Texte intégralBag&rsquo
settlements have been a part of dynamic spatial systems, which seasonally depend on and reciprocally interact with Anatolian towns throughout centuries. This thesis presents the transforming setting of &lsquo
bag&rsquo
settlements related to changing values and meanings through an ontological assessment. Therefore, the thesis assumes that the main values of spatial organization, farmland pattern, ecological formation, settlement character, socio-cultural structure and lifestyle of the inhabitants constitute the unique entity of &lsquo
bag&rsquo
settlements. In time, the pressures of changing socio-economic conditions have destroyed the interaction between Anatolian towns and &lsquo
bag&rsquo
settlements. The thesis explains the changing role of &lsquo
bag&rsquo
settlements, changing and conflicting land uses and the loss of unique &lsquo
bag&rsquo
character in the case of Mugla-Karabaglar. Karabaglar is a &lsquo
bag&rsquo
settlement in the southwestern Turkey, where Mugla town residents live seasonally. It is a third grade natural site, the unique character, the natural and cultural assets and the lifestyle of which must be preserved. However, with transformation of the main values, meanings and practices, Karabaglar could no longer perpetuated its initial existence of being. Karabaglar is significant for two reasons: it has natural and cultural beings that need to be conserved, and as it still goes through a transformation process. The master&rsquo
s thesis of the author evaluated conservation plan of Karabaglar. Differently, this doctoral thesis evaluates the dynamics of the spatio-temporal transformation process in Karabaglar. It presents the changing role and significance of Karabaglar within the town-country continuum. This situation brings forward the conservation problematic of the character, landscape and uniqueness of Karabaglar. Within this respect, the thesis contributes to the literature of &lsquo
bag&rsquo
settlements in terms of defining their being and changing role throughout the history.

Texte intégralOrganization by compartmentalization is a general property of natural systems coordinating biological events in space and time. Over the past three decades, it has been demonstrated that multiple micrometric to nano-metric sub-compartments co-exist at a single membrane level. Such membrane organization seems critical for most all cell bioactivities and therefore critical for development and survival of potentially all living organisms. Plants respond to pathogens by activating highly regulated plasma membrane-bound signalling pathways. Plant plasma membrane (PM) displays a great diversity of compartments, but underlying functions and molecular mechanisms governing such organization are not well understood. To get insight in how and why plant PM is compartmentalized, we choose to study the plant PM nanodomain goup 1 REMORIN during the interaction between N. benthamiana and the Potato Virus X (PVX). Using a multidisciplinary approach we decipher a molecular mechanism involved in defining REMORIN PM domains localization. Making mutants we provide a functional link between REMORIN PM organization at single molecule level, its phosphostatus, regulation of plasmodesmata permeability and PVX cell-to-cell movement restriction. We then provide evidences that during N.benthamiana PVX sensing, PM organization appears critical for the modulation plant defence mechanisms and cell signaling. This study provides a unique mechanistic insight into how tight control of protein spatio-temporal organization at PM level is crucial to confer membrane domains identity and functionality

Texte intégralThis thesis work is devoted to visualization by optical microscopy of thermo- and photo-induced phase transitions, in switchable spin transition single crystals. The study of crystals of the compound [{Fe (NCSe) (py) 2} 2 (m-bpypz)] showed the possibility to control reversibly the dynamics of the HS/LS interface through a photo-thermal effect generated by an irradiation of the whole crystal or using a spatially localized light spot on the crystal surface. The investigations of the effect of the light intensity on the transition temperature have highlighted the importance of the coupling between the crystal and the thermal bath in these experiments. Concomitantly, we developped a reaction diffusion model allowing to describe and iden-tify the relevant physical parameters involved in the control of the movement of HS/LS interface. The obtained results are very encouraging and reproduce the main features of the experimental data. However the origin of the interface orientation observed by the optical microscopy in the crystal of the compound [{Fe (NCSe) (py) 2} 2 (m-bpypz)] re-mained mysterious, and needed an elastic approach to be handled. At this end, an electro-elastic model including the volume change at the spin transition was developed. By taking into account for the anisotropy of the unit cell deformation at the transition, we were able to reproduce quantitatively the experimental HS/LS interface orientation. This result confirms the crucial role of the lattice symmetry and its elastic properties in the emergence of a stable interface orientation. The last part of the thesis is devoted to the investigation of photo-induced effects at very low temperatures (~10K). There, we visualized for the first time the real time transformation of a single crystal under LIESST (Light Induced Excited Spin State Trapping) effect as well as its subsequent relaxation at higher temperatures. We have also studied the light induced instabilities through investigation on the LITH (Light Induced Thermal Hysteresis) loops. Around the latter, a fascinating world made of nonlinear effects, and patterns formation emerged, recalled the well known Turing structures. These results lead to new horizons that will give access to new theories and original experimental observations that will enrich the topics opening the new avenues to study of nonlinear phenomena in spin crossover solids
