Abstract:
Le début de la cryoconservation d’embryons de mammifères remonte aux années 70. Depuis, les techniques se sont multipliées et diversifiées pour essayer de répondre aux problèmes que posaient certaines espèces, certains stades de développement embryonnaire et les embryons fragilisés comme les embryons produits in vitro, issus de clonage ou encore les embryons biopsiés. Sont principalement utilisées, la congélation lente qui est basée sur l’équilibration progressive entre les cryoprotecteurs et le compartiment aqueux de l’embryon, la vitrification qui transforme rapidement la phase liquide du cytoplasme embryonnaire en phase solide amorphe appelée «état vitreux», et l’OPS (Open Pulled Straw) qui est une vitrification très rapide. Chaque technique a ses avantages et ses inconvénients, mais la congélation lente est actuellement la technique de référence sur le terrain en ovin, caprin et bovin avec des embryons produits in vivo. Par contre, pour les porcins, seule la technique d’OPS permet une bonne cryconservation des embryons. Dans l’espèce équine, aucune technique ne donne de très bons résultats : les poulains nés après transfert d’embryons cryoconservés sont encore rares.

Magnetic solid phase extraction has been used as pretreatment technique for the analysis of several compounds because of its advantages when it is compared with classic methods. This methodology is based on the use of magnetic solids as adsorbents for preconcentration of different analytes from complex matrices. Magnetic solid phase extraction minimizes the use of additional steps such as precipitation, centrifugation, and filtration which decreases the manipulation of the sample. In this review, we describe the main procedures used for synthesis, characterization, and application of this pretreatment technique which were applied in food analysis.

Molecularly imprinted techniques have been rapidly developed in recent decade. In this paper, novel mixed-template molecularly imprinted techniques are introduced. The development and applications of mixed-template molecularly imprinted techniques in recent decade are reviewed, including dual-template molecularly imprinted technique and multi-template molecularly imprinted technique, and its application in chromatographic solid phase, solid phase extraction and microsphere extraction. Moreover, the trends of mixed-template molecularly imprinted techniques in sample pretreatment are prospected.

Abstract:
Les travaux decrits dans ce memoire se rapportent a la synthese et a l'analyse structurale de molecules organiques accrochees sur resine de merrifield. Au cours des deux premieres parties, nous avons mis au point deux nouvelles syntheses traceless sur phase solide d'isoxazolidines et de 1h1,2,3-triazoles diversement substitues. Les deux voies d'acces reposent sur une strategie commune, dont l'etape-cle est une cycloaddition 1,3-dipolaire entre un dipolarophile (alcene ou alcyne) et, respectivement, une nitrone ou un azoture. Les cycloadduits sont construits en un nombre minimal d'etapes, au cours desquelles les differentes sources de diversite structurale sont introduites successivement. Le caractere traceless des deux syntheses est apporte par l'utilisation d'une resine de type rem, qui permet un decrochage fonctionnalisant de chacun des heterocycles. Enfin, dans un troisieme volet, nous avons developpe une nouvelle procedure de rmn du 1 3c en phase gel de resines fonctionnalisees. Grace a l'utilisation d'une sequence d'impulsions aux delais optimises, il est possible d'obtenir un spectre totalement exploitable en moins de vingt minutes. Ce temps d'experience particulierement court rend la methode compatible avec l'analyse de routine. La procedure ainsi mise au point peut etre employee en complement d'autres techniques analytiques telles que la microspectroscopie infrarouge.

Abstract:
A third of adults in the UK admit to using controlled substances at least once hi their lives and the most commonly abused drug was found to be cannabis, followed by the opiates and ecognine derivatives. Forensic laboratories are continually trying to reduce the turnaround time for samples submitted for analysis, to less than the current time of four days. One of the main aims of this work was to reduce the time taken to prepare, extract and analyse hair for target analytes. A new method combining supercritical fluid extraction and solid phase micro-extraction (SFE/SPME) is introduced. This coupled procedure utilises the advantages of using supercritical carbon dioxide as an extraction solvent and collecting the extract on a selected SPME fibre. Together the combined techniques provide a method that is solvent-free and highly selective, whilst reducing sample preparation to a minimum. Digested hair samples are examined by SEM to find the best method. Alkali hydrolysis and enzyme digestion using proteinase K were found to be the most effective pre-extraction methods. Solid hair samples with the addition of a derivatization reagent (BSTFA) gave the best results using the coupled technique. After optimisation and validation, the extraction from both real and spiked samples resulted in recoveries between 60% to 80% for the target analytes A9 -tetrahydrocannabinol (A9- THC) , cannabinol (CBN) and cannabidiol (CBD). Other forensic applications such as the extraction of polycyclic aromatic hydrocarbons and pesticides from environmental and food samples and the examination of fire debris for the presence of accelerants are also carried out. PAH could easily be extracted from complex matrices to give recoveries of up to 97% for some analytes at the optimized conditions of 90°C at 4500 psi over a 10 minute dynamic extraction period. Pesticide extraction gave recoveries of 60 - 80% while the extraction of accelerants from fire debris clearly showed that accelerants such as diesel can be identified over the coincidental PAH and hydrocarbon peaks found in burnt debris. The coupling of SFE to SPME provides an exciting technique that has a wide range of applications and creates potential for a fully automated on-line system.

Des nouvelles techniques chimiques et analytiques en phase solide ont ete developpees, permettant d'engendrer des chimiotheques d'une grande diversite, utilisables dans les tests de criblage a haut debit des societes pharmaceutiques. Plusieurs schemas reactionnels d'assemblage d'acides carboxyliques et de n-nucleophiles ont ete etudies et mis au point. Un nouveau reactif d'activation, derive d'hobt, immobilise sur polymere a ete synthetise, selon un procede applicable a tout type de support polymerique insoluble fonctionnalise avec des groupements -nh#2. Une grande efficacite de ce derive d'hobt pour l'assemblage d'amides a ete obtenue par incorporation d'un groupement electroattracteur sulfonamide sur le noyau benzenique du benzotriazole. Nous avons mis au point une strategie permettant la synthese d'amides en utilisant ce reactif de couplage, avec des bonnes conversions et puretes, en utilisant une tres large diversite de monomeres. L'utilisation d'acides carboxyliques et de n-nucleophiles a ete par la suite etudiee en vue de l'assemblage d'heterocycles. Un procede comprenant 4 etapes a ete mis au point pour la synthese en phase solide de benzimidazoles 1,2-disubstitues, et un criblage chimique des monomeres susceptibles a etre utilises a ete realise. Ce schema reactionnel permettrait d'aboutir a une chimiotheque de dimensions relativement importantes (pres de 10000 produits). Nous nous sommes interesses au techniques d'analyse non destructive des produits sans clivage de leur support de synthese, et nous avons contribue au developpement de la technique de resonance magnetique nucleaire sous l'angle magique, rmn mas. Cette technique a ete d'abord utilisee au suivi d'un schema reactionnel modele conduisant aux olefines selon la reaction de heck, et elle a ete par la suite appliquee a l'analyse des resines intermediaires intervenant dans la synthese en phase solide de benzimidazoles.

Abstract The perspectives and techniques developed in the previous chapters will now be applied to calculation of wave propagation in solids. Their application to longitudinal and shear waves will be both familiar and simple. Even more satisfying is the success of those same techniques in finding solutions for waves in a system that does not obey the wave equation and exhibit a phase speed that varies with frequency. Measurement of the frequencies of the normal modes of thin bars will be used to determine the bars’ elastic constants to high precision. The relationship between measured modal frequencies and the elastic moduli is particularly simple because the torsional, flexural, and longitudinal modes of bars can be selectively excited and detected. The technique of resonant ultrasound spectroscopy will allow the extraction of moduli from resonance frequencies even for samples with dimensions that are not as conducive as those of thin bars by a process that is significantly more computationally intensive. The flexural rigidity of wires under tension will be analyzed to determine the normal modes of a “stiff string,” and those effects will be discussed in relation to the tuning of pianos

Abstract:
In this research work, an electrical impedance spectroscopy technique has been developed to measure the solids concentration of slurry mixtures. This new measurement technique is based on the fact that the AC frequency responses of solids are distinct from those for the liquid phase. In this technique, the electrical impedance spectrum of slurry mixtures is measured over the frequency range of 0.1 Hz to 1 MHz. Changes in the frequency response at different excitation signal amplitudes reflect changes in the slurry composition and the electrical conductivity of the liquid phase, and by analyzing the spectra using artificial intelligence data analysis techniques, such as Multiplayer Perceptron Artificial Neural Network, one can determine the volumetric fraction of the solids phase. The technique has been successfully tested for various slurries under different conductivities of liquid phase and temperatures. In contrast with previous techniques based on conductivity or capacitance, this new technique can be used for on-line measurement of solids concentration in slurry streams even when the conductivity of the liquid phase changes substantially with time.

Abstract:
Liquid-solid phase transition is accompanied by a latent heat release, both in isothermal and non-isothermal phase transformations. The latent heat and the discontinuous phase change function increase the difficulty of obtaining a solution for the Fourier heat conduction equation. Celentano et al. [Int. J. Numer. Meth. Eng. 37(20), 1994] proposed a temperature-based finite element model for solving multidimensional transient heat conduction involving phase change. The present work addresses the computational aspects of the Celentano et al. model. The importance of a line search algorithm for improving the convergence of the Newton-Raphson iterations are explained in detail. While performing the iterations in this kind of fixed domain methods, the phase front moves back and forth fictitiously. The introduced phase change matrix handles the latent effect efficiently. The phase fractions are evaluated at the integration points instead of the nodal points. Several numerical examples are presented and the benefits and difficulties of the solution technique are elaborately discussed.
