Abstract:
Realizou-se a avaliação de pirogênios em produtos veterinários de uso parenteral, pelo método da hipertermia em coelhos, calculando-se, para o teste das amostras, doses com concentrações de três a sete vezes superiores à terapêutica. Preconizou-se como resposta positiva o aumento de temperatura de 0,6°C. Utilizou-se também o ensaio do lisado de amebócitos do Limulus (LAL) por geleificação, semiquantitativo, executando o teste de interferentes, validando o procedimento e estabelecendo a máxima diluição válida para a análise de cada produto. Paralelamente, efetuou-se avaliação comparativa de amostras com o método do LAL cromogênico, quantitativo, demonstrando correlação e reprodutibilidade dos resultados. Avaliaram-se vinte e oito produtos de diferentes classes farmacológicas, observando-se que dois não cumpriram as especificações, sendo reprovados. Sugere-se que as especificações estudadas sejam adotadas, contribuindo para aprimorar o controle de contaminantes, garantindo a qualidade e a segurança dos produtos veterinários.

Abstract:
Lactobacillus acidophilus used in three different applications, compounding pharmacies (LA1), fermented dairy (LA2), and allopathic compoundings (LA3) were tested to evaluate the existence of significant differences between them and in different growth conditions. In the evaluation of resistance to different commercial use antibiotics, all strains were sensitive to the antibiotics ampicillin, chloramphenicol, doxycycline, and tetracycline. LA1 was considered moderately sensitive (MS) to erythromycin and LA3 was MS to clindamycin and erythromycin. LA3 was classified between MS to resistant to erythromycin. All three strains were resistant to gentamicin. When evaluating acid pH resistance, the three origins presented similar behavior, with a decrease in cell viability at pH 2, maintaining constant viability at pH 3 and 4. In the test of resistance to the gastrointestinal tract conditions and hydrophobicity, LA2 presented better results. The three strains showed production of inhibitory compounds against pathogenic bacteria and deconjugated tauroconjugated bile salts (TDCA). It was concluded that, depending on the origin, Lactobacillus acidophilus may present different behaviors that will determine its growth and, consequently, its action in vivo. Due to the practicality of access, economy, and the satisfactory results in the tests performed, LA2 can be considered the strain of choice among those studied.

Um das principais cianobactérias de ocorrência local é o gênero Microcystis. As células de Microcystis têm tolerância a baixas salinidades, porém com o aumento abrupto da salinidade, as células podem sofrer lise. Muitas vezes, em função da hidrodinâmica local, essas florações de cianobactérias podem atingir a região da Praia do Cassino, possibilitando o contato com banhistas. Devido à natureza Gram-negativa da composição da parede celular das cianobactérias, esse contato com as células pode resultar em na ocorrência de casos de irritação epicutânea e reações alérgicas. O agente causador é o lipídeo A (endotoxina), encontrado no lipopolissacarídeo (LPS) da membrana externa das cianobactérias. Com o objetivo de detectar concentrações ambientais de LPS das florações de cianobactérias foi utilizado o método cromogênico cinético de ponto final do teste do Lisado do Amebócito de Limulus polyphemus (teste LAL). O teste LAL foi realizado em amostras de água de superfície coletadas nas regiões de São Lourenço do Sul, Praia do Laranjal (Pelotas), Museu Oceanográfico da FURG, Yacht Club do Rio Grande e Praia do Cassino (Rio Grande), entre os meses de dezembro de 2005 e março de 2006. Nestas amostras também foram estimados a abundância celular dos principais grupos do fitoplâncton, a concentração de clorofila-a, e medida a salinidade local no momento da coleta. Para avaliar a possível interferência dos sais nos resultados do teste LAL, também foram realizados cultivos da cianobactéria Microcystis crescendo em diferentes salinidades, onde também foram estimados a abundância celular, e as concentrações de clorofila-a, LPS e microcistinas. Os ajustes metodológicos realizados no teste LAL durante o trabalho resultaram em uma sensibilidade para sua aplicação nas amostras ambientais. As maiores concentrações de endotoxinas detectadas nas amostras ambientais (109,5 EU.mL-1, 71,8 EU.mL-1 e 93,7 EU.mL-1) se correlacionaram positivamente com as maiores abundâncias celulares (aproximadamente 600.000 células.mL-1, 400.000 células.mL-1 e 300.000 células.mL-1, respectivamente), todas com a presença da cianobactéria Microcystis.<br>Cyanobacterial blooms have been frequently observed in the waters of the Patos Lagoon estuary (RS). One of the main cyanobacterium of local occurrence is the genus Microcystis. The Microcystis cells present some tolerance to low salinity, but with the rapid salt increase the cells may suffer breakdown. Usually due to the local hydrodynamics these blooms may reach the recreational waters of the Cassino Beach, exposing swimming bathers to its contact. Due to the Gram-negative nature of the cyanobacterial cell membrane, this contact with the cells may result in case occurrences of epicutaneous irritation and allergic reactions. The causing agent is the lipid A (endotoxin), found on the lipopolysaccharide (LPS) of the external membrane of cyanobacteria. With the objective to detect environmental concentrations of LPS from cyanobacterial blooms, it was used the kinetic chromogenic endpoint method of the Limulus polyphemus Amebocyte Lisate test (LAL test). The LAL test was performed in surface water samples collected from the regions of São Lourenço do Sul, Laranjal Beach (Pelotas), FURG’s Oceanographic Museum, Yacht Club Rio Grande, and Cassino Beach (Rio Grande) betwen December 2005 and March 2006. In these samples it was also estimated the cellular abundance of the main phytoplanktonic groups, the chlorophyll-a concentration, and measured the local salinity. To evaluate the possible interference of salinity in the LAL test results, it was also cultivated the cyanobacterium Microcystis growing in different salinities. From these cultures it was estimate the cellular abundance, and chlorophyll-a, LPS and microcystin concentrations.. The methodological adjustments performed in the LAL test during this work resulted in a sensitivity for its application in environmental samples. The highest endotoxin concentrations detected in the environmental samples (109.5 EU.mL-1, 71.8 EU.mL-1 and 93.7 EU.mL-1) have positively correlated with the highest cell abundances (approximately 600,000 cells.mL-1, 400,000 cells.mL-1 and 300,000 cells.mL-1, respectively), all of them with the presence of the cyanobacteria Microcystis.

Abstract:
Este trabalho teve como objetivo geral a produção de um equipamento pequeno, multifuncional, de baixo custo e útil para as áreas de engenharia química e de química. Quanto à área de engenharia química, manipulam-se amostras em dispositivos miniaturizados que possuem operações unitárias comuns, como por exemplo, misturadores, formação de spray e remoção de partículas. Em química, efetua-se o pré-tratamento de amostras gasosas e possível detecção de compostos orgânicos voláteis (VOCs). A metodologia utilizada seguiu os protocolos para produção de equipamento. O equipamento projetado e construído apresenta três partes distintas e com as seguintes características. As amostras, que podem ser liquidas ou gasosas, são admitidas ou em fluxo contínuo ou de modo estacionário. A manipulação de dispositivos miniaturizados, como por exemplo, mini-misturadores, ocorre em três eixos distintos, o que permite o uso de dispositivos tridimensionais. Para todas as amostras a detecção é baseada em micro-balança de quartzo, com 5 cristais piezelétricos de quartzo (PQC) medindo simultaneamente e independentemente a aquisição de dados online. Quanto a amostras gasosas, é possível a detecção de VOCs na faixa de ppm e em dispositivo miniaturizado (mini manifold) para distribuição do fluxo uniformemente nos 5 PQCs. Através do uso do spray é possível a detecção de gases, líquidos e particulados. O componente sensor nos PQCs é filme fino depositado por plasma e a base de HMDS, hexametildissilazana. Esses filmes foram modificados por exposição à radiação ultravioleta (UVC) e feixe de íons (Ar+ ou He2+), os filmes após exposição mostraram-se mais eficientes na discriminação de VOCs. O equipamento foi construído com modos convencionais de produção, como por exemplo, usinagem com máquinas operatrizes como torno mecânico e fresadoras, e com partes e peças de fácil aquisição no mercado nacional.<br>This work aimed the production of a small equipment, multi-functional, lowcost and useful for chemical engineering and chemistry areas. Regarding the chemical engineering area samples are handled in miniaturized devices having common unit operations such as mixing, spray forming and removing particles. In chemistry, the pretreatment of gaseous samples and detection of volatile organic compounds (VOCs) could be made. The methodology followed the protocols for production equipment. The equipment designed and built features three distinct parts with the following characteristics. Samples can be liquid or gaseous, they can be admitted or continuous flow or stationary mode. The manipulation of miniaturized devices, such as minimixers occurs in three distinct axes, which allows the use of three-dimensional devices. For all samples the detection is based on quartz microbalance with 5 piezoelectric quartz crystals (PQC) the online data are acquired by measuring simultaneously and independently. As for gaseous samples, it is possible the VOCs detection in the ppm range and miniaturized device (mini manifold) to flow evenly distribution in 5 PQCs. Through the use of spray it is possible to detect gases, liquids and particulates. The sensing component in PQCs is thin film deposited by plasma and the base of HMDS hexamethyldisilazane. These films were modified by exposure to ultraviolet radiation (UVC) and ion beam (Ar+ or He2+) films after exposure were more efficient in the discrimination of VOCs. The equipment was built with conventional production methods, such as, machining with machine tools such as lathes and milling machines,, and parts and pieces of easy acquisition in the domestic market.

Entretanto, a agress?o est? comumente associada ? viol?ncia, que ? considerada um comportamento mal-adaptativo, como car?ter destrutivo das pessoas. Presente desde as etapas iniciais do desenvolvimento, torna-se relevante compreender o fen?meno ps?quico da agress?o na inf?ncia, como forma de conhecer e definir formas de interven??es adequadas na cl?nica psicol?gica infantil com o objetivo de a??es de preven??o. Para tanto, foram elaboradas duas se??es de estudo: uma te?rica e uma emp?rica. A se??o te?rica apresenta uma revis?o da contribui??o dos te?ricos da psican?lise para compreens?o da agressividade. Discute-se a import?ncia da agress?o para os diferentes autores e tamb?m se contextualiza como a agressividade est? presente nas fases do desenvolvimento infantil, a partir de um enfoque no per?odo da lat?ncia. Nesta etapa do desenvolvimento, que se caracteriza por um equil?brio entre impulso e defesas mais elaboradas, a utiliza??o de mecanismos defensivos, como a sublima??o, instrumentaliza a crian?a para superar situa??es dif?ceis de car?ter agressivo. Na se??o emp?rica, ? retratado um estudo quantitativo, do tipo transversal, que busca identificar a presen?a de evid?ncias de conte?dos agressivos nas respostas ao Teste dos Contos de Fadas (TCF), instrumento projetivo tem?tico. Participaram do estudo 72 crian?as dos sexos feminino e masculino, com idades entre 6 e 11 anos, de escolas p?blicas e privadas da cidade de Porto Alegre. A fim de obter dados que caracterizem os participantes, foi utilizada uma Ficha de Dados Sociodemogr?ficos. Para excluir casos com suspeita de comprometimento intelectual, foi administrado o Teste Matrizes Progressivas Coloridas de Raven - Escala Especial. Todo o material verbalizado pelas crian?as ao Teste dos Contos de Fadas (TCF) foi avaliado por tr?s ju?zes (J1, J2 e J3) que realizaram avalia??es com base no sistema de categoriza??o de respostas. Para avaliar o grau de concord?ncia entre eles, foi utilizada a estat?stica Kappa. Posteriormente, foi realizado um levantamento de freq??ncias dos tipos de conte?dos agressivos identificados nas respostas e, para a compara??o entre grupos e associa??es das vari?veis conforme escola, sexo e idade, tem?tica/conflito da s?rie de cart?es, foi utilizado o Teste Exato de Fisher. Os resultados mostram que o grau de concord?ncia entre os ju?zes, na sua maioria, foi satisfat?rio, uma vez que a concord?ncia alcan?ada foi, predominantemente, de valores moderados, substancias e quase perfeitos na maioria das categorias da agress?o. Os conte?dos agressivos com maior freq??ncia de respostas foram: Medo de Agress?o (f = 54), Agress?o Tipo A (f=49) e Agress?o como Retalia??o (f=34). Foi identificada associa??o significativa entre a vari?vel Agress?o Tipo A e sexo masculino (p=0,042). Foi poss?vel identificar as freq??ncias da vari?vel agress?o em seis das sete s?ries de cart?es (Chapeuzinho Vermelho, Lobo, An?es, Bruxa, Gigante, Cenas do Conto Chapeuzinho Vermelho). Abstract:
Creep properties for stress mitigation simulation in post weld heat treatment (PWHT) were studied. Materials tested in this study were both deposited metal and base metal of a nickel base alloy, an austenitic stainless steel, and a low alloy steel. First, creep tests were conducted at 625 °C, which is a holding temperature in PWHT of a low alloy steel. Test specimens were subjected to certain stress and the amount of creep strain was measured. A Norton-Bailey law model was used to simulate the amount of creep strain. Material parameters for the Norton-Bailey law model were determined from the creep strain measurement data. Stress relaxation tests were also conducted at 625 °C. Test specimens were subjected to a constant total strain and stress relaxation was measured. The experimentally measured stress relaxation data were calculated using the Norton-Bailey law model with the material parameters determined from the creep tests. The calculation data showed good agreement with the experiment data. Accordingly, the material parameters determined in this study can be considered valid.
