PURPOSE: Ten years after the first clinical studies, the clinical impact of myeloid growth factors in acute myeloid leukemia is still unclear. One of the objectives of the Groupe Ouest-Est Leucémies Aigues Myeloblastiques (GOELAM) 2 trial was to evaluate the benefit of granulocyte colony-stimulating factor (GCSF) given only after the two courses of intensive consolidation chemotherapy (ICC) used to maintain complete remission (CR). PATIENTS AND METHODS: One hundred ninety-four patients who were in CR after induction treatment were randomly assigned to receive G-CSF (100 patients) or no G-CSF (94 patients) after two courses of ICC (ICC 1, high-dose cytarabine plus mitoxantrone; ICC 2, amsacrine plus etoposide). G-CSF (filgrastim) was administered from the day after chemotherapy until granulocyte recovery at a daily dose of 5 μg/kg. RESULTS: In the G-CSF group, the median duration of neutropenia (< 0.5 × 109/L) was dramatically reduced, both after ICC 1 (12 v 19 days, P < .001) and after ICC 2 (20 v 28 days, P < .001). The median duration of hospitalization was also significantly shorter in the G-CSF group (24 v 27 days after ICC 1, P < .001; 29 v 34 days after ICC 2, P < .001). The median duration of intravenous antibiotics was significantly reduced after ICC 1 and ICC 2, and the median duration of antifungal therapy was significantly reduced after ICC 1. However, the incidence of microbiologically documented infections, the toxic death rate, the 2-year disease-free survival, and the 2-year overall survival were not affected by G-CSF administration. Moreover, the median interval between ICC1 and ICC2 was reduced by only 2 days, and the number of patients undergoing ICC2 was not increased in the G-CSF arm. CONCLUSION: G-CSF should be administered routinely after ICC to reduce the duration of neutropenia and hospitalization. However, G-CSF did not seem to significantly increase the feasibility of this two-course program or modify overall outcome.

Several researchers have recommended that level-specific fit indices should be applied to detect the lack of model fit at any level in multilevel structural equation models. Although we concur with their view, we note that these studies did not sufficiently consider the impact of intraclass correlation (ICC) on the performance of level-specific fit indices. Our study proposed to fill this gap in the methodological literature. A Monte Carlo study was conducted to investigate the performance of (a) level-specific fit indices derived by a partially saturated model method (e.g., [Formula: see text] and [Formula: see text]) and (b) [Formula: see text] and [Formula: see text] in terms of their performance in multilevel structural equation models across varying ICCs. The design factors included intraclass correlation (ICC: ICC1 = 0.091 to ICC6 = 0.500), numbers of groups in between-level models (NG: 50, 100, 200, and 1,000), group size (GS: 30, 50, and 100), and type of misspecification (no misspecification, between-level misspecification, and within-level misspecification). Our simulation findings raise a concern regarding the performance of between-level-specific partial saturated fit indices in low ICC conditions: the performances of both [Formula: see text] and [Formula: see text] were more influenced by ICC compared with [Formula: see text] and SRMRB. However, when traditional cutoff values ( RMSEA≤ 0.06; CFI, TLI≥ 0.95; SRMR≤ 0.08) were applied, [Formula: see text] and [Formula: see text] were still able to detect misspecified between-level models even when ICC was as low as 0.091 (ICC1). On the other hand, both [Formula: see text] and [Formula: see text] were not recommended under low ICC conditions.

Aims: To evaluate the carotid ultrasound intra- and interobserver agreements in a common clinical scenario when making manual measurements of the intima-media thickness (IMT) and peak systolic (PSV) and end diastolic (EDV) velocities in the common (CCA) and the internal carotid (ICA) arteries. Material and methods: Three different experienced operators per- formed two time-point carotid ultrasounds in 21 patients with cardiovascular risk factors. Each operator measured freehand the CCA IMT three consecutive times in each examination. The CCA and ICA hemodynamic parameters were acquired just once. For our purpose we took the average (IMTmean) and maximum (IMTmax) IMT values. Quantitative variables were analyzed with the t-student, and ANOVA test. Agreements were evaluated with the Intraclass Correlation Coefficient (ICC). Results: IMTmean intraobserver agreement was better on the left (ICC: 0.930-0.851-0.916, operators 1-2-3) than on the right (ICC: 0.789-0.580-0.673, operators 1-2-3). IMTmax agreements (Left ICC: 0.821-0.723-0.853, operators 1-2-3; Right ICC: 0.669- 0.421-0.480, operators 1-2-3) were lower and more variable. Interobserver agreements for IMTmean (ICC: 0.852-0.860; first-second ultrasound) and IMTmax (ICC: 0.859-0.835; first-second ultrasound) were excellent on the left, but fair-good and more variable on the right (IMTmean; ICC: 0.680-0.809; first-second ultrasound; IMTmax; 0.694-0.799; first-second ultra- sound). Intraobserver agreements were fair-moderate for PSVs and good-excellent for EDVs. Interobserver agreements were good-excellent for both PSVs and EDVs. Overall, 95% confidence intervals were narrower for the left IMTmean and CCA velocities. Conclusions: Intra and interobserver agreements in carotid ultrasound are variable. In order to improve carotid IMT agreements, IMTmean is preferable over IMTmax.

Abstract Personality has been linked to risk of dementia. Most studies ask individuals to rate their own personality traits or for a knowledgeable informant to perform the rating; few collect data from both. When informants are asked to give an estimate of the patient’s lifelong personality traits, they often describe personality before disease onset. When asked to self-rate, patients may instead assess their personality as they see themselves, providing a personality-state measure. The goal of this study was to assess agreement between two independent measures of personality and evaluate whether stage of cognitive impairment and characteristics of patients or caregivers impact concordance. In 79 consecutive patient-caregiver dyads presenting to our center (mean age:76.8±8.4; 44.1% female; 6% cognitively normal, 41% MCI; and 53% dementia) with in-depth psychosocial and neuropsychological evaluations, we found informants rated patients lower on openness (O) (ICC=0.434; 95%CI: 0.235-0.598) and agreeableness (A) (ICC=0.491; 95%CI: 0.302-0.643) and higher on extraversion (O) (ICC=0.396; 95%CI: 0.191-0.568) and neuroticism (N) (ICC=0.444; 95%CI: 0.247-0.607). Greater discordance was observed in established dementia (ICCE=0.497; 95%CI: 0.222-0.700; ICCA=0.337; 95%CI:0.031-0.586; ICCN=0.422; 95%CI: 0.191-0.683), compared with MCI (ICCO=0.568; 95%CI: 0.282-0.762). We explored the effect of patient and caregiver mood and caregiver burden on personality ratings. Although personality is typically described as a trait, we present evidence that in the eyes of patients, personality ratings may represent a state that changes across the spectrum of cognitive impairment. Understanding how patients and caregivers differentially perceive personality may assist in developing novel psychotherapeutic interventions and approaches dealing with behavioral manifestations of dementia.

Abstract The International Criminal Court (ICC or the Court) is currently suffering from an evidence problem. Since the time the ICC became functional, some judges have criticized the investigative practices of the Office of the Prosecutor (OTP), and some cases have crumbled due to the quality of evidence. This article tackles these problems from the perspective of third-party investigations and their role in ICC cases. It examines the difference between private and public investigators under the legal framework of the Rome Statute to find whether and how third-party investigations can be used in (and by) the ICC. Evidence collected by third-party investigators will likely face challenges of admissibility in being introduced into trial. Those challenges could, however, be overcome if third-party investigations are regulated within the legal framework of the ICC. Ultimately, this article aims to provide a legal framework to optimize the use of third-party investigations in the ICC.

Interstitial cells of Cajal (ICC) have important functions in regulation of motor activity in the gastrointestinal tract. In murine small intestine, ICC are gathered in the regions of the myenteric plexus (ICC-MY) and the deep muscular plexus (ICC-DMP). These two classes of ICC have different physiological functions. ICC-MY are pacemaker cells and generate the slow-wave electrical rhythmicity of gastrointestinal organs. ICC-DMP form synaptic connections with the varicose nerve terminals of enteric motor neurons and are involved in reception and transduction of motor neurotransmission. Gene expression underlying specific functions of ICC classes is incompletely understood. In the present study, we used recently developed highly selective techniques to isolate the two functional ICC classes from enzymatically dispersed intestinal muscles by fluorescence-activated cell sorting. The transcriptomes of ICC-MY and ICC-DMP were investigated using oligonucleotide microarray analysis. Differential expression of functional groups of genes defined by standard gene ontology terms was also studied. There were substantial numbers of genes expressed more abundantly in ICC than in the tunica muscularis, and we also detected marked phenotypic differences between ICC-MY and ICC-DMP. Notably, genes related to cell junction, process guidance, and vesicle trafficking were upregulated in ICC. Consistent with their specific functions, metabolic and Ca2+ transport genes were relatively upregulated in ICC-MY, whereas genes for signaling proteins involved in transduction of neurotransmitter functions were relatively upregulated in ICC-DMP. Our results may lead to the identification of novel biomarkers for ICC and provide directions for further studies designed to understand ICC function in health and disease.

Interstitial cells of Cajal (ICC) play a vital role in the gastrointestinal motility. However, information on ICC in lower vertebrates is rare. Here, ICC and ICC-like features of the gastric wall in the bullfrog (Rana catesbeiana) were observed by light microscopy and transmission electron microscopy. The lengths and distances of the ICC/ICC-like features were measured by morphometric analysis. The gastric wall contained mucosa, submucosa, tunica muscularis, and serosa. The gastric glands contained mucous cells and oxynticopeptic cells. The ICC with 1–3 processes were located among smooth muscle cells (SMC) of the tunica muscularis. Moreover, the ICC-like features were observed among oxynticopeptic cells of the mucosa. The processes of ICC established direct contacts with SMC. Also, the gap junctions were observed between the processes of ICC and nerve fiber bundles in the tunica muscularis. The multivesicular bodies, including shedding exosomes, were frequently observed between ICC and SMC. In addition, ICC-like features and their processes were observed in close proximity to oxynticopeptic cells and blood vessels. Our findings illustrated that ICC are present in the gastric tunica muscularis, and ICC-like features were in the mucosal lamina propria of the gastric wall of R. catesbeiana. These histological evidences supported the notion that ICC are implicated in gastric motility.

Cílem studie je představit výzkumné šetření v oblasti hodnocení rozvoje interkulturní komunikační kompetence (dále ICC1) u studentů učitelství anglického jazyka pro základní školy v rámci studijního předmětu Intercultural Communicative Competence. Fenomén interkulturality klade nové nároky nejen na pedagogickou praxi, ale i výzkum. Zaměřujeme se tedy na tematiku interkulturní výchovy a vzdělávání s klíčovým cílem rozvoje ICC. Předpokladem pro výzkumné uchopení problematiky je konceptualizace konstruktu ICC a jeho následná didaktizace, tj. rozvoj a hodnocení. V rámci realizovaného výzkumného šetření jsme využili kvantitativního i kvalitativního přístupu ke sběru i analýze dat; jako techniky sběru dat byl využit dotazník YOGA Form a ohniskové skupiny, jejichž transkripty byly podrobeny obsahové analýze. Výzkumný soubor tvořilo 17 studentů učitelství anglického jazyka (15 žen a 2 muži; průměrný věk: 24 let). Cílem výzkumného šetření bylo odhalit, zda u studentů na základě absolvování studijního předmětu Intercultural Communicative Competence došlo k rozvoji jednotlivých dimenzí ICC a pokud ano, tak jakých dimenzí a v jaké míře. Výsledky výzkumu potvrdily, že u studentů došlo k rozvoji všech dimenzí ICC, avšak v různé míře. Signifikantní rozvoj byl zaznamenán v oblasti dimenzí znalostí a dovedností. V rámci dimenze povědomí a postojové pak došlo k iniciaci rozvojových tendencí, což s ohledem na obtížnost jejich didaktizace hodnotíme kladně, jelikož i podpora rozvoje reflektivního potenciálu studentů ve vztahu k rozvoji ICC (i iniciaci toho rozvoje) hraje důležitou roli. Na základě výsledků výzkumu diskutujeme možné implikace těchto výstupů, tj. zejména rozvoj dimenze znalostní a dovednostní, pro teorii i praxi.

Specific functions of interstitial cells of Cajal (ICC) have been linked to distinct classes that differ by morphology and distribution. In the small intestine, slow wave-generating ICC are located in the myenteric region (ICC-MY), whereas ICC that mediate neuromuscular neurotransmission occur either throughout the circular muscle layer (intramuscular ICC, ICC-IM) or in association with the deep muscular plexus (ICC-DMP). Selective isolation of ICC to characterize specific properties has been difficult. Recently, neurokinin-1 receptors have been detected in murine ICC-DMP and neurons but not in ICC-MY. Here we identified and isolated ICC-DMP/IM by receptor-mediated internalization of fluorescent substance P and Kit immunofluorescence. Specificity of labeling was verified by confocal microscopy. Mouse and human ICC-DMP/IM were detected in suspension by fluorescent microscopy and harvested for RT-PCR with micropipettes. The isolated cells expressed Kit but not markers for neurons, smooth muscle, or antigen-presenting cells. ICC-DMP expressed neurokinin-1 receptor, M2 and M3 muscarinic receptors, P2Y1 and P2Y4 purinergic receptors, VIP receptor 2, soluble guanylate cyclase-1 subunits, and protein kinase G. L- or T-type Ca2+ channels were not detected in these cells. ICC-MY and ICC-DMP were simultaneously detected and enumerated by flow cytometry and sorted to purity by fluorescence-activated cell sorting. Morphological changes of interstitial cells of Cajal (ICC) have been proposed to characterize motility disorders. However, a global view of the network orientations of ICC subgroups has not been established to illustrate their three-dimensional (3-D) architectures in the human colon. In this research, we integrate c-kit immunostaining, 3-D microscopy with optical clearing, and image rendering to present the location-dependent network orientations with high definition. Full-depth colonic tissues were obtained from colectomies performed for nonobstructing carcinoma. Specimens of colon wall were prepared away from the tumor site. C-kit and nuclear fluorescent staining were used to identify the ICC processes and cell body. Optical clearing was used to generate transparent colon specimens, which led to panoramic visualization of the fluorescence-labeled ICC networks at the myenteric plexus (ICC-MY), longitudinal (ICC-LM) and circular (ICC-CM) muscles, and submucosal boundary (ICC-SM) up to 300 μm in depth via confocal microscopy with subcellular level resolution. We observed four distinct network patterns: 1) periganglionic ICC-MY that connect with ICC-LM and ICC-CM, 2) plexuses of ICC-LM within the longitudinal muscle and extending toward the serosa, 3) repetitive and organized ICC-CM layers running parallel to the circular muscle axis and extending toward the submucosa, and 4) a condensed ICC-SM layer lining the submucosal border. Among the four patterns, the orderly aligned ICC-CM layers provide an appropriate target for quantitation. Our results demonstrate the location-dependent network orientations of ICC subgroups and suggest a practical approach for in-depth imaging and quantitative analysis of ICC in the human colon specimen.

Aim: Integrated analysis of genomics, epigenomics, transcriptomics and clinical information contributes to identify specific molecular subgroups and find novel biomarkers for pancreatic cancer. Materials & methods: The DNA copy number variation, the simple nucleotide variation, methylation and mRNA data of pancreatic cancer patients were obtained from The Cancer Genome Atlas. Four molecular subgroups (iC1, iC2, iC3 and iC4) of pancreatic cancer were identified by integrating analysis. Results: The iC1 subgroup harbors better prognosis, higher immune score, lesser DNA copy number variation mutations and better genomic stability compared with iC2, iC3 and iC4 subgroups. Three new genes ( GRAP2, ICAM3 and A2ML1) correlated with prognosis were identified. Conclusion: Integrated multi-omics analysis provides fresh insight into molecular classification of pancreatic cancer, which may help discover new prognostic biomarkers and reveal the underlying mechanism of pancreatic cancer.

Depletion of interstitial cells of Cajal (ICC) networks is known to occur in several gastrointestinal motility disorders. Although confocal microscopy can effectively image and visualize the spatial distribution of ICC networks, current descriptors of ICC depletion are limited to cell numbers and volume computations. Spatial changes in ICC network structural properties have not been quantified. Given that ICC generate electrical signals, the organization of a network may also affect physiology. In this study, six numerical metrics were formulated to automatically determine complex ICC network structural properties from confocal images: density , thickness , hole size , contact ratio , connectivity and anisotropy . These metrics were validated and applied in proof-of-concept studies to quantitatively determine jejunal ICC network changes in mouse models with decreased ( 5-HT 2 B receptor knockout (KO)) and normal ( Ano1 KO) ICC numbers, and during post-natal network maturation. Results revealed a novel remodelling phenomenon occurring during ICC depletion, namely a spatial rearrangement of ICC and the preferential longitudinal alignment. In the post-natal networks, an apparent pruning of the ICC network was demonstrated. The metrics developed here enabled the first detailed quantitative analyses of structural changes that may occur in ICC networks during depletion and development.

Interstitial cells of Cajal (ICC) have been proposed as stretch receptors for vagal afferent nerves in the stomach based on immunohistochemical studies. The aim of the present study was to use electron microscopy and the anterograde degeneration technique to investigate ultrastructural features and survival dependency of ICC associated with vagal afferent innervation of the cat esophagus. This is the first report on the ultrastructural characteristics of ICC in the cat esophagus. Intramuscular ICC (ICC-IM) were identified throughout the musculature, whereas ICC in the myenteric plexus were rare. ICC-IM were particularly numerous in septa aligned with smooth muscle bundles. They were in synapse-like contact with nerve varicosities and in gap junction contact with smooth muscle cells. Smooth muscle cells also made contact with ICC through peg and socket junctions. Precision damage through small-volume injection of saline in the center of the nodose ganglion from the lateral side, known to selectively affect sensory nerves, was followed within 24 h by degeneration of a subset of nerve varicosities associated with ICC-IM, as well as degeneration of the associated ICC-IM. Smooth muscle cells were not affected. Nerves of Auerbachs plexus and associated ICC were not affected. In summary, ICC-IM aligning the esophageal muscle bundles form specialized synapse-like contacts with vagal afferent nerves as well as gap junction and peg-and-socket contacts with smooth muscle cells. This is consistent with a role of ICC-IM as stretch receptors associated with vagal afferent nerves; the ICC-vagal nerve interaction appears essential for the survival of the ICC.

Background/Aims: Anoctamin1 (Ca2+-activated Cl- channel, ANO1) is a specific marker of the interstitial cells of Cajal (ICC) in the gastrointestinal tract, and are candidate proteins that can function as pacemaker channels. Recently, novel selective ANO1 inhibitors were discovered and used to study Ca2+-activated Cl- channels. Therefore, to investigate whether ANO1 channels function as pacemaker channels, selective ANO1 inhibitors were tested with respect to the pacemaker potentials in ICC. Methods: Whole-cell patch-clamp recording, RT-PCR, and intracellular Ca2+ ([Ca2+]i) imaging were performed in cultured ICC obtained from mice. Results: Though CaCCinh-A01 (5 µM), T16Ainh-A01 (5 µM), and MONNA (5 µM) (selective ANO1 inhibitors) blocked the generation of pacemaker potentials in colonic ICC, they did not do so in small intestinal ICC. Though nifulmic acid (10 µM) and DIDS (10 µM) (classical Ca2+-activated Cl- channel inhibitors) also had no effect in small intestinal ICC, they suppressed the generation of pacemaker potentials in colonic ICC. In addition, knockdown of ANO1 reduced the pacemaker potential frequency in colonic ICC alone. Though ANO1 inhibitors suppressed [Ca2+]i oscillations in colonic ICC, they did not do so in small intestinal ICC. T-type Ca2+ channels were expressed in the both the small intestinal and colonic ICC, but mibefradil (5 µM) and NiCl2 (30 µM) (T-type Ca2+ channel inhibitors) inhibited the generation of pacemaker potentials in colonic ICC alone. Conclusion: These results indicate that though ANO1 and T-type Ca2+ channels participate in generating pacemaker potentials in colonic ICC, they do not do so in small intestinal ICC. Therefore, the mechanisms underlying pacemaking in ICC might be different in the small intestine and the colon.

This study aims to measure the validity and reliability construct satisfaction and quality of library services based on the user using LIBQUAL (library quality) method with a touch of marketing management. Data analysis by Cronbach Alpha and Pearson Correlation. The research was conducted in Tanjungpura University (UNTAN) by taking samples of visitors (students) at six (6) faculty libraries namely three (3) social faculty of 150 people and three (3) science faculty of 150 people. The sample is chosen by purposive sampling method. Reliability test results show that the five US, IC, LP, LQ and KU variables show high instrument reliability. The result of pearson correlation test shows that the measurement instrument used has problem validity on some question items. At minimum service item with correlation value smaller than 0.30 is IC2, IC5 and LQ5 so it is declared invalid. At service expectation items with correlation value smaller than 0.30 ie AS8, IC1, IC2, IC4, LP1, KU1, KU2, KU4, and KU8. And at present service items with correlation values smaller than 0.30 ie IC1, IC3, LP1, LP3, LQ1, LQ3, LQ4 and KU1.

In the 21st century, intercultural communication is inevitable owing to cutting-edge technologies and transportation. Thus, over the past decades, there has been an ever-increasing interest in and attention to intercultural communicative competence (ICC) in English language teaching and learning. This study reports findings from a systematic research synthesis of 20 empirical studies regarding learning and teaching ICC in the Asian English as a foreign language (EFL) context. The research questions incl ude: 1) In what contexts was ICC investigated?; 2) Who were the participants in the reviewed studies?; 3) What kinds of ICC topics were explored?; 4) What activities and tasks were involved in ICC teaching and learning?; 5) What suggestions are recommended for future research on ICC? The findings expand upon learning contexts, participants, ICC-related topics, activities involved in ICC teaching and learning. Furthermore, this paper provides preservice and inservice teachers with valuable insights as well as suggestions and recommendations for teaching ICC in teacher education. This systematic research synthesis would be of value in developing and improving ICC-focused teaching and learning in English education.

Bulk RNA-seq data of 255 ICC patients were analyzed. Infiltration levels of SPP1+CD68+ tumor-associated macrophages (TAMs) were examined by dual immunofluorescence (IF) staining in 264 resected ICC samples. The correlation between SPP1+ TAMs and clinicopathological features as well as their prognostic significance was evaluated.ResultsAmong the 10 patients, five received biopsy at baseline, and others were biopsied at different timings following ICBs. Single-cell transcriptomes for 5,931 cells were obtained. A tighter cellular communication network was observed in ICB-treated ICC. We found a newly emerging VEGF signaling mediated by PGF-VEGFR1 between cancer-associated fibroblasts (CAFs) and endothelial cells in ICC following ICBs. SPP1 expression was dramatically upregulated, and SPP1+ TAM gene signatures were enriched in TAMs receiving ICB therapy. We also identified SPP1+ TAMs as an independent adverse prognostic indicator for survival in ICC.ConclusionOur analyses provide an overview of the altered tumor ecosystem in ICC treated with ICBs and highlight the potential role of targeting CAFs and SPP1+TAMs in developing a more rational checkpoint blockade-based therapy for ICC.
