In Pseudomonas protegens CHA0 and other fluorescent pseudomonads, the Gac/Rsm signal transduction pathway is crucial for the expression of secondary metabolism and the biological control of fungi, nematodes, and insects. Based on the findings of a previous metabolomic study, the role of intracellular γ-aminobutyrate (GABA) as a potential signal in the Gac/Rsm pathway was investigated herein. The function and regulation of a gabDT (c01870-c01880) gene cluster in strain CHA0 were described. The gabT gene encoded GABA transaminase (GABAT) and enabled the growth of the bacterium on GABA, whereas the upstream gabD gene (annotated as a gene encoding succinic semialdehyde dehydrogenase) had an unknown function. A gacA mutant exhibited low GABAT activity, leading to the markedly greater intracellular accumulation of GABA than in the wild type. In the gacA mutant, the RsmA and RsmE proteins caused translational gabD repression, with concomitant gabT repression. Due to very low GABAT activity, the gabT mutant accumulated GABA to high levels. This trait promoted a planktonic lifestyle, reduced biofilm formation, and favored root colonization without exhibiting the highly pleiotropic gacA phenotypes. These results suggest an important role of GABA in the Gac/Rsm-regulated niche adaptation of strain CHA0 to plant roots.

Access to microarcsecond astrometry is now routine in the radio, infrared (IR), and optical domains. In particular, the publication of the second data release ( Gaia DR2) from the Gaia mission made it possible for every astronomer to work with easily accessible, high-precision astrometry for 1.7 billion sources to twenty-first magnitude over the full sky. ▪ Gaia provides splendid astrometry, but at the limits of the data small systematic errors are present. A good understanding of the Hipparcos– Gaia astrometry concept, and of the data collection and processing, provides insights into the origins of the systematic errors and how to mitigate their effects. ▪ A selected set of results from Gaia highlight the breadth of exciting science and unexpected results, from the Solar System to the distant Universe, to creative uses of the data. ▪ Gaia data release 2 provides, for the first time, a dense sampling of Galactic phase space with high-precision astrometry, photometry, and radial velocities, allowing researchers to uncover subtle features in phase space and the observational Hertzsprung–Russell diagram. ▪ In the coming decade, we can look forward to more accurate and richer Gaia data releases, and new photometric and spectroscopic surveys coming online that will provide essential complementary data. ▪ The longer term promises exciting new opportunities for microarcsecond astrometry and beyond, including the plans for an IR version of Gaia that would offer the dense sampling of phase space deep into the Milky Way's nuclear regions.

AbstractWe present first results from our study of the host galaxies and environments of quasars in Galaxy And Mass Assembly (GAMA), a multiwavelength photometric and spectroscopic survey for ∼300,000 galaxies over ∼300 deg2, to a limiting magnitude of r ∼ 20 mag. We use a GAIA-selected sample of ∼350 quasars at z < 0.3 in GAMA. For all the quasars, we determine all surrounding GAMA galaxies and measure their star formation (SF) rate and SF history, and the host galaxy morphology and group membership of the quasars. As a comparison sample of inactive galaxies, we use 1000 subsets of galaxies in GAMA, matched in redshift and galaxy stellar mass to the quasars. We find that quasar activity does not depend on the large-scale environment (cluster/group/void), although quasars tend to prefer satellite location in their environment. Compared to inactive galaxies, quasars are preferentially hosted in bulge-dominated galaxies and have higher SF rates, both overall and averaged over the last 10 and 100 Myr. Quasars also have shorter median SF timescales, shorter median time since the last SF burst, and higher metallicity than inactive galaxies. We discuss these results in terms of triggering mechanisms of the quasar activity and the role of quasars in galaxy evolution.

This study aimed to evaluate apple firmness measured using both the penetrometer and acoustic methods. The methodologies were applied to Royal Gaya and Golden Smoothee apples (Malus domestica. Borkh) harvested from 12 different orchards in Catalonia (Spain), on six different dates, and over three seasons. The relationship between firmness and physicochemical quality parameters was analyzed at harvest and post-harvest. The results obtained showed a noticeable correlation between Magness Taylor firmness and acoustic measurements in Royal Gala, but no correlation was found for Golden Smoothee. Fruit variety and storage had a clear influence on correlations between the two measurements. Measurements of Magnes Taylor and acoustic firmness seemed to correspond to different physical properties. Acoustic measurements seemed to be a good tool for evaluating changes in tissue firmness during long-term storage, especially for Golden Smoothee apples. The maturity of the fruit seemed to play a more important role in the measurement of Magnes Taylor firmness than in the measurement of acoustic firmness for the Royal Gaya variety.

Context. Since July 2014, the Gaia mission has been engaged in a high-spatial-resolution, time-resolved, precise, accurate astrometric, and photometric survey of the entire sky. Aims. We present the Gaia Science Alerts project, which has been in operation since 1 June 2016. We describe the system which has been developed to enable the discovery and publication of transient photometric events as seen by Gaia. Methods. We outline the data handling, timings, and performances, and we describe the transient detection algorithms and filtering procedures needed to manage the high false alarm rate. We identify two classes of events: (1) sources which are new to Gaia and (2) Gaia sources which have undergone a significant brightening or fading. Validation of the Gaia transit astrometry and photometry was performed, followed by testing of the source environment to minimise contamination from Solar System objects, bright stars, and fainter near-neighbours. Results. We show that the Gaia Science Alerts project suffers from very low contamination, that is there are very few false-positives. We find that the external completeness for supernovae, CE = 0.46, is dominated by the Gaia scanning law and the requirement of detections from both fields-of-view. Where we have two or more scans the internal completeness is CI = 0.79 at 3 arcsec or larger from the centres of galaxies, but it drops closer in, especially within 1 arcsec. Conclusions. The per-transit photometry for Gaia transients is precise to 1% at G = 13, and 3% at G = 19. The per-transit astrometry is accurate to 55 mas when compared to Gaia DR2. The Gaia Science Alerts project is one of the most homogeneous and productive transient surveys in operation, and it is the only survey which covers the whole sky at high spatial resolution (subarcsecond), including the Galactic plane and bulge.

ABSTRACT Gassericin A is a circular bacteriocin produced by Lactobacillus gasseri strain LA39. We found a 33,333-bp plasmid, designated pLgLA39, in this strain. pLgLA39 contained 44 open reading frames, including seven genes related to gassericin A production/immunity (gaa), as well as genes for replication, plasmid maintenance, and conjugative transfer. pLgLA39 was transferred from LA39 to the type strain of L. gasseri (JCM 1131) by filter mating. The transconjugant exhibited >30-fold-higher more resistance to gassericin A and produced antibacterial activity. Lactobacillus reuteri LA6, the producer of reutericin 6, was proved to harbor a plasmid indistinguishable from pLgLA39 and carrying seven genes 100% identical to gaa. This suggests that pLgLA39 might have been transferred naturally between L. gasseri LA39 and L. reuteri LA6. The seven gaa genes of pLgLA39 were cloned into a plasmid vector to construct pGAA. JCM 1131T transformed with pGAA expressed antibacterial activity and resistance to gassericin A. pGAA was segregationally more stable than a pGAA derivative plasmid from which gaaA was deleted and even was more stable than the vector. This suggests the occurrence of postsegregational host killing by the gaa genes. pLgLA39 carried a pemIK homolog, and segregational stabilization of a plasmid by the pLgLA39-type pemIK genes was also confirmed. Thus, pLgLA39 was proved to carry the genes for at least two plasmid maintenance mechanisms, i.e., gaa and pemIK. Plasmids containing a repA gene similar to pLgLA39 repA were distributed in several L. gasseri strains.

The Zurich-Basel PhD program Science and Policy involves early-stage scientists in science-policy boundary work through internships and co-supervision at academic and policy institutions. In a Delphi study, students reported to have acquired new skill sets for science-policy dialogue. However, challenges remain, including the lack of time for policy work during a PhD project or the different incentive systems in academia and policy.

The astrometric European Space Agency (ESA) Gaia mission was launched in December 19, 2013. One of the tasks of the Gaia mission is production of an astrometric catalog of over one billion stars and more than 500000 extragalactic sources. The quasars (QSOs), as extragalactic sources and radio emitters, are active galactic nuclei objects (AGNs) whose coordinates are well determined via Very Long Baseline Interferometry (VLBI) technique and may reach sub-milliarcsecond accuracy. The QSOs are the defining sources of the quasi-inertial International Celestial Reference Frame (ICRF) because of their core radio morphology, negligible proper motions (until sub-milliarcsecond per year), and apparent point-like nature. Compact AGNs, visible in optical domain, are useful for a direct link of the future Gaia optical reference frame with the most accurate radio one. Apart from the above mentioned activities, Gaia has other goals such as follow-up of transient objects. One of the most important Gaia's requirements for photometric alerts is a fast observation and reduction response, that is, submition of observations within 24 hours. For this reason we have developed a pipeline. In line with possibilities of our new telescope (D(cm)/F(cm)=60/600) at the Astronomical Station Vidojevica (ASV, of the Astronomical Observatory in Belgrade), we joined the Gaia-Follow-Up Network for Transients Objects (Gaia-FUN-TO) for the photometric alerts. Moreover, in view of the cooperation with Bulgarian colleagues (in the frst place, SV), one of us (GD) initiated a local mini-network of Serbian { Bulgarian telescopes useful for the Gaia-FUN-TO and other astronomical purposes. During the next year we expect a new 1.4 m telescope at ASV site. The speed of data processing (from observation to calibration server) could be one day. Here, we present an overview of our activities in the Gaia-FUN-TO which includes establishing Serbian { Bulgarian mini-network (of five telescopes at three sites, ASV in Serbia, Belogradchik and Rozhen in Bulgaria), the Gaia-FUN-TO test observations, and some results.

Context. The European Space Agency’s Gaia satellite was launched into orbit around L2 in December 2013. This ambitious mission has strict requirements on residual systematic errors resulting from instrumental corrections in order to meet a design goal of sub-10 microarcsecond astrometry. During the design and build phase of the science instruments, various critical calibrations were studied in detail to ensure that this goal could be met in orbit. In particular, it was determined that the video-chain offsets on the analogue side of the analogue-to-digital conversion electronics exhibited instabilities that could not be mitigated fully by modifications to the flight hardware. Aims. We provide a detailed description of the behaviour of the electronic offset levels on short (<1 ms) timescales, identifying various systematic effects that are known collectively as “offset non-uniformities”. The effects manifest themselves as transient perturbations on the gross zero-point electronic offset level that is routinely monitored as part of the overall calibration process. Methods. Using in-orbit special calibration sequences along with simple parametric models, we show how the effects can be calibrated, and how these calibrations are applied to the science data. While the calibration part of the process is relatively straightforward, the application of the calibrations during science data processing requires a detailed on-ground reconstruction of the readout timing of each charge-coupled device (CCD) sample on each device in order to predict correctly the highly time-dependent nature of the corrections. Results. We demonstrate the effectiveness of our offset non-uniformity models in mitigating the effects in Gaia data. Conclusions. We demonstrate for all CCDs and operating instrument/modes on board Gaia that the video-chain noise-limited performance is recovered in the vast majority of science samples.

Resumen
AbstractGaia is an ESA cornerstone mission conducting a full sky survey over its 5 year operational period. Gaia performs astrometric, photometric and spectroscopic measurements. The data processing is entrusted to scientists and engineers who have formed the Gaia Data Processing and Analysis Consortium (DPAC). The photometric science alerts started in 2014. The first intermediate data release (Gaia DR1) took place 14 September 2016 and it has been extensively used by the community. Gaia DR2 is scheduled for April 2018. Gaia is expected to be able to continue observations roughly for another 5 years after the nominal phase. The procedure to grant funding for the extension period has been initiated. In case funding is granted, the total operational time of Gaia may be 10 years.

To reveal the key enzyme genes involved in γ-aminobutyric acid (GABA) metabolic pathways response to elevated metabolite storage in embryos during barley germination, this study investigated the GABA content, cloned GABA metabolic pathway genes and analyzed their expression levels, respectively. In barley embryos, GABA content continued to rise during the soaking process and then decreased after the germination. Three genes including glutamic acid decarboxylase (GAD), GABA transaminase (GABA-T) and succinic semialdehyde dehydrogenase (SSADH) involved in the GABA pathway were cloned and characterized from the barley embryos, respectively. Before the germination, the expression of GAD gene was up-regulated, while GABA-T gene expression was down-regulated. After the germination, GAD gene expression was lowered, but GABA-T gene expression was rapidly increased. The SSADH gene expression remained stable after soaking of 4 h, and then down-regulated. There is evidence that the high GABA content in germinating barley seeds is parallel with the upregulation of the GAD gene, and down-regulation of GABA-T gene. These results indicate that the expression level of the genes involved in GABA pathway is a crucial factor in GABA accumulation during soaking and germination. This study is beneficial for the development of GABA-rich barley products by germination. © 2021 Friends Science Publishers

Abstract Introduction Acute myeloid leukemia (AML) is a malignant clonal hematopoietic stem cell disease. Ecotropic viral integration site-1(EVI-1) has been recognized as one of the most dominant oncogenes associated with murine and human myeloid leukemia. EVI-1 harbors several hallmark functions that are normally associated with leukemogenesis. Overexpression EVI-1 upregulates the transcription of the transcription factor gene GATA-2, which plays a critical role in the maintenance of hematopoietic stem cells.EVI-1 also binds to the transcription factors GATA-1,PU.1,RUNX-1,SCL and LMO2 thereby inhibiting their activity and blocking the differentiation of hematopoietic progenitors. Arsenic trioxide (ATO), which was used as a traditional Chinese medicine, has shown excellent therapeutic efficiency for acutepromyelocyticleukemia. A previous study has demonstrated that ATO targets EVI-1 protein to induce apoptosis . However, the molecular mechanisms underlying this regulation of AML cells by ATO have not been fully elucidated. We aim to expand our understanding of the cellular regulation and effect of ATO on EVI-1 and several related transcription factors (such as GATA1, GATA2, RUNX-1, LMO2, PU-1) in human acute monocytic leukemia THP-1 cells. Methods The endogenous expression of EVI-1 is higher in THP-1 cells, which was confirmed through gene analysis sequencing and comparison with Genbank.THP-1 cells were treated with various concentrations of ATO( 0,1,3,5¦ÌM) for 24h, 48h and 72h. Expressions of EVI-1,and related transcription factor genes(GATA-2 ,GATA-1, RUNX-1,MPO,LMO,PU.1,SCL2) were determined by qRT-PCR. Results Compared to controls, THP-1 cells with high expression of EVI-1 gene were selected to confirm that overexpression of EVI-1 can promote the expression of GATA-2 , and reduce the level of GATA-1, RUNX-1, MPO, LMO, PU.1, SCL2 simultaneously in vitro(Fig.1).To determine whether ATO is able to directly regulate EVI-1 and related transcription factor genes expression in THP-1 cells, we examined their mRNA expression by qRT-PCR after treatment with different concentrations of ATO. We found that ATO has the ability to down regulate EVI-1 gene in a dose-dependent and time-dependent manner( Fig.2a).Moreover, GATA-2 gene expression decreased when THP-1 cells were treated with ATO(Fig.2b). We examined the related transcription factors expression and found that ATO suppressed the expression levels of PU.1, RUNX-1, SCL and LMO2 (Fig.2c, 2d, 2e) in THP-1 cells. Interestingly, exposure to increasing concentrations of ATO for increasing lengths of time was associated with gradual promotion in the expression of GATA-1 mRNA( Fig 2f). Summary We verified that the endogenous expression of EVI-1 is higher in THP-1 cells, which confirmed THP-1 cells can be a vitro model to investigate EVI-1 gene functions. Meanwhile, related transcription factor gene GATA-2, which is regarded as EVI-1 regulatory element, is upregulated in THP-1 cells. Moreover, GATA-1, which is essential for erythroid differentiation, and other related transcription factor genes (PU.1, RUNX-1, SCL and LMO2) decreased in THP-1 cells. ATO dose-and time-dependently decreased expressions of EVI-1 and GATA-2 gene and regulated those related transcription factors in EVI-1-overexpressing cells compared with control cells. The study aimed to find out: (1) differences in science learning outcomes between students who were taught with delayed and direct feedback giving techniques, (2) the interaction effects of feedback techniques and spatial cognitive style on science learning outcomes, (3) differences in science learning outcomes in the group of students who have high spatial cognitive style, those who are taught using delayed and direct feedback giving techniques, (4) differences in science learning outcomes in students who have a low spatial cognitive style, are taught using delayed and reversed direct giving techniques . The method used is tretment by level with 2 x 2 ANAVA design. The results of this study show that the learning outcomes of science in students who are taught by the technique of giving direct natural science learning outcomes are higher than the delayed giving technique. There is an influence of the interaction between the reciprocal giving technique and the cognitive style of the students towards the learning outcomes of science. Students who have a high spatial cognitive style learned by the technique of giving direct feedback to science learning outcomes are higher than the delayed giving technique. For students who have a low spatial cognitive style, learning techniques for giving direct feedback to science learning outcomes are lower than delayed feedback techniques. Penelitian bertujuan untuk mengetahui: (1) perbedaan hasil belajar IPA antara peserta didik yang dibelajarkan dengan teknik pemberian balikan langsung dan balikan tertunda, (2) pengaruh interaksi teknik pemberian balikan dan gaya kognitif spasial terhadap hasil belajar IPA, (3) perbedaan hasil belajar IPA pada kelompok peserta didik yang memiliki gaya kognitif spasial tinggi, yang dibelajarkan dengan menggunakan teknik pemberian balikan langsung dan balikan tertunda, (4) perbedaan hasil belajar IPA pada peserta didik yang memiliki gaya kognitif spasial rendah, dibelajarkan dengan menggunakan teknik pemberian balikan langsung dan balikan tertunda. Metode yang digunakan adalah tretmen by level dengan rancangan desain ANAVA 2 x 2. Hasil dari penelitian ini menunjukan hasil belajar IPA pada peserta didik yang dibelajarkan dengan teknik pemberian balikan langsung hasil belajar IPA lebih tinggi daripada teknik pemberian balikan tertunda. Terdapat pengaruh interaksi antara teknik pemberian balikan dan gaya kognitif peserta didik terhadap hasil belajar IPA. Peserta didik yang memiliki gaya kognitif spasial tinggi yang dibelajarkan dengan teknik pemberian balikan langsung hasil belajar IPA lebih tinggi daripada teknik pemberian balikan tertunda. Untuk peserta didik yang memiliki gaya kognitif spasial rendah, dibelajarkan dengan teknik pemberian balikan langsung hasil belajar IPA lebih rendah daripada teknik pemberian balikan tertunda.

In view of the global grand challenges, fundamental research institutions are increasingly being asked to provide context for the application of their research findings and to incorporate transdisciplinary forms of knowledge production. But how can the involvement of stakeholders from outside academia be captured and evaluated within the research process? And how can they be engaged in meaningful science-stakeholder dialogue? “Good” processes are a prerequisite for meeting these changing requirements and for ensuring a successful knowledge transfer at the interface between science and society.Societal challenges are increasing on a global scale, requiring an intensification of knowledge transfer (KT) between scientific and societal actors on multiple levels. These transfers bring up new demands regarding the way in which knowledge is produced and transferred, which mechanisms are utilized to ensure the quality of these knowledge interactions, and how such interactions can be evaluated. Capturing and evaluating KT, however, also opens up new reflections about what a meaningful “impact” is and how KTs are shaped and driven. The results presented here were reached by means of a formative and summative evaluation approach, and include an accompanying research effort that aimed to capture central KT processes from start to “finish” by a tailored KT process assessment framework. This framework was applied to the ongoing activities of twelve in-house KT projects (from 2014 to 2017) that were conducted in a fundamental natural science research institution within the field of earth system science. Our findings indicate that, among other things, the continuous assessment of the underlying processes of KT allows for processes and outcomes to be directly influenced, while also providing scope for institutional learning. Better insights into the definition of “societal relevance” may in fact not start with the result, but rather with the question of how the research will be conducted and for whom.

AbstractIn addition to its main goal of measuring the parallaxes of 109 stars in the Galaxy, ESA’s Gaia mission is also probing the time domain on a range of astrophysically interesting time-scales. The photometric measurements are processed for transient variability, and events that are discovered are published as a public alerts stream, known as Gaia Science Alerts. This talk gave an overview of the project to date, and highlighted its unique characteristics as a transient survey. It discussed briefly some of the recent scientific results which illustrate the broad range of science investigations enabled by the data. Gaia Alerts is particularly suited to the study of Galactic populations, as it covers the Galactic Plane with high astrometric precision and photometric accuracy.
