Resumen
The benefit of the entomopathogen Metarhizium anisopliae to early root development was evaluated. Two inoculating methods, conidia-suspension-drenching (T1) and conidia-coating (T2), were used when sowing peanut. The results showed that taproot length and lateral root number in T1 significantly increased compared to the control (T0) in days 4–10 after treatment, whereas no significant difference was found between T2 and T0. The fungal density by T1 and T2 fluctuated in the first 8 days, followed by a gradual decline. The ratio of the taproot length or lateral root number in T1 and T0 was significant relative to the fungal persistence. It suggested that M. anisopliae promotes peanut root development and should be considered as important factor in plant protection besides pest controls.

Texto completo
Resumen
To improve the culture system of bovine somatic cell nuclear transfer (SCNT) embryos, we studied the effects of activin A on developmental competence of bovine SCNT embryos during the early development stage based on the traditional culture method, and analyzed the expression level of the genes related to blastocyst hatching (Na/K-ATPase, Glut-1) and related to activin A signalling pathway (ActRII and Smad2). We generated the bovine SCNT embryo using a Holstein cow oocyte as recipient cytoplasm and a foetal ear fibroblast (Holstein cow, 120 days) as donor cell. The embryos were cultured as follows: experiment 1, the addition of activin A at the concentrations of 0 (control), 20 (M1&ndash;20), 40 (M1&ndash;40) or 80 ng/ml (M1&ndash;80) to the media during the first three days and no addition during the subsequent 5 days; experiment 2, no addition of activin A to the media during the first 3 days and the addition of activin A at the concentrations of 0 (control), 20 (M2&ndash;20), 40 (M2&ndash;40) or 80 ng/ml (M2&ndash;80) during the subsequent 5 days. The results indicated that the blastocyst formation rate and hatching rate, and total blastomere numbers as well as ICM/TE obtained in experiment 1 were not significantly different from the control group (P &gt; 0.05). In contrast, these values obtained in experiment 2 were significantly higher than in the control group (P &lt; 0.05). In addition, the relative abundance (ratio to GAPDH mRNA) of each gene (Glut-1, ActR II and Smad2) was not significantly different among the treatments in the experiment. The expression levels of 4 genes (Na/K-ATPase, Glut-1, ActR II and Smad2) in blastocysts obtained in experiment 2 were higher than those obtained in experiment 1. In conclusion, the present study suggests that the addition of activin A to the culture media from day 4 to day 8 can enhance the developmental competence of bovine SCNT embryos.

Resumen
Potential of Trichoderma harzianum IS005-12 (TH-IS005-12) to promote seed germination and seedling growth of Italian ryegrass (IRG) forage was evaluated in vitro. Non-desiccated seeds and those pre-harvestly desiccated with total herbicide were treated 25 days (freshly harvested) and 178 days after harvest (mature) with TH-IS005-12 spore suspensions at 0 (T0), 1.8 × 107 (T1) and 1.2 × 109 (T2) spore/mL. TH-IS005-12 promoted the early and final germination and seedling growth in all non-desiccated and desiccated, freshly harvested as well as mature IRG seeds. It was more effective in pre-harvestly desiccated freshly harvested seeds where T2 treatment increased final germination rate for 24%, root number per seedling 1.6-fold and seedling vigour 1.9-fold compared to the untreated control. Moreover, TH-IS005-12 showed an inhibitory activity against seedborne fungi Alternaria alternata and A. ventricosa suppressing their growth in vitro by 82% and 77%, respectively.

Aoki, Yoshinao, Arisa Usujima y Shunji Suzuki. "High night temperature promotes downy mildew in grapevine via attenuating plant defence response and enhancing early Plasmopara viticola infection". Plant Protection Science 57, No. 1 (3 de diciembre de 2020): 21–30. http://dx.doi.org/10.17221/92/2020-pps.

Texto completo
Resumen
The night temperature is one of the critical environmental factors affecting the grape berry quality. The objective of this study was to clarify whether a high night temperature promotes downy mildew on grapevines. The high night temperature conditions suppressed the gene expression of the pathogenesis-related proteins in the grapevine cultured cells and grapevine seedlings compared with the control night temperature conditions. The Plasmopara viticola colony formation on the leaves of the seedlings exposed to the control night temperature conditions became slightly noticeable on day 5 after inoculation, whereas a large number of colonies were clearly observed on the leaves of the seedlings exposed to the high night temperature conditions. On day 10 after inoculation, the leaf defoliation and withering were marked in the P. viticola infected seedlings exposed to the high night temperature conditions. The high night temperature conditions promoted the P. viticola zoospore germination. These results suggest that the high night temperature promoted the grape downy mildew by attenuating the constitutive plant defence response as well as enhancing the P. viticola early infection. Viticulturists should be vigilant regarding pest management strategies against the P. viticola primary infection of grapevines when high night temperatures continue for a long time.

Texto completo
Resumen
Maize yield is severely affected by soil salinity. In an effort to engineer maize for improved salt tolerance, embryogenic calli of maize were co-bombarded with plasmids containing <I>Oryza sativa</I> Na+/H+ antiporter gene (<I>OsNHX1</I>) and <I>ba</I>r genes. For the molecular analysis of putative transgenic samples, PCR, Southern and Northern blots were carried out. The maize plants over-expressing <I>OsNHX</I>1 accumulated more biomass when grown in the presence of 200mM NaCl in greenhouse conditions. Higher Na+ and K+ content was observed in transgenic leaves than in wildtype leaves when treated with 100~200mM NaCl, while the osmotic potential and the proline content in transgenic leaves was lower than in wild-type maize. A field trial revealed that the transgenic maize plants produced higher grain yields than the wild-type plants at the vegetative growth stage. These results demonstrate that the <I>OsNHX1</I> gene was successfully transferred into <I>Zea may</I>s, and the salt-tolerance of transgenic maize was improved by overexpression of the <I>OsNHX1</I> gene.

Texto completo
Resumen
The hormone leptin has a variety of functions. Originally known for its role in satiety and weight loss, leptin more recently has been shown to augment tumor growth in a variety of cancers. Within gliomas, there is a correlation between tumor grade and tumor expression of leptin and its receptor. This suggests that autocrine signaling within the tumor microenvironment may promote the growth of high-grade gliomas. Leptin does this through stimulation of cellular pathways that are also advantageous for tumor growth and recurrence: antiapoptosis, proliferation, angiogenesis, and migration. Conversely, a loss of leptin expression attenuates tumor growth. In animal models of colon cancer and melanoma, a decline in the expression and secretion of leptin resulted in a reduction of tumor growth. In these models, positive mental stimulation through environmental enrichment decreased leptin secretion and improved tumor outcome. This review explores the link between leptin and glioblastoma.
