Target identification has been considered as a chief parameter in drug discovery as it fully characterizes on-target and off-target effects of drug binding. Cell signaling receptors, structural proteins, and post-translational modifications of histones by histone deacetylases are the most widespread targets that are progressively being explored. The FDA approved histone deacetylases inhibitors and the majority of HDACi in and out of clinical trials based on the activities of 11 isoforms of the enzyme in non-selective influence approach. Unfortunately, reported HDACi does not possess a high degree of structural specificity and ultimately lessens the therapeutic index with many dose limiting toxicities. This chapter illustrates how different approaches are incorporated into the novel inhibitors discovery that are truly isoform-selective and which are specifically involved in targeting only a particular isozyme. Further, it highlights the aspects relating to provide a wider therapeutic index with an improved toxicity profile of lead like epigenetic modulators.

Phthalates are a group of manmade chemicals which may be regarded as heroes having profound application in industries belongs to dieters of 1, 2-benzenedicarboxylic acid (phthalic acid). They are mainly used as plasticizer. Phthalates may be of high and low molecular weight, and have find their existence in the manufacturing of various products that are of daily use to humans and pose threat to non-occupational humans besides of those working in the industries where phthalate based products are manufactured. Phthalates are now widely used and therefore it is difficult to avoid their exposure; thus, its adverse effects are very certain. The literature present revealed that phthalate exposure has made alteration both to females and males, like reproductive alterations: including damaging the sperm quality, semen concentration and sperm DNA, breast cancer in females, AGD, endocrine disruption. The phthalate exposure has been shown to alter the behavior, which is an indication of nervous system damage. Phthalate exposure has been revealed to affect the respiration. Phthalate after exposure has been found to get metabolized in various end points and their detection by various techniques has made an insight towards understanding the mechanism of phthalates toxicity. Phthalates both of low and high molecular weight have found their way to fauna and created a mess with the physiology of animals or humans. Thus, precaution is better, as said prevention is better than cure. The suggestive remedy for phthalate exposure is to make less use of products that contain phthalate material, more importantly during pregnancy and developmental stage of infants. Utmost care has been taken in regard to the literature cited. The data included in this chapter has been taken from those research articles with indexing of Science Citation Index Expanded, Scopus as for authenticity, and also using the Web of Science Database, which provides interactive citation.

Titanium dioxide (TiO2), owing to its non-toxicity, chemical stability, and low cost, is one of the most valuable ceramic materials. TiO2 derived coatings not only act like a ceramic protective shield for the metallic substrate but also provide cathodic protection to the metals against the corrosive solution under Ultraviolet (UV) illumination. Being biocompatible, TiO2 coatings are widely used as an implant material. The acid treatment of TiO2 promotes the attachment of cells and bone tissue integration with the implant. In this chapter, the applications of TiO2 as a corrosion inhibitor and bioactive material are briefly discussed. The semiconducting nature and high refractive index of TiO2 conferred UV shielding properties, allowing it to absorb or reflect UV rays. Several studies showed that a high ultraviolet protection factor (UPF) was achieved by incorporating TiO2 in the sunscreens (to protect the human skin) and textile fibers (to minimize its photochemical degradation). The rutile phase of TiO2 offers high whiteness, and opacity owing to its tendency to scatter light. These properties enable TiO2 to be used as a pigment a brief review of which is also addressed in this chapter. Since TiO2 exhibits high hardness and fracture toughness, the wear rate of composite is considerably reduced by adding TiO2. On interacting with gases like hydrogen at elevated temperatures, the electrical resistance of TiO2 changes to some different value. The change in resistance can be utilized in detecting various gases that enables TiO2 to be used as a gas sensor for monitoring different gases. This chapter attempts to provide a comprehensive review of applications of TiO2 as an anti-corrosion, wear-resistant material in the mechanical field, a UV absorber, pigment in the optical sector, a bioactive material in the biomedical field, and a gas sensor in the electrical domain.

In particular, it lacks an open-source, interactive interface for measuring the health risks associated with sourcing, manufacturing, selecting, installing, using, maintaining, and disposing of building-based polymers. Because of their high degree of chemical synthesis, polymers are typically more toxic than wood, glass, or concrete; yet architects, engineers, builders, clients, and the general public remain poorly informed about the deadly accumulation of synthetic polymers that originate in the building industry and that pervade our air, water, and bodies. This question should be central to the very definition and practice of life-cycle assessment, and this chapter outlines a process for developing an industry-based life-cycle index of human health in building (LCI-HHB). After all, traditional LCAs are of little help to anyone not healthy enough to enjoy them.

Broad-spectrum antibacterial agents kill most bacteria including gram-positive rods and cocci, gram-negative rods and cocci, and often anaerobes too. Narrow-spectrum agents kill a narrow range of microbes, e.g. benzylpenicillin is mainly active against gram-positive cocci. By and large a narrow-spectrum antimicrobial is less likely to disrupt a patient’s normal flora than a broad-spectrum agent. Hence, if the likely organism is causing an infection it is best to give a narrow-spectrum antimicrobial to treat that specific organism. If a patient presents ‘septic’ and the source of infection is unknown, relevant cultures should be taken followed by broad-spectrum antimicrobial cover. This can later be modified either when the source of infection is found or as a result of microbiology culture results. ● Agents mostly active against gram-positive bacteria include: ■ Penicillin (Also active against Neisseria spp.). ■ Fusidic acid. ■ Macrolides (Also active against Legionella, Campylobacter, Bordetella spp.). ■ Clindamycin. ■ Glycopeptides. ■ Oxazolidinones. ■ Streptogramins. ● Agents mainly active against gram-negative bacteria include: ■ Polymyxin. ■ Trimethoprim. ■ Aminoglycosides (also active against staphylococci and show synergy when combined with beta-lactams against/glycopeptides against streptococci). ■ Monobactams. ■ Temocillin. ● Broad-spectrum antimicrobials include: ■ Beta-lactam plus beta-lactamase inhibitor combinations. ■ Cephalosporins. ■ Carbapenems. ■ Chloramphenicol, Tetracyclines/Glycyclines. A bactericidal agent is a compound that actively kills multiplying bacteria. A bacteriostatic compound inhibits the growth of bacteria. Whether or not an antimicrobial is bactericidal or bacteriostatic depends on a variety of things, including the type of agent, its concentration, and the organism it is being used to treat. It is especially important to try and use a bactericidal agent if the patient’s immune system is impaired or the infection is at a site where it is difficult for the immune system to access, e.g. the heart valves in bacterial endocarditis, the meninges in meningitis. Examples of each are given here: ● Bactericidal agents include beta-lactams, glycopeptides, fluoroquinolones, and aminoglycosides. ● Bacteriostatic agents include macrolides, clindamycin, tetracyclines, trimethoprim, and sulphonamides. The therapeutic index of a drug is the ration of the concentration of drug likely to be toxic to the patient divided by the concentration of drug likely to be clinically effective.
