Texte intégralThis research is an investigative study of a descriptive, comparative and interpretive nature under the auspices of the Historical Linguistics. This study presents the analysis of the vocalization of postvocalic /l/ and /r/ in speech community of Jaraguá, located in the midwest of Goiás, Brazil. In the process of vocalization, the /l/ is replaced by [y] and /r/, whose analyzed variant is [ö], with [y] and [w], providing falling diphthongs (vowel + semivowel). The postvocalic vocalization occurs in CVC and CV syllables within words, for example, salgada > saygada, alma > ayma, garfo > gayfu ~ gawfu; at the end of words the process of erasure or resyllabication predominated, as in enxoval > inxovali ~ inxová, cobertor > cubeytori ~ cubeytô. The sample group consists of 17 speakers (male and female) above 65 years old, born near the town, or people that have lived for more than 50 years in the place, with little or no level of literacy. We applied the technique of data collection based on the methodological assumptions of Ethnography, adopting a semi-structured interview. The themes of the interviews were directed to narratives and dialogues about family, childhood, migration, religious, cultural and professional activities, and especially about history and the beginning of the locality. The focus is primarily phonological, so it is used the terminology of this area of Linguistics. The theoretical section presents the main theories and methods of Historical Linguistics from the perspectives of Paixão de Sousa (2005-2009), Silva (1973), Coutinho (1975), Campbell (2004), Maurer Jr. (1951), Faraco (2005), Callou e Leite (1993) and Ladefoged e Maddieson (1996). The discussion and presentation of the Brazilian Portuguese Language and its structural system is based on theorists such as Ilari (1990), Ilari e Basso (2006), Mattos e Silva (2004), Camara Jr. ([1970] 2008a - [1953] 2008b), Amaral (1976), Jakobson (1967) and others. Theoretically, the process of postvocalic vocalization is shown and discussed on the perspectives by Camara Jr. ([1953] 2008a - [1970] 2008b), Coutinho (1975), Elia (1979), Silva (2008), Melo (1981), Moura (1993), Vieira (1983), Silva Neto (1988) among others. Given these theoretical and methodological assumptions, it is considered, in the analysis of data, the synchronic and diachronic criteria of postvocalic vocalization, in the intralinguistic perspective. In a diachronic point of view, we observe the evolution from Latin to Portuguese, and from the synchronic point of view, the occurrences of the speech community of Jaraguá, comparing them to recent studies from other regions of the country.
A presente pesquisa caracteriza-se por ser um estudo investigatório de natureza descritiva, comparativa e interpretativa sob os auspícios da Linguística Histórica. Este estudo apresenta a análise da vocalização dos fonemas /l/ e /r/ pós-vocálicos na comunidade de fala de Jaraguá, localizada na região Centro-Oeste do Estado de Goiás, Brasil. Nesse processo de vocalização, o /l/ é substituído por [y] e o /r/, cuja variante analisada é [ö], por [y] e [w], criando ditongos decrescentes (vogal + semivogal). A vocalização ocorre em sílabas CVC e VC, no interior de palavras em posição pós-vocálica, por exemplo, salgada > saygada, alma > ayma, garfo > gayfu ~ gawfu; em final de palavras predominam os processos de apagamento ou de ressilabação, como em enxoval > inxovali ~ inxová, cobertor > cubeytori ~ cubeytô. O grupo de amostra é constituído por 17 falantes (homens e mulheres), acima de 65 anos de idade, nascidos nas proximidades do município, ou com mais de 50 anos de residência na localidade, com pouco ou nenhum nível de letramento. Empregou-se a técnica de coleta de dados baseada nos pressupostos metodológicos da Etnografia, adotando a entrevista semi-estruturada. Os temas das entrevistas foram direcionados às narrativas e diálogos sobre família, infância, migrações, atividades religiosas, culturais, profissionais e, principalmente, sobre a história e o surgimento do lugar. O foco deste trabalho é fundamentalmente fonológico, portanto são utilizadas as terminologias dessa área da Linguística. A fundamentação teórica apresenta as principais teorias e métodos da Linguística Histórica a partir das perspectivas de Paixão de Sousa (2005-2009), Silva (1973), Coutinho (1975), Campbell (2004), Maurer Jr. (1951), Faraco (2005), Callou e Leite (1993) e Ladefoged e Maddieson (1996). A discussão e apresentação da Língua Portuguesa do Brasil e do seu sistema estrutural encontram-se baseadas em teóricos como Ilari (1990), Ilari e Basso (2006), Mattos e Silva (2004), Camara Jr. ([1970] 2008a [1953] 2008b), Amaral (1976), Jakobson (1967) e outros. Teoricamente, o processo da vocalização pós-vocálica é mostrado e discutido nas perspectivas de Camara Jr. ([1953] 2008a [1970] 2008b), Coutinho (1975), Elia (1979), Silva (2008), Melo (1981), Moura (1993), Vieira (1983), Silva Neto (1988) dentre outros. Diante desses postulados teóricos e metodológicos, são considerados, na análise dos dados, os critérios diacrônicos e sincrônicos da vocalização pós-vocálica, na perspectiva intralinguística. Do ponto de vista diacrônico são observadas as evoluções do Latim ao Português; e do ponto de vista sincrônico, as ocorrências da fala da comunidade de Jaraguá, comparando-as com estudos recentes de outras regiões do país.

Texte intégralThe aim of presented doctoral thesis was investigation of phytochemical composition and biological activity of water and methanol extracts of fresh and air- dried rose hips, as well as purée and jam made according to traditional recipe of hips of six wild growing Rosa L. species: R. canina, R. dumalis, R. dumetorum, R. tomentosa, R. arvensis, and R. sempervirens. Examination of phytochemical composition included LC-MS/MS analysis of 64 selected phenolic compounds, quinic acid (organic acid) and three triterpenoids. Also, total phenolic and flavonoid contents, as well as vitamin C contents were determined spectrophotometrically. Biological activity evaluation of extracts of six Rosa species included in vitro investigation of antioxidant, anti- inflammatory, anti-acetylcholinesterase and cytotoxic activity. According to obtained results, fresh and air-dried rose hips, as well as their preserves present valuable source of vitamin C and phenolic compounds, especially ellagic acid, which was the most abundant examined phenolic compound. Furthermore, high concentration of ursolic acid was detected in some Rosa tomentosa and R. rumetorum extracts, while notable concentration of quinic acid was present in all examined extracts. Extracts of all examined species, apart from Rosa arvensis, showed considerable antioxidant activity in terms of radical scavenging ability, reduction potential and inhibition of lipid peroxidation. Moreover, extracts exhibited moderate anti- inflammatory activity by means of inhibition of the main arachidonic acid metabolites formed incyclooxygenase-1 (12-HHT, TXB2, PGE 2 ) and 12-lipoxygenase (12-HETE) pathway, especially prostaglandin E2 . Also, investigation of anti- cetylcholinesterase activity revealed moderate activity of extracts of all examined species. In addition, predominantly water extracts of fresh andair-dried rose hips, as well as purée of R. canina, R. tomentosa and R. sempervirens showed inhibitory activity toward HeLa, while R.sempervirens extracts also inhibited HT-29 cell growth. Presented results indicate significant potential of examined rose hips and their preserves for use as nutraceuticals and functional food.

Texte intégralEn el presente trabajo de tesis, se realiza un planeamiento estratégico para la Constructora San Jorge E. I. R. L., en el que se analiza la situación actual de la empresa, y se plantea un horizonte de tiempo de 10 años para su ejecución, implementación y puesta en marcha a fin de llegar a una situación futura deseada. Este plan estratégico se ha iniciado con el establecimiento de la visión, misión, valores y código de ética de la constructora. Luego se ha realizado el análisis del entorno para Constructora San Jorge E. I. R. L., del cual se obtuvieron siete oportunidades y ocho amenazas. Del análisis del sector construcción y de la competencia, se obtuvieron diez factores claves de éxito. Asimismo, del análisis interno de la constructora se obtuvieron ocho fortalezas y seis debilidades. Estos tres análisis han permitido conocer y comprender el entorno de la Constructora San Jorge E. I. R. L., y plantear nueve estrategias: (a) Establecer alianzas estratégicas con los actuales y nuevos proveedores de insumos y materiales de construcción, (b) Desarrollar relaciones orientadas a la obtención de mejores condiciones financieras con los bancos privados, (c) Desarrollar el talento profesional del personal, (d) Ejecutar proyectos de inversión privada, (e) Penetrar en mercados nacionales, (f) Desarrollar un programa de optimización de gastos e inversión por obra, (g) Desarrollar alianzas estratégicas con empresas extranjeras del mismo rubro para tener mayor competitividad en el país, (h) Implementar certificaciones de calidad para poder competir con empresas extranjeras, y (i) Implementar sistemas de contingencia frente a la problemática social del país. Estas estrategias permitirán llegar a la visión de la Constructora San Jorge E. I. R. L. a través de seis objetivos de largo plazo: (a) al 2025, incrementar las ventas a S/. 30‟000,000 de soles. En el año 2014, las ventas totalizaron S/. 15‟000,000 de soles y el incremento promedio de ventas de los tres últimos años fue de 73%; (b) al 2025, obtener una utilidad sobre ventas de 10%. En el año 2014, la utilidad sobre ventas fue de 3.4%; (c) al 2025, formar parte del ranking de las 20 mayores empresas constructoras del Perú, según América Economía; (d) al 2025, contar con una certificación ISO 9001 y/o OHSAS 18001; (e) al 2025, desarrollar cinco acciones en beneficio de la comunidad vinculada, promoviendo el cuidado del medio ambiente, dado que a la fecha la constructora no cuenta con un plan de responsabilidad social; y; (f) al 2025, desarrollar y poner en marcha al menos tres nuevas mejoras tecnológicas que enriquezcan la eficiencia de los procesos y de los recursos. Actualmente no se cuentan con mejoras tecnológicas debido a la carencia de un área de Innovación & Desarrollo. También se han establecido objetivos de corto plazo, los cuales permitirán realizar la revisión, evaluación, y control de los resultados. El entorno es favorable para Constructora San Jorge E. I. R. L.; sin embargo, en su organización interna resulta necesario contar con áreas importantes, como Recursos Humanos, Marketing, Finanzas, e Investigación y Desarrollo; además de contar con un sistema informático integral que permita mejorar su gestión. Para los objetivos de corto plazo, se asignaron los recursos necesarios para el cumplimiento de los mismos, así como también las políticas para cada una de las estrategias retenidas. Se plantea también una nueva estructura organizacional con la inclusión de áreas necesarias para la mejora de la constructora. Finalmente, en el proceso estratégico, la etapa de evaluación y control se realizará a través del Tablero de Control Balanceado (Balanced Scorecard)
In this thesis work, we make a strategic planning for Constructora San Jorge E.I.R.L., in which the current situation of the company is analyzed and we propond a horizon of time of ten years for the execution, implementation and setting up in order to reach a desired future situation. This strategic plan has begun with the establishment of vision, mission, values and code of ethics of the enterprise. After we have made the analysis the of environment of the Constructora San Jorge E.I.R.L., in we obtained which seven oportunities and eight threats. Of the Analysis of the construction sector and competition, we obtained ten key success factors. Also, of the internal analysis of the construction we obtained eight strengths and six weaknesses. These three tests have allowed us to know and understand the environment of Constructora San Jorge E.I.R.L., and raise nine strategies: (a) Establish strategic alliances with current and new suppliers of equipment and materials, (b) Develop oriented relationships at achieving better conditions financing with private banks, (c) develop the professional talent of staff, (d) Run private investment projects, (e) Penetrating national markets, (f) Develop an optimization program expenditures an investment by work, (g) Develop strategic alliances with foreign companies in the same industry to be more competitive in the country, (h) Implement quality certifications to compete with foreign companies, and (i) Implement contingency systems taked social problems of the country. These strategies will allowed reach the vision of Constructora San Jorge E.I.R.L. through six long-term objectives: (a) By 2025, increase sales to S/. 30‟000,000 soles. In 2014, sales totaled S/. 15‟000,000 soles and the average increase in sales of the last three years was 73%, (b) In 2025, get a return on sales of 10%. In 2014, the return on sales was 3.4%, (c) By 2025, part of the ranking of the 20 largest construction companies in Peru, according to America Economic (d) By 2025, have an ISO 9001 certification and / or OHSAS 18001, (e) By 2025, develop five actions to benefit the associated community, promoting environmental stewardship, since the date the builder does not have a social responsibility plan, (f) By 2025, develop and launch at least three new technological improvements that enhance the efficiency of processes and resources. Currently they not have technological improvements due to the lack of an area of Innovation & Development. We have also short-term objectives, which will allow us to make the review, evaluation, and control of the results. The environment is favorable for Constructora San Jorge E.I.R.L.; However, in its internal organization is important canting with areas such as Human Resources, Marketing, Finance, and Research and Development; in addition to a integral computerized system to improve its management. For short-term goals the resources necessary to fulfill them were assigned, as well as policies for each one of the retained strategies. A new organizational structure also arises with the inclusion of necessary areas for improvement of the enterprise. Finally, in the strategic process, the evaluation stage and control will be make through the Balanced Scorecard (Balanced Scorecard)

The purpose of this work is to carry out the phytochemical study and evaluate biological activities of the essential oil and extracts of the Pimenta pseudocaryophyllus (Gomes) L.R. Landrum (Myrtaceae) leaves. Chapter 1 provides a review of the main botanical aspects of the genus Pimenta, its chemical composition and pharmacological potential. Chapter 2 describes the occurrence of infraspecific variability in P. pseudocaryophyllus. For this the essential oils from 12 specimens of P. pseudocaryophyllus that occur naturally in three different locations of the Brazilian Cerrado underwent a qualitative and quantitative analysis by gas chromatography-mass spectrometry (GC/MS). The multivariate statistical analysis identified three chemotypes of P. pseudocaryophyllus, characterized by the predominance of citral, (E)-caryophyllene and (E)-methyl isoeugenol constituents. Chapter 3 presents the phytochemical assessment of the hexane, dichloromethane, ethyl acetate and aqueous fractions obtained from the crude ethanol extract of P. pseudocaryophyllus leaves. Chromatographic and spectrophotometric (1H and 13C) methods enabled the isolation and identification of lupeol, α-and -amyrin, oleanolic, ursolic and betulinic acids, quercetin, quercitrin, isoquercitrin, afzelin, avicularin, guaijaverin, catechin, ellagic acid and gallic acid. Chapter 4 assesses the antimicrobial activity of the ethanol extracts, fractions (hexane, dichloromethane, ethyl acetate and aqueous), essential oils and isolated compounds of citral (CQ) and (E)-methyl isoeugenol (MQ) chemotypes of P. pseudocaryophyllus. The broth microdilution test was applied to determine the minimum inhibitory concentration (MIC) against strains of Gram(+) and Gram(-) bacteria, and of Candida and Cryptococcus spp fungi. The CQ and MQ dichloromethane fraction, CQ ethyl acetate fraction, and CQ essential oil were the most active (MIC = 62.5 to 500 mg / mL) against Gram(+). The oleanolic acid proved to also be responsible for this action. The CQ and MQ extracts of P. pseudocaryophyllus were active (MIC = 7.8 to 500 mg / mL) against most strains of Candida spp and Cryptococcus spp, where gallic acid, ellagic acid and quercitrin contributed to this action. The data obtained indicate the antimicrobial potential of CQ and MQ P. pseudocaryophyllus against some infectious diseases. Chapter 5 describes the antinociceptive activity, in mice, of CQ and MQ ethanol extracts, CQ fractions and CQ essential oil, and the anti-inflammatory activity of CQ crude ethanol extract of P. pseudocaryophyllus. The abdominal writhing test induced by intraperitoneal administration (ip) of acetic acid (1.2% v / v) and the ear edema test induced by croton oil (2.5% V/V) were performed. The CQ crude ethanol extract (2000, 1000 and 500 mg/kg, po) showed a dose-dependent antinociceptive effect. Among the fractions, only the aqueous fraction was not active. The CQ essential oil (60, 200 and 600 mg / kg, po) presented significant antinociceptive activity, probably because of its high citral concentration. Pretreatment with CQ crude ethanol extract (2000, 1000 and 500 mg / kg) reduced edema by 14.3 ± 0.4 mg (control) to 10.9 ± 0.5, 11.3 ± 0, 4, 10.5 ± 0.5 mg, respectively. The analgesic effect may be related to an anti-inflammatory action observed in the ear edema test.
O presente trabalho teve como objetivos realizar o estudo fitoquímico e avaliar atividades biológicas do óleo essencial e de extratos da folha de Pimenta pseudocaryophyllus (Gomes) L. R. Landrum (Myrtaceae). O Capítulo 1 traz uma revisão bibliográfica do gênero Pimenta onde são abordados os principais aspectos botânicos, a composição química e o potencial farmacológico. O Capítulo 2 descreve a ocorrência de variabilidade infraespecífica em P. pseudocaryophyllus. Para tanto, foi realizada a análise qualitativa e quantitativa, por cromatografia gasosa acoplada a espectrometria de massas (CG/EM), do óleo essencial das folhas de 12 espécimes de P. pseudocaryophyllus que ocorrem naturalmente em três diferentes localidades do Cerrado brasileiro. A análise estatística multivariada dos dados identificou três quimiotipos de P. pseudocaryophyllus, caracterizados pela predominância dos constituintes citral, (E)-cariofileno e (E)-metilisoeugenol. O Capítulo 3 apresenta os resultados da avaliação fitoquímica das frações hexano, diclorometano, acetato de etila e aquosa obtidas a partir do extrato etanólico bruto das folhas de P. pseudocaryophyllus. A partir da utilização de técnicas cromatográficas e espectrofotométricas (RMN 1H e 13C) foi possível isolar e identificar os triterpenos pentacíclicos lupeol, α- e -amirina, os ácidos oleanólico, ursólico e betulínico, os flavonóides quercetina, quercitrina, isoquercitrina, afzelina, avicularina, guaijaverina e catequina, e os ácidos gálico e elágico. O Capítulo 4 expõe a avaliação da atividade antimicrobiana dos extratos etanólicos brutos, frações (hexano, diclorometano, acetato de etila e aquosa), óleos essenciais e substâncias semipurificadas de P. pseudocaryophyllus quimiotipos citral (QC) e (E)- metilisoeugenol (QM). Foi utilizado o teste da microdiluição em caldo, que permitiu determinar a Concentração Inibitória Mínima (CIM) frente a cepas de bactérias Gram(+), Gram(-) e fungos Candida spp. e Cryptococcus spp. As frações diclorometano (QC e QM), acetato de etila (QC) e o óleo essencial do quimiotipo citral foram os mais ativos (CIM = 62,5-500 μg/mL) frente as bactérias Gram(+), o ácido oleanólico mostrou ser um dos responsáveis por essa ação. O QC e QM foram ativos (CIM = 7,8 – 500 μg/mL) frente à maioria das cepas fúngicas, sendo que ácido gálico, ácido elágico e quercitrina contribuíram para essa ação. Os dados obtidos indicaram que P. pseudocaryophyllus QC e QM apresentam potencial antimicrobiano. O Capítulo 5 descreve a avaliação da atividade antinociceptiva, em camundongos, dos extratos etanólicos brutos (QC e QM), frações e óleo essencial (QC) de P. pseudocaryophyllus e a atividade anti-inflamatória do extrato etanólico bruto (QC) de P. pseudocaryophyllus. Foram utilizados os testes das contorções abdominais induzidas pela administração intra-peritonial (i.p.) de ácido acético (1,2%, v/v) e do edema de orelha induzido pelo óleo de cróton (2,5%, V/V) em camundongos. O extrato etanólico bruto QC (2000, 1000 e 500 mg/kg, v.o.) demonstrou efeito antinociceptivo dose-dependente. Dentre as frações, apenas a aquosa não foi ativa. O óleo essencial QC (60, 200 e 600 mg/kg, v.o.) demonstrou significativa atividade antinociceptiva, provavelmente por causa do seu elevado teor em citral. O pré-tratamento com o extrato etanólico bruto QC nas dose 2000, 1000 e 500 mg/kg reduziu o edema de 14,3±0,4 mg (controle) para 10,9±0,5; 11,3±0,4; 10,5±0,5 mg, respectivamente. A ação analgésica pode estar relacionada a uma ação anti-inflamatória, observada no teste de edema de orelha.

Texte intégralThe process of multiple motile cilia formation (multiciliogenesis) is composed of many different steps. Recently, we demonstrated that microRNAs of the miR-449 family control several of these steps. During my work, I focused on the role played by miR-449 in two aspects of the development of the mucociliary epithelium. In multiciliated cells, a dense actin network underlying the apical aspect of the cell membrane (actin cap) is required for the anchoring of the multiple basal bodies, and therefore for proper ciliogenesis. Small GTPases play important role in the formation of the actin cap. In the course of my work, I took part in the identification of transcripts coding the small GTPase R-Ras as bona fide targets of miR-449. I demonstrated that apical and subapical actin network reorganization and multiciliogenesis were impaired when R-Ras mRNA was protected from miR-449 binding. Moreover, the actin cap formation and multiciliogenesis were rescued when the translation of protected R-Ras transcripts was prevented. I also contributed to the finding that a new miR-449 target, the KIT receptor tyrosin kinase ligand STEEL, is involved in the process through which the multiciliated cells reach their final position within the developing frog epidermis. STEEL, which likely acts as a guidance molecule for the KIT-expressing multiciliated cells, needs to be repressed by miR-449 within the migrating cells to ensure their proper directional migration. Altogether, my work contributed to elucidate the complex role played by the miR-449 miRNA in the process of vertebrate multiciliogenesis
