Resumen
The development and optimisation of conditions for oxidative Wacker-type cyclisations followed by establishing the reaction scope are reported. Building upon the achievements in the field of oxidative Wacker-type reactions that has recently gathered interest, hydroxylamines and hydrazines were converted to isoxazolidines and pyrazolidines respectively. Secondary hydroxylamines cyclised yielding syn-isoxazolidines with excellent diastereoselectivities, whereas secondary hydrazines cyclised yielding anti-pyrazolidines but still maintained a high level of diastereoselectivity. Additionally, an enantioselective variant was explored. Isoxazolidines were successfully transformed to the corresponding 1,3-amino alcohols, which were further converted to amino sugar derivatives.

Resumen
Les 3-amino-3-désoxyglycosides (ADGi) constituent une classe essentielle de glycosides, présents dans la structure de nombreux composés bioactifs importants. Les 3-amino-3-désoxyglycals (ADGa) sont des donneurs prometteurs pour la synthèse de ces ADGi. La partie glycal des ADGa permet de les envisager à la fois comme donneurs de 2-désoxy-ADGi par glycosylation en présence d’un accepteur, ou de 1,2-trans-ADGi via une séquence époxydation/glycosylation. Ces travaux de thèse ont été consacrés au développement d’une nouvelle voie d’accès à ces ADGa, mettant à profit deux réactions clés : un réarrangement de Ferrier, suivi d’une réaction d’aza-Wacker (AW) en conditions oxydantes avec un lien amovible de type carbamate. Cette réaction d’AW permet, en une seule étape, de former la liaison C3-N et de regénérer le glycal. En modifiant des conditions issues de la littérature utilisant la 1,4-benzoquinone (p-BQ), nous avons obtenu les ADGa correspondant aux séries D-Glucal, D-Galactal, L-Rhamnal et L-Fucal, en utilisant le DMF comme solvant. Les ADGa étant obtenus avec des protections orthogonales, une preuve de concept de leur déprotection sélective a pu être réalisée sur l’ADGa modèle. Finalement, l’ADGa dérivé du D-Galactal a été engagé dans une séquence d’époxydation / glycosylation pour la synthèse d’un disaccharide avec d’excellents rendements et sélectivités. L’utilisation du DMF comme solvant nous a ensuite permis de définir des conditions pour cette cyclisation d’AW en utilisant le dioxygène de l’air comme réoxydant du Pd (0). La sélectivité de l’étape d’amidopalladation a également été étudiée pour les deux systèmes. Après deutération des adduits de Ferrier, nous avons démontré que l’amidopalladation était trans dans les deux systèmes réoxydants, en partant des carbamates dérivés du D-Glucal et du D-Galactal. Finalement, deux sous-produits d’intérêts ont pu être étudiés. Le premier, obtenu lors de l’optimisation avec la p-BQ, correspond formellement à une fonctionnalisation 1,3 du carbamate modèle, et ses conditions de synthèse ont été optimisées. Le second est un dérivé d’hexa-2,4-diènal, qui a pu être isolé et pleinement caractérisé
3-Amino-3-deoxyglycosides (ADGi) constitute an essential class of glycosides found in the structure of several important bioactive compounds. 3-Amino-3-deoxyglycals (ADGa) are promising donors for these ADGi. The glycal moiety allows ADGa to serve as donors of 2-deoxy-ADGi by glycosylation in the presence of an acceptor, or as donors of 1,2-trans-ADGi through an epoxidation / glycosylation sequence. This thesis work focuses on developing a new synthetic route for these ADGa, taking advantage of two key reactions: a Ferrier reaction followed by an aza-Wacker (AW) reaction in oxidizing conditions with carbamate type tether. This AW step enables C3-N bond formation and glycal reformation in a single step. Based on modified conditions from the literature using 1,4-benzoquinone (p-BQ), we obtained ADGa corresponding to the D-Glucal, D-Galactal, L-Rhamnal, and L-Fucal series, using DMF as the solvent. These ADGa were obtained orthogonally protected, and a proof of concept for their selective deprotection was demonstrated on the model ADGa. Finally, the ADGa derived from D-Galactal was subjected to an epoxidation / glycosylation sequence to synthesize a disaccharide with excellent yield and selectivity. The use of DMF as a solvent also allowed us to establish conditions for the AW cyclization, using dioxygen from the air as a reoxidant for Pd(0). The selectivity of the amidopalladation step was then examined for the two systems. After deuteration of the Ferrier adducts, we demonstrated that amidopalladation proceeded trans in both reoxidizing systems, starting from carbamates derived from D-Glucal and D-Galactal. Finally, two by-products of interest were studied. The first is a formal 1,3-functionalization of the model carbamate, and the reaction conditions for its synthesis were optimized. The second is a hexa-2,4-dienal derivative, which was isolated and fully characterized

Resumen
La condensation du 1,8-bis(triméthylsilyl)octa-2,6-diène (BISTRO) l'anhydride succinique conduit à la ((d,l)-6,9-divinyl-1 -oxaspiro-4,4-nonane-2-one. La réaction d'alkylation de cette spirolactone a été étudiée. Des calculs de structure complètent l'étude expérimentale. Plus particulièrement, l'alkylation de cette lactone par un dérivé benzocyclobuténique suivie de la thermolyse du composé obtenu, conduit à un 17α-vinyl stéroi͏̈de de manière stéréoséleclive. L'ajout de deux étapes (méthoxycarbonylation et décarboxylation) à la première synthèse décrite, permettent d'accéder à des stéroi͏̈des présentant la configuration naturelle. Les synthèses proposées donnent accès au squelette stéroi͏̈dique en 5 étapes du butadiène. L'oxydation du groupement vinyle 17α par la réaction de Wacker a été étudiée. Cette dernière a révélé une oxydation inhabituelle de type anti Markovnikov. La transformation des stéroi͏̈des obtenus ainsi que les essais d'introduction méthyle angulaire en position 13 sont décrits. .
Acylation of l,8-bis(trimethylsilyl)-2,6-octadiene) with succinic anhydride affords (dl))-6,9-divinyl-l-oxaspiro|[4,4]nonane-2-one. Alkylation of this spirolactone with a benzocyclobuten derivative followed by leads to a 17α -vinyl steroid. Experimental and computationnal studies on the alkylation step are described. Addition of two steps (methoxycarbonylation and decarboxylation) affords the corresponding steroids with the natural configuration. This work proposes a new synthesis of steroids in only five steps from butadiene. Studies on the Wacker oxydation of the 17α vinylic group are described. Anti Markovnikov oxydalion is observed. Synthetic transformations and attemps to introduce angulary 13β methyl group are described. The use of simple and economical reactions allowed us to synthesize 84 new steroid structures

La réaction de cyclisation de type Wacker, de composés polyols et aminopolyols insaturés constitue un outil puissant et efficace pour la synthèse d’hétérocycles oxygénés ou azotés.Dans ce travail de thèse, nous proposons l’étude d’une réaction catalysée par un complexe de palladium(II) de type domino-cyclisation, mettant en jeu une réaction de couplage. Cette séquence catalytique revient à une fonctionnalisation d’un hétérocycle par une chaîne latérale, tout en créant deux centres stéréogènes en une seule étape. L’influence de la nature des réactifs mis en jeu, ainsi que des conditions expérimentales sur l’activité et la diastéréosélectivité de la réaction sont discutées. Les applications vers la synthèse de produits naturels (anisomycine) ou d’analogues (varitriol) sont présentées.La réaction d’oxycarbonylation catalysée par un complexes de palladium(II) est une transformation intéressante de polyols insaturés en lactones bicycliques, présentant un motif de type tétrahydrofurane avec une excellente stéréosélectivité-cis. Le premier exemple de réaction d’oxycarbonylation catalysée par des complexes de palladium chiraux dans les liquides ioniques est décrit. Une étude approfondie de la nature des ligands démontre que les bis(oxazolines) chirales constituent les meilleurs ligands du palladium pour la cyclisation du pent-4-ène-1,3-diol racémique 69a. Le dédoublement cinétique du composé 69a sous atmosphère de monoxyde de carbone, en présence d’un complexe chiral de palladium(II) et de p-benzoquinone employant l’acide acétique ou le liquide ionique [bmim]NTf2 comme solvant, a permis d’isoler le 2,6-dioxabicyclo-[3.3.0]octane-3-ones avec jusqu’à 57% d’excès énantiomérique pour l’énantiomèrede configuration (R,R)-70a, et jusqu’à 80% d’excès énantiomérique pour l’énantiomèrede configuration (S,S)-70a
Intramolecular Wacker-type cyclization of unsaturated polyols and aminopolyols represents a powerful method in the synthesis of oxygen- or nitrogen-containing heterocycles. The thesis offers an insight into the systematic study of domino intramolecular palladium(II)-catalyzed cyclization and coupling reaction allowing the implementation of side chains into heterocyclic skeletons along with the formation of two stereocenters in a single step. Different types of coupling partners and reaction conditions were examined, the influence of substrate substituents on diastereoselectivity is discussed. The applications in the synthesis of naturally occuring compounds or their analogs are outlined (anisomycin, varitriol). Palladium(II)-catalyzed oxycarbonylation represents an interesting transformation of unsaturated polyols into bicyclic lactones with tetrahydrofuran structural motif with excellent cis-stereoselectivity. The first example of the use of ionic liquids as reaction media in the asymmetric Pd(II)-catalyzed oxycarbonylation is described. Based on a ligand screening, the chiral bis(oxazoline)-type ligands were successfully used in the Pd(II)-promoted bicyclisationof racemic pent-4-ene-1,3-diol (±)-69a. The kinetic resolution of (±)-69a in the presenceof chiral catalyst and p-benzoquinone under carbon monoxide atmosphere using acetic acid and/or ionic liquid as solvent afforded enantioenriched 2,6-dioxabicyclo[3.3.0]octane-3-ones(R,R)-70a (57% ee) and (S,S)-70a (80% ee), respectively

La thèse présentée étudie la réactivité de deux composés, l'a-chloro-a,a-difluorotoluène (1) et l'a-chloro-a,a-difluoroanisole (2), afin d'en développer la valorisation. La première partie reporte des essais de substitution nucléophile sur la liaison carbone/chlore des composés (1) et (2). Ces réactions sont difficiles et cette faible réactivité de la liaison carbone/chlore est attribuée à l'influence électronique des atomes de fluor voisins. La substitution du chlore par le groupement triméthylsilyle (deuxième chapitre) a permis de synthétiser deux analogues gem-difluorés du réactif de Ruppert. Les deux composés triméthylsilylés (PhCF2SiMe3 et PhOCF2SiMe3) sont de bons intermédiaires pour des réactions de gem-difluoration. Enfin la synthèse de dérivés homoallyliques gem-difluorés a fourni un accès facile à de nombreux produits gem-difluorés a, b ou g fonctionnalisés et a mis en évidence l'influence de l'atome de fluor sur la régiochimie de réactions palladocatalysées
The aim of this thesis is the study of the chemical reactivity of two compounds: a-chloro-a,a-difluorotoluene (1) and a-chloro-a,a-difluoroanisole (2). The first part of the manuscript shows nucleophilic substitution reactions. These reactions require special conditions and only phenoxy ot thiophenoxy groups can be used as nucleophiles. These results are explained by neighbouring fluorine atoms which induce the C/Cl bond lack of reactivity. Magnesium promoted chlorine substitution by trimethylsilyl group allows access to two gem-difluorinated analogues of Ruppert's reagent (CF3SiMe3). These two products (PhCF2SiMe3 and PhOCF2SiMe3) are good intermediates in gem-difluoration reactions. Finally, reaction between allyltributyltin and compounds (1) or (2) gives an easily access to many a, b or g functionnalized gem-difluorinated products. Fluorine influence upon the regiochemistry of pallado-catalysed reactions is also demonstrated

Resumen
La condensation du 1,8-bis(triméthylsilyl)octa-2,6-diène (BISTRO) sur l’anhydride chloroacétique suivie d’un traitement en milieu basique, conduit à l’époxyde divinylcyclopentane spiranique. Ce dernier permet d’accéder facilement à des précurseurs possédant un hétéroatome. La formation du thioacétate donne accès à une série de 11-thia stéroïdes possédant une variété de substituants sur le cycle A. La très bonne sélectivité de la réaction de Diels-Alder conduit majoritairement aux composés de stéréochimie naturelle trans-anti-trans. Nous avons également mis au point une synthèse efficace d’un azabenzocyclobutène qui nous a permis de réaliser la première synthèse totale de 3-aza stéroïdes possédant un cycle A aromatique en seulement cinq étapes à partir du BISTRO avec un rendement global de 42 %. L’utilisation de notre azabenzocyclobutène et de la stratégie mise au point pour obtenir des 11-hétéro stéroïdes nous a permis de réaliser les premières synthèses totales de 3-aza-11-thia et de 3-aza-11-oxa stéroïdes en seulement cinq étapes à partir du BISTRO avec des rendements respectifs de 18 et 19 %
Condensation of 1,8-bis(trimethylsilyl)octa-2,6-diene with chloroacetic anhydride followed by basic treatment, led to the spirodivinylcyclopentanic epoxide, which is a good precursor of molecules bearing an heteroatome. The formation of the thioacetate gives access to a set of 11-thia steroids diversely substituted on the A ring. The very good selectivity of Diels-Alder reactions led to the isomers with the natural trans-anti-trans stereochemistry. We also achieved the first efficient synthesis of an azabenzocyclobutene, precursor of 3-aza steroids bearing an aromatic A ring through a five steps sequence from BISTRO and more than 42 % overall yield. Moreover, using our azabenzocyclobutene and the same strategy developed previously, we have achieved the first total synthesis of 3-aza-11-thia and 3-aza-11-oxa steroids through a five steps sequence from BISTRO with respectively 18 and 19 % overall yield

Resumen
Le premier objectif de ce travail a été d’évaluer le potentiel catalytique d’un ligand tétraphosphine : le cis-cis-cis-1,2,3,4-tétrakis-(diphénylphosphinométhyl)cyclopentane ou Tédicyp. Nous avons étudié l’activité catalytique de ce ligand dans des réactions de Suzuki et de Negishi catalysées par le palladium. Le complexe Tédicyp-palladium a une nouvelle fois montré son efficacité avec des quantités aussi faibles que 0. 000 1 % molaire. Le deuxième projet de ce travail a été de synthétiser un nouveau ligand tétraphosphine : le cis,cis,cis-1,2,4-tris-(diphénylphosphinométhyl)-3-(diphénylphosphinoéthyl)cyclopentane, ou Tridicyp. Sa synthèse a été réalisée en sept étapes avec un rendement global de 75 %. Nous avons ensuite évalué l’activité catalytique de ce nouveau ligand tétrapodal dans diverses réactions catalysées par le palladium (Tsuji-Trost, Heck, Suzuki, Sonogashira, Negishi). Les résultats obtenus sont excellents et bien supérieurs à ceux obtenus avec des ligands classiques
The first aim of this work was to evaluate the catalytic potential in homogeneous catalysis of a tetraphosphine ligand : the cis-cis-cis-1,2,3,4-tetrakis-(diphenylphosphinomethyl)cyclopentane or Tedicyp. We have explored the potentiel of this catalyst by investigation of well known palladium-catalyzed reactions such as Suzuki and Negishi cross-coupling reactions. The complex gave very good results, in terms of substrate / catalyst ratio, compared to the traditional complexes. The second target was to synthesize a new tetraphosphine ligand : cis,cis,cis-1,2,4-tris-(diphenylphosphinomethyl)-3-(diphenylphosphinoethyl)cyclopentane, or Tridicyp. It has been synthesized in seven steps from commercially available dicyclopentadiene. The efficiency of this tetraphosphine ligand for palladium-catalysed coupling reactions has been studied. Satisfactory results in terms of substrate / catalyst ratio were obtained for Tsuji-Trost, Heck, Suzuki, Sonogashira and Negishi reactions

Resumen
Cette thèse a porté sur la mise au point de l’addition régiosélective et stéréosélective d’allyléthers métaux sur des oxazolidines issues du (S)-phénylglycinol, de façon à avoir accès à des aminoalcools de structures différentes en jouant sur la nature du métal. Des aminoalcools possédant un nouveau centre stéréogène contrôlé et une double liaison éther d’énol de configuration Z ont été obtenus avec un allyléther lithié, et l’utilisation d’un zincique fournit des aminoalcools possédant deux nouveaux centres stéréogènes syn ou anti selon le substituant de l’oxazolidine. Une nouvelle réaction de cyclisation de ces aminoalcools par catalyse au palladium a ensuite été explorée afin d’obtenir différentes oxazolidines bicycliques. Une excellente régio- et stéréosélectivité a été constatée pour la formation de ces oxazolidines possédant un H ou un Me en jonction de cycle. L’étude de divers paramètres a mis en évidence leur influence sur le déroulement de la réaction. Ces oxazolidines bicycliques se sont révélées être de bons précurseurs pour la synthèse asymétrique d’hétérocycles azotés. Nous avons obtenu des dérivés énantiopurs de la proline, substitués en position 5 et en position 4 et 5.
