Resumen
The translation and connotation of landscape architecture (LA) caused a lot of controversy in academia when the term ‘LA’ was introduced to China. In this study, we summarized the different opinions of Chinese scholars based on the origin, evolution, and professional contents of LA, which can be divided into ‘Jing Guan Jian Zhu’ (Chinese Pinyin), ‘Jing Guan She Ji’, and ‘Feng Jing Yuan Lin’. Finally, this article provides strategies and suggestions for enhancing the development of LA when the first-level discipline is established in China, and the aim is to narrow the gap with the international community.

Resumen
The papers are dedicated to the concepts, main ideas, texts, and forms of practice of the syncretic Taoist movement Chongxuan-pai – “Twofold mystery”, which was developed in the 7–10th centuries. This school borrowed a number of logical constructions of Madhyamika Buddhism, including the system of four-level dialectical negation, as well as the idea of absolute “emptying of consciousness” for overcoming attachments both to mundane life and to any mental concepts. In part, these ideas are reflected in the Taoist-Buddhist practice of attaining “purity and quietness, a specific tradition of meditation, with its most important text “Canon of Purity and Quietness” (Qingjing jing) (given in this article in the author’s translation). Despite its traditional structure, thematic allusions with “Dao De Jing” and precepts attributed to Lao-jun, this Canon pays special attention to the technique of “inner contemplation” (nei guan), which also gravitates to the Buddhist practice of vipasyana and is called to eliminate binary opposition (pure-polluted, movement-rest) in the practitioner’s consciousness. The “Canon of purity and Quietness” is still highly revered in central China’s Taoist schools today. The basis of the meditative practice according to this Canon is the gradual ascending from “looking inward”, “looking outward” and “looking away” to “contemplation of emptiness”. The highest stage of “emptying the emptiness” leads to the complete deactualization of the ego and the breaking of all ties with the world. In this way, the exegetical idea of the “Twofold mystery” is realized as a form of Taoist practice influenced by Buddhist ideas.

Abstract In the 11th century, the dissemination of Buddhism in the territories of Northern Song 北宋 dynasty (960–1127) China, Khitan 契丹 Liao 遼 (907/916–1125), and Xixia 西夏 (Tangut 党項, 1038–1227) kingdoms each reached a peak. What united the learned peoples of these three kingdoms in terms of religious and intellectual development was the comparatively widespread study and adoption of the teachings of Huayan Buddhism, or studies of and commentaries to the translations of the Avataṃsaka-sūtra into Chinese in 60-, 80-, and 40-rolls (Huayan jing 華嚴經, esp. T nos. 278, 279, 293). In this article I address some of my earlier research concerning two treatises composed by Bensong 本嵩 (active ca. 1083–1085), the Huayan guan tongxuan ji 華嚴觀通玄記 (Record of the Profundities of Total Meditative Insight [or Contemplation] of the Gaṇḍavyūha chapter of the Avataṃsaka-sūtra Flower Garland Sūtra) and Fajie guan sanshimen song 法界觀三十門頌 (Verses Praising the Thirty Contemplative Approaches or Gates presented in the Gaṇḍavyūha Chapter on Entry into the Realm of Reality), among other examples from the period, to illustrate how these Huayan teachings were actually the product of Huayan and Chan Buddhist ideological frameworks, which fruitfully can be called Huayan-Chan. In order to demonstrate why the rubric ‘Huayan-Chan’ can be productive, I examine a range of commentarial Buddhist texts in Chinese to show who the patriarchs of Huayan-Chan were considered to be during the 11th century across the ethnically and linguistically diverse region of Northern China, and bring to the fore what some of these key teachings were. My main goal is to present the specific circumstances within which Huayan-Chan developed within the three kingdoms of Northern Song China, the Khitan Liao, and the Tangut Xixia.

Resumen
The process of cutting sheet steel plates is a mandatory work that must be done in a manufacturing process that uses plate-based materials. Cutting of sheet steel plates is usually done using a shearing machine, a cutting grinding machine, plate scissors and weld cutting. The purpose of this study focuses on improving the performance capability of the machine / tool, especially in hand grinding machines so that their function is not only limited to cutting workpieces under the hand, but its function can be maximized to cut sheet plates, box pipes, hollows more easily and quickly by adding jig components as supporting components. The stages that must be done are including design, cutting materials in accordance with the design plan, parakitan (Asembly), finishing, the next stage is testing the working ability of the tool. As for the manufacturing site, testing of the working method of the tool is carried out in the laboratory of bench work and plate majoring in mechanical engineering of Banyuwangi State Polytechnic. From the test results, the plate cutting time using a sliding cutting jig tool becomes faster at approximately 2 -3 minutes than a grinder without being installed on the aid tool. From the results of research that has been done, it is seen that the function of tool performance from hand grinding becomes more optimal and able to contribute to the lecture practicum of bench and plate work in the laboratory of bench and plate work department of Mechanical Engineering Banyuwangi State Polytechnic.

Resumen
The so-called Dao Guan Hu (bottom-filled ewer), also referred to as Dao Liu Hu (reverse-flow ewer), and Dao Zhu Hu (reverse-filled ewer), is a type of pouring vessel designed with Stevin’s Law, a formula in physics which employs a communicating tube to balance out the equilibrium of the liquid levels via a vacuum lock. The structure has a small hole at the bottom of a ewer, a jar, or a trompe-l'œil figure connected to a hollow tube inside the vessel. The liquid will not leak out when turning the vessel upright after it is filled. The current evidence attests that China started producing such wares in the ninth century during the late Tang dynasty. The production continued throughout the Song, Liao, Jin, Ming, and Qing dynasties, and the products were traded to Europe as export ceramics in the seventeenth century. They were also found on the Korean peninsula as Goryeo celadon in the twelfth century and in addition as Buncheong ware during the Joseon dynasty in the fifteenth century. The blue and white teapots with overglaze decoration retrieved from a shipwreck assemblage near Hội An in Vietnam also testify the production of this type of vessels with the same structure in Vietnam in the fifteenth and early sixteenth centuries. In the early eighteenth century, the Meissen porcelain manufactory in Germany copied peach-shaped white porcelain pots with overglaze polychrome enamels from imported Chinese bottom-filled prototypes. Dutch potters also decorated imported white porcelain Dao Guan Hu from China with overglaze polychrome enamels.

The article explores Jiang Menglin’s philosophy of life and his notion of rensheng guan (“view on life”) in the period between his studies in the USA and the year of the May Fourth events in 1919. In the first part, the paper traces the origins of Jiang’s idea back to the then-prevalent version of pragmatism propagated by John Dewey and other pragmatist thinkers gathered at Columbia University, while in the subsequent parts it aims to illuminate the later developments of Jiang’s own version of pragmatism in the context of the May Fourth intellectual discourse. While the article aims at presenting a positive outline of Jiang’s philosophy, it also endeavours to expose its less explicit aspects through its apophatic (exposition by negation or denial) expositions in Jiang’s writings from the period. Finally, it focuses on Jiang’s contributions to the debate on suicide that developed after Lin Deyang’s suicide in November 1919.

Resumen
Due to the similarity of passengers source in long distance transportation, competitions among passenger dedicated line(PDL), existing railway lines and air transportation are severely. First of all, analyzing the element which influence travelers choice, then a competition model for these three modes of transportation are gotten, which mainly take ticket price, comfort and travel time into account. In the process of competition, the ticket price adjustment can affect the service quality. Therefore, in this paper using the average crowding degree to measure service quality, under the promise the guarantee of the service quality of transportation modes, a PDL ticket price model based on the service quality is formulated, and the LINGO software be used to solve this model. At last the PDL of Jing-guang is chosen as an example to calculate the ticket price of PDL. The result shows that in order to meet the required of service quality of each transportation mode, the ticket price of PDL should no less than RMB 700.

The β-glucanase gene (GLU, from Paenibacillus polymyxa CP7) was cloned into a specific expression plasmid (MUC2-GLU-LV). Transgenic mice were prepared by microinjection. Polymerase chain reaction and Southern blot analysis of genomic DNA extracted from the tail tissue of transgenic mice showed that the mice carried the β-glucanase gene. Northern blot analysis indicated that β-glucanase was specifically expressed in the intestine of the transgenic mice. The β-glucanase activity in the intestinal contents was found to be 1.23±0.32 U mL−1. The crude protein, crude fat digestibility of transgenic mice were increased by 9.32 and 5.09% (P<0.05), respectively, compared with that of the non-transgenic mice, while moisture in feces was reduced by 12.16% (P<0.05). These results suggest that the expression of β-glucanase in the intestine of animals offers a promising biological approach to reduce the anti-nutritional effect of β-glucans in feed.

This paper contrasts the rhythm of Li Bai's “GUAN SHAN YUE”(关山月)and that of Fletcher's English version “The MOON OVER The PASS” and finds out the functional equivalence between the Ping (平) and Ze (仄) (level and oblique tones) of Chinese poetry and the lightly stressed and heavily stressed syllables of English poetry, the number of Yan (言) (the number of characters) and the number of feet, and the rhymes. To promote the realization of musical beauty, formal beauty and emotional expression in the translation of Chinese poetry and reproduce the rhythm of ancient Chinese poetry, five-character and seven-character poem with regular line can be translated into iambic pentameter; pre-Tang poem and Song Ci with irregular line can be translated according to the analogy of character and syllable, and the feet of the translated poem can mainly be iambs; the rhyme of the translated poem can be couplet rhyme, cross rhyme or alternate rhyme. Based on this strategy, the author translates her self-created poem “MAN JIANG HONG ZHAN YI”(满江红·战疫)into English to prove the feasibility of the rhythm reproduction strategy of the English translation of ancient Chinese poetry.

AbstractThe Abbey Celebrating the Tang [Qingtang guan 慶唐觀], a Daoist temple on Mount Longjiao in southern Shanxi Province, played a special role in the religious history of China in the Tang dynasty. Because of the myth that Laozi himself emerged from this mountain during the war to found the Tang state, this abbey was closely linked to the political legitimation of the Tang. Even plants in this abbey were regarded as the harbingers of the fate of the state. The emperor Xuanzong erected a huge stele in the Abbey Celebrating the Tang, demonstrating the support enjoyed from the royal house. Images of the six emperors, from Tang Gaozu to Xuanzong, were also held in the abbey. After the collapse of the Tang dynasty in 907, the Abbey Celebrating the Tang lost its political, legitimizing privileges, but its connection with the local community continued to develop well into the Song, Liao, Jin, and later dynasties. The creation and transformation of the Abbey Celebrating the Tang not only show the political influence of popular religion in ancient medieval China but also provide an interesting case of how a Daoist temple grew in popularity and prestige after it lost favor with the state.

A new kind of cathode materials, Li3V2(PO4)3/C nanocomposites, has been prepared via one-step solid-state reaction using ultra low-cost asphalt as both reduction agents and carbon sources. The asphalt is contained 60.37% of fixed carbon and 0.18% of other impurity.It is purchased from Zhen jiang Xin Guang Metallurgical Subsidiary Material Plant. Structural analysis shows that the obtained Li3V2(PO4)3/C nanocomposites contain abundant Li3V2(PO4)3 nanorods and micro/nano particles encapsulate with carbon shells. The Li3V2(PO4)3/C nanocomposites achieve enhanced dischargeability, reversibility, and cycleability. Electrochemical tests show that the Li3V2(PO4)3/C nanocomposite has initial discharge capacities of 170 mAhg-1 at 0.1C in the voltage range of 3.0 to 4.8 V. The improved electrochemical properties of the Li3V2(PO4)3/C nanocomposites are attributed to the presence of Li3V2(PO4)3/C nanorods and the electronically conductive carbon shell. This one-step solid state reaction using low-cost asphalt as carbon sources is feasible for the preparation of the Li3V2(PO4)3/C nanocomposites which can offer favorable properties for commercial applications.

ObjectivesIndividuals with obesity especially excessive visceral adiposity have high risk for incident hypertension. Recently, a new algorithm named relative fat mass (RFM) was introduced to define obesity. Our aim was to investigate whether it can predict hypertension in Chinese population and to compare its predictive power with traditional indices including body mass index (BMI), waist circumference (WC) and waist-to-height ratio (WHtR).DesignA 6-year prospective study.SettingNine provinces (Hei Long Jiang, Liao Ning, Jiang Su, Shan Dong, He Nan, Hu Bei, Hu Nan, Guang Xi and Gui Zhou) in China.ParticipantsThose without hypertension in 2009 survey and respond in 2015 survey.InterventionLogistic regression were performed to investigate the association between RFM and incident hypertension. Receiver operating characteristic (ROC) analysis was performed to compare the predictive ability of these indices and define their optimal cut-off values.Main outcome measuresIncident hypertension in 2015.ResultsThe prevalence of incident hypertension in 2015 based on RFM quartiles were 14.8%, 21.2%, 26.8% and 35.2%, respectively (p for trend <0.001). In overall population, the OR for the highest quartile compared with the lowest quartile for RFM was 2.032 (1.567–2.634) in the fully adjusted model. In ROC analysis, RFM and WHtR had the highest area under the curve (AUC) value in both sexes but did not show statistical significance when compared with AUC value of BMI and WC in men and AUC value of WC in women. The performance of the prediction model based on RFM was comparable to that of BMI, WC or WHtR.ConclusionsRFM can be a powerful indictor for predicting incident hypertension in Chinese population, but it does not show superiority over BMI, WC and WHtR in predictive power.

Abstract There are few effective therapeutic options for metastatic lung adenocarcinomas (LUADs) that lack actionable mutations in receptor tyrosine kinases, and there is an urgent need to develop a better understanding of the biological basis of LUAD growth and metastasis. Our group has developed mouse models of LUAD metastasis and found that LUAD metastasis is epithelial-to-mesenchymal transition (EMT)-dependent. EMT causes cancer cells to switch their axis of polarity from apical-basal to front-rear, which orients organelles and actinbased cytoskeletal structures in ways that facilitate purposeful cancer cell motility and metastasis. The EMT-activating transcription factor ZEB1 silences microRNAs that target key inhibitors of the polarity axis switch. These data support the general belief that EMT-dependent metastasis is a cell-autonomous process. However, our new findings here show that ZEB1 coordinates the components of secretory trafficking machinery, Rab6A and Rab8A, via silencing miR-148a that target these effectors, to drive polarized secretory trafficking toward the leading edge of LUAD cells, to enhance surface exposure of MMP14 so that triggers ECM degradation, accelerates focal adhesion turnover and promotes cancer cell migration. Moreover, the ZEB1-driven secretory trafficking increases the secretions of cytokines and immunomodulation factors, to generate an immunosuppressive tumor microenvironment, and to promote LUAD metastasis. These findings advance a paradigm in which EMT drives LUAD metastasis through a cell non-autonomous mechanism. The novelty rests in our results that demonstrate a transcriptional governance of polarized vesicular transport, providing the potential to target ZEB1-driven secretory pathways for the purpose of blocking metastasis in LUAD. Rechargeable post Li-ion batteries based on aluminum (Al) are gaining attention due to high abundance of Al, the high theoretical volumetric capacity, and high safety associated with the low flammability [1,2]. In comparison to Li-ion batteries (LiBs), where the solid-electrolyte interphase (SEI) has been studied for decades, little is known on SEI formation in dependence of the used electrolyte for Al electrodes. In this study, we present the electrochemical performance of Al foils with two different thicknesses (0.025 mm and 0.075 mm), which were used as negative electrode in Al-ion batteries in AlCl3/1-ethyl-3-methylimidazolium chloride [EMIm]Cl electrolyte. Atomic force microscopy (AFM) was used to determine changes in the morphology of Al foils during the charge - discharge process to obtain information on the SEI and its microstructural morphology [3]. Spatially-resolved information on the electrochemical activity of interphase layers on cycled Al foils can be obtained via scanning electrochemical microscopy (SECM), which so far has only be studied at LiBs [4]. First results reveal the correlation between the morphological changes of SEI layer during cycling and its electrochemical behavior depending on the Al foil surface properties, which will be presented and discussed in this contribution. Abstract Background: Gastric cancer (GC) ranks the fifth most common cancer and the fourth most frequent cause of cancer-related deaths. Genomically stable gastric cancers (GSGCs) represent 20% of all GCs and GSGC patients have the worst prognosis among all GC subtypes. Although two HER2-targeted antibody-drug conjugates (ADCs), RC48 and DS8201, have shown promising clinical efficacies, most GSGC patients do not benefit from these ADCs due to the infrequent and heterogeneous HER2 expression. Therefore, there remains a significant and unmet medical need for developing novel ADCs targeting GSGC tumors. Methods: In this presented study, a quantitative and unbiased approach was first used to identify potential targets for GSGC in a panel of human GSGC cell lines in reference with two normal human gastric epithelial cells. Cell membrane localizations of potential targets on GSGC cell lines were determined by immunofluorescent (IF) staining. To correlate it with clinically-relevant GSGC tumor samples, immunohistochemical (IHC) staining of potential targets were performed in human diffuse GC tumor tissues along with normal gastric tissues. Next, GSGC cellular internalization capability of antibodies against potential targets was evaluated. Then, an optimized target was selected for developing GSGC-targeted ADCs. A series of ADC formulations with different linkers and payloads were constructed and characterized. Their anti-tumor activities against GSGC cell lines were subsequently screened in order to identify the optimized ADC formulation. Eventually, the anti-tumor activity and biosafety profile of the optimized ADC formulation were determined in suppressing tumor growth, progression and metastasis by using established orthotopic and peritoneal metastasis models of GSGC. Results: We first identified CD54 as a potential molecular target for human GSGCs using comparative flow cytometric analysis. We next evaluated its potential as an ADC target for GSGC-targeted therapy by determining its overexpression levels, cell membrane localizations, and cell internalization activity in GSGC cells. Furthermore, we constructed and characterized a panel of CD54-targeted ADCs and identified an optimized ADC formulation by quantitatively determining their half maximum inhibitory concentrations (IC50s) and validated its anti-tumor efficacy against GSGC tumors. Meanwhile, there was no evidence to indicate histopathological damages to normal organs in the optimized ADC treated group. Conclusions: In this study, we identified a molecular target for GSGC using an unbiased and quantitative screening assay and explored its potential application in developing GSGC-targeted ADCs. In order to maximize its efficacy, we optimized the ADC formulations against GSGC cells using different ADC linkers and payloads. Our study provides a promising targeted therapeutic candidate for the clinical treatment of GSGC. Objective. To clarify the effectiveness and mechanism of the Chinese herbal formula Xingpi Kaiyu Fang (XPKYF) which is composed of American ginseng (Xi-Yang-shen), Radix curcumae (Yu-Jin), Acori tatarinowii rhizoma (Shi-Chang-pu), and Hypericum perforatum (Guan-Ye-lian-qiao) in depressed rats. Methods. The rat model of depression was established by chronic unpredictable mild stress (CUMS) method for 6 weeks. Rats were randomly divided into six groups: control group, CUMS group, CUMS+XPKYF (3.6g/kg/d, 7.2g/kg/d, 14.4g/kg/d) groups, and CUMS+sertraline (4.5mg/kg/d) group. The sucrose preference test and the forced swimming test were performed to assess the rats’ depression behavior. Mitochondrial ultrastructure was observed by transmission electron microscope and adenosine triphosphate (ATP) content, sodium potassium ATPase (Na/K-ATPase) activity, and mitochondrial respiratory chain complexes activities in hippocampus and gastrocnemius muscle were measured at the 14th and 42nd day. Results. Rats subjected to six weeks of CUMS exhibited decreased sucrose preference ratio and prolonged immobility time. CUMS reduced ATP content in hippocampus, decreased Na/K-ATPase activity and respiratory chain complex I, III, and IV activities in hippocampus and gastrocnemius muscle, and damaged mitochondrial ultrastructure of hippocampus and gastrocnemius muscle. XPKYF at 14.4g/kg, the efficacy trend of which was better than the other drug groups, could prevent the stress-induced depressed behavior changes, inhibit the decrease of Na/K-ATPase activity in hippocampus, inhibit the decrease of respiratory chain complex III activities in hippocampus and gastrocnemius muscle, and protect mitochondria from ultrastructural damage. Conclusions. Energy deficiency and damaged mitochondrial ultrastructure were found in hippocampus and gastrocnemius muscle of depressed rats established by CUMS. XPKYF could partly reverse alterations in ATP, Na/K-ATPase, and respiratory chain complexes of hippocampus and gastrocnemius muscle and protect mitochondria from ultrastructural damage. This provides another experimental evidence for the clinical application of XPKYF in the treatment of depression.

Abstract. Gross primary productivity (GPP) quantifies the amount of carbon dioxide (CO2) fixed by plants through photosynthesis. Although as a key quantity of terrestrial ecosystems, there is a lack of high-spatial-and-temporal-resolution, real-time and observation-based GPP products. To address this critical gap, here we leverage a state-of-the-art vegetation index, near-infrared reflectance of vegetation (NIRV), along with accurate photosynthetically active radiation (PAR), to produce a SatelLite Only Photosynthesis Estimation (SLOPE) GPP product for the contiguous United States (CONUS). Compared to existing GPP products, the proposed SLOPE product is advanced in its spatial resolution (250 m versus >500 m), temporal resolution (daily versus 8 d), instantaneity (latency of 1 d versus >2 weeks) and quantitative uncertainty (on a per-pixel and daily basis versus no uncertainty information available). These characteristics are achieved because of several technical innovations employed in this study: (1) SLOPE couples machine learning models with MODIS atmosphere and land products to accurately estimate PAR. (2) SLOPE couples highly efficient and pragmatic gap-filling and filtering algorithms with surface reflectance acquired by both Terra and Aqua MODIS satellites to derive a soil-adjusted NIRV (SANIRV) dataset. (3) SLOPE couples a temporal pattern recognition approach with a long-term Cropland Data Layer (CDL) product to predict dynamic C4 crop fraction. Through developing a parsimonious model with only two slope parameters, the proposed SLOPE product explains 85 % of the spatial and temporal variations in GPP acquired from 49 AmeriFlux eddy-covariance sites (324 site years), with a root-mean-square error (RMSE) of 1.63 gC m−2 d−1. The median R2 over C3 and C4 crop sites reaches 0.87 and 0.94, respectively, indicating great potentials for monitoring crops, in particular bioenergy crops, at the field level. With such a satisfactory performance and its distinct characteristics in spatiotemporal resolution and instantaneity, the proposed SLOPE GPP product is promising for biological and environmental research, carbon cycle research, and a broad range of real-time applications at the regional scale. Cu2(ZnSn)(Se)4 (CTZSS) has an significant advantages over CIGS as thin film solar cell due to its optimum band gap. Producing thin film layers by electroplating techniques is chiefly attractive due to low production cost and high throughput 1-3. Several publication reports CTZS electrodeposition; however, the electrodeposition method still has an important challenge such as a annealing process due to partial pressure variation4. We introduce in this work an advance lower electrolyte concentration which suitable to single bath electroplating similar to prvious work with CIGS5. The electrolyte is significantly more dilute in comparison to common electrolytes designated in the literature1-4. We use pulsing technique with current pulsing technique to improve the deposit adhesion and quality. Theelectrolyte composition is: 0.0042 M CuSO4, 0.0031 M ZnSO4, 0.0051 M SnCl2, 0.0062 M Na2S2O3, and 0.19 mM Na2S2O3. PHydrion is used as a buffer (pH=2), and 0.61 M LiCl is used as supporting electrolyte. Electroplating experiments was carried out at a rotating disk electrode system which provides a better controlled electroplating condition. Rotating disk function is to have better control to experiment parameters and to compare Dc current efficiency with pulsing current. electrochemical behavior study and the deposit equality investigation are provided in Fig. 1. The effects of pulsing time and current density on the CTZS alloy atomic composition, deposit quality, and its adhesion to the back contact are discussed. The ongoing pandemic of COVID-19 has forced us to consider using available drugs in the shortfall of vaccines and established treatment. Cotrimoxazole is one of the oldest drugs presently used in the prevention and treatment of opportunistic infections in the human immune deficiency virus (HIV) etc. It is a combination of two drugs Trimethoprim and Sulfamethoxazole. This cost-effective old drug is well tolerated among the population with the concomitant use of folic acid; moreover, it also looks after the secondary infections. For the last fifteen years, on-surface synthesis became an extremely popular and powerful method to fabricate nanostructures which otherwise would not have been accessible by conventional wet organic syntheses. The syntheses, realized in the chamber of high vacuum scanning tunneling microscope (STM), permitted the fabrication and the characterization of atomically defined nanostructures such as polycyclic aromatic hydrocarbons (PAH), graphene nanoribbons or 2D covalent networks.1 Among the organic molecules studied by STM, porphyrins are of particular importance because of their electronic and optical properties. Initial studies were mainly devoted to the supramolecular organization of porphyrins and porphyrin derivatives on surface and it is only in 2004 that noble metal surfaces were used as catalytic surface to perform reactions on the porphyrin cores.2 Since then, examples of open-shell porphyrin derivatives3 and conjugated oligomers4 were reported. While the synthesis of fused bis- and tetra-4,5-bis-(2,4,6-trimethylphenoxy)anthracen-9-yl porphyrins containing Ni(II) was reported by Anderson about 10 years ago,5 the formation π-extended porphyrin bearing unsubstituted anthracenyl moieties were never achieved. The formation of such compounds became possible by on-surface synthesis. Herein, we report on the synthesis of fused bis- and tetra-anthracenylporphyrin derivatives via the cyclodehydrogenation the parent meso-substituted molecules on a catalytic Au(111) surface.6 Depending on the annealing temperature, anthracenylporphyrins with the four anthracene fully linked to the porphyrin core or over-oxidized porphyrin in which the anthracene are connected two by two in 2-2’ position were observed by STM (Figure 1). Abstract Cytokines are potent stimulators of the immune system and have long been investigated and used as therapeutics to treat cancer. However, free cytokines have short half-life and the lack of tumor targeting often results in systemic toxicity. IL-15 is a stimulatory cytokine that shares the common beta and gamma receptors with IL-2. In contrast to IL-2, IL-15 is more selective for NK and effector memory T cells, with less proliferative effect on Tregs, making it a more preferred therapeutic candidate for cancer. QL415 is an anti-PD-L1 x IL-15 fusion protein designed to enhance tumor accumulation and prolong blood circulation, thereby widening the therapeutic window. IL-15 and IL-15 receptor alpha sushi domain are covalently linked to the C-terminus of our proprietary PD-L1 antibody. Fc-mediated effector functions were ablated using mutations in the CH2 domain, to prevent depletion of effector immune cells. In vitro studies showed QL415 to be highly potent in promoting proliferation and pSTAT5 signaling of IL-15 -responding NK92 and M07e cells. QL415 selectively induced proliferation of NK and CD8+ T cells from human PBMC in vitro and mouse CD8+ and NK1.1 cells in vivo. In a MC38 mouse colon cancer model expressing human PD-L1, QL401 was highly effective in suppressing tumor growth, in contrast to PD-L1 monoclonal antibody or a non-targeted IL-15 control. Follow up tumor rechallenge study showed protective memory against cognate MC38 but not B16F10. In a separate MC38 tumor model using a mouse surrogate molecule of QL415, immunophenotyping of antigen presenting cells offered preliminary evidence of enrichment of conventional dendritic cells 1 in the tumor draining lymph nodes. Therefore, QL415 not only directly stimulates effector immune cells, but also indirectly through antigen presenting cells, promotes robust anti-tumor response. In non-human primates, Q415 was tolerated up to 1.5 mg/kg, well above the effective therapeutic dose. In light of these favorable preclinical efficacy and safety results, a phase 1 dose escalation and expansion study has been initiated to evaluate the safety, tolerability, and early efficacy of QL415.
