Texto completoColorectal cancer (CRC) is a major public health problem yet it remains the third most common and the second deadliest cancer for both men and women in the United States and in Europe, owing to diagnosis at advanced stage and variable response to the treatments. However, the incidence and mortality appear to be steadily declining in countries with programmatic screening and it remains critical to characterize CRC molecular subtypes to identify new biomarkers. CRC display a wide clinicopathological and molecular heterogeneity and arise from different carcinogenesis pathway. Approximately 20% to 30% of CRC occur by the means of the serrated neoplasia pathway. CRC from the serrated pathway are associated with frequent epigenetic instability (CIMP, CpG island methylator phenotype) which causes most sporadic microsatellite instability (MSI) CRC through epigenetic inactivation of MLH1 and frequent BRAF mutation. CRC from the serrated pathway frequently display a mucinous pattern and are prone to express secreted mucins MUC2 and MUC5AC. Early identification of serrated pathway precursor lesions is currently challenging since the progression to adenocarcinoma in this pathway is particularly brief.We assessed in this study clinicopathological and molecular features of colorectal carcinomas and polyps and we analyzed the interest of mucin genes MUC2 and MUC5AC to classify serrated polyps and to identify the precursor lesions of CIMP/MSI carcinomas._x000D_A series of 418 CRC and 330 polyps was included in the study, with 218 serrated polyps and 112 conventional adenomas. We assessed the molecular profile (KRAS/BRAF mutations, MSI, CIMP, MGMT methylation, MLH1) and the expression and methylation profile of MUC2 and MUC5AC genes and further correlation with clinical and pathological data were performed.We show that MUC5AC hypomethylation is a specific marker of CIMP/MSI CRC, independently of other clinical or pathological factors. We show moreover than MUC5AC hypomethylation is an early event of carcinogenesis, specific to microvesicular hyperplastic polyps (MVHP) and sessile serrated adenoma (SSA) suggesting a filiation between these two lesions and also a progression of MVHP to SSA, SSA with dysplasia, and then to MSI/CIMP CRC. MUC5AC hypomethylation was moreover highly specific of BRAF mutated, CIMP or MSI serrated lesions.In conclusion, our results suggest that the MUC5AC mucin is involved/plays a role in the development/progression of colorectal tumors of the serrated pathway. MUC5AC hypomethylation occurs early during carcinogenesis and may be of interest for the diagnosis of serrated polyp especially in the presence of ambiguous morphology and to early detection of polyps carrying a malignant potential. Our results suggest moreover that some MVHP may progress to SSA and CIMP/MSI CRCs highlighting the importance to perform biopsy of serrated polyps during endoscopy to identify microvesicular subtype among colorectal hyperplastic polyps, which could require a minimal surveillance

Texto completoFrom a commonsense perspective, the multicellular organisms such as men and horses are thought of as typical examples of biological individuals. But contemporary philosophers of biology present a number of definitions of biological individuality which stimulate an intense debate over deciding the level of organization to which the biological individuality corresponds. This research proposes a historical analysis, which doesn’t aim to solve the conceptual issues, but to understand the diversity of biological thinking about individuality. From the regeneration of the polyp discovered by Abraham Trembley in the middle of the 18th century to the philosophical renovation achieved by Henri Bergson at the beginning of the 20th century, the biological thinking called into question the basis on which the notion of biological individuality was built. The invention and the reinterpretation of the concepts such as the organism, the cell, the division of labor, the milieu, the natural selection and the life, allowed to ask if the biological individuality can be defined as indivisibility, secondly if it can be identified with the organism, and finally if one can determine the level of organization to which the biological individuality corresponds. The diversity of concepts of biological individuality doesn’t mean that this term is dispensable for the biological research. It allows to present the biological and philosophical problems in more precise terms, so that the biologists and philosophers can cooperate to tackle them

Many applications of amorphous polymers require a thermo-mechanically coupled large-deformation elasto-viscoplasticity theory which models the strain rate and temperature dependent response of amorphous polymeric materials in a temperature range which spans the glass transition temperature of the material. We have formulated such a theory, and also numerically implemented our theory in a finite element program. The material parameters in the theory have been calibrated for poly(methyl methacrylate), polycarbonate, and Zeonex - a cyclo-olefin polymer. The predictive capabilities of the constitutive theory and its numerical implementation have been validated by comparing the results from a suite of validation experiments against corresponding results from numerical simulations. Amorphous chemically-crosslinked polymers form a relatively new class of thermallyactuated shape-memory polymers. Several biomedical applications for thermally-actuated shape-memory polymers have been proposed/demonstrated in the recent literature. However, actual use of such polymers and devices made from these materials is still quite limited. For the variety of proposed applications to be realized with some confidence in their performance, it is important to develop a constitutive theory for the thermo-mechanical response of these materials and a numerical simulation-based design capability which, when supported with experimental data, will allow for the prediction of the response of devices made from these materials under service conditions. We have developed such a theory and a numerical simulation capability, and demonstrated its utility for modeling the thermo-mechanical response of the shape-memory polymer tBA-PEGDMA. An elastomeric gel is a cross-linked polymer network swollen with a solvent, and certain thermally-responsive gels can undergo large reversible volume changes as they are cycled about a critical temperature. We have developed a thermodynamically-consistent continuum-level theory to describe the coupled mechanical-deformation, fluid permeation, and heat transfer of such gels. We have numerically implemented our theory in a finite element program by writing thermo-chemo-mechanically coupled elements. We show that our theory is capable of simulating swelling, squeezing of fluid by applied mechanical forces, and thermally-responsive swelling/de-swelling of such materials.

Texto completoAirway epithelium is covered by a thin layer of mucus that plays a protective role against inhaled pathogens. Mucous hypersecretion contributes to the pathophysiology of chronic airway diseases including nasal polyposis. Activation of the EGF receptor induces mucin production in experimental models but the relevance of this pathway to human diseases is unknown. The potential role of eosinophils, which are recruited to nasal polyp epithelium, in mucin synthesis is controversial. Here, we show that nasal polyp epithelium expresses mucin MUC5AC and EGF receptors. Importantly, we show that activation of the EGF receptor by its ligands induces mucin MUC5AC production in primary cultures of nasal polyp epithelial cells. We also show that human eosinophils, isolated from peripheral blood and activated in vitro, release products that induces activation of the EGF receptor leading to mucin synthesis. Because EGF receptor inhibitors are not yet approved for use in subjects with nasal polyps, we assessed the effects of corticosteroids on mucin and EGFR expression in a clinical study. Intranasal steroids reduced polyp size and eosinophilic infiltration but had little effects on mucin and EGF receptor expression. In conclusion, these studies suggest that EGF receptor regulates mucin synthesis in polyp epithelium and that EGF receptors inhibitors could be useful for targeting mucous hypersecretion in nasal polyps
