Mein Ziel ist eine Begründung der Physik aus der Operationalisierung ihrer Grundmaße. Wir beginnen mit der klassischen und relativistischen Kinematik. Dann greifen wir einen programmatischen Entwurf von Heinrich Hertz auf und gelangen über die Quantifizierung von Energie und Impuls zu den Bewegungsgleichungen und dem Wirkungsfunktional. Den Abschluß bildet eine relativistische Revision der Energie, Impuls und Massenvorstellung. Das Ziel ist nicht primär, den mathematischen Aufbau der Mechanik zu ändern oder zu verbessern, sondern ein tieferes physikalisches Verständnis der Mechanik zu erlangen, so wie Einstein mit seinen Gedankenexperimenten zur relativistischen Kinematik. Ausgehend von anerkannten Messhandlungen und Naturprinzipien wird der mathematische Formalismus erst erzeugt und somit neu beleuchtet. Die Arbeit steht damit im selben Kontext wie die aktuellen Bemühungen zur "Fundierung der Quantenmechanik", nur eben mit Bezug auf die Interpretation der klassischen und relativistischen Kalküle.
My objective is a foundation of physics from the operationalization of its basic observables. We begin with classical and relativistic kinematics. Seizing on a programmatic proposal by Heinrich Hertz we arrive via quantification of energy-momentum at the equations of motion and the action functional. Finally we present a relativistic revision of energy, momentum and inertial mass. The goal is not primarily to change or improve the mathematical structure of mechanics, but to gain a deeper physical understanding of mechanics like Einstein with his gendanken experiments on relativistic kinematics. Starting from undisputed measurement operations and natural principles the mathematical formalism is only created and thus shown in a new light. This work is in the same context as current efforts to the "foundation of quantum mechanics", just with respect to the interpretation of the classical and relativistic calculuses.

The research network “Basic Concepts for Convection Parameterization in Weather Forecast and Climate Models” was organized with European funding (COST Action ES0905) for the period of 2010–2014. Its extensive brainstorming suggests how the subgrid-scale parameterization problem in atmospheric modeling, especially for convection, can be examined and developed from the point of view of a robust theoretical basis. Our main cautions are current emphasis on massive observational data analyses and process studies. The closure and the entrainment–detrainment problems are identified as the two highest priorities for convection parameterization under the mass–flux formulation. The need for a drastic change of the current European research culture as concerns policies and funding in order not to further deplete the visions of the European researchers focusing on those basic issues is emphasized.

This project traces the ways universities articulate a desire for diversity through the gateway genres of college admissions, composition course placement, and first-year-composition (FYC). Together, these genres serve as points of access for a theoretical study that seeks to better understand the ideological function of writing programs to socialize borderline college applicants into the rhetorically constructed role of a Diverse College Student. I focus on what I call bootstraps genres--reoccurring rhetorical situations that call for students to recount social hardships like racism and classism as personal hardships to be overcome through personal heroics. Despite being immersed in rhetorics of individualism, the college application essay, the directed self-placement guide, and the literacy narrative all call for the mimetic construction of disadvantage as an appeal to college-readiness. As new college students move through the initiation rituals of admissions, orientation, and FYC, they are presented with rhetorical tasks that are both raced and classed. Bootstraps genres ask students to first read the university's desire for diversity and then fulfill that desire through personal stories of difference and disadvantage.

Tuberculosis, an infectious disease caused by Mycobacterium tuberculosis, has been a global health problem for years. The emergence of drug resistance in this organism generates the necessity of exploring novel targets and developing new drugs. Topoisomerases are enzymes found in all kingdoms of life responsible for overcoming the topological barriers encountered during essential cellular processes. The genomes of mycobacteria encode only one type IA topoisomerase (MtopI), which has been validated as a novel TB drug target. The goal of this study is to obtain new information on the mechanism and resistance of endogenous and synthetic inhibitors of MtopI. Rv1495 is a M. tuberculosis toxin that belongs to the MazEF family (MazE is the antitoxin and MazF is the toxin), with endoribonuclease activity. Rv1495 (MazF homolog in M. tuberculosis) toxin has been shown to interact directly with the C-terminal domain of MtopI for mutual inhibition. In this study the interaction of Rv1495 with the positively charged C-terminal tail in Mtop I is reported. This new information is useful for rational design and discovery of antibiotics for mycobacteria. Ethacridine, an FDA approved drug has shown activity against MtopI. In this project we studied the mechanisms of resistance associated with this drug as well the use of Ethacridine in combination with Moxifloxacin, to potentiate the bactericidal effect of this current second line drug for TB treatment. Results from sequencing of the genomic DNA isolated from the resistant mutants suggested the involvement of the Holliday-junction Ruv resolvase. Further studies showed that co-treatment with Ethacridine can enhance the moxifloxacin-mediated killing of M. smegmatis. FP-11g, a novel fluoroquinophenoxazine inhibitor of bacterial topoisomerase I, has shown promising activity against M, tuberculosis. We explored the bactericidal activity and resistance mechanisms associated to FP-11g using M. smegmatis as model organism. Additionally, the inhibitory effect of FP-11g was also evaluated on M. abscessus. FP-11g at concentration 4X MIC showed complete bactericidal activity against M. smegmatis after 24 hours. Inhibitory activity against M. abscessus was also observed. Results from sequencing of the genomic DNA isolated from the M. smegmatis resistant mutants revealed mutations in genes associated with general drug resistance.

When we talk about collective entities, action is the most common kind of ascription. We regularly say things such as ?China suspends all coal imports from North Korea?; ?Uber is investigating harassment claims by ex-employee?; ?Supreme Court considers case of a shot fired in U.S. that killed a teenager in Mexico?; ?Malaysia recalls ambassador to North Korea?; ?SpaceX launches rocket from NASA?s historic moon pad.? Are those ascriptions true? For sure, they could all be metaphoric. We could take collective entities as agents just as a way of speaking. In this work, I argue in favor of a realist position regarding collective entities and their status of agent; rendering some of these sentences true. Recently, many philosophers are addressing this topic, but the discussion tends to be guided by the problem of collective intentionality, the problem of how collective entities can have mental states. My work tries to bring more elements of philosophy of action to the investigation of collective action. I take as a guide the problem of action individuation, because this topic addresses questions of central importance for collective action. Especially the question of aggregate actions, actions that are composed of other actions, which seems to be the paradigmatic case of collective action, insofar as they are presumably composed of individuals? actions. The problem of action individuation leads us to two central concepts on the nature of action: basic action and intention. In this work, I will show how an investigation on basic action can help us locate the place of individuals? contributions in collective action and how an investigation on intention can locate a fundamental element of action that is irreducible and distinctively collective in collective action cases. After exploring these two core concepts, I provide a definition of action that take seriously the place of intention as a guide to identify when an event constitutes an action.

This thesis explores current research on autonomy. Autonomy is defined in three categories: freedom to pose questions and encouraging student choice, trust in students to solve problems posed by those questions, and allowing student reflection on progress. Autonomy is one solution to promote intrinsic motivation in students. Autonomy supportive classrooms feature mainly language learning atmospheres in research. However, it is also pertinent to contemporary art education as it promotes an environment for student creativity. Student performance and the modes of measurement for this research project are based on common formative and summative assessment measures. Will autonomy in the classroom combat issues of poor performance? While there is significant research occurring in the field of language learning regarding student autonomy in the classroom, very little is found in the Basic Digital Photography area. This study employed an action research project in order to determine if student autonomy is an effective tool in the basic digital photography classroom. Data collected include survey results, student journals, homework completion rates, and test scores to compare the results of student performance from a teacher directed unit to a more autonomous one. A new plan of action for incorporating findings into another unit is proposed as the autonomous unit as it currently stands revealed that autonomy is not an effective tool in the manner it was utilized in this Basic Digital Photography classroom.

This thesis concerns the question of what it is for a subject to act. It answers this question in three steps. The first step is taken by arguing that any satisfactory answer must build on the idea that an action is something predicable of the acting subject. The second step is taken by arguing in support of an answer which does build on this idea, and does so by introducing the idea that acting is doing something which is an exercise of a particular kind of disposition on the part of the acting subject. The third step is taken by arguing that the disposition in question must be of a kind which is exercised in conditions in which the acting subject thinks they are acting. From this vantage point the thesis develops many further commitments: That action is constitutively subject to a mode of explanation that mentions the kind of disposition just mentioned; that any case of acting requires a veridical representation of a means by which the action is performed; and that a problem about the underspecified nature of desire ascriptions can be solved by appeal to the conceptual materials made available by these investigations. The thesis finally develops several objections to the account it gives, both substantive and methodological, and explains why these objections ought to be rejected.

The research employed a qualitative research paradigm. The ethnographic approach was used to conduct the research. Data collection was done though various ethnographic techniques, classroom observation, in-depth interviews and document analysis. The population was 85 women who participated in a Adult Basic Education and Training programme in Eersterivier in the Cape Peninsula area a questionnaire was used to collect demographic information of the participants Data was analysed by thematic analysis and coded, categorised and discussed according to the aim and objectives of the study in relation to previous studies The main findings were why exploring the existing literacy practice women were depended on others for literacy assistance, which made them avoid literacy events and become vulnerable in this process to cope with the everyday life.

While the Constitutional Court has repeatedly enforced a duty to meaningfully engage with communities where the provision of basic social services and goods is at stake, uncertainty about the form, extent and quality of community engagement continues to negatively impact on development projects. The much publicised "toilet saga" in the Makhaza informal settlement in the Western Cape is a telling example. This problem is partly attributable to a lack of clarity about the legal foundations and basis of this duty. The research project explores such an approach by clarifying the proper legal and constitutional basis of the right to meaningfully participate in development projects and the delivery of basic social goods, and in so-doing, to place public participation in socio-economic rights on a sound legal and constitutional foundation. The project further seeks to develop a framework or best practice for the practical implementation of this right in the context of urban development projects.

In this dissertation we present the quantitative response of an epidemic of the so-called SIR-type, in a population consisting of a local component and a migrant component. Each component can be divided into three classes, the susceptible individuals, usually denoted by S, who are uninfected but may contract the disease, infected individuals (I) who are infected and can spread the disease to the susceptible individuals and the class (R) of recovered individuals. If a susceptible individual becomes infected, it moves into the infected class. An infected individual, at recovery, moves to the class R. Firstly we develop a model describing two interacting populations with vaccination. Assuming the vaccination rate in both groups or components are constant, we calculate a threshold parameter and we call it a vaccination reproductive number. This invariant determines whether the disease will die out or becomes endemic on the (in particular, local) population. Then we present the stability analysis of equilibrium points and the effect of vaccination. Our primary finding is that the behaviour of the disease free equilibrium depend on the vaccination rates of the combined population. We show that the disease free equilibrium is locally asymptotically stable if the vaccination reproductive number is less than one. Also our stability analysis show that the global stability of the disease free equilibrium depends on the basic reproduction number, not the vaccination reproductive number. If the vaccination reproductive number is greater than one, then the disease free equilibrium is unstable and there exists three endemic equilibrium points in our model. Two of these three endemic equilibria are so-called boundary equilibrium points, which means that the infection is only in one group of the population. The third one which we focus on is the general endemic point for the whole system. We derive a threshold condition that determines whether the endemic equilibria is locally asymptotically stable or not. Secondly, by assuming that the rate of vaccination in the migrant population is constant, we apply optimal control theory to find an optimal vaccination strategy in the local population. Our numerical simulation shows the effectiveness of the control strategy. This model is suitable for modeling the real life situation to control many communicable diseases. Models similar to the model used in the main contribution of our dissertation do exist in the literature. In fact, our model can be regarded as being in-between those of [Jia et al., Theoretical Population Biology 73 (2008) 437-448] and [Piccolo and Billings, Mathematical and Computer Modeling 42 (2005) 291-299]. Nevertheless our stability analysis is original, and furthermore we perform an optimal control study whereas the two cited papers do not. The essence of chapter 5 and 6 of this dissertation is being prepared for publication.

In this thesis I argue that by taking account of economic disparities and backlogs in intergovernmental infrastructure grants to municipalities in South Africa, government will effectively meet its constitutional obligation to equitably allocate infrastructure grants to local government according to the principles of parity, proportionality and priority (Young, 1994). Municipalities will thus be able to provide basic services to households in keeping with the Bill of Rights of the Constitution of South Africa (1996). Adapting the Petchey et al (2004) provincial capital expenditure grant model to the provision of municipal services using secondary data and information from local municipalities in the Cape Winelands District, I evaluate whether government&rsquo
s existing municipal grants are allocated equitably and, whether they account for disparities that differentiate municipalities from each other. The findings from my analysis show that the current approach to financing municipal infrastructure does not sufficiently account for disparities and thus, undermines the requirement for equitability, adequacy and efficiency of intergovernmental allocations. Consequently, the right of citizens to basic municipal services is compromised and the macroeconomic structure is weakened. Furthermore the institutional arrangements for local government autonomy is undermined because municipalities cannot ensure stability, predictability, flexibility and economic efficiency of infrastructure budgets. As municipalities receive part of their finance from national government through infrastructure grants, I used data from five local municipalities to examine the extent to which there is equitability and efficiency in the way this finance is allocated. To this end I constructed and applied a composite disparity index for each municipality to my adapted municipal infrastructure grant model to analyze and observe the impact of economic disparities in grant allocations. The findings show that a grant model that accounts for economic disparities satisfies the constitutional, economic and institutional considerations that should inform municipal grant allocation decisions. I conclude the thesis by highlighting the limitations and possibilities of using a municipal infrastructure grant model that accounts for economic disparities and, I propose some recommendations for applying such a model in South Africa.

This dissertation research aims to analyze the material Ejercicios Amigos Dos used by students learning Spanish as a foreign language in a Swedish school. Specifically, it examines the place of speaking skills, in the context of basic language skills. This study was an exploratory and descriptive analysis of the material Ejercicios Amigos Dos.Qualitative method used was action research, which was based on the real needs of the author of this study to enhance their teaching.Sheet one and Sheet two containing educational criteria for the analysis of teaching materials were used to measure the place of speaking skills.The main findings in the analysis showed that speaking skills are taken into account in the teaching material, however there are some micro skills missing. Speaking skills do not take a prominent role in the teaching material. Writing skills are emphasized to a greater extent.

This study explores the contribution that the learners and the teacher made to curriculum development in a process approach to English as a second language for adults. The research was carried out with a group of workers attending a basic English course in an adult education programme on the East Rand. This study covers the entire sixteen month period from 1986 to 1987, and was carried out by the teacher-as-researcher. The research methodology was a form of illuminative evaluation, using syllabus accounts. These accounts provided a record of what took place in the classroom and were organised into three main themes. Subsequent analysis of the accounts focussed on learner and teacher roles in determining the learning experiences generated in the classroom. These findings were then re-examined in the light of the literature on process approaches. Conclusions were drawn about these roles and the implications for teacher development and learner training. Subsequently these conclusions were presented as illuminative data to reflect upon the curriculum framework underlying process approaches. The conclusions drawn in this study challenge the assumption that learners and the teacher jointly and equally negotiate learning in a process approach. Instead they suggest that the teacher is very much in control as the overall manager of curriculum development, and that learners are secondary agents in this endeavour. The roles of both parties are also more complex and more interlinked than the literature implies. Following this, conclusions were drawn about the roles that learners and the teacher played in curriculum development in this study. The research went on to examine the broader implications of these findings, by assessing the feasibility of a process approach for large-scale adult basic education work in South Africa. From this assessment, a new 'hybrid' approach was proposed which retains important features of a process approach while modifying it to make it more accessible and appropriate for South Africa's needs. Finally, the study identified future research directions.

Esta dissertação tem como objetivo apresentar alternativas de ações que aprimorem o processo de implementação dos Conteúdos Básicos Comuns (CBC) em uma escola estadual participante do Projeto Escola-referência, pertencente à Superintendência Regional de Ensino de São João del-Rei (SRE/SJDR). Apesar de analisar as ações dos professores e EEB, enfatizou-se principalmente o papel do diretor escolar como elemento fundamental para a implementação das políticas educacionais na escola, bem como para a garantia do uso dos CBC como currículo oficial. Este estudo sobre a temática curricular fez-se importante, pois se partiu da compreensão de que o currículo é o centro das relações pedagógicas ocorridas na sala de aula e da atuação profissional que a autora exerce na função de inspetora escolar na SRE/SJDR. Assim, para cumprir o objetivo, buscou-se conhecer o desenho da política curricular oficial e como esta foi apropriada pela escola-referência, passando pela SRE/SJDR como primeira instância de implementação. A metodologia de pesquisa utilizada foi a análise de documentos referentes à política de implementação dos CBC, além da consulta a documentos, como livro de ponto, livro de atas, termos de visitas e relatórios de gestão; entrevistas com diretores, professores e especialistas de educação básica, diretora educacional e analistas educacionais da SRE, aplicação de questionário aos professores que atuam no Ensino Fundamental da escola analisada e observação nãoparticipante. À medida que o caso foi configurado, algumas questões de pesquisa foram levantadas e discutidas buscando alguns referenciais teóricos que abordam a implementação de políticas curriculares. Destarte, os dados encontrados na pesquisa demonstram que há uma proeminência da atuação do diretor escolar enquanto ator primordial na implementação da política pública curricular, vez ser ele quem coordena, articula e monitora sua equipe de trabalho na execução de suas funções, garantindo o seu envolvimento na implementação dos CBC.
The present dissertation has as its goal to present alternative actions which improve the implementation process of the Common Basic Syllabus (CBC, in Portuguese) at a state school that participates in the Reference School Project, belonging to the Regional Education Oversight of São João del-Rei (SRE/SJDR). Although we have analyzed the actions by the teachers and EEB, we have emphasized mainly the role of the school principal as a fundamental element to the implementation of the educational policies at school, as well as to guarantee the use of the CBC as the official syllabus. This study about curriculum is important, since we have comprehended that the curriculum is the center of the pedagogical relations that occur in the classroom and of the professional role which the author has as a school inspector at the SRE/SJDR. Thus, in order to fulfill our goal, we have aimed to understand the design of the official curricular policy and how it was appropriated by the reference school, viewing the SRESJDR as its first stage of implementation. As such, some conditionals and some variables were identified in both contexts which have contributed to the implementation of the CBC the way it was identified in the research. The research methodology utilized was the analysis of documents referring to the implementation policy of the CBCs and consultations to documents and well as check-in book, book of minutes, terms of visit and management reports/ interviews with principals, teachers and basic education specialists, school principal and education analysts at the SRE, survey with the elementary school teachers of the studied school and non-participant observation. As the case was configured, some research issues were raised and discussed regarding the implementation of curricular policies, Therefore, the data found in the research show that there is a prominence in the role of the school principal as a primary actor in the implementation of a curricular public policy, given that they are the ones Who coordinate, articulate and monitor their work teams in executing their functions and guarantees their involvement in the implementation of the CBC.

Il est courant de faire remonter l’origine du «problème corps-esprit» à Descartes et à sa distinction radicale des substances pensante et étendue. La question est bien connue : comment l’esprit, n’étant pas corporel, pourrait-il agir sur le corps et le mouvoir, et comment le corps pourrait-il agir en retour sur l’esprit, en causant ses sentiments et ses passions ? Si Descartes ne voyait là aucune difficulté qui aurait mérité d’abandonner la distinction des substances, ou au contraire la thèse d’une interaction causale réelle entre l’esprit et le corps, tel n’aura pas été le cas de ses lecteurs et interprètes. Ces derniers n’ont eu de cesse de faire valoir l’incohérence de son «dualisme interactionniste», souvent invoquée pour rendre compte de l’abandon, à l’âge classique, des relations causales entre l’esprit et le corps, et à l’époque contemporaine, du dualisme des substances au profit d’une ontologie physicaliste. Dans ce travail, nous revenons sur les difficultés engendrées par le cartésianisme concernant les rapports de l’esprit et du corps, en combinant trois perspectives qui associent réceptions classiques et contemporaines : celle des premiers objecteurs de Descartes ; celle de ses successeurs cartésiens, en s’intéressant à leur traitement de la difficulté liée à l’interaction âme-corps ; et celle des philosophes de l’esprit contemporains, dans le cadre de leur réflexion autour du Mind-body problem. Les problèmes philosophiques ainsi mis au jour sont utilisés pour relire les thèses de Descartes et proposer une nouvelle évaluation des doctrines sur la base des phénomènes psychophysiques qu’elles permettent ou non d’expliquer
It is common to trace back the origin of the “mind-body problem” to Descartes and to his radical distinction between extended and thinking substance. The question is well known: how can the immaterial mind act on the body and move it, and how can the body, in turn, act on the mind by causing its feelings and passions? While Descartes did not regard this as the source of any difficulties that might have necessitated the rejection of the distinction between substances or, inversely, of the theory of real causal interaction between mind and body, his readers and interpreters did. They constantly insisted on the inconsistency of his “interactionist dualism,” which is often invoked as a reason for discarding the theory of causal relation between mind and body in the classical period, and for replacing substance dualism with a physicalist ontology in the contemporary period.In this work, we return to the difficulties generated by Cartesian philosophy about the relationship between mind and body from three interrelated perspectives, which combine classical and contemporary receptions of Descartes: that of the first objectors to Descartes; that of his successors, with special consideration of their treatment of the difficulties involved in explaining mind-body interaction; and that of contemporary philosophers of mind, whose reflections on the mind-body problem are examined. Our approach to the philosophical issues thus brought to light allows us to revisit Descartes’s theses, and to propose a new evaluation of the doctrines on the basis of the psychophysical phenomena that they are capable of accounting for or not

La projection de la grève dans l’ordre international permet d’apprécier la richesse et la diversité de l’expression collective des travailleurs. Son développement à l’échelle internationale a généré une mise en concurrence accrue des différents droits nationaux. L’absence d’une réglementation matérielle traduit la difficulté juridique de maîtriser la notion et engendre un important conflit de lois et de juridictions. La nécessité de l’adoption de règles de conflit déterminant la loi et le juge compétent est donc devenue indispensable afin d’apporter une réponse à cette problématique conflictuelle. Le caractère de droit fondamental de la grève requiert cependant de faire l’objet d’une protection particulière lorsque sa valeur est remise en cause par une loi étrangère désignée par la règle de conflit. A ce titre l’exception d’ordre public classique permet d’évincer utilement la loi étrangère contraire à l’ordre public du for. L’exigence de la présence de liens de rattachement pertinents avec l’ordre juridique du for et d’un certain degré de gravité de la violation alléguée au droit fondamental de grève du for garantissent une intervention pondérée de l’exception d’ordre public. Elle constitue ainsi un moyen efficace de protéger le droit de grève tout en restant un mécanisme dont l’intervention reste exceptionnelle
The development of the strike in the international order can appreciate richness and diversity of the collective expression of workers, it also increased competition from different national laws. The absence of a regulation reflects the difficulty of controlling the legal concept and it creates a conflict of laws and courts. The need for the adoption of rules of conflict, which determines the law and the competent court, has therefore become necessary to provide an answer to this conflict problem. The character of the fundamental right to strike, however, requires to be special protection when its value is questioned by an authority designated by the conflict rule foreign law. As such, the conventional public policy exception allows oust usefully foreign law contrary to public policy. The requirement of the presence of links relevant connecting with the legal order and a certain degree of gravity of the alleged violation of the fundamental right to strike ensures a weighted intervention public policy exception. It constitutes an effective means of protecting the right to strike while remaining a mechanism whose intervention is exceptional

Today’s elementary school students face myriad traumatic issues including poverty, violence, physical and emotional abuse, homelessness, and parental substance abuse. These adverse childhood experiences are responsible for an increased risk of academic failure and behavioral problems in childhood, adolescence, and into adulthood. Social-emotional learning (SEL) programs, provided through school and community partnerships, attempt to address these needs in both school-based and out-of-school-time (OST) learning settings. The purpose of this action research study was to examine one northern California-based nonprofit organization’s OST SEL program for third through fifth grade students and determine actions and interventions for greater program effectiveness. Students, parent/guardians, site administrators, school-staff, and community members engaged in focus groups, completed surveys, participated in validation groups, and acted as research associates throughout the iterative plan, act, observe, reflect action research cycle conducted during the winter of 2020. Qualitative data from focus group meetings included identified themes from authenticated and coded transcripts while quantitative data included descriptive statistical analysis of pre-program and end-of-program surveys. As the lead researcher for the study, I worked with action research participants to make program modifications and identify new actions for program improvement. Based on themes and data trends, as well as the application of self-determination theory’s basic psychological needs theory, findings demonstrate that student self-management skills improved during the 4-week action research cycle, as did their sense of autonomy, competence, and relatedness. At the same time, findings suggest additional growth opportunities in the areas of responsible decision-making and program improvement through development and implementation of integrated and universal SEL supports in classrooms, schools, families, and the larger community. Linking the literature to these findings, recommendations for future action research cycles include age and developmental considerations regarding instruction and application of responsible decision-making skills, and integration of all five SEL competencies for greater harmonization of emotion and thought. To that end, the nonprofit organization’s educational design team and I have begun redesigning the curriculum. Program modifications address child development of perceptual and higher-level memory and cognitive skills, and intentional integration of instruction and practice in all five SEL competencies throughout each program module. Teaching, combined with real-time application of planning and decision-making skills, will include opportunities for active role-playing, adult guidance, and experiences in which students learn and grow from mistakes. In addition to student program modifications to build responsible decision-making skills, literature supports the call for universal SEL in the form of partnerships between schools, families, and community organizations for resource coordination resulting in more positive youth outcomes. Again working with the educational design team, the nonprofit organization and I have taken initial steps to establish a city wellness task force, bringing together a diverse group of stakeholders to partner in wellness for children and families in the community. We also continue to iterate as we develop a community of practice for educators, focused on building SEL knowledge and practice. By focusing on continuous improvement through an ongoing action research process, this study not only serves as an opportunity to celebrate successes, but highlight growth opportunities to advance the work of the northern California-based nonprofit organization and its programs. Utilizing study findings in combination with supporting literature, we are taking immediate action towards more positive outcomes for those we serve. This study also provides tools and guidance for other community partners in their design and implementation of effective SEL programs for the social and emotional well-being of elementary school students and families, and the communities in which they live.

Abstract The object of the study is the curriculum reform in basic education in the 2000's as local interpretations. The objective of the study is to create an image on how local municipal processes were formed and what kind of discourses in terms of educational policy, public administration and local level occurred. The empirical data of the study was collected in two cities, Oulu and Raahe, during 2002–2005. Part of the data was created in the processes and its so-called natural corpus whereby part of the material was acquired for the study. The analysis of the data was based on the constructionist approach, in which language is perceived as a social matter and in which the contextuality of meanings is recognized. In this case, the theoretical background of the study and earlier research on the curricula theory and practice, as well as research on the status and development of the school system form a part of qualitative hermeneutic interpretation. Characteristics of both action and ethnographic research can be seen in the study. Curricula have been used to direct the Finnish education for approximately one hundred years. The method of implementation has however changed in such a way that curriculum does not hold an established status in comprehensive schools. The unestablished status of curriculum can be seen also in this study. The concept of curriculum was defined in a number of different ways in the subjects' speech. Local processes differed from each other and experiences from previous reforms were not utilized. However, the teachers, principals and administrative personnel participating in the processes had a positive attitude towards the reform. The study will focus on how curriculum is defined in the subjects' speech concerning the school, municipality and department of education. The speech emphasises co-operation. The idea of up-to-date school and teachership could be maintained through curriculum work. Speech concerning the reform of the school emphasised the uniformity of comprehensive schools, which is linked with co-operation between different school levels, curriculum co-operation and school common to all pupils. The speech also highlighted the individuality of pupils. Speech concerning the municipality and the department of education raised the issues of multi-professionalism and regionality. Local strategic planning was raised in both processes. The example from Oulu shows that curriculum work may function as a tool for implementing the strategy of the department of education. This signifies a change in the administrative culture that has strengthened the position of local interpretation. The study also examined the subjects' speech in the perspective of educational policy discourses of the previous decade. In addition to national and local goals, international aspects can be identified. On the basis of this interpretation, operators in curriculum work were identified and named the voice of expertise and education, voice of developing local administration and voice of market economy. The voice of market economy is loud because it also affects the attitudes of the experts and developing local administration. In the background, conflicting assumptions on the society, individual and leadership can be perceived. These conflicts can be approached as alternatives in current discussion in which teaching personnel should also become more consciously involved
Tiivistelmä Tutkimuksen kohteena on 2000-luvun opetussuunnitelmauudistus perusopetuksessa paikallisina tulkintoina. Tutkimuksen tarkoituksena on ollut luoda kuvaa siitä, millaiseksi paikalliset kuntaprosessit muodostuivat ja millaisia koulutuspoliittisia, julkishallinnollisia ja paikallisia diskursseja niissä ilmeni. Tutkimuksen empiirinen aineisto on koottu kahdessa kaupungissa, Oulussa ja Raahessa, vuosien 2002–2005 aikana. Osa aineistoa on prosesseissa syntynyttä ns. luonnollista aineistoa ja osa tutkimusta varten hankittua. Tutkimusaineiston analyysi perustuu konstruktionistiseen lähestymistapaan, jossa kieli ymmärretään yhteiskunnallisena asiana ja jossa tunnustetaan merkitysten kontekstuaalisuus. Tällöin myös tutkimuksen teoreettinen tausta, opetussuunnitelmateoriaa ja käytäntöjä tarkasteleva aiempi tutkimus sekä koulun tilaa ja kehittymistä kuvaava tutkimus, ovat osa kvalitatiivista hermeneuttista tulkintaa. Tutkimuksessa voi nähdä sekä toimintatutkimuksen että etnografisen tutkimuksen piirteitä. Opetussuunnitelmilla on pyritty ohjaamaan suomalaista koulua noin sadan vuoden ajan. Toteuttamisen tapa on kuitenkin vaihdellut niin, ettei opetussuunnitelmalla ole vakiintunutta asemaa peruskoulussa. Vakiintumattomuus näkyy myös tässä tutkimuksessa. Opetussuunnitelmakäsitys määrittyi hyvin monin eri tavoin osallistujien puheessa. Paikalliset prosessit erosivat toisistaan, eikä niissä hyödynnetty edellisten uudistusten kokemuksia. Prosesseihin osallistuneet opettajat, rehtorit ja hallinnon henkilöstö ottivat uudistuksen kuitenkin myönteisesti vastaan. Tutkimuksessa tarkastellaan millaiseksi opetussuunnitelma määrittyy osallistujien koulua, kuntaa ja opetustoimea koskevassa puheessa. Puheessa korostui yhteistyö. Opetussuunnitelmatyöllä voitiin ylläpitää ajantasaisen koulun ja opettajuuden ihannetta. Koulua koskevassa muutospuheessa korostui peruskoulun yhtenäisyys, joka liitettiin kouluasteiden yhteistyöhön ja opetussuunnitelmayhteistyöhön sekä kaikille oppilaille yhteiseen kouluun. Puhe korosti oppilaiden yksilöllisyyttä. Kuntaa ja opetustoimea koskevassa puheessa nousivat esille moniammatillisuus ja seutukunnallisuus. Paikallinen strateginen suunnittelu nousi esille molemmissa prosesseissa. Oulun esimerkki osoittaa, että opetussuunnitelmatyö voi toimia opetustoimen strategian toteuttamisen välineenä. Tämä kertoo hallintokulttuurin muutoksesta, joka on vahvistanut paikallista tulkintaa. Tutkimuksessa tarkastellaan myös osallistujien puhetta suhteessa edellisen vuosikymmenen koulutuspoliittisiin diskursseihin. Puheessa voi erottaa kansallisten ja paikallisten tavoitteiden lisäksi kansainvälisiä vaikutteita. Tämän tulkinnan kautta erottuvat opetussuunnitelmatyössä vaikuttavat toimijatahot, jotka tutkimuksessa nimetään asiantuntijuuden ja sivistyksen ääneksi, kehittyvän paikallishallinnon ääneksi sekä markkinatalouden ääneksi. Markkinatalouden ääni on kantava, sillä se vaikuttaa myös asiantuntijoiden asenteissa ja kehittyvässä paikallishallinnossa. Sen taustalla voi kuitenkin nähdä ristiriitaisia oletuksia yhteiskunnasta, yksilöstä ja johtamisesta. Näitä ristiriitoja voi lähestyä ajankohtaisen keskustelun vaihtoehtoina, jossa myös opetushenkilöstön tulisi olla entistä tietoisemmin mukana

This thesis analyzes the Joint Action Plan - PAR and its deployment in the action set in the Plan Hall, under the auspices of the size of educational management in the Municipality of Portel in the state of Pará in the period 2007-2010. It also consists in raising the possible interference of the Integrated Planning, Budget and Finance of the Ministry of Education - SIMEC - PAR module, the process of conducting the municipal education.The overall objective of the thesis is to analyze the systemic process of implementation of PAR as strategic management tool municipal educational systems for teaching and its result in the construction of public policy and management of the municipal education of a city, geographically isolated in space Amazon, and the worst results in IDEB - the city of Para Portel in Marajó archipelago.The research methodology is a case study, a qualitative, and is characterized by analytical and comprises a literature review, documentary research and field research on the City of Portel in Marajó Archipelago , providing direct contact with staff or local committees , followed by semi-structured interviews with staff setting-up of the PAR . From the first theoretical-practical inserts revealed a systemic articulated public policy, technical, operational and bureaucratic, with a demand for action beyond the capacity of the municipal entity from the point of view of educational administration
A presente tese analisa o Plano de Ações Articuladas - PAR e seu desdobramento no conjunto de ações previstas no Plano municipal, sob a égide da dimensão da gestão educacional no Município de Portel, no Estado do Pará no período de 2007 a 2010. Consiste também em levantar as possíveis interferências do Sistema Integrado Planejamento, Orçamento e Finanças do Ministério da Educação SIMEC- módulo PAR, no processo de condução da educação municipal. O objetivo geral da tese é analisar o processo sistêmico de implantação do PAR, como instrumento estratégico de gestão educacional dos sistemas municipais de ensino e seu resultado na construção de políticas públicas e de gerenciamento do sistema municipal de ensino de um município, geograficamente isolado no espaço amazônico, e com os piores resultados no IDEB o município paraense de Portel, no arquipélago de Marajó. A metodologia da pesquisa é um estudo de caso, de cunho qualitativo, e se caracteriza pela modalidade analítica, compreende a revisão bibliográfica, a pesquisa documental e a pesquisa de campo no Município de Portel, no Arquipélago de Marajó, proporcionando contato direto com a equipe ou comitês locais, acompanhada de entrevistas semiestruturadas com a equipe elaboradora do PAR. A partir das primeiras inserções teórico-práticas evidenciou-se uma política pública sistêmica articulada, técnico-operacional e burocrática, com uma demanda de ações além da capacidade do ente municipal sob o ponto de vista da gestão educacional

O presente relatório retrata todo o processo de ensino e aprendizagem relativo ao meu desempenho no decorrer da prática de ensino supervisionada, que decorreu na Escola Básica Conde de Vilalva, com uma turma do 3º ciclo do ensino básico durante todo o ano letivo de 2016/2017, e na Escola Secundária Gabriel Pereira com a turma do ensino secundário ao longo do 3º período. Descrevo e analiso, de forma reflexiva, as decisões tomadas em várias dimensões do perfil de desempenho profissional: ao nível do planeamento e da avaliação do ensino e da aprendizagem, da condução do ensino e das atividades de participação na escola e relação com a comunidade. Apresento as estratégias desenvolvidas e os resultados obtidos sobre a investigação-ação que desenvolvi na turma sobre a problemática do desenvolvimento das competências sociais e relacionais nas aulas de Educação Física. As conclusões que retirei com a aplicação das estratégias no decorrer da investigação-ação foram bastante benéficas para a minha formação profissional; ABSTRACT: This report describes the whole process of teaching and learning related to my performance during the supervised teaching practice, which took place at the Conde de Vilalva Basic School, with a class from the 3rd cycle of basic education throughout the academic year 2016/2017 , and Gabriel Pereira Secondary School with the secondary school class throughout the 3rd period. I describe and analyze, in a reflexive manner, the decisions taken in various dimensions of the professional performance profile: in the planning and evaluation of teaching and learning, in the conduction of teaching and in the activities of participation in the school and relationship with the community. I present the strategies developed and the results obtained on the action research that I developed in the class on the problematic of the development of the social and relational competences in the classes of Physical Education. The conclusions I drew from the application of the strategies in the course of the action research were very beneficial for my professional training.

L'objectif de ma thèse est de participer à la construction d'un nouveau robot humanoïde qui peut réaliser des interactions intuitives avec l'humain à travers l'observation et l'imitation. Pour cela, j'ai conduit une série d'études expérimentales chez le jeune adulte pour caractériser les propriétés cinématiques des mouvements du bras réalisés pendant des interactions motrices et sociales, autant d'éléments qui seront les patterns de référence pour le futur robot. En se concentrant sur le comportement non-verbal, nous avons testé comment les contraintes externes et internes (difficulté, prédictibilité, temporalité) façonne la cinématique des mouvements du bras et de la main dans une simple action séquentielle de prise et de pose d'un objet (étude 1 et 2). Les résultats révèlent des modulations précoces dans la cinématique de la phase d'atteinte et de saisie, en fonction de la taille et de la stabilité du réceptacle terminal sur lequel l'objet devait être placé. Ces modulations observées dans le premier élément de la séquence sont en contradiction avec les modèles d'optimisation de trajectoire utilisés en robotique pour les séquences d'action. Ils suggèrent un couplage fort entre les paramètres moteurs dans une stratégie de planification encapsulée qui rétro-propage les contraintes contextuelles sur les éléments précoces de la séquence. Pour confirmer ces résultats, une seconde série d'étude a été conduite en utilisant des tâches cinématiques et vidéos pour montrer que les intentions motrices humaines pouvaient être lues à travers la détection de ces modulations cinématiques précoces. En utilisant un système de classification artificiel, nous avons testé si les indices de bas niveau pouvaient permettre une catégorisation des essais. Les résultats montrent qu'en absence de capacité cognitive particulière, le réseau de neurone pouvait catégoriser les intentions significativement au-dessus du niveau de la chance en observant les 500 premières millisecondes de l'action (étude 3). La troisième partie de mon travail de thèse s'est tournée vers les mesures en eye-tracking. Nous avons révélé ici que la stratégie proactive de fixations oculaires utilisée pendant l'observation de l'action était similaire à celle utilisée pendant son exécution (étude 4). De plus, les catégorisations correctes d'intentions motrices étaient caractérisées par des saccades plus précises et des fixations plus longues sur l'objet. Les mouvements oculaires sont connus pour jouer un rôle important dans les interactions sociales. Ainsi, dans une dernière expérience (étude 5), nous avons mis en place un jeu compétitif en face à face révélant des effets spécifiques du contexte social qui modifie la cinématique des mouvements d'atteinte selon le type de situations interactives. Dans le manuscrit de thèse je propose une discussion qui replace les résultats dans les modèles neuronaux et cognitifs de l'intégration sensori-mmotrice. Lorsque c'est le cas, des directions futures sont suggérées à la fois pour les modèles cognitifs de contrôle moteur et pour le développement e systèmes artificiels neuro-inspirés intégrant des capacités d'interaction sociale intuitive
The aim of my PhD thesis was to participate in the construction of a new humanoid robot that can sustain intuitive interactions with humans through observation and imitation. As such, I conducted a series of experimental studies in young adults to better characterize the kinematic properties of those arm movements performed during motor and social interactions, elements that are the reference patterns for the to-come robot. Focusing on non-verbal behavior, we tested how external and internal constraints (difficulty, predictability, timing) shaped the kinematics of both arm and hand movements in a very simple pick and place sequential action (study 1 and 2). The results revealed early modulations in kinematics in the reach-to-grasp phase depending of the size and the stability of the target pad on which the object had to be placed. These modulations observed within the first element of the sequence were in contradiction with the current optimized trajectory models used in robotics for action sequences. They suggest in fact a strong coupling of the motor parameters within an encapsulated planning strategy that back-propagates the contextual constraints on to the early elements of the motor sequence. To confirm these findings, a second serie of studies were conducted using kinematic and video based tasks to show that human motor intentions can be read through the detection of these early kinematic modulations (study 3). Using basic artificial classification, we tested whether low-revel motor indices could afford trial categorization without the need for higher-level process such as motor imagery. results indicated that indeed without cognitive abilities the neural network could categorize the intention of an observed action within the first 500ms, significantly above chance level (study 4). The third place of my PhD work turned to eye tracking. Here, we revealed that the proactive strategy of eye-fixations used during action observation were similar to those made during executed actions. Additionally, good categorization of motor intention was characterized by more accurate saccades and longer object fixations. Eye movements are known to play an important role in social intercations. Hence, in a final experiment (study 5), we setup a face-to-face competitive game to reveal the specific effects thet the social context may play on the kinematic properties of reaching during different types of interactive situations. In the PhD mansucript, I propose a general discussion that sets these results within the current cognitive and neuronal models of sensori-motor integration. When appropriate, future directions are suggested both for cognitive models of motor control and for the development of neuro-inspired articicial systems constitued with intuitive social interaction skills

P2X receptors are ATP-gated cation channels. P2X1 receptors are widely expressed throughout the body and have a range of functional roles, e.g. contraction of mesenteric arteries and regulation of blood clotting. The recent crystallisation of the zebra fish P2X4 receptor has provided a major advance in understanding the molecular basis of receptor properties. However, how agonists or antagonists are co-ordinated and the extent of the proposed ligand binding site have not been addressed at a structural level. A mutagenesis based approach was used to propose a model of the ATP binding site and has highlighted some residues involved in antagonist action at P2X receptors. The aim of this thesis was to investigate the molecular basis of antagonist action at the P2X1 receptor using site-directed mutagenesis and P2X receptor chimeras. The wild-type, mutant and chimaeric P2X receptors were expressed in Xenopus laevis oocytes and the currents were characterised using two electrode voltage clamp. Initially, suramin was shown to act as a competitive antagonist and PPADS as a non-competitive antagonist at the P2X1 receptor. The contribution of residues V74 to G96 to human P2X1 receptor properties were determined using cysteine scanning mutagenesis. This region contains a residue that has been shown to be important in suramin action at the P2X4 receptor (K78) but cysteine mutation of the residues V74 to G96 had either no effect or slightly increased antagonism by suramin or PPADS. Also, a further residue was found to be important in ATP potency (F92) and the use of partial agonists and modification with cysteine reactive methanethiosulfonate (MTS) reagents identified additional residues important in channel activation. Mapping these residues onto a homology model of the P2X1 receptor showed the depth of the agonist binding site and highlighted the importance of the rear/inner cavity of the binding pocket in the gating of the channel subsequent to agonist binding. The cysteine rich head region of the P2X receptor, which is adjacent to the proposed ATP binding pocket, is absent in the antagonist insensitive Dictyostelium receptors. P2X1 and P2X2 receptors have ~1400-fold difference in sensitivity to a suramin analogue NF449. Chimeras and point mutations in the cysteine rich head region were made between the P2X1 and P2X2 receptors and they identified the region between the third and fourth conserved cysteine residues of the P2X1 receptor as being important in conferring the difference in sensitivity. In particular, the positively charged residues at the base of the cysteine rich head region of the P2X1 receptor accounted for the highly selective antagonism of NF449 at the P2X1 receptor. Additionally, these residues were shown to play a role in the molecular basis of suramin and PPADS action at the P2X1 receptor. Reciprocal chimeras and mutations in the P2X2 receptor produced modest increases in antagonist sensitivity. In silico docking models highlighted possible sites of action for NF449 and suramin on the P2X1 receptor showing that the base of the cysteine rich head region may be important in the binding of antagonists. In summary, this research furthered understanding of ligand action at the P2X1 receptor.

The function of the human endometrium is to accept the implanting embryo and provide an environment for its subsequent growth and development by forming the maternal side of the placenta. The endometrium undergoes rapid cyclic changes of cell proliferation, differentiation and renewal and briefly becomes receptive to the embryo allowing its attachment and subsequent invasion during the implantation window. These processes are controlled systemically by steroid hormones which are thought to activate and control local molecular effectors including chemotatic cytokines known as chemokines. Chemokines are known to recruit leukocytes by chemotaxis, but also have a wider multifunctional in the control of cell function. Limited studies have shown expression of the chemokine ligand CXCL12 and its receptor CXCR4 in the human endometrium. However, little is known about the precise role of these molecules in endometrial function. To investigate this further, real-time RT-PCR was used to measure the expression of CXCL12 and CXCR4 mRNA in endometrial biopsies throughout the menstrual cycle. Both CXCR4 and CXCL12 were expressed by the endometrium at all stages of the cycle. Expression of CXCL12 mRNA was relatively low and did not change throughout the menstrual cycle. However, CXCR4 mRNA significantly increased during the early proliferative phase of the cycle, with a second slight increase during the mid secretory phase. Expression of CXCL12 and CXCR4 in cultured human endometrial epithelial and stromal cells was investigated using real-time RT-PCR used to measure levels of CXCL12 and CXCR4 mRNA expression. CXCL12 expression was found to be higher in stromal cells compared to epithelial cells, while expression of CXCR4 was highest in the endometrial epithelial cells. Expression of CXCR4 mRNA was also investigated in Ishikawa and HEC-1-B cell lines. Both cell lines were found to express CXCR4 mRNA, though levels were higher in Ishikawa cells compared to HEC-1-B cells, which was more comparable to their primary epithelial cell counterparts. Although the functions of CXCL12 in the endometrium remain unknown, CXCL12 has been shown to increase the expression of interleukin 6 (IL6), interleukin8 (IL8), matrix metalloproteinase 2 (MMP2), matrix metalloproteinase 9 (MMP9) and vascular endothelial growth factor A (VEGFA) in various non-endometrial cell types. As all of these factors have previously been shown to be of potential importance to endometrial function, their basal mRNA expression was quantified in primary epithelial and stromal endometrial cells and Ishikawa and HEC-1-B cell lines using real-time RT-PCR. To investigate effects of CXCL12 on these endometrial factors, all four endometrial cell types were incubated with CXCL12 and the expression of IL6, IL8, MMP2, MMP9 and VEGFA was measured using real-time RT-PCR. Basal levels of IL8 and MMP9 mRNA were found to be significantly higher in endometrial epithelial cells in comparison to stromal cells, while VEGFA was significantly higher in stromal cells in comparison to epithelial cells. Expression of IL6, IL8, MMP2 and MMP9 was several orders of magnitude lower in the Ishikawa and HEC-1-B cell lines in comparison to their primary epithelial counterparts suggesting that these cell lines are not good models to investigate endometrial function. Incubation with CXCL12 had no significant effect on the expression of any of these factors, in primary epithelial, stromal, Ishikawa or HEC-1-B cells, which suggests CXCL12 may not regulate their expression in the endometrium. To investigate the potential wider effects of CXCL12 on the expression of endometrial factors, a genome-wide microarray expression analysis was carried out in Ishikawa cells. Incubation of these cells with CXCL12 resulted in the identification of fifty one mRNA transcripts that were shown to be significantly and reproducibly up-regulated by CXCL12. Fourteen of these transcripts were produced by unknown genes, but thirty seven transcripts could be identified and potential functions attributed to them. Several of these genes have previously been shown to be of potential importance to endometrial function or to be known components of the CXCR4 signaling pathways. Overall, the results of this study have increased our knowledge of the expression and cellular distribution of CXCL12 and CXCR4 in the endometrium. This study has also identified several potential roles for CXCL12 and CXCR4 in endometrial function which would warrant further investigation.

Synovial sarcoma is a highly invasive soft tissue malignancy of young people, for which current treatment provides limited benefit. A chromosomal translocation t(X;18) is characteristic of this disease, and the resulting fusion SS18-SSX acts as an oncoprotein that promotes synovial sarcoma oncogenesis through unclear mechanisms. The work presented in this thesis aims to understand the molecular basis of SS18-SSX oncogenic action. Using patient-derived synovial sarcoma cells, we characterized a multicomponent SS18-SSX complex and discovered a scaffolding role for SS18-SSX in connecting two previously unlinked molecules, activating transcription factor 2 (ATF-2) and transducin-like enhancer of split 1 (TLE1). The latter recruits Polycomb-group (PcG)/histone deacetylase (HDAC) repressors to SS18-SSX transcriptional complex, to silence ATF-2 target genes. In accordance with this model, HDAC inhibitors effectively reverse PcG/HDAC-mediated epigenetic repression, rescue the expression of SS18-SSX target genes, and suppress synovial sarcoma tumor cell survival. Beyond this finding, we further showed that treatment with HDAC inhibitors prevents TLE1 recruitment and triggers SS18-SSX oncoprotein degradation, thereby disrupting this aberrant transcriptional complex. Collectively, these findings advance our understanding of the molecular mechanisms underlying SS18-SSX action in synovial sarcoma pathogenesis. This thesis also provides further support for the clinical evaluation of HDAC inhibitors alone or in combination with other agents to treat synovial sarcoma.

Quinolones are a clinically-useful class of antibacterial agents known to target DNA gyrase, a bacterial type II topoisomerase. Gyrase is unique among topoisomerases in its ability to introduce negative supercoils into DNA using the energy derived from ATP hydrolysis. The active enzyme is composed of two GyrA and two GyrB subunits, forming an A2B2 tetramer of molecular weight 374 kDa. The mechanism of supercoiling by gyrase involves the ATP-driven passage of one segment of DNA through a gyrase-stabilised double-stranded break in another. Tyrosine 122 of E. coli GyrA becomes covalently attached to DNA when gyrase breaks the phosphodiester bonds of DNA during supercoiling. When this residue is mutated to serine or phenylalanine, gyrase can no longer cleave or supercoil DNA, but can bind DNA normally. Rapid-gel filtration experiments have shown that quinolones can still bind to proteins bearing these mutations, suggesting that DNA cleavage by gyrase is not required for quinolone binding. Transcription by T7 and E. coli RNA polymerases is blocked by the presence of a gyrase-quinolone-DNA complex. Mapping of the transcription termination sites in the presence of gyrase and quinolones shows that blocking occurs about 10 to 20 base-pairs upstream of the gyrase cleavage site. Blocking of transcription by T7 RNA polymerase by a gyrase-quinolone complex on DNA does not occur when the active-site tyrosine of gyrase is mutated to serine, which indicates that the polymerase blocking requires DNA cleavage. Analysis of transcription in the absence of drug suggest that RNA polymerase does not displace gyrase from the template. DNA gyrase is also the target of the CcdB protein which is encoded by the F plasmid. When its action is not prevented by CcdA protein, CcdB is a potent cytotoxin. Using in vitro transcription by T7 RNA polymerase, it has been shown that CcdB complexed with gyrase can block transcription in a similar manner to the gyrase-quinolone complex. Furthermore, in the presence of CcdA, CcdB can no longer induce gyrase to block transcription.

This thesis has evaluated the roles of dorsal and ventral processing streams in recognition and use of objects. Four main empirical studies are presented. First, to investigate how the cortical brain processes semantic and action knowledge in different object-related tasks, I examined structural data from stroke patients (Chapter 2) and functional data from healthy individuals (Chapter 3) using a voxel-wise statistical analysis method. Using data of different modalities (structural CT, fMRI) from different sources (patients’ lesions; healthy subjects’ functional activity) handled with a systematic analysis approach, I attempted to find convergent evidence to support the dissociation of semantic and action processing. Second, I also looked into the potential differentiation within the mechanisms underlying object-related action (Chapter 4) and object naming (Chapter 5) separately. Overall, comparable findings were provided from the voxel-based morphometric analysis of patients’ lesion data and the fMRI study with healthy participants: an association was observed between ventral brain structures and the retrieval of semantic knowledge/object recognition while a dorsal fronto-parietal-occipital network was found to support the processing of action knowledge/object-oriented action. Specific dissociations were also observed within the representations for object-oriented actions as well as the mechanisms underlying naming of objects.

Actin isoform specific mutations have been identified as causes for various human diseases. These include twelve missense mutations in γ-nonmuscle actin leading to early onset autosomal dominate nonsyndromic hearing loss and twenty two missense mutations in α-smooth muscle actin leading to thoracic aortic aneurysms and dissections (TAAD). The molecular mechanisms leading to these human pathologies are mainly unknown, principally due to the inability to isolate pure mutant γ-nonmuscle actin and α-smooth muscle actin in quantities required for biochemical analysis. To begin to address these problems, I have individually expressed the human nonmuscle actin isoforms, β– and γ– nonmuscle actin, in a baculovirus expression system and characterized their biochemical properties. Surprisingly, despite a conserved amino acid difference of only 4 residues at or near the N-terminus, Ca-γ-actin exhibits slower monomeric and filamentous biochemical properties than β-actin. In the Mg-form, the difference between the two is smaller. Mixing experiments with Ca-actins reveal the two will readily co-polymerize. Calcium bound in the high affinity binding site of γ-actin may cause a selective energy barrier relative to β-actin that retards the equilibration between G– and F-monomer conformations resulting in a slower polymerizing actin with greater filament stability. This difference may be particularly important in sites such as the γ-actin-rich cochlear hair cell stereocilium where local mM calcium concentrations may exist. In hair cells γ-nonmuscle actin seems to play a central role in stereocilia maintenance. To determine how the deafness causing D51N-γ-mutant actin mutation leads to deafness, I expressed and characterized it in the γ-actin background. The D51N mutation, lethal when cloned into yeast, displayed decreased filament stability and polymerization kinetics of an actin more dynamic than γ-actin. This result suggests that the hearing effects of the γ-actin mutations on the hearing apparatus are not simply caused by an inability to polymerize. The observed increased polymerization rates and decreased filament stability may have major implications for the human disease, as the mutation may alter the ability of the γ-actin to fulfill its maintenance functions. To address the basis by which TAAD mutations cause vascular dysfunction I introduced two of the know human mutations, N115T and R116Q, into yeast actin, 86% identical to human α-smooth muscle actin. I then generated yeast strains expressing each of these mutations as the sole actin in the cell to assess their effect on actin function in vivo and in vitro. Both mutant strains exhibited reduced ability to grow under a variety of stress conditions, although the N115T cells were more severely affected. In vitro the mutations caused exhibited altered thermostability and nucleotide exchange rates indicating effects on monomer conformation with R116Q the most severely affected. The N115T actin demonstrated a biphasic elongation phase during polymerization, while R116Q actin demonstrated a markedly extended nucleation phase. Allele-specific effects were also seen on critical concentration, rate of depolymerization and filament treadmilling. R116Q filaments were hypersensitive to severing by the actin-binding protein cofilin. In contrast, N115T filaments were hyposensitive to cofilin, despite near normal binding affinities of actin for cofilin. The mutant specific effects on actin behavior suggest that individual mechanisms may contribute to TAAD. Understanding the mechanisms of actin dependent human diseases requires elucidation of the effects of the mutations on the behavior of actin per se, its regulation, and the impact on actin mediated processes within the cell. The work provided in this thesis and future studies will provide the information required to understand the pathways involved in these diseases and form innovative treatments for deafness and TAAD.

This work has as main objective to analyze the relations of power and actions, and their influence in the teaching practices and teacher of Geography, and therefore the implications on the educational role of teaching Geography. With training and practice teaching subject matter of the present research. From of the assumption that there are internal and external factors that influence the processes of training and development activities of teachers and students, makes it a genealogical approach aiming to arrest and understand the relationships interdependence in the core of each institution involved directly in the process of training or teaching-learning and between them. Therefore, this paper starts by a dialectical approach about the importance and interference of the institutions involved in the Brazilian educational process, family, state, school and university. Working concepts and some of its specificities, with the aid of a wide geographical literature, pedagogical, philosophical and sociological ,the research emerges and consolidates with the study and empirical analysis. Thus, in addition to literature, which focuses this field of research, we obtained data and information through official documents, interviews and questionnaires, with the participation of two teachers who exercise effective in basic education; three teachers trainers Degree in Geography; twenty-one undergraduates 8th period Course of Geography and forty students in the 9th grade of Elementary School . Informed by analysis of the content, the empirical work revealed that teachers and students of the two levels of education, basic and higher, are aware of the need for redefinition of training, study and practice. But at the same time, says the barriers experienced in education are from the educational system itself, but rather, the socioeconomic and political structure of the country. We hope that this work will motivate other educational subjects to deepen their studies and develop research that contributes to the proposals of experts, teachers and students are considered and not just heard and dismissed.
Esta dissertação tem como objetivo principal, analisar as relações de poderes e ações e, suas interferências na formação e práticas docentes de professores de Geografia, e, portanto, as implicações sobre a função educativa do ensino de Geografia. Sendo a formação e a prática docentes objetos de estudo da presente pesquisa. Partindo do pressuposto de que existem fatores internos e externos que influenciam os processos de formação e desenvolvimento das atividades docentes e discentes, realiza-se uma abordagem genealógica objetivando apreender e entender as relações de interdependência no bojo de cada instituição envolvida diretamente no processo de formação e ou ensino aprendizagem e entre as mesmas. Portanto, esse trabalho inicia-se por meio de uma abordagem dialética sobre a importância e interferências das instituições envolvidas no processo educacional brasileiro, família, Estado, escola, e Universidade. Trabalhando conceitos e algumas de suas especificidades, com o auxílio de uma ampla literatura geográfica, pedagógica, filosófica e sociológica a pesquisa desponta e consolida-se com o estudo e análise empírica. Assim, além da pesquisa bibliográfica, que focaliza esse campo de investigação, obtivemos dados e informações por meio de documentos oficiais, entrevistas e questionários, com a participação de dois professores que exercem efetivo exercício no Ensino Básico; três professores formadores do Curso de Licenciatura em Geografia; vinte e um licenciados do 8º período do Curso de Geografia e com quarenta alunos da 9º Ano do Ensino Fundamental. Orientado pela análise desse conteúdo, o trabalho empírico revelou que professores e alunos dos dois níveis de ensino, básico e superior, são conscientes das necessidades de ressignificação da formação, do estudo e da prática. Mas ao mesmo tempo, afirma que os entraves vividos na educação são provenientes do próprio sistema educacional, mas, sobretudo, da estrutura socioeconômica e política do país. Esperamos que esse trabalho venha a motivar outros sujeitos educacionais a aprofundarem seus estudos e desenvolvam pesquisas que contribuam para que as proposições de especialistas, docentes e discentes sejam consideradas e, não simplesmente ouvidas e descartadas.

In my essay, I will explore how, as a primary teacher in a leisure home, I may try to communicate the value base we are to convey equal to the central content. It is in the sign of measurability that the school is now in the approach we applied to the students, that they have also begun to apply to themselves and their fellow human beings. They place their intrinsic value outside themselves and on results and other measurable things. I want to be able to work for them to attribute to them a self-worth and a good self-esteem, regardless of their study results or other measurable things. They live and breathe and it should suffice to acknowledge their existence and worth as fellow human beings. The questions that underlie this essay arose because I felt that we lack a dimension in discussions about the school that can highlight how we as teachers work with motivation or self-esteem. After all, trying to get students to approach new things they can't yet and trust in their own and the common life is, after all, perhaps the most important thing we can work on. I used two dilemmas where the students in their everyday situations at school express their despair and frustration at not really feeling that they are worth living as they are. I will then, through my questions, try to find out if there is any way to find a better way to equip them for life and its challenges. I start from a hermeneutic perspective to emphasize the importance of self-reflection as an important part of being able to meet and deal with myself, subject and object at the same time. I have chosen to use Gert Biesta's thoughts on what functions are at play when we talk about education. He wants us to focus on what these functions do for us so that we can discuss what is desirable based on it and not by simply looking at "what works" and what can be measured. Together with Biesta, Hannah Arendt's thoughts on the active life helps me in my search for an educational look at education that values people as complex beings and not just certain measurable parts of us.
I min essä kommer jag utforska hur jag som grundlärare inom fritidshem kan försöka kommunicera värdegrunden vi ska förmedla likställt med det centrala innehållet. Det är i mätbarhetens tecken som skolan nu befinner sig, och det mätbara synsätt vi applicerar på eleverna har de även börjat tillämpa på sig själva och sina medmänniskor. De lägger sitt egenvärde utanför sig själva, på resultat och andra mätbara saker. Jag vill kunna jobba för att eleverna ska tillskriva sig själva ett egenvärde och en god självkänsla oavsett studieresultat eller annat mätbart. De lever och andas, det borde räcka för att bekräfta deras existens och värde som medmänniskor. De frågor som ligger till grund för denna uppsats uppstod genom att jag ansåg att vi saknar en dimension i diskussioner om skolan som kan lyfta fram hur vi som lärare jobbar med motivation eller självkänsla.  Att försöka få eleverna att ge sig på nya saker de ännu inte kan och ha tillit till det egna och det gemensamma livet är trots allt kanske det viktigaste vi kan arbeta med. Jag har utgått från två dilemman där eleverna i deras vardagssituationer i skolan uttrycker sin förtvivlan och frustration över att inte riktigt känna att de duger som de är. Jag kommer sedan genom mina frågeställningar försöka undersöka om det finns något sätt att bättre rusta dem inför livet och dess utmaningar. Jag utgår från ett hermeneutiskt perspektiv för att poängtera vikten av självreflektion som en viktig del i att kunna möta och bemöta mig själv som subjekt och objekt samtidigt. Jag har valt att ta hjälp av Gert Biestas tankar om vilka funktioner som är i spel då vi pratar om utbildning. Han vill att vi ska fokusera på vad dessa funktioner gör för oss för att på så sätt kunna diskutera vad som är önskvärt och inte genom att enkom titta på ”vad som fungerar” och vad som kan mätas. Tillsammans med Biesta hjälper mig Hannah Arendts tankar om det aktiva livet mig i sökandet efter en pedagogisk blick på utbildning som värdesätter människor som komplexa varelser och inte bara vissa mätbara delar av oss.

In this thesis I examine and explore whether and if Integrated Water Resources Management (IWRM) inspired water reforms respond to- and address the diverse realities of women and men in informal (and formal) rural economies of Sekororo, South Africa and Ward 17 in Gwanda, Zimbabwe which are both in the Limpopo basin. South Africa and Zimbabwe, like other southern African countries, embarked on IWRMinspired water reforms, culminating in the promulgation of the National Water Acts in 1998, four years after the attainment of South Africa’s democracy in 1994 and 18 years after Zimbabwe attained independence in 1980. I argue that the adoption of IWRM, which emphasises second generation water issues such as demand management, water quality, environmental flow requirements etc, and not the development of water infrastructure, begs the question whether such reforms can make a meaningful contribution to the development agenda in countries where, during apartheid and colonialism, the water rights (among other rights) of millions of blacks were compromised because of unjust legislation and skewed underinvestment in water infrastructure

Dance is a universal and ancient human behavior; however, our understanding of the basis of this behavior is surprisingly weak. In this dissertation, I explore the cognitive and evolutionary foundations of human dance, providing evidence of two ways in which the production and perception of dance actions are rooted in the functions of more general cognitive systems.In doing so, I aim to both inform our understanding of dance, and use the study of dance to elucidate broader issues in cognition. Chapter 1 demonstrates that the ability to entrain, or move in time with an auditory beat, is not unique to humans. In addition, across hundreds of species, I find that all animals able to entrain can also vocally imitate sound. This supports the hypothesis that entrainment relies on cognitive machinery that originally evolved to support vocal imitation. Chapter 2 investigates the perception of dance-like actions. Previous work shows that we infer the goals of observed actions by calculating their efficiency as a means to external effects, like reaching an object or location. However, dance actions typically lack an external effect or external goal. In two experiments, I show that for dance-like actions, adults infer that the agents’ goal is simply to produce the movements themselves. Furthermore, this inference is driven by the actions’ inefficiency as a means to external goals. This inefficiency effectively rules out external goals, making movement-based goals the best explanation. Thus, perception of both dance and non-dance actions appears to rely the same type of efficiency-based goal inference. Chapter 3 builds on these findings, showing that the inference that the movements are the goal is closely related to our concept of dance. First, I find that participants view movement-based goals as more consistent with dance than with other activities. Second, I find that simply construing actions as having movement-based goals leads participants to view the actions as more dancelike, even when all participants have seen the exact same actions. Thus, even our categorization of actions as dance versus non-dance is rooted in the same cognitive processes as support our understanding of other intentional actions.

Calponins (CaPs) are actin-binding proteins that stabilize actin filaments. Mammals express three genetic isoforms in a tissue-specific manner. CaPs share an N-terminal calponin-homology (CH) domain followed by short tandem sequence repeats termed calponin-like (CLIK ) repeats. The extreme C-termini of CaPs contain isoform-specific tail sequences. Basic calponin (hlCaP) was the first isoform to be discovered 22 years ago in an attempt to identify troponin-like proteins in smooth muscle. The scope of this thesis was to reveal the mechanisms that regulate the actin-binding activity of hlCaP.

The reason is the depletion of hydrocarbons and the relatively facile production of hydrogen from various renewable sources of energy. Hydrogen can be combusted in an efficient way in a fuel cell with water as emission product. The overall goal of the project was to deevlop the knowledge base for solid-state hydrogen storage technology suitable for stationary and mobile applications. The aim of this research was to develop a novel composite hydrogen storage material with high wt% storage capacity, high intrinsic safety, appropriate thermodynamics, high mechanical strength, reversibility of the system and fast kinetics based on a well known "

Résumé :

The Orange Basin was formed during the late Jurassic to early Cretaceous periods due to Gondwana breakup and rifting and later drifting apart of the African and South American plates. The basin consists of siliciclastic sandstone which took its sediment supply from river system with a rivalling delta to the north of the basin. Geological and petrophysical studies were carried out to evaluate the reservoir potential of the wells in the study area. This study considered five wells (A-G1, A-W1, A-K1, A-K2 and A-Y1) in the Orange Basin with attention to the Albian age sandstone. Only three of the studied wells (A-G1, A-W1 and A-K1) have core samples for analysis. The methods used for the execution of this study include the description and calibration of spot cores with conventional standard logging record responses, wireline log interpretation using sequence stratigraphy approach, detailed petrographic (SEM, HR-TEM, XRD and thin section) and geochemical (pore water geochemistry, FTIR and XRF) analyses, and petrophysical analysis to unravel the complexities with regard to facies association, depositional environment and diagenesis. Linking diagenesis to depositional facies and sequence stratigraphy has given a clearer picture to the spatial and temporal distribution of diagenetic alterations and thus of evolution of reservoir quality in the studied wells. Three depositional lithofacies were identified based on a detailed core description [fine grained sandstone (F1), very fine grained sandstone (F2) and mudstone (F3)]. Fluvio-deltaic and shallow marine environments were also interpreted from the core description based on the sedimentary structures and mineral assemblage while the log interpretation shows that the different reservoir units range between LST, TST and HST but mostly of LST. Mineralogical predictions were made possible in the wells without core samples (A-K2 and A-Y1) through the use of density-neutron cross plot, these reveal that the two wells contain some considerable amount of clay minerals like kaolinite, chlorite and illite.

A thorough document review will be done of the policies and related materials around BMCs, where events will be constructed from the findings. Interviews will be conducted for verification purposes, to verify the desktop findings and to assimilate any conflicts of opinion that might have not been documented.

The reservoir quality (RQ) of well E-BA1 was characterized using thin sections and core samples in a petrographic study. Well E-BA1 is situated in the Bredasdorp Basin, which forms part of the Outeniqua Basin situated in the Southern Afircan offshore region. Rifting as a result of the break up of Gondwanaland formed the Outeniqua Basin. The Bredasorp Basin is characterized by half-graben structures comprised of Upper Jurassic, Lower Cretaceous and Cenozoic rift to drift strata. The current research within the thesis has indicated that well E-BA1 is one of moderate to good quality having a gas-condensate component.

This research into Chronic Fatigue Syndrome (CFS) presents the results of 60 first-person psycho-phenomenological interviews with 30 New Zealand women. The participants were recruited from the Canterbury and Wellington regions, 10 had recovered. Taking a non-dual, non-reductive embodied approach, the phenomenological data was analysed semiotically, using a graph-theoretical cluster analysis to elucidate the large number of resulting categories, and interpreted through the enactive approach to cognitive science. The initial result of the analysis is a comprehensive exploration of the experience of CFS which develops subject-specific categories of experience and explores the relation of the illness to universal categories of experience, including self, ‘energy’, action, and being-able-to-do. Transformations of the self surrounding being-able-to-do and not-being-able-to-do were shown to elucidate the illness process. It is proposed that the concept ‘energy’ in the participants’ discourse is equivalent to the Mahayana Buddhist concept of ‘contact’. This characterises CFS as a breakdown of contact. Narrative content from the recovered interviewees reflects a reestablishment of contact. The hypothesis that CFS is a disorder of action is investigated in detail. A general model for the phenomenology and functional architecture of action is proposed. This model is a recursive loop involving felt meaning, contact, action, and perception and appears to be phenomenologically supported. It is proposed that the CFS illness process is a dynamical decompensation of the subject’s action loop caused by a breakdown in the process of contact. On this basis, a new interpretation of neurological findings in relation to CFS becomes possible. A neurological phenomenon that correlates with the illness and involves a brain region that has a similar structure to the action model’s recursive loop is identified in previous research results and compared with the action model and the results of this research. This correspondence may identify the brain regions involved in the illness process, which may provide an objective diagnostic test for the condition and approaches to treatment. The implications of this model for cognitive science and CFS should be investigated through neurophenomenological research since the model stands to shed considerable light on the nature of consciousness, contact and agency. Phenomenologically based treatments are proposed, along with suggestions for future research on CFS. The research may clarify the diagnostic criteria for CFS and guide management and treatment programmes, particularly multidimensional and interdisciplinary approaches. Category theory is proposed as a foundation for a mathematisation of phenomenology.

The Sevier River Basin situated in south central Utah is characterized by its semiarid climate, snowmelt-driven runoff, and high dependency on agricultural economy. High evapotranspiration and low precipitation make agricultural production challenging, but naturally stored water in the snowpack in the mountains alleviates water stresses during high water demand seasons. The snowmelt-driven river flow along the main channel is highly exploited for irrigation for farms near the Sevier River. Reservoir operations and river diversions result in heavily regulated flows from the upper to the lower basins. The return flows of over-irrigated water in the upper basin increase salinity of surface water. Long-term applications of salinity water in agriculture eventually produce high soil salinity in the agricultural areas near Delta in the lower basin, which deteriorated farmers’ crop productivity. Farmers cropping near Delta struggle with both water and salinity stresses. Indeed, crop prices and yields are always their concerns. For them, efficient water management can be achieved with consideration of hydrologic, agronomic, and economic aspects of water resources. The overall goal of this research was to develop a decision supporting framework for efficient water and land allocations that considered hydrologic processes, crop response to water in salinity-affected farms, and farmers’ profit and financial risk. This research introduces a methodology for predicting water availability in a given cropping year from the snowpack in the mountains, and agronomic simulations with satellite images follow for quantifying crop response to water. The hydrologic predictions and the agronomic simulations are finally incorporated into an economic analysis that provides efficient water and land allocations with multiple crop selections. In a rural river basin, data limitation is a common concern for water resources engineers; thus simple but robust methodologies are proposed for hydrologic prediction. In the same context, satellite images are used for the estimation of crop yields in individual farms near Delta with no prior crop experimental plots. Historical records of crop prices are used for the economic analysis. The methodologies developed in this research provide a comprehensive decision analysis framework for efficient water management where water is scare and available from snowmelt only, the economy depends on agriculture only, and salinity is present in both soil and water due to long-term irrigation. The case study is for the agricultural area near Delta in the Sevier River Basin, but its applicability is not limited and is flexibly applicable to other agricultural regions.
