Deorphanization of ORL-1/LC132 in 1995 by reverse pharmacology in two simultaneously published landmark studies added a new member to the opioid family of G-protein coupled receptors. Meunier and Reinscheid used cells expressing recombinant ORL-1 (human) or LC132 (rat) and the presumed intracellular inhibition of cyclic AMP formation to ‘fish’ for endogenous peptide ligands in rat whole-brain and pig hypothalamic extracts. Both studies reported the isolation of a 17-amino-acid peptide, which was named nociceptin and orphanin FQ by the two authors, respectively. The behaviour of the isolated peptide was a complete surprise, as a general hyperalgesia was observed when the peptide was administered at supraspinal sites. We now know that this peptide has, in fact, anti-opioid action, particularly in the medulla. The endogenous peptide exerts a multitude of effects both in the nervous system and, unlike classical opioids, has efficacy in neuropathic pain.

This book is the first in-depth examination of revenge in the Odyssey. The principal revenge plot of the Odyssey—Odysseus’ surprise return to Ithaca after twenty years away and his vengeance on Penelope’s suitors—is the act for which he is most celebrated. This story forms the backbone of the Odyssey. But is Odysseus’ triumph over the suitors as univocally celebratory as is often assumed? Does the poem contain and even suggest other, darker interpretations of Odysseus’ greatest achievement? This book offers a careful analysis of several other revenge plots in the Odyssey—those of Orestes, Poseidon, Zeus, and the suitors’ relatives. It shows how these revenge stories color one another with allusions (explicit and implicit) that connect them and invite audiences to interpret them in light of one another. These stories—especially Odysseus’ revenge upon the suitors—inevitably turn out to have multiple meanings. One plot of revenge slips into another as the offender in one story becomes a victim to be avenged in the next. As a result, Odysseus turns out to be a much more ambivalent hero than has been commonly accepted. And in the Odyssey’s portrayal, revenge is an unstable foundation for a community. Revenge also ends up being a tenuous narrative structure for an epic poem, as a natural end to cycles of vengeance proves elusive. This book offers a radical new reading of the seemingly happy ending of the poem.

Results of the randomized, international INSIGHT START clinical trial provide definitive proof of the benefit of antiretroviral therapy initiation in asymptomatic individuals with CD4+ counts greater than 500 cells/mm3. There are six different classes of antiretroviral agents: two types of reverse transcriptase inhibitors, two types of entry inhibitors, one class of inhibitors of HIV protease, and one class of inhibitors of HIV integrase. Combination antiretroviral therapy is recommended for all people living with HIV. The primary goal of combination antiretroviral therapy is to achieve viral suppression. Each antiretroviral class targets a unique step in the replication cycle of HIV-1.

This chapter critically evaluates the likelihood that planet Earth will cross one or more global environmental tipping points, resulting in a degraded state that would be difficult to reverse. Ecological tipping points occur when components of a system change rapidly due an initial forcing that is amplified by positive feedbacks, resulting in a regime shift. The chapter examines the evidence in support of biological and geophysical boundaries that clearly delimit a “safe operating space” for people and biodiversity. For individual ecosystems, abrupt state transitions have been documented. However, apart from the climate system, there is scant evidence (or theoretical justification) to support the view that global aggregates like biodiversity, chemical cycles, or resource extraction have planetary thresholds that define the boundaries of a global safe operating space. Acknowledging the absence of clear evidence for thresholds or boundaries at the global level does not diminish the seriousness of anthropogenic impacts. It does, however, imply that local-scale mitigation actions will be most effective.

The emphasis of this chapter is on how mines are taxed, not on how tax revenues should be used. However, it is noted that one of the influencing factors that drives fiscal system policy evolution is the issue of whether subnational governments should have additional taxing power or be given special revenue dispensation. It is the author’s experience that devolving additional tax power to subnational governments is often not possible because of constitutional constraints or because central authorities wish to maintain control. The chapter provides an introduction to the various taxation approaches applied to the mining sector and it includes a description of the principal tax types and investment tax incentives. The author concludes that when devising mineral sector fiscal systems governments should carefully assess their fiscal options using a holistic approach that anticipates commodity price cycles, and that mining companies should anticipate fiscal system changes.

This book reviews the rapid rise of crime and violence in Latin America over the last few decades and offers an explanation to a striking paradox: In the midst of decreasing poverty, economic growth, and democratization crime has risen throughout the region. Drawing from large data sets, I argue that this is because crime has become a profitable industry in weak states with outdated criminal justice systems unable to withstand the challenge posed by new criminal enterprises. Prosperity has enhanced consumer demand for illicit goods, fueling the growth of secondary and illegal markets, including markets for stolen goods and narcotics that can provide an income for millions of youngsters willing to take the risk of arrest and loss of life. While some countries have experienced moderate increases in criminality others have experienced catastrophic rates of violence, resulting in two types of stable equilibria: Low- and high-crime countries. I explain why different equilibria, between the profit opportunities provided by criminality and a weak criminal justice system, have triggered a rapid upward spiral of crime and a sharp increase in the intensity of violence in some states but a moderate upward trend in others, and why certain countries have transitioned from low- to high-crime environments with vicious cycles of high criminality that are very difficult to reverse. The resulting severe, undesired outcomes are studied in this book: serious predatory crime diversification, consolidation of organized crime, ineffective justice reforms, weak policing, and overcrowded prisons.

The Spanish composer Sebastián de Vivanco (ca. 1551–1622) was born, like his revered contemporary Tomás Luis de Victoria, in Avila. Having secured prestigious cathedral and university posts at Salamanca, Vivanco saw through the press, between 1607 and 1614, three luxury choirbooks containing 18 Magnificats, 10 masses, and 72 motets, spread over a total of more than 900 printed pages. The first of these choirbooks, all of which were printed by the Fleming ArtusTaberniel and his wife Susana Muñoz, is a cycle of Magnificats providing polyphony for the odd- and even-numbered verses in all eight tones, plus one extra Magnificat in each of the much-used first and eighth tones. If Vivanco has been eclipsed for too long by his great contemporary and compatriot, it is in the complexity and ingenuity of the many canons to be found in these Magnificats that Vivanco outshines even Victoria.

Abdominal injuries are common following blunt and penetrating trauma. They can result in a spectrum of severity from benign to potentially life-threatening conditions. Soon after injury, haemorrhage is the predominant concern, and leading cause of morbidity and mortality. Active haemorrhage resulting in shock requires emergent operative intervention and aggressive haemostatic resuscitation. However haemodynamically-stable patients benefit from non-operative management of solid organ injuries with or without angiographic embolization. Sepsis usually occurs as a result of intra-abdominal infections from missed bowel perforations or anastomotic leaks. Sterile systemic hyperinflammatory conditions can result from major hepatic necrosis or pancreatic injuries, and closely mimic infectious conditions. Damage control surgery is a valuable adjunct to the operative management of major abdominal trauma. This concept recognizes that the time and procedures required to perform definitive operative repair may be detrimental when physiological derangements are excessive. By limiting operations to controlling haemorrhage and enteric contamination, further deterioration, and the ‘vicious bloody cycle of trauma’ can be avoided. The operative and critical care management of patients with abdominal trauma should be closely integrated to correct physiological derangements with rapid stabilization and reversal of hypoperfusion. Abdominal compartment syndrome, characterized by intra-abdominal hypertension and resultant remote organ dysfunction, is a risk in patients undergoing high-volume fluid resuscitation. Emergent decompressive laparotomy is indicated in patients with abdominal compartment syndrome and results in rapid reversal of physiological compromise. Paramount to optimal management of abdominal injuries is the close integration of operative and critical care approaches.

The governance of natural resource wealth is considered to constitute a key determinant in whether the extraction of natural resources proves to be a blessing or a curse. In response to this challenge, a variety of international initiatives have emerged to codify successful policies pursued by countries, and promote global norms and best practices to guide decision-makers. These initiatives, such as the Extractives Industry Transparency Initiative, have seen success in spreading and embedding governance norms, ranging across revenue transparency, contract disclosure, and the creation of instruments such as resource funds and building institutions for checks and balances. However, evidence for causal impact remains weak and sometimes limited to anecdotal cases. The end of the super-cycle of commodity prices, and the prospect of permanently lower prices for fossil fuels, creates new challenges for resource-rich countries but may also allow space and time for reflection, lesson-learning and improvements in governance.

Lifeline for about one-sixth of the world’s population in the subcontinent, the Indian summer monsoon (ISM) is an integral part of the annual cycle of the winds (reversal of winds with seasons), coupled with a strong annual cycle of precipitation (wet summer and dry winter). For over a century, high socioeconomic impacts of ISM rainfall (ISMR) in the region have driven scientists to attempt to predict the year-to-year variations of ISM rainfall. A remarkably stable phenomenon, making its appearance every year without fail, the ISM climate exhibits a rather small year-to-year variation (the standard deviation of the seasonal mean being 10% of the long-term mean), but it has proven to be an extremely challenging system to predict. Even the most skillful, sophisticated models are barely useful with skill significantly below the potential limit on predictability. Understanding what drives the mean ISM climate and its variability on different timescales is, therefore, critical to advancing skills in predicting the monsoon. A conceptual ISM model helps explain what maintains not only the mean ISM but also its variability on interannual and longer timescales.The annual ISM precipitation cycle can be described as a manifestation of the seasonal migration of the intertropical convergence zone (ITCZ) or the zonally oriented cloud (rain) band characterized by a sudden “onset.” The other important feature of ISM is the deep overturning meridional (regional Hadley circulation) that is associated with it, driven primarily by the latent heat release associated with the ISM (ITCZ) precipitation. The dynamics of the monsoon climate, therefore, is an extension of the dynamics of the ITCZ. The classical land–sea surface temperature gradient model of ISM may explain the seasonal reversal of the surface winds, but it fails to explain the onset and the deep vertical structure of the ISM circulation. While the surface temperature over land cools after the onset, reversing the north–south surface temperature gradient and making it inadequate to sustain the monsoon after onset, it is the tropospheric temperature gradient that becomes positive at the time of onset and remains strongly positive thereafter, maintaining the monsoon. The change in sign of the tropospheric temperature (TT) gradient is dynamically responsible for a symmetric instability, leading to the onset and subsequent northward progression of the ITCZ. The unified ISM model in terms of the TT gradient provides a platform to understand the drivers of ISM variability by identifying processes that affect TT in the north and the south and influence the gradient.The predictability of the seasonal mean ISM is limited by interactions of the annual cycle and higher frequency monsoon variability within the season. The monsoon intraseasonal oscillation (MISO) has a seminal role in influencing the seasonal mean and its interannual variability. While ISM climate on long timescales (e.g., multimillennium) largely follows the solar forcing, on shorter timescales the ISM variability is governed by the internal dynamics arising from ocean–atmosphere–land interactions, regional as well as remote, together with teleconnections with other climate modes. Also important is the role of anthropogenic forcing, such as the greenhouse gases and aerosols versus the natural multidecadal variability in the context of the recent six-decade long decreasing trend of ISM rainfall.

The world's largest demonstrator of a revolutionary energy system in desalination for drinking water production is in operation. MIDES uses Microbial Desalination Cells (MDC) in a pre-treatment step for reverse osmosis (RO), for simultaneous saline stream desalination and wastewater treatment. MDCs are based on bio-electro-chemical technology, in which biological wastewater treatment can be coupled to the desalination of a saline stream using ion exchange membranes without external energy input. MDCs simultaneously treat wastewater and perform desalination using the energy contained in the wastewater. In fact, an MDC can produce around 1.8 kWh of bioelectricity from the energy contained in 1 m3 of wastewater. Compared to traditional RO, more than 3 kWh/m3 of electrical energy is saved. With this novel technology, two low-quality water streams (saline stream, wastewater) are transformed into two high-quality streams (desalinated water, treated wastewater) suitable for further uses. An exhaustive scaling-up process was carried out in which all MIDES partners worked together on nanostructured electrodes, antifouling membranes, electrochemical reactor design and optimization, life cycle assessment, microbial electrochemistry and physiology expertise, and process engineering and control. The roadmap of the lab-MDC upscaling goes through the assembly of a pre-pilot MDC, towards the development of the demonstrator of the MDC technology (patented). Nominal desalination rate between 4-11 Lm-2h-1 is reached with a current efficiency of 40 %. After the scalability success, two MDC pilot plants were designed and constructed consisting of one stack of 15 MDC pilot units with a 0.4 m2 electrode area per unit. The existence of a political executive is a key feature of any political system: from eminently democratic regimes to totalitarian dictatorships, and every shade in between. In recent decades the long-term trend of putting constitutional and democratic constraints on the political executive is reversed. In addition to an autocratic turn in some countries, internationalization, securitization and a growing need for coordination shift the balance between governments and other political institutions. This re-empowerment of the political executive has contributed to its rediscovery in political science. As the 37 chapters in this volume demonstrate, the past two decades have seen an impressive outpouring of research on political executives. Edited and written by 49 of the most distinguished scholars in the field, this Oxford Handbook of Political Executives combines substantive stocktaking with setting new agendas for the next generation of political executive research. The book is organized around five themes. Part I, Theorizing and Researching Political Executives is devoted to the theoretical and methodological approaches in the study of political executives. Part II looks at The Composition and Life Cycle of Political Executives, from the formation to the termination of successive governments. Part III discusses The Dynamics and Developments within the Executive, such as hierarchical relations and internal political processes. Part IV focusses on The Dynamics and Developments between the Executive and the Broader Political Context, including interactions with the bureaucracy and parliament. Part V is devoted to Political Executives Beyond the Democratic Nation State, such as the European Union and autocratic regimes.
