Three experiments carried out in the southeast of Mexico (Yucatan) are reported. In the first, yield of P. purpureum increased from 3,760 to 14,526 kgDM/ha with age 6 weeks (young) and 28 weeks (mature). Maturity increased lignin from 43 to 108g/kgDM. Potential degradabilities of leaf and stem were 788g & 782g/kgDM with young, and 725 & 403g/kgDM with mature. Young yielded more potentially degradable DM (70 & 35kgDM/ha/d) than mature. In the second, molasses urea block (MUB) supplementation of bulls grazing during the dry season, did not improve the potential degradability of pasture (525g/kgDM). Estimated pasture intake was 68gDM/kgW0.75 /d, and measured rumen pool size of grazing bulls was 79 gDM/kgW 0.75. Intake of MUB was low (236gDM/d) and only increased rumen NH 3-N from 100 to 128mg/l. Availability of pasture was 255kg DM/animal. It is suggested that need of supplementation could be low if there is sufficient pasture to allow selective grazing. In the third, hay diets of young or mature P. purpureum were studied in six zebu (Bos indicus) bulls fed ad libitum. Intakes were, 91 and 44gDM/kgW0.75/d, digestibilities, 591 and 453g/kgDM, estimated energy intakes 768 and 307kJME/kgW0.75/d and Metabolizable Protein, 4.1 and 1.3g/kgW0.75/d with young and mature forages respectively. Forage maturity resulted in negative N balance (352 vs -113mgN/kgW0.75/d), decreased potential (756 vs 568g/kgDM) and fractional rate (0.07 vs 0.03%) of rumen DM degradation, but gave higher DM degradabilities of the non-soluble fraction (378 vs 283g/kgDM). Forage maturity reduced rumen outflow rates of liquid (0.09 vs 0.05%), and solid (0.038 vs 0.015%) phases, increasing mean retention times of digesta (22 vs 65h), and the rumen pool size (84 vs 120gDM/kgW0.75) and reduced the eating (380 vs 273min) and rumination daily times (433 vs 398min). The Purine Derivatives (PD) excretion in urine reflected differences between diets (651 vs 354mMol/kgW0.75/d). However, rumen Microbial N Supply estimated by the PD method was very low in comparison with estimates from digestibility and N balance measurements. It is suggested this effect could be explained by greater recycling of PD to the rumen and a lower endogenous excretion by zebu cattle.

Traditional Dwellings in Ankara Citadel have an important place among both Anatolian and Ankara Traditional Dwellings, reflecting certain characteristics of their own. Citadel Area, as one of the oldest settlements in Ankara, has been hosting both residential and commercial activities for a very long period of time. Today, the area has regained its popularity
as a commercial, cultural and tourism center with the potential of its remaining values from the past and conservation movements held in the area starting from 1970&

s. Within these circumstances, the residential pattern inside Citadel area, the boundaries of which are strictly defined by historical fortress walls, carries the highest risk in the area. At this point, main factors threatening the existing pattern can be categorized in four titles, which also constitute the definition of the problem that the thesis is going to deal with. First factor is the rapid change in original use of traditional dwellings in the Outer Citadel due to the pressure derived from the increase in tourism, commercial and cultural activities in the area. Second factor is the assignment of gastronomic function as the most popular and major function among new functions of restored traditional dwellings in the area. The third factor is the disappearance of original characteristics and values of traditional dwellings due to restoration projects, which is caused by the discordant relation of the spatial, functional requirements of gastronomic use with the original characteristics, spatial capacity of traditional dwellings. Last factor is the present physical situation of traditional dwellings, which doesn&

t correspond with their physical situation in the projects approved by the Board of Conservation of Ankara. Outer Citadel is the study area for the problem with its clear boundaries, which is actually a buffer zone between the commercial, cultural, tourism zone outside the Citadel Area and Inner Citadel, in which the existing residential pattern, mostly preserving its original characteristics and use, is located. This thesis mainly intends to point out how the original characteristics, values and spatial quality of traditional dwellings are affected from the gastronomic function in both positive and negative ways within the context of contemporary understanding and methods of conservation. Discordances and problems between the approved restoration projects and the restoration implementations are another subject of the thesis. The thesis starts with an introduction chapter, informing about the theoretical framework established for the study, the definition and aim, which also informs about the methodology followed. Second chapter consists of a historical research on Ankara Citadel and examinations on the present situation of the area, traditional dwellings in Ankara Citadel and other developments in the city which have an effect on Citadel Area and traditional dwellings. Third chapter includes the analyses of traditional dwellings chosen for the study with their evaluations, in which the original characteristics and values these dwellings are also indicated. Fourth chapter consists of the analyses of the restoration projects of the traditional dwellings again with their evaluations, considering both the present situation and the original characteristics and values. Thesis ends with a conclusion chapter, which makes a general evaluation of the study within the context of modern conservation approaches.

Studiens syfte är att skapa en ökad förståelse om hur svenska förskolebarn i åldrarna fyra och fem år interagerar med och uppfattar de olika artefakterna på sin utegård. Med artefakter menas de fysiska redskap som finns på förskolans utegård. Vi har valt att inrikta oss på om barnen leker enskilt eller tillsammans med andra, om de leker utifrån given funktion eller skapad funktion samt hur de ser på artefakter med hänsyn till estetiska omdömen. Vi har använt oss av metoden the Mosaic approach som lyfter fram barns perspektiv. I vår studie har barn fotograferat utegården. Tio barn intervjuades om utegården där barnens fotografier användes för att samtala om och barnen fick sedan guida oss runt på utegården. I resultatet visade det sig att när barnen samtalade om bilderna kom de fram till många olika användningsområden och åsikter kring artefakterna. Under guidningar fick vi se hur fyra barn interagerade med artefakterna och vi fick även en omfattande bild av utegården. Det visade sig att barnen använde sand ihop med artefakterna, vilket kan bero på att sand är lättillgängligt och enkelt att förflytta. Det visade sig även att barn tänker på sin säkerhet eftersom barnen visade en oro att ramla ner från artefakter som var högt placerade. Vår slutsats är att utegården erbjuder enskildhet och lek tillsammans med andra samt att barnen använder givna och skapade funktioner tillsammans med artefakterna. Studiens resultat visar att artefakterna skapar en trivsam utegård, vilket visar sig när barnen gärna samtalar om och visar de olika artefakterna.
The study's purpose is to create a better understanding of how Swedish preschool children aged four to five years interact with and perceive the various artifacts on their patio. With artifacts means the physical tools that are on the preschool’s patio. We have chosen to focus on if the children play alone or with others, if they play by the given function or created function, and how they see the artifacts considering aesthetic judgments. We have used the method of the Mosaic approach that emphasizes the child's perspective. In our study have the children photographed the patio. Ten children were interviewed about the patio where the children's photographs were used to discuss. Four children guided us around the patio. The results showed that when the children talked about the pictures they came up with many different uses and opinions about the artifacts. During guided tours, we saw how four children interacted with the artifacts and we even got a comprehensive image of the patio. It turned out that the children used the sand together with the artifacts, which may be due to the sand is easily accessible and easy to move. It was also found that children think about their safety because the children showed a concern of falling from the artifacts that were highly placed. Our conclusion is that the patio offers privacy and play together with other children, and to use the given and created functions together with the artifacts. The results of the study show that the artifacts create a pleasant patio, which appears when the children gladly talk about and show the different artifacts.

After the students in high school have learned to integrate a function, the calculation of the volume of a solid of revolution, like a rotated parabola, is taken as a good applied example. The next step is to calculate the volume of an object of reality which is interpreted as a solid of revolution of a given function f(x). The students do all these calculations in the same way and get the same result. Consequently the teachers can easily decide if a result is right or wrong. If the students have learned to work with a graphical or CAS calculator, they can calculate the volume of solids of revolution in reality by modelling a possible fitted function f(x). Every student has to decide which points of the curve that generates the solid of revolution can be taken and which function will suitably fit the curve. In Austrian high schools teachers use GeoGebra as a software which allows you to insert photographs or scanned material in the geometric window as a background picture. In this case the student and the teacher can control if the graph of the calculated function will fit the generating curve in a useful way.

The Givens orthogonalization algorithm is an efficient alternative to the normal equations method for solving many adjustment problems in photogrammetry. The Givens method is one of a class of methods for solving linear systems known generally as orthogonalization or QR methods. It allows for sequential processing and greatly simplifies the computation of statistics on the observations and residuals. The underlying reason for these advantages is the immediate availability of the orthogonal Q matrix, which is computed as the data are processed and is intimately related to the statistics needed for blunder detection. One of these statistics, the F statistic computed from externally studentized residuals, is both easily obtained and well-suited for blunder detection. The Givens method requires nearly four times the number of computations as compared to the normal equations approach in order to reach a solution. However, depending on the size of the problem, blunder detection through the normal equations requires far more computer time than is required when starting with a Givens decomposition. The method allows a user to review intermediate results, test residuals and modify the solution without having to compute a full solution. Adjustments of a level net and a single-photo resection are used to demonstrate the method. Because of the advantage in computational time, the Givens method is superior to the normal equations approach when rigorous blunder detection is required.

The non-profit sector is growing, where NPOs compete for scarce resources. Meanwhile, innovation in communication technology requires resources and creativity. Can social media work as an efficient tool for communicating organizational goals and values and how does it enhance trust in NPOs relationship with donors? How do NPOs use ICT and social media in communication to donors and how do they implement functional and emotional communication online?  The research area is approached through a deductive, qualitative, and constructivist perspective. Interviews with two leading NPOs were held, as well as a number of interviews with potential donors. The theoretical framework builds upon Morgan and Hunts and MacMillan et al’s models of trust and commitment. Functional and emotional communication was developed through various theories on online communication. The main findings of this thesis are that Swedish NPOs mainly focus on emotional communication in order to obtain emotional commitment before further developing trust, which is created from functional communication. The investigated NPOs use functional communication to a very little extent.

Stockholm’s subway system is considered one of the longest art galleries in the world.Each station has its own identity, but at the same time, all metro stations have the sameuniform functional light approach.This light could not prevent the passengers from gathering and walking randomlyat the subway station’s platform. Moreover, the uniform light is neglecting the artwork onthe station’s platform; hence, this could lead to losing the identity feature of the station. Ingeneral, the demand for using public transportation in sustainable urban cities is increasing.This thesis studies how to use light as an information giver for the passengers on the subwaystation platform:By creating light safety zones and by enhancing the wayfinding to minimize conflict points.From the result of the questionnaire and the observations in two case studies, adesign principle with eight parameters has been developed to be applied in new lightingdesign for the T-Centralen platform station. It aims to restore the identity of the station,and create different zones at the T-Centralen subway platform, to enhance the movementof the passengers and to make it more organized by combining different layers of functional,atmospheric and dynamic light.

Der Schwerpunkt der vorliegenden Arbeit liegt auf Methoden zur Lösung nichtlinearer Optimierungsprobleme mit der Anforderung, jedes globale Optimum garantiert zu finden und mit einer im Voraus festgesetzten Genauigkeit zu approximieren. Eng verbunden mit dieser deterministischen Optimierung ist die Berechnung von Schranken für den Wertebereich einer Funktion über einem gegebenen Hyperquader. Verschiedene Ansätze, z. B. auf Basis der Intervallarithmetik, werden vorgestellt und analysiert. Im Besonderen werden Methoden zur Schrankengenerierung für multivariate ganz- und gebrochenrationale Polynome mit Hilfe der Darstellung in der Basis der Bernsteinpolynome weiterentwickelt. Weiterhin werden in der Arbeit schrittweise die Bausteine eines deterministischen Optimierungsverfahrens unter Verwendung der berechneten Wertebereichsschranken beschrieben und Besonderheiten für die Optimierung polynomialer Aufgaben näher untersucht. Die Analyse und Bearbeitung einer Aufgabenstellung aus dem Entwicklungsprozess für Fahrzeuggetriebe zeigt, wie die erarbeiteten Ansätze zur Lösung nichtlinearer Optimierungsprobleme die Suche nach energieeffizienten Getrieben mit einer optimalen Struktur unterstützen kann. Eukaryotic genomes are organized into chromatin, a highly dynamic complex of DNA and proteins, which plays a critical role in the regulation of gene expression. This thesis focuses on the study of a non-histone chromatin protein, the SET domain-containing H3K9 methyltransferase (HMTase) SU(VAR)3-9, and its role in the packaging and regulation of a euchromatic locus, the histone genes cluster (HIS-C). SU(VAR)3-9 was discovered in Drosophila melanogaster, but it is highly conserved from yeast to mammals. It has two conserved domains, the chromo- and the SET domains, and both are required for its function in gene silencing. The SET domain is responsible for the catalytic activity of SU(VAR)3-9, while the exact function of the chromo domain is still unclear. To gain an insight on the role(s) of SU(VAR)3-9 in the regulation of gene silencing, we first characterized a collection of Su(var)3-9 EMS-induced mutants that had been isolated in a genetic screen for strong, dominant suppressors of position-effect variegation (PEV). These mutants were characterized at the molecular, enzymatic, and cellular level, and their effect on gene silencing was also examined. We found that all mutants have single amino acid substitutions in the conserved preSET/SET/postSET domain, and that they all display a dramatic or complete loss of HMTase activity, strongly suggesting that suppression of PEV is linked to SU(VAR)3-9’s ability to methylate H3K9. The HIS-C is a natural, euchromatic target of SU(VAR)3-9, and mutations in Su(var)3-9 can alter its chromatin structure (Ner et al., 2002). To investigate the exact role(s) of SU(VAR)3-9 in the regulation of this locus, we analyzed the effects of a series of Su(var)3-9 missense mutants on the chromatin architecture of the HIS-C and on the expression of the histone genes. We detected a drastic reduction in the levels of H3K9me2 and HP1 associated with the his genes in all Su(var)3-9 missense mutants, although the mutant SU(VAR)3-9s still associate with the HIS-C. In addition, these mutants have elevated amounts of histone H2A and histone H3 RNA, suggesting that the enzyme function of SU(VAR)3-9 is critical for the regulation of the histone genes.

The present dissertation investigates similes and their importance in Eichendorff’s narrative work. The sources of the investigation consist of seven of Eichendorff’s narratives. Their 734 similes make up the corpus, which is presented in its entirety in the appendix. The context of the similes is partly included as well. Initially, I define the concept of “simile” more precisely, partly distancing myself from the definitions found in classical dictionaries of literary terms. After this, I describe my procedure for analysis in detail. This turned out to be necessary, since there was no similar study to be found on this topic in the extensive literature on Eichendorff. The search for models of types of similes brought me back to antique rhetoric as well as to Middle High German epic poems. In the first analysis, the types of similes occurring in the corpus are presented. The syntactic structures of image receivers and image givers are used as criteria. Four structures of similes occurred: a) classical similes and b) similes with image givers, which represent adverbial clauses and c) as / as if – clauses or are d) subject-related. The frequency and the development of frequency of types of similes are presented as well. In the second step of the thesis, I investigate whether similes tend to depict conditions/qualities or procedures/actions. It turned out that similes reflecting conditions/qualities, i.e. epic similes, dominated strongly. The high number of similes could possibly be explained by the functions carried out by epic similes in narrative texts. In the third part, I concern myself with the question whether the similes of the corpus are imaginative representations only and what kind of sensorial perceptions they express. Admittedly, the dominating percentage of the similes proved to be images, but more than fifteen per cent consist of sounds and other sensorial perceptions. Furthermore, imaginative similes, but also sounding similes express motion, so that they illustrate pictures in motion and sounding motion respectively. These come close to synaesthesia, whereas only five similes illustrate „pure“ synaesthesia. In contrast, subject-related similes are perceptions of different sensations and feelings, illustrating the inner life of a character not shared by any other character. Finally, the semantic content of the similes is investigated in order to determine the metamorphosis, i.e. the trope transfer from proprium to improprium. It turned out that only the classical simile originating in antique rhetoric is suitable for a semantic analysis. Above all, there is great variation in the trope transfer. The metamorphosis human being → nature dominates strongly, which makes the narrative text appear as a palimpsest, in which yet another world glimmer in front of the human being behind every character. However, the many trope transfers that convey reality → unreality could be interpreted as transitions and as a “magical code” of Eichendorff. Furthermore, the semantic analysis uncovers content and motives of classical similes. It becomes clear that pre-constructed – and only pre-constructed - content is imitated here. Consequently, it can be asserted that Eichendorff’s great number of similes constitute or at least contribute to the formulaic manner (according to Kohlschmidt) and the intertextuality (according to Nienhaus) in Eichendorff’s narrative work. Above all, the subject-related simile type turns out to be a typical representative of Romanticism because of its subjectivism. Together with its preformed semantic content, it constitutes the “romantic formula” of Eichendorff’s work.

In recent years, a focus issue of scientific computing in the performance of the computation. A creative development for the high-performance computing is CUDA which adopts GPU to be an auxiliary device for the numerical computation, especially, parallel computation. We therefore survey the elementary architecture of CUDA in this thesis. A numerical experiment in GPU by using Matlab with software Jacket for solving nonlinear optimization problem is demonstrated. Some numerical results are illustrated the efficiency of GPU.

The growing number of uncharacterised sequences in public databases has turned the prediction of protein function into a challenging research field. Traditional annotation methods are often error-prone due to the small subset of proteins with experimentally verified function. Goal of this thesis was to analyse the function and evolution of protein domains in order to understand molecular processes in the cell. The focus was on signalling domains of little understood function, as well as on functional sites of protein domains in general. Glucosaminidases (GlcNAcases) represent key enzymes in signal transduction pathways. Together with glucosamine transferases, they serve as molecular switches, similar to kinases and phosphatases. Little was known about the molecular function and structure of the GlcNAcases. In this thesis, the GlcNAcases were identified as remote homologues of N-acetyltransferases. By comparing the homologous sequences, I was able to predict functional sites of the GlcNAcase family and to identify the GlcNAcases as the first family member of the acetyltransferase superfamily with a distinct catalytic mechanism, which is not involved in the transfer of acetyl groups. In a similar approach, the sensor domain of a plant hormone receptor was studied. I was able to predict putative ligand-binding sites by comparing evolutionary constraints in functionally diverged subfamilies. Most of the putative ligand-binding sites have been experimentally confirmed in the meantime. Due to the importance of enzymes involved in cellular signalling, it seems impossible to find substitutions of catalytic amino acids that turn them catalytically inactive. Nevertheless, by scanning catalytic positions of the protein tyrosine phosphatase families, I found many inactive domains among single domain and tandem domain phosphatases in metazoan proteomes. In addition, I found that inactive phosphatases are conserved throughout evolution, which led to the question about the function of these catalytically inactive phosphatase domains. An analysis of evolutionary site rates of amino acid substitutions revealed a cluster of conserved residues in the apparently redundant domain of tandem phosphatases. This putative regulatory center might be responsible for the experimentally verified dimerization of the active and inactive domain in order to control the catalytic activity of the active phosphatase domain. Moreover, I detected a subgroup of inactive phosphatases, which presumably functions in substrate recognition, based on different evolutionary site rates within the phosphatase family. The characterization of these new regulatory modules in the phosphatase family raised the question whether inactivation of enzymes is a more general evolutionary mechanism to enlarge signalling pathways and whether inactive domains are also found in other enzyme families. A large-scale analysis of substitutions at catalytic positions of enzymatic domains was performed in this work. I identified many domains with inactivating substitutions in various enzyme families. Signalling domains harbour a particular high occurrence of catalytically inactive domains indicating that these domains have evolved to modulate existing regulatory pathways. Furthermore, it was shown that inactivation of enzymes by single substitutions happened multiple times independently in evolution. The surprising variability of amino acids at catalytic positions was decisive for a subsequent analysis of the diversity of functional sites in general. Using functional residues extracted from structural complexes I could show that functional sites of protein domains do not only vary in their type of amino acid but also in their structural location within the domain. In the process of evolution, protein domains have arisen from duplication events and subsequently adapted to new binding partners and developed new functions, which is reflected in the high variability of functional sites. However, great differences exist between domain families. The analysis demonstrated that functional sites of nuclear domains are more conserved than functional sites of extracellular domains. Furthermore, the type of ligand influences the degree of conservation, for example ion binding sites are more conserved than peptide binding sites. The work presented in this thesis has led to the detection of functional sites in various protein domains involved in signalling pathways and it has resulted in insights into the molecular function of those domains. In addition, properties of functional sites of protein domains were revealed. This knowledge can be used in the future to improve the prediction of protein function and to identify functional sites of proteins
Durch den rasanten Anstieg unbekannter Proteinsequenzen in öffentlichen Datenbanken ist die Vorhersage der Proteinfunktion zu einem herausfordernden Forschungsgebiet geworden. Herkömmliche Annotationsmethoden sind häufig fehlerhaft, da nur einem kleinen Teil der Proteine experimentell eine Funktion zugewiesen werden konnte. Ziel der hier vorliegenden Arbeit war es, die Funktion und Evolution von Proteindomänen in Hinblick auf die molekularen Vorgänge innerhalb der Zelle zu untersuchen. Der Schwerpunkt lag auf Signaldomänen mit unbekannter Funktion und auf funktionell wichtigen Positionen in Domänen. Glucosaminidasen (GlcNAcasen) spielen eine wichtige Rolle in Signaltransduktionswegen. Zusammen mit den Glucosamintransferasen dienen sie als molekulare Schalter, ähnlich den Kinasen und Phosphatasen, jedoch war sehr wenig über ihre molekulare Funktion, sowie über ihre Struktur bekannt. In dieser Studie wurde die entfernte Verwandtschaft der GlcNAcasen zu den Acetyltransferasen gezeigt. Durch den Vergleich von homologen Sequenzen konnte ich funktionelle Positionen vorhersagen und die GLcNAcasen als erstes Mitglied der Acetyltransferasen-Superfamilie mit einem neuen katalytischen Mechanismus identifizieren, der nicht den Transfer von Acetylgruppen vermittelt. In einem ähnlichen Ansatz wurde die Sensordomäne eines Hormonrezeptors aus Pflanzen untersucht. Dabei konnte ich durch den Vergleich von evolutiven Zwängen in funktionell unterschiedlichen Subfamilien Liganden-bindende Positionen bestimmen. Die meisten dieser Vorhersagen wurden inzwischen experimentell bestätigt. Aufgrund der entscheidenden Bedeutung von enzymatischen Domänen in Signaltransduktionsprozessen erscheint es unmöglich, Substitutionen von katalytischen Aminosäuren zu finden, die die Domäne inaktivieren würden. Dennoch habe ich in einer Analyse der katalytischen Positionen in der Proteintyrosinphosphatase-Familie viele inaktive Domänen in Einzel- und Tandem-Domänen-Phosphatasen in den Proteomen von Metazoa gefunden. Ich habe zusätzlich beobachtet, dass die inaktiven Domänen in der Evolution konserviert sind, was die Frage aufwirft, welche Funktion diese katalytisch inaktiven Domänen haben. Eine Analyse der Evolutionsraten von Aminosäuresubstitutionen identifizierte eine Ansammlung von konservierten Positionen in der scheinbar überflüssigen inaktiven Domäne von Tandemphosphatasen. Dieser möglicherweise regulatorische Bereich könnte für die Dimerisierung der aktiven und inaktiven Domäne verantwortlich sein, welche experimentell nachgewiesen wurde, sowie für die Regulation der katalytischen Aktivität der Phosphatasedomäne. Außerdem habe ich durch die unterschiedlichen Evolutionsraten eine Untergruppe der inaktiven Phosphatasen entdeckt, die wahrscheinlich an der Substraterkennung beteiligt ist. Die Charakterisierung dieser neuen regulatorischen Module in der Phosphatase- Familie führte zu der Frage, ob die Inaktivierung von Enzymen ein allgemeiner Mechanismus in der Evolution ist, um Signaltransduktionswege zu erweitern, und ob es auch in anderen Enzymfamilien inaktive Domänen gibt. Dazu wurde eine umfassende Analyse durchgeführt, um Substitutionen an katalytischen Positionen in enzymatischen Domänen zu untersuchen. Ich habe in vielen Domänen aus unterschiedlichen Enzymfamilien inaktivierende Substitutionen gefunden. Einen besonders hohen Anteil an katalytisch inaktiven Domänen gibt es in Signaldomänen, was zeigt, daß diese Domänen entstanden sind, um existierende regulatorische Netze zu modifizieren. Es konnte ferner gezeigt werden, daß die Inaktivierung von Enzymen durch einzelne Subsitutionen mehrmals unabhängig voneinander in der Evolution stattgefunden hat. Die Variabilität von Aminosäuren an katalytischen Positionen war ausschlaggebend für eine anschließende, allgemeinere Analyse von funktionellen Positionen. Mit Hilfe von funktionellen Positionen, die aus strukturellen Komplexen extrahiert wurden, konnte ich zeigen, dass funktionelle Positionen nicht nur in der Aminosäure, sondern auch in ihrer Lokalisation innerhalb der Struktur variieren. Im Laufe der Evolution haben sich Domänen aus Duplikationsprozessen gebildet, sich neuen Bindungspartnern angepasst und neue Funktionen entwickelt, was sich nun in der hohen Variabilität ihrer funktionellen Positionen widerspiegelt. Dennoch gibt es große Unterschiede zwischen Domänenfamilien. Die Analyse hat gezeigt, dass funktionelle Positionen von nuklearen Domänen viel stärker konserviert sind, als jene von extrazellulären Domänen. Die hier vorgestellte Studie beschreibt funktionelle Positionen in verschiedenen an Signaltransduktionswegen beteiligten Proteindomänen und liefert Einblicke in ihre molekulare Funktion. Außerdem wurden Eigenschaften von funktionell wichtigen Positionen aufgezeigt. Diese Erkenntnisse können in Zukunft zur Optimierung der Vorhersage von Proteinfunktionen und zur Identifikation von funktionellen Positionen genutzt werden

Women’s procreative capacities and the appreciation of birth experiences have always been recognised in civil society and the early days of psychology. Given that our culture is one that emphasises a woman’s capacity to bear children as one of the greatest social achievements, the social responsibility to procreate and ensure collective survival becomes a potent mandate, especially so for Black African women. To fulfil this social responsibility, traditional African culture dictates a very specific process of pregnancy which involves a series of watershed moments, each of which requires that social support, of whatever form, should be available. This study explored the role that social support from significant others and health professionals play in mediating psychological issues during pregnancy, childbirth and postnatally among Black African women in Madadeni Township in KwaZulu-Natal. The study was rooted in the interpretive, qualitative paradigm and a phenomenological research design was used. Purposive sampling was used to select participants who were aged from 18 to 25 years with a baby older than two weeks but less than six months. Semi structured interviews were conducted in the mother tongue of the participants until the point of saturation where no new information arose from the six participants interviewed. Thematic content analysis was used to extract recurrent themes across participants. The results indicate that social support, especially from parents and partners, plays a defining role in helping women to cope with the stress experienced during pregnancy, childbirth and postnatally.

Upon adoptive transfer into congenic histocompatible hosts, naïve CD8+ T cells stimulated ex vivo by TCR in the presence of IL-4 persist in the hosts and become long-lived memory cells. A unique subset of memory CD8+ T cells resides in the liver (TLM) and this subset is highly reduced in the IL-15Ralpha-knockout (ko) liver. TLM cells are similar to effector memory (TEM) cells in that they are both CD62LlowCCR7-, but are unlike TEM cells in that they express reduced IFN-gamma inducibility, cytolytic activity and TCR-induced proliferation. CFSE dilution assay results indicate that TLM cells undergo IL-15Ralpha-dependent proliferation. Immunofluorescent staining revealed that the TLM cells form a large number of cell clusters in the liver of WT but not IL-15Ralpha-ko mice. TLM cells form clusters through clonal expansion because adoptive transfer of an admixture of TCR+ IL-4-activated Vbeta8+ and Vbeta5+ CD8+ T cells results in clusters composed exclusively of Vbeta5+ or Vbeta8+ cells. Clonal expansion of CD8+ T cells is also found in the liver of Listeria monocytogenes-immune mice. TLM cell clusters in the liver are located around the sinusoid, closely associate with hepatic stellate cells, and are in close association with IL-15+ and IL-15Ralpha+ dendrite-like processes. Sorted CD62Llow TLM cells can migrate to other organs after re-transfer and display increased CD62L expression and become phenotypically similar to central memory (TCM) cells. Our results bring to light a previously unappreciated role of the liver in the growth and maintenance of memory CD8+ T cells. These results also indicate that clonal growth of memory CD8+ T cells in the liver is a normal physiological process and that caution should be exercised in interpreting focal lymphoid growth in clinical liver biopsy specimens as pathology and not normal physiology.

The subject of the present study is an analysis of five ditransitive verbs: give, lend, send, offer and show. The study focuses on the position of the two objects and on the factors that have an impact on the object ordering. An attempt is here made to provide a systematic overview of the position of the two objects with respect to their realization (i.e. substantival or pronominal). As regards the realization of the two objects, four types are distinguished: i. both Oi /Oprep and Od realized by nouns; ii. both Oi /Oprep and Od realized by pronouns; iii. Oi /Oprep realized by a noun and Od by a pronoun; iv. Oi /Oprep realized by a pronoun and Od by a noun. The position of the objects is assumed to be associated with the distribution of communicative dynamism or in other words with the principle of end-focus, i.e. that given information tends to precede new information. The second principle that operates in the ordering the two objects is the principle of end-weight. Of the three (or four, including intonation) factors whose interplay determines the FSP function of a clause element, in the case of ditransitive complementation the most important role is played by the contextual factor. Nicknames have been analysed, recorded and processed in many diverse ways by different languages, scholars and communities. In Zulu, many works of similar type have all been the size of an article up until 1999. This research on the subject is one of the first done in this depth. Nicknames form part of a Zulu person's daily life. They identify him/her more than the real or legal name. They shape him/her more than any other mode of address. They influence behaviour, personality, interaction based activities and the general welfare of an individual. They discipline, they praise, they mock too. Surprisingly, they are regarded as play items. They are even termed playnames (izidlaliso). But they are as serious as any item that makes an individual to be a significant figure in the community. They are unique in the sense that they stick more obstinately on the victim should he/she try to get rid of them. They are capable of staying for life. They only vanish to give others a chance to feature on the same individual. They are so poetic. A talented onomastician can tell a full story about an individual without him grabbing what is being said about him just because the story is spiced with just a single figurative nickname. They haunt the whole arena of the parts of speech in a language, especially the Zulu language. They modify the well known meaning of words into special references that paint in bright colours the character of an individual. Zulu nicknames processes visit all possible languages and adapt items from into Zuluised special terms that a capable of inheriting an onomastic status. They originate even from the most sensitive sources like people's private lives. The only challenging area about nicknames is that bearers do not want to expose them to peale who are not known to them, even if they do not fall into a category of nicknames for ridicule. Finally, nicknames have been exposed here as linguistic items that organise the community into makers and bearers, and then users of nicknames.

Der Schwerpunkt der vorliegenden Arbeit liegt auf Methoden zur Lösung nichtlinearer Optimierungsprobleme mit der Anforderung, jedes globale Optimum garantiert zu finden und mit einer im Voraus festgesetzten Genauigkeit zu approximieren. Eng verbunden mit dieser deterministischen Optimierung ist die Berechnung von Schranken für den Wertebereich einer Funktion über einem gegebenen Hyperquader. Verschiedene Ansätze, z. B. auf Basis der Intervallarithmetik, werden vorgestellt und analysiert. Im Besonderen werden Methoden zur Schrankengenerierung für multivariate ganz- und gebrochenrationale Polynome mit Hilfe der Darstellung in der Basis der Bernsteinpolynome weiterentwickelt. Weiterhin werden in der Arbeit schrittweise die Bausteine eines deterministischen Optimierungsverfahrens unter Verwendung der berechneten Wertebereichsschranken beschrieben und Besonderheiten für die Optimierung polynomialer Aufgaben näher untersucht. Die Analyse und Bearbeitung einer Aufgabenstellung aus dem Entwicklungsprozess für Fahrzeuggetriebe zeigt, wie die erarbeiteten Ansätze zur Lösung nichtlinearer Optimierungsprobleme die Suche nach energieeffizienten Getrieben mit einer optimalen Struktur unterstützen kann.
