In this work, Oxford Nanopore sequencing is tested as an accessible method for quantifying heterogeneity of amplified DNA. This method enables rapid quantification of deletions, insertions, and substitutions, the probability of each mutation error, and their locations in the replicated sequences. Amplification techniques tested were conventional polymerase chain reaction (PCR) with varying levels of polymerase fidelity (OneTaq, Phusion, and Q5) as well as rolling circle amplification (RCA) with Phi29 polymerase. Plasmid amplification using bacteria was also assessed. By analyzing the distribut

In some nonlinear unsteady inverse problems, the inverse solution will oscillate violently in the whole time domain due to the sharp change of the sensitivity coefficients. To deal with this problem, a new sequential function specification method with variable future time steps is proposed in this paper. The future time steps are adjusted by the error amplification coefficients which are defined as the reciprocal of the square sum of the sensitivity coefficients. Resumen
Numerical weather forecasting errors grow with time. Error growth results from the amplification of small perturbations due to atmospheric instability or from model deficiencies during model integration. In current NWP systems, the dimension of the forecast error covariance matrices is far too large for these matrices to be represented explicitly. They must be approximated. This paper focuses on comparing the growth of forecast error from covariances modeled by the Met Office operational four-dimensional variational data assimilation (4DVAR) and ensemble transform Kalman filter (ETKF) methods

Resumen
Abstract The emerging field of precision oncology utilizes next-generation sequencing (NGS) to guide decisions based on individual genetic signatures. As demand for these applications grows, minimizing errors and biases introduced during library preparation and sequencing is critical. Advances in sequencing chemistries have improved base quality scores, shifting the focus to library preparation to improve data quality. PCR amplification is a key potential source of error during NGS library preparation; single-base substitutions, insertions, and deletions (indels) introduced by PCR in addition

Resumen
Thermus thermophilusMutS, a thermostable mismatch-recognizing protein, is utilized in PCR to suppress nonspecific amplification by preventing synthesis from mismatched primers.T. thermophilusRecA also decreases nonspecific amplification by promoting proper hybridization between the primer and template. We observed that MutS and RecA function under the same reaction conditions and that MutS and RecA do not preclude each other. Furthermore, there were some DNA sequences for which only one of the 2 proteins effectively suppressed nonspecific amplification. The simultaneous use of MutS and RecA is

Resumen
ABSTRACT Plasmid F′128 was formed by an exchange between chromosomal Rep sequences that placed lac near dinB between many pairs of Rep sequences. Plasmid F′128 is critical for selection-enhanced lac reversion (adaptive mutation), which requires prior lac amplification. The structure of F′128 supports the idea that amplification is initiated by Rep-Rep recombination and that general mutagenesis requires coamplification of dinB (error-prone polymerase) with lac.

Resumen
A system-level implementation of a parallel-path hybrid switched-capacitor amplifier is presented in this paper. The proposed parallel-path amplifier incorporates a gain and slew rate-boosting switching path in parallel with an embedded assisted SAR path, aiming for IoT applications. As an alternative concept to the conventional analog topologies, the proposed amplifier combines nonlinear and linear paths to provide coarse and fine amplifications. In the coarse amplification, a high current is provided through a switching path for a fraction of time, which improves the slew rate and open-loop
