Abstract Objectives We propose a one-shot, privacy-preserving distributed algorithm to perform logistic regression (ODAL) across multiple clinical sites. Materials and Methods ODAL effectively utilizes the information from the local site (where the patient-level data are accessible) and incorporates the first-order (ODAL1) and second-order (ODAL2) gradients of the likelihood function from other sites to construct an estimator without requiring iterative communication across sites or transferring patient-level data. We evaluated ODAL via extensive simulation studies and an application to a dataset from the University of Pennsylvania Health System. The estimation accuracy was evaluated by comparing it with the estimator based on the combined individual participant data or pooled data (ie, gold standard). Results Our simulation studies revealed that the relative estimation bias of ODAL1 compared with the pooled estimates was <3%, and the ratio of standard errors was <1.25 for all scenarios. ODAL2 achieved higher accuracy (with relative bias <0.1% and ratio of standard errors <1.05). In real data analysis, we investigated the associations of 100 medications with fetal loss during pregnancy. We found that ODAL1 provided estimates with relative bias <10% for 85% of medications, and ODAL2 has relative bias <10% for 99% of medications. For communication cost, ODAL1 requires transferring p numbers from each site to the local site and ODAL2 requires transferring (p×p+p) numbers from each site to the local site, where p is the number of parameters in the regression model. Conclusions This study demonstrates that ODAL is privacy-preserving and communication-efficient with small bias and high statistical efficiency.

The outer dynein arm of Chlamydomonas flagella, when isolated under Mg(2+)-free conditions, tends to dissociate into an 11 to 12S particle (12S dynein) containing the gamma heavy chain and a 21S particle (called 18S dynein) containing the alpha and beta heavy chains. We show here that functional outer arms can be reconstituted by the addition of 12S and 18S dyneins to the axonemes of the outer armless mutants oda1-oda6. A third factor that sediments at integral 7S is required for efficient reconstitution of the outer arms on the axonemes of oda1 and oda3. However, this factor is not necessary for reconstitution on the axonemes of oda2, oda4, oda5, and oda6. SDS-PAGE analysis indicates that the axonemes of the former two mutants lack a integral of 70-kD polypeptide that is present in those of the other mutants as well as in the 7S fraction from the wild-type extract. Furthermore, electron micrographs of axonemal cross sections revealed that the latter four mutants, but not oda1 or oda3, have small pointed structures on the outer doublets, at a position in cross section where outer arms normally occur. We suggest that the 7S factor constitutes the pointed structure on the outer doublets and facilitates attachment of the outer arm. The discovery of this structure raises a new question as to how the attachment site for the outer arm dynein is determined within the axoneme.

Previous work has revealed a cytoplasmic pool of flagellar precursor proteins capable of contributing to the assembly of new flagella, but how and where these components assemble is unknown. We tested Chlamydomonas outer-dynein arm subunit stability and assembly in the cytoplasm of wild-type cells and 11 outer dynein arm assembly mutant strains (oda1-oda11) by Western blotting of cytoplasmic extracts, or immunoprecipitates from these extracts, with five outer-row dynein subunit-specific antibodies. Western blots reveal that at least three oda mutants (oda6, oda7, and oda9) alter the level of a subunit that is not the mutant gene product. Immunoprecipitation shows that large preassembled flagellar complexes containing all five tested subunits (three heavy chains and two intermediate chains) exist within wild-type cytoplasm. When the preassembly of these subunits was examined in odastrains, we observed three patterns: complete coassembly (oda 1, 3, 5, 8, and 10), partial coassembly (oda7 and oda11), and no coassembly (oda2, 6, and 9) of the four tested subunits with HCβ. Our data, together with previous studies, suggest that flagellar outer-dynein arms preassemble into a complete Mr ≃ 2 × 106 dynein arm that resides in a cytoplasmic precursor pool before transport into the flagellar compartment.

The concept odal can be regarded in a narrow sense, i.e. as the inherited landed property of a family. But here it is argued that odal should be viewed in a wide sense - as a mentality that is of great importance to the understanding of Late Iron Age society in Sweden. The article focuses on the material expressions which belonging to a family and possessing a farm could take in the individual farmstead in the Mälar Valley. The Viking Age is interpreted as a time period in which there was a need to make the odal visible. The acts of burying dead relatives on top of the graves of early ancestors, erecting runestones, and possibly also erecting mounds are regarded as ways of guarding, marking, and confirming the possession of the odal in the odal man's own eyes and in his neighbours' and consequently also the odal man's position in society.

Outer dynein arms (ODAs) are multiprotein complexes that drive flagellar beating. Based on genetic and biochemical analyses, ODAs preassemble in the cell body and then move into the flagellum by intraflagellar transport (IFT). To study ODA transport in vivo, we expressed the essential intermediate chain 2 tagged with mNeonGreen (IC2-NG) to rescue the corresponding Chlamydomonas reinhardtii mutant oda6. IC2-NG moved by IFT; the transport was of low processivity and increased in frequency during flagellar growth. As expected, IFT of IC2-NG was diminished in oda16, lacking an ODA-specific IFT adapter, and in ift46 IFT46ΔN lacking the ODA16-interacting portion of IFT46. IFT loading appears to involve ODA16-dependent recruitment of ODAs to basal bodies followed by handover to IFT. Upon unloading from IFT, ODAs rapidly docked to the axoneme. Transient docking still occurred in the docking complex mutant oda3 indicating that the docking complex stabilizes rather than initiates ODA–microtubule interactions. In full-length flagella, ODAs continued to enter and move inside cilia by short-term bidirectional IFT and diffusion and the newly imported complexes frequently replaced axoneme-bound ODAs. We propose that the low processivity of ODA-IFT contributes to flagellar maintenance by ensuring the availability of replacement ODAs along the length of flagella.

Axonemal dyneins are multisubunit enzymes that must be preassembled in the cytoplasm, transported into cilia by intraflagellar transport, and bound to specific sites on doublet microtubules, where their activity facilitates microtubule sliding-based motility. Outer dynein arms (ODAs) require assembly factors to assist their preassembly, transport, and attachment to cargo (specific doublet A-tubule sites). In Chlamydomonas, three assembly factors—ODA5, ODA8, and ODA10—show genetic interactions and have been proposed to interact in a complex, but we recently showed that flagellar ODA8 does not copurify with ODA5 or ODA10. Here we show that ODA5 and ODA10 depend on each other for stability and coexist in a complex in both cytoplasmic and flagellar extracts. Immunofluorescence and immuno–electron microscopy reveal that ODA10 in flagella localizes strictly to a proximal region of doublet number 1, which completely lacks ODAs in Chlamydomonas. Studies of the in vitro binding of ODAs to axonemal doublets reveal a role for the ODA5/ODA10 assembly complex in cytoplasmic maturation of ODAs into a form that can bind to doublet microtubules.

35 strains of Chlamydomonas mutant missing the entire outer dynein arm were isolated by screening slow-swimming phenotypes. They comprised 10 independent genetic loci (odal-10) including those of previously isolated mutants oda38 and pf28. The 10 loci were distinct from pf13 and pf22, loci for nonmotile mutants missing the outer arm. These results indicate that at least 12 genes are responsible for the assembly of the outer dynein arms. There were no mutants lacking partial structures of the outer arm, suggesting that lack of a single component results in failure of assembly of entire outer arms. Temporary dikaryons derived from mating of two different oda strains often, but not always, recovered the wild-type motility within 2 h of mating. Hence, outer arms can be transported and attached to the outer doublets independently of flagellar growth.

Research on damage diagnosis or safety monitoring based on structural vibration response is one of the hot issues in the engineering field. The characteristic information of the structure is obtained by analyzing the structure response data. In the process of data analysis, the choice of data length is very important, which is related to the validity of the structure monitoring results. At present, the selection of data length is usually subjective, which reduces the rigor of the structure monitoring process. Therefore, a method based on improved multiscale permutation entropy (IMPE) is proposed to determine the optimal data analytical length (ODAL) of vibration data. This method creatively applies multiscale permutation entropy (MPE) to the field of data length analysis when processing nonlinear and nonstationary signals and optimizes MPE with the help of the improved coarse-grained method to obtain IMPE. IMPE is sensitive to different data lengths, and the entropy changes with the increase of the data length and tends to be stable. Here, the stable value is defined as a standard entropy. The entropy satisfying 97% of the standard entropy is used as the effective entropy, and the corresponding data length value of the effective entropy is selected as the ODAL of the vibration data. This method is suitable for many fields, provides a reliable data analytical length for data analysis, and has good engineering practicability.

To learn more about how dyneins are targeted to specific sites in the flagellum, we have investigated a factor necessary for binding of outer arm dynein to the axonemal microtubules ofChlamydomonas. This factor, termed the outer dynein arm-docking complex (ODA-DC), previously was shown to be missing from axonemes of the outer dynein armless mutants oda1 and oda3. We have now partially purified the ODA-DC, determined that it contains equimolar amounts ofM r ∼105,000 and ∼70,000 proteins plus a third protein of M r ∼25,000, and found that it is associated with the isolated outer arm in a 1:1 molar ratio. We have cloned a full-length cDNA encoding theM r ∼70,000 protein; the sequence predicts a 62.5-kDa protein with potential homologs in higher ciliated organisms, including humans. Sequencing of corresponding cDNA from strain oda1 revealed it has a mutation resulting in a stop codon just downstream of the initiator ATG; thus, it is unable to make the full-length M r ∼70,000 protein. These results demonstrate that the ODA1 gene encodes the M r ∼70,000 protein, and that the protein is essential for assembly of the ODA-DC and the outer dynein arm onto the doublet microtubule.

Assembly of outer dynein arms (ODAs) requires multiple steps and involves multiple proteins in addition to dynein subunits. The Chlamydomonas ODA10, ODA5, and ODA8 loci genetically interact and are hypothesized to function as an axonemal accessory complex, but only ODA5p was previously characterized. We positionally cloned ODA10 and identified the gene by rescuing an oda10 mutant with a hemagglutinin-tagged cDNA. ODA10 sequence predicts a conserved coiled-coil protein homologous to mouse ccdc151. ODA10p is present in cytoplasm and flagella, remains axonemal after detergent treatment, and is extracted with 0.6 M NaCl. Both outer arm dynein and ODA10p rebound to the axonemes when desalted extracts are mixed with oda10-mutant axonemes. Sucrose gradient separation of these extracts shows that ODA10p sediments near the top of the gradient, not with 23S outer dynein arm proteins. Unexpectedly, dynein and ODA10p fractions are able to bind individually to oda10 axonemes. ODA10p is present on oda8-mutant flagella at wild-type levels. However, ODA10p does not assemble into oda5 flagella and is absent from oda5 cytoplasm, suggesting a necessity of ODA5p for stability of ODA10p in vivo. The results suggest that ODA10p does not function as a part of a traditionally defined docking complex.

Of the uncloned ODA genes required for outer dynein arm assembly in Chlamydomonas, ODA5 and ODA10 are of particular interest because they do not encode known subunits of the outer arm or the outer dynein arm-docking complex (ODA-DC), and because genetic studies suggest their products interact. Beginning with a tagged oda5 allele, we isolated genomic and cDNA clones of the wild-type gene. ODA5 predicts a novel, 66-kDa coiled-coil protein. Immunoblotting indicates Oda5p is an axonemal component that assembles onto the axoneme independently of the outer arm and ODA-DC and is uniquely missing in oda5 and oda10 axonemes. Oda5p is released from the axoneme by extraction with 0.6 M KCl, but the soluble Oda5p does not cosediment with the outer dynein arm/ODA-DC in sucrose gradients. Quantitative mass spectrometry by using isotope coded affinity tagging revealed that a previously unidentified adenylate kinase is reduced 35–50% in oda5 flagella. Direct enzymatic assays demonstrated a comparable reduction in adenylate kinase activity in oda5 flagella, and also in oda10 flagella, but not in flagella of other oda mutants. We propose that Oda5p is part of a novel axonemal complex that is required for outer arm assembly and anchors adenylate kinase in proximity to the arm.

We have used an insertional mutagenesis/ gene tagging technique to generate new Chlamydomonas reinhardtii mutants that are defective in assembly of the outer dynein arm. Among 39 insertional oda mutants characterized, two are alleles of the previously uncloned ODA3 gene, one is an allele of the uncloned ODA10 gene, and one represents a novel ODA gene (termed ODA12). ODA3 is of particular interest because it is essential for assembly of both the outer dynein arm and the outer dynein arm docking complex (ODA-DC) onto flagellar doublet microtubules (Takada, S., and R. Kamiya. 1994. J. Cell Biol. 126:737– 745). Beginning with the inserted DNA as a tag, the ODA3 gene and a full-length cDNA were cloned. The cloned gene rescues the phenotype of oda3 mutants. The cDNA sequence predicts a novel 83.4-kD protein with extensive coiled-coil domains. The ODA-DC contains three polypeptides; direct amino acid sequencing indicates that the largest of these polypeptides corresponds to ODA3. This protein is likely to have an important role in the precise positioning of the outer dynein arms on the flagellar axoneme.

The newness of this industry presents an opportunity to investigate the lived experience of remote employees working in an up-and-coming virtual organization. Through interviews with six ODAs, we explored motivations, day-to-day workflow, and development of work identities. Analysis uncovered unique challenges ODAs faced when performing the “human-based” tasks of online dating, which differed starkly from other popular services being bought and sold in the gig economy (e.g., rideshare, food delivery). Findings also show how ODAs engage in pragmatic and critical sensemaking as they navigate the specific challenges associated with ODA labor, and those created by remote work and gig labor, more generally.

Oxydiacetic acid (Oda) and iminodiacetic acid (Ida) have been combined with two diamines, namely ethylenediamine (en) and o-phenylenediamine (o-phen), to give three molecular crystals of compositions [(Oda2−)(en2+)]·H2O (Odaen), [(Ida−)(0.5en+)] (Idaen) and [(Oda−)(o-phen+)] (Odaophen). Single crystal X-ray structure determination of the three molecular salts revealed that the hydrogen bonding interactions form distinct supramolecular chains. In Odaen, water molecules and Oda2− anions generate one-dimensional supramolecular chains where two chains overlap each other in zigzag form. In Odaophen, Oda− anions generate a one-dimensional linear supramolecular chain. In Idaen, both intermolecular and intramolecular proton transfer occurs. Ida− anions generate a one-dimensional zigzag chain. These one-dimensional supramolecular chains extend to form different two-dimensional supramolecular chains by the interaction of diaminium and monoaminium cations. Intermolecular close contacts are also examined using Hirshfeld analysis to reveal similarities and differences in the crystal structures.

We find that two Chlamydomonas outer arm dynein assembly loci, oda6 and oda9, are located on the left arm of linkage group XII, in the vicinity of the previously mapped locus for a 70,000 Mr dynein intermediate chain protein. Restriction fragment length polymorphism mapping indicates that this dynein gene is very closely linked to the oda6 locus. A cDNA clone encoding the 70,000 Mr protein was isolated, sequenced, and used to select genomic clones spanning the corresponding locus from both wild-type and oda6 libraries. When wild-type clones were introduced into cells containing an oda6 allele, the mutant phenotype was rescued, while no rescue was observed after transformation with oda6 clones. Genetic analysis further revealed that newly introduced gene copies were responsible for the rescued phenotype and thus confirms that ODA6 encodes the 70,000 Mr dynein intermediate chain protein. The inability of oda6 mutants to assemble any major outer arm dynein subunits shows that this protein is essential for assembly of stable outer dynein arms. This is the first use of transformation with a wild-type gene to identify the product of a Chlamydomonas mutant.

Structural health monitoring (SHM) is a modern technique f or damage identification in the e xis ting structure. The structural stiffness, frequency, damping, and dominant mode shapes represent the actual operating conditions of the structure. The main principle of structural health monitoring is to identif y the mod al parameters from experime ntal resu lts both damaged and undamaged conditions. Damage is much effective to decrease stiffness and strength of structural components and it changes dynamic behaviour and damping ratio of whole structures. Bruel & Kjaer experi mental modal analys is technique is r ecently used for civil engineering structures for modal parameters estimation. The paper describes the initial structural health monitoring of a steel frame . The modal parameters were estimated for undamaged condition s a nd these result s are verified and up dated by the numerical FEM tool SAP2000. For the undamaged structure , mode shapes and frequencies were calibrated properly. In the second step, damaged was initiated by dismantling one element from the lower part of the frame. The estimat ed m odal parameter s were compared to the initial one. The mode shapes and frequencies are quite different for some specific mode due to damage initiation . One extra mode was created for the damaged frame due to damage initiation. The 4 th mode was not found f or the initial m easurement because of presence of lower beam. ABSTRACT The outer dynein arm of Chlamydomonas flagella contains three heavy chains (α, β, and γ), each of which exhibits motor activity. How they assemble and cooperate is of considerable interest. Here we report the isolation of a novel mutant, oda2-t, whose γ heavy chain is truncated at about 30% of the sequence. While the previously isolated γ chain mutant oda2 lacks the entire outer arm, oda2-t retains outer arms that contain α and β heavy chains, suggesting that the N-terminal sequence (corresponding to the tail region) is necessary and sufficient for stable outer-arm assembly. Thin-section electron microscopy and image analysis localize the γ heavy chain to a basal region of the outer-arm image in the axonemal cross section. The motility of oda2-t is lower than that of the wild type and oda11 (lacking the α heavy chain) but higher than that of oda2 and oda4-s7 (lacking the motor domain of the β heavy chain). Thus, the outer-arm dynein lacking the γ heavy-chain motor domain is partially functional. The availability of mutants lacking individual heavy chains should greatly facilitate studies on the structure and function of the outer-arm dynein.

Purpose This paper aims to explore the literature on office design approaches (ODAs) in relation to employee health. The overall goal is to facilitate the practical use and theoretical development of design approaches to healthy offices. Design/methodology/approach A scoping review of 7,432 papers collected from 4 electronic databases and 5 scientific journals resulted in the selection of 18 papers for content analysis. Findings Various ODAs relating to building design features and health were identified. The findings highlight challenges for this emergent field, including a paucity of literature on ODAs, a lack of definitions of health and healthy offices, ambiguous design strategies and a lack of a holistic ODA. Originality/value ODAs are potentially valuable resources but an under-considered topic for healthy office development. To the best of the authors’ knowledge, this study is the first scoping review to map and compare different design approaches in the context of office design and its main contribution is in encouraging researchers and practitioners to bring a salutogenic and holistic perspective to their design approaches.

Montmorillonite (MMT) was modified by octadecylamine (ODA) surfactant. The surfactant contents were varied, i.e. 0.5, 1.0 and 2.0 times the cation exchange capacity (CEC) of the MMT. XRD and FTIR spectra of the organoclay revealed that ODA molecules intercalated into MMT layers. The MMT-ODA was melt-mixed with natural rubber (NR) using a two roll mill. Effects of surfactant content and organoclay content on cure characteristics, mechanical properties, and morphologies of NR nanocomposites were investigated. Morphologies of the NR nanocomposites, with increasing surfactant content, revealed the exfoliated structure and the good dispersion of the organoclay in the NR matrix. These caused the enhancement of mechanical properties of the NR nanocomposites. With increasing the MMT-ODA2 content up to 5 phr, scorch time and cure time of the NR nanocomposites decreased while their tensile strength increased.

Oda padeda reguliuoti žmogaus kūno temperatūrą, per ją vyksta įvairių medžiagų absorbcija bei sekrecija. Neigiami išoriniai veiksniai turi įtakos raginio odos sluoksnio pažeidimų atsiradimui, o tai sutrikdo barjerines odos funkcijas. Įvairios odos ligos gali pažeisti pėdas. Pagrindiniai problemų sukėlėjai: bakterijos, virusai, grybeliai. Siekiant atkurti lipidinį raginio odos sluoksnio barjerą, naudojami ingredientai, kurių struktūra panaši į natūraliai esančių raginio sluoksnio lipidų struktūrą. Sausų pėdų korekcijai dažnai naudojama apikosmetika. Vienas iš apikosmetikos ingredientų yra bičių vaškas, pasižymintis gydomosiomis ir antibakterinėmis savybėmis. Šio tyrimo tikslas – ištirti emulsijos su bičių vašku poveikį dehidratuotai pėdų odai. Tyrimo rezultatai parodė, kad emulsija su bičių vašku teigiamai veikia dehidratuotą pėdų odą. Po 10 savaičių emulsijos naudojimo, buvusi dehidratuota visų tiriamųjų pėdų oda tapo normali.

Axonemal dynein complexes are preassembled in the cytoplasm before their transport to cilia, but the mechanism of this process remains unclear. We now show that mice lacking Pih1d3, a PIH1 domain–containing protein, develop normally but manifest male sterility. Pih1d3−/− sperm were immotile and fragile, with the axoneme of the flagellum lacking outer dynein arms (ODAs) and inner dynein arms (IDAs) and showing a disturbed 9+2 microtubule organization. Pih1d3 was expressed specifically in spermatogenic cells, with the mRNA being most abundant in pachytene spermatocytes. Pih1d3 localized to the cytoplasm of spermatogenic cells but was not detected in spermatids or mature sperm. The levels of ODA and IDA proteins were reduced in the mutant testis and sperm, and Pih1d3 was found to interact with an intermediate chain of ODA as well as with Hsp70 and Hsp90. Our results suggest that Pih1d3 contributes to cytoplasmic preassembly of dynein complexes in spermatogenic cells by stabilizing and promoting complex formation by ODA and IDA proteins.

<p>Sub-Saharan Africa (SSA) countries have benefited for more than fifty years from international aid in the form of loans and/or donations. Nevertheless, they seem not to benefit from these massive financial resources (ODA) they receive because their economic and social situation is not very good. This study aims to assess the impact of ODA on economic growth in SSA and to see if its effect on growth is conditioned by the quality of the economic policy. The estimates are conducted on a dynamic panel of twenty-three SSA countries running from 1985 to 2014. With macroeconomic data from the World Bank's CD-ROM (World Development Indicators, 2015), the Generalized Method of Moment (GMM) system from Blundel and Bond (1998) was used. The results show that the impact of ODA on growth is not significant. Subsequently, when squared aid (ODA2) is included in the estimate, ODA becomes significant, meaning that a substantial amount of assistance is required to be effective in raising the economic growth rate of the SSA countries. In addition, the effectiveness of ODA is conditioned by the quality of the economic policy. This seems to be bad in SSA, hence the negative impact of the aid on economic growth.</p>

Axonemal protein complexes, such as outer (ODA) and inner (IDA) dynein arms, are responsible for the generation and regulation of flagellar and ciliary beating. Studies in various ciliated model organisms have shown that axonemal dynein arms are first assembled in the cell cytoplasm and then delivered into axonemes during ciliogenesis. In humans, mutations in genes encoding for factors involved in this process cause structural and functional defects of motile cilia in various organs such as the airways and result in the hereditary disorder primary ciliary dyskinesia (PCD). Despite extensive knowledge about the cytoplasmic assembly of axonemal dynein arms in respiratory cilia, this process is still poorly understood in sperm flagella. To better define its clinical relevance on sperm structure and function, and thus male fertility, further investigations are required. Here we report the fertility status in different axonemal dynein preassembly mutant males (DNAAF2/ KTU, DNAAF4/ DYX1C1, DNAAF6/ PIH1D3, DNAAF7/ZMYND10, CFAP300/C11orf70 and LRRC6). Besides andrological examinations, we functionally and structurally analyzed sperm flagella of affected individuals by high-speed video- and transmission electron microscopy as well as systematically compared the composition of dynein arms in sperm flagella and respiratory cilia by immunofluorescence microscopy. Furthermore, we analyzed the flagellar length in dynein preassembly mutant sperm. We found that the process of axonemal dynein preassembly is also critical in sperm, by identifying defects of ODAs and IDAs in dysmotile sperm of these individuals. Interestingly, these mutant sperm consistently show a complete loss of ODAs, while some respiratory cilia from the same individual can retain ODAs in the proximal ciliary compartment. This agrees with reports of solely one distinct ODA type in sperm, compared to two different ODA types in proximal and distal respiratory ciliary axonemes. Consistent with observations in model organisms, we also determined a significant reduction of sperm flagellar length in these individuals. These findings are relevant to subsequent studies on the function and composition of sperm flagella in PCD patients and non-syndromic infertile males. Our study contributes to a better understanding of the fertility status in PCD-affected males and should help guide genetic and andrological counselling for affected males and their families.

A new 3d-4 f heteronuclear coordination polymer containing cobalt and neodymium, [Nd2Co3(oda)6(H2O)6]·3H2O}n (1) (H2oda = oxydiacetic acid), has been synthesized and structurally characterized. In 1, the oxydiacetate dianions (oda2−) act as pentadentate ligands, and each of them chelates one Nd3+ ion and bridges two Co2+ ions. The infinite connection of metal ions and ligands results in a highly ordered 3D hexagonal channel framework. The photoluminescent, EPR and magnetic properties of the complex were also investigated.

Redox-based regulatory systems are essential for many cellular activities. Chlamydomonas reinhardtii exhibits alterations in motile behavior in response to different light conditions (photokinesis). We hypothesized that photokinesis is signaled by variations in cytoplasmic redox poise resulting from changes in chloroplast activity. We found that this effect requires photosystem I, which generates reduced NADPH. We also observed that photokinetic changes in beat frequency and duration of the photophobic response could be obtained by altering oxidative/reductive stress. Analysis of reactivated cell models revealed that this redox poise effect is mediated through the outer dynein arms (ODAs). Although the global redox state of the thioredoxin-related ODA light chains LC3 and LC5 and the redox-sensitive Ca2+-binding subunit of the docking complex DC3 did not change upon light/dark transitions, we did observe significant alterations in their interactions with other flagellar components via mixed disulfides. These data indicate that redox poise directly affects ODAs and suggest that it may act in the control of flagellar motility.

Oda – organas, užtikrinantis apsaugą nuo išorės poveikio ir išlaikantis reikiamą organizmo skysčių balansą. Oda sensta kartu su visu organizmu. Senstančios odos koregavimui plačiai naudojamas masažas, įvairūs pilingai, injekcinės procedūros, aparatinės metodikos. Tyrimo tikslas – įvertinti elektrostimuliacijos ir klasikinio masažo poveikį brandžiai veido odai. Tyrime dalyvavo šešios 40-50 metų moterys. Visos tiriamosios atsitiktinai buvo suskirstytos į dvi grupes, kurioms buvo taikomos skirtingos veido odos poveikio priemonės: trims klasikinis veido masažas (KM grupė), kitoms trims tiriamosioms – audinių elektrostimuliacija (ES grupė). Tiriamųjų odos būklė buvo vertinama ,,Skin Analyzer Soft plus“ aparatu, audinių deguonies įsotinimas ir kraujotaka buvo registruojama neinvaziniu artimosios infraraudonosios spektroskopijos būdu. Atlikus skirtingas veido odos stimuliacijas – elektrostimuliaciją ir klasikinį veido masažą, nustatyta: klasikinis veido masažas pagerino veido odos drėgmę U zonoje, veido odos riebumas padidėjo visoms tiriamosioms. Raukšlių gylis visoms tiriamosioms kaktos zonoje sumažėjo, jų plotis padidėjo, o raukšlių gylis akių zonoje nepakito, tačiau jų plotis šioje zonoje padidėjo. Arterinė audinių kraujotaka po masažo procedūrų reikšmingai pagerėjo tik vienai tiriamajai. Elektrostimuliacija reikšmingai padidino veido audinių kraujotaką visoms tiriamosioms. Klasikinis veido masažas reikšmingai pagerino deguonies įsisavinimą tik vienai tiriamajai. Vertinant deguonies įsisavinimo kaitą elektrostimuliacijos poveikyje, užfiksuotas reikšmingas deguonies įsisavinimo sumažėjimas dviem tiriamosioms po atliktų procedūrų.

Inner dynein arms, but not outer dynein arms, require the activity of KHP1(FLA10) to reach the distal part of axonemes before binding to outer doublet microtubules. We have analyzed the rescue of inner or outer dynein arms in quadriflagellate dikaryons by immunofluorescence microscopy of p28(IDA4), an inner dynein arm light chain, or IC69(ODA6), an outer dynein arm intermediate chain. In dikaryons two strains with different genetic backgrounds share the cytoplasm. As a consequence, wild-type axonemal precursors are transported to and assembled in mutant axonemes to complement the defects. The rescue of inner dynein arms containing p28 in ida4-wild-type dikaryons progressively occurred from the distal part of the axonemes and with time was extended towards the proximal part. In contrast, the rescue of outer dynein arms in oda2-wild-type dikaryons progressively occurred along the entire length of the axoneme. Rescue of inner dynein arms containing p28 in ida4fla10-fla10 dikaryons was similar to the rescue observed in ida4-wild-type dikaryons at 21 degrees C, whereas it was inhibited at 32 degrees C, a nonpermissive temperature for KHP1(FLA10). In contrast, rescue of outer dynein arms in oda2fla10-fla10 dikaryons was similar to the rescue observed in oda2-wild-type dikaryons at both 21 degrees and 32 degrees C and was not inhibited at 32 degrees C. Positioning of substructures in the internal part of the axonemal shaft requires the activity of kinesin homologue protein 1.

The outer dynein arms (ODAs) of the flagellar axoneme generate forces needed for flagellar beating. Elucidation of the mechanisms underlying the chemomechanical energy conversion by the dynein arms and their orchestrated movement in cilia/flagella is of great importance, but the nucleotide-dependent three-dimensional (3D) movement of dynein has not yet been observed. In this study, we establish a new method for reconstructing the 3D structure of the in vitro reconstituted ODA–microtubule complex and visualize nucleotide-dependent conformational changes using cryoelectron microscopy and image analysis. As the complex went from the rigor state to the relaxed state, the head domain of the β heavy chain shifted by 3.7 nm toward the B tubule and inclined 44° inwards. These observations suggest that there is a mechanism that converts head movement into the axonemal sliding motion.

Demembranated sperm flagella from Gallus domesticus have been prepared by the rapid-freeze, deep-etch, rotary replica technique in order to study the three-dimensional morphology of the outer dynein arm (ODA)-ATPase complex. In general, the ODAs resemble most closely those from the sea urchin Strongylocentrotus described by W. S. Sale et al. In the ‘rigor’ condition, the ODA consists of a major subunit (the head), from which a slender link extends to the adjacent B-tubule (the B-link). A smaller, intermediate subunit lies adjacent to the head, distally, and a further extension of the complex, the minor subunit, continues distally beneath the head domain of the next arm complex, where it attaches to the A-tubule. In the presence of ATP and vanadate (‘relaxed’ condition), the attachment point of the B-link to the head is shifted to a more proximal position, and the minor subunit is no longer visible. This is interpreted as resulting from a rotation of the head. These features are demonstrated stereoscopically, and from several viewpoints. Image enhancement has been used to clarify and define the repetitive features of the dynein arrays. In addition, some of the axonemes have been imaged from highly contrasted 20 nm thin sections; the detection of B-links in such sections means that these slender structures cannot be considered artefacts of the rapid-freeze, deep-etch protocol.

This article examines the role of lecturers in encouraging and supporting students likely to be predisposed to challenges related to incomprehension, incapacity and isolation embedded in the virtual learning environment. This article used a constructivist lens to gain through interviews an understanding into the intuition, thoughts, ideals, beliefs and inclination of lecturers about the nature and extent of their supportive role in the e-learning environment. The key findings revealed that the role of lecturers in promoting e-learning is varied. The discrepancy seems to be emanating from the lack of clear understanding of the meaning, the depth, the breadth and thrust of e-learning pedagogy at the University of South Africa (UNISA). The UNISA Strategy 2015-2030 introduced incremental changes in the form of Open Distance e-Learning (ODeL), which highlights the infusion of e-learning in the Open Distance Learning (ODL) context. Given its ODeL mandate, and the prescripts of the UNISA Strategy 2015-2030 and as part of the academic project the institution committed to introduce e-learning from 2015. Further, the institutional resolve to adopt a hybrid model that incorporates traditional and electronic teaching modes has caused uncertainty in the teaching space with some lecturers leaning towards e-learning whilst others lean towards traditional methods. The article recommends that UNISA needs to clarify its standpoint by adopting definitions, demarcations, strategies and tools that will enhance the understanding, adoption and usability of e-learning platforms and systems.

The nexin-dynein regulatory complex (N-DRC) forms a cross-bridge between the outer doublet microtubules of the axoneme and regulates dynein motor activity in cilia/flagella. Although the molecular composition and the three-dimensional structure of N-DRC have been studied using mutant strains lacking N-DRC subunits, more accurate approaches are necessary to characterize the structure and function of N-DRC. In this study, we precisely localized DRC1, DRC2, and DRC4 using cryo–electron tomography and structural labeling. All three N-DRC subunits had elongated conformations and spanned the length of N-DRC. Furthermore, we purified N-DRC and characterized its microtubule-binding properties. Purified N-DRC bound to the microtubule and partially inhibited microtubule sliding driven by the outer dynein arms (ODAs). Of interest, microtubule sliding was observed even in the presence of fourfold molar excess of N-DRC relative to ODA. These results provide insights into the role of N-DRC in generating the beating motions of cilia/flagella.

Grow Together: Save the World While Building a Meaningful ‘Bee-lationship’ Paul Sherfey Keywords: insects; interspecies relationships; human-animal studies; community gardens Collective gardens – in which individuals work collectively to cultivate and care for a common gardening area – have become a growing phenomenon in recent years. At these sites, the cultivation of community is often as important as the cultivation of organic, local produce. However, observation and digital research carried out in the context of a transnational study of such gardens demonstrates that this community is not limited to human participants, but instead also includes other animal species at these sites. The article investigates the relationships cultivated with one such group – insects. How might we understand the interest shown by gardeners in building hotels and cafés, sowing meadows and arranging festivals for insects? Do participants only see insects for their use-value, or is there something more occurring in the relationship they cultivate, and how it is represented and discussed? Beginning with a discussion of the built environment of the studied collective gardens, the article analyses how certain design choices are specifically oriented towards the use and benefit of insects – especially bees. Progressing from physical space to digital space, the empirical discussion then investigates this interest in bees and their welfare further through several paradigmatic examples. In so doing, discourses communicated in manifestos, social media and news interviews are analysed. This is done in order to explore the worldviews from which individuals and groups understand the importance of bees, as well as the backgrounds that influence their actions and the fantasies for the future that provide a focal point towards which to orient their efforts. Finally, I contrast the discourses about bees with the lack of similar discourse about another group of insects which are readily observable at many sites – wasps. I discuss how differing cultural heritages related to each affect how they are valued and reflect on the possibilities available to us as humans to see ourselves and our future as being dependent on one species, while being comparably indifferent to the presence and important contributions of the other.

Abstract The use of high-throughput sequencing techniques has allowed the identification of numerous mutations in genes responsible for severe astheno-teratozoospermia due to multiple morphological abnormalities of the sperm flagella (MMAF). However, more than half of the analysed cases remain unresolved suggesting that many yet uncharacterised gene defects account for this phenotype. Based on whole-exome sequencing data from a large cohort of 167 MMAF-affected subjects, we identified two unrelated affected individuals carrying a homozygous deleterious mutation in CFAP70, a gene not previously linked to the MMAF phenotype. One patient had a homozygous splice variant c.1723-1G>T, altering a consensus splice acceptor site of CFAP70 exon 16, and one had a likely deleterious missense variant in exon 3 (p.Phe60Ile). The CFAP70 gene encodes a regulator protein of the outer dynein arms (ODA) strongly expressed in the human testis. In the sperm cells from the patient carrying the splice variant, immunofluorescence (IF) experiments confirmed the absence of the protein in the sperm flagellum. Moreover, IF analysis showed the absence of markers for the ODAs and the central pair complex of the axoneme. Interestingly, whereas CFAP70 staining was present in sperm cells from patients with mutations in the three other MMAF-related genes ARMC2, FSIP2 and CFAP43, we observed an absence of staining in sperm cells from patients mutated in the WDR66 gene, suggesting a possible interaction between two different axonemal components. In conclusion, this work provides the first evidence that loss of CFAP70 function causes MMAF and that ODA-related proteins may be crucial for the assembly and/or stability of the flagellum axoneme in addition to its motility.

β-N-Oxalyl-l-α,β-diaminopropionic acid (β-ODAP) is a non-protein amino acid present in Lathyrus sativus (grass pea) and other Lathyrus species, in parallel with its nontoxic isomer, α-ODAP. When consuming grass pea for several months as staple food, β-ODAP may cause neurolathyrism, a motor neuron degeneration syndrome. Therefore, the independent quantification of both ODAP isomers instead of only the total amount in grass pea allows the identification of less toxic varieties and the development of tools to support breeding for improving grass pea quality. In this work, a simple and fast HPLC-MS/MS method was developed without sample derivatization, using a hydrophilic interaction chromatography (HILIC) column and an isocratic gradient of eluents for 18 min, which allowed the determination of both α- and β-ODAP. The proposed method was fully validated and applied to the determination of α- and β-ODAP contents in a diverse collection of 107 grass pea accessions representative of the main grass pea-growing geographical regions in the world, with the prompt identification of contrasting accessions. β-ODAP content in the analyzed grass pea samples ranged from 0.45 ± 0.02 to 6.04 ± 0.45 mg g−1. The moderate correlation found between α- and β-ODAP contents (0.65) in this collection reinforces the importance of the independent quantification of both ODAP isomers.

A novel potentiometric sensor based on ionophore (Cd(NH2CH2CH2OCH2CH2OCH2CH2NH2)Ag3(CN)5) for the determination ofβ-N-oxalyl-l-α,β-diaminopropionic acid (ODAP) is developed. The ODAP-selective membrane sensor demonstrates high sensitivity and short response time. The detection limit of the ODAP-selective membrane sensor is about2×10-6 mol L-1and the response time is shorter than 6 s. The linear dynamic range of the ODAP-selective membrane sensor is between ODAP concentrations of1.0×10-2and1×10-6 mol L-1. The ODAP-selective membrane sensor exhibits good operational stability for at least one week in dry conditions at 4–6°C. It has a reproducible and stable response during continuous work for at least 10 h with a relative standard deviation of 0.28% (n= 18).
