The ability of the zona pellucida to mediate the species specific recognition of spermatozoa is due to a glycoprotein constituent of this structure, known as ZP3. In view of its important role in the initiation of fertilization, ZP3 has been identified as a suitable candidate for the development of a contraceptive vaccine. Raft membranes are implicated in cell adhesion. Here, we demonstrated that rafts isolated from hypermotile capacitated pig sperm as low-density Triton X-100 insoluble membranes had the ability to bind specifically to homologous zonae pellucidae (ZP). This binding was dependent on pig ZP3alpha sulfoglycoprotein, a major player in intact sperm binding.
