Resumen

Acute lung injury (ALI) is one of the most prevalent respiratory syndromes of excessive inflammatory reaction during lung infection. Candida albicans ( C. albicans) infection is among the leading causes of ALI. MicroRNAs (miRNAs) regulate the expression of target mRNAs, including those involved in inflammatory processes, by binding to the 3′UTR. To date, the roles of miRNAs in C. albicans-induced ALI remain unclear. In this study, we investigated the role of miR-384-5p in C. albicans-induced ALI and its underlying molecular mechanism. RT-PCR, Western blot, ELISA, Myeloperoxidase (MPO) assay, microRNA target analysis, transient transfection, and luciferase reporter assay were utilized. In vivo study was conducted on mouse model. The expression of miR-384-5p was upregulated and positively correlated with inflammatory cytokine production in lung tissues and RAW264.7 and J774A.1 macrophages infected with C. albicans. The miR-384-5p inhibitor alleviated the inflammatory reaction induced by C. albicans. Target prediction analysis revealed that PGC1β was a target of miR-384-5p, which was further validated by the PGC1β 3′-UTR luciferase assay and the inverse correlation between the expression of miR-384-5p and PGC1β in C. albicans-infected ALI tissues and macrophages. Moreover, macrophages transfected with miR-384-5p mimic exhibited reduced levels of PGC1β. The suppression of the expression of PGC1β by C. albicans infection in the macrophages was abrogated by miR-384-5p inhibitor. Then, we demonstrated that PGC1β played an inhibitory role in C. albicans-induced production of inflammatory cytokines. Furthermore, suppression of miR-384-5p in macrophages inhibited the activation of the NF-κB, MAPK, and Akt signaling pathways triggered by C. albicans, but not the STAT3 pathway. These results demonstrate that miR-384-5p contributes to C. albicans-induced ALI at least in part by targeting PGC1β and enhancing the activation of the NF-κB, MAPK, and Akt inflammatory signaling pathways. Thus, targeting miR-384-5p might exert a protective effect on C. albicans-induced ALI.

AbstractChronic hepatitis C virus (HCV) infection has been associated with development of mixed cryoglobulinemia type 2 (MC2), a lymphoproliferative disorder characterized by B cell monoclonal expansion and immunoglobulin M/k cryoprecipitable immunoglobulin production. A short sequence (codons 384-410) of the HCV E2 protein, which has the potential to promote B cell proliferation, was investigated in 21 patients with HCV-related MC2 and in a control group of 20 HCV carriers without MC2. In 6 of the 21 (29%) patients with MC2, all the clones isolated from plasma, peripheral blood mononuclear cells, and liver showed sequence length variation compared with the hypervariable region 1 (HVR1) consensus sequence; 5 patients had an insertion at codon 385, and 1 patient had a deletion at codon 384. Inserted residues at position 385 were different within and between patients. No such mutations were observed in any of the HVR1 clones from control patients without MC2, and the difference between the 2 groups was statistically significant (P = .02). Analysis of 1345 HVR1 sequences obtained from GenBank strongly supported the conclusion that the observed insertions and deletion represent a rare event in HCV-infected patients, suggesting that they are significantly associated with MC2. The physical and chemical profiles of the 385 inserted residues detected in the MC2 patients were consistent with the possibility that these mutations, which occurred in a region containing immunodominant epitopes for neutralizing antibodies and binding sites for B lymphocytes, may be selected by functional constraints for interaction with host cells.

Aim. The purpose of this study is to determine the association between eotaxin 426 C/T, −384 A/G, 67 G/A, eNOS −786 T/C, 4 a/b, and MMP-13 rs640198 G/T and prognosis of patients with known CAD.Methods. From total of 1161 patients referred to coronary angiography, 532 patients with angiographically confirmed CAD were selected. Their long-term outcome was followed up using hospital database. Subsequent events were assessed in this study: death or combined endpoint-myocardial infarction, unstable angina pectoris, revascularization, heart failure hospitalization, and cardioverter-defibrillator implantation.Results. The multivariate Cox regression model identified age, smoking, and 3-vessel disease as significant predictors of all-cause death. Further analysis showed that eotaxin 67 G/A (GA + AA versus GG) and eotaxin −384 A/G (GG versus GA + AA) were significant independent prognostic factors when added into the model: HR (95% CI) 2.81 (1.35–5.85),p=0.006; HR (95% CI) 2.63 (1.19–5.83),p=0.017; eotaxin −384 A/G was significantly associated with the event-free survival, but it did not provide the prognostic information above the effect of two- or three-vessel disease.Conclusion. The A allele in eotaxin 67 G/A polymorphism is associated with worse survival in CAD patients.

Resumen

Babesia gradually attains resistance against the allopathic medicines due to their frequent use. To overcome the resistance, herbal therapy is getting more attention. The current study was planned to monitor the efficacy of Peganum harmala against babesiosis in cattle. The blood was collected from ear vein of cattle having high infestation of ticks. A thin blood smear was prepared and stained. Through survey 74 animals were found positive for tick-borne pathogens with an overall prevalence of 19.27%. Babesia sp. was the most prevalent (9.38%; 36/384) haemoparasite, followed in order by Theileria sp. (5.20%, 20/384) and Anaplasma sp. (4.69%, 18/384). All blood samples were subjected to PCR and it was found that 40/384 (10.41%) blood samples were harbouring Babesia bovis and Babesia bigemina. From positive animals for Babesia 30 cattles were randomly selected and divided into 3 groups of 10 animals each viz A, B and C, all groups were tested with the water extract of P. harmala @ 7.5 mg kg-1, 10 mg kg-1 and 12.5 mg kg-1 body weight by intramuscular route, at interval of 12, 24 and 36 h. Ten healthy animals were selected as control in group D. Three animals with moderate infection, 8-9 animals of group A, B and C were recovered, during 48 h after treatment. Two animals each from A, B and one from group C were not cured and died. Five animals from group A were not cured till 36 h of the treatment. Group D of healthy untreated animals were well as they were. Results showed that there was a significant increase in WBCs, including increase in leukocytes and neutrophils in animals infected, whereas number of RBCs and HB decreases. This study concluded that infected cattle treated with water extract of P. harmala 12.5 mg kg-1, recovered early (P<0.01) compared to that of 7.5 and 10 mg kg-1.

La Commission de réforme du droit du Canada (CRD) a publié, en 1982, un document de travail intitulé Euthanasie, aide au suicide et interruption de traitement. En 2009, la députée Francine Lalonde a déposé à la Chambre des communes le projet de loi C-384 intitulé Loi modifiant le Code criminel (droit de mourir dignement). Ce projet de loi a été défait. Une période de près de trente ans sépare ces deux documents. Quelles sont les préoccupations que partagent ces documents ? Le projet de loi envisage-t-il l’aide au suicide d’une manière qui diffère de celle du document de la CRD ? L’examen du document de la CRD, du projet de loi C-384 et des travaux de la Chambre des communes fait ressortir des difficultés persistantes dans nos discussions publiques. Certaines suggestions s’imposent à propos du débat souhaitable dans une matière aussi contentieuse que l’euthanasie et l’aide au suicide.

Resumen

While c-MYC is well established as a proto-oncogene, its structure and function as a transcription factor have made c-MYC a difficult therapeutic target. To identify small-molecule inhibitors targeting c-MYC for anticancer therapy, we designed a high-throughput screening (HTS) strategy utilizing cellular assays. The novel approach for the HTS was based on the detection of cellular c-MYC protein, with active molecules defined as those that specifically decreased c-MYC protein levels in cancer cells. The assay was based on a dual antibody detection system using Förster/fluorescence resonance energy transfer (FRET) and was utilized to detect endogenous c-MYC protein in the MYC amplified cancer cell lines DMS273 and Colo320 HSR. The assays were miniaturized to 1536-well plate format and utilized to screen the GlaxoSmithKline small-molecule collection of approximately 2 million compounds. In addition to the HTS assay, follow-up assays were developed and used to triage and qualify compounds. Two cellular assays used to eliminate false-positive compounds from the initially selected HTS hits were (1) a cellular toxicity assay and (2) an unstable protein reporter assay. Three positive selection assays were subsequently used to qualify compounds: (1) 384-well cell cycle flow cytometry, (2) 384-well cell growth, and (3) c-MYC gene signature reverse transcription quantitative PCR (RT-qPCR). The HTS and follow-up assays successfully identified three compounds that specifically decreased c-MYC protein levels in cancer cells and phenocopied c-MYC siRNA in terms of cell growth inhibition and gene signatures. The HTS, triage, and three compounds identified are described.

Resumen

The diversity of Colletotrichum lindemuthianum is a major limiting factor in control of anthracnose on bean (Phaseolus vulgaris), and race characterization of this pathogen is an important tool in breeding programs. Race characterization has been carried out on isolates from North, Central, and South America; Europe; and Asia, but little or no information exists on the diversity of C. lindemuthianum in Africa. In this work, 12 isolates from the major bean-growing areas of Burundi, Central Africa, were characterized. Their virulence was tested on 12 bean differential cultivars (1) and on 4 bean cultivars commonly grown in Burundi: 2 from local germ plasm (Muyinga-1 and Urubonobono) and 2 from Colombia (A 321 and Calima). Detached unifoliate bean leaves from 8-day-old plants were placed on a humid surface in trays and sprayed until runoff with a suspension of 106 spores ml-1. Trays covered with transparent plastic sheets to keep a minimum relative humidity of 92% were incubated at 20°C. Seven days after inoculation, symptoms were evaluated for severity on a scale of 1 to 9. Leaves scored as 1 to 3 were considered resistant. Races were characterized according to a numerical binary system (1). Nine races were identified: 9, 69, 87, 384, 385, 401, 448, 449, and 485. Seven of these races (9, 69, 87, 384, 401, 448, and 485) were described for the first time in Africa. Races 401 and 485 have not yet been reported in the literature. The most susceptible differential cultivars were Michelite, PI 207262, To, and Mexico 222. Muyinga-1, Urubonobono, and A 321 were sensitive to nine, six, and five races, respectively. There is a high diversity of C. lindemuthianum in Burundi, and the local germ plasm tested is very susceptible to the characterized races. Breeding programs in Burundi should focus on lines and cultivars, such as Tu, AB 136, G 2333, and Calima, that are resistant to all the races characterized in this study. Reference: (1) M. A. Abstract Background: Additional instruments have become available since instruments for DNA melting analysis of PCR products for genotyping and mutation scanning were compared. We assessed the performance of these new instruments for genotyping and scanning for mutations. Methods: A 110-bp fragment of the β-globin gene including the sickle cell anemia locus (HBB c. 20A>T) was amplified by PCR in the presence of LCGreen Plus or SYBR Green I. Amplicons of 4 different genotypes [wild-type, homozygous, and heterozygous HBB c. 20A>T and double-heterozygote HBB c. (9C>T; 20A>T)] were melted on 7 different instruments [Applied Biosystems 7300, Corbett Life Sciences Rotor-Gene 6500HRM, Eppendorf Mastercycler RealPlex4S, Idaho Technology LightScanner (384 well), Roche LightCycler 480 (96 and 384 well) and Stratagene Mx3005p] at a rate of 0.61 °C/s or when this was not possible, at 0.50 °C steps. We evaluated the ability of each instrument to genotype by melting temperature (Tm) and to scan for heterozygotes by curve shape. Results: The ability of most instruments to accurately genotype single-base changes by amplicon melting was limited by spatial temperature variation across the plate (SD of Tm = 0.020 to 0.264 °C). Other variables such as data density, signal-to-noise ratio, and melting rate also affected heterozygote scanning. Conclusions: Different instruments vary widely in their ability to genotype homozygous variants and scan for heterozygotes by whole amplicon melting analysis. Instruments specifically designed for high-resolution melting, however, displayed the least variation, suggesting better genotyping accuracy and scanning sensitivity and specificity.

ABSTRACT The structure of the nucleocapsid protein of bunyaviruses has not been defined. Earlier we have shown that Tula hantavirus N protein oligomerization is dependent on the C-terminal domains. Of them, the helix-loop-helix motif was found to be an essential structure. Computer modeling predicted that oligomerization occurs via helix protrusions, and the shared hydrophobic space formed by amino acids residues 380-IILLF-384 in the first helix and 413-LI-414 in the second helix is responsible for stabilizing the interaction. The model was validated by two approaches. First, analysis of the oligomerization capacity of the N protein mutants performed with the mammalian two-hybrid system showed that both preservation of the helix structure and formation of the shared hydrophobic space are crucial for the interaction. Second, oligomerization was shown to be a prerequisite for the granular pattern of transiently expressed N protein in transfected cells. N protein trimerization was supported by three-dimensional reconstruction of the N protein by electron microscopy after negative staining. Finally, we discuss how N protein trimerization could occur.

Resumen

The genus Chionanthus (Oleaceae Hoffmans. & Link) includes deciduous or evergreen trees and shrubs distributed widely in tropical and sub-tropical areas, including a few temperate species. Although Chionanthus species are planted as ornamental garden plants and commercialized for natural products, genetic information for Chionanthus spp. is lacking. We created microsatellite-enriched libraries of Chionanthus retusus Lindl. & Paxton, assembled 1072 contigs, and detected 1010 repeats. The frequency of the repeats decreased with the increase in repeat length, and the most abundant motifs were: AG, AC, AAG, ACC, AT, and ACTC. We screened 384 markers on 12 accessions of four related taxa that included C. retusus, Chionanthus virginicus L., Chionanthus pygmaeus Small, and Osmanthus americanus (L.) Benth. & Hook. A total of 195 simple sequence repeat (SSR) markers amplified and discriminated six accessions of C. retusus and 57 SSR markers amplified and discriminated across the four Oleaceae species screened. To identify the best markers to use in future experiments, the “Unique Pattern Informative Combination” (UPIC) values were calculated for all the markers and the 100 markers that were most effective are reported here. The percentage of heterozygous loci across the 384 markers was lowest for C. retusus (29.3%) and highest for O. americanus (68.9%). The SSR markers developed here could assist in taxonomy and hybridization investigations for breeding programs and authentication of varieties used as medicinal plants.

The vacuolar H+-ATPase (V-ATPase) in intercalated cells contributes to luminal acidification in the kidney collecting duct and nonvolatile acid excretion. We previously showed that the A subunit in the cytoplasmic V1 sector of the V-ATPase (ATP6V1A) is phosphorylated by the metabolic sensor AMP-activated protein kinase (AMPK) in vitro and in kidney cells. Here, we demonstrate that treatment of rabbit isolated, perfused collecting ducts with the AMPK activator 5-aminoimidazole-4-carboxamide-1-β-d-ribofuranoside (AICAR) inhibited V-ATPase-dependent H+ secretion from intercalated cells after an acid load. We have identified by mass spectrometry that Ser-384 is a major AMPK phosphorylation site in the V-ATPase A subunit, a result confirmed by comparing AMPK-dependent phosphate labeling of wild-type A-subunit (WT-A) with that of a Ser-384-to-Ala A subunit mutant (S384A-A) in vitro and in intact HEK-293 cells. Compared with WT-A-expressing HEK-293 cells, S384A-A-expressing cells exhibited greater steady-state acidification of HCO3−-containing media. Moreover, AICAR treatment of clone C rabbit intercalated cells expressing the WT-A subunit reduced V-ATPase-dependent extracellular acidification, an effect that was blocked in cells expressing the phosphorylation-deficient S384A-A mutant. Finally, expression of the S384A-A mutant prevented cytoplasmic redistribution of the V-ATPase by AICAR in clone C cells. In summary, direct phosphorylation of the A subunit at Ser-384 by AMPK represents a novel regulatory mechanism of the V-ATPase in kidney intercalated cells. Regulation of the V-ATPase by AMPK may couple V-ATPase activity to cellular metabolic status with potential relevance to ischemic injury in the kidney and other tissues.

Resumen

The rotational symmetry, cohesive energies, and thermal stabilities of the toroidal forms, as well as helically coiled forms derived from C 240, C 360, and C 540, are studied by molecular dynamics. For various rotational symmetric helically coiled forms, the dependence of the polygon patterns in the inner ridge line on the coil length is studied. For the short-coil-length limit, the surface consists of pentagons, hexagons, and heptagons. For C 240 and C 360 helices the surface patterns are changed with increasing coil length. By stretching the helically coils along the fiber axis, we found that the surface of C 384 helix, eightfold rotational symmetric form derived from torus C 240, consisting of pentagons and heptagons without heptagons becomes stable as the coil length becomes longer.

Abstract The crystal structure of (CH3)3 NHBF 4 was studied in three solid phases by X-ray diffraction techniques. The structures of the ionic plastic phase (Phase I) stable above 453 K and Phase II between 384 and 453 K are CsCl-type cubic (a = 5.772(4) Å) and tetragonal (a = 9.815(5) and c = 6.895(5) Å), respectively. The room temperature phase (Phase III) forms a monoclinic lattice (space group P21/m, a = 5.7017(8), b = 8.5724(9), c = 7.444(1) Å, and ß = 102.76(1)°). BF4− ions in this phase were shown to be disordered in four orientations.

Tujuan penelitian ini untuk mendapatkan determinan penggunaan kontrasepsi pada wanita usia subur di Provinsi Jawa Tengah. Penelitian ini merupakan penelitian kuantitatif dengan rancangan cross sectional study (potong lintang). Data yang digunakan raw data SDKI tahun 2017 provinsi Jawa Tengah terbaru, meliputi jumlah anak hidup (MV201), umur istri (MV013), pendidikan suami (SM 108), pendidikan istri (MV 106), status pekerjaan istri (MV 717), tempat tinggal (MV 025), indeks kekayaan (MV 190), akses sumber informasi KB melalui radio (MV 384 A), akses sumber informasi KB melalui televisi (MV 384 B), akses sumber informasi KB melalui majalah (MV 384 C), penggunaan kontrasepsi (MV 313). Pengolahan data dilakukan dengan menggunakan program SPSS 22. Setelah itu dianalisis, meliputi karakteristik masing- masing variabel penelitian dengan distribusi frekuensi yang ditampilkan dalam persentase masing- masing kelompok data. Selanjutnya analisis bivariat untuk melihat hubungan antara variabel independen dengan variabel dependen dengan menggunakan Chi Square. Hasilnya ada beberapa variabel yang merupakan determinan penggunaan kontrasepsi/memiliki hubungan dalam penggunaan kontrasepsi, variabel tersebut antara lain jumlah anak yang dilahirkan, pendidikan istri, pekerjaan istri, tempat tinggal, mendapatkan informasi KB melalui media televisi dan membaca informasi KB pada majalah dengan penggunaan kontrasepsi. Sementara variabel yang tidak memiliki hubungan dengan penggunaan kontrasepsi antara lain umur istri, pendidikan suami, indek kekayaan, mendengarkan informasi KB dari radio dengan penggunaan kontrasepsi. Rekomendasinya sosialisasi tentang program KB secara menyeluruh dengan bahasa menarik dan mengajak terutama pada media radio. sosialisasi juga tidak terbatas pada kaum perempuan, agar laki-laki semakin memahami akan program KB dan berpartisipasi dalam pengambilan keputusan Kata Kunci : Determinan, Penggunaan Kontrasepsi, Wanita Usia Subur

Intracellular recordings, ATP and cytosolic calcium measurements from CA1 pyramidal cells in rat hippocampal slices were used to examine the mechanisms by which temperature alters hypoxic damage. Hypothermia (34°C) preserved ATP (1.7 vs. 0.8 nM/mg) and improved electrophysiologic recovery of the CA1 neurons after hypoxia; 58% of the neurons subjected to 10 min of hypoxia (34°C) recovered their resting and action potentials, while none of the neurons at 37°C recovered. Increasing the glucose concentration from 4 to 6 mM during normothermic hypoxia improved ATP (1.3 vs. 0.8 nM/mg) and mimicked the effects of hypothermia; 67% of the neurons recovered their resting and action potentials. Hypothermia attenuated the membrane potential changes and the increase in intracellular Ca2+(212 vs. 384 nM) induced by hypoxia. Changing the glucose concentration in the artificial cerebrospinal fluid primarily affects ATP levels during hypoxia. Decreasing the glucose concentration from 4 to 2 mM during hypothermic hypoxia worsened ATP, cytosolic Ca2+, and electrophysiologic recovery. Ten percent of the neurons subjected to 4 min of hypoxia at 40°C recovered their resting and action potentials; this compared with 60% of the neurons subjected to 4 min of normothermic hypoxia. None of the neurons subjected to 10 min of hypoxia at 40°C recovered their resting and action potentials. Hyperthermia (40°C) worsens the electrophysiologic changes and induced a greater increase in intracellular Ca2+(538 vs. 384 nM) during hypoxia. Increasing the glucose concentration from 4 to 8 mM during 10 min of hyperthermic hypoxia improved ATP (1.4 vs. 0.6 nM/mg), Ca2+(267 vs. 538 nM), and electrophysiologic recovery (90 vs. 0%). Our results indicate that the changes in electrophysiologic recovery with temperature are primarily due to changes in ATP and that the changes in depolarization and Ca2+are secondary to these ATP changes. Both primary and secondary changes are important for explaining the improved electrophysiologic recovery with hypothermia.

Resumen

Campylobacter jejuni and Campylobacter coli are globally recognized as a major cause of bacterial foodborne gastroenteritis. A cross-sectional study was conducted from October 2015 to May 2016 in Mekelle city to isolate, identify, and estimate the prevalence of C. jejuni and C. coli in raw meat samples and to determine their antibiotic susceptibility pattern. A total of 384 raw meat samples were randomly collected from bovine (n = 210), goat (n = 108), and chicken (n = 66), and isolation and identification of Campylobacter spp. were performed using standard bacteriological techniques and PCR. Antibiotic susceptibility test was performed using disc diffusion method. Of the total 384 raw meat samples, 64 (16.67%) were found positive for Campylobacter spp. The highest prevalence of Campylobacter spp. was found in chicken meat (43.93%) followed by bovine meat (11.90%) and goat meat (9.25%). The most prevalent Campylobacter spp. isolated from meat samples was C. jejuni (81.25%). The overall prevalence of Campylobacter in restaurants, butcher shops, and abattoir was 43.93%, 18.30%, and 9.30%, respectively. 96.8%, 81.25%, 75%, and 71% of the Campylobacter spp. isolates were sensitive to norfloxacin, erythromycin, chloramphenicol, and sulphamethoxazole-trimethoprim, respectively. However, 96.9%, 85.9%, and 50% of the isolates were resistant to ampicillin, amoxicillin, and streptomycin, respectively. Strains that developed multi-drug resistant were 68.7%. The result of this study revealed the occurrence of Campylobacter in bovine, goat, and chicken meats. Hence, there is a chance of acquiring infection via consumption of raw or undercooked meat. Thus, implementation of hygienic practices from a slaughterhouse to the retailers, proper handling and cooking of foods of meat are very important in preventing Campylobacter infection.

ISSN 0804–4643 Steroid 11β-hydroxylase deficiency (11β-OHD) is an autosomal recessive hereditary defect and one of the causes of congenital adrenal hyperplasia. The 11β-hydroxylase enzyme is encoded by the CYP11B1 gene on chromosome 8q22. Twelve types of mutations of this gene have been reported previously in patients with 11β-OHD, including one Japanese patient. We detected an additional previously uncharacterized mutation: R384G mutation, a single C→G base substitution in the codon 384 of the exon 7 changing an arginine (CGA) to a glycine (GGA) by sequencing the CYP1 1B1 gene of Japanese siblings with 11β-OHD. We analyzed also the CYP11B1 gene of their unaffected family members. Their grandfather was homozygous for the normal gene, whereas their grandmother, parents and younger brother were heterozygous for the mutation. These results indicate that the cause of 11β-OHD is heterogeneous even in the Japanese population, which is ethnically homogeneous. Many diseases in humans are caused by mutations that decrease the stability of specific proteins or increase their susceptibility to aggregation. Consequently, the availability of high-throughput methods for assessing protein stability and aggregation properties under physiological conditions (e.g., 37 °C) is necessary to analyze physicochemical properties under conditions that are closer to in vivo models. Therefore, the authors have explored the use of isothermal denaturation (ITD) in a 384-well format to evaluate the reproducibility of the method in assessing the stability of proteins at temperatures below the melting temperature and detecting the binding of ligands. Under the conditions tested, the authors were able to assess the stability of citrate synthase and malate dehydrogenase at different constant temperatures and detect the binding of oxaloacetate and nicotinamide adenine dinucleotide to these 2 enzymes, respectively, using the 384-well format. The ITD experiments detected ligand binding to these proteins at about 4 times lower concentration compared with techniques that measure changes in melting temperature. The data show that ITD can be applied to screen libraries of a relatively large number of compounds or detect small stability differences between protein variants. The contact resistances investigated in this study of anisotropic conductive adhesive film joints using Au/Ni bumps and flexible substrates are found to be increased by the drop impact energy and also by the combined effect of heat/humidity and the impact energy. The samples humidified at 85 °C/85% RH for 384 h, on which impact energy of 50 J was induced, exhibit the most severe results. The contact resistance increases by 700%, which had been about 0.062 Ω in the as-bonded condition. The samples without humidification showed a sluggish and gentle increase in contact resistance with induced drop impact energy. The contact resistance was found to be increased by 400% after absorbing 90 J energy. Scanning electron microscopy images show particle deformation due to abrasion and friction between the contacting surfaces resulting from the sudden impact. Joints are also observed with no connections, which signify open circuits. Almost 25% of circuits were found open in the samples (after 384 h in a humid environment), which have suffered severe mechanical shock. Breaking of the conductive layer of the particle and exposing the underlying polymeric portion was also observed.

We compared the ability of two closely related truncated E2 glycoproteins (E2660) derived from hepatitis C virus (HCV) genotype 1a strains Glasgow (Gla) and H77c to bind a panel of conformation-dependent monoclonal antibodies (MAbs) and CD81. In contrast to H77c, Gla E2660 formed disulfide-linked high molecular mass aggregates and failed to react with conformation-dependent MAbs and CD81. To delineate amino acid (aa) regions associated with protein aggregation and CD81 binding, several Gla–H77c E2660 chimeric glycoproteins were constructed. Chimeras C1, C2 and C6, carrying aa 525–660 of Gla E2660, produced disulfide-linked aggregates and failed to bind CD81 and conformation-dependent MAbs, suggesting that amino acids within this region are responsible for protein misfolding. The presence of Gla hypervariable region 1 (aa 384–406) on H77 E2660, chimera C4, had no effect on protein folding or CD81 binding. Chimeras C3 and C5, carrying aa 384–524 or 407–524 of Gla E2660, respectively, were recognized by conformation-dependent MAbs and yet failed to bind CD81, suggesting that amino acids in region 407–524 are important in modulating CD81 interaction without affecting antigen folding. Comparison of Gla and H77c E2660 aa sequences with those of genotype 1a and divergent genotypes identified a number of variant amino acids, including two putative N-linked glycosylation sites at positions 476 and 532. However, introduction of G476N–G478S and/or D532N in Gla E2660 had no effect on antigenicity or aggregation.

St Jerome's third Latin translation of the Sefer Tehillim or ‘Book of Psalms’ is called the iuxta Hebraeos or Hebraicum, because he based it on the original Hebrew in which it was composed in order to obtain the greatest authenticity possible. Preceded by the so-called Romanum version of c. 384, which was primarily a translation of the Greek Septuagint, and the Gallicanum of c.392 which was a revision of it based on Origen's hexaplaric Septuagint text, the Hebraicum version of c. 400 represents an attempt by Jerome to produce a Latin translation as close as possible to the Hebrew text. However, despite its greater accuracy with respect to the Hebrew original, the Hebraicum was apparently never used in the liturgy, and was preserved solely as a patristic text in bibles or psalters for scholarly use.

ABSTRACT The hepatitis C virus glycoprotein E2 receptor-binding domain is encompassed by amino acids 384 to 661 (E2661) and contains two hypervariable sequences, HVR1 and HVR2. E2 sequence comparisons revealed a third variable region, located between residues 570 and 580, that varies widely between genotypes, designated here as igVR, the intergenotypic variable region. A secreted E2661 glycoprotein with simultaneous deletions of the three variable sequences retained its ability to bind CD81 and conformation-dependent monoclonal antibodies (MAbs) and displayed enhanced binding to a neutralizing MAb directed to E2 immunogenic domain B. Our data provide insights into the E2 structure by suggesting that the three variable regions reside outside a conserved E2 core.

El objetivo de este trabajo fue identificar maíces (Zea mays L.) con estabilidad en el rendimiento de grano y potencial para la industria de la masa y la tortilla, mediante la evaluación de la interacción genotipo ambiente de ocho genotipos cultivados en ocho ambientes del subtrópico de México. Durante los ciclos PV-2015 y 2016 se evaluaron maíces con valor agregado: tres de alto contenido de aceite (H-327 AO, H-388 O, H-389 O), uno de alta calidad proteínica (H-390 C), un amarillo normal (H-387 A) y tres testigos comerciales (H-377, H-383 C, DK2027Y). Mediante el modelo de regresión de sitios se identificó al híbrido H-389 O como uno de los materiales de mayor rendimiento (12.07 t ha-1) y más estable, con buena calidad de grano y producción de tortillas suaves, tanto frescas como almacenadas. En 2015 una mayor precipitación incrementó la acumulación de almidón, la densidad y la dureza del grano. El genotipo H-387 A sobresalió por su alto contenido de carotenoides (12.11 μg g-1 maíz) respecto al testigo amarillo (2.72 μg g-1), lo que le confiere propiedades nutracéuticas. El hibrido H-390 C no destacó por su contenido de lisina y triptófano. Aunque poco estables en rendimiento, el resto de los genotipos presentaron características de grano aptas para el procesamiento y rendimiento aceptable de tortillas (1.5 kg kg-1 de maíz).

There exist relatively few drug classes on the market to treat dermatophyte infections. This investigation was designed to develop and validate high-throughput methodology for screening and confirmation of chemicals for activity against Trichophyton tonsurans. Growth characteristics were examined on two platforms (96- and 384-well) in three media at eight spore concentrations over a period of up to 120 h. Microspectrophotometry was used to automate plate reads. The 384-well platform was used to screen more than 7000 compounds from six chemical libraries. Z-scores for optical density relative to positive growth controls were used to flag compounds of interest and activity confirmed in separate assays. The final conditions selected for both screening and confirmation with minimum inhibitory concentration (MIC) determination were growth for 48 h at 32 °C in SabDex with 1 × 104 spores per reaction. Sensitivity and specificity averaged 99.2% (range, 95.2%−100%) and 99.8% (range, 99.1%−100%), respectively. MICs for known antifungals were similar to those reported by others using Clinical and Laboratory Standards Institute methods. Several novel compound classes were identified to have activity against T. tonsurans with potency comparable to known antifungals. A robust, reproducible assay is described that permits high-throughput screening in T. tonsurans.

The samples humidified at 85°C/85% RH for 384 h, on which a load of 3.164 Kg was dropped four times from a height of 0.4 m, exhibit the most severe results. The contact resistance increases by 700%, which had been about 62 mΩ in the as-bonded condition. The samples without humidification showed a sluggish and gentle increase in contact resistance with the induced mechanical shock. The contact resistance was found to be increased by 400% after the sixth drop from a height of 0.5 m. Scanning electron microscope images show particle deformation due to abrasion and friction between the contacting surfaces resulting from the sudden impact. Joints are also observed with no connections, which signify open circuits. Almost 25% of the circuits were found open in the samples (after 384 h in a humid environment), which have suffered severe mechanical shock (load drops four times from 0.4 m height). Breaking of the conductive layer of the particle and exposing the underlying polymeric portion were also observed.

The unicellular green alga Chlamydomonas reinhardtii responds to sulfate deprivation by producing an arylsulfatase (Lien, T., and O. Schreiner. 1975. Biochim. Biophys. Acta. 384:168-179; Schreiner, O., 1975. Biochim. Biophys. Acta. 384:180-193) and by developing the capacity to transport sulfate more rapidly (our unpublished data). The arylsulfatase activity, detectable 3 h after the transfer of the cells to low sulfate medium (less than or equal to 10 microM sulfate), is a periplasmic protein released into the culture medium by cw15, a cell wall-less mutant of C. reinhardtii. We have purified the derepressible arylsulfatase to homogeneity and have raised monospecific antibodies to it. The protein monomer (67.6 kD) associates into a dimer, and the enzyme activity shows an alkaline pH optimum and a Km of 0.3 mM for p-nitrophenylsulfate. Studies focused on arylsulfatase biosynthesis demonstrate that it is glycosylated and synthesized as a higher molecular mass precursor. The mature protein contains complex N-linked oligosaccharides and the primary translation product has an apparent molecular mass approximately 5 kD larger than the deglycosylated monomer. Since translatable RNA encoding the arylsulfatase can only be detected in cells after sulfate starvation, it is likely that accumulation of the enzyme is regulated at the level of transcription, although posttranscriptional processes may also be involved.

Crystals of tris(N,N-dimethylaminomethyl)phosphine oxide (tmpo) are hexagonal, space group P63; a = 8.723(5), c = 10.636(5) Å, Z = 2, R = 0.039 for 384 observed reflections. The complex [Co(tmpo)2(H2O)2](ClO4)2, forms orthorhombic crystals, space group Pbca, a = 19.16(1), b = 10.576(2), c = 117.043(4) Å, Z = 4, R = 0.069 for 1 409 observed reflections. The molecule is centrosymmetric, with the Co atom lying in the center. The tmpo is coordinated through the oxygen atom and one of the nitrogen atoms. The coordination of the ligand brings about very small changes in the bond lengths and angles except for the P=O bond. The coordinated water molecules play a major part in the structure of [Co(tmpo)2(H2O)2](ClO4)2.

Resumen

Coccomyxa subellipsoidea C-169 (C-169) is an oleaginous microalga which is promising for renewable biofuel production. MicroRNAs (miRNAs), as the pivotal modulators of gene expression at post-transcriptional level, are prospective candidates for bioengineering practice. However, so far, no miRNA in C-169 has been reported and its potential impact upon CO2 supplementation remains unclear. High-throughput sequencing of small RNAs from C-169 cultured in air or 2% CO2 revealed 124 miRNAs in total, including 118 conserved miRNAs and six novel ones. In total, 384 genes were predicted as their potential target genes, 320 for conserved miRNAs and 64 for novel miRNAs. The annotated target genes were significantly enriched in six KEGG pathways, including pantothenate and CoA biosynthesis, C5-branched dibasic acid metabolism, 2-oxocarboxylic acid metabolism, butanoate metabolism, valine, leucine and isoleucine biosynthesis and alpha-linolenic acid metabolism. The miRNAs’ target genes were enriched in lipid metabolism as well as RNA-interacting proteins involved in translation, transcription and rRNA processing. The pioneering identification of C-169 miRNAs and analysis of their putative target genes lay the foundation for further miRNA research in eukaryotic algae and will contribute to the development of C-169 as an oleaginous microalga through bioengineering in the future.

Teremos como foco de análise as três Filipícas, com a intenção de captar, nesses discursos, o sentido da identidade grega ligada ao sistema políade, único sistema político visto por Demóstenes como digno para os helenos e o único capaz de preservar a liberdade da Hélade. Em contrapartida, iremos examinar como, ao mesmo tempo, Demóstenes, ao falar do ser grego, da natureza da polis, constrói toda uma retórica segundo a qual Filipe II, rei da Macedônia, é simbolizado como bárbaro e inimigo da Hélade.

Resumen

The purpose of this paper is to contribute to a vision for leadership in business, banking, and politics based on a recovery of virtue. It draws principally on the works of the classical philosophers Aristotle (384–322 BCE) and Plato (c. 427–347 BCE) in line with the contemporary resurgence of Aristotle associated with Alasdair MacIntyre and others. In the context of an ethical analysis of the Irish banking crisis (2008-2016), the paper will show how virtue ethics can contribute to the avoidance of a repetition of the disastrous financial crisis of 2008 in Ireland and globally. It proposes a holistic approach that integrates virtue and culture, ethics and governance.

Resumen

Despite the widespread use of telemetry to track the movements of many different avian species, there are few published studies describing tracking methods for large psittacine birds. Due to their powerful bills and inquisitive demeanours, large parrots may damage valuable transmitters and confound telemetry studies. We undertook a captive trial of three attachment methods (collar, harness and tail-mount) and a novel weak-link harness design for black cockatoos (Calyptorhynchus latirostris, C. baudinii and C. banksii naso). Mean retention times for the transmitter packages ranged from 44 to 384 days. There was no skin or feather damage to the birds associated with transmitter attachments. The results showed sufficient transmitter retention times to allow for the collection of valuable movement and survival data, with no obvious ill effects on animal welfare, and are a first step towards using transmitters on wild cockatoos.

O objetivo do trabalho foi estudar a eficiência de alguns inseticidas para o controle de Chrysodeixis includens (Lepidoptera: Noctuidae) em duas safras na cultura da soja. Desenvolveu-se dois experimentos, sendo um na safra 2006/07 (experimento 1) e outro na safra 2007/08 (Experimento 2), no município de Chapadão do Sul (MS), sendo delineados em blocos ao acaso com 7 tratamentos e 4 repetições (experimento 1) e 6 tratamentos e 4 repetições (experimento 2). No experimento 1, os tratamentos foram: 1. Testemunha; 2. Profenofós + Lufenuron (151 g i.a. ha-1); 3. Metomil (7,5 g i.a. ha-1); 4. Teflubenzuron (384 g i.a. ha-1); 5. Thiodicarb (24 g i.a. ha-1); 6. Clorpirifós (28,8 g i.a. ha-1); 7. Fenitrotion + esfenvarelate (384 g i.a. ha-1). No experimento 2, foram utilizados os tratamentos: 1. Testemunha; 2. Teflubenzuron (12 g i.a. ha-1); 3. Metomil (129 g i.a. ha-1); 4. Profenofós + Lufenuron (15+150 g i.a. ha-1); 5. Flubendiamid (12 g i.a. ha-1); 6. Flubendiamid (14,4 g i.a. ha-1). Foi observada uma baixa eficiência dos inseticidas no controle de C. includens na cultura da soja. Os inseticidas Fenitrotion+Esfenvarelate, Tiodicarb, Metomil e Clorpirifós são os mais eficientes para o controle de lagartas pequenas de falsa-medideira (Chrysodeixis includens). Para as lagartas grandes os inseticidas mais eficientes são Flubendiamide e Profenofós+Lufenuron.

Resumen

ABSTRACT The survival of Escherichia coli and Salmonella strains during diced white radish kimchi fermentation was studied. Kimchi batches inoculated with the pathogens were fermented at 4, 15, and 25°C for 42 to 384 h. Cell counts of E. coli and Salmonella were enumerated on E. coli–coliform count plates and xylose lysine deoxycholate agar, respectively. Baranyi (primary model) and polynomial (secondary model) models, validated by root mean square error, were used to describe the kinetic behavior of the pathogens. In the primary model, both the death phase shoulder (E. coli: 208.18 to 8.25 h, 4 to 25°C; Salmonella: 79.91 to 0.97 h, 4 to 25°C) and bacterial cell counts (log CFU per gram per hour) decreased with increasing temperature (P < 0.05) (death rate: E. coli: −0.02 to −0.09, 4 to 25°C; Salmonella: −0.01 to −0.10, 4 to 25°C), the results being equally significant in the secondary model. The root mean square error (0.480 to 0.485) showed that the model performance was good. The fermentation temperature and time are the critical factors that control pathogenic E. coli and Salmonella in kimchi.
