Texto completoIn forty years only, DNA sequencing technologies triggered a revolution in biological analysis with the beginning of the genomic era. Nevertheless, this booming technological field is still hampered by unmet technological needs for DNA sample preparation and quality control. The most recent third generation sequencing technologies require very long DNA fragments of more than 50,000 base pairs, the manipulation and characterization of which remains a technological challenge. In the prospect of accelerating and improving state of the art protocols for sample preparation, we developed an instrument, based on the µLAS technology that allows the concentration and separation of high molecular weight DNA fragments with high sensitivity. With this technology, we developed a method for the isolation and sequencing of target genomic regions in complex genomes. We report the isolation, the sequencing and the assembly of a locus of 31.5 kb extracted from the genome of the plant Medicago Truncatula. We finally developed a prototype for high molecular weight DNA purification in complex samples, which is based on a size-accordable DNA valve for the size selection in the range 200 – 50,000 bp. Texto completoNew synthesis routes have been developed for the preparation of glycerilic hydroxyurethanes and polyhydroxyurethanes without the use of isocyanates. They were prepared from biosourced cyclocarbonates, highly reactive towards amines, diamines and polyamines. The providers of cyclocarbonate function are glycerol carbonate and glycerol acrylate carbonate which is a bifunctional reactive. Glycerilic hydroxyurethanes and polyhydroxyurethanes with high oxygenated and nitrogenated potential were then designed via these new glycerilic cyclocarbonates’ intermediates, substitutes of fossil cyclic carbonates such as ethylene or propylene carbonates. The strategy development of high molecular weights glycerilic polyhydroxyurethanes is based on the sequenced reactivity of the glycerol acrylate carbonate: Aza-Michaël reaction on the ethylenic function of the acrylate and aminolysis reaction of the 5-membered cyclic carbonate function. According to the nature of the amine, polyhydroxyurethanes up to 40,000 g/mol were obtained. They can be obtained as foams, gels, resins, depending on the predominance of linear or cross-linked bridges. Intramolecular chemical networks were elaborated by hydrogen bonding between O-H or N-H sites with the oxygen atom of the carbamoyl function of the glycerilic skeleton. That is the reason why carbamoyle functions are less susceptible to hydrolysis and why the chemical and mechanical resistance properties of the glycerilic polyhydroxyurethanes are increased

Texto completoThis main objective of this work is the study of interactions between prokaryotic compartments of meso-and bathypelagic zones with different size classes of dissolved organic carbon (DOC). Several measurements of monomers assimilation (3H-Glucose), of complex molecules degradation (3H-EPS and HMW-DOC) and prokaryotic heterotrophic production were realized through the water column of NW Mediterranean Sea (Gulf of Lion, DYFAMED and ANTARES station) and NE Atlantic Ocean (PAP site). During these studies, all measurements realized below 1000 m depth, were carry out under in situ condition of hydrostatic pressure (> 10 MPa, HP) and compared to their decompressed counterpart measurements, realized at atmospheric pressure (0.1 MPa, ATM). These coupled measurements determine the role of hydrostatic pressure on deep sea microbial activity following the Pressure effect (Re) ration (=HP measurement / ATM measurement). The results show that microbial activities measured under HP condition during stratified water period, were more important than those measured under ATM condition (mean Pe = 4.01, n=120), and confirm the abilities of deep sea prokaryotes to degrade complex organic molecules. Moreover, the cell-specific activity of deep sea prokaryotes in 3H-EPS degradation are 6 time more active than the surface, indicating the ability of autochthonous deep sea prokaryotes to degrade complex molecules under high conditions of pressure

Texto completoHyaluronan (HA) is a biopolymer used as a therapeutical agent for its chemo-physical properties. The aim of this thesis was to investigate the pharmacological properties of high molecular weight HA. In vitro and in vivo data indicate that high molecular weight HA has significant protective effects against benzalkonium chloride (a preservative incriminated in several iatrogenic pathologies), sodium laurylsulfate (toxic used in reglementary studies of ocular toxicity), and UVB radiations (responsible for traumas of the ocular surface). This modulation is attributable to pharmacological activities of HA, with antiapoptotic and anti-inflammatory effects we observed in vitro, which could explain the ocular surface-protective role of HA we observed in vivo

Texto completoThe aim of this study was to produce and characterize composites made from wood flour and Ultra High Molecular Weight Polyethylene (UHMWPE). Firstly, the composites were initially prepared with wood sawdust provided from a sawmill and this study highlights particularly the use of raw materials in a powder form. Composites were prepared by a simple dry mixing technique and then compression molded. The wood content was up to 30%wt. The effect of wood content on morphology, density, hardness, and mechanical properties (tensile, torsion and flexion) was investigated. The results show that wood flour addition increased substantially all the moduli (up to 97%). The results show also that good dispersion and adhesion were achieved and wood flour addition increased significantly the mechanical properties of the composites.

Texto completoAn increasing demand for materials used for stab protection has been expressed to provide more protective, flexibility and lightweight. Researchers have mainly focused on studies about stab resistance of soft body armour based upon technical textile fibres and 2D fabrics. However, the soft protective materials based on 3D fabrics have been rarely study in recent research works, especially those revealing that 3D woven architectures can play a decisive role during stab impact. 3D warp interlock fabrics (3DWIFs) can be used in a soft vest for anti-stab applications. The overall aim of this current research has been oriented to explore different design of 3DWIFs that provide the more efficient protective solution. Hence, this thesis has been concentrated on both the manufacturing process parameters and the resulted product parameters of the 3DWIFs made with HMWPE yarns. The production process parameters have been studied to optimize the manufacturing and the mechanical properties of 3DWIFs. The product parameters of 3DWIFs have been investigated to find the optimized combination for the best protective resistance against stabbing. The four main categories of 3D warp interlock fabrics architectures as : A/T, A/L, O/T, and O/L, were woven by twisted high molecular weight polyethylene (HMWPE) yarns. The mechanical characteristics of 3DWIFs were systematically tested and compared. Besides, a dedicated experimental study has been performed on 3DWIFs submitted to low-speed impact, including single-stab and double-stab properties in terms of depth of penetration and trauma. The double-pass stabbing tests are complementary to single-pass stabbing tests. It was experimentally concluded that the orthogonal/through-the-thickness interlock fabric has a good stab resistance. Texto completoAccurate measurements of physical activity in free living are needed to establish what dose of physical activity is necessary for obtaining a specific health benefits. The first aim of this work was to validate the Actiheart (which combines heart rate and accelerometry sensors) with doubly labeled water (DLW). We show a good level of concordance between physical activity energy expenditure (PAEE) estimated by Actiheart and DLW. Individualization of the relationship between heart rate and PAEE by an incremental test is needed for an accurate estimate of the PAEE at the individual level and to evaluate changes induced by an intervention. In laboratory, we show that the accuracy of Actiheart is activitydependent. This requires the establishment of their recognition from new sensors and mathematical models. Adiponectin, hormone secreted by adipose tissue, has a role in energy metabolism and its secretion decreases with obesity. However the effects of physical activity remain in contradiction in published studies. The second objective of this work was to evaluate the effect of physical activity and intervention with weight control on plasma adiponectin. We show that the total and high molecular weight adiponectin were negatively associated with modifications of the physical activity level. Further work is however necessary to understand the mechanisms underlying this modulation of plasma adiponectin which does not seem related to changes in synthesis in adipose tissue or muscle
