Macrophages are major players in the pathogenesis of pulmonary arterial hypertension (PAH).To investigate whether lung macrophages and pulmonary-artery smooth muscle cells (PASMCs) collaborate to stimulate PASMC growth and whether the CCL2-CCR2 and CCL5-CCR5 pathways inhibited macrophage–PASMC interactions and PAH development, we used human CCR5-knock-in mice and PASMCs from patients with PAH and controls.Conditioned media from murine M1 or M2 macrophages stimulated PASMC growth. This effect was markedly amplified with conditioned media from M2 macrophage/PASMC co-cultures. CCR2, CCR5, CCL2 and CCL5 were upregulated in macrophage/PASMC co-cultures. Compared to inhibiting either receptor, dual CCR2 and CCR5 inhibition more strongly attenuated the growth-promoting effect of conditioned media from M2-macrophage/PASMC co-cultures. Deleting either CCR2 or CCR5 in macrophages or PASMCs attenuated the growth response. In mice with hypoxia- or SUGEN/hypoxia-induced PH, targeting both CCR2 and CCR5 prevented or reversed PH more efficiently than targeting either receptor alone. Patients with PAH exhibited CCR2 and CCR5 upregulation in PASMCs and perivascular macrophages compared to controls. The PASMC growth-promoting effect of conditioned media from M2-macrophage/PASMC co-cultures was greater when PASMCs from PAH patients were used in the co-cultures or as the target cells and was dependent on CCR2 and CCR5. PASMC migration toward M2-macrophages was greater with PASMCs from PAH patients and was attenuated by blocking CCR2 and CCR5.CCR2 and CCR5 are required for collaboration between macrophages and PASMCs to initiate and amplify PASMC migration and proliferation during PAH development. Dual targeting of CCR2 and CCR5 may hold promise for treating human PAH.

Myoendothelial gap junctional signaling mediates pulmonary arterial endothelial cell (PAEC)-induced activation of latent TGF-β and differentiation of cocultured pulmonary arterial smooth muscle cells (PASMCs), but the nature of the signal passing from PAECs to PASMCs through the gap junctions is unknown. Because PAECs but not PASMCs synthesize serotonin, and serotonin can pass through gap junctions, we hypothesized that the monoamine is the intercellular signal. We aimed to determine whether PAEC-derived serotonin mediates PAEC-induced myoendothelial gap junction-dependent activation of TGF-β signaling and differentiation of PASMCs. Rat PAECs and PASMCs were monocultured or cocultured with (touch) or without (no-touch) direct cell-cell contact. In all cases, tryptophan hydroxylase 1 (Tph1) transcripts were expressed predominantly in PAECs. Serotonin was detected by immunostaining in both PAECs and PASMCs in PAEC/PASMC touch coculture but was not found in PASMCs in either PAEC/PASMC no-touch coculture or in PASMC/PASMC touch coculture. Furthermore, inhibition of gap junctions but not of the serotonin transporter in PAEC/PASMC touch coculture prevented serotonin transfer from PAECs to PASMCs. Inhibition of serotonin synthesis pharmacologically or by small interfering RNAs to Tph1 in PAECs inhibited the PAEC-induced activation of TGF-β signaling and differentiation of PASMCs. We concluded that serotonin synthesized by PAECs is transferred through myoendothelial gap junctions into PASMCs, where it activates TGF-β signaling and induces a more differentiated phenotype. This finding suggests a novel role of gap junction-mediated intercellular serotonin signaling in regulation of PASMC phenotype.

Myoendothelial gap junctions are involved in regulating systemic arterial smooth muscle cell phenotype and function, but their role in the regulation of pulmonary arterial smooth muscle cell (PASMC) phenotype is unknown. We therefore investigated in cocultured pulmonary arterial endothelial cells (PAECs) and PASMCs whether myoendothelial gap junctional signaling played a role in PAEC-dependent regulation of PASMC phenotype. Rat PAECs and PASMCs were cocultured on opposite sides of a porous Transwell membrane that permitted formation of heterotypic cell-cell contacts. Immunostaining showed expression of the gap junctional protein connexin 43 (Cx43) on projections extending into the membrane from both cell types. Dye transfer exhibited functional gap junctional communication from PAECs to PASMCs. PASMCs cocultured with PAECs had a more contractile-like phenotype (spindle shape and increased expression of the contractile proteins myosin heavy chain, H1-calponin, and α-smooth muscle cell-actin) than PASMCs cocultured with PASMCs or cocultured without direct contact with PAECs. Transforming growth factor (TGF)-β1 signaling was activated in PASMCs cocultured with PAECs, and the PASMC differentiation was inhibited by TGF-β type I receptor blockade. Inhibition of gap junctional communication pharmacologically or by knock down of Cx43 in PAECs blocked TGF-β signaling and PASMC differentiation. These results implicate myoendothelial gap junctions as a gateway for PAEC-derived signals required for maintaining TGF-β-dependent PASMC differentiation. This study identifies an alternative pathway to paracrine signaling to convey regulatory signals from PAECs to PASMCs and raises the possibility that dysregulation of this direct interaction is involved in the pathogenesis of hypertensive pulmonary vascular remodeling.

To address developmental regulation of pulmonary vascular O2 sensing, we tested the hypotheses that 1) fetal but not adult pulmonary artery smooth muscle cells (PASMCs) can directly sense an acute increase in O2, 2) Ca2+-sensitive K+(KCa) channel activity decreases with maturation, and 3) PASMC KCa channel expression decreases with maturation. We used fluorescence microscopy to confirm that fetal but not adult PASMCs are able to sense an acute increase in O2tension. Acute normoxia induced a 22 ± 2% decrease in cytosolic Ca2+ concentration ([Ca2+]i) in fetal PASMCs and no change in [Ca2+]i in adult PASMCs ( P < 0.01). The effects of K+channel antagonists were studied on fetal and adult PASMC [Ca2+]i. Iberiotoxin (10−9 M) caused PASMC [Ca2+]i to increase by 694 ± 22% in the fetus and caused no change in adult PASMCs. KCa channel expression and mRNA levels in distal pulmonary arteries from fetal and adult sheep were examined. Both KCachannel protein and mRNA expression in the distal pulmonary vasculature decreased with maturation. We conclude that maturation-dependent changes in PASMC O2 sensing render the fetal PASMCs uniquely sensitive to an acute increase in O2 tension at a biologically critical time point.

Background/Aims: The enhanced proliferation of pulmonary arterial smooth muscle cells (PASMCs) is a central pathological component in pulmonary arterial hypertension (PAH). Both the Warburg effect and platelet-derived growth factor (PDGF) are involved in the proliferation of PASMCs. However, the mechanism underlying the crosstalk between the Warburg effect and PDGF during PASMC proliferation is still unknown. We hypothesized that PDGF promotes the Warburg effect via activating the phosphatidylinositol 3-kinase (PI3K) signaling pathway and hypoxia-inducible factor 1-α (HIF-1α) in proliferative PASMCs. Methods: PASMCs were derived from pulmonary arteries of SD rats; cell viability, the presence of metabolites, and metabolic enzyme activities assay were determined by MTT assays, kit assays and western blot analysis, respectively. Results: PDGF promoted PASMC proliferation in a dose- and time-dependent manner, accompanied by an enhanced Warburg effect. Treatment with PDGFR antagonists, Warburg effect inhibitor and PDK1 inhibitor significantly inhibited PI3K signaling activation, HIF-1α expression and PASMC proliferation induced by PDGF, respectively. Furthermore, treatment with PI3K signaling pathway inhibitors remarkably suppressed PDGF-induced PASMC proliferation and the Warburg effect. Conclusion: microplate reader (Biotek, Winooski The Warburg effect plays a critical role in PDGF-induced PASMC proliferation and is mediated by activation of the PI3K signaling pathway and HIF-1α.

Peroxisome proliferator-activated receptor-γ (PPARγ) and Rho-kinase (ROCK) regulate smooth muscle cell (SMC) proliferation and contribute to vascular remodeling in adult pulmonary hypertension. Whether these pathways interact to contribute to the development of vascular remodeling in persistent pulmonary hypertension of the newborn (PPHN) remains unknown. We hypothesized that ROCK-PPARγ interactions increase SMC proliferation resulting in vascular remodeling in experimental PPHN. Pulmonary artery SMCs (PASMCs) were harvested from fetal sheep after partial ligation of the ductus arteriosus in utero (PPHN) and controls. Cell counts were performed daily for 5 days with or without PPARγ agonists and ROCK inhibition. PPARγ and ROCK protein expression/activity were measured by Western blot in normal and PPHN PASMCs. We assessed PPARγ-ROCK interactions by studying the effect of ROCK activation on PPARγ activity and PPARγ inhibition (siRNA) on ROCK activity and PASMC proliferation. At baseline, PPHN PASMC cell number was increased by 38% above controls on day 5. ROCK protein expression/activity were increased by 25 and 34% and PPARγ protein/activity decreased by 40 and 50% in PPHN PASMC. ROCK inhibition and PPARγ activation restored PPHN PASMC growth to normal values. ROCK inhibition increased PPARγ activity by 50% in PPHN PASMC, restoring PPARγ activity to normal. In normal PASMCs, ROCK activation decreased PPARγ activity and PPARγ inhibition increased ROCK activity and cell proliferation, resulting in a PPHN hyperproliferative PASMC phenotype. PPARγ-ROCK interactions regulate SMC proliferation and contribute to increased PPHN PASMC proliferation and vascular remodeling in PPHN. Restoring normal PPARγ-ROCK signaling may prevent vascular remodeling and improve outcomes in PPHN.

Vascular remodeling resulting from altered pulmonary arterial smooth muscle cell (PASMC) growth is a contributing factor to the pathogenesis of hypoxic pulmonary hypertension. PASMC growth requires an alkaline shift in intracellular pH (pHi) and we previously showed that PASMCs isolated from mice exposed to chronic hypoxia exhibited increased Na+/H+ exchanger (NHE) expression and activity, which resulted in increased pHi. However, the mechanism by which hypoxia caused these changes was unknown. In this study we tested the hypothesis that hypoxia-induced changes in PASMC pH homeostasis are mediated by the transcriptional regulator hypoxia-inducible factor 1 (HIF-1). Consistent with previous results, increased NHE isoform 1 (NHE1) mRNA and protein, enhanced NHE activity, and an alkaline shift in pHi were observed in PASMCs isolated from wild-type mice exposed to chronic hypoxia (3 wk at 10% O2). In contrast, these changes were absent in PASMCs isolated from chronically hypoxic mice with partial deficiency for HIF-1. Exposure of PASMCs to hypoxia ex vivo (48 h at 4% O2) or overexpression of HIF-1 in the absence of hypoxia also increased NHE1 mRNA and protein expression. Our results indicate that full expression of HIF-1 is essential for hypoxic induction of NHE1 expression and changes in PASMC pH homeostasis and suggest a novel mechanism by which HIF-1 mediates pulmonary vascular remodeling during the pathogenesis of hypoxic pulmonary hypertension.

The proliferation of pulmonary artery smooth muscle cells (PASMCs) is an important cause of pulmonary vascular remodeling in pulmonary hypertension (PH). It has been reported that miR-137 inhibits the proliferation of tumor cells. However, whether miR-137 is involved in PH remains unclear. In this study, male Sprague-Dawley rats were subjected to 10% O2 for 3 weeks to establish PH, and rat primary PASMCs were treated with hypoxia (3% O2) for 48 h to induce cell proliferation. The effect of miR-137 on PASMC proliferation and calpain-2 expression was assessed by transfecting miR-137 mimic and inhibitor. The effect of calpain-2 on PASMC proliferation was assessed by transfecting calpain-2 siRNA. The present study found for the first time that miR-137 was downregulated in pulmonary arteries of hypoxic PH rats and in hypoxia-treated PASMCs. miR-137 mimic inhibited hypoxia-induced PASMC proliferation and upregulation of calpain-2 expression in PASMCs. Furthermore, miR-137 inhibitor induced the proliferation of PASMCs under normoxia, and knockdown of calpain-2 mRNA by siRNA significantly inhibited hypoxia-induced proliferation of PASMCs. Our study demonstrated that hypoxia-induced downregulation of miR-137 expression promoted the proliferation of PASMCs by targeting calpain-2, thereby potentially resulting in pulmonary vascular remodeling in hypoxic PH.

Chronic hypoxia-induced pulmonary arterial hypertension (HPH) is closely associated with profound vascular remodeling, especially pulmonary arterial medial hypertrophy and muscularization due to hyperplasia of pulmonary artery smooth muscle cells (PASMCs). Aberrant Wnt signaling has been associated with lung diseases, but its role in pulmonary hypertension is unclear. This study evaluated the effect of Wnt5a on hypoxia-induced proliferation of human PASMCs and its possible mechanism. The results show that hypoxia (3% O2, 48 h) induced proliferation of human PASMCs, accompanied with a significant decrease in Wnt5a gene expression, increase in β-catenin and Cyclin D1 expression, as well as β-catenin nuclear translocation. Treatment with recombinant mouse Wnt5a significantly inhibited hypoxia-induced proliferation of human PASMCs, upregulation of Cyclin D1 and β-catenin expression, as well as the nuclear translocation of β-catenin. These effects were inhibited by Wnt5a antibody. Knocking down β-catenin or Cyclin D1 gene expression inhibited hypoxia-induced human PASMC proliferation, whereas overexpression of β-catenin increased hypoxia-induced human PASMC proliferation and counteracted the inhibitory effect of Wnt5a. These results suggest that Wnt5a has an antiproliferative effect on hypoxia-induced human PASMC proliferation by downregulation of β-catenin and its target gene Cyclin D1. Hypoxia-induced downregulation of Wnt5a may be a way to facilitate hypoxia-induced human PASMC proliferation. The results of this study will help to understand the novel strategies for PH treatment involving Wnt signaling.

Intracellular Ca2+ levels play a critical role in the regulation of vasodilation and vasoconstriction by stimulating pulmonary artery smooth muscle cell (PASMC) proliferation, which is important in the pathogenesis of pulmonary arterial hypertension (PAH); however, L-type Ca2+ channel antagonists are useful in only few patients with PAH. The present study sought to assess the effect of mibefradil, which blocks T-type Ca2+ channels, on PASMC proliferation and Ca2+ channel profile. Human PASMCs were stimulated with 25 ng/mL platelet-derived growth factor-BB (PDGF-BB) with and without 10 µM mibefradil or 100 nM sildenafil. After 48 or 72 h, PASMC proliferation and Ca2+ channel expression were assessed by MTT assays and western blot analysis, respectively. PDGF-BB-induced PASMC proliferation at 72 h (p<0.01), which was inhibited by both sildenafil and mibefradil (p<0.01). Transient receptor potential Ca2+ channel 6 (TRPC6) expression was significantly increased with PDGF-BB stimulation (p=0.009); however, no changes in TRPC1, TRPC3, CAV1.2, and CAV3.2 levels were observed. Although both TRPC1 and CAV1.2 expression levels were increased in PDGF-stimulated PASMCs on mibefradil and sildenafil treatment, it was not statistically significant (p=0.086 and 1.000, respectively). Mibefradil inhibits PDGF-BB-stimulated PASMC proliferation; however, the mechanism through which it functions remains to be determined. Further studies are required to elucidate the full therapeutic value of mibefradil for PAH.

Alteration in the tone of pulmonary arteries may lead to disease such as pulmonary hypertension often associated with major cardiac complications. This dysfunction is partly in the pulmonary arterial smooth muscle cells (PASMCs) where the excitation-contraction coupling is modified by ion channel behaviour to increase the contractile force. Mathematical models of systemic smooth muscle cells (SMCs) that incorporate electrophysiological and chemomechanical mechanisms to understand the underlying cellular physiology have been successfully employed. Models of pulmonary arterial smooth muscle cells (PASMCs) are only beginning to emerge. Mathematical model prototyping with available experimental data and model investigation from different parameter values is a time-consuming and complex process. This thesis is concerned with the development and validation of mathematical models of excitation-contraction coupling in three types of PASMCs of the rat species, one homogeneous type originating from the distal pulmonary arteries and two from proximal pulmonary arteries. Some key novel additions from previous vascular SMC models include the distinct modelling of Ca2+ in the subplasmalemmal cytosolic region, incorporation of subunit-specific currents from the K+ channel family and a generic G-protein receptor model able to reproduce complex Ca2+ profiles. The main pulmonary and systemic arteries statistically differ in its response to phenylephrine in a wire myograph. The ionic currents of the models were validated against experimental data largely from rat species. The models replicate the recordings of Ca2+ and the resting potential (Em) profiles arising from agonist-induced cytosolic Ca2+ ([Ca2+]i) stimulation (G-protein activation), nifedipine, ryanodine, caffeine and niflumic acid. The distal PASMC model was sensitive to an increase in [Ca2+]i from G-protein activation although were less likely to reproduce Ca2+ oscillations than proximal PASMCs. The proximal models determined the likely proximal PASMC type in literature experiments recording [Ca2+]i and Em. I have developed software that enables other users to simulate Ca2+ and Em changes in SMC studies and the ability to parse a master file describing the mathematical model into different language formats to increase productivity. These models provide a foundation for further studies to better understand PASMC function in the context of normal physiology as well as pathological conditions.

The Policy Aware Social Miner (PASM) project focuses on creating awareness of how seemingly harmless social data might reveal sensitive information about a person, which could be potentially abused. It seeks to define good practices around social data mining. PASM allows people to create policies governing the use of their personal information on social networks. Using linked data, PASM semantically enhances the usage restrictions to ensure that potentially sensitive information is identified and appropriate policies are enforced. PASM also enables people to provide refutations for other information about them that is found on the Web. PASM encourages consumers of social information on the Web to use the mined data appropriately by enforcing data policies before returning the search results. PASM provides a solution to the following issue of privacy in social data mining - although people know that searches for data about them are possible, they have no way to either control the data that is put on the Web by others or indicate how they would like to restrict use of their own data. In a user study conducted to measure the performance of PASM in identifying sensitive posts as compared to the study participants, PASM obtained an F-Measure of 84% and an accuracy of 80%. Interestingly, PASM demonstrated a higher recall than precision, a property that was valued by the study participants as all but one participant indicated that they would prefer receiving false positives rather than false negatives.

Várias ligas de Al em cavacos, com diferentes granulometrias foram dissolvidas de modo convencional, com ataque por água régia. Este tipo de decomposição resulta, muitas vezes, em resíduos sólidos, cujas quantidades dependem dos teores de silício. A fim de se comparar os resultados, procedeuse, com esses resíduos, de dois modos distintos: ou foram desprezados ou foram decompostos com ácido fluorídrico, eliminando-se o silício, dissolvendo-se o restante com ácido nítrico e agrupando-o à primeira solução para a determinação de Fe, Cu, Mn, Mg, Cr, Ni, Zn, Pb, Sn, Ti e Zr por ICP-OES. Foi avaliada a influência de quantidades crescentes de Al na determinação destes elementos e, também, a provável interferência espectral de Cu na determinação do Zn (com e sem separação eletrolítica de Cu). A dissolução eletrolítica de ligas de alumínio em cavacos foi estudada pressionando-os contra um eletrodo de platina para que se comportassem como se fossem o próprio ânodo e a eletrólise ocorresse. Uma célula foi construída utilizando-se um tubo (corpo de seringa de 2,5 mL) com perfurações ao redor da base aberta, onde foi colocada uma malha sintética (perlon) no fundo e, sobre esta, a amostra. Na parte superior foi introduzido um pequeno tarugo de acrílico com um fio de platina passando pelo seu interior e com um orifício por onde passava o fluxo (4,5 mL min.-1) da solução eletrolítica (ácido nítrico diluído). Na extremidade inferior, prendeu-se, com o fio, uma plaquinha, também de platina, que pressionava a amostra. A solução saía pelos orifícios inferiores para um béquer de onde era recirculada e onde estava colocado o cátodo, um fio de Pt (de forma de losango), sendo novamente captada e introduzida na parte superior do tarugo do ânodo. Com esse circuito, foram obtidos bons resultados utilizando-se ácido nítrico 1 mol L-1, corrente de 0,6A e tempo de eletrólise de 10 minutos. Sendo a dissolução parcial, é necessário que se conheçam as porcentagens dos elementos por dissolução ácida total de uma liga de composição similar ou por um material de referência certificado. Normalizaram-se os dados pelas porcentagens conhecidas dos constituintes das ligas. Compararam-se os cálculos feitos dos seguintes modos: 1-) pela razão entre a concentração do elemento com a do Al, também determinada; 2-) pela a razão entre a concentração do elemento com a soma das concentrações de todos os elementos determinados, inclusive o Al; 3-) pela razão entre a concentração do elemento e a soma das concentrações só dos outros elementos, sem o Al. O desvio padrão e o erro relativo para os elementos Fe, Cu, Mg, Ni e Cr, quando em teores > 0,1%, foram < 10%; para teores de Zn < 0,2%, foram menores de 10% e para teor de 4,8% foram < 15%. Já para o Mn, quando em teor > 0,2% foram <10% (exceto para a liga B.C.S.268/1). O desvio padrão relativo para o Zr, com teor ao redor de 0,002% foi <10%. Os resultados para o elemento Pb nas ligas B.C.S. 216/1 e 380 tiveram desvio padrão e erro relativo <10%, mas o mesmo não ocorreu na liga B.C.S. 268/1. O elemento Sn apresentou resultados piores, ao redor de 20%. O elemento Ti só deu bons resultados quando em teor de cerca de 0,2%. Para estas determinações foram avaliadas as dissoluções químicas dos metais nas ligas pelo ácido nítrico em diferentes concentrações e temperaturas. Além disso, foram avaliadas duas outras células para a verificação da possibilidade de dissolução eletrolítica das ligas de Al on-line com a determinação por ICP-OES.
Several aluminum alloys with different chips size were dissolved using aqua regia (HCl:HNO3,3:1). The residues of the dissolution procedure were either rejected or used in silicon volatilization with hydrofluoric acid before determination of Fe, Cu, Mn, Mg, Cr, Ni, Zn, Pb, Sn, Ti and Zr by ICP-OES. The influence caused by crescent amounts of aluminum in the determination of other elements and the probable spectral interference by Cu in determination of Zn (with or without electrolytic separation of Cu) was evaluated. The electrolytic dissolution of aluminum alloys was performed by pressing chips against a platinum electrode for the electrolysis to occur, so that way the chips behaved as the anode. A tube (body of syringe of 2.5mL) with perforations around the open base was used to contain the sample and Pt electrode. The alloy was placed on synthetic wool (perlon) in this tube. An acrylic dowel with a platinum wire across its interior was fixed in the superior part a tube with Teflon film, with an orifice through which the electrolytic solution flowed (HNO3 1 mol L-1; 4.5mL min-1). A platinum dish was fixed at the inferior extremity of the platinum wire to press the sample. The solution came out through the inferior orifices of the tube to a flask, from where it was again captured (with a peristaltic pump) to go into the superior part of the tube. In this flask, a Pt wire cathode was also placed. A current of 0,6A and ten minutes of electrolysis provided good results. Due to the partial dissolution, the knowledge of percentages of previous elements analyzed after total dissolution, or using certified reference material are necessary. The results were normalized by known percentages. The calculations were made by: 1-) the ratio between the concentration of the element determind, with the determined concentration of aluminum; 2-) the ratio between concentration of the element and the sum of the concentrations of the elements determined, including Al; 3-) the ratio between the concentration of element and the sum of the concentrations of the elements determined, without the aluminum. The relative standard deviations and accuracy were <10% for the elements Fe, Cu, Mg, Ni and Cr when the concentrations were >0.1%; for the Zn were 15%, when the concentration >0.2%, and <10% for concentration <0.2%. For Ti with concentration of about 0.1%, were 15%. Relative standards deviations for the Zr, with concentration of 0.002%, were always <10%. The element Pb, Sn and Mn didnt provide good results. For these determinations, the chemical dissolution of the elements with different nitric acid concentrations and temperature was evaluated. Two other cells for the electrolytic dissolution of Al alloys followed by online ICP-OES determination were examined.

De la décennie 80 aux années 2000, la qualité de l’enseignement en Afrique subsaharienne s’est progressivement dégradée sous l’influence de multiples facteurs. Déjà caractérisés par un faible niveau d’efficacité interne, de scolarisation et d’acquis scolaires, eux-mêmes parsemés de disparités et d’inégalités, les transformations du secteur vont être accentuées par la hausse de la population scolarisable. Face à cette situation, les pouvoirs publics adopteront à l’aune de l’an 2000 une réforme des curricula par l’Approche Par Compétences (APC). Attachée à des enjeux de justice et d’amélioration des apprentissages, la mise en œuvre de l'APC n’a pas donné lieu à une large évaluation dans la littérature économique. C’est fort de ce constat, que cette thèse s’est fixée pour objectifs d’analyser son transfert et son impact par la comparaison des expériences ivoirienne et sénégalaise. Pour y parvenir, elle prend appui sur les mix methods. Aussi, les chapitres 1 et 2 identifient les caractéristiques et les particularités de chaque système, puis les déterminants et les enjeux de la réforme. Quant au chapitre 3, il analyse son transfert et son effectivité. A sa suite, le chapitre 4 évalue son impact sur les indicateurs d’efficacité interne et les acquis scolaires à travers un modèle multiniveaux. Les résultats obtenus suggèrent que l'APC ne permet pas d’expliquer l’amélioration de l’efficacité interne, qui a été le fait d'une révision des règles de régulation inter-cycles intervenue dans le cadre de la politique universelle d'éducation. Concernant la qualité des apprentissages, l’analyse économétrique corrobore l’évaluation qualitative du transfert
From 1980 to 2000, the education quality in sub-Saharan Africa decreased gradually under multiple influence. Already characterized by a low level of internal efficiency, schooling and learning outcomes, themselves dotted with regional disparities, gender and unequal access, the transformations of the education sector will be accentuated by the rise in school-age population. To deal with this situation, Governments adopt a curriculum reform at the beginning of 2000 through the Skills-Based Approach.Attached to social justice issues and learning quality, the Skills-Based Approach’ implementation has not always been conducive to rigorous evaluation in the economic literature. Due this situation, our thesis tempts to analyze its transfer and impact by comparing the Ivorian and Senegalese experiences. To achieve this goal, our work has been based on mix methods. In doing so, chapters 1 and 2 successively identify each system particularities and then the curriculum determinants and main objectives. Consecutively, chapter 3 analyzes its transfer, articulation and effectiveness in each educational system, whereas Chapter 4 assesses its impact on internal efficiency and learning quality by a multilevel model.By comparing the results from each methods, we observe that the Skills-based Approach does not explain internal efficiency improvement, which is rather the consequence of inter-cycle transition rules revision. Concerning learning quality, the econometric analysis corroborates the transfer assessment, and suggests a new approach to educational product quality: it insists to pay particular attention to the way in which educational policy is conceived and disseminated

O planejamento e gerenciamento da manutenção de software é um problema tanto para usuários quanto para engenheiros de software. O processo que conduz a execução das demandas registradas pelos usuários pode interferir diretamente no custo da manutenção. O presente estudo apresenta o Processo para Agrupamento de Solicitações de Manutenção (PASM), descrevendo suas diretrizes para guiar a priorização, agrupamento e execução de diversos tipos de manutenção de software. O PASM foi aplicado na Divisão de Tecnologia da PUC Minas e entre os benefícios identificados encontram-se o aumento de 18,76% das horas trabalhadas em projetos de manutenção e um declínio de 27,73% nas horas trabalhadas em solicitações urgentes.

The Federal scientific center for bast crops has long been creating varieties with high resistance to 2 diseases (rust and Fusarium wilt), and now successfilly solves the problem of forming resistance to 3 (rust, Fusarium and Anthracnose) and even to 4 diseases (rust, Fusarium, Anthracnose and Pasmo). New proposals for phytosanitary monitoring and forecasting racionalize plant protection in relation to flax production. Ecologized biological preparations (for example, Vitaplan, Sternifag) are effective against flax diseases (Bacteriosis, Anthracnose, Mottling, etc.) and contribute to the preservation of the flax crop.
