Texto completoThe CCAAT enhancer binding protein beta (C/EBPβ) transcription factor regulates differentiation, proliferation, and functionality of many cell types, including various cells of the immune system. A detailed molecular understanding of how C/EBPβ directs alternative cell fates remains largely elusive. Ectopic expression of C/EBPβ has been previously shown to reprogram committed B cell progenitors into inflammatory macrophages. We took advantage of this reprogramming system in order to examine how C/EBPβ regulates (trans)differentiation. To determine which C/EBPβ protein modules are important for reprogramming, C/EBPβ wild type isoforms and mutants were ectopically expressed in primary mouse B cell progenitors. The data showed that the translationally regulated long isoforms LAP* and LAP, but not the N-terminally truncated isoform LIP can reprogram lymphoid cells into myeloid cells. Furthermore, we found that conserved regions 2,3 and 4 in the C/EBPβ protein transactivation domain are necessary and sufficient for B-to-myeloid cell conversion. Interestingly, the reprogrammed myeloid cells were found to represent a heterogeneous mixture of different myeloid cell types. Detailed analyses of the reprogrammed CD11b+ cells revealed that discrete conserved regions in C/EBPβ activated distinct pro- and anti-inflammatory genes and triggered divergent differentiation programs. Moreover, not only structural C/EBPβ mutants, but also post-translational modification (PTM) site mutations led to different reprogramming outcomes. These data suggest that C/EBPβ orchestrates myeloid diversification by integrating PTMs with structural plasticity as signal dependent adaptable modular properties to determine cell fate.

Texto completoThe proteasome system of the cell is responsible for the degradation of proteins and plays a central role in the generation of epitopes which are presented to cytotoxic T-lymphocytes (CTLs) on MHC-class-I molecules. The stimulation of cells by interferon-gamma (IFNgamma) leads to the formation of immunoproteasomes that show an improved generation of many MHC-class-I epitopes compared to constitutive proteasomes. In healthy mice, immunoproteasomes are mainly expressed in the lymphatic tissues, whereas the non-lymphatic organs predominantly contain constitutive proteasomes. In this project the effect of Listeria monocytogenes infection on murine 20S proteasomes derived from the liver, spleen, small intestine and colon were investigated. The structure of the isolated proteasomes was analyzed by two-dimensional gel electrophoresis and western blots while the function was studied by in vitro processing of three oligomeric peptide substrates. Identification and quantification of the processing products was performed by HPLC-ESI-ion trap mass spectrometry. The project showed for the first time, that after infection 20S proteasomes isolated from non-lymphatic organs as well as from non-lymphatic cells displayed structural and functional plasticity: immunoproteasomes were induced post infection which could be correlated with the enhanced generation of immuno-relevant fragments. This was independent of the direct presence of Listeria monocytogenes in the organs and solely controlled by the cytokine IFNgamma. In addition, an increased posttranslational modification with the monosaccharide N-acetylglucosamine could be detected in liver-derived proteasomes after infection. Furthermore, a detailed analysis of the mass spectrometry data was established according to the cleavage site usage of constitutive and immunoproteasomes. The result was that immunoproteasomes are involved in improved generation of the immuno-relevant fragments by the faster cleavage and the changed usage of existing cleavage sites after infection.

Texto completoLipopolysaccharide (LPS) binding protein (LBP) is an acute phase protein with the ability to bind and transfer LPS of Gram-negative bacteria. This soluble pattern recognition molecule represents an important defense principle of the host. Regulation of the hepatic acute phase response and its termination are important mechanisms for limiting systemic inflammatory activity of the host. Here were analyze the cooperation of Interleukin (IL)-1, IL-6, and Dexamethasone (Dex) at LBP expression in the hepatoma cell lines HuH-7 and Hep G2. The major inducer of LBP expression is IL-6. Within the LBP promoter numerously highly consensus binding sites such as AP-1, C/EBP-beta? and STAT3 are present, that confer transcriptional activity as shown by truncation and mutation experiments. Additionally, activate NF-kappaB sites activated by IL-1 were detected at the LBP promoter. By mutation experiments of the promoter furthermore were found differentially active glucocorticoid response elements (GREs). The promoter contains GREs enhancing the activity as well as inhibitory ones. The enhancing effect towards LBP expression by Dex was mediated by IL-6. Dex stimulated the expression of the IL-6 receptor and therefore upregulated the IL-6 pathway. Transforming Growth Factor (TGF)-beta is able to inhibit LBP expression in stimulated cells. An AP-1 binding site was identified mediating inhibitory TGF-beta effects towards LBP promoter activity. Furthermore it was shown that a growth factor independence (Gfi)-1 binding site localized near the AP-1 site is essential for mediating the TGF-beta inhibitory effect. The relevancy of the Gfi-1 site fore mediating TGF-beta effects indicates a novel mechanism for understanding inhibitory TGF-beta effects at the transcriptional level. In summary the complex regulation of LBP were elucidate which may help to eventually develop novel intervention strategies for acute phase, sepsis, and septic shock.

Texto completoThe growing demand for traditional bicycle and e-bikes is an opportunity for the market of locking and antitheft systems for these types of vehicles. This dissertation intends to study the smart padlocks market, understand its strengths and limitations; create an intelligent padlock concept that tries to fill the gaps in this market, considering the customer’s needs; and finally, perform the mechanical design and automation of the system.The benchmarking of smart padlocks allowed us to understand the main characteristics of these locking systems, advantages, limitations and extras. User’s main features were identified and, using the Mudge diagram, these needs were ranked in descending order of value to the client. The QFD diagram was used to identify critical features for the customer, specify the correlation between customer needs and project requirements, and order the project requirements in descending order of improvement priority. The generated padlock’s concept is easy to mount on a typical bike, with high compatibility between bicycles and without the need for an external object to lock it, with a worm drive locking mechanism. The communication between the lock and the user is made by Bluetooth and GSM/GPRS to allow a global reach. The bicycle can be unlocked remotely, unlocking the padlock with the user’s approach or manual, in case of lack of battery. The application makes it possible to lock and unlock the bike, allowing it to be shared with third parties; tracks statistics such as the distance travelled, average speed and calories burned and can track the bike by GPS.The mechanical dimensioning of the worm drive system allowed the determination of the parameters of the worm wheel and the worm. The worm wheel has a prime diameter of 133 mm, with an outer diameter of 136 mm and an inner diameter of 129 mm, with 76 teeth. The worm has a prime diameter of 10 mm, an outer diameter of 13 mm and an inner diameter of 6 mm. The maximum worm length is 34 mm and the minimum length is 18 mm.The research and development in this field allows the improvement of smart padlocks, increasing security and alternatives available on this market, providing a better and more personalized experience for the user.
