In the first part, we study quadratic Leibniz algebras. First, we prove that these algebras are symmetric. Then, we use the T*-extension and the double extension to prove some properties of this type of Leibniz algebras. Besides, since we observe that the skew-symmetry of the Leibniz bracket gives rise to other types of invariance for a bilinear form on a Leibniz algebra: The left invariance and the right invariance. We focus on the study of left (resp. right) Leibniz algebras with symmetric, non degenerate and left (resp. right) invariant bilinear form. In particular, we prove that these algebras are Lie admissibles. The second part of this work is dedicated to the study of quadratic Lie triple systems and pseudo-euclidien Jordan triple systems. Texto completoThe goal of this thesis is to explore relative Rota-Baxter operators in the context of ternary algebras of both Lie and Jordan types. We mainly consider Lie triple systems, 3-Lie algebras and ternary Jordan algebras. The study covers their structure, cohomology, deformations, and their connection with the Yang-Baxter equations. The work is divided into three main parts. The first part aims first to introduce and study a graded Lie algebra whose Maurer-Cartan elements are Lie triple systems. It turns out to be the controlling algebra of Lie triple systems deformations and fits with the adjoint cohomology theory of Lie triple systems introduced by Yamaguti. In addition, we introduce the notion of relative Rota-Baxter operators on Lie triple systems and construct a Lie 3-algebra as a special case of L∞-algebras, where the Maurer-Cartan elements correspond to relative Rota-Baxter operators. In the second part, we introduce the concept of twisted relative Rota-Baxter operators on 3-Lie algebras and construct an L∞-algebra, where the Maurer-Cartan elements are twisted relative Rota-Baxter operators. This allows us to define the Chevalley-Eilenberg cohomology of a twisted relative Rota-Baxter operator. In the last part, we deal with a representation theory of ternary Jordan algebras. In particular, we introduce and discuss the concept of coherent ternary Jordan algebras. We then define relative Rota-Baxter operators for ternary Jordan algebras and discuss solutions ofthe ternary Jordan Yang-Baxter equation involving relative Rota-Baxter operators. Moreover, we investigate ternary pre-Jordan algebras as the underlying algebraic structure of relative Rota-Baxter operators

En dernier lieu, nous étudions la conjecture de Helgason pour la frontière de Shilov des espaces hermitiens symétriques. Plus précisément, nous caractérisons l'image par de la transformation de Poisson des hyperfonctions et des fonctions $L^p$ sur la frontière de Shilov.

Texto completoBreast cancer (BC) is a highly complex, heterogeneous and multifactorial disease. Compared to other subtypes, triple negative (TNBC) represents one of the most advanced stages of the disease, and is characterized by a strong inter- and intra-tumor heterogeneity and a high mutational burden, making treatments very difficult and inefficient. The absence of estrogen and progesterone receptors expression, and the absence of HER2 receptor overexpression, makes chemotherapy (example: doxorubicin), the prior treatment in patients with this type of BC. Unfortunately, the complete anatomopathological response rate, in response to these chemotherapies, rarely exceeds 50% with a long-term benefit affecting only 30 to 50% of patients. The LIIC laboratory in which I did my thesis aims to show that a donor of nitric oxide (NO), glyceryl trinitritrate (GTN), a drug used in cardiology, can improve the therapeutic efficacy of anti-cancerous chemotherapy. The aim of my thesis project was therefore to determine whether the association of GTN with doxorubicin could potentiate the antitumor activity of this chemotherapy in TNBC, and to determine the mechanism by which this combination acts by emphasizing the immune microenvironment in general, and more particularly on immunosuppressive myeloid cells, MDSCs.The results showed that the association of GTN with doxorubicin significantly improved the anti-tumor efficacy of this chemotherapy, in a TNBC model induced by the injection of 4T1 breast cells. This effect is due, in part, to the ability of GTN to increase the differentiation of LTCD4 + in the anti-tumor linage Th1, to increase the intra-tumor recruitment of CD8 + / PD-1 + cells and of G-MDSCs down-expressing PD-L1. Nevertheless, the main mechanism by which GTN acts is essentially based on its ability to reprogram, in the presence of ROS, these G-MDSCs towards a less immunosuppressive phenotype. Indeed, the results revealed that GTN, via the S-nitrosylation of STAT5, was able to modulate the lipid metabolism of these cells, which is dependent on the protein FATP2 (Fatty Acid Transport Protein 2). Thus, a decrease in FATP2, as well as all the upstream (STAT5) and downstream (PGE2) components of this signaling pathway was observed in response to GTN +/- doxorubicin. All of these effects were inhibited in the presence of an ROS inhibitor, the N-acetyl cysteines (NAC). NAC also significantly delayed tumor progression when used in combination with doxorubicin / GTN.This work, based on the use of two molecules commonly used in the clinic, undoubtedly opens up a new treatment perspective for patients with TNBC and this combination can quickly be offered to clinicians

Texto completoTwo different topics are encountered in this PhD thesis : on the one hand, the study of foliations using groupoids, and on the other, energy functions for Morse-Smale systems. In the first part, the links between transverse and global models for the holonomy of a foliation (respectively germs and foliation groupoids) are explicited. A notion of compact generation is defined for a large class of topological groupoids, which extends Andr´e Haefliger’s definition for transverse holonomy in a Morita-invariant way. This new definition is better fitted for global foliation groupoids. In the second part, we explain how to reorder the critical values of a Morse-Lyapunov function for a discrete Morse-Smale system; whenever it is sufficiently non-degenerate. The described method uses the suspension of the system, on which is used an adapted version of a general Lyapunov reordering method, called shearing. This adapted version involves a precise control of the behaviour of a Bott-Lyapunov function near a periodic orbit of a Morse-Smale field

Texto completoThe intricate interplay between the immune response and tumor progression, particularly in the context of triple-negative breast cancer (TNBC), has been a focal point of oncological research over the last decade. Our preceding investigation highlighted the role of the HLF gene within a quartet gene signature, identifying its inverse association with the infiltration of immune cells within the tumor milieu. During my thesis, we investigated more precisely its implication in the immune response and TNBC tumor development. The bioinformatic analysis of clinical data from patients with TNBC demonstrated that low levels of HLF expression were associated with an overexpression of genes related to the mechanisms of attraction and activation of the immune system. The implication of neutrophils in cancer is contentious, as they can function either as active anti-tumoral partners or as pro-tumoral agents promoting immunosuppression and tumor growth. The duality of the immune system’s role in cancer, oscillating between antitumor action and support for tumor growth, reflects its great complexity within the tumor microenvironment. In this study, we employed CRISPR/Cas9 technology to inactivate the HLF gene in TNBC cell lines, enabling us to explore the repercussions on the chemokine network regulating the presence and activity of different immune populations in the tumor microenvironment. Finally, in vivo experiments conducted on immunodeficient mouse models demonstrated that reducing HLF in certain TNBC subtypes hindered tumor progression. Altogether, our results suggest that a reduction of HLF expression can lead in the early steps of tumor development to the awakening of the anti-tumoral function of the immune system to enforce the necessary immunological tone to tumor elimination. Thus, HLF emerges as a promising therapeutic target to promote the engagement and effectiveness of a competent immune response against aggressive subtypes of TNBC

Texto completoTriple negative breast cancer (TNBC) is the most aggressive form of breast cancers. It accounts for 15-20% of breast cancers. No progress in survival has been achieved since the advent of standard chemotherapy protocols. TNBC is an important clinical challenge. They have the worst outcome among breast cancer subgroups. Given their poor prognosis, their assumed hetetogeneity, and absence of any alternative specific targeted therapy, chemotherapy remains the only TNBC treatment. Despite an often good initial response to treatment, more than a half of patients do not achieve a pathological complete response, with a frequent and fast tumor relapse. Several therapeutic approaches have been identified preclinically, but none of these molecules have been shown to be effective on all of these patients. There is a urge for the identification of new treatments.An interesting strategy is the repurposing of medical compounds that were initially not intended for the treatment of a given disease. This strategy takes advantage of the extremely expensive initial research and development effort. This process is potentially efficient and cost-effective as previous clinical trials have been performed and pharmacokinetics/pharmacodynamics and toxicity have been already explored. In order to develop new treatment schemes we addressed the following question: Is there available drugs with strong activity in TNBC? To do so, we performed a high-throughput drug screening on 12 TNBC cell lines to reflect the dramatic heterogeneity of the disease. From this drug discovery program, several interesting compounds were identified with significant anti-tumor potential against TNBC. More particularly, psychoactive compounds regulating serotonin metabolism (ie antidepressant drugs and notably serotonin selective reuptake inhibitors-SSRIs) were found to be highly effective “hits”.My thesis work turned to the comprehension of serotonin implication in TNBC physiopathology to understand if modulating its metabolism could be of therapeutic interest for TNBC management. Different biological aspects were investigated concerning serotonin effects on TNBC cellular models (serotonin adjunction in vitro or endogenous synthesis inhibition). In addition, I established a comprehensive map of the serotonergic landscape in TNBC (biosynthetic capacity, transporters, receptors) that led to the identification of therapeutic targets that would be of interest in the treatment of cancer: HTR1D and HTR1B. Indeed, by blocking these promising targets (with chemical inhibitors or siRNA knockdown) we observed a strong reduction in cell viability in our large panel of TNBC cell lines. Remarkably, we found that their expression levels were associated to poor prognosis in breast cancer, and notably in TNBC subtype with huge dichotomy observed in the outcome, allowing future stratification of TNBC patient management and selection for further targeted therapies. These results pinpoint HTR1D and HTR1B as strong prognosis biomarkers in TNBC. Immunohistochemistry staining was also conducted to confirm the presence of these targets at the protein level in tumor samples. Moreover, I could identify a microRNA regulating one of these receptors: has-miR-599. Consistently, expression levels of this microRNA demonstrated a prognostic impact on TNBC survival. While ex vivo data of one SSRI and the dual antagonist of HTR1D/HTR1D receptors shown encouraging efficacy, their preclinical evaluation assessed in a TN PDX model could not allow to demonstrate any significant effect on tumor growth in vivo. As a matter of fact, serotonin metabolism is a complex system and TNBC heterogeneity does not permit to conclude on the therapeutic proof of concept of the serotonergic modulation in TNBC with this first attempt. Texto completoThe Corporate social responsibility (CSR) is the concept according to which the companies have an obligation towards social actors others that the shareholders and beyond the legal and lawful regulations. In this concept, the definition of the performance is not only economic and financial. Indeed, the RSE corresponds to the application to the companies of the sustainability concept; it invites them to take into account, beyond the economic and financial aspects, the social and environmental aspects of their activities. The management control is in the middle of this relation between global responsibility and economic performance for the companies. Our questionings are interested in the step to adopt by a company to implement sustainability trategy and objectives which are incarnated in the management instrumentation, like with the design and the implementation of piloting tools adapted to measure a multidimensional performance. We carried out three years of research intervention within Batigère, a group of Social Firm for Housing leader on his sector, within the framework of a CIFRE contract (Industrial Convention of Formation by Research). Batigère posts clearly in its strategic orientations sustainability concerns centered on three dimensions: the economic one, the social one and the environmental one. Our research was done in two phases: an exploratory phase during which we studied indepth Batigère operation Batigère like its instrumentation of management, and a phase of ingenieric research during which we took part in the process of design and implementation of his system of scorecards. Initially, our case study highlighted the possibility of developing a coherent step of global responsibility really incarnating itself in the management instrumentation. With our opinion, the tensions which can remain between economic performance and global responsibility must be assumed and managed within the framework of the total system of control of the company, if one is interested in the effectiveness of the policies implemented in the social field. In the second time, the results of our of ingenieric research show that for the design of a system of scorecards integrating the CSR, two approaches OVAR and Balanced Scorecard (BSC), although they have dissimilarities, can be complementary. Indeed, by combining BSC tool and its forms of balance with the coherent and participative step recommended by OVAR method, we could implement a coherent scorecards system allowing the piloting of a multidimensional performance. In the third time, we prolonged our research on the appropriation by the actors of the scorecards system lately designed. It is indeed the play of the individuals activating the management instruments which is essential. We show through quotations drawn from the talks carried out with the actors as well as extracts of emails received during tool in situ operation that: firstly, the scorecards system contributed to the homogenization of the efficiency representation and the performance criteria within the company, and secondly, it allowed to legitimate the function of the financial controller by offering an adequate tool for the financial piloting of the operations while widening control towards a multidimensional performance. We show finally that technological dimensions (design features and automation parameters) of a management tool have a considerable impact on the assimilation by the actors of a tool lately designed. We qualified the step of CSR integration in the system of control implemented within Batigère of “combinative method”. It has two specificities: a coherent system of control which articulate financial performance and social performance, and a step of systems design of scorecards which ensure a vertical and horizontal coherence. We tried to model this combinative method, by specifying the principles subjacent with each stage of the method as well as the actors having taken part in the process, in order to register the contribution of our ingenieric research in the production of knowledge

L'étude des propriétés des hadrons dans la matière nucléaire est un sujet d'intérêt actuel. Les kaons chargés positivement possèdent un anti-quark étrange qui ne peut pas être réabsorbé et constituent donc une sonde intéressante du milieu nucléaire.
La première partie de cette thèse étudie les sections efficaces de production de pions et de kaons lors de collisions proton noyau pour une énergie incidente de l'ordre du GeV.
Dans un premier temps, le modèle de Dynamique Quantique Moléculaire utilisé pour simuler les collisions proton noyau est décrit. Puis, les sections efficaces totales des processus élémentaires implémentées dans le modèle sont présentées. Ensuite, les effets de la fonction spectrale du nucléon sur la production de pions et de kaons sont mis en évidence dans les sections efficaces doublement différentielles. Finalement, les conséquences des corrélations observées entre les nucléons du noyau sont discutées.
II - Mesures de sections efficaces de production de particules chargées légères
lors de réactions induites par protons de 62,9 MeV sur cible de plomb
Pour développer des options nouvelles de gestion des déchets nucléaires, des recherches sont menées sur les systèmes hybrides. La seconde partie de cette thèse s'inscrit dans le cadre des mesures de données nucléaires liées à la conception de tels systèmes. Un pouvoir prédictif suffisant des codes théoriques, de 20 à 150 MeV, nécessite de nouvelles mesures de sections efficaces pour les contraindre.
L'expérience réalisée a pour but de déterminer les sections efficaces de production de particules chargées légères (p, d, t, 3He, a) lors de réactions induites par protons de 62,9 MeV sur une cible de plomb 208Pb.
Le contexte général de cette partie est présenté en premier lieu. Puis, le dispositif expérimental composé de 7 télescopes triple (Si, Si, CsI(Tl)) est décrit. Les chapitres suivants sont consacrés au dépouillement des données (identification des particules, étalonnage des détecteurs) et à l'extraction des sections efficaces. Finalement, une étude comparative de nos résultats expérimentaux à ceux des codes théoriques est menée.

Texto completoSymptoms of post-traumatic stress disorder (PTSD) are a common comorbidity in veterans seeking treatment of chronic musculoskeletal pain (CMP). However, little is known regarding the mutual influence of PTSD and CMP in this population. Using cross-sectional and longitudinal data from a randomized clinical trial evaluating a stepped care intervention for CMP in Iraq/Afghanistan veterans (ESCAPE), this dissertation examined the relationships between PTSD and CMP along with other factors including depression, anxiety, catastrophizing and health-related quality of life. The Classification and Regression Tree (CART) analysis was conducted to identify key factors associated with baseline PTSD besides CMP severity. A series of statistical analyses including logistical regression analysis, mixed model repeated measure analysis, confirmatory factor analysis and cross-lagged panel analysis via structural equation modeling were conducted to test five competing models of PTSD symptom clusters, and to examine the mutual influences of PTSD symptom clusters and CMP outcomes. Results showed baseline pain intensity and pain disability predicted PTSD at 9 months. And baseline PTSD predicted improvement of pain disability at 9 months. Moreover, direct relationships were found between PTSD and the disability component of CMP, and indirect relationships were found between PTSD, CMP and CMP components (intensity and disability) mediated by depression, anxiety and pain catastrophizing. Finally, the coexistence of PTSD and more severe pain was associated with worse SF-36 Physical Component Summary (PCS) and Mental Component Summary (MCS) scores. Together these findings provided empirical support for the mutual maintenance theory.
