The emergency control of Menglou~Qifang inverted siphon, which is about 72 km long, is the key to the safety of the Northern Hubei Water Transfer Project. Given the complicated layout of this project, traditional emergency control method has been challenged with the fast hydraulic transient characteristics of pressurized flow. This paper describes the application of model predictive control (MPC), a popular automatic control algorithm advanced in explicitly accounting for various constraints and optimizing control operation, in emergency condition. For the fast prediction to the pipe-canal combination system, a linear model for large-scale inverted siphon proposed by the latest research and the integrator-delay (ID) model for open canals are used. Simulation results show that the proposed MPC algorithm has promising performance on guaranteeing the safety of the project when there are sudden flow obstruction incidents of varying degrees downstream. Compared with control groups, the peak pressure can be reduced by 17.2 m by MPC under the most critical scenario, albeit with more complicated gates operations and more water release (up to 9.75 × 104 m3). Based on the linear model for long inverted siphon, this work highlights the applicability of MPC in the emergency control of large-scale pipe-canal combination system.

ABSTRACT QuantiFERON-TB (QIFN) (CSL Limited) is a whole-blood assay for the recognition of infection withMycobacterium tuberculosis. QIFN measures gamma interferon (IFN-γ) production when purified protein derivatives (PPDs) of mycobacteria are incubated with venous blood samples. The specificity of QIFN in medical students before and after BCG immunization was assessed, and sensitivity in patients with tuberculosis was assessed. Antigens were PPD derived from M. tuberculosis and twoM. tuberculosis-specific proteins, ESAT-6 and MPT-64. Of 60 medical students, all of whom had 0-mm tuberculin skin tests (TSTs) at study entry, 58 (97%) were initially classified as negative forM. tuberculosis infection by PPD QIFN. Five months after BCG immunization, 7 of 54 students (13%) had a TST result of ≥10 mm and 11 of 54 students (20%) tested positive by PPD QIFN. ESAT-6- and MPT-64-stimulated IFN-γ responses in the medical students were negative prior to and after BCG immunization. For patients with active tuberculosis, 12 of 19 (63%) were positive by PPD QIFN, 11 of 19 (58%) were positive by ESAT-6 QIFN, and 0 of 12 were positive by MPT-64 QIFN. In conclusion, PPD QIFN was negative in 97% of a low-risk population who had not received BCG and who had negative TSTs. The specificities of both the TST and PPD QIFN were reduced following BCG immunization. PPD QIFN and ESAT-6 QIFN were of similar and moderate sensitivity in patients with active tuberculosis, but ESAT-6 QIFN is likely to be more specific because it is not influenced by past BCG exposure.

We evaluated an in vitro test of cell-mediated immunity, the tuberculin gamma interferon assay, QuantiFERON-TB (QIFN), in 455 individuals from three groups: group I, 237 immigrants from high-risk countries; group II, 127 health care workers undergoing Mantoux testing; group III, 91 patients being investigated for possible active tuberculosis (79 patients) or Mycobacterium avium-Mycobacterium intracellulare complex infection (12 patients). The QIFN results were compared either to those of the Mantoux test or to microbiological and clinical diagnosis, as appropriate. In each group the correlation between the diameter of induration for the skin test and the magnitude of QIFN response was significant and of moderate strength (Spearman’s rank correlation coefficient; ρ = 0.59 to 0.61; P < 0.001). For group I, the agreement between QIFN and Mantoux results was 89% for Mantoux-negative and 64% for Mantoux-positive individuals. For group II, when ≥10-mm-diameter induration was taken as positive, the agreement was 81% for Mantoux-negative and 67% for Mantoux-positive individuals. For group III, agreement was 81% for Mantoux-negative and 86% for Mantoux-positive patients. For patients being evaluated for active tuberculosis, the performance of the Mantoux test was not statistically different from that of the QIFN assay. In patients with active tuberculosis, the assay had a sensitivity of 77%, not significantly higher for extrapulmonary than pulmonary cases (83% versus 74%). QIFN sensitivity was not significantly different for smear-negative or smear-positive cases (80% versus 71%). The QIFN assay is a potential replacement for the Mantoux test. The acceptability of these performance values and those of similar evaluations will determine the place this test will have in detecting evidence of mycobacterial infection.

Abstract More than 100 Nobel laureates recently signed a letter urging Greenpeace to end its opposition to genetically modified organisms (GMOs). With an ever-increasing demand for food, China finds itself confronted with the same question: is it time to rethink and reassess its stance towards GMO? Rice is one of the most important food crops in China and around the world. In recent years, Chinese scientists have made important progress in rice functional genomics research including identifying genes and regulatory mechanisms and platforms and technologies for breeding new varieties. However, in contrast to the progress in scientific research, the commercial, social and policy framework remains largely unresolved. National Science Review (NSR) spoke with Qifa Zhang (Zhang), Professor at Huazhong Agricultural University, Academician of the Chinese Academy of Sciences and Foreign Associate of National Academy of Sciences, USA. In this interview, Prof. Zhang shares with us his views and insights on the frontier research in rice studies and commercialization of transgenic rice in China.

Objective: To analyze the distribution characteristics of syndrome of TCM of the patients with COVID-19 in Kunming, China. Methods: To classify and summarize the TCM syndrome types of COVID-19 patients via Cluster analysis combining with tongue image, chest CT and clinical expertise of 36 Covid-19 patients in Kunming. Results: In the 36 cases of Kunming region confirmed COVID-19 patients, 17 cases had fever (47.2%), 18 cases had cough (50%), 16 cases felt bitter taste in the mouth (44.4%), 18 cases felt dry throat (50.0%), 17 cases had poor appetite (47.2%), 15 cases had nausea (41.7%); 12 cases had diarrhea (33.3%), 15 cases had insomnia (41.7%); 12 cases had chest tightness (33.3%); 3 cases had dyspnea (8.3%); 6 cases had nasal congestion and running nose (16.7%); 15 cases had fatigue (41.7%); 6 cases had headache and body pain (16.7%); 5 cases had red tongue (13.9%); 18 cases had pale red tongue (50%); 8 cases had tongue with red edge and tip (22.2%); 3 cases had dark red tongue (8.3%); 2 cases had cyanosis (5.6%); 3 cases had swollen tongue (8.3%); 18 cases had dentate tongue (50%); 4 cases had yellow tongue coating (11.1%); 5 cases had yellow sticky tongue coating (13.9%); 12 cases had white sticky tongue coating (33.3%); 6 cases had thin white tongue coating (16.7%); 2 cases had no tongue coating (5.6%). The chest CT results showed that: There were five cases without lesions. The lesions were located in the upper lobe of one lung in 13 cases located in the lower lobe of one lung in seven cases, located in the upper middle lobe in three cases, located in the lower lobe in five cases, and in the upper middle and lower lobes of double lung in 13 cases. There were 14 cases of Shaoyang syndrome, 17 cases of wet Resistance Tir-juao Syndrome. According to the time of onset, the disease was followed by Shaoyang Syndrome (1 day), the Wet blocked tri-jiao Syndrome (3 days), epidemic poison retention lung syndrome and syndrome of flaring heat in qifen and yingfen (5 days), and dampness-toxicity lung-stagnation syndrome (6 days). Conclusion: The TCM syndromes of COVID-19 in Kunming are mainly the Wet Resistance Tri-Jiao Syndrome and Shaoyang syndrome, followed by dampness-toxicity lung-stagnation syndrome, epidemic poison retention lung and syndrome of flaring heat in qifen and yingfen.

I cannot overstate that 2020 was one of the toughest years on our industry’s ecosystem. The huge scale of the crisis wreaked by the COVID-19 pandemic has affected us all in such an unprecedented way and added a hard-hitting dimension to our industry’s fragile outlook. The prolonged periods of lockdown and the hiatus mandated in several countries, including some of the high-consuming ones, resulted in the largest oil glut ever, which exacerbated the economics severely. The flat oil-supply curve erased almost all the growth of the last decade and has put our endurance, agility, and resilience to a hard test. The offshore segment, from which approximately 33% of global oil production comes, was confronted by this turmoil, and escalating challenges surrounded the viability and sustainability of deepwater developments like never before. Unlike previous downturns experienced by our industry, the record plunge in oil demand has put the effectiveness of previous measures of recovery, such as deferral of investments or waiting for an upturn in oil prices, under the microscope. As the headwinds of the pandemic continue reshaping the global economy, our industry, and the deepwater segment in particular, have no time to lose to rock the boat and upend assumptions in the post-COVID-19 world. Imminent substantive changes within the industry are inevitable to reduce uncertainty and risk and to prepare for the new decade and beyond of delivery in light of the rapid evolution of renewables. Major deepwater operators have made remarkable achievements in lowering the average breakeven price to around $50 per barrel from around $70 per barrel in the past 5 years. Increasing cost savings are vital, and innovative money-saving engineering notions are an essential tactic to achieve a lower breakeven price. Additional proportionate, strategic, and reinventive initiatives are needed for the deepwater sector to adapt to the new energy landscape and search for new areas of growth. They will govern the future of the sector and differentiate the pioneers from followers. I am in no doubt that redefining fundamental strategies toward the development of partnerships in joint deepwater projects, embracing cutting-edge fit-for-purpose and cross technologies, and prioritizing net-zero emissions are keys to unlocking transformational gains and leveraging the opportunity to promote a sustainable future for the deepwater sector. In this study, we have studied the interaction and mechanism of survivin in regulating leukemia stem-like KG-1a cell escape the synergistic cytotoxic effect of arsenic trioxide and aclacinomycin. MATERIALS AND METHODS: The anti-proliferation effect was detected by CCK-8 and colony-forming assay, protein-protein chip assay was used to analysis the expression level of survivin before and after combination treatment. The induction of apoptosis and cell cycle arrest in KG-1a cell line were detected by FACS, the expression of related signal pathway protein was detected by western blot. RESULTS: The anti-proliferation of KG-1a cells caused by arsenic trioxide or aclacinomycin showed a time- and dose-independent manner. However, protein-protein chip assay showed that the expression of survivin had a significant increase after the combination treatment (p<0.05), but when survivin was suppressed, the apoptosis rate had a more significant increase than the single drug treatment. Meanwhile, more prominent cell cycle arrest was observed in the combination treatment, further study found that suppression of survivin combined with chemotherapy may activate the related apoptosis pathway protein but suppress the PI3K/AKT signal pathway. CONCLUSION: survivin play an important role in regulating leukemia stem cell escape the synergistic cytotoxic effect of arsenic trioxide and aclacinomycin. The mechanism of regulation process may through activate the related apoptosis pathway protein and supperss the PI3K/AKT signal pathway. National Curriculum for English in Pakistan, reveals its intentions for acquiring language related international requirements concerning with career choices and furthering education. Formative assessment is at the center of students’ learning and is devised in the curriculum document providing basis for its implementation. Curriculum implementation is pivotal to classroom practices of teachers. Curriculum guides teachers to implement formative assessment. Professional commitment contributes enriching teacher’ classroom practices. Focus of the study was to examine formative assessment practices of teachers, as a part of curriculum implementation, in accordance with curriculum document and its relationship with professional commitment of secondary school English teachers. Study secures its significance for teachers, curriculum developers, and governmental agencies. Quantitative survey was the method adopted for the study and explanatory correlational design was used. Teachers, concerned with teaching of English at secondary level, from entire secondary schools working under the supervision of the Punjab province were the study population. Sampling procedure employed multistage stratified random sampling technique and 576 teachers were selected as study sample. Two questionnaires were developed that included questionnaire regarding implementation of formative assessment (QIFA) and teachers’ professional commitment scale (TPCS). Survey was conducted as a data collection strategy. In statistics, Mean and Spearman correlation were employed. Results revealed that professional commitment of teachers and their formative assessment practices were moderate. Results regarding relationship between the variables; professional commitment of teachers and their formative assessment practices were significant correlated. The study recommended school based teacher trainings to sensitize and enhance teachers’ awareness and practices towards formative assessment practices as a part of implementation process. Governmental agencies are needed to focus the situation of professional commitment of teachers and implementation of formative assessment as a part of implementation.

54 Background: Direct cytolysis of tumor cells by CD8+ T cells results from the net effect of at least two biochemical pathways: (1) stimulatory signaling from the activated T cell receptor (TCR) complex in response to its recognition of a tumor neoantigen presented in the context of autologous MHC class I, and (2) suppressive signaling from immune checkpoints, such as the response of PD1 to binding its ligand, PDL1. Because the PD1:PDL1 immune checkpoint is significant for therapy only when there is tumor cell-specific TCR activation and signaling, it is not surprising that simple measurements of either PD1 or PDL1 in tumor biopsies are, at best, imperfect predictive biomarkers. Instead, a more precise test that quantifies PD1 signaling due to PDL1 binding only in the subset of CD8+ T cells exhibiting activated TCR signaling should provide a more accurate assessment of the extent of immune checkpoint suppression of tumor immunity and therefore be a more predictive biomarker of response to PD1/PDL1-targeted immunotherapy. Methods: We have developed a multiplexed immunofluorescence microscopy test capable of simultaneous quantitation of TCR activation (phospho-CD3zeta), immune checkpoint signaling via PD1 (phospho-SHP1 and -SHP2), and the net stimulation or inhibition resulting from the integration of these two pathways (phospho-ZAP70). Results: Specific antibodies to these biomarkers have been qualified, including peptide inhibition studies to establish antibody specificity, and their performance established by fit-for-purpose studies of in vitro models of CD8+ T cell activation. This multiplex biomarker panel is suitable for clinical use with formalin-fixed, paraffin embedded core needle biopsies of tumor and quantitative immunofluorescence microscopy (qIFA). Conclusions: The additional biomarkers of tumor immunity are expected to add an important context for interpreting PD1/PDL1 measurements. Funded by NCI Contract No. HHSN261200800001E.

Gravestones have a major significance in history and archaeology for being authentic historical documents that reveal many aspects of ancient societies pertaining to their population, races, religious culture, language, professions, trades, and many other aspects. Given that the heritage of Gaza has been subjected to destruction due to political conflict and negligence, the researcher embarked on several tours to take an inventory of gravestones in Gaza’s cemeteries in reality, which are of great importance with respect to Gaza’s eminent figures and their families during the end of the Ottoman era.The total number of gravestones tallied in Gaza reached 139, dating back to the period between AH 1230/1814 CE and the British occupation of Palestine in AH 1336/1917 CE, which is the date of the most recent gravestones. On one of the highest hills of Bab al-Bahr (Gate of the Sea) and Al-Sheikh Shaban cemetery, 45 tombstones were found belonging to the families: El-Husseini, Abu Khadra, Abu Shaban, Saq Allah, El-Bar'asi, Abu Assi, Murshid, El-Ja'farawi, El-Ghusain, Si-Salem, Al-Radwan, and others. In the Bin Marwan cemetery, 37 tombstones were discovered belonging to the families: El-Nakhal, El-Shawwa, Wafa El-Alami, El-Bitar, El-Wahidi, El-Khaznadar, and others. In El-Tiflisi (Abu al-Kass) cemetery in the Shuja'iyya neighborhood, 18 tombstones were discovered belonging to the families: Bseiso, Hathat, and sheikhs of El-Hasanat tribe, El-Sunna', El-Qadirat, and others. In Addiriyah and El-Tamrtashi cemetery, 9 tombstones were found belonging to the families: El-Tamrtashi, Murad, El-Batsh, El-Jabali, and others; 3 tombstones belonging to Al-Ghusain family were found in the cemetery of Al-Ghusain school; and 2 tombstones belonging to Al-Husseini’s and El-Jaouni’s families were found in El-Mufti cemetery. In the cemetery of the Orthodox Church of Saint Porphyrius, 25 gravestones were found belonging to the families: Zarifa, El-Tarzi, Farah, Qifa, El-Madbak‎‏, El-Sayegh, Musaad, Shuhaiber, El-Tawil, and El-Jildah.

Abstract Non-point source (NPS) pollution has caused serious threats to water quality on a global scale. However, the investigation using a single measure with multi-scenarios for the long-term simulation in karst agricultural watershed is still lacking. In this study, the Annualized Agricultural Non-Point Source Pollution (AnnAGNPS) model was developed to verify the applicability in a karst agricultural watershed. Then, the model was used to determine the best management measures (BMPs) and the reduction rate characteristics under long-term effects (about 10 years) in the watershed. The AnnAGNPS model perform well in simulating in NPS pollution with R2 (0.95 for runoff, 0.93 for TN, and 0.93 for TP, respectively) and NSE (0.95 for runoff, 0.53 for TN, and 0.57 for TP, respectively). The output of total nitrogen (TN) and total phosphorous (TP) primarily occurred in the rainy season (up to 80%). The loss of mass of TN and TP were mainly observed in orchards and woodlands in the upstream of each sub-basin. The results from AnnAGNPS model demonstrated that different BMPs had significant impacts on the reduction of NPS pollution. Furthermore, a same BMPs measure showed it was closely related to land use in the watershed. In the Qifeng River watershed, stubble tillage (ST) showed to be useful with relatively good reduction rates (16.64% for sediment, 17.85% for TN, and 17.80% for TP, respectively). The simulation results indicated that AnnAGNPS was a valuable tool after validation for the planning and management of the watershed in karst areas.

Xing Du is first author on ‘SMARCA2 is regulated by NORFA–miR-29c, a novel pathway that controls granulosa cell apoptosis and is related to female fertility’, published in JCS. Xing is a postdoc in the lab of Qifa Li at Nanjing Agricultural University, Jiangsu, China, investigating the genetic mechanism and epigenetic regulation of sow prolificacy, and the molecular mechanism of mammalian ovarian granulosa cell apoptosis.
