Abstract This study is designed to clarify the role of an orphan nuclear hormone receptor, RORα, on thyroid hormone (TH) receptor (TR)-mediated transcription on a TH-response element (TRE). A transient transfection study using various TREs [i.e., F2 (chick lysozyme TRE), DR4 (direct repeat), and palindrome TRE] and TR and RORα1 was performed. When RORα1 and TR were cotransfected into CV1 cells, RORα1 enhanced the transactivation by liganded-TR on all TREs tested without an effect on basal repression by unliganded TR. By electrophoretic mobility shift assay, on the other hand, although RORα bound to all TREs tested as a monomer, no (or weak) TR and RORα1 heterodimer formation was observed on various TREs except when a putative ROR-response element was present. The transactivation by RORα1 on a ROR-response element, which does not contain a TRE, was not enhanced by TR. The effect of RORα1 on the TREs is unique, because, whereas other nuclear hormone receptors (such as vitamin D receptor) may competitively bind to TRE to exert dominant negative function, RORα1 augmented TR action. These results indicate that RORα1 may modify the effect of liganded TR on TH-responsive genes. Because TR and RORα are coexpressed in cerebellar Purkinje cells, and perinatal hypothyroid animals and RORα-disrupted animals show similar abnormalities of this cell type, cross-talk between these two receptors may play a critical role in Purkinje cell differentiation.

We studied the effects of thyroid hormone (T3) on nuclear protein-DNA interactions by using dimethyl sulfate (DMS) and DNase I ligation-mediated PCR footprinting. We examined an endogenous gene the growth hormone (GH) gene, and a stably transfected plasmid containing the chicken lysozyme silencer (F2) T3 response element (TRE) gene, F2-TRE-TK-CAT, both in pituitary tumor (GC) cells. The 235-1 cell line, which expresses prolactin (PRL) and Pit-1, but not the T3 receptor (TR) or GH, was used as a control. DMS and DNase I footprinting identified protected G residues in the Pit-1, Sp1, and Zn-15 binding sites of the GH gene in GC, but not in 235-1, cells. There was no specific protection of the tripartite GH TRE at -180 bp against either DMS or DNase I in the absence or presence of T3 in either cell line. However, T3 increased protection of the Pit-1 and Sp1 binding sites against DMS in GC cells. In GC cells stably transfected with a plasmid containing F2-TRE-TK-CAT or TRalpha, chloramphenicol acetyltransferase expression was T3 inducible and DMS footprinting revealed both F2 TRE TR-binding half sites in a pattern suggesting the binding of TR homodimers before and during T3 exposure. We conclude that the GH gene is accessible to specific nuclear proteins in GC, but not in 235-1, cells and that T3 enhances this interaction, although there is no evidence of TR binding to the low-affinity rat GH TRE. The presence of TR binding to the high-affinity F2 TRE before and during T3 exposure suggests that reversible interaction of T3 with DNA-bound TRs, rather than transient T3-TR contact with TREs, determines the level of T3-stimulated transcriptional activation.

The THRB gene encodes the well-described thyroid hormone (T3) receptor (TR) isoforms TRβ1 and TRβ2 and two additional variants, TRβ3 and TRΔβ3, of unknown physiological significance. TRβ1, TRβ2, and TRβ3 are bona fide T3 receptors that bind DNA and T3 and regulate expression of T3-responsive target genes. TRΔβ3 retains T3 binding activity but lacks a DNA binding domain and does not activate target gene transcription. TRΔβ3 can be translated from a specific TRΔβ3 mRNA or is coexpressed with TRβ3 from a single transcript that contains an internal TRΔβ3 translation start site. In these studies, we provide evidence that the TRβ3/Δβ3 locus is present in rat but not in other vertebrates, including humans. We compared the activity of TRβ3 with other TR isoforms and investigated mechanisms of action of TRΔβ3 at specific thyroid hormone response elements (TREs) in two cell types. TRβ3 was the most potent isoform, but TR potency was TRE dependent. TRΔβ3 acted as a cell-specific and TRE-dependent modulator of TRβ3 when coexpressed at low concentrations. At higher concentrations, TRΔβ3 was a TRE-selective and cell-specific antagonist of TRα1, -β1, and -β3. Both TRβ3 and TRΔβ3 were expressed in the nucleus in the absence and presence of hormone, and their actions were determined by cell type and TRE structure, whereas TRΔβ3 actions were also dependent on the TR isoform with which it interacted. Analysis of these complex responses implicates a range of nuclear corepressors and coactivators as cell-, TR isoform-, and TRE-specific modulators of T3 action.

Technology-rich learning environments (TREs) provide opportunities for learners to engage in complex interactions involving a multitude of cognitive, metacognitive, and affective states. Understanding learners’ distinct learning progressions in TREs demand inquiry approaches that employ well-conceived theoretical accounts of these multiple facets. The present study investigated learners’ interactions with BioWorld, a TRE developed to guide students’ clinical reasoning through diagnoses of simulated patients. We applied person-oriented analytic methods to multimodal data including verbal protocols, questionnaires, and computer logs from 78 task solutions. Latent class analysis, clustering methods, and latent profile analysis followed by logistic regression analyses revealed that students’ clinical diagnosis ability was positively correlated with advanced self-regulated learning behaviors, high confidence and cognitive strategy use, critical attention to experts’ feedback, and their positive emotional responses to feedback. The study results have the potential to contribute to a theory-guided approach to designing TREs with a data-driven assessment of multidimensional growth. Building on the study results, we introduce and discuss an ecological learner model for assessing multidimensional learner traits which can be used to design a TRE for adaptive scaffolding.

Fatty acid synthase (FAS) is a key enzyme of hepatic lipogenesis responsible for the synthesis of long-chain saturated fatty acids. This enzyme is mainly regulated at the transcriptional level by nutrients and hormones. In particular, glucose, insulin, and T3 increase FAS activity, whereas glucagon and saturated and polyunsaturated fatty acids decrease it. In the present study we show that, in liver, T3 and insulin were able to activate FAS enzymatic activity, mRNA expression, and gene transcription. We localized the T3 response element (TRE) that mediates the T3 genomic effect, on the FAS promoter between −741 and −696 bp that mediates the T3 genomic effect. We show that both T3 and insulin regulate FAS transcription via this sequence. The TRE binds a TR/RXR heterodimer even in the absence of hormone, and this binding is increased in response to T3 and/or insulin treatment. The use of H7, a serine/threonine kinase inhibitor, reveals that a phosphorylation mechanism is implicated in the transcriptional regulation of FAS in response to both hormones. Specifically, we show that T3 is able to modulate FAS transcription via a nongenomic action targeting the TRE through the activation of a PI 3-kinase-ERK1/2-MAPK-dependent pathway. Insulin also targets the TRE sequence, probably via the activation of two parallel pathways: Ras/ERK1/2 MAPK and PI 3-kinase/Akt. Finally, our data suggest that the nongenomic actions of T3 and insulin are probably common to several TREs, as we observed similar effects on a classical DR4 consensus sequence.

Object Surface-based registration (SBR) with facial surface scans has been proposed as an alternative for the commonly used fiducial-based registration in image-guided neurosurgery. Recent studies comparing the accuracy of SBR and fiducial-based registration have been based on a few targets located on the head surface rather than inside the brain and have yielded contradictory conclusions. Moreover, no visual feedback is provided with either method to inform the surgeon about the estimated target registration error (TRE) at various target locations. The goals in the present study were: 1) to quantify the SBR error in a clinical setup, 2) to estimate the targeting error for many target locations inside the brain, and 3) to create a map of the estimated TRE values superimposed on a patient's head image. Methods The authors randomly selected 12 patients (8 supine and 4 in a lateral position) who underwent neurosurgery with a commercial navigation system. Intraoperatively, scans of the patients' faces were acquired using a fast 3D surface scanner and aligned with their preoperative MR or CT head image. In the laboratory, the SBR accuracy was measured on the facial zone and estimated at various intracranial target locations. Contours related to different TREs were superimposed on the patient's head image and informed the surgeon about the expected anisotropic error distribution. Results The mean surface registration error in the face zone was 0.9 ± 0.35 mm. The mean estimated TREs for targets located 60, 105, and 150 mm from the facial surface were 2.0, 3.2, and 4.5 mm, respectively. There was no difference in the estimated TRE between the lateral and supine positions. The entire registration procedure, including positioning of the scanner, surface data acquisition, and the registration computation usually required < 5 minutes. Conclusions Surface-based registration accuracy is better in the face and frontal zones, and error increases as the target location lies further from the face. Visualization of the anisotropic TRE distribution may help the surgeon to make clinical decisions. The observed and estimated accuracies and the intraoperative registration time show that SBR using the fast surface scanner is practical and feasible in a clinical setup.

ABSTRACT Protein arginine methyltransferase 1 (PRMT1) acts as a transcription coactivator for nuclear receptors through histone H4 R3 methylation. The in vivo function of PRMT1 is largely unknown. Here we investigated the role of PRMT1 in thyroid hormone (T3) receptor (TR)-mediated transcription in vivo during vertebrate development. By using intestinal remodeling during T3-dependent Xenopus laevis metamorphosis for in vivo molecular analysis, we first showed that PRMT1 expression was upregulated during metamorphosis when both TR and T3 were present. We then demonstrated a role for PRMT1 in TR-mediated transcription by showing that PRMT1 enhanced transcriptional activation by liganded TR in the frog oocyte transcription system and was recruited to the T3 response element (TRE) of the target promoter in the oocyte, as well as to endogenous TREs during frog metamorphosis. Surprisingly, we found that PRMT1 was only transiently recruited to the TREs in the target during metamorphosis and observed no PRMT1 recruitment to TREs at the climax of intestinal remodeling when both PRMT1 and T3 were at peak levels. Mechanistically, we showed that overexpression of PRMT1 enhanced TR binding to TREs both in the frog oocyte model system and during metamorphosis. More importantly, transgenic overexpression of PRMT1 enhanced gene activation in vivo and accelerated both natural and T3-induced metamorphosis. These results thus indicate that PRMT1 functions transiently as a coactivator in TR-mediated transcription by enhancing TR-TRE binding and further suggest that PRMT1 has tissue-specific roles in regulating the rate of metamorphosis.

This research analyzes and seeks to understand the increase in informal activities in general and mobile commerce as a particular case and the conflict between the public and employees of these itinerant trade. This analysis is performed as a result of the dynamics inherent in the capitalist system that is based on socioeconomic contradictions, among them the need to generate a surplus of labor to thus have its disposal an army reserve. In that sense this analysis takes into account the urbanization process as RMSP suffered by one of the promotion of unemployment which had their causes in several variables. The phenomenon of the increase in practice of these activities, named in this study as "independent work", practiced by itinerant workers in trains and other public spaces of RMSP, has widened from the 80's resulting from economic crises that hit the country causing huge recession unemployment and an economy that can be described as the results of politics applied at that moment. This unemployment is particularly accentuated in the 90s resulting from the economic opening process undertaken by the Brazilian Government as a way of integrating the country in the process of globalization. The way this integration, was made, marked by unbridled economic openness, reflected in several aspects of the economy of the country: it has a new territoriality of plants provided by the development of technical-scientific-media informational affectry RMSP mainly because this the most industrialized in the country, it also involves a process of productive restructuring in which the industrial sector loses its economic space and it is substituted by tertiary activities; as a consequence implemented a new model of administrative organization and the production process adopted by enterprises, the model flexible also called post-fordist, requiring a new profile with more and better professional qualifications to perform the tasks, now with a greater degree of complexity and flexibility. These changes reflected directly in the levels of unemployment now reaching ever higher rates. Informal activities in general and labor have become mobile, thus a way to ensure survival for the huge number of unemployed, to consolidate is not as alternatives but rather as a temporary work, by effective work. Just as the growing number of than practitioners in itinerant trade grew the conflicts that came to rely even violently with police actions undertaken. The territory of the CPTM trains as public spaces and great appeal to the trade due to the large flow of passengers, has become one of those places where this picture was reflected socioeconomic and conflict is fierce. This territory, which offers easier to enable practitioners of this activity a condition of legalization of the organization and activities to provide decent conditions for a significant number of workers was the locus chosen in this research as part of the complex and contradictory dynamics of capitalist economy
Esta pesquisa analisa e procura compreender o aumento das atividades informais de maneira geral e do comércio ambulante como um caso específico e os conflitos entre o poder público e esses trabalhadores do comércio ambulante. Esta análise é realizada enquanto resultante de uma dinâmica inerente ao próprio sistema capitalista que se assenta em contradições socioeconômicas, dentre elas a necessidade de geração de um excedente de mão-de-obra para, dessa maneira, ter a sua disposição um exército de reserva. Nesse sentido essa análise leva em consideração o processo de urbanização sofrido pela RMSP como um dos pressupostos de promoção do desemprego que teve suas causas em várias variáveis. O fenômeno do aumento na prática dessas atividades, denominadas nessa pesquisa como trabalho autônomo , praticada pelos trabalhadores ambulantes nos trens e outros espaços públicos da RMSP, se acentuou a partir das décadas de 80 resultantes de crises econômicas que atingiram o Brasil provocando enorme recessão econômica e um desemprego que pode ser caracterizado como conjuntural. Esse desemprego acentua-se de sobremaneira na década de 90 resultante do processo de abertura econômica empreendido pelo Estado brasileiro como forma de integração do País ao processo de globalização. A forma como se deu essa integração, marcada por uma abertura econômica desenfreada, repercutiu em vários aspectos da economia do País: empreendeu-se uma nova territorialidade das plantas industriais proporcionado pelo desenvolvimento dos meios técnico-científico-informacionais atingindo principalmente a RMSP por ser esta a mais industrializada do País; a mesma também passa por um processo de reestruturação produtiva em que perde espaço o caráter industrial e ganham espaço as atividades terciárias; implanta-se um novo modelo de organização administrativa e do processo de produção adotado pelas empresas, o modelo flexível também denominado pós-fordista, que exigem um novo perfil de profissional com maior e melhor qualificação para executar as tarefas, agora com um maior grau de complexidade e flexibilidade. Essas transformações repercutiram diretamente nos níveis de desemprego que passaram a atingir índices cada vez maiores. As atividades informais em geral e o trabalho ambulante tornaram-se, dessa forma, numa forma de garantir a sobrevivência para esse enorme contingente de desempregados, passando a se consolidarem não como alternativas temporárias e sim como trabalho de fato. Da mesma forma que crescia o número de praticantes no comércio ambulante cresciam os conflitos que passaram a contar inclusive com ações policiais empreendidas violentamente. O território dos Trens da CPTM, enquanto espaços públicos e de grande apelo para o comércio devido ao grande fluxo de passageiros, passou a ser um desses lugares onde este quadro socioeconômico se refletiu e os conflitos se acirraram. Esse território que oferece condições mais fáceis de possibilitar aos praticantes dessa atividade uma condição de organização e legalização da atividade visando proporcionar condições dignas a um número significativo de trabalhadores foi o lócus escolhido nesta pesquisa como uma das partes da complexa e contraditória dinâmica da economia capitalista

The present dissertation aims to analyze the commerce situation in the wagons of the train compositions crossing the West Zone of São Paulo Metropolitan Region, foccusing on Line 8 (Diamond), which is a part of CPTM (Companhia Paulista de Trens Metropolitanos) structure. Considering that, a historical contextualization of the phenomenon (informal work) in which my object is inserted was developed. The analysis of the literature addressing the subject as to its concepts, as well as the field observation of the commerce inside the compositions in different days and times, substantiate this study to faithfully de-scribe the informal work carried out, which despite the illegality, represents the sub-sistence of those individuals. The relation of those illegal activities and the society in a general perspective was approached, based on the field observations and on the interviews of a selected group of workers. The data collected underlie the description and analysis of the set of relations concerned as the objective aimed to this dissertation
Esta presente dissertação tem como intenção primordial, a análise da situação que se encontra o comércio no interior dos vagões das composições que cruzam parte da porção oeste da Região Metropolitana de São Paulo através da linha 8 (Diamante) que faz parte da estrutura da Companhia Paulista de Trens Metropolitanos (CPTM). Para tanto, trabalhei a contextualização histórica do surgimento do fenômeno (Trabalho Informal) no qual meu objeto está inserido. A análise de obras que já conceituaram o fenômeno em conjunto com vários dias de observação do comércio no interior das composições, são o fundamento desse estudo que, espero, tenha conseguido descrever com fidedignidade o trabalho ali realizado que, apesar de ilegal, garante a subsistência daqueles que a praticam. Temos aqui, uma visão de como essas atividades ilegais se relacionam com a sociedade mais ampla. Através das observações de um trabalho de campo e de uma entrevista aplicada a um grupo desses trabalhadores, esta dissertação intentou em descrever e analisar o conjunto dessas relações

Resumen
Esta obra é composta de textos, crônicas, versos e poesias de autores regionais que tiveram suas vidas marcadas e transformadas a partir do ano de 1910, com a chegada da ferrovia, evento que mudou não só a face da cidade de Ipueiras, no interior cearense, mas também seus hábitos, costumes e modo de vida. As cidades e vilas por onde a ferrovia passou convidam a viajar pelas páginas do tempo, seguindo os trilhos da antiga estrada de ferro, mais conhecida como Linha Norte. Ligando a cidade de Camocim aos Sertões dos Inhamuns, cortando milhas e milhas de sertões bravios com suas imponentes locomotivas, chamadas carinhosamente de “Maria Fumaça”, semelhantes a verdadeiros “Dragões de Ferro”, cuspindo fogo e sibilando, anunciando que o progresso finalmente havia chegado ao Sertão. É um trabalho, encabeçado pela Secretaria de Educação de Ipueiras, que buscou parceria de autores ipueirenses já consagrados, como o jornalista Frota Neto, a cordelista Dalinha Catunda, como também de pessoas que se interessam por nossa história, como a Mestra em Geografia Luciana Andrade Catunda, o historiador Antônio Genilson Vieira de Paiva, o professor Francisco de Assis Lima (Fury), o ipueirense Tadeu Fontenele e, ainda, um belíssimo texto do saudoso cronista ipueirense Bérgson Frota (in memoriam), já publicado anteriormente no jornal O Povo. Este livro traz também, em comemoração ao centenário ocorrido em 2017, do ilustre ipueirense Gerardo Mourão, um belíssimo texto intitulado “Gerardo Mello Mourão e o Trem”, de autoria do seu biógrafo, o reconhecido escritor cearense José Luís Lira. Se pudéssemos comparar esta jornada literária a uma locomotiva, poderíamos dizer que o combustível seria a saudade e o maquinista seria uma mistura nostálgica das figuras daqueles tempos. No primeiro vagão, viria uma imensa carga de informações sobre a história do surgimento da Linha Norte, com o impacto da sua chegada em nossa localidade, segundo o olhar do Professor Genilson Paiva. O segundo vagão traria um relato da Mestra em Geografia Luciana Catunda sobre “O algodão e a ferrovia na mobilidade do território cearense”, um destaque para o escoamento da produção algodoeira, o chamado “ouro branco”. No terceiro vagão, viriam as valiosas crônicas do professor Fury, que nos transportam até a antiga estação ferroviária, com o seu vai e vem de viajantes: aventureiros, comerciantes, migrantes em busca de melhores oportunidades ou, simplesmente, passageiros que faziam de cada chegada e partida do trem sempre um grande acontecimento, que atraía olhares curiosos, vendedores ambulantes e até mesmo as moças da cidade, que, como nos romances de Leon Tolstói, sonhavam em encontrar ali um grande amor. No quarto vagão viria o encanto e a beleza do trabalho de Dalinha Catunda, neta do “chefe da estação”, seu Gonçalo Ximenes Aragão, que nos embala em seus versos e poesias, onde a alegria da chegada dava lugar à tristeza da partida, até a chegada do próximo trem na estação. Dalinha nos conta a história da passagem de um rei por Ipueiras – não um rei qualquer, mas um rei nordestino – o Rei do Baião. No ano de 1966, no prédio da estação, cantou para aquela gente que delirava com os acordes da sanfona do “Velho Lua”. O quinto vagão vem carregado de nostalgia e de personagens da infância e juventude do jornalista Frota Neto, que se perpetuaram na história do trem e nos fazem sentir como se lá também estivéssemos vivenciando aquele cotidiano que ainda hoje se faz presente pelas estórias e anedotas de figuras como a Dona Maria Capoeira – cafezeira e quituteira da estação ferroviária, testemunha de grandes momentos desta história, agora existente somente na memória dos antigos e pela presença dos velhos trilhos e estações ferroviárias que insistem em resistir à força do tempo e do descaso das políticas governamentais e de preservação patrimonial. No sexto vagão, Tadeu Fontenele nos fala sobre o “Show do Luiz Gonzaga em Ipueiras” e faz uma narrativa linda da sua aventura com sua mãe, Dona Ineizita, na garupa da sua bicicleta, do Centro da cidade até o Bairro da Estação, para não perder a apresentação do ídolo. O sétimo vagão traz uma crônica do jovem escritor ipueirense Bérgson Frota, cujo título, “O Tempo do Trem em Ipueiras”, bem poderia dar nome a este pequeno livro, de tanto que se identifica com o sentimento comum aos demais autores. O oitavo vagão viria trazendo o orgulho de nossa Ipueiras ter como filho o ilustre poeta Gerardo Mourão, cujo centenário de nascimento ocorreu em 2017, comparado a Dante Alighieri, eleito pela Guilda Órfica, secular irmandade internacional de poetas, o maior poeta do século XX, conforme trajetória sintetiza tão bem o Professor Zé Luís Lira. Finalmente, no último vagão, os professores Ronaldo Moreira e Paula Gaspar nos presenteiam com recortes da história da Escola Juarez Catunda, localizada no Bairro da Estação, equipamento que viabilizou a realização deste projeto, através do Programa Mais Cultura nas Escolas, onde os idealizadores optaram, dentre tantas alternativas, por nos trazer estes Escritos Sobre a Estação Ipueiras.
