Radioisotopic dating can provide critical constraints for understanding the rates of tectonic, dynamic and biologic processes operating on our planet. Improving the interpretation and implementation of geochronologic data by coupling it with numerical modeling studies is the central theme of this thesis. Each chapter works to address a variety of problems in the Earth sciences. In each study, the interpretation of geochronologic data is aided with a numerical model that simulates the long-term behavior and/or initial conditions of the U-Pb system and provides an effective means of exploring the parameters that influence a calculated date. The record provided by geochronologic data is then coupled with models to quantitatively determine rates of geologic process on Earth. This approach permits geochronologic data to move beyond just establishing a relative time line of events. Using this dual modeling approach, Chapters 2-5 work to measure the long-term cooling and erosion rate of the lithosphere, specifically constraining the time scales and rate of transition between the stages of mountain belt formation and stability. Chapter six works to constrain the timing and duration of Central Atlantic Magmatic Province flood basalt volcanism and its relationship to the end Triassic mass extinction. The seventh and final chapter uses high precision U-Pb geochronology to evaluate the reliability of an Ar-Ar standard often used as a fluence monitor. The overall focus of this thesis has been to push the envelope of geochronologic precision and accuracy while coupling the data with modeling studies to yield new insight into Earth systems.

Full textThe world is producing 2.5 quintillion bytes daily, known as big data. Volume, value, variety, velocity, and veracity define the five characteristics of big data that represent a fundamental complexity for many machine learning algorithms, such as clustering, image recognition, and other modern learning techniques. With this large data, hyperparameter estimations do not take the form of the sample mean (not linear). Instead, they takethe form of average over m-tuples, known as the U-statistic estimator in probabilityand statistics. In this work, we treat the collection of U-statistics, known as the Uprocess, for two types of dependent variables, the Markovian data, and locally stationary random variables. Thus, we have divided our work into two parts to address each type independently. In the first part, we deal with Markovian data. The approach relies on regenerative methods, which essentially involve dividing the sample into independent and identically distributed (i.i.d.) blocks of data, where each block corresponds to the path segments between two visits of an atom called A, forming a renewal sequence. We derive the limiting theory for Harris recurrent Markov chain over uniformly bounded and unbounded classes of functions. We show that the results can be generalized also to the bootstrappe dU statistics. The bootstrap approach bypasses the problems faced with the asymptotic behavior due to the unknown parameters of limiting distribution. Furthermore, the bootstrap technique we use in this thesis is the renewal bootstrap, where the bootstrap samplevis formed by resampling the blocks. Since the non-bootstrapped blocks are independent, most proofs reduce to the i.i.d. case. The main difficulties are related to the randomsize of the resampled blocks, which creates a problem with random stopping times. This problem is degraded by replacing the random stopping time with their expectation. Also, since we resample from a random number of blocks, and the bootstrap equicontinuity can be verified by comparing with the initial process, the weak convergence of the bootstrap U-process must be treated very carefully. We successfully derive the results in the case of the k-Harris Markov chain. We extend all the above results to the case where the degreeof U-statistic grows with the sample size n, with the kernel varying in a class of functions. We provide the uniform limit theory for the renewal bootstrap for the infinite-degree U-process with the help of the decoupling technique combined with symmetrization techniques in addition to the chaining inequality. Remaining in the Markovian setting, we extend the weighted bootstrap empirical processes to a high-dimensional estimation. We consider an exchangeably weighted bootstrap of the general function-indexed empirical U-processes. In the second part of this thesis, dependent data are represented by locally stationary random variables. Propelled by the increasing representation of the data by functionalor curves time series and the non-stationary behavior of the latter, we are interested in the conditional U-process of locally stationary functional time series. More precisely, we investigate the weak convergence of the conditional U-processes in the locally stationary functional mixing data framework. We treat the weak convergence in both caseswhen the class of functions is bounded or unbounded, satisfying some moment conditions. Finally, we extend the asymptotic theory of conditional U-process to the locallystationary functional random field {Xs,An : s ∈ Rn} observed at irregular spaced locations in Rn = [0,An]d ∈ Rd, and include both pure increasing domain and mixed increasing domain. We treat the weak convergence in both cases when the class of functions is boundedor unbounded, satisfying some moment conditions. These results are established underfairly general structural conditions on the classes of functions and the underlying models

Full textCatchment hydrological functions of water collection, storage and release have geochemical signatures in stream water largely mirroring those found in critical zone compartments. These signatures are strongly controlled by the different bio-geo-physico-chemical processes that occur within the regolith-plant interface. Until now, investigations into the critical zone’s regolith and hydrological processes research have largely remained uncoupled –leading to a widespread use of non-conservative tracers with multiple origins and thereby stymieing our capability for identifying water pools and flow paths. Here we study the mixing of water in the subsurface through a unique portfolio of complementary groups of tracers (trace elements O-Hand Sr-Nd-Pb-U isotopes) which enables investigating regolith evolution processes and solutes transport within the critical zone. We report the interest of this approach to strengthen water flowpaths and end-members characterization

Full textThe kinematics and geodynamics associated with the compressional deformation in the Brooks Range and British-Barn Mountains, respectively in Alaska (USA) and North Yukon (Canada), in a back-arc setting has long been debated. In particular, the tectonic history of the Arctic Alaska continental block located in the foreland of the Brooks Range has been diversely interpreted in the plate reconstructions proposed for the Canadian basin. The Brooks Range mountain chain develops from the Lower Cretaceous to the Cenozoic. Constraining its exhumational history and its link with shortening evolution is essential in this arctic area where plate tectonics, associated to the Canadian basin opening in the North, has led to controversial geodynamic models. Tectonic coupling with the active Pacific subduction in the south as well as major seismogenic strike-slip faulting make this orogen a key area where to refine the regional plate reconstructions and understanding of orogenic processes. The Cenozoic evolution of the Brooks Mountains is poorly understood although it is a nearly unique example of arctic orogen, which have potentially recorded major climate changes like Paleocene-Eocene Thermal Maximum, the Oligocene cooling and Quaternary glaciations. This orogen is a key to study and quantify the climate impact on the north-alaskan topographic growth. This study combined low-temperature thermochronometry (FT, U-Th/He) on granitic and sedimentary rocks, which were thermo-kinematically modeled using Pecube in order to define the orogenic evolution of this arctic region since 100 Ma. In parallel, a field-based structural study (North Yukon and Brooks Range) was combined with the analysis of subsurface data. Thermochronological data, coupled to thermometric RSCM analyses and thermal modeling first define a slow exhumation period (0.2 km/Ma) from Upper Cretaceous up to the Eocene. With the Eocene, exhumation rates drastically increased to reach 1.25-1.29km/Ma as the deformation also migrates from SW toward NE. In both alaskan and canadian parts of mountains ranges a clear Oligocene exhumational event is identified. This event is linked to out-of-sequence crustal duplexing in the internal part of the range in the Brooks Range, contemporaneous with the propagation of deformation offshore, along British and Barn Mountains

Full textAt a crossroads of economical, sociological, cultural and sanitary issues, dietary analysis is of major importance for public health institutes. When international trade facilitates the transportation of foodstuffs produced in very different environmental conditions, when conspicuous consumption encourages profitable strategies (GMO, pesticides, etc.), it is necessary to quantify the sanitary risks engendered by such economic behaviors. We are interested in the evaluation of chronic types of exposure (at a yearly scale) to food contaminants, the long-term toxicity of which is already well documented. Because dietary risk and benefit is not limited to the abuse or the avoidance of toxic substances, nutritional intakes are also considered. Our work is thus organized along three main lines of research. We first consider the statistical analysis of very high long-term types of exposure to one or more chemical elements present in the food, adopting approaches in keeping with extreme value theory. Then, we adapt classical techniques borrowed from the statistical learning field concerning minimum volume set estimation in order to identify dietary habits that realize a compromise between toxicological risk and nutritional benefit. Finally, we study the asymptotic properties of a number of statistics that can assess the characteristics of the distribution of individual exposure, which take into account the possible survey scheme from which the data originate

Full textOn the one hand, we construct such tests based on bootstrap and permutation approaches. Their asymptotic performance are studied in a point process framework through the analysis of the asymptotic behavior of the conditional distributions of both bootstrapped and permuted test statistics, under the null hypothesis as well as under any alternative. A simulation study is performed verifying the usability of these tests in practice, and comparing them to existing classical methods in Neuroscience. We then focus on the permutation tests, well known for their non-asymptotic level properties. Their p-values, based on the delayed coincidence count, are implemented in a multiple testing procedure, called Permutation Unitary Events method, to detect the synchronization occurrences between two neurons. The practical validity of the method is verified on a simulation study before being applied on real data. On the other hand, the non-asymptotic performances of the permutation tests are studied in terms of uniform separation rates. A new aggregated procedure based on a wavelet thresholding method is developed in the density framework. Based on Talagrand's fundamental inequalities, we provide a new Bernstein-type concentration inequality for randomly permuted sums. In particular, it allows us to upper bound the uniform separation rate of the aggregated procedure over weak Besov spaces and deduce that this procedure seems to be optimal and adaptive in the minimax sens

Full textThe concept of Digital Fabrication Laboratories (Fablabs), initially an industrial prototyping platform for local entrepreneurships is rapidly finding its way into the education arena and used as a platform for learning and innovation. With the current impact of digital technology on the societies, there are calls from organizations like the National Research Council (NRC) for the education sector to cater for the so-called 21st Century Skills, while the industrial sector to rethink design and production in order to cater for sustainability (inclusive of eco-design and circular economy) and to embrace the latest technologies in preparation for the Industries 4.0. Fablab platforms established in universities (Ub-Fablabs) could therefore play a vital role as a ‘support platform’ to meet these demands.Since there has been very little or no research into the capacities of Ub-Fablabs to meet these demands, this research took a two-fold approach toi) investigate and align cognitive, affective and psychomotor skills (inclusive of mechanical, electrical and embedded software operational skills) embedded in the design process in Ub-Fablabs.ii) assess the capacity of Ub-Fablabs to prepare citizens for the future design and production industries. In part 1 of the research, the researcher closely observed the design and production of a rock milling machine (RMM) where utilisation of a range of wood, stone and synthetic materials technologies were displayed. The design and production activities were recorded using field notes, video-recording and still photography. To keep trace of the design and production activities, an iterative design process model, the ‘Nawita Design Process Model (NDPM) was developed and utilised. Data was analysed using an adapted protocol analysis and results were graphed using pie and bubble-chart graphing. Results showed that materialising the creative ideas incubated in stage 1 of the NDPM unleashed a stunning peak of cognitive, affective and psychomotor skills in stages 2, 3 and 4 of NDPM. These processes and skills are undoubtedly constituted of the 21st Century Skills. In part 2 of the research, an adapted online content analysis was used to collect data from 90% of the Ub-Fablabs worldwide. An Ub-Fablab Capacity Indicator Scale (Ub-Fablab CIS) was developed and used to score on four potential aspects of Ub-Fablab if it has to be used as a support platform to incubate proactive minds for the future integration of design and production industries. These components are i) Technological infrastructure; ii) Constructionist pedagogical approach; iii) Collaboration through digital networking and iv) sustainability (inclusive of eco-design and circular economy). Results revealed that all Ub-Fablabs have a strong stand in providing technological infrastructures and a strong constructionist pedagogical approach, but may need to improve on its capacity for collaboration through networking and catering for sustainability. The findings of this research provides a strong standing for Ub-Fablabs to be utilised to augment efforts to equip citizens with 21st Century skills and incubate proactive minds for the future integration of design and production industries

The objective of this study is to examine, in light of the expression of multiple p53 family member isoforms, the specific role of p73 in malignant conversion, cellular response to DNA damage, and direct or indirect cooperation with other p53 family members in a clonal model of epidermal carcinogenesis. We first focused on the role of p73 in malignant conversion. Whether sporadic or siRNA induced, loss of p73 in initiated p53+/+ keratinocytes lead to conversion to squamous cell carcinoma (SCC) in vivo which was reversible upon reconstitution of TAp73α but not ΔNp73α. Second, we investigated the cellular response to ionizing radiation (IR) in the presence and absence of p73, showing that loss of p73 at malignant conversion was associated with resistance to IR in vitro. The loss of radiation sensitivity and malignant conversion was characterized by reduced steady state DNA binding levels of transcriptionally active p63 isoforms to the p21 promoter, failure to induce specific p53 family transcriptional targets, and failure to arrest in G1. Reconstitution of TAp73α, but not ΔNp73α, increased steady state DNA binding capabilities of TAp63β, TAp63γ, and ΔNp63γ, and steady state levels of p53 family target mRNA, but did not restore cellular sensitivity to IR. We thus uncovered a functional cooperation between TA isoforms of p73 and p63 and showed that p73-mediated DNA damage response was uncoupled from its tumor suppressive role. We observed preferential DNA binding of the inhibitory ΔNp63α isoform both in vitro and invivo in SCC suggesting that in the absence of TAp73α a balance is tipped toward DNA binding of the inhibitory isoforms. Third, we studied the role of the p53 family inkeratinocyte response to UVB. Tumorigenic cells lacking p73 that were resistant to IR remained sensitive to UVB, accompanied by DNA binding of the TAp63γ isoform, suggesting that keratinocyte response to UVB is not dependent upon p73 and suggesting a hierarchy of p53 family member responses to DNA damage. Finally, we examined TAp73α interaction with the p53 family inhibitor Mdm2. Mdm2 was in complex with DNA-bound p53 family members in malignant cells, but reconstitution of cells withTAp73α correlated with removal of Mdm2 from the complex, making them more like primary keratinocytes or initiated cells. Like the initiated cells, cells expressing TAp73α were refractory to treatment with the Mdm2-p53 inhibitor Nutlin-3 while cells lacking p73 expression or expressing ΔNp73α were sensitive. Thus, we suggest that p73 may be acting as a molecular shield to keep p53 family member inhibitors, such as ΔNp63α andMdm2, at bay. Further understanding of p53 family interplay in tumor development and DNA damage response could lead to new therapies or optimization of current therapeutic strategies in solid tumors of epithelium, particularly where deregulation or loss of p63 and p73 expression is associated with increased tumor invasiveness, treatment resistance, and poor patient prognosis.
