Résumé :

ObjectivesJuvenile idiopathic arthritis (JIA) is a heterogeneous group of conditions unified by the presence of chronic childhood arthritis without an identifiable cause. Systemic JIA (sJIA) is a rare form of JIA characterised by systemic inflammation. sJIA is distinguished from other forms of JIA by unique clinical features and treatment responses that are similar to autoinflammatory diseases. However, approximately half of children with sJIA develop destructive, long-standing arthritis that appears similar to other forms of JIA. Using genomic approaches, we sought to gain novel insights into the pathophysiology of sJIA and its relationship with other forms of JIA.MethodsWe performed a genome-wide association study of 770 children with sJIA collected in nine countries by the International Childhood Arthritis Genetics Consortium. Single nucleotide polymorphisms were tested for association with sJIA. Weighted genetic risk scores were used to compare the genetic architecture of sJIA with other JIA subtypes.ResultsThe major histocompatibility complex locus and a locus on chromosome 1 each showed association with sJIA exceeding the threshold for genome-wide significance, while 23 other novel loci were suggestive of association with sJIA. Using a combination of genetic and statistical approaches, we found no evidence of shared genetic architecture between sJIA and other common JIA subtypes.ConclusionsThe lack of shared genetic risk factors between sJIA and other JIA subtypes supports the hypothesis that sJIA is a unique disease process and argues for a different classification framework. Research to improve sJIA therapy should target its unique genetics and specific pathophysiological pathways.

Résumé :

Systemic juvenile idiopathic arthritis (sJIA) is a rare subtype of juvenile idiopathic arthritis, whose clinical features are systemic fever and rash accompanied by painful joints and inflammation. Even though sJIA has been reported to be an autoinflammatory disorder, its exact pathogenesis remains unclear. In this study, we integrated a meta-analysis with a weighted gene co-expression network analysis (WGCNA) using 5 microarray datasets and an RNA sequencing dataset to understand the interconnection of susceptibility genes for sJIA. Using the integrative analysis, we identified a robust sJIA signature that consisted of 2 co-expressed gene sets comprising 103 up-regulated genes and 25 down-regulated genes in sJIA patients compared with healthy controls. Among the 128 sJIA signature genes, we identified an up-regulated cluster of 11 genes and a down-regulated cluster of 4 genes, which may play key roles in the pathogenesis of sJIA. We then detected 10 bioactive molecules targeting the significant gene clusters as potential novel drug candidates for sJIA using an in silico drug repositioning analysis. These findings suggest that the gene clusters may be potential genetic markers of sJIA and 10 drug candidates can contribute to the development of new therapeutic options for sJIA.

Résumé :

Systemic juvenile idiopathic arthritis (sJIA) is an often severe, potentially life-threatening childhood inflammatory disease, the pathophysiology of which is poorly understood. To determine whether genetic variation within the MHC locus on chromosome 6 influences sJIA susceptibility, we performed an association study of 982 children with sJIA and 8,010 healthy control subjects from nine countries. Using meta-analysis of directly observed and imputed SNP genotypes and imputed classic HLA types, we identified the MHC locus as a bona fide susceptibility locus with effects on sJIA risk that transcended geographically defined strata. The strongest sJIA-associated SNP, rs151043342 [P = 2.8 × 10−17, odds ratio (OR) 2.6 (2.1, 3.3)], was part of a cluster of 482 sJIA-associated SNPs that spanned a 400-kb region and included the class II HLA region. Conditional analysis controlling for the effect of rs151043342 found that rs12722051 independently influenced sJIA risk [P = 1.0 × 10−5, OR 0.7 (0.6, 0.8)]. Meta-analysis of imputed classic HLA-type associations in six study populations of Western European ancestry revealed that HLA-DRB1*11 and its defining amino acid residue, glutamate 58, were strongly associated with sJIA [P = 2.7 × 10−16, OR 2.3 (1.9, 2.8)], as was the HLA-DRB1*11—HLA-DQA1*05—HLA-DQB1*03 haplotype [6.4 × 10−17, OR 2.3 (1.9, 2.9)]. By examining the MHC locus in the largest collection of sJIA patients assembled to date, this study solidifies the relationship between the class II HLA region and sJIA, implicating adaptive immune molecules in the pathogenesis of sJIA.

BackgroundSystemic juvenile idiopathic arthritis (sJIA) is a childhood arthritis with signs of autoinflammation and high risk of macrophage activation syndrome (MAS). Immunopathogenesis of sJIA is based on excessive activation of innate immunity, which is characterized by hyperproduction of pro-inflammatory cytokines - interleukin (IL)-1 and IL-18, which play a key role in induction of synthesis of other pro-inflammatory mediators. Macrophage activation mediates the evidence of hyperferritinemia, which stimulates a pathological immune response with the development of MAS. In clinical practice, the study of the cytokine profile and its comparison with the acute-phase inflammatory response markers in patients (pts) with sJIA can make possible assessment the nature of the course process and predict the possibility of MAS.ObjectivesTo study distinctive features of some biomarkers in pts with sJA in relation with history of MAS.MethodsTotal IL-18, IP-10, IL1RA, IL1b, IL6, CRP and ferritin levels were determined in pts with sJIA and compared between between groups regarding to disease activity and history of MAS. Serum levels of cytokines, ferritin were measured using standard commercial enzyme-linked immuno-sorbent assay. CRP were estimated by commercial nephelometric method.ResultsA total of 39 pts were included to study, the ratio of boys and girls was 1:1.4. Median age of sJIA onset was 4.4 years [2.9; 6.5], age at the time of examination - 11.4 years [7.35; 13.5]. The pts were divided into 3 groups: group I - with MAS (8 pts), group II - with active sJIA without MAS (16 pts), group III - with inactive sJIA (15 pts). Total IL-18 levels were significantly higher in pts with MAS (group I) (median 6036.5 pg/ml [IQR 1932.5; 9160.3], and remained persistently elevated even in the majority of pts with inactive disease (group III) (median 1625.25 pg/ml [IQR 634.1; 2451.6]. CRP was higher in group I (CRP: median 60.6 mg/l [IQR 41.2; 109.4]), than in group II (CRP: median 15.65 mg/l; IQR 10.4; 38.4), without a statistically significant difference (p=0.5), but the level of ferritin was significantly higher in group I (median 893.25 ng/ml [IQR 253.9; 7725.25]), than in group II (median 62.1 ng/ml [IQR 19.8; 211.8], p=0.009). Generally, we observed positive correlation between CRP and ferritine (r=0.629, р˂0.05), CRP and IL-18 (r=0.352, р˂0.05), CRP and IL1RA (r=0.693, р˂0.05); between ferritin and IL-18 (r=0.566, р˂0.05), between ferritin and IL1RA (r=0.545, р˂0.05). The strongest correlation was for ferritin and overall for groups I and II. We didn’t observe significantly difference between IP-10, IL1b, IL6 between groups.ConclusionSerum levels of IL-18 were elevated in the majority of pts with sJIA regardless of activity, but were significantly higher in pts with active disease and, especially, in pts with MAS. Increased level of IL-18 may reflect increased disease activity or development of MAS, that confirms the key role of IL-18 in sJIA and MAS. According to our data, in clinical practice the measure of serum level of ferritin more useful for early detecting MAS in pts with sJIA due to more stronger correlation between the levels of proinflammatory cytokines and ferritin, than between proinflammatory cytokines and CRP. We were unable to identify a statistically significant difference in the level of IL1RA in active sJIA and MAS, despite a significant correlation between the level of CRP and IL1RA, ferritin and IL1RA, which may be associated with small number of pts.Disclosure of InterestsNone declared

ZUSAMMENFASSUNGIm letzten Jahrzehnt hat eine neue Komplikation der systemischen juvenilen Arthritis mehr und mehr Beachtung in Fachkreisen und als „sJIA Lung Disease“ (sJIA-LD) Einzug in die Literatur gefunden. Die Kinder mit sJIA-LD präsentieren sich mit initial oft unspezifischen respiratorischen Symptomen, Hypoxie und Hautausschlag. Ein häufiges eindrückliches erstes Zeichen sind Trommelschlegelfinger mit digitalen Erythemen. Möglicherweise scheint die sJIA-LD gehäuft aufzutreten, wenn Kinder ein junges Alter bei sJIA-Diagnose hatten sowie ein oder mehrere Makrophagen-Aktivierungssyndrome in der Vorgeschichte, hohe Interleukin-18-Spiegel im Serum und eine Unverträglichkeit gegenüber Biologika (IL-1- oder IL-6-Blocker). Die Mortalität variiert stark zwischen den einzelnen Fallsammlungen, in der größten Kohorte ist sie mit 36 % hoch 1.

Systemic juvenile idiopathic arthritis (sJIA) is a severe orphan autoimmune disease characterized by fever, polymorphic skin rash, hepatosplenomegaly, serositis, generalized lymphadenopathy, and destructive arthritis. Proinflammatory cytokines, including interleukin-1 (IL-1), IL-6, IL-17, IL-18, and tumor necrosis factor (TNF), play a key role in sJIA pathogenesis. sJIA is refractory to conventional immunosuppressive antirheumatic drugs. Before the development of biologicals, sJIA patients had to receive corticosteroids constantly. The implementation of biologicals, such as tocilizumab (IL-6 inhibitor) and canakinumab (IL-1 inhibitor) into clinical practice radically changed the disease course, reduced the need for corticosteroids, and significantly decreased the risk of disability. The administration of biologicals at sJIA onset ensured faster achievement of inactive disease or remission compared to conventional therapy, as well as more patients responding to them. Implementation of biologicals into clinical practice dictated the need to identify optimal conditions for initiating therapy in sJIA patients to improve treatment efficacy and safety. Résumé :

Evaluation of the safety profile of vaccines in patients with rheumatic diseases requires an assessment of their impact on disease activity. The effect of antipneumococcal vaccination on the activity of systemic juvenile idiopathic arthritis (sJIA) has not been studied so far. Objective. To evaluate the dynamics of sJIA activity after immunization with pneumococcal 13-valent conjugate vaccine (PCV13) of patients receiving biologicals. Patients and methods. This study included patients with sJIA in remission or active disease receiving biologicals during inpatient treatment and vaccinated with PCV13. To evaluate the effect of immunization on sJIA activity, we measured serum levels of high-sensitivity C-reactive protein (hs-CRP) and calprotectin. In addition to that, we assessed the number of new cases, when concentration of these markers was above the upper limit (UL) 4 weeks after PCV13 administration. Results. In 18 out of 53 patients in remission (34%) and 3 out of 25 patients with active sJIA (12%), hs-CRP levels were undetectable (0.1 mg/L) at both time-points (baseline and after 4 weeks). Among those with detectable hs-CRP levels in at least one time-point 4 weeks after vaccination, patients in remission (n = 35) had no significant changes in hs-CRP (median -0.17 mg/mL; 95% CI -0.84…0.41), whereas patients with active sJIA (n = 22) demonstrated a 3-fold decrease in hs-CRP level (median -0.94 mg/mL (95% CI -3.93…0.05). We observed no significant difference in calprotectin levels in the groups. Concentration of hs-CRP above the UL 4 weeks after vaccination was detected in 2 out of 53 sJIA patients in remission (4%) and none of the patients with active sJIA. Сoncentration of calprotectin above the UL 4 weeks after vaccination was detected in 8 out 53 (15%) and 5 out 25 (20%) sJIA patients in remission and with active disease, respectively. Conclusion. Vaccination against pneumococcal infection in patients with sJIA led to an increase in the level of highly sensitive laboratory markers of sJIA activity in 4–20% of patients. Systemic juvenile idiopathic arthritis (sJIA) is a severe autoinflammatory disorder with a still not clearly defined molecular mechanism. To better understand the disease, we used scattered datasets from public domains and performed a weighted gene coexpression network analysis (WGCNA) to identify key modules and hub genes underlying sJIA pathogenesis. Two gene expression datasets, GSE7753 and GSE13501, were used to construct the WGCNA. Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analyses were applied to the genes and hub genes in the sJIA modules. Cytoscape was used to screen and visualize the hub genes. We further compared the hub genes with the genome-wide association study (GWAS) genes and used a consensus WGCNA to verify that our conclusions were conservative and reproducible across multiple independent datasets. A total of 5,414 genes were obtained for WGCNA, from which highly correlated genes were divided into 17 modules. The red module demonstrated the highest correlation with the sJIA module ( r = 0.8 , p = 3 e − 29 ), whereas the green-yellow module was found to be closely related to the non-sJIA module ( r = 0.62 , p = 1 e − 14 ). Functional enrichment analysis demonstrated that the red module was mostly enriched in the activation of immune responses, infection, nucleosomes, and erythrocytes, and the green-yellow module was mostly enriched in immune responses and inflammation. Additionally, the hub genes in the red module were highly enriched in erythrocyte differentiation, including ALAS2, AHSP, TRIM10, TRIM58, and KLF1. The hub genes from the green-yellow module were mainly associated with immune responses, as exemplified by the genes KLRB1, KLRF1, CD160, and KIRs. We identified sJIA-related modules and several hub genes that might be associated with the development of sJIA. Particularly, the modules may help understand the mechanisms of sJIA, and the hub genes may become biomarkers and therapeutic targets of sJIA in the future.

Objective.We sought to identify specific microRNA (miRNA) for systemic juvenile idiopathic arthritis (sJIA) and to determine the involvement of these miRNA in regulating the expression of cytokines.Methods.Microarray profiling was performed to identify differentially expressed miRNA in sJIA plasma. Levels of candidate miRNA and mRNA were assessed by real-time PCR, and cytokines were measured by ELISA. Dual-luciferase reporter assay was used to validate the direct interaction between miR-26a and interleukin 6 (IL-6).Results.Forty-eight miRNA were differentially expressed in the plasma of patients with sJIA compared with healthy controls (HC). Five miRNA were selected for further validation. The expression level of miR-26a was exclusively elevated in the plasma of patients with sJIA as compared with 4 rheumatic diseases and 2 subtypes of JIA (oligoarticular and polyarticular). The levels of IL-6, IL-1β, and tumor necrosis factor-α in the plasma of patients with sJIA were increased, and only IL-6 presented a positive correlation with miR-26a (r = 0.539, p < 0.0001). After stimulation with IL-6, miR-26a expression was upregulated in THP-1 cells, while the supernatant level of IL-6 was downregulated by transfection of miR-26a mimics. Consistently, direct target relationship between miR-26a and IL-6 was confirmed.Conclusion.This study demonstrates that miR-26a is expressed specifically and highly in sJIA plasma and suggests that miR-26a may regulate the levels of cytokines in sJIA. Our findings highlight miR-26a as a potential biomarker for the diagnosis as well as differential diagnosis of sJIA.

ObjectivesThe International League of Associations for Rheumatology classification criteria define systemic juvenile idiopathic arthritis (SJIA) by the presence of fever, rash and chronic arthritis. Recent initiatives to revise current criteria recognise that a lack of arthritis complicates making the diagnosis early, while later a subgroup of patients develops aggressive joint disease. The proposed biphasic model of SJIA also implies a ‘window of opportunity’ to abrogate the development of chronic arthritis. We aimed to identify novel SJIA biomarkers during different disease phases.MethodsChildren with active SJIA were subgrouped clinically as systemic autoinflammatory disease with fever (SJIAsyst) or polyarticular disease (SJIApoly). A discovery cohort of n=10 patients per SJIA group, plus n=10 with infection, was subjected to unbiased label-free liquid chromatography mass spectrometry (LC-MS/MS) and immunoassay screens. In a separate verification cohort (SJIAsyst, n=45; SJIApoly, n=29; infection, n=32), candidate biomarkers were measured by multiple reaction monitoring MS (MRM-MS) and targeted immunoassays.ResultsSignatures differentiating the two phenotypes of SJIA could be identified. LC-MS/MS in the discovery cohort differentiated SJIAsyst from SJIApoly well, but less effectively from infection. Targeted MRM verified the discovery data and, combined with targeted immunoassays, correctly identified 91% (SJIAsyst vs SJIApoly) and 77% (SJIAsyst vs infection) of all cases.ConclusionsMolecular signatures differentiating two phenotypes of SJIA were identified suggesting shifts in underlying immunological processes in this biphasic disease. Biomarker signatures separating SJIA in its initial autoinflammatory phase from the main differential diagnosis (ie, infection) could aid early-stage diagnostic decisions, while markers of a phenotype switch could inform treat-to-target strategies.

The autoinflammatory syndromes are a group of disorders defined as “unmotivated recurrent inflammatory events” haracterized by an apparently spontaneous manifestations of inflammation, without autoreactive T cells or auto-antibody involvement. These conditions usually present aberrant responses associated to molecular pathogenic patterns and deregulated production of inflammatory cytokines, such as interleukin-1β (IL-1β), interleukin-18 (IL-18) and Tumor Necrosis Factor-α (TNF- α). Systemic juvenile idiopathic arthritis (sJIA) and Adult onset Still’s disease (AOSD) are particular kinds of autoinflammatory disorders with still unknown etiology, mainly associated to the possible pathogenic role of IL-1β, since the efficiency of pharmacological treatments with IL-1β blockers agents. The typical patients’ neutrophilia would presume the PMNs involvement at the onset of these syndromes. For this reason we have analyzed the PMNs activation state at baseline, together with their response to extracellular pro-inflammatory stimuli. In comparison with healthy granulocytes, sJIA and AOSD PMNs didn’t present differences in term of cellular activation state and IL-1β production, and our results also revealed that neither healthy nor sJIA/AOSD PMNs can produce elevated levels of this cytokine, as opposed to monocytes, which can release IL-concentrations more than 10 fold higher than PMNs. We have also analyzed the inflammatory response mediated by monocytes, after activation of three different inflammasome complexes, in sJIA, AOSD patients and patients affected by the hereditary periodic fever syndromes (HPFs) Familial Mediterranean Fever (FMF) and Cryopyrin associated periodic syndrome (CAPS). Monocytes from HPFs affected patients have shown the clear involvement of one specific inflammasome complex. sJIA and AOSD monocytes, instead, have not presented significant differences with healthy monocytes, leaving some doubts about the principal cellular source of IL-1β in these syndromes, also considering previous experiments which indicated that sJIA monocytes released even lower IL-1β concentrations than healthy monocytes. An important difference has been observed about the S100 calcium-binding protein A12 (S100A12) production. Indeed, patients PMNs have produced higher levels of S100A12 after stimulation in comparison with healthy ones. This result could be related to the typical elevated S100A12 serum concentration observed in affected patients. Taken together, our observations indicate that the role of PMNs in sJIA and AOSD is not directly related to the inflammasome activity and IL-1β production, but it deserves further investigations about a possible different action mechanism, involved in the pathogenesis of these syndromes.

Cette thèse a pour objectif de proposer une approche renouvelée des verbes russes en –sja (Vsja), improprement qualifiés de « réfléchis ». Les approches existantes peinent à rendre compte des valeurs multiples et contradictoires que peuvent assumer ces verbes et notamment de celles ne pouvant en aucun cas relever de la réfléxivité stricto sensu.Le modèle proposé s’inscrit dans la théorie des opérations énonciatives (TOE) et unifie toutes les valeurs de Vsja. Tout d’abord, la comparaison de –sja, sebjá et de la particule atone sebe a permis de mieux dessiner les spécificités de Vsja. Puis l’analyse des faits diachroniques a permis de rappeler le lien ancien qui, en slave, unit –sja à la valeur de la voix moyenne. Cependant, –sja, en synchronie, ne peut être considéré comme un opérateur de voix notamment parce que certains emplois de Vsja (kusat’sja, par exemple) n’entraînent aucun changement de voix ou de diathèse. Dans notre modèle, –sja est la trace d’une opération énonciative visant à centraliser une relation – et une seule – entre Vsja et un autre actant de l’énoncé, qui n’est pas forcément le sujet grammatical, mais sans exclure obligatoirement la relation au second actant dans le cas des phrases biactancielles. Celle-ci est seulement périphérisée et, selon le sémantisme de Vsja et des actants en présence, peut/doit être maintenue ou exclue. Et si cette centralisation, qui intervient juste après l’opération d’instanciation, engendre, sur le plan sémantique, des effets de sens de type voix moyenne, ce modèle permet surtout d’envisager Vsja en dehors du système des voix verbales, tout comme il permet de résoudre l’ensemble des contradictions apparentes entre les différentes valeurs de Vsja
The aim of this study is to propose a new approach to –sja verbs in Russian, improperly described as “reflexive”. The existing models fail to account for all the various and sometimes contradictory values assumed by –sja verbs, in particular for those Vsja which cannot be considered as having a “reflexive” meaning.The proposed model lies within the framework of the Théorie des opérations énonciatives (TOE) and unifies all the values of –sja verbs. The comparison between –sja, sebjá and the atonic particle sebe helped to understand the specificity of –sja. Then a short analysis of diachronical facts enabled us to stress the historical link in Slavic between –sja and the middle voice. Nevertheless, the marker cannot be considered synchronically as a “voice operator” notably because certain Vsja (e. g. kusat’sja) do not present a voice or diathesis shift. In our model, –sja is the sign of an enunciative operation whose aim is to centralize the relation between Vsja and one actant only, that is not necessarily the grammatical subject, and resulting in the other relation being simply peripheralized. Such an approach allows to solve all the apparent contradictions within the so-called Russian “reflexive” verbs and allows also to envisage Vsja outside the system of the verbal voices

Den här studien undersöker det ryska verbprefixet v(o)- och cirkumfixet v(o)- -sja i det konkreta fysiska rummet. Syftet med den kontrastiva studien är att undersöka och beskriva betydelser. Tvåspråkig data från en samtida rysk-svensk ordbok analyseras med Krongauz metod. En lista över verbaffixens betydelser byggs upp genom att jämföra lexikala betydelser och morfosyntaktiska konstruktioner för verben i båda språken. Resultatet visar att affixens betydelser kan delas in i följande kategorier: Spatiala rörelser in i ett slutet rum, Spatiala rörelser till en avgränsad yta, Spatiala rörelser mot en närhet, Vidhäftning och Platser i det fysiska rummet.
This present study investigates the Russian verbal prefix v(o)- and circumfix v(o)- -sja in the concrete physical space. The aim of the contrastive study is to explore and describe meanings. Bilingual data, extracted from a contemporary Russian-Swedish dictionary, is analysed by using Krongauz’s method. A list of meanings of the Russian verbal affixes is built by comparing similarities and differences between lexical meanings and morphosyntactic structures for the verbs in both languages. The result shows that the meanings of the affixes can be divided into the following categories: Spatial movements into an enclosed space, Spatial movements onto a delimited surface, Spatial movements towards a vicinity, Adhesion and Locations in physical space.

The aim of this research is to determine whether the national consciousness is a determinant of the educational systems of the Republic of China (Taiwan) and of the Whites in die Republic of South Africa. By making use of the Bereday method results are put alongside each other and compared so as to determine similarities and differences. By studying the literature and arguing national consciousness was found to be the available facts the a determinant of the relative educational systems. The way in which each community expresses its national consciousness in time and space determines the way in which it is reflected in the educational systems. In the Republic of China the core of national consciousness is to be found in the 'Three principles of the people' and with the Whites of the Republic of South Africa it is Christian nationalism. The educational system of the Republic of China is therefor determined by the 'Three principles of the people' and that of the Whites of the Republic of South Africa by Christian nationalism. Research shows that both the national consciousness and the educational system are subject to the dynamics of change. The contemporary world has changing views on the being and substance of national consciousness. In both the Republic of China and the Republic of South Africa there are signs of this. Ongoing research should indicate how this will effect the different educational systems.

In this final bachelor work there is the designed shopping centre "PRISMA", which is foreseen to be constructed in the free plot of Pakruojis.First part consists of brief description of the plan of the plot, building characteristics, its construction solutions and engineering networks. In addition, there are calculationsof coefficients of heat-transferring walls. In the constructions part, there is a designed collectable reinforced floor slab, 12 m and 6 m metallic beams and one chosen reinforced column. In the technological part, there is given a card of reinforced columns estimations. Selection of mechanisms is also descripted. What is more, it consists of requirements of column construction’s quality control and main technical safety. Throughout the drawings there are demand graphs of works and workers. Economical part consists of sheets with quantity of works, and accordingly prepared local estimate of building with sheets of mechanisms and materials.

The importance of small and medium-sized enterprises (SMEs) in the economy should not be overlooked. The main function of SMEs is to contribute to the economic activities in a country, through the provision of goods and services to the public or other firms. These goods and services could be traded internationally, thus increasing a country?s export performance. It is important for economic growth that SMEs grow within their respective economies. One way that SMEs can achieve growth is through internationalisation. Firms are internationalising faster than ever before (because of advances in telecommunications and transportation) and internationalisation theories that can provide practical guidance to firms are more important today than in the past. A firm?s ability to internationalise no longer only depends on the quality of the product, the delivery terms and competitive prices. Internationalisation increasingly depends on the ability and willingness of financial institutions to grant credit. Obtaining trade finance has become a major hindrance to SME internationalisation, especially in Africa. By overcoming the difficulties in obtaining trade finance, African SMEs will be able to expand into foreign markets. The purpose of this study is to determine how African SMEs can overcome trade finance barriers to internationalisation. SMEs can do so by mitigating the risks involved in every international transaction and by becoming ?trade finance ready?. A trade finance facility that is well suited for African SMEs (because it revolves around identifying and mitigating the risks involved with their international transactions) is structured trade and commodity finance. In trading with China, African SMEs can obtain structured trade and commodity finance from a specialist financial institution that focuses on the Chinese market (which is the focus of this study). China Construction Bank (Johannesburg branch), through their association with Rand-Asia Trade Finance, provides structured trade and commodity finance to African SMEs. The key to receiving structured trade and commodity finance is that these SMEs, together with China Construction Bank (Jhb) and Rand-Asia Trade Finance have to mitigate the risks involved with their international transactions so that the SMEs can become ?trade finance ready?.

Die vorliegende Arbeit beschäftigt sich mit der analytischen Modellierung und Optimierung synthetischer Jet-Aktuatoren, welche zur aktiven Strömungsbeeinflussung genutzt werden können. Ein in der Literatur bekanntes eindimensionales Modell wird ausführlich hergeleitet und an gemessene Geschwindigkeitsspektren verschiedener Jet-Aktuatoren angepasst. Der Einfluss jedes Modellparameters wird separat untersucht. Außerdem wird ein empirischer Zusammenhang zwischen Membranresonanzfrequenz und Luftkammervolumen angegeben, mit dessen Hilfe synthetische Jet-Aktuatoren mit größtmöglichen Strömungsgeschwindigkeiten durch die Düse konstruiert werden können.

This thesis proposes to discuss the concept of social order and status of the Sjiq ,according to the negative estimation , such as ‘It’s rotten here’, to judge their own community by the Sjiq in Alang Busi. The discussion begins by defining the local terms, gaya and utux, for that they are the most connected to the concept of order. The core concept of gaya is reproduction. The Sjiq keep their reproductivity by staying ‘inside’, and segregate from the de-reprodutive ‘outside’ owns by utux. by obeying gaya, phrases of rudan(reproductive elders). The contrast between ‘inside’ and ‘outside’ provides them the basis to define the relationships between human and utux, individuals, houses, and groups. If there were any member of ‘inside’ in psaniq (de-reprodutive status), they would try to have dmahul, the animal sacrifice ritual, to cut off the relationship/road/bridge between human and utux, and return to the normal status. Meanwhile, it also connects members who have the common reproduction relationship. During the ritual, it reinforces not only the opposition and heterogeneity between ‘inside’ and ‘outside’, but also the connection and homogeneity among ‘inside’, and reverses the de-reprodutive status to the reprodutive status. Keeping ‘inside’ and ‘outside’ in the proper way of segregation is the concept of order for the Sjiq.

The author discusses the distribution of the reflexive anaphora sebja and the middle voice affix -sja/-s’ in Russian. The Russian data are discussed assuming that in languages where the middle voice morphology can encode reflexivity, anaphors can be canceled in standard object position, where Accusative Case is assigned. This entails that direct object anaphors, when realized, are not in the structural position where they receive Accusative Case; it is argued that they can surface in Focus positions. The final part of the paper is dedicated to the analysis of the formal status of the middle voice affix.

Zakon o revidiranju in zadnja novela ZGD-1K sta vnesla novo definicijo subjektov javnega interesa (SJI) v slovenski pravni prostor. Ta pojem zavzema vse javne delniške družbe in gospodarske družbe iz finančnega področja (banke in zavarovalnice), za katere veljajo bolj stroga pravila računovodskega poročanja in delovanja. Slovenija je evropsko definicijo SJI razširila še na vse gospodarske družbe, v katerih ima država ali lokalna skupnost večinski delež glasovalnih pravic. Že od novele ZGD-1J, morajo SJI v poslovnem poročilu priložiti posebno izjavo o nefinančnem poslovanju, ki vsebuje pomembne podatke o okoljskih, socialnih in kadrovskih zadevah, spoštovanju človekovih pravic ter v zadevah v zvezi z bojem proti korupciji in podkupovanju z obrazložitvami izvajanih politik na tem področju. Epidemija COVID-19 nam je pokazala, da smo ranljivi in kako pomembna je socialna in pravna država ter ustrezen odnos do narave. Zato poslovanje gospodarskih subjektov v in po epidemiji zahteva ponoven premislek o vzdržnem poslovnem modelu, kjer ni na prvem mestu zgolj lukrativni namen družb ustvarjati dobiček na trgu, ampak tudi ustrezen odnos do naravnega in predvsem družbenega okolja. Pričakovati je, da bodo nefinančna poročila razširjena na SJI, predvsem družbe, katerih glasovalne pravice so v državni in lokalni lasti in od njih se pričakuje poslovanje, ki bi upoštevalo ustrezen odnos do naravnega in družbenega okolja. Tako avtorja v prispevku podajata tudi nekatere predloge in primere dobrih praks, za povečanje družbene odgovornosti SJI in s tem zmanjševanje revščine.

Abstract.—Lake Conroe has long been synonymous with the controversy surrounding control of the exotic plant hydrilla Hydrilla verticillata. By the time hydrilla was first identified in Lake Conroe in 1975 (2 years after impoundment), the plant covered 470 acres. By 1979, hydrilla increased to 4,500 acres and was causing problems for boaters, skiers, and swimmers. As a result of efforts by the Lake Conroe Association and its supporters and despite objections by Texas Parks and Wildlife Department (TPWD) staff, the Texas Legislature directed the Texas Agricultural Experiment Station and TPWD to conduct a study to determine the ability of grass carp Ctenopharyngodon idella to control hydrilla and the effects of hydrilla removal on fish populations, the fishery, and the limnology of the reservoir. Between September 1981 and September 1982, 270,000 diploid grass carp were released into Lake Conroe. By October 1983 hydrilla and all other aquatic vegetation had disappeared from the reservoir. Hydrilla growth was suppressed in Lake Conroe for the next 13 years; however, in 1996, approximately 3 acres of hydrilla were discovered. For the next 8 years (1997–2004), herbicide treatments funded primarily by the San Jacinto River Authority (SJRA) and conducted by SJRA and TPWD successfully limited hydrilla expansion. At the same time, native vegetation establishment by TPWD, U.S. Army Engineer Environmental Research and Development Center’s Lewisville Aquatic Ecosystem Research Facility, and several angling groups led to diversification and expansion of the native aquatic plant community in Lake Conroe and provided an alternative to a monoculture of hydrilla for fish habitat. However, by 2005, herbicide treatments alone were unable to control hydrilla expansion. As a result, TPWD and SJRA determined the need to develop a comprehensive hydrilla management plan for Lake Conroe. Therefore, a plan based on the principles of Integrated Pest Management was developed with the cooperation of angler organizations, property owner associations, local businesses, and other user groups. The plan integrated use of ecological, biological, chemical, and mechanical strategies for control of hydrilla while promoting diversity of the aquatic plant community. The stated time line of the plan was to reduce the surface coverage of hydrilla at Lake Conroe to less than 40 acres by spring of 2008, while preserving a diverse aquatic plant community.

Résumé :

Sweeping Jet Actuator (SJA) was introduced as a potential active flow control method for reducing three-dimension (3D) flow separations in a compressor cascade. Unlike some other actuators, SJA needs no valves or moving parts to convert its steady compressed air source into sweeping jets that oscillates from side to side through the millimeter-sized outlet nozzle. The rather simple and small structure makes it possible to place SJA into the blades. In this study, a 3D numerical simulation using unsteady RANS codes was conducted to investigate the effects of SJA on the flow pattern and the aerodynamic loss mechanism in a compressor cascade. Firstly, the reliability of a commercial Computational Fluid Dynamic (CFD) code was validated and the computed results showed good agreements with experimental data from the literature. Secondly, some possible affecting factors, such as actuating pressure, position of SJA exit and jet skew angle, were analyzed and discussed in detail. Moreover, the effectiveness of active flow control under different locations and stream directions of SJA was studied for obtaining a further understanding of the mechanism of SJA for controlling flow separations. In addition, the generation and interaction of internal secondary flows in the compressor cascade were also investigated, and the oscillating jet process of SJA was presented. The numerical results indicate that using SJA delays effectively the corner flow separation, thus decreases the aerodynamic loss of the compressor cascade. For the optimum scheme within the present research, the reduction of overall time-averaged total pressure loss coefficient achieves about 5.6% compared with the original case without SJA. The streamwise position of SJA has a more remarkable influence in improving performance than the other SJA schemes. The considerable improvements of flow separation in the corner region is considered to be one of the main reasons in overall performance increase.

Synthetic jet actuators (SJA) are one of the newly developed actuators that have demonstrated its great potentials in active flow applications, particularly in closed-loop flow controls. The SJA contains a piezoelectric membrane in a cavity, which vibrates and generates a periodic jet at the exit of the cavity through an orifice that is mounted flush with the solid wall of the flow field. In order to design the feedback control laws, it is crucial to be able to quantitatively capture the dynamics of SJA. In this paper, the dynamics of SJAs with six different orifice sizes are experimentally investigated. A synthesis using system identification for the purpose of constructing mathematical models of these zero mass-flux actuators is offered. The experimental study includes two output parameters, the acoustic sound pressure generated by the SJA and the mechanical membrane vibration of the SJA. State-space models for these outputs (sound pressure and mechanical vibration) are developed as a function of orifice size. These results form a foundation for future intelligent design of SJA.

Considering the shortcoming and inability of the conventional piezoelectric synthetic jet actuator (PE-SJA), a novel multi-diaphragm PE-SJA was designed and made in this paper, and some detailed experimental study was carried out upon it. Using the hotwire anemometry, the optimum resonance frequency of the actuator was obtained. Then, under this special frequency, velocity magnitude was measured at different positions along the axis of the orifice. And then the orifice exit velocity of different numbers of diaphragm was measured, analysized and compared, which showed that the mean and peak axis velocity of one diaphragm PE-SJA were only the half of those of the five diaphragms PE-SJA., while the performance of three diaphragms PE-SJA was almost the same as that of the five diaphragms., which meant that although the performance of the PE-SJA increased with the number of the piezoelectric diaphragm, it was not in linearly ways. From the experimental result it was also shown that: For frequency f = 560Hz, the optimum diameter and depth of the orifice was 2.5mm and 2mm, respectively. Therefore, the optimum depth/diameter ratio was 0.8 for the present actuator and frequency. And then the performance of the multi-diaphragm PE-SJA with taper-jet exit orifice was much better than that of the straight orifice. Finally, the PIV was used to measure the structure of vortex of the flowfield.

Synthetic jet actuators have been used for control of flow separation in high performance airfoils for increasing lift and low drag. The synthetic jets have zero net mass flux but impart net momentum to the flow. In this study, LES simulations was used to study the time history of outlet velocity profile and net momentum imparted to the flow by a synthetic jet actuator (SJA). The SJA studied is not symmetric and operates with the aid of a piezoelectric (PZT) ceramic circular plate actuator. A three-dimensional mesh for the computational domain of the SJA and surrounding volume was developed and was used to evaluate the details of the airflow conditions inside the SJA as well as at the outlet. The vibration of the PZT ceramic actuator was used as a boundary condition in the computational model to drive the SJA. Particular attention was given to the time evolution of the SJA outlet velocity profile. The time history of the outlet velocity profile of the synthetic jet was curve-fitted to an empirical equation. The resulting outlet velocity could be used as an inlet velocity for active close loop control of airflow separation around an airfoil at high angle of attack.

Abstract A preliminary 2D numerical investigation of the active control of unsteady cavitation by means of one single synthetic jet actuator (SJA) is presented. The SJA has been applied to hinder the intrinsic instabilities of a cloud cavitating flow of water around a NACA 0015 hydrofoil with an angle of attack of 8° and ambient conditions. It has been placed inside the inception region at a distance of 16% of the chord from the leading edge. Concerning the numerical approach, a Eulerian homogeneous mixture/mass transfer model has been used, in combination with an extended Schnerr-Sauer cavitation model and a Volume of Fluid (VOF) interface tracking method. The synthetic jet has been modeled by means of a user-defined velocity boundary conditions based on a sinusoidal waveform. A sensitivity analysis has been first performed in order to evaluate the influence of the main control parameters, namely the momentum coefficient Cμ, the dimensionless frequency F+ and the jet angle αjet. By combining the cavitating vapor content and the impact on the hydrodynamic performance, the best performing SJA configuration has been retrieved. Then, a deeper analysis of the vapor cavity dynamics and the vorticity field has been conducted in order to understand the modification of the main flow produced by the synthetic jet. The best SJA configuration was observed at Cμ = 0.0002, F+ = 0.309 and αjet = 90°, which led to a reduction of both the average vapor content and the average torsional load in the measure of 34.6% and 17.8% respectively. A reduction of the average pulsation frequency of the pressure upstream confirmed the beneficial effect of the SJA. The analysis of the coupled dynamics between vapor cavity-vorticity and their POD-based modal structures highlighted that the benefit of the SJA lies on preventing the growth of a thick sheet cavity which tends to cause the development of the highly cavitating cloud dynamics after the cavity breakup. This is mainly due to an additional vorticity close to the hydrofoil surface just downstream the SJA, as well as a local pressure modification close the SJA during the blowing stroke.
