Ovarian cancer is a deadly disease attributed to late-stage detection as well as recurrence and the development of chemoresistance. Ovarian cancer stem cells (OCSCs) are hypothesized to be largely responsible for the emergence of chemoresistant tumors. Although chemotherapy may initially succeed at decreasing the size and number of tumors, it leaves behind residual malignant OCSCs. In this study, we demonstrate that aldehyde dehydrogenase 1A1 (ALDH1A1) is essential for the survival of OCSCs. We identified a first-in-class ALDH1A1 inhibitor, compound 974, and used 974 as a tool to decipher the mechanism of stemness regulation by ALDH1A1. The treatment of OCSCs with 974 significantly inhibited ALDH activity, the expression of stemness genes, and spheroid and colony formation. An in vivo limiting dilution assay demonstrated that 974 significantly inhibited CSC frequency. A transcriptomic sequencing of cells treated with 974 revealed a significant downregulation of genes related to stemness and chemoresistance as well as senescence and the senescence-associated secretory phenotype (SASP). We confirmed that 974 inhibited the senescence and stemness induced by platinum-based chemotherapy in functional assays. Overall, these data establish that ALDH1A1 is essential for OCSC survival and that ALDH1A1 inhibition suppresses chemotherapy-induced senescence and stemness. Targeting ALDH1A1 using small-molecule inhibitors in combination with chemotherapy therefore presents a promising strategy to prevent ovarian cancer recurrence and has the potential for clinical translation.

Abstract Background A chimerically configured gracilis and profunda artery perforator (PAP) flap is highly prevalent based on recent computed tomography (CT)-imaging data. The purpose of this study is to further characterize the vascular anatomy of this novel flap configuration and determine the feasibility of flap dissection. Methods To characterize flap arterial anatomy, lower extremity CT angiograms performed from 2011 to 2018 were retrospectively reviewed. To characterize venous anatomy and determine the feasibility of flap harvest, the lower extremities of cadavers were evaluated. Results A total of 974 lower extremity CT angiograms and 32 cadavers were included for the assessment. Of the 974 CT angiograms, majority (966, 99%) were bilateral studies, yielding a total of 1,940 lower extremities (right-lower-extremity = 970 and left-lower-extremity = 970) for radiographic evaluation. On CT angiography, a chimerically configured gracilis and PAP flap was found in 51% of patients (n = 494/974). By laterality, chimeric anatomy was present in 26% of right lower extremities (n = 254/970) and 25% of left lower extremities (n = 240/970); bilateral chimeric anatomy was found in 12% (n = 112/966) of patients. Average length of the common arterial pedicle feeding both gracilis and PAP flap perforasomes was 31.1 ± 16.5 mm (range = 2.0–95.0 mm) with an average diameter of 2.8 ± 0.7 mm (range = 1.3–8.8 mm).A total of 15 cadavers exhibited chimeric anatomy with intact, conjoined arteries and veins allowing for anatomical tracing from the profunda femoris to the distal branches within the tissues of the medial thigh. Dissection and isolation of the common pedicle and distal vessels was feasible with minimal disruption of adjacent tissues. Chimeric flap venous anatomy was favorable, with vena commitante adjacent to the common pedicle in all specimens. Conclusion Dissection of a chimeric medial thigh flap consisting of both gracilis and PAP flap tissues is feasible in a cadaveric model. The vascular anatomy of this potential flap appears suitable for future utilization in a clinical setting.

Abstract Introduction: An attractive, yet unrealized, goal in cancer therapy is repurposing psychiatric drugs that can readily penetrate the blood-brain barrier for the treatment of deadly primary brain tumors and brain metastasis. The phenothiazine (PTZ) class of anti-psychotic agents were originally developed to inhibit dopamine- and serotonin-receptors (DR/5-HTRs). PTZs have demonstrated anti-cancer properties in numerous studies through a variety of proposed mechanisms, including on-target (DR/5-HTR) and off-target (e.g. calmodulin (CaM) inhibition) effects. PTZs inhibit DR/5-HTRs in the low- to mid-nanomolar range whereas anti-cancer effects tend to be observed only at a higher (low micromolar) dose range. Thus, clinical efficacy of existing PTZs may be limited by toxicity of on-target effects (sedation) of these drugs as DR/5-HTR antagonists. However, it has been unclear whether anti-cancer effects of PTZs are entirely separate from their activity as DR/5-HTR antagonists. Here we evaluate the in vitro anti-cancer efficacy of a novel PTZ analog (CWHM-974, a N-benzyl-linked fluphenazine (FLU) derivative), originally developed to treat brain-resident fungal infections, that is a 100-1000-fold less potent DR/5-HTR antagonist than FLU. Experimental procedures: To evaluate the anti-cancer efficacy of FLU and CWHM-974, we assessed cytotoxicity following 72h exposure against a broad panel of cancer cell lines. To investigate CaM as a target for CWHM-974, we evaluated the correlation between the sensitivity to CMW-974 and sensitivity to the CaM inhibitor W-7 as well as CaM gene expression levels in a panel of cancer cell lines. We also tested the anti-cancer efficacy of a sulfoxide analog of CWHM-974 (SLU-0010894) which lacks CaM binding activity. Results: We confirmed the anti-cancer efficacy of FLU and demonstrated that CWHM-974 is consistently 2-10-fold more potent than FLU in a broad panel of cancer cell lines. Among the most sensitve cancer cell lines are melanoma, which is a cancer that frequently metastasizes to the brain. Consistent with the hypothesis that CaM is a relevant target for CWHM-974, we found that: 1) CaM mRNA (CALM1) levels are correlated with sensitivity to CWHM-974; 2) the activities of CWHM-974 and W7 are correlated, and 3) most importantly, SLU-0010894 does not exhibit anti-cancer activity. Conclusions: The anti-cancer efficacy of the PTZ derivative CWHM-974 is seperable from DR/5-HTR antagonism. Our data suggests that further optimization of this compound may preserve the desirable pharmacologic properties for targeting brain cancers while reducing toxicity due to DR/5-HTR antagonism. Moreover, we provide a variety of evidence suggesting that anti-cancer efficacy of CWHM-974 is associated with CaM binding. Wnt (a portmanteau of Wingless and Int-1) signaling in the adult heart is largely quiescent. However, there is accumulating evidence that it gets reactivated during the healing process after myocardial infarction (MI). We here tested the therapeutic potential of the Wnt secretion inhibitor LGK-974 on MI healing. Ischemia/reperfusion (I/R) injury was induced in mice and Wnt signaling was inhibited by oral administration of the porcupine inhibitor LGK-974. The transcriptome was analyzed from infarcted tissue by using RNA sequencing analysis. The inflammatory response after I/R was evaluated by flow cytometry. Heart function was assessed by echocardiography and fibrosis by Masson’s trichrome staining. Transcriptome and gene set enrichment analysis revealed a modulation of the inflammatory response upon administration of the Wnt secretion inhibitor LGK-974 following I/R. In addition, LGK-974-treated animals showed an attenuated inflammatory response and improved heart function. In an in vitro model of hypoxic cardiomyocyte and monocyte/macrophage interaction, LGK974 inhibited the activation of Wnt signaling in monocytes/macrophages and reduced their pro-inflammatory phenotype. We here show that Wnt signaling affects inflammatory processes after MI. The Wnt secretion inhibitor LGK-974 appears to be a promising compound for future immunomodulatory approaches to improve cardiac remodeling after MI.

Abstract High grade serous ovarian cancer (HGSOC) is the leading cause of death from gynecologic malignancies. Ovarian cancer stem cells (OCSCs) are hypothesized to contribute to the emergence of chemoresistant tumors. Aldehyde dehydrogenase 1A1 (ALDH1A1) is an intracellular enzyme that oxidizes toxic aldehydes to carboxylic acids and plays a role in controlling cell differentiation pathways. We have previously used ALDH1A1 activity as a functional marker to identify OCSCs and demonstrated that OCSCs persist after chemotherapy and drive recurrent, drug resistant disease. However, the mechanism by which ALDH1A1 maintains stemness phenotype remains poorly understood. To examine the effect of ALDH1A1 on cellular survival signals in OCSCs, we generated a novel ALDH1A1-specific small molecule inhibitor named compound 974. Treatment of HGSOC cell lines with compound 974 reduced ALDH enzyme activity (p<0.01) and inhibited stem-like properties including spheroid formation (p<0.01) and clonogenic survival (p<0.05). Compound 974 treatment inhibited (p<0.01) stemness genes Prom1, Bmi1, Oct4 and Nanog. ALDH1A1 inhibition also reduced (p<0.05) cisplatin IC50 and synergized (p<0.05) with cisplatin treatment. To further examine the effect of compound 974 to inhibit ALDH1A1 and consequently tumor initiation, mice were injected with 106, 105 and 104 OVCAR3 cells treated in vitro with compound 974 (5µM for 48h). In a parallel study, mice were injected with 106, 105 and 104 of ALDH1A1 knockdown or scrambled control cells. In both cases, tumor initiation was significantly delayed compared to the control, and extreme limiting dilution analysis further revealed that compound 974 reduced (p<0.05) CSC frequency compared to control. To better understand the mechanism by which ALDH1A1 maintains OC stemness, OVCAR3 cells treated with vehicle or compound 974 were analyzed by RNA-sequencing and bioinformatics. Transcriptomic analysis revealed that compound 974 significantly (FDR < 0.05, fold change > 2) downregulated expression of markers strongly associated with CSC phenotypes (NF-κB, CD44, Klf4, Sox9, Fzd7) and chemoresistance (ABCB1). Ingenuity Pathway Analysis for downstream regulators of differentially expressed genes revealed that the senescence pathway was significantly altered. Analysis by qRT-PCR revealed that compound 974 inhibited (p<0.01) chemotherapy-induced expression of senescence marker p21 and senescence associated secretory phenotype (SASP) genes including IL6, IL8, CXCL1 and CXCL3. Compound 974 also inhibited(p<0.05) the chemotherapy-induced beta-galactosidase activity. Collectively, our findings further support the functional role for ALDH1A1 in OCSC and reveal a novel role for the enzyme in chemotherapy-induced senescence and disease relapse. The secreted Wnt signaling molecules are essential to the coordination of cell-fate decision making in multicellular organisms. In adult animals, the secreted Wnt proteins are critical for tissue regeneration and frequently contribute to cancer. Small molecules that disable the Wnt acyltransferase Porcupine (Porcn) are candidate anticancer agents in clinical testing. Here we have systematically assessed the effects of the Porcn inhibitor (WNT-974) on the regeneration of several tissue types to identify potentially unwanted chemical effects that could limit the therapeutic utility of such agents. An unanticipated observation from these studies is proregenerative responses in heart muscle induced by systemic chemical suppression of Wnt signaling. Using in vitro cultures of several cell types found in the heart, we delineate the Wnt signaling apparatus supporting an antiregenerative transcriptional program that includes a subunit of the nonfibrillar collagen VI. Similar to observations seen in animals exposed to WNT-974, deletion of the collagen VI subunit, COL6A1, has been shown to decrease aberrant remodeling and fibrosis in infarcted heart tissue. We demonstrate that WNT-974 can improve the recovery of heart function after left anterior descending coronary artery ligation by mitigating adverse remodeling of infarcted tissue. Injured heart tissue exposed to WNT-974 exhibits decreased scarring and reduced Col6 production. Our findings support the development of Porcn inhibitors as antifibrotic agents that could be exploited to promote heart repair following injury.

In this study, simultaneous second-harmonic generation (SHG), sum frequency generation (SFG), and Raman conversion based on MgO-doped periodically poled lithium niobate (MgO:PPLN) for multi-wavelength generation is demonstrated. The approach used is based on a single MgO:PPLN crystal poled with a uniform period of 10.2 µm that phase matches SHG and SFG, simultaneously. Using a simplified double-pass geometry, up to 0.8 W of blue light at 487 nm is achieved by a frequency-doubling 974 nm laser diode pump, and 0.5 W of orange light at 598 nm is generated by frequency mixing 974 nm pump with C-band (1527–1565 nm) tunable laser source. At high pump powers of the 974 nm laser source, other unexpected peaks at 437, 536, 756, 815 and 1038 nm were observed, of which the 1038 nm line is due to Stimulated Raman Scattering within the MgO:PPLN crystal. The resulting multi-wavelength light source may find a wide range of applications in biomedicine and basic research.

Introduction: Preeclampsia remains one of the most common causes of maternal and perinatal mortality worldwide. A significant role in the pathogenesis of this pathology is assigned to placental ischemia and endothelial dysfunction. Therefore, the aim of the present study was to study the effectiveness of asialized erythropoietin and arginase II selective inhibitors: KUD-259 and KUD-974 in the correction of morphofunctional disorders of the cardiovascular system in experimental preeclampsia. Materials and methods: The study was performed in 260 female Wistar rats, each weighing 250–300 g. An ADMA-like preeclampsia was reproduced in the experiment. To assess the emerging morphofunctional disorders, the following parameters were used: blood pressure, coefficient of endothelial dysfunction, microcirculation in the placenta, proteinuria, fluid content in the omentum, concentration of terminal metabolites in the blood plasma, and morphometric parameters of fetuses. Results and discussion: The administration of arginase II selective inhibitor KUD-974 in combination with asialized erythropoietin leads to a pronounced correction of emerging changes: a decrease in systolic and diastolic blood pressure in 1.5 and 1.7 times, a decrease in proteinuria in 3.6 times and a decrease in fluid content in the omentum. When arginase II selective inhibitor KUD 974 and asialized erythropoietin are used with methyldopa, the positive effects of the former are enhanced. Conclusion: Arginase II selective inhibitors KUD-259 and KUD-974 and asialized erythropoietin have a pronounced positive effect on the correction of morphofunctional disorders in animals with ADMA-like preeclampsia.

La Escala de Autoeficacia General se ha empleado para evaluar diversas poblaciones alrededor del mundo, puesto que, esta variable es importante para desarrollar comportamientos sociales alternativos a los psicopatológicos. El objetivo: fue obtener las evidencias psicométricas de la Escala de Autoeficacia General. Metodología: 1100 universitarios fueron evaluados con la Escala de Autoeficacia General de Baessler y Schwarzer modificada por Anicama y Cirilo (2012). Resultados: Se halló una distribución normal y coeficientes de correlación ítem-test corregidas mayores a .30. Se identificaron índices de ajuste adecuados para un modelo unidimensional con covarianza de ítems (x2/gl=4.465; GFI=.974; CFI=.973; TLI=.964; RMSEA=.056; SRMR=.0273; AIC=191.329). Asimismo, se hallaron correlaciones inversas con el Cuestionario de Ansiedad Social para adultos (CASO) de Caballo et al. y con la Escala de Estrés ante enfermedades transmisibles APA de Anicama et al. El Alfa de Cronbach y el Omega de McDonald’s tuvieron valores mayores a 0.70. Conclusiones: Este estudio reafirma la validez y fiabilidad psicométrica de la Escala de Autoeficacia General en una muestra de universitarios de Lima.

AbstractThe Egmond Gospels, a much cherished manuscript in the Netherlands, have provoked a wealth of publications, descriptions, commentaries, catalogue entries etc. Most noted are the two dedication miniatures representing count Thierry of Holland (d. 988) and his wife Hildegard, offering the Evangeliary to the abbey of Egmond. The MS is now in the Koninklijke Bibliotheek, The Hague, cod. 76 F I. In the present article an attempt has been made to describe the colour programme of these two miniatures more accurately with the help of a stereomicroscope and the Munsell Color Atlas. Some suggestions have been made about the pigments which were probably used. A new date for the miniatures has been proposed, taking into consideration their historical context and their iconography. The most likely period is from 974 to 980. In this period double portraits are being introduced in the West, in the Ottonian world. An incentive for this new style was undoubtedly the arrival of the Byzantine princess Theophano who married Otto II in 972. In the Byzantine world imperial double portraits were very common. After the death of her father-in-law Otto I, in 973 and the official recognition of her co-rulership in 974, her role became more prominent, and may have stimulated the making of Western double portraits, if not as propaganda then at least to imitate Byzantine court life. Egbert, son of Thierry II and Hildegard, became chancellor of the Reich in 976 and archbishop of Trier in 977. He played an important role in Ottonian art as a patron and promoter, in which Byzantine influence is clearly discernible. Iconographical elements in the Egmond Gospels, such as the proskynesis, the disproportion between donors and 'authorities', the symmetry, the double portrait etc. betray influence from Byzantine iconography. Byzantine influence, indirectly via the Ottonians, was thus an active force in the execution of these miniatures, wherein Egbert is likely to have played an active role, even if the artist and the workshop are unknown.

Abstract During meiosis, axial elements are generated by the condensation of sister chromatids along a protein core as precursors to the formation of the synaptonemal complex (X). Functional axial elements are essential for wild-type levels of recombination and proper reductional segregation at meiosis I. Genetic and cytological data suggest that three meiosis-specific genes, HOP1, RED1 and MEK1, are involved in axial element formation in the yeast Saccharomyces cerevisiae. HOP1 and RED1 encode structural components of axial elements while MEK1 encodes a putative protein kinase. Using a partially functional allele of MEK1, new genetic interactions have been found between HOP1, RED1 and MEK1. Overexpression of HOP1 partially suppresses the spore inviability and recombination defects of mekl-974; in contrast, overexpression of RED1 exacerbates the mek1-974 spore inviability. Co-overexpression of HOP1 and RED1 in mek1-974 diploids alleviates the negative effect of overexpressing RED1 alone. Red1p/Red1p as well as Hop1p/Red1p interactions have been reconstituted in two hybrid experiments. Our results suggest a model whereby Mekl kinase activity controls axial element assembly by regulating the affinity with which Hoplp and Redlp interact with each other.

Abstract Background: Transcription factors of the Forkhead box O (Fox O) family have important roles in cellular proliferation, apoptosis, differentiation, and stress resistance. In pancreatic β-cells, FoxO1 protein plays an important role in β-cells development. The molecular mechanism of transcriptional regulation of basal FoxO1 gene expression in pancreatic β-cells is not fully understood. Objectives: Explore the potential transcription factors regulating FoxO1 promoter activity using pancreatic β-cell line (RINm5F cells) Methods: Promoter screening method, luciferase reporter gene analysis, transient expression assay system, and deletion analysis of a -974/-18 bp 5’ upstream region of the mouse FoxO1 gene were used in this study. Results: An inhibition domain (-974/-321) and an activation domain (-321/-18) was identified through deletion analysis of a -974/-18 bp 5’ upstream region of the mouse FoxO1 gene. Using the promoter screening method, several transcription factors were selected. Luciferase reporter studies showed that these factors could regulate FoxO1 promoter activity in RINm5F cells. Among these factors, cAMP response-element binding protein (CREB) could positively regulate FoxO1 promoter activity. Signal transducer and activator of transcription 1 (STAT1) played a negative role on FoxO1 promoter. In addition, ETS oncogene family member Elk-1 did not affect the FoxO1 promoter activity. Conclusion: Two transcription factors (CREB and STAT1) could effectively regulate the mouse FoxO1 gene promoter activity.

Chickpea (Cicer arientinum L.) is one of the most dominant pulse crops in India, which contributes 38 percent of the area and 50 percent production of pulses compare to the total pulse production of India. Chickpea contains protein-2.1%, carbohydrates-61.5%, and fat-4.5% and more iron, calcium and niacin content. The main constrain of chickpea production due to parasitic nematodes (Meloidogyne incognita) is about 14% of total global production in annual yield loss. Pseudomonas fluorescens is a bacterial bio-agent that can help in nematode suppression in chickpea plants. This experiment was conducted to experience the differences, if any, in manganese content concerning chickpea inoculated with M. incognita with a combination of Pseudomonas fluorescens as a bioagent, where different treatments of nematode, bacteria, and chemicals are used sustaining the enhancement of disease resistance in chickpea cultivars RSG 974, GG 5, GNG 2144. The total manganese content of chickpea variety GNG 2144 was found highest in treatment, where only bacteria (P. fluorescens) was inoculated, i.e., 6.44 mg/100g of a root, followed by GG 5, i.e., 5.63 mg/100g of root and RSG 974 was, i.e., 4.14 mg/100g of root respectively. Application of Pseudomonas fluorescence combined or alone gradually increased the manganese concentration in roots of chickpea plants i.e., RSG 974 (4.14 mg/100g), GG 5(5.63 mg/100g), GNG 2144 (6.44 mg/100g) compared to the health check.

Abstract To improve our understanding of Feline calicivirus (FCV) infection in cats in Northeast China, 1584 serum samples from 974 domestic cats and 610 stray cats were collected between 2012 and 2015. The samples were tested for FCV antibodies using a commercially available ELISA kit. The results revealed an overall seroprevalence of 37.56% (595/1584), a seroprevalence in domestic cats of 32.85% (320/974) and a seroprevalence in stray cats of 45.08% (275/610). Risk factor analysis indicated that species was the only risk factor for the presence of FCV (OR=1.678, 95% CI=1.362-2.066, P<0.001); age, season, region and gender were not risk factors. This is the first report of FCV infection in stray cats in China, and the results of this study can aid in FCV infection control in the felidae family.

Objective: Floating bioadhesive tablets of hydrochlorothiazide were developed to prolong gastric residence time leading to an increase in drugbioavailability where here a combination of floating and bioadhesion mechanism is combined.Methods: Tablets are prepared by direct compression technique using polymers xanthan gum, carbopol 974 P, HPMC K15M, HPMC K100M, magnesiumStearate USP-NF (Avicel PH 102), microcrystalline cellulose Ph 102, Talc, and sodium bicarbonate.Results: Tablets were evaluated for their physical characteristics, namely, hardness, thickness, friability and weight variation, drug content, andfloating properties. The best formulation subjected for kinetic treatment, i.e., zero order, first order, peppas, Higuchi, and Hixon-crowel. The R valuesare 0.9366, 0.9364, 0.9680, 0.9974, and 0.9283, respectively.Conclusion: Optimized formulae F4 containing polymers HPMC K4M and CARBOPOL 974 P showed more bioadhesion with a controlled release over12 hrs. Therefore, formulation F4 identified as a successful formulation for the development of floating bioadhesive tablets.Keywords: Hydrochlorothiazide, Floating tablets, Gastroretentive drug delivery system.
