Jatropha (Jatropha Curcas L) is a plant that has been used empirically as a fever reducer. Fever is one of the symptoms of the disease which is characterized by a significant increase in body temperature. The novelty of this study was due to the effectiveness of Jatropha Curca L. leaf extract as an antipyretic. The purpose of this study was to determine the dose of antipyretic effectiveness of Jatropha Curcas (Jatropha Curcas L) Leaf Extract in Male Mice (Mus Musculus). Extraction by maceration using 96% ethanol solvent. 15 male mice (Mus Musculus) were divided into 5 groups, namely negative control (Na-cmc 1%), positive control (Paracetamol), and the treatment group given Jatropha leaf extract with doses of 150, 200, 250 mg/kgBW. Peptone 10% 1 ml orally as a fever inducer. And observed at 30, 60, 90 and 120 minutes after treatment. The data obtained were analyzed using one-way ANNOVA followed by the LSD test to determine differences between groups. The results showed that the leaf extract of Jatropha Curcas (Jatropha Curcas L) had an antipyretic effect where the most effective dose was at a dose of 250 mg/kgBW because it has a very stable temperature reducing power and effectiveness is comparable to Paracetamol. The conclusion of this study is that Jatropha Curcas L extract at a dose of 250 mg/kg can be used as an antipyretic.

ABSTRACT
 The study investigating acute toxicity test of Jatropha curcas L. on Nile tilapia seeds (Oreochromis niloticus L.), had been conducted in May 2017, in Balai Riset Perikanan dan Perairan Umum (BRPPU) Mariana, Banyuasin. This study was aimed to determine the concentration of Jatropha curcas seed powder needed to cause 50% death of Nile tilapia seeds (LT50). This acute toxicity test used biological test with exposure time of 96 hours. The concentrations of Jatropha curcas tested were 0 (control), 240, 288, 346, 415, 498, 597, and 716 ppm which were applied for 96 hours. Data were analyzed using Spearman-Karber method with SPSS. Result showed LC50 of 24 hours was 551,271 ppm, LC50 of 48 hours was 466,513 ppm, and LC50 of 96 hours was 393,892 ppm. Meanwhile, LT50 for 597 ppm of Jatropha curcas was 1285, 166 minutes, and LT50 for 761 ppm was 784,121 minutes. The higher the concentration of Jatropha curcas seeds given the fastest time needed to cause the death of Nile tilapia seeds, Jatropha curcas seeds had moderate toxicity.

Introduction:Recurrent aphthous stomatitis (RAS), also known as canker sores, is an inflammation that occurs in the oral mucosa. Frangipani leaf extract and jatropha leaf extract are known to contain saponins, tannins and flavonoids that function as wound healers and antimicrobials. Purpose:Determine whether the administration of frangipani leaf extract (Plumeria acuminata Ait) is more effective in curing recurrent aphthous stomatitis than jatropha leaf extract (Jatropha curcas L). Materials and Methods: The method used is experimental study with a purposive sampling of 30 people. frangipani leaf extract (Plumeria acuminata Ait) and jatropha leaf extract (Jatropha curcas L) were applied to each of 15 samples. Statistical calculations using the Independent T-test. Result and Discussion: The results showed that the mean difference in diameter reduction of recurrent aphthous stomatitis in the first sample group of frangipani leaf extract (Plumeria Acuminata Ait) was 1.13 mm, while in the second sample group Jatropha leaf extract (Jatropha curcas L) was 0.47 mm. From the results of the Independent T-Test test on frangipani leaf extract (Plumeria acuminata Ait) obtained a sig value of 0.002 (p<0.05) and jatropha leaf extract (Jatropha curcas L) of 0.002 (p<0.05), indicating that there is the difference between the use of frangipani leaf extract (Plumeria acuminata Ait) and jatropha leaf extract (Jatropha curcas L) in the treatment of recurrent aphthous stomatitis. Conclusion: frangipani leaf extract (Plumeria acuminata Ait) was more effective in curing recurrent aphthous stomatitis (SAR) than jatropha leaf extract (Jatropha curcas L)

Jatropha curcas L. is an important resource for bioenergy. There are many kinds of energy plants in the world, but they are not utilized sufficiently. Because of the lack of knowledge on resource numbers and distribution of energy plant limited their development. In this paper, we discussed the natural distribution of Jatropha curcas L. in Guangxi province. At the same time, we carried out assessment on the energy plant resource numbers of Jatropha curcas L. in Guangxi. At last, we provided some suggestions for developing of Jatropha curcas L. in China.

ABSTRAKStaphylococcus epidermidis merupakan salah satu bakteri penyebab penyakit kulit seperti jerawat. Getah jarak pagar (Jatropha curcas L.) merupakan bahan alami yang terdapat di batang tanaman yang memiliki efek sebagai antibakteri, karena mengandung senyawa aktif berupa flavonoid, alkaloid, tanin dan saponin. Tujuan penelitian ini adalah untuk mengetahui konsentrasi getah jarak pagar (Jatropha curcas L.) dalam menghambat pertumbuhan bakteri Staphylococcus epidermidis. Penelitian ini menggunakan rancangan acak lengkap (RAL) dan dianalisis dengan uji One Way ANOVA dan uji Duncan dengan 4 konsentrasi 25%, 50%, 75%, dan 100%, kontrol positif yaitu kloramfenikol dan kontrol negatif yaitu akuades steril. Hasil statistik diameter zona hambat adanya perbedaan yang signifikan (p0,05) pada setiap konsentrasi dilihat dari nilai diameter zona hambat. Konsentrasi getah jarak pagar (Jatropha curcas L.) yang mampu menghambat pertumbuhan bakteri Staphylococcus epidermidis terdapat pada konsentrasi 100% (11,58 mm) dibandingkan konsentrasi 75% (9,10 mm), konsentrasi 50% (8,60 mm), dan konsentrasi 25% (7,10 mm), sedangkan untuk kontrol positif (18,07 mm) dan kontrol negatif (0,00 mm). Hal ini dapat disimpulkan bahwa getah jarak pagar (Jatropha curcas L.) berpengaruh pada pertumbuhan bakteri Staphylococcus epidermidis. Kata Kunci : Getah Jarak Pagar (Jatropha curcas L.), Staphylococcus epidermidis, Antibakteri. ABSTRACTStaphylococcus epidermidis on of the bacteria that causes skin diseases such as acne. Jatropha sap (Jatropha curcas L.) is a natural ingredient found in plat stems which has an antibacterial effect because it contains active compounds in the from of flavonoids, alkaloids, tannins, and saponins. The purposes of this study was to determine the concentration of jatropha sap (Jatropha curcas L.) in inhibiting the growth of Staphylococcus epidermidis bacteria. This study used a completely randomized design and analyzed by One Way ANOVA test and Duncan test with 4 concentration of 25%, 50%, 75%, and 100%, positive control namely chloramphenicol and aquades as negative control. The statistical results of the inhibition zone diameter showed a significant difference (p0,05) at each concentration seen from the value of the inhibition zone diameter. The concentration of jatropha sap (Jatropha curcas L.) which can inhibit the growth of Staphylococcus epidermidis bacteria is found at a concentration of 100% (11,5 mm) compared to a concentration of 75% (9,1 mm), a concentration of 50% (8,6 mm), and a concentration of 25% (7,1 mm), positive control (18,0 mm) and negative control (0,0 mm). it can be concluded that the jatropha sap (Jatropha curcas L.) has an effect on the growth of Staphylococcus epidermidis bacteria. Bacillus cereus merupakan bakteri patogen yang dapat mengakibatkan terjadinya diare. Getah Jarak Pagar (Jatropha curcas L.) memiliki kandungan senyawa flavonoid, alkaloid, saponin dan tanin yang bersifat sebagai antibakteri. Penelitian ini untuk mengetahui aktivitas daya hambat antibakteri getah Jarak Pagar (Jatropha curcas L.) konsentrasi 25%, 50%, 75% dan 100% dengan pelarut akuades steril menggunakan metode difusi Kirby-Bauer/Paper disk. Hasil pengujian menunjukkan bahwa getah Jarak Pagar (Jatropha curcas L.) memiliki daya hambat antibakteri terhadap bakteri Bacillus cereus ditandai dengan adanya zona bening yang terbentuk di sekitaran kertas cakram. Berdasarkan hasil uji statistik one way ANAVA dapat dilihat bahwa nilai Ftabel ≤ Fhitung (4,25 ≤ 160,136) hal ini menyatakan adanya pengaruh antibakteri daya hambat getah Jatropha curcas L. terhadap pertumbuhan bakteri Bacillus cereus. Hasil uji Post Hoc Duncan menunjukkan konsentrasi dengan nilai rata rata tertinggi yaitu konsentrasi 100% (11,63 mm) kategori kuat, dilanjutkan dengan konsentraasi 75% (10,83% mm) kategori kuat, 50% (9,78% mm) kategori sedang, dan 25% (8,48% mm) kategori sedang. Dapat disimpulkan bahwa getah Jarak Pagar (Jatropha curcas L.) memiliki daya hambat terhadap pertumbuhan bakteri Bacillus cereus.Kata Kunci: Getah jarak pagar (Jatropha curcas L.), Bacillus cereus, antibakteri.

This study was conducted to assess the phytochemical, antistaphylococcal and antioxidant properties of the leaf and stem bark of Jatropha curcas and Jatropha gossypifolia. Ethanolic extract of the leaf and stem bark of Jatropha curcas and Jatropha gossypifolia was obtained using standard methods. Qualitative and quantitative phytochemical properties of theJatropha plant was assessed using standard methods. Antistaphylococcal and minimum inhibitory concentration of the extracts were assessed against Staphylococcal aureus obtained from different sources using agar diffusion method. Ferrous ion and hydroxyl radical scavenging activity of the extracts were also determined using standard methods. Quantitative phytochemical screening revealed that the distribution of phytochemicals in the two Jatropha species do not follow a regular trend. However, saponin was highest in Jatropha curcas stem bark (35.64 mg/g) while Jatropha gossypifolia stem bark and leaf have the highest flavonoid (31.35 mg/g) and alkaloids (23.20 mg/g) respectively. The antistaphylococcal effect of the combined extract was higher and significantly different (P≤0.05) than when used singly. The highest antistaphylococcal effect (19.83mm) was recorded for the combination of Jatropha curcas and Jatropha gossypifolia leaf (JCL and JGL) against Staphylococcus aureus obtained from blood. Antioxidant assay of extracts revealed a concentration dependent effect. The antioxidant activities of the extracts vary from one extract to the other. The results obtained from this study indicates that bioactive compounds present in Jatropha curcas and Jatropha gossypifolia can be exploited as source of effective antistaphylococcal and antioxidant compounds.

Latar belakang :
 Kandungan ekstrak tumbuhan telah digunakan sudah sejak lama sebagai salah satu metode alternatif/komplementer terutama dalam penyembuhan luka, hanya saja belum terinci sehingga perlu pengujian baik secara in vitro maupun in vivo.
 Tujuan :
 Untuk mengetahui sejauhmana efektifitas tanaman Jatropha Curcas L. terhadap penyembuhan luka dengan pendekatan dari beberapa penelitian yang telah ada sebelumnya.
 Metode :
 Metode dari tinjauan literature ini menggunakan pedoman Prisma dengan penelusuran artikel ilmiah menggunakan data based Pubmed, Google Scholar dan Science Direct. Melakukan pencarian pada Pubmed dengan kata kunci Extract Jatropha Curcas L.  Dari beberapa artikel penelitian yang telah diulas menunjukkan bahwa efektifitas dari ekstrak tanaman jarak pagar (Jatropha Curcas L.) memiliki pengaruh terhadap penyembuhan luka pada hewan coba tikus Wistar.

Agricultural University, Sardarkrushinagar during the year 2002 and 2003. Pollen grains were creamish yellow and sticky. They were Spheroidal or globular in distilled water and acetocarmine with three to four germspores. The mean pollen size in distilled water was 90.5 and 83.5 μm in lower and upper tier of stamen, respectively, while in acetocarmine 82 and 89 μm in upper and lower tier of stamens, respectively. The pollen viability was quite high (95.04 to 98.77%). Maximum pollen germination (35.49%) and pollen tube length (265.19 μ) was recorded in 15 % sucrose solution with 0.005% boric acid.

CoordenaÃÃo de AperfeiÃoamento de Pessoal de NÃvel Superior<br>O pinhÃo manso (Jatropha curcas L.) à considerado uma fonte potencial para a produÃÃo de biocombustÃveis, tendo por base a alta produtividade e qualidade do Ãleo extraÃdo de suas sementes. Entretanto, seu uso à inviabilizado para alimentaÃÃo humana e animal devido à presenÃa de fatores antinuticionais como a curcina e, em especial, os Ãsteres de forbol. A utilizaÃÃo de ferramentas biotecnolÃgicas, como a cultura de tecidos, pode vir a minimizar ou solucionar essas limitaÃÃes. Nesse contexto, o objetivo desse trabalho foi propagar in vitro plantas inteiras via cultivo de Ãpices caulinares obtidos a partir de sementes de pinhÃo manso germinadas in vivo e in vitro; determinar a reprodutibilidade de um protocolo de regeneraÃÃo disponÃvel na literatura e definir, atravÃs de estudos histolÃgicos, a origem da planta regenerada in vitro. Para a propagaÃÃo in vitro utilizou-se Ãpices caulinares, como explantes, que foram colocados em meio MS suplementado com as citocininas BAP (6-benzilaminoprina), KIN (cinetina) e 2-iP (2-isopenteniladenina) na concentraÃÃo de 2,0 mg.L-1. O BAP apresentou os melhores resultados no desenvolvimento dos Ãpices caulinares. Este e a giberelina foram utilizados juntos no mesmo meio ou separados, para a induÃÃo do alongamento dos mesmos. O Ãcido indol-3-butirÃco (AIB) foi usado para o enraizamento. A suplementaÃÃo do meio de cultura com BAP foi eficiente no desenvolvimento dos Ãpices caulinares e o alongamento apresentou Ãxito com o uso do GA3 individualmente. O enraizamento com o AIB foi conseguido. A fim de determinar a reprodutibilidade do protocolo de organogÃnese, cotilÃdones de sementes de pinhÃo manso germinadas in vitro foram colocados em meio MS suplementado com BAP e AIB para a induÃÃo da formaÃÃo de partes aÃreas, estas foram alongadas com BAP e enraizadas com AIB. AlÃm disso, os explantes cotiledonares foram colocados com a face abaxial e adaxial em contato com o meio de cultivo a fim de estabelecer a melhor posiÃÃo do mesmo na regeneraÃÃo in vitro. Plantas inteiras de J. curcas L. foram obtidas a partir da utilizaÃÃo do protocolo testado. A melhor posiÃÃo do segmento cotiledonar foi a face adaxial em contato com o meio, onde 56% dos explantes formaram partes aÃreas. A anÃlise histolÃgica foi feita com a coleta de explantes cotiledonares em intervalos sequenciais de 0, 5, 10, 15 e 25 dias de permanÃncia do meio de regeneraÃÃo. O estudo anatÃmico possibilitou o acompanhamento da organogÃnese do pinhÃo manso, no qual cÃlulas parenquimÃticas prÃximas à regiÃo adaxial do explante cotiledonar se dividiram e formaram as partes aÃreas. Os dados obtidos nesse trabalho mostraram que ambos os protocolos utilizados, a partir de Ãpices caulinares e de explantes cotiledonares, foram eficientes no processo de regeneraÃÃo; as anÃlises histolÃgicas sugerem que a regeneraÃÃo ocorre via organogÃnese direta e possivelmente apresenta origem multicelular.<br>The physic nut (Jatropha curcas L.) is considered a potential source for the biofuel production, because of its high productivity and oil quality extracted from the seeds. However, its use is unfeasible for human and animal food due to antinutritional factors like the curcin protein and specially the secondary metabolites â phorbol esters. The application of biotechnological tools, like tissue culture, can be used to overcome these limitations. In this context, the aim of this work was to propagate whole plants in vitro using shoot cultures obtained from physic nut seeds germinated in vitro and in vivo; to determine the reproducibility of a regeneration protocol available in the literature, and to define through histological studies, the origin of the regenerated plant in vitro. For the in vitro propagation, shoots were used as explants sources, which were placed on MS medium supplemented with the cytokinins BAP (6- benzylaminopurine), KIN (kinetin) e 2-iP (2- isopentenyl adenine), 2,0 mg.L-1. BAP showed the best results in the development of the shoots. This regulator was used alone or in association with gyberelins for the elongation of the shoots. The indole-3-butyric-acid was used for the rooting. The supplementation of the medium culture with BAP was efficient in the development of the shoots and the elongation was superior with the use of GA3 alone. The rooting was achieved with AIB. To determine the reproducibility of an organogenesis protocol, cotyledons from physic nut seeds germinated in vitro were placed on MS medium supplemented with BAP and AIB for the promotion of aerial which were elongated with BAP and rooting with AIB. The cotyledonary explants were placed with the abaxial and adaxial face in contact with the medium to establish the best position of it in the in vitro regeneration. Whole plants of J. curcas L. were obtained using the protocol tested. The best position of the cotyledonar segment was the adaxial face in contact with the medium, where 56% of the explants formed shoots. The histological analysis was made with cotyledonary explants collected in sequential intervals of 0, 5, 10, 15 e 25 days of permanency in the regeneration medium. The anatomical study allowed the accompaniment of the organogenesis in physic nut, where parenchymal cells next to the adaxial region of the cotyledonar explants were divided and formed the shoots. The data obtained from this work showed that both protocols used, from shoots tips and cotyledonar explants, were efficient in the regeneration process; the histological analysis suggests that regeneration occurs via direct organogenesis and possibly presents multicellular origin.

Previous issue date: 2013<br>The physic nut (Jatropha curcas L.) is considered a potential source for the biofuel production, because of its high productivity and oil quality extracted from the seeds. However, its use is unfeasible for human and animal food due to antinutritional factors like the curcin protein and specially the secondary metabolites – phorbol esters. The application of biotechnological tools, like tissue culture, can be used to overcome these limitations. In this context, the aim of this work was to propagate whole plants in vitro using shoot cultures obtained from physic nut seeds germinated in vitro and in vivo; to determine the reproducibility of a regeneration protocol available in the literature, and to define through histological studies, the origin of the regenerated plant in vitro. For the in vitro propagation, shoots were used as explants sources, which were placed on MS medium supplemented with the cytokinins BAP (6- benzylaminopurine), KIN (kinetin) e 2-iP (2- isopentenyl adenine), 2,0 mg.L-1. BAP showed the best results in the development of the shoots. This regulator was used alone or in association with gyberelins for the elongation of the shoots. The indole-3-butyric-acid was used for the rooting. The supplementation of the medium culture with BAP was efficient in the development of the shoots and the elongation was superior with the use of GA3 alone. The rooting was achieved with AIB. To determine the reproducibility of an organogenesis protocol, cotyledons from physic nut seeds germinated in vitro were placed on MS medium supplemented with BAP and AIB for the promotion of aerial which were elongated with BAP and rooting with AIB. The cotyledonary explants were placed with the abaxial and adaxial face in contact with the medium to establish the best position of it in the in vitro regeneration. Whole plants of J. curcas L. were obtained using the protocol tested. The best position of the cotyledonar segment was the adaxial face in contact with the medium, where 56% of the explants formed shoots. The histological analysis was made with cotyledonary explants collected in sequential intervals of 0, 5, 10, 15 e 25 days of permanency in the regeneration medium. The anatomical study allowed the accompaniment of the organogenesis in physic nut, where parenchymal cells next to the adaxial region of the cotyledonar explants were divided and formed the shoots. The data obtained from this work showed that both protocols used, from shoots tips and cotyledonar explants, were efficient in the regeneration process; the histological analysis suggests that regeneration occurs via direct organogenesis and possibly presents multicellular origin.<br>O pinhão manso (Jatropha curcas L.) é considerado uma fonte potencial para a produção de biocombustíveis, tendo por base a alta produtividade e qualidade do óleo extraído de suas sementes. Entretanto, seu uso é inviabilizado para alimentação humana e animal devido à presença de fatores antinuticionais como a curcina e, em especial, os ésteres de forbol. A utilização de ferramentas biotecnológicas, como a cultura de tecidos, pode vir a minimizar ou solucionar essas limitações. Nesse contexto, o objetivo desse trabalho foi propagar in vitro plantas inteiras via cultivo de ápices caulinares obtidos a partir de sementes de pinhão manso germinadas in vivo e in vitro; determinar a reprodutibilidade de um protocolo de regeneração disponível na literatura e definir, através de estudos histológicos, a origem da planta regenerada in vitro. Para a propagação in vitro utilizou-se ápices caulinares, como explantes, que foram colocados em meio MS suplementado com as citocininas BAP (6-benzilaminoprina), KIN (cinetina) e 2-iP (2-isopenteniladenina) na concentração de 2,0 mg.L-1. O BAP apresentou os melhores resultados no desenvolvimento dos ápices caulinares. Este e a giberelina foram utilizados juntos no mesmo meio ou separados, para a indução do alongamento dos mesmos. O ácido indol-3-butiríco (AIB) foi usado para o enraizamento. A suplementação do meio de cultura com BAP foi eficiente no desenvolvimento dos ápices caulinares e o alongamento apresentou êxito com o uso do GA3 individualmente. O enraizamento com o AIB foi conseguido. A fim de determinar a reprodutibilidade do protocolo de organogênese, cotilédones de sementes de pinhão manso germinadas in vitro foram colocados em meio MS suplementado com BAP e AIB para a indução da formação de partes aéreas, estas foram alongadas com BAP e enraizadas com AIB. Além disso, os explantes cotiledonares foram colocados com a face abaxial e adaxial em contato com o meio de cultivo a fim de estabelecer a melhor posição do mesmo na regeneração in vitro. Plantas inteiras de J. curcas L. foram obtidas a partir da utilização do protocolo testado. A melhor posição do segmento cotiledonar foi a face adaxial em contato com o meio, onde 56% dos explantes formaram partes aéreas. A análise histológica foi feita com a coleta de explantes cotiledonares em intervalos sequenciais de 0, 5, 10, 15 e 25 dias de permanência do meio de regeneração. O estudo anatômico possibilitou o acompanhamento da organogênese do pinhão manso, no qual células parenquimáticas próximas à região adaxial do explante cotiledonar se dividiram e formaram as partes aéreas. Os dados obtidos nesse trabalho mostraram que ambos os protocolos utilizados, a partir de ápices caulinares e de explantes cotiledonares, foram eficientes no processo de regeneração; as análises histológicas sugerem que a regeneração ocorre via organogênese direta e possivelmente apresenta origem multicelular.

Se analiza la variabilidad morfológica de Jatropha curcas en 20 poblaciones cultivadas en las regiones de Lambayeque, Piura, Tumbes, San Martín y Ucayali. Se muestrearon 123 individuos de 2-4 cm de dap; se registraron ocho parámetros morfométricos como: altura total de la planta, número de ramas de la planta, número de lóbulos del limbo, longitud del limbo, del pecíolo, del fruto, largo y ancho de la semilla. Los parámetros fueron analizados mediante la prueba de comparaciones de medias ANOVA (p menor 0.05); a los que mostraron diferencias significativas se realiza el análisis de componentes principales.
Existen diferencias significativas de seis parámetros entre las poblaciones; respecto a la altura total de la planta, el mayor valor se registra para la población de La Libertad, y el menor valor para la población de San Isidro; respecto a la cantidad de ramas, el mayor valor se registra para Olmos, y el menor valor para Chazuta; respecto a la longitud del limbo y el pecíolo, el mayor valor se registra para Tambo Grande, y el menor valor para Olmos; respecto a la cantidad de lóbulos, el mayor valor se registra para Tambo Grande, y el menor valor para Curimaná; respecto a la longitud del fruto, el mayor valor se registra para la población de Oidor y el menor valor en la población Yarinacocha. Los resultados del Análisis de Componentes Principales mostraron que el 66.9% de la varianza correspondió a los 2 primeros componentes. No se observaron grupos claramente definidos; sin embargo, las poblaciones de la región costera formaron grupos distintos con respecto a las poblaciones de la región amazónica.
Palabras Claves: piñón blanco, morfometría, análisis de varianza, análisis de componentes principales.<br>--- We analyzed the morphological variation of Jatropha curcas in 20 populations grown in the regions of Lambayeque, Piura, Tumbes, San Martin and Ucayali. A total of 123 individuals with DBH 2-4 cm were sampled, eight morphometric parameters were evaluated such as total plant height, number of branches of the plant, number of lobes of the lamina, length of blade, petiole, fruit and seed. The parameters were analyzed by Paired t test ANOVA (p less than 0.05), those with significant differences were performed principal components analysis.
There are significant differences among the populations in seven parameters; regarding the total plant height, the highest value recorded was in La Libertad, and the lowest value for the population of San Isidro; regarding the number of branches, the highest value recorded was in Olmos, and the lowest value for Chazuta; regarding the length of the blade and petiole, the highest value recorded was found in Tambo Grande, and the lowest value in Olmos, regarding the number of lobes, the highest value recorded was in Tambo Grande, and the lowest value in Curimaná; regarding the length of the fruit, the highest value recorded was found in Oidor and the lowest in the population Yarinacocha. The Principal Component Analysis showed that 70.9% of the variance corresponds to the two first components. Groups clearly defined were not observed; however, the populations of the coastal region formed groups distinct from those of the jungle region.

Já a técnica da micropropagação pode produzir mudas sadias, em escala comercial com um curto período. Com isso objetivou-se com esta pesquisa, desenvolver protocolos para a propagação do pinhão manso via sementes, estaquia e micropropagação, visando à obtenção de mudas de qualidades. O projeto foi dividido em três partes: a primeira parte objetivou-se produzir plantas através de sementes, onde foram realizados testes pré-germinativos em diferentes substratos (comercial, vermiculita, areia); a segunda parte utilizou-se estacas, onde foram testadas partes do ramo (apical, mediana, basal) e diferentes substratos (comercial Bioplant®, vermiculita, areia) em duas épocas, agosto/2011 e março/2012. Na terceira parte foram desenvolvidas técnicas de micropropagação, envolvendo as etapas de assepsia dos explantes in vitro através do controle de contaminação utilizando concentrações de cloro ativo e tempo de imersão, problemas frequentes que inviabilizam a utilização da micropropagação para espécies lenhosas, como o pinhão manso; etapa de desenvolvimento onde estudou concentrações da citocinina 6-Benzilaminopurina (BAP) e dos sais do meio de cultivo Murashige e Skoog (1962) (MS); e a fase de enraizamento in vitro utilizando concentrações do ácido indol butírico (AIB). Concluiu-se que na propagação por sementes o melhor tratamento pré-germinativo foi a escarificação mecânica + imersão em água por 12 horas, e o uso do...<br>The propagation of physic nut can be realized by seed, vegetative by cuttings and micropropagation. When performed by seed, plants are more vigorous but begin production late. Using cuttings have an early production and keeps the characteristics of the parent plant, however, there is less initial vegetative growth. Since the technique of micropropagation can produce healthy seedlings in commercial scala with a short period. Therefore, objective was the research, develop protocols for the propagation of physic nut seeds, cuttings and micropropagation, in order to obtain seedlings qualities. The project was divided into three parts: the first part the objective was to produce plants by seed, where tests were performed pre-germination on different substrates (commercial, vermiculite, sand), the second part was used cuttings, where they were tested parts of the branch (apical, middle, basal) and different substrates (commercial, vermiculite, sand) in two seasons, from August/2011 and March/2012. In the third part have been developed of micropropagation techniques, involving the stages of aseptic explants in vitro through contamination control using ative chloro concentrations and time of immersion, frequent problems that hinder the use of micropropagation for woody species, like physic nut, development studying concentrations of cytokinin 6-benzylaminopurine (BAP) and salts of Murashige e Skoog (1962) (MS) medium and in vitro rooting concentrations of auxin indol butyric acid (IBA). Conselho Nacional de Desenvolvimento Científico e Tecnológico<br>In this work was determined, for the first time, the volatile composition from fresh and dried leaves of six accessions (named PM-2, PM-7, PM-10, PM-11, PM-12 e PM-14) and a cultivar (EPAMIG, named here EMB) of Jatropha curcas L. The
volatile compounds was extracted by hydrodistillation and then analyzed by GCMS. Thus, it was possible to observe that the (Z)-3-hexen-1-ol was the major compound in the fresh leaves (>70% in all accessions). Alcohols also predominated in the dried leaves, mainly (Z)-2-penten-1-ol and (Z)-3-hexen-1-ol, but others constituents were identified in significant proportions included benzenoid compounds such as benzaldehyde. The influence of drying on volatile composition from leaves of J. curcas was evaluated on the PM-2 accession. Besides, this work also describes the development of a High Performance Liquid Chromatograpy (HPLC) method for differentiation of six accessions and a cultivar (EPAMIG) from fresh and dried leaves of J. curcas. The methanol extracts of the leaves were analyzed by HPLC-DAD and after stages of optimizations, we obtained the fingerprint chromatograms. For a comparative analysis of the
accessions and a cultivar, were applied chemometric tools of Principal Component Analysis (PCA) and Hierarchical Cluster Analysis (HCA), which it was possible to discriminate the samples from dried leaves according to their origins.
The methanol extracts of the leaves were submitted to cytotoxicity assay against four tumor cell lines (HCT-8, HL-60, SF-295 and MDA-MB435), presenting IC50 values of 25.13 μg/ml, against HL-60 cell line for EMB sample considered
promising by National Institute of Cancer, USA (NCI). The biological activity of this extracts was evaluated on the development of Spodoptera frugiperda, considered as a primary pest in corn crop. It was observed 60% larval mortality.<br>No presente trabalho foi determinado, pela primeira vez, os compostos voláteis das folhas frescas e secas de Jatropha curcas L. pela técnica de hidrodestilação, para seis diferentes acessos (denominados de PM-2, PM-7, PM-10, PM-11, PM-
12 e PM-14) e uma cultivar (EPAMIG, denominada de EMB). As análises foram realizadas através de CG/EM, obtendo-se o (Z)-3-hexen-1-ol (75,7±18,2%) como composto majoritário nas folhas frescas, com percentuais superiores a 70% entre os acessos. Nas folhas secas obteve-se como compostos majoritários o (Z)-2- penten-1-ol (14,9±11,3%), (Z)-3-hexen-1-ol (9,6±9,1%) e o benzaldeído (3,9±2,7%). Um experimento de secagem foi realizado para o acesso PM-2 com intuito de verificar o efeito da secagem na composição dos voláteis das folhas de J. curcas. Neste trabalho também foi desenvolvido um método por Cromatografia Líquida de Alta Eficiência (CLAE) para diferenciação dos seis acessos e da cultivar (EPAMIG) para folhas frescas e secas de J. curcas. Os extratos
metanólicos das folhas foram analisados por CLAE-DAD e após etapas de otimizações, foram obtidos os cromatogramas fingerprints. Para uma análise comparativa entre os acessos e a cultivar, aplicaram-se ferramentas quimiométricas de Análise dos Componentes Principais (PCA) e Análise de Agrupamentos Hierárquicos (HCA), pelas quais foi possível diferenciar as
amostras das folhas secas de acordo com suas procedências. Os extratos metanólicos também foram submetidos a ensaios de citotoxicidade in vitro para 04 linhagens de células tumorais humanas (HCT-8, HL-60, SF-295 e MDA-MB- 435), obtendo-se CI50 25,13 μg/mL, na linhagem HL-60 para a amostra da cultivar (EPAMIG). Este resultado está dentro do intervalo proposto pelo National Institute of Cancer, USA (NCI) para considerar extratos como promissores. A atividade biológica destes extratos também foi avaliada sobre o desenvolvimento de Spodoptera frugiperda, principal praga do milho, apresentando 60% de mortalidade larval.

Conselho Nacional de Desenvolvimento CientÃfico e TecnolÃgico<br>Physic nut (Jatropha curcas L.) is an important crop due to its ability of storing high content of oil in the seeds, which can serve as raw material for biodiesel production. Because of the presence of toxic constituents like phorbol esters (PEs) and curcins, the seed cake produced as a result of oil extraction cannot be utilize for animal feed. Development of the genotypes better suited for the industrial applications and biodiesel production as well as with lower level of toxic constituents is being hampered by a lack of understanding about the a) proteins related to the biosynthesis and degradation of fatty acids (FAs) and triacylglycerides (TAGs), b) role of proteins deposited during seed development and c) proteins related to the synthesis and storage of toxic compounds during seed development. Agreeing with this, we have performed the anatomical analysis of the developing seeds of J. curcas, followed by the proteome analysis of the endosperm isolated from the seeds of J. curcas at five different developmental stages, which resulted into the identification of the 1517, 1256, 1033, 752 and 307 proteins, from Stage 6, 7, 8, 9 and 10, respectively, summing up to a total of 1760 proteins. Proteins with similar expression pattern were grouped into five different clusters and protein quantification based on spectral counts was determined. Besides identification of the proteins involved in the biosynthesis and degradation of the FAs and TAGs, we also identified a large number of proteins involved in the metabolism of the carbohydrates, which are important for supplying energy and carbon source for the synthesis of TAGs in heterotrophic seeds. Among the members of different classes of seed storage proteins (SSPs), we have identified four SSPs named as nutrients reservoir, which in contrast to the other SSPs showed decreasing deposition pattern during seeds development and revealed to have special role during seed development. In addition, peptidases belong to different mechanistic classes were identified, which have a range of functions, highlighting the role in reserve mobilization during germination. Isoforms of curcin were also identified in this proteome analysis which were absent in our previous proteome analysis of the other tissues from these seeds, suggesting that the deposition of these toxic proteins only occur in the endosperm. Similarly, several enzymes involved in the biosynthesis of diterpenoid precursors were identified in this proteome analysis but, like in our previous proteome analysis of the other tissues from J. curcas seeds,we were unable to identify any terpene synthase/cyclase, enzymes responsible for the synthesis of PEs, which collectively suggesting that the synthesis of PEs may not occur in seeds of this plant. In conclusion, the strategy used here enabled us to provide a first in depth proteome analysis of the endosperm from J. curcas developing seeds, which along with providing information regarding important aspects of the seed development, also set the foundation of a proteomic approach to study biotechnologically important plant species.<br>PinhÃo manso (Jatropha curcas L.) à uma cultura importante devido à sua habilidade em armazenar alto conteÃdo de Ãleo nas sementes, as quais podem servir como matÃria-prima para a produÃÃo de biodiesel. Devido à presenÃa de constituintes tÃxicos como Ãsteres de forbol e curcina, a torta da semente produzida como resultado da extraÃÃo do Ãleo nÃo pode ser utilizada na alimentaÃÃo animal. O desenvolvimento de genÃtipos mais adequados a aplicaÃÃes industriais e à produÃÃo de biodiesel assim como apresentando baixos nÃveis de constituintes tÃxicos està sendo prejudicado pela falta de entendimento sobre a) proteÃnas relacionadas a biossÃntese e degradaÃÃo de Ãcidos graxos e triacilglicerÃis, b) o papel de proteÃnas depositadas durante o desenvolvimento da semente e c) proteÃnas relacionadas à sÃntese e reserva de compostos tÃxicos durante o desenvolvimento da semente. Diante disso, nÃs realizamos uma anÃlise anatÃmica de sementes em desenvolvimento de J. curcas, seguido por uma anÃlise proteÃmica do endosperma isolado de sementes dessa espÃcie em cinco diferentes estÃgios de desenvolvimento, o que resultou na identificaÃÃo de 1517, 1256, 1033, 752 e 307 proteÃnas, dos estÃgios 6, 7, 8, 9 e 10, respectivamente, somando um total de 1760 proteÃnas. ProteÃnas com padrÃo de expressÃo similar foram agrupadas em cinco grupos diferentes e a quantificaÃÃo das proteÃnas baseada na contagem dos espectros foi determinada. AlÃm da identificaÃÃo das proteÃnas envolvidas na biossÃntese e degradaÃÃo de FAs e TAGs, nÃs identificamos um grande nÃmero de proteÃnas envolvidas no metabolismo de carboidratos, as quais sÃo importantes para o fornecimento de energia e fontes de carbono para a sÃntese de TAGs em sementes heterotrÃficas. Entre os membros de diferentes classes de proteÃnas de reservas de sementes (SSPs), nÃs identificamos quatro SSPs denominadas reservatÃrios de sementes, que em contraste as outras SSPs mostraram decrÃscimo no padrÃo de deposiÃÃo e revelaram ter um papel especial durante o desenvolvimento da semente. Em adiÃÃo, peptidases pertencentes a diferentes classes mecanÃsticas foram identificadas destacando o papel da mobilizaÃÃo de reservas durante a germinaÃÃo. Isoformas da curcina ausentes em nossas anÃlises proteÃmicas prÃvias de outros tecidos da semente foram identificadas sugerindo que a deposiÃÃo dessas proteÃnas tÃxicas sà ocorre no endosperma. Similarmente, vÃrias enzimas envolvidas na biosÃntese de precursores de diterpenÃides foram identificadas nessa anÃlise proteÃmica, mas como em nossas prÃvias anÃlises proteÃmicas de outros tecidos de sementes de J. curcas, nÃs nÃo fomos capazes de identificar sintases/ciclases de terpenos, enzimas responsÃveis pela sÃntese de PEs, o que coletivamente sugere que a sÃntese desses compostos pode nÃo ocorrer nas sementes dessa planta. Em conclusÃo, a estratÃgia utilizada nos fornece a primeira anÃlise proteÃmica profunda do endosperma de sementes em desenvolvimento de J. curcas, o que alÃm de fornecer informaÃÃes sobre aspectos importantes do desenvolvimento da semente, tambÃm estabelece a base para uma pesquisa proteÃmica com o objetivo de estudar espÃcies vegetais importantes biotecnologicamente.

When performed by seed, plants are more vigorous but begin production late. Using cuttings have an early production and keeps the characteristics of the parent plant, however, there is less initial vegetative growth. Since the technique of micropropagation can produce healthy seedlings in commercial scala with a short period. Therefore, objective was the research, develop protocols for the propagation of physic nut seeds, cuttings and micropropagation, in order to obtain seedlings qualities. The project was divided into three parts: the first part the objective was to produce plants by seed, where tests were performed pre-germination on different substrates (commercial, vermiculite, sand), the second part was used cuttings, where they were tested parts of the branch (apical, middle, basal) and different substrates (commercial, vermiculite, sand) in two seasons, from August/2011 and March/2012. In the third part have been developed of micropropagation techniques, involving the stages of aseptic explants in vitro through contamination control using ative chloro concentrations and time of immersion, frequent problems that hinder the use of micropropagation for woody species, like physic nut, development studying concentrations of cytokinin 6-benzylaminopurine (BAP) and salts of Murashige e Skoog (1962) (MS) medium and in vitro rooting concentrations of auxin indol butyric acid (IBA). Previous issue date: 2014<br>Physic nut (Jatropha curcas L.) is an important crop due to its ability of storing high content of oil in the seeds, which can serve as raw material for biodiesel production. Because of the presence of toxic constituents like phorbol esters (PEs) and curcins, the seed cake produced as a result of oil extraction cannot be utilize for animal feed. Development of the genotypes better suited for the industrial applications and biodiesel production as well as with lower level of toxic constituents is being hampered by a lack of understanding about the a) proteins related to the biosynthesis and degradation of fatty acids (FAs) and triacylglycerides (TAGs), b) role of proteins deposited during seed development and c) proteins related to the synthesis and storage of toxic compounds during seed development. Agreeing with this, we have performed the anatomical analysis of the developing seeds of J. curcas, followed by the proteome analysis of the endosperm isolated from the seeds of J. curcas at five different developmental stages, which resulted into the identification of the 1517, 1256, 1033, 752 and 307 proteins, from Stage 6, 7, 8, 9 and 10, respectively, summing up to a total of 1760 proteins. Proteins with similar expression pattern were grouped into five different clusters and protein quantification based on spectral counts was determined. Besides identification of the proteins involved in the biosynthesis and degradation of the FAs and TAGs, we also identified a large number of proteins involved in the metabolism of the carbohydrates, which are important for supplying energy and carbon source for the synthesis of TAGs in heterotrophic seeds. Among the members of different classes of seed storage proteins (SSPs), we have identified four SSPs named as nutrients reservoir, which in contrast to the other SSPs showed decreasing deposition pattern during seeds development and revealed to have special role during seed development. In addition, peptidases belong to different mechanistic classes were identified, which have a range of functions, highlighting the role in reserve mobilization during germination. Isoforms of curcin were also identified in this proteome analysis which were absent in our previous proteome analysis of the other tissues from these seeds, suggesting that the deposition of these toxic proteins only occur in the endosperm. Similarly, several enzymes involved in the biosynthesis of diterpenoid precursors were identified in this proteome analysis but, like in our previous proteome analysis of the other tissues from J. curcas seeds,we were unable to identify any terpene synthase/cyclase, enzymes responsible for the synthesis of PEs, which collectively suggesting that the synthesis of PEs may not occur in seeds of this plant. In conclusion, the strategy used here enabled us to provide a first in depth proteome analysis of the endosperm from J. curcas developing seeds, which along with providing information regarding important aspects of the seed development, also set the foundation of a proteomic approach to study biotechnologically important plant species.<br>Pinhão manso (Jatropha curcas L.) é uma cultura importante devido à sua habilidade em armazenar alto conteúdo de óleo nas sementes, as quais podem servir como matéria-prima para a produção de biodiesel. Devido à presença de constituintes tóxicos como ésteres de forbol e curcina, a torta da semente produzida como resultado da extração do óleo não pode ser utilizada na alimentação animal. O desenvolvimento de genótipos mais adequados a aplicações industriais e à produção de biodiesel assim como apresentando baixos níveis de constituintes tóxicos está sendo prejudicado pela falta de entendimento sobre a) proteínas relacionadas a biossíntese e degradação de ácidos graxos e triacilgliceróis, b) o papel de proteínas depositadas durante o desenvolvimento da semente e c) proteínas relacionadas à síntese e reserva de compostos tóxicos durante o desenvolvimento da semente. Diante disso, nós realizamos uma análise anatômica de sementes em desenvolvimento de J. curcas, seguido por uma análise proteômica do endosperma isolado de sementes dessa espécie em cinco diferentes estágios de desenvolvimento, o que resultou na identificação de 1517, 1256, 1033, 752 e 307 proteínas, dos estágios 6, 7, 8, 9 e 10, respectivamente, somando um total de 1760 proteínas. Proteínas com padrão de expressão similar foram agrupadas em cinco grupos diferentes e a quantificação das proteínas baseada na contagem dos espectros foi determinada. Além da identificação das proteínas envolvidas na biossíntese e degradação de FAs e TAGs, nós identificamos um grande número de proteínas envolvidas no metabolismo de carboidratos, as quais são importantes para o fornecimento de energia e fontes de carbono para a síntese de TAGs em sementes heterotróficas. Entre os membros de diferentes classes de proteínas de reservas de sementes (SSPs), nós identificamos quatro SSPs denominadas reservatórios de sementes, que em contraste as outras SSPs mostraram decréscimo no padrão de deposição e revelaram ter um papel especial durante o desenvolvimento da semente. Em adição, peptidases pertencentes a diferentes classes mecanísticas foram identificadas destacando o papel da mobilização de reservas durante a germinação. Isoformas da curcina ausentes em nossas análises proteômicas prévias de outros tecidos da semente foram identificadas sugerindo que a deposição dessas proteínas tóxicas só ocorre no endosperma. Similarmente, várias enzimas envolvidas na biosíntese de precursores de diterpenóides foram identificadas nessa análise proteômica, mas como em nossas prévias análises proteômicas de outros tecidos de sementes de J. curcas, nós não fomos capazes de identificar sintases/ciclases de terpenos, enzimas responsáveis pela síntese de PEs, o que coletivamente sugere que a síntese desses compostos pode não ocorrer nas sementes dessa planta. Em conclusão, a estratégia utilizada nos fornece a primeira análise proteômica profunda do endosperma de sementes em desenvolvimento de J. curcas, o que além de fornecer informações sobre aspectos importantes do desenvolvimento da semente, também estabelece a base para uma pesquisa proteômica com o objetivo de estudar espécies vegetais importantes biotecnologicamente.

Por ser um cultivo perene ainda incipiente, a avaliação da produtividade é importante para o seu melhoramento genético. As estimativas de parâmetros genéticos são importantes para a caracterização da estrutura genética da população, para o entendimento genético dos caracteres e para a predição dos ganhos com seleção. O presente estudo avaliou o potencial de produtividade de 78 acessos (assumidos como famílias de meios irmãos) de J. curcas, com 48 e 60 meses, em dois anos de produção (2012/13 e 2013/14). O ensaio foi instalado em Araponga (latitude 20o 39’ S, longitude 42o 32’ W e altitude 823 m), MG, em quatro experimentos, todos em blocos casualizados com quatro repetições e parcelas de quatro plantas, utilizando o espaçamento 2x2m, e duas testemunhas comuns aos experimentos, totalizando 1376 plantas. Os caracteres avaliados foram produtividade de frutos (PF), de grãos (PG) e de óleo (PO), em kg.ha-1, teor de óleo da semente (TO), em %, peso de 50 frutos (P50F) e 100 sementes (P100S), em g, comprimento (CF) e largura (LF) do fruto, e comprimento (CS) e largura (LS) da semente, em mm. Processou-se a estimação de parâmetros genéticos e a predição dos ganhos genéticos com a seleção de indivíduos e clones. Para as produtividades e teor de óleo, os resultados evidenciaram a existência de variabilidade genética entre as famílias e herdabilidades individuais de moderada a média magnitude, entre 0,19 e 0,51, adequadas para a seleção da principal característica a ser explorada, PO (0,50). Com base nos valores de repetibilidade ( > 0,60) dos caracteres PG e PO, duas medições ou colheitas consecutivas são suficientes, uma vez que as famílias mantiveram seus desempenhos relativos ao longo dos dois anos. A seleção, pelo valor genotípico, das 20 plantas superiores quanto à produtividade de óleo proporcionou ganho médio de 162,7%, superior à seleção de plantas pelos valores genéticos aditivos (157%), indicando maiores possibilidades de sucesso com a implantação de plantios clonais.<br>Jatropha curcas L. is one of the most promising oilseed for the production of biodiesel and bio-kerosene. For being an incipient perennial crop, evaluating productivity is important for your breeding. Estimates of genetic parameters are important for the characterization of the genetic structure of the population to the genetic understanding of the characters and for the prediction of gains from selection. This study evaluated the 78 accessions productivity potential (assumed to families half-sib) of J. curcas, with 48 and 60 months, two years of production (2012/13 and 2013/14). The experiment was conducted in Araponga (latitude 20o 39 'S, longitude 42o 32' W and altitude 823 m), MG, in four trials, all randomized blocks with four replicates and four plants per plot, using the spacing 2x2m, and two common checks to the experiments, totaling 1376 plants. The characters evaluated were fruit yield (PF), grains (PG) and oil (PO), in kg ha-1, seed oil content (TO), in%, weight of 50 fruits (P50F) and 100 seeds (P100S) in g, length (FL) and width (LF) of the fruit, and length (CS) and width (LS) seed, in mm. Was estimated genetic parameters and prediction of genetic gain with selection of individuals and clones. For the yield and oil content, the results showed the existence of genetic variability between individual families and heritability of moderate to medium magnitude, between 0.19 and 0.51, suitable for selecting the main feature to be explored, PO ( 0.50). Based on the repeatability values (> 0.60) for the PG and PO characteristics , two consecutive measurements or harvests are sufficient, since the families retained their relative performances throughout the two years. The selection by the genotypic value, the top 20 plants on the oil yield provided average gain of 162.7%, higher than the plant selection by additive genetic values (157%), indicating greater chances of success with the implementation of clonal plantations .

Fundamentado nesse fato, muitos países, incluindo o Brasil, aumentaram os esforços para financiar pesquisas com espécies
vegetais produtoras de biocombustíveis. Dentre essas espécies, Jatropha curcas L. destaca-se pelo alto potencial para produção de óleo. Visando à propagação clonal e massal de acessos elite de J. curcas, técnicas de cultura de tecidos têm sido aplicadas. Entretanto, diferentes autores frisaram a necessidade de ampliação de estudos envolvendo ferramentas da cultura de tecidos vegetais, dentre elas a poliploidização in vitro. A indução de poliploides in vitro é uma importante ferramenta no melhoramento genético de plantas, por possibilitar a obtenção de maior variabilidade de materiais com possíveis incrementos de características agronômicas desejáveis. Nesse sentindo, um dos objetivos deste trabalho foi estabelecer um protocolo de indução in vitro de plântulas poliploides da variedade / Gonçalo/ (2C = 0.85 pg, 2n = 2x = 22 cromossomos). Os resultados de citometria de fluxo evidenciaram que tetraploides e mixoploides foram obtidos a partir de ápices caulinares tratados com diferentes concentrações de colchicina e tempos de exposição. Fundamentado em três parâmetros (número de plântulas sobreviventes, tetraploides e mixoploides), o tratamento envolvendo 0,5 mM de colchicina a um pulso de 96 horas foi considerado o mais adequado. Com base nesse resultado, um
segundo experimento de poliploidização foi realizado somente com a dosagem de 0,5 mM com pulsos de 96, 120, 144 e 168 horas. Os resultados confirmaram que a concentração de 0,5 mM por 96 horas é ideal para indução de tetraploides de J.
curcas / Gonçalo/ . Plântulas mixoploides também foram encontradas nos tratamentos com0,5 mM de colchicina aos pulsos de 96, 120 e 144 horas. Visto a relevância da
propagação clonal e massal de acessos elite de J. curcas, inclusive das plântulas poliploides obtidas no primeiro estudo, o presente trabalho também teve como objetivo induzir a calogênese in vitro a partir de explantes foliares. Os resultados
mostraram que a combinação ácido 2,4-diclorofenoxiacético (2,4-D)/cinetina (KN) promoveu maior número de explantes com calos friáveis quando comparado à combinação ácido indol-3-butírico (AIB)/KN. Dessa forma, o procedimento
envolvendo a combinação auxina/citocinina é fundamental para gerar calos friáveis, os quais representam a base para estabelecimento de um sistema embriogênico in vitro de acessos elite. Pela primeira vez, um sistema de cultura de tecidos foi adaptado para indução, propagação e recuperação de tetraploides e mixoploides de J. curcas / Gonçalo/ . ampliação de bancos de germoplasma in vitro da espécie<br>The use of bioenergy has been suggested as a strategy for reducing the utilization of fossil fuels. Thereafter, many countries, including Brazil, have increased efforts to fund the research of plant species for biofuel production. Among these species, Jatropha curcas L. stands out mainly due to its high potential for oil production. Thus, tissue culture techniques have been applied aiming at the mass and clonal propagation of J. curcas elite lines. However, different authors have emphasized the necessity of expanding studies that involve plant tissue culture tools such as in vitro polyploidization. In this sense, the present study sought to establish a protocol for in vitro induction of polyploid seedlings from shoot tips of the variety Gonzalo (2C = 0.85 pg, 2n = 2x = 22 chromosomes). Flow cytometry revealed mixoploids and tetraploids obtained from shoot tips treated with different colchicine concentrations and exposure times. Based on three criteria (survival rate of the explants, and numbers of tetraploid and mixoploid plantlets), the treatment with 0.5 mM colchicine in a 96 hour pulse was statistically considered the most appropriate. Based on these data, a second polyploidization experiment was carried out, using a dosage of 0.5 mM colchicine in pulses of 96, 120, 144 and 168 hours. The results confirmed that the concentration of 0.5 mM for 96 hours was the ideal treatment for J. curcas Gonçalo tetraploid induction. Mixoploid seedlings were also detected in the treatments with 0.5 mM colchicine in pulses of 96, 120 and 144 hours. Considering the relevance of mass and clonal multiplication of elite J. curcas accessions, and the polyploid seedlings obtained in the first investigation, this study further aimed to establish in vitro callogenesis from leaf explants. The results showed that the combination of 2,4-dichlorophenoxyacetic acid (2,4-D) and kinetin (Kn) provided
greater number of explants with friable calli in relation to the combination indol-3-butyric acid (IBA) and Kn. Thus, 2,4-D/Kn was the treatment of choice to generate friable calli, which are essential for somatic embryogenesis. Ultimately, a tissue culture system was adapted for induction, propagation and recovery of in vitro tetraploids from the J. curcas variety Gonçalo, as well as other inbred lines. Besides providing a new protocol for polyploidy generation, this study also contributed to the germplasm of J. curcas and the achievement of genetic variability through obtainment of tetraploid and mixoploid seedlings

O maracujazeiro-amarelo é considerado uma espécie sensível aos sais (Ayers & Westcot, 1999). Assim o declínio produtivo da cultura deve ocorrer quando a condutividade elétrica do ambiente radicular das plantas atingisse valores superiores a 1,3 dS m-1. Entretanto, algumas pesquisas têm demonstrado que, apesar da salinidade provocar reflexos negativos no crescimento, na capacidade produtiva da cultura e na qualidade dos frutos (Soares et al., 2008; Cavalcante et al., 2009), a utilização de corretivos pode amenizar os efeitos da salinidade, resultando em maior desenvolvimento das mudas e produção de frutos pelas plantas (Souza et al., 2008). Alguns trabalhos mostraram que a utilização de rejeitos de mineração como constituinte de substratos para emergência e produção de mudas de várias espécies vegetais, como Carica papaya L. (Alencar, 2004); Moringa oleifera Lam. (Alves, 2005); Jatropha curcas L. (Trajano et al., 2010); Cnidoscolus quercifolius Pohl. (Farias Júnior, 2011) e Mimosa caesalpiniifolia Benth. (Rodrigues, 2011), pode ser uma alternativa promissora para diminuição dos impactos ambientais destes rejeitos. Contudo, estudos avaliando o efeito destes rejeitos em solos salinizados são quase inexistentes. O presente trabalho teve o objetivo de avaliar o efeito do gesso agrícola e doses de rejeitos de vermiculita e de caulim, sobre os atributos químicos de um solo salino-sódico e o crescimento do maracujazeiro amarelo.

A degradação ambiental ocorre principalmente nas regiões áridas, semiáridas e subúmidas secas do planeta, e resulta de fatores climáticos e antrópicos tais como as atividades pecuárias, de agricultura e de mineração (LIMA, 2004). Especificamente na região semiárida do nordeste do Brasil, que se estende por mais de 980 km2 distribuídos em todos os Estados nordestinos (exceto o Maranhão) e no norte de Minas Gerais, o desequilíbrio ambiental atinge mais de 20 milhões de hectares, o que equivale a mais de 12% da região Nordeste e quase 22% da região semiárida nordestina (MEDEIROS et al., 2012; SILVA et al., 2004). Estes autores informam que este quadro é mais crítico no Ceará e na Paraíba, onde a degradação ambiental é observada em mais da metade dos seus territórios. Esta degradação tem um forte componente humano, pois 21,3 milhões de habitantes habitam essa região, além dos rebanhos constituídos de 23,9 milhões de bovinos, 8,8 milhões de caprinos e 8,0 milhões de ovinos que se alimentam da vegetação nativa (IBGE, 2007; MEDEIROS et al., 2012), sendo por isso considerada a região semiárida mais populosa e com maior grau de antropismo do planeta (DRUMOND et al., 2000). As atividades humanas, tais como a pecuária e a mineração praticadas de maneira inadequada, podem desencadear processos de degradação ambiental, caracterizada pelo empobrecimento da flora e da fauna, erosão do solo e deterioração da qualidade da água dos rios e dos reservatórios (AZEVÊDO, 2011). Especificamente no solo, a densidade e a porosidade da camada superficial são os atributos mais prejudicados e precisam ser melhorados para favorecer a regeneração da vegetação, pois desequilíbrios nesses atributos prejudicam o desenvolvimento do sistema radicular e no estabelecimento e crescimento das plantas (NUNES, 2012). A mineração é uma atividade de forte impacto ambiental, pois implica geralmente na remoção da camada superficial do solo na área da jazida e na deposição superficial de rejeitos (BARRETO, 2001). A exploração da vermiculita, um produto utilizado na construção civil, melhoria das propriedades físicas de solos agrícolas, produção de cerâmica e remediação de solos contaminados por petróleo (NASCIMENTO, 2008). Há várias jazidas em exploração na região Nordeste do Brasil, uma delas no município de Santa Luzia-PB. Em visita à Mineradora Pedra Lavrada, que extrai a vermiculita no município de Santa Luzia, constatou-se a presença de crateras resultantes da extração do minério e a deposição de rejeitos no entorno da unidade de beneficiamento. Estes rejeitos não têm valor comercial que justifique o seu processamento. Este material de menor valor, doravante denominado de coproduto, afeta diretamente o ambiente pela ocupação de áreas de Caatinga, causando poluição visual e soterrando a vegetação nativa. Além disto, provoca efeitos indiretos no ambiente quando é carreado pelo vento e pelas águas pluviais, poluindo a água e assoreando rios e reservatórios. Este material de menor valor comercial é gerado quando a rocha é moída, e a vermiculita é separada de acordo com a sua granulometria. Dos cinco co-produtos gerados, apenas os de menor granulometria (poeira fina e ultrafina) são compatíveis para a produção de mudas, pois as pedras e pedaços menos friáveis do minério não apresentam, por motivos óbvios, potencial para a nutrição de plantas. Estes dois co-produtos provêm de um material friável do grupo das micas que formam silicato hidratado de magnésio, ferro e alumínio e constitui uma fonte de Ca, K e Mg para as plantas (NASCIMENTO, 2008). A utilização destes co-produtos se mostrou adequada para compor até 50% do substrato de produção de mudas de maracujá Passiflora edulis Sims. f. flavicarpa Deg.) (LEITE, 2012) e pinhão manso (Jatropha curcas L.) (TRAJANO, 2010), propiciando ao substrato boas características de porosidade e capacidade de retenção de umidade, o que sugere a possibilidade de seu uso na produção de mudas de outras espécies. As mudas produzidas com estes co-produtos poderiam ser direcionadas para plantios em geral ou para a revegetação de áreas degradadas pela mineração, num círculo virtuoso em que a degradação ambiental provocada pela mineração em si e pela deposição dos co-produtos no ambiente seria parcialmente revertida pela utilização dos co-produtos gerados.
