper proposes a reading of American poet Eileen Myles’ 2007 poetry collection Sorry, Tree in the interrelated contexts of Lauren Berlant’s understanding of intimacy and her concept of intimate citizenship, Lee Edelman’s understanding of sexuality and negativity, as well as Giorgio Agamben’s sense of the contemporary as always untimely. An openly lesbian and queer author, Myles gestures towards the sawed and the disempowered, olering an intimate, yet unmistakably political negotiation of a minoritarian lesbian position as both denant and transformative of the (hetero)normative status quo through acts of observation, engagement, and participation that do not necessarily have to be conspicuous or successful to elect reconceptualization of the social; rather, Myles suggests that individual agency in the public world also resides in failure as illuminative of the fact that one’s desire for presence is continuously actualized through entanglement with negativity.

Abstract:
Dion Boucicault’s three Irish plays: The Colleen Bawn (1860), Arrah-na-Pogue (1864) and The Shaughraun (1874), while not critically significant, owe their perennial popularity to their appeal to Irish romantic nationalism and to their memorable character types. While Boucicault’s character Myles Murphy or Myles na gCopaleen (Myles of the Ponies), an example of his native Irish hero, was the first of a series of rogue heroes that John Millington Synge developed in his character of Christy Mahon, Boucicault also owes the character of Myles to American native heroes like Sam Patch, Davy Crockett and Mose the Bowery B’hoy. While the plays are not great drama, they are good theatre and a less self-conscious national theatre has found room for both Boucicault and Synge.

Abstract:
Abstract Conditional mutants of the myeloblastic leukemic M1 cell line, expressing the chimeric mycer transgene, have been established. It is shown that M1 mycer cells, like M1, undergo terminal differentiation coupled to growth arrest and programmed cell death (apoptosis) after treatment with the physiologic differentiation inducer interleukin-6. However, when beta-estradiol is included in the culture medium, M1 mycer cells respond to differentiation inducers like M1 myc cell lines, where the differentiation program is blocked at an intermediate stage. By manipulating the function of the mycer transgene product, it is shown that there is a 10-hour window during myeloid differentiation, from 30 to 40 hours after the addition of the differentiation inducer, when the terminal differentiation program switches from being dependent on c-myc suppression to becoming c-myc suppression independent, where activation of c-myc has no apparent effect on mature macrophages. M1 mycer cell lines provide a powerful tool to increase our understanding of the role of c-myc in normal myelopoiesis and in leukemogenesis, also providing a strategy to clone c-myc target genes.
