Francesco di Giorgio Martini (c. 1480-98) elabora método para concepção de templos, a partir de esquemas geométrico-proporcionais pautados por círculos e quadrados, alinhado às prescrições vitruvianas sobre symmetria e analogia arquitetura e corpo humano. Henry Millon e Richard Betts iniciaram estudos de cotejamento destes esquemas com obras e desenhos coevos. Interessa, aqui, continuar tais estudos aplicados às igrejas de San Bernardino degli Zoccolanti e Santa Maria delle Grazie al Calcinanio, ambas atribuídas a Martini. Pretende-se responder lacunas apontadas pelos historiadores e demonstrar que, longe da repetição estereotípica, o método formulado por Martini concerne flexibilidade e adequação a diversidade de exigências impostas pela ideação arquitetônica, bem como avanço técnico e compositivo para superação das referências antigas e coetâneas.

In December 1499 the Hypnerotomachia Poliphili appeared in Venice under the imprint of Aldus Manutius, in an edition whose woodcuts and typography make it one of the masterpieces of Italian printing. The enigmatic dream recounted in the book describes Poliphilo's struggle to win his love and his final happy union with his beloved Polia. The study focuses on the group of architectural representations in the book. Free from a slavish attempt to illustrate, they follow the course of the love story within the framework of an inner visual logic. A second part analyzes the section of the Temple of Venus Physizoa, an architectural representation that was omitted in Lotz's famous study of 1956. Issues of the history of technology, such as the modern cartographic method introduced by Alberti in the 1450s, are addressed in relation to this section. From a comparison with representations of space from the 1490s, however, one may conclude that, contrary to the thesis proposed by L. Lefaivre, there can have been no direct connection between the author or the artist of the Hypnerotomachia and the architectural theorist. At the same time, the Master of Polifilo appears to belong to the group of painter-architects, along with Francesco di Giorgio Martini, Leonardo, and Raphael.

Abstract Four central concepts of the writings of Francesco di Giorgio (1439 –1501) are discussed: spirienza, dottrina, ingegno, and scienzia. The spirienza can be both »autoptic study« and »experimentation«. It is rooted in Francesco di Giorgio’s original training and has had a lasting influence on his method. The dottrina, »erudition«, serves both Francesco di Giorgio personally and architects in general to move from practitioner to learned architect. The ingegno, talent, is interpreted as the innate creativity and ability to find innovative solutions, it thus plays a central role as a counterpart to the dottrina. Finally, scienzia means a systematic method in the practice of architecture. This gains great importance, especially in the later Treatise, when Francesco di Giorgio emphasizes that in architecture two methods must complement each other: archaeological investigation and the study of (ancient) literature.

Francesco di Giorgio, the Sienese architect and artist, visited Rome c. 1470. By looking at his plan of the ‘porticho del Champitolio’, it is possible to reconstruct not only what Francesco di Giorgio saw on the Monte Tarpeo, but also what Poggio Bracciolini, Flavio Biondo, Pietro del Massaio and others saw there. It was apparently a notable site, an evocative ruin worthy of commentary, artistic representation and imaginative reconstruction. Whatever temple remains continued to be visible, however, these were insufficient to suggest that they were originally part of the temple. By the end of the fifteenth century, the temple had been lost. None the less, Francesco di Giorgio unwittingly documented the last standing columns of the Temple of Jupiter Optimus Maximus.

The characteristic structural forms of large Renaissance churches-domes, drums, pendentives, and barrel vaults-were the products of innovation in theory and practice during the later fifteenth century in Italy that culminated in Bramante's projects for the new Saint Peter's. Significant ideas were contributed by Leon Battista Alberti, Francesco di Giorgio, and Leonardo da Vinci. Francesco di Giorgio's geometrical methods of design for churches as described in his second treatise incorporate a procedure for calculating the thickness of walls bearing vaults. Francesco di Giorgio tested the procedure in his own churches, and it was later used by Bramante.

The purpose of this article is to describe and analyze part of the contents of an Italian manuscript and model book compiled in Venice by Angelo dal Cortivo between 1489 and 1536-the Zichy Codex, now in Szabó Ervin Központi Könyvtár (Erwin Szabo Public Library), Budapest. This analysis demonstrates that the contents of the manuscript represent the earliest version thus far discovered of Francesco di Giorgio Martini's first treatise as found in Saluzzianus 148, Biblioteca Reale, Turin. The article proceeds from an account of the history of the manuscript, including an extensive bibliography and unpublished documents for the life of its compiler, to comparisons of the material in the Zichy Codex with that in Saluzzianus 148 of the Biblioteca Reale, Turin. A list, paragraph by paragraph, of the contents and their parallels in the Turin manuscript is found in Table I. Table II compares the subject matter organization of the two texts. Textual and visual material from the Zichy Codex is discussed, showing that it contains material not found in other Francesco di Giorgio texts, that the understanding of Vitruvius found in the Zichy Codex is poorer than that demonstrated in Saluzzianus 148, and that a purposeful re-organization of the material in the Zichy Codex occurred before the writing of the Turin manuscript. A date in the 1460s is proposed for the Francesco di Giorgio material found in the Zichy Codex.

The role of Francesco di Giorgio (1439-1501) in developing the forms of artillery fortification marking the transition from late medieval defenses to the mature bastioned forts of the 16th century is becoming clearer as additional research has enhanced our knowledge of the chronology of his interventions, the maturation of design elements, and the interlocking personal, institutional, and political factors in his work for the Aragonese Kingdom of Naples. These efforts by Francesco di Giorgio and his associates focused on Naples, Otranto, Gallipoli, Taranto, Manfredonia, Monte Sant'Angelo, Reggio Calabria, Ortona, Matera, and Brindisi. Archival sources, investigation of the sites, and surviving graphic materials contribute substantially to identifying this "school" of military architects and the evolution of design brought about by the technological challenge of gunpowder, firearms, and siege artillery.

Alla luce degli studi di Giorgio Fulco sulla componente prudenziale attiva nell’opera, il saggio riflette sulle ambiguità volutamente ricercate da Pona, con lo scopo di verificare quanto esse siano spie di un discorso allusivo o semplici momenti ludici. In particolare, oggetto di questo lavoro è un’analisi di tre passi programmatici e dallo spiccato valore autocensorio, volta a indagare il loro farsi nella storia redazionale della Lucerna e l’influenza esercitata da due dei principali modelli dell’opera, l’Argenis di John Barclay e la Filena di Nicolò Franco.

Arrivò a Venezia molto giovane per lavorare nella biblioteca di Francesco Barbaro e passò gran parte della sua vita in Italia insegnando. Nell'ultimo libro del suo manuale, Rhetoricorum Libri V, egli, dedicando un'attenzione particolare all'elocutio, elabora un insieme molto interessante di precetti, intrecciando la teoria degli stili con la dottrina ermogenea delle ιδεαι fino ad allora sconosciuta agli autori di manuali latini. Giorgio Trebisonda aveva già trattato brevemente questa questione, una prima volta, in una lettera indirizzata al suo maestro Vittorino da Feltre e concernente esplicitamente i genera dicendi, una seconda volta, in una lunga lettera mandata a Girolamo Bragadin per dargli consigli su come ottenere la suavitas dicendi. Lamentando di non potere fare riferimento a fonti latine e di conseguenza di trovarsi nella duplice difficoltà di dovere tradurre in latino la terminologia specifica della dottrina Ermogenea e di dovere trovare esempi latini per illustrarla, Giorgio Trebisonda cerca di superare questa difficoltà sostituendo a Demostene, l'autore paradigmatico in Ermogene, Cicerone, dalle cui opere attinge la maggior parte del materiale che inserisce nella dottrina Ermogenea con un risultato veramente eccezionale.

En Nápoles la arquitectura de la época de la Corona de Aragón empezó con la reconstrucción del Castelnuovo, impulsada por Alfonso el Magnánimo, y realizada sobre todo por el arquitecto mallorquín Guillem Sagrera. Otros maestros catalanes, activos en la misma fábrica, difundieron elementos arquitectónicos como escaleras, logias, portales y ventanas de formas originales y decoraciones tardogóticas, en los centros urbanos del interior de la Campania y del bajo Lazio (por aquel entonces unido al reino de Nápoles). Se trataba de un gusto que terminaría siendo suplantado por el renacentista toscano que, a partir del último cuarto del siglo XV, se impuso en Nápoles y palautinamente en las regiones del Reino. La iglesia de Santa Caterina a Formello, inspirada en la arquitectura de Francesco di Giorgio y la capilla Caracciolo di Vico anexa a la iglesia de San Giovanni a Carbonara, derivada de una idea tal vez bramantesca, son importantes obras del primer Quinientos, período que en la arquitectura civil fue dominado por la figura del arquitecto Giovanni Mormando. Fue bajo el virreinado de Pedro de Toledo (1532-1553), sin embargo, cuando la ciudad de Nápoles conoció una verdadera renovación: el ensanche de los Quartieri Spagnoli, la calle Toledo, el nuevo Palacio del Virrey, la iglesia y el hospital de San Giacomo degli Spagnoli, el palacio de los Tribunales y el fuerte de Sant’Elmo, obra del arquitecto Pedro Luis Escrivá, que se ocupó también de otras fortificaciones en el Sur de Italia, son las obras impulsadas por este Virrey.

BackgroundApremilast, an inhibitor of the phosphodiesterase 4, is indicated for Psoriatic Arthritis (PsA) treatment. The 3 years retention rate, an outcome indirectly related to efficacy, observed in clinical trials [1] is 55,5%. A single subsequent real world setting study [2] suggested a lower efficacy as it reported that the six months retention rate was about 57%.ObjectivesThe main aim of this retrospective observational study is the assessment of apremilast 3 years retention rate in a real world PsA patients’ cohort. Moreover, the secondary objective is reporting the reasons of apremilast suspension and the most relevant factor related to treatment persistence.MethodsIn thirteen Italian rheumatological referral centers, all PsA consecutive patients who received apremilast were enrolled. Anamnestic data, treatment history and PsA disease activity (DAPSA) at baseline and after 6 and 12 months were recorded. The Kaplan-Meier curve and the Cox analysis computed the apremilast retention rate and treatment persistence-related risk factors. A p-value < 0.05 was considered statistically significant.ResultsThe three-hundred-twenty-four enrolled patients (median age 60 [InterQuartile Range IQR 52-67] yrs; female prevalence 57,0%) median observation period was 17 [IQR 7-36] months (6848 patients-months). The apremilast retention rate at 6, 12 and 36 months was, respectively, 95%, 86% and 66% (Figure 1). The main discontinuation reasons were: primary inefficacy (40% of interruptions), secondary inefficacy (18%) and gastro-intestinal intolerance (17%%). The oligo-articular onset was the only factor associated to apremilast persistence (Hazard ratio 0.57 IQR 0.34-0.96). Sex, age, and sever comorbidities (cancer, chronic infections etc) were not related to treatment discontinuation. The basal DAPSA (24.1, IQR 18.5-32.0) decreased after 6 and 12 months (respectively 14.5, IQR 10.1-22.6 and 10.5, IQR 8,0-15.2). Remission or minimal disease activity (DAPSA < 15) was achieved after 12 months in 38.0% of patients.ConclusionAlmost two third of PsA patients receiving apremilast were still in treatment after 3 years. The study’s data, confirmed its efficacy and safety profile. Apremilast appear a good treatment choice in patients with oligo articular PsA or burdened by severe comorbidities.References[1]Mease et al. Abstract Increasing clinical use of immune checkpoint inhibitors testifies to the importance of modulating the immune TME to obtain meaningful anti-tumor immune responses. Acting only on T lymphocytes may, however, not be sufficient, e.g. in immunologically-cold tumors or due to de novo or acquired resistance. Moreover, immune-related AEs remain hurdles of T cell therapies. To overcome these limitations and to awaken the immune system in an agnostic way against the tumor, we have developed a genetically modified cell-based autologous hematopoietic stem cell platform (Temferon) delivering immunotherapeutic payloads into the TME through Tie-2 expressing monocytes (TEMs), a subset of tumor infiltrating macrophages. TEM-GBM is an ongoing open-label, Phase 1/2a dose-escalating study evaluating the safety & efficacy of Temferon in up to 21 newly diagnosed patients with glioblastoma & unmethylated MGMT promoter assigned to 7 different cohorts (3 pts each) differing by Temferon dose (0.5-4.0x106/kg) and conditioning regimen (BCNU+ or Busulfan+Thiotepa). By Oct 15th, 2021, 15 pts (cohort 1-5) had received escalating doses of Temferon with a median follow up of 267 days (range: 60-749). Rapid engraftment and hematological recovery from nonmyeloablative conditioning occurred in all pts. Temferon-derived differentiated cells, as determined by the presence of vector genomes in the DNA, were found at increasing proportions in PB and BM, reaching up to 30% at 1 month for the highest cohorts tested (2.0x106/kg) and persisting up to 18 months, albeit at lower levels. Despite the significant proportion of engineered cells, only very low median concentrations of IFNα were detected in the plasma (D+30, 5.9; D+90, 8.8pg/mL) and in the cerebrospinal fluid (D+30, 1.5; D+90, 2.4pg/mL), indicating tight regulation of vector expression. SAEs were mostly attributed to conditioning chemotherapy (e.g. infections) or disease progression (e.g. seizures). 1 SUSAR (persistent GGT elevation) has occurred. Median OS is 14 mth from surgery (11 mth post Temferon). Four pts from the low dose cohorts underwent 2nd surgery. These recurrent tumors contained gene-marked cells and expressed IFN-responsive genes, indicative of local IFNα release by TEMs. In 1 pt, a stable lesion (as defined by MRI) had a higher proportion of T cells & TEMs, an increased IFN-response signature and myeloid re-programming revealed by scRNAseq, as compared to a synchronous, progressing tumor. TCR sequencing of blood and tumor samples showed a post-treatment increase in the cumulative frequency of tumor-associated T cell clones identified in 1st and 2nd surgery specimens (up to 4 out of 9 subjects). These results provide initial evidence for on-target activity of Temferon in GBM, to be consolidated with longer follow up in the higher dose cohorts. BackgroundAxial Spondyloarthritis (axSpA) can be distinguished in radiographic axSpA (r-axSpA) and non-radiographic (nr-axSpA). Secukinumab (SEC) is a novel treatment for axSpA, but data from real-life are still missing.ObjectivesTo evaluate, in a multicentric-Italian-cohort of axSpA patients on SEC followed for 24-months: a)the long-term effectiveness and safety of SEC; b)the drug-retention-rate and low-disease-activity (LDA) measured as BASDAI<4/ASDAS<2.1 and very-low-disease-activity (VLDA) measured as BASDAI<2/ASDAS<1.3; c)any differences in outcomes according to: line of biological treatment (naïve/non-naïve), gender (male/female), subtype of axSpA (r-axSpA/nr-axSpA).MethodsConsecutive patients with active axSpA (diagnosis according Assessment of SpondyloArthritis International Society ASAS criteria), who started SEC treatment, were evaluated prospectively. Data on disease characteristics, previous/ongoing treatments and imaging were collected. Disease-activity/functional/clinical scores and biochemical values were recorded at baseline (T0), at 6 (T6), 12 (T12), and 24 (T24) months. Effectiveness was evaluated over-time with descriptive statistics; multivariate Cox and logistic regression models were used to evaluate predictors of drug-discontinuation and LDA at T6. Infections, adverse events were recorded.Results249 patients (47.8% male; median age 51 years) were enrolled; 40.9% had HLA-B27; 53.8% had r-axSpA and 46.2% nr-axSpA. SEC was prescribed in 28.9% naïve and in 71.1% non-naïve patients. SEC effectiveness was shown as an improvement in several outcomes, such as ASDAS [T0=3.5(2.9-4.4) vs. T24=1.9(1.2-2.4);p=0.02] and BASDAI [T0=6.5(5.0-7.5) vs. T24=2.8(1.8-4.0);p=0.03]. After 24-months of treatment, 90.7% of naïve and 75.3% of non-naïve patients achieved LDA (BASDAI<4). At T24 naïve-patients showed better physical-functioning and lower disease-activity than non-naïve (Table 1). Similarly, at T24 we observed better physical functioning and lower inflammatory activity in males vs. females and in nr-axSpA vs. r-axSpA subjects. Retention-rate at T24 was 75% in the whole population, with some difference depending on gender (log-rank 9.319; p=0.002) (Figure 1). V članku je na primeru doslej manj raziskanega mestnega urada prokuratorjev ali oskrbnikov koprske stolnice (Procuratore della Fabrica della Cattedrale di Capodistria) predstavljena vloga koprskega plemstva pri javnem umetnostnem naročništvu. Največji vpliv na izbiro arhitektov in izvajalcev umetnostnih naročil in s tem na končno podobo koprske stolnice v prvi polovici 18. stoletja so imeli prokuratorji Benvenuto markiz Gravisi (1679–1746), Alvise grof Tarsia (1697–1774) in Francesco Barbabianca (1695–1753). Pri izbiri arhitektov, kot sta Giorgio Massari (1687–1766) in Domenico Schiavi (1718–1795), so si pomagali z nasveti in povezavami koprskih izobražencev, kot sta Girolamo markiz Gravisi (1720–1812) in Gian Rinaldo grof Carli (1720–1795), ki so jih ti vzpostavili med študijem na univerzi v Padovi ali preko članstev v akademijah. Tako so se prokuratorji v letih 1748–1749 lahko v zvezi s prezidavo kora koprske stolnice posvetovali tudi s tedaj vodilno avtoriteto na arhitekturnem področju v Beneški republiki, Giovannijem markizom Polenijem (1683–1761), profesorjem eksperimentalne fizike na univerzi v Padovi.

In his treatises dedicated to the Arts, Alberti develops a notion of beauty that distinguishes an innate beauty, pulchritudo, and a form of auxiliary light and a complement to beauty, the ornamentum. The former, intrinsic to the conformation of the buildings, receives the addition of ornaments as garments that clothe the naked body. The notion is guided by the principle of decorum, a parameter which is the foundation of beauty and that regulates the ornaments. Considering the implication of such principles, Alberti prescribes the appropriate ornaments to each genre of building, but he also suggests that the work ought to be constructed naked and only later clothed. Although the author´s asseverations indicate that the state of nudity should be temporary, the appreciation of a kind of beauty inherent to the structure of edifices find peculiar resonances still during the Renaissance, as seen in the buildings projected by Francesco di Giorgio during his stay at the court of Urbino. In the treatises of the Cinquecento, too, one may find notions that lead to Alberti as undoubtedly albertian is the idea of order as adherent ornament which Serlio and Vignola share with their contemporaries and around which much of the architectural debate since the fifteenth century is focused. This work aims to examine such fundamental questions concerning the notions of beauty indebted to Alberti´s precepts.
