ABSTRACTPseudomonas aeruginosais an opportunistic human pathogen and has been established as a model organism to study bacterial biofilm formation. At least three exopolysaccharides (alginate, Psl, and Pel) contribute to the formation of biofilms in this organism. Here mutants deficient in the production of one or more of these polysaccharides were generated to investigate how these polymers interactively contribute to biofilm formation. Confocal laser scanning microscopy of biofilms formed in flow chambers showed that mutants deficient in alginate biosynthesis developed biofilms with a decreased proportion of viable cells than alginate-producing strains, indicating a role of alginate in viability of cells in biofilms. Alginate-deficient mutants showed enhanced extracellular DNA (eDNA)-containing surface structures impacting the biofilm architecture. PAO1 ΔpslAΔalg8overproduced Pel, and eDNA showing meshwork-like structures presumably based on an interaction between both polymers were observed. The formation of characteristic mushroom-like structures required both Psl and alginate, whereas Pel appeared to play a role in biofilm cell density and/or the compactness of the biofilm. Mutants producing only alginate, i.e., mutants deficient in both Psl and Pel production, lost their ability to form biofilms. A lack of Psl enhanced the production of Pel, and the absence of Pel enhanced the production of alginate. The function of Psl in attachment was independent of alginate and Pel. A 30% decrease in Psl promoter activity in the alginate-overproducing MucA-negative mutant PDO300 suggested inverse regulation of both biosynthesis operons. Overall, this study demonstrated that the various exopolysaccharides and eDNA interactively contribute to the biofilm architecture ofP. aeruginosa.

We compute necessary conditions on Yetter–Drinfeld modules over the groups PGL(2, q) = PGL(2, 𝔽q) and PSL(2, q) = PSL(2, 𝔽q) to generate finite-dimensional Nichols algebras. This is a first step towards a classification of pointed Hopf algebras with group of group-likes isomorphic to one of these groups. As a by-product of the techniques developed in this work, we prove that any finite-dimensional pointed Hopf algebra over the Mathieu groups M20 or M21 = PSL(3, 4) is the group algebra.

ABSTRACTBiofilm formation is a complex process in which many factors are involved. Bacterial swarming motility and exopolysaccharides both contribute to biofilm formation, yet it is unclear how bacteria coordinate swarming motility and exopolysaccharide production. Psl and Pel are two key biofilm matrix exopolysaccharides inPseudomonas aeruginosa. This opportunistic pathogen has three types of motility, swimming, twitching, and swarming. In this study, we found that elevated Psl and/or Pel production reduced the swarming motility ofP. aeruginosabut had little effect on swimming and twitching. The reduction was due to decreased rhamnolipid production with no relation to the transcription ofrhlAB, two key genes involved in the biosynthesis of rhamnolipids. Rhamnolipid-negativerhlRandrhlABmutants synthesized more Psl, whereas exopolysaccharide-deficient strains exhibited a hyperswarming phenotype. These results suggest that competition for common sugar precursors catalyzed by AlgC could be a tactic forP. aeruginosato balance the synthesis of exopolysaccharides and rhamnolipids and to control bacterial motility and biofilm formation inversely because the biosynthesis of rhamnolipids, Psl, and Pel requires AlgC to provide the sugar precursors and an additionalalgCgene enhances the biosynthesis of Psl and rhamnolipids. In addition, our data indicate that the increase in RhlI/RhlR expression attenuated Psl production. This implied that the quorum-sensing signals could regulate exopolysaccharide biosynthesis indirectly in bacterial communities. In summary, this study represents a mechanism that bacteria utilize to coordinate swarming motility, biosurfactant synthesis, and biofilm matrix exopolysaccharide production, which is critical for biofilm formation and bacterial survival in the environment.

ABSTRACTPseudomonas aeruginosais an opportunistic human pathogen that is especially adept at forming surface-associated biofilms.P. aeruginosacauses catheter-associated urinary tract infections (CAUTIs) through biofilm formation on the surface of indwelling catheters.P. aeruginosaencodes three extracellular polysaccharides, PEL, PSL, and alginate, and utilizes the PEL and PSL polysaccharides to form biofilmsin vitro; however, the requirement of these polysaccharides duringin vivoinfections is not well understood. Here we show in a murine model of CAUTI that PAO1, a strain harboringpel,psl, andalggenes, and PA14, a strain harboringpelandalggenes, form biofilms on the implanted catheters. To determine the requirement of exopolysaccharide duringin vivobiofilm infections, we tested isogenic mutants lacking thepel,psl, andalgoperons and showed that PA14 mutants lacking these operons can successfully form biofilms on catheters in the CAUTI model. To determine the host factor(s) that induces the ΔpelDmutant to form biofilm, we tested mouse, human, and artificial urine and show that urine can induce biofilm formation by the PA14 ΔpelDmutant. By testing the major constituents of urine, we show that urea can induce apel-,psl-, andalg-independent biofilm. Thesepel-,psl-, andalg-independent biofilms are mediated by the release of extracellular DNA. Treatment of biofilms formed in urea with DNase I reduced the biofilm, indicating that extracellular DNA supports biofilm formation. Our results indicate that the opportunistic pathogenP. aeruginosautilizes a distinct program to form biofilms that are independent of exopolysaccharides during CAUTI.

Pseudolycoriella cavatica (Skuse, 1888), a widely distributed species, was recorded from New Zealand for the first time, and recognised as a senior synonym of Spathobdella setigera Hardy, 1960 syn. n. Apart from Psl. kaikoura and Psl. cavatica all New Zealand Pseudolycoriella species group in four different clusters: the Psl. bispina complex, the Psl. jejuna complex, the Psl. macrotegmenta complex, and the Psl. zealandica complex. The monophyly of those four species complexes was confirmed by a genetic analysis based on two mitochondrial genes (COI and 16S) and one nuclear gene (28S). A key to the species is given.

ABSTRACT Pseudomonas aeruginosa forms biofilms, which are cellular aggregates encased in an extracellular matrix. Molecular genetics studies of three common autoaggregative phenotypes, namely wrinkled colonies, pellicles, and solid-surface-associated biofilms, led to the identification of two loci, pel and psl, that are involved in the production of carbohydrate-rich components of the biofilm matrix. The pel gene cluster is involved in the production of a glucose-rich matrix material in P. aeruginosa strain PA14 (L. Friedman and R. Kolter, Mol. Microbiol. 51:675-690, 2004). Here we investigate the role of the pel gene cluster in P. aeruginosa strain ZK2870 and identify a second genetic locus, termed psl, involved in the production of a mannose-rich matrix material. The 11 predicted protein products of the psl genes are homologous to proteins involved in carbohydrate processing. P. aeruginosa is thus able to produce two distinct carbohydrate-rich matrix materials. Either carbohydrate-rich matrix component appears to be sufficient for mature biofilm formation, and at least one of them is required for mature biofilm formation in P. aeruginosa strains PA14 and ZK2870.

Summary This study was conducted to explore basic physical and mechanical properties of parallel strand lumber (PSL) made exclusively from southern pine and yellow-poplar, respectively, and to examine their relationships using statistical analysis. Small specimens were prepared from commercial southern pine PSL and yellow-poplar PSL billets and tested for specific gravity, moisture content, dimensional stability, bending properties, shear strength and compressive strength. Results indicate average specific gravity of southern pine PSL is higher than that of yellow-poplar PSL, while their average moisture content and dimensional stability are very similar. Southern pine PSL has higher average modulus of elasticity but lower average modulus of rupture than yellow-poplar PSL. While average longitudinal shear strength does not exhibit differences between southern pine PSL and yellow-poplar PSL, average compressive strength of southern pine PSL is higher than that of yellow-poplar PSL. There are positive correlations among modulus of elasticity, modulus of rupture and specific gravity. PSL improves some properties of solid wood from which PSL is made.

The aims of this research are to know how the design of PSL with Solar Cell Power can provide a reliable service of The PSL. Besides that, monitoring of the performance of PSL is expected to be carried out centrally, this is to facilitate employees in observing the performance of the lights, also to anticipate damage PSL components. The design of PSL are expected to meet these expectations. This PSL designed can provide data on the performance of the main components of the PSL with Solar Cell Power, namely: Solar Cell, Batteries and LEDs. Economic analysis is taken into account of the design of this PSL. Net Present Value (NPV) are used in analyzing the Smart Design of PSL, which is by adding up all cash flow and investment costs.

Planum semilunatum (PSL) cells of the rat ampullae were studied by light, transmission, and scanning electron microscopy. The observed ultrastructure of rat PSL cells was similar to that described in other species, but is in disagreement with earlier reports of rat PSL cells, most probably because of previous divergent definitions of the PSL. Regions of PSL cells were easily distinguished from other nonsensory epithelia at the lateral ends of the crista ampullaris. The PSL region consisted of irregular-shaped columnar to cuboidal pentagonal or hexagonal cells that interdigitated with one another by lateral membrane infoldings. In the PSL region the subepithelial reticular layer appeared thickened and formed wartlike impressions in the basal surface of the PSL cells. These morphological characteristics of the subepithelial reticular layer were unique to the PSL region in the ampulla and may reflect special adaptations of the PSL region to mechanical stress. Furthermore, the thick subepithelial reticular layer may have implications for transport across the PSL region.

Pakistan Super League (PSL) is a well-known T20 cricket league with millions of viewers. With this large viewer base, predicting the outcome of PSL matches opens a new research avenue for academic researchers. In this paper, we collect PSL data from relevant sources and generate a validated data set for machine learning experiments. We implement the “PSL Eye” solution which employs Neural Networks (NNs) to predict the match winning team. We preprocess the dataset to eliminate the extra variables then we tune the hyper parameters of NN. After acquiring the optimal values of hyper parameters, we run our NN based PSL Eye to obtain the final results. The overall accuracy of PSL-Eye with testing data set is 82% which is very promising and shows the importance of NN in predicting PSL match outcome.

All species are keyed, figured and distribution data is given. Ostroverkhovana Komarova, 2002 is regarded as a junior synonym of Pseudolycoriella Menzel & Mohrig, 1998. Phytosciara (Prosc.) plusiochaeta (Hippa & Vilkamaa, 1991) is reported from North America for the first time.

ABSTRACTPolysorbate 80 (PS80) is a nonionic surfactant and detergent that inhibits biofilm formation byPseudomonas aeruginosaat concentrations as low as 0.001% and is well tolerated in human tissues. However, certain clinical and laboratory strains (PAO1) ofP. aeruginosaare able to form biofilms in the presence of PS80. To better understand this resistance, we performed transposon mutagenesis with a PS80-resistant clinical isolate, PA738. This revealed that mutation ofalgCrendered PA738 sensitive to PS80 biofilm inhibition. AlgC contributes to the biosynthesis of the exopolysaccharides Psl and alginate, as well as lipopolysaccharide and rhamnolipid. Analysis of mutations downstream of AlgC in these biosynthetic pathways established that disruption of thepsloperon was sufficient to render the PA738 and PAO1 strains sensitive to PS80-mediated biofilm inhibition. Increased levels of Psl production in the presence of arabinose in a strain with an arabinose-induciblepslpromoter were correlated with increased biofilm formation in PS80. InP. aeruginosastrains MJK8 and ZK2870, known to produce both Pel and Psl, disruption of genes in thepslbut not thepeloperon conferred susceptibility to PS80-mediated biofilm inhibition. The laboratory strain PA14 does not produce Psl and does not form biofilms in PS80. However, when PA14 was transformed with a cosmid containing thepsloperon, it formed biofilms in the presence of PS80. Taken together, these data suggest that production of the exopolysaccharide Psl byP. aeruginosapromotes resistance to the biofilm inhibitor PS80.

Objective.The optimal treatment strategy for anti-aminoacyl-tRNA synthetase antibody–positive polymyositis/dermatomyositis-associated interstitial lung disease (anti-ARS-PM/DM-ILD) is yet to be established. We aimed to evaluate the efficacy of glucocorticoids and calcineurin inhibitors (CNI) in patients with ARS-PM/DM-ILD.Methods.Progression-free survival (PFS) and overall survival rates were retrospectively evaluated in 32 consecutive patients with ARS-PM/DM-ILD. Disease progression was defined as deterioration in PM/DM-ILD (including recurrence). Predictive factors associated with PFS were analyzed by Cox hazards analysis. The efficacy of first-line prednisolone (PSL) plus CNI therapy was compared with that of PSL monotherapy using propensity score–matched analysis.Results.Overall, 20 (62.5%) and 12 (37.5%) patients received first-line therapy with PSL + CNI and PSL, respectively. The 2-year PFS and 5-year survival rates in the overall cohort were 68.8% and 96.9%, respectively. On multivariate analysis, arterial oxygen pressure (HR 0.86) and PSL monotherapy (vs PSL + CNI; HR 7.29) showed an independent association with PFS. Baseline characteristics of propensity score-matched PSL + CNI and PSL groups were similar. The 2-year PFS rate was significantly higher in the matched PSL + CNI group than in the matched PSL group. All patients who experienced disease progression during first-line therapy were subsequently treated with second-line therapies. The 5-year survival rates of both the matched PSL + CNI and PSL groups were favorable.Conclusion.Propensity score–matched analysis demonstrated that first-line PSL + CNI therapy for patients with ARS-PM/DM-ILD significantly improved the PFS compared with PSL monotherapy, although there was no significant difference regarding longterm survival.

Prednisolone (PSL), a type of corticosteroid used to treat autoimmune diseases, can increase the risk of infection and osteoporosis. Saireito (114), a Kampo medicine, has an immunosuppressive effect; with its use, the dose of steroids can be reduced. However, its mechanism when used with PSL is still unclear. We used peripheral blood mononuclear cells (PBMCs) from healthy adults to examine the effect of 114 and PSL treatment on PBMC proliferation, T-cell subsets, and cytokine production. PBMCs were cotreated with concanavalin A and 300 μM 114 (either Tsumura & Co. (TJ) or Kracie Holdings (KR)) and 0.0001–1.0 μM PSL for 96 h to create the T-cell mitogen. We then measured the PBMC proliferation; ratio of CD4+ T cells, CD8+ T cells, and T-follicular helper (Tfh) cells; and concentration of cytokines (TNF, IFN-γ, IL-6, IL-10, IL-17A, and IL-21). The proliferation of PBMCs was dose dependently suppressed in both the PSL and PSL + 114 groups ( p < 0 . 05 ). Combination therapy increased the IC50 in the PSL group (0.0947 μM) by 2.02 and 1.64-fold in the PSL + TJ114 and PSL + KR114 groups, respectively. Both the PSL + 114 groups had an increased ratio of CD4+ T cells compared to the PSL group, with no effect on the ratio of CD8+ T and Tfh cells. Furthermore, the PSL + 114 groups showed increased IL-6 and IL-10 compared to the PSL monotherapy group, although the difference was not significant. There was no significant difference in the TNF, IFN-γ, IL-17A, and IL-21 concentrations between the PSL and PSL + 114 groups. The elevated IC50 with 114 cotreatment suggests diminished immunosuppressive action. Moreover, increased cytokine production by Th2 with 114 cotreatment suggests a restoration of T-cell balance in Th1-mediated autoimmune diseases. However, increased IL-6 suggests potential exacerbation of IL-6-mediated diseases, such as rheumatoid arthritis. Therefore, it is necessary to monitor these clinical parameters when using 114 in combination with PSL.

ABSTRACT The ability to form biofilms in the airways of people suffering from cystic fibrosis is a critical element of Pseudomonas aeruginosa pathogenesis. The 15-gene psl operon encodes a putative polysaccharide that plays an important role in biofilm initiation in nonmucoid P. aeruginosa strains. Biofilm initiation by a P. aeruginosa PAO1 strain with disruption of pslA and pslB (ΔpslAB) was severely compromised, indicating that psl has a role in cell-surface interactions. In this study, we investigated the adherence properties of this ΔpslAB mutant using biotic surfaces (epithelial cells and mucin-coated surfaces) and abiotic surfaces. Our results showed that psl is required for attachment to a variety of surfaces, independent of the carbon source. To study the potential roles of Psl apart from attachment, we generated a psl-inducible P. aeruginosa strain (Δpsl/p BAD -psl) by replacing the psl promoter region with araC-p BAD , so that expression of psl could be controlled by addition of arabinose. Analysis of biofilms formed by the Δpsl/p BAD -psl strain indicated that expression of the psl operon is required to maintain the biofilm structure at steps postattachment. Overproduction of the Psl polysaccharide led to enhanced cell-surface and intercellular adhesion of P. aeruginosa. This translated into significant changes in the architecture of the biofilm. We propose that Psl has an important role in P. aeruginosa adhesion, which is critical for initiation and maintenance of the biofilm structure.

A novel kind of parallel strand lumber (PSL) was produced, in which bamboo slivers with viscoelastic deformation were mixed with poplar veneer strands at a weight ratio of 1:4 as raw materials, then the flexural characteristics of PSL were investigated. The results showed that stretching treatment was an effective method to generate viscoelastic deformation of bamboo slivers, which would induce prestress in the bamboo slivers, reinforced PSL. The modulus of rupture (MOR) and modulus of elasticity (MOE) of prestressed bamboo slivers reinforced PSL were 7.84% and 11.76% higher than that of control PSL respectively. The flexural toughness index of prestressed bamboo slivers reinforced PSL was increased by 27.55% compared with control PSL. The presence of prestress in bamboo slivers reinforced PSL significantly enhanced its flexural stiffness and toughness, while it had no obvious effect on its flexural strength.

Aim: pH-sensitive liposomes (pSL) have emerged as promising nanocarriers due to their endo/lysosome-escape abilities, however, their pH sensitivity is compromised by poly(ethylene glycol) (PEG) coating. This study investigates whether an intracellular PEG-detachment strategy can overcome this PEG dilemma. Materials & methods: First, PEG2000 was conjugated with a phospholipid via an acid-labile hydrazide–hydrazone bond (–CO–NH–N = CH–), which was postinserted into pSL, forming PEG-cleavable pSL (CL-PEG-pSL). Their endo/lysosomal-escape abilities in MIA PaCa-2 cells, pharmacokinetics and tumor accumulation abilities were studied using PEG-pSL as reference. Results: CL-PEG-pSL showed rapid endo/lysosome-escape abilities in the cancer cells and higher tumor accumulation in MIA PaCa-2 xenograft model in contrast to PEG-pSL. Conclusion: Cleavable PEGylation is an efficient strategy to ameliorate the PEG dilemma of pSL for cancer drug delivery.

Parallel strand lumber (PSL) is an attractive structural wood composite which may have prospective use in building constructions. Conducting nonlinear analysis for the bending of PSL beams is a critical step in the determination of ultimate strength and deflection of them, which is an essential requirement of the building design philosophy based on probability of ultimate state. For the purposes of this article, an inelastic theoretical model regarding the load-carrying capacity of the PSL bending component has been developed. Based on the uniaxial loading tests, the stress-strain behaviors of PSL composite in the grain direction were measured. 4-point bending experiments were also performed in this study to investigate the failure mechanism of the PSL components. The results show that the tensile stress-strain relationship of PSL materials in the grain direction remains linear until breaking, while the compressive stress-strain relationship exhibits nonlinear characteristics once the compressive stress exceeds the proportional limit, which can be expressed by a quadratic polynomial. The failure mode of the PSL beam can be summarized that the fibres in the top of the broken section were buckling and those in the bottom of the section were broken when failure occurred. Significant nonlinear behavior was exhibited based on the load-deflection curves of the PSL beams. To predict the nonlinear bending performance of the PSL beams, a theoretical model that could consider the nonlinear stress-strain relations of PSL and predict the damage modes of the PSL beams was developed. Well agreements can be observed between the results of calculations and experiments.

Objective.Although prednisolone (PSL) and immunosuppressants are key drugs for Takayasu arteritis (TA) treatment, there is limited evidence on the optimal PSL dose. The aim of this study was to investigate the correlation between the initial PSL dose and relapse in TA.Methods.We enrolled 105 patients with TA who satisfied the criteria of the Japanese Circulation Society and American College of Rheumatology from 1990 to 2015. The clinical characteristics and outcomes of patients with TA were retrospectively evaluated. The relapse-free period was assessed according to the difference in initial treatments.Results.Relapse was observed in 57 (59.4%) of 96 patients treated with immunosuppressive therapy at diagnosis during a median followup of 56 months. Male sex and younger age of onset were significantly associated with relapse. Although ≤ 30 mg/day PSL monotherapy was preferably prescribed for patients with lower inflammatory markers, compared with > 30 mg/day (87.2% vs 52.6%), a significantly higher relapse rate was observed in the ≤ 30 mg/day group (HR 1.78; p = 0.047). Further, the relapse-free period was longer in patients treated with ≥ 50 mg/day PSL compared with those treated with ≤ 40 mg/day PSL. Combination therapy improved the relapse-free period compared with PSL monotherapy in the short term. The initial PSL dose was not associated with adverse events.Conclusion.A higher dose of PSL was associated with a significant decrease in the relapse rate. The effect of combination therapy on relapse needs to be further investigated. Lower-dose PSL monotherapy is an undesirable strategy for remission induction in TA, despite low disease activity.

We conclude by discussing the significance of PSL for organizational theory and leadership preparation and professional development.

AbstractWe introduce a new method to study rational conjugacy of torsion units in integral group rings using integral and modular representation theory. Employing this new method, we verify the first Zassenhaus conjecture for the group PSL(2, 19). We also prove the Zassenhaus conjecture for PSL(2, 23). In a second application we show that there are no normalized units of order 6 in the integral group rings of M10 and PGL(2, 9). This completes the proof of a theorem of Kimmerle and Konovalov that shows that the prime graph question has an affirmative answer for all groups having an order divisible by at most three different primes.

We reduce a case of the hidden subgroup problem (HSP) in $\SL$, $\PSL$, and $\PGL$, three related families of finite groups of Lie type, to efficiently solvable HSPs in the affine group $\AGL$. These groups act on projective space in an ``almost'' 3-transitive way, and we use this fact in each group to distinguish conjugates of its Borel (upper triangular) subgroup, which is also the stabilizer subgroup of an element of projective space. Our observation is mainly group-theoretic, and as such breaks little new ground in quantum algorithms. Nonetheless, these appear to be the first positive results on the HSP in finite simple groups such as $\PSL$.

Public Street Lighting (PSL) is a part of road complementary buildings installed on the left, right and/or in the middle of the road to illuminate the surrounding environment. PSLs in residential areas usually do not have a small amount, so it will be difficult if the supervision is done manually. Therefore, we need a system that can monitor and maintain PSL effectively and efficiently. This study aims to prove that the Operations Dashboard for ArcGIS (ODA) can be utilized in monitoring PSL based on Geographic Information Systems (GIS). This PSL monitoring system can be used by the government and private parties to conduct PSL maintenance activities more efficiently and effectively, by providing information about PSL in realtime through widgets contained in ODA, and with the help of a mobile application, Collector for ArcGIS (CFA) as a sensor replacement that functions to carry out data collection in the field. The results of this study indicate that ODA can be used to monitor PSL by showing information about the status, location, type, and number of PSLs damaged.

Which groups can be uniquely determined by the structure of their complex group algebras? It is proved that every quasisimple group except covers of the alternating groups is uniquely determined up to isomorphism by the structure of [Formula: see text], the complex group algebra of [Formula: see text]. One of the next natural groups to be considered are the characteristically simple groups. In this paper, as the first step in this investigation we prove that if [Formula: see text] is an odd prime number, then [Formula: see text] is uniquely determined by the structure of its complex group algebra.

The vertices of Γ(G) are the primes dividing the order of G and two distinct vertices p, q are joined by an edge if there is an element in G of order pq. It is proved that if p > 11 and p ≢ 1 (mod 12), then PSL(2,p) is uniquely determined by its prime graph. Also it is proved that if p > 7 is a prime number and Γ(G) = Γ(PSL(2,p2)), then G ≅ PSL(2,p2) or G ≅ PSL(2,p2).2, the non-split extension of PSL(2,p2) by ℤ2. In this paper as the main result we determine finite groups G such that Γ(G) = Γ(PSL(2,q)), where q = pk. As a consequence of our results we prove that if q = pk, k > 1 is odd and p is an odd prime number, then PSL(2,q) is uniquely determined by its prime graph and so these groups are characterizable by their prime graph.

Background:Previous cohort studies showed that the use of prednisolone (PSL) was a risk factor for clinical fractures in patients with rheumatoid arthritis (RA). However, there are few reports on relationship between PSL dose and clinical fractures.Objectives:The present study aimed to determine the effect of PSL dose on the incidence of clinical fractures in the RA patients treated with PSL.Methods:We evaluated anthropoetric parameters, bone mineral density (BMD), disease activity score 28-erythrocyte sedimentation rate (DAS28-ESR), RA medication (methotrexate (MTX) dose, use of biologic disease modified anti-rheumatic-drugs (bDMARDs), and PSL dose) and the incidence of clinical fractures during nine years in RA patients who participant the TOMORROW study (UMIN000003876), which is a 10-years prospective cohort study. Data on clinical fracture was self-reported on the questionnaires. In this analysis, the data of RA patients treated with PSL at least once during nine-year period were evaluated. We analyzed the average dose of PSL until the incidence of the clinical fractures. The risk factor for clinical fractures were analyzed by using Cox proportional hazard model with adjustment for age, sex, body mass index (BMI), and smoking history.Results:We analyzed the data of 67 RA patients treated with PSL. Among them, median age was 61.8 year, 56 patients (83.6%) were female, 47 patients (70.1%) were never smoker and median disease duration was 12.1 year. The number of patients treated with PSL at baseline was 48 (69.1%). During 9 years, 23 clinical fractures were observed in 67 patients, and the incidence of clinical fracture was 0.046/person-year. In 19 patients, who were not treated with PSL at baseline but treated with PSL at least once during 9 years, 5 clinical fractures were observed. In 67 RA patients, Cox proportional hazard analysis revealed that baseline disease activities, BMD at thoracic vertebrae and medication were not significant risk factors for clinical fractures. Background A previous report using a partial sciatic nerve ligation (PSL) model for neuropathic pain in rats demonstrated that consumption of soy-containing diets preoperatively and postoperatively suppressed development of mechanical and heat allodynia, as well as hyperalgesia. The current study examined whether dietary soy suppresses these neuropathic sensory disorders when consumed either before or after PSL injury. Methods Male Wistar rats were grouped into seven different feeding regimens. These rats were fed SOY (RMH-1000, PMI Feeds, St. Louis, MO), a diet containing 85% soy protein since weaning, and were then switched to noSOY (Bio-Serv Co., Frenchtown, NJ), a diet devoid of soy at certain time points before PSL injury (14, 7, 1 days, or 15 and 0 h). Postoperatively, these rats were fed SOY or noSOY diets. Levels of mechanical and heat allodynia and hyperalgesia were determined preoperatively and 3, 8, and 14 days after PSL injury. Results Compared with groups fed preoperative noSOY, consumption of SOY before PSL injury significantly blunted postoperative levels of allodynia and hyperalgesia. Administering the SOY diet both before and after PSL injury provided no additional suppression of neuropathic pain. No pain suppression was noted in rats fed a noSOY diet preoperatively and SOY diet after PSL injury. Switching from SOY to noSOY feeding within 15 h of PSL injury was sufficient to allow for the full development of allodynia and hyperalgesia. Conclusions Consumption of a soy-containing diet suppressed the development of neuropathic pain after PSL injury. The pain-suppressing properties of dietary soy were the result of a preemptive effect (i.e., when consumed preoperatively), but not a palliative effect (i.e., when consumed postoperatively). This effect of soy-containing diets appears to be short-lived, since switching to a noSOY diet 15 h before ligation abrogated the suppressive effect of soy.

ABSTRACTExopolysaccharide Psl is a critical biofilm matrix component inPseudomonas aeruginosa, which forms a fiber-like matrix to enmesh bacterial communities. Iron is important forP. aeruginosabiofilm development, yet it is not clearly understood how iron contributes to biofilm development. Here, we showed that iron promoted biofilm formation via elevating Psl production inP. aeruginosa. The high level of iron stimulated the synthesis of Psl by reducing rhamnolipid biosynthesis and inhibiting the expression of AmrZ, a repressor ofpslgenes. Iron-stimulated Psl biosynthesis and biofilm formation held true in mucoidP. aeruginosastrains. Subsequent experiments indicated that iron bound with Pslin vitroand in biofilms, which suggested that Psl fibers functioned as an iron storage channel inP. aeruginosabiofilms. Moreover, among three matrix exopolysaccharides ofP. aeruginosa, Psl is the only exopolysaccharide that can bind with both ferrous and ferric ion, yet with higher affinity for ferrous iron. Our data suggest a survival strategy ofP. aeruginosathat uses exopolysaccharide to sequester and store iron to stimulate Psl-dependent biofilm formation.IMPORTANCEPseudomonas aeruginosais an environmental microorganism which is also an opportunistic pathogen that can cause severe infections in immunocompromised individuals. It is the predominant airway pathogen causing morbidity and mortality in individuals affected by the genetic disease cystic fibrosis (CF). Increased airway iron and biofilm formation have been proposed to be the potential factors involved in the persistence ofP. aeruginosain CF patients. Here, we showed that a high level of iron enhanced the production of the key biofilm matrix exopolysaccharide Psl to stimulate Psl-dependent biofilm formation. Our results not only make the link between biofilm formation and iron concentration in CF, but also could guide the administration or use of iron chelators to interfere with biofilm formation inP. aeruginosain CF patients. Furthermore, our data also imply a survival strategy ofP. aeruginosaunder high-iron environmental conditions.
