There is a direct relationship between the development of colon cancer and exogenous factors, such as diet. The immunomodulatory effect of probiotic bacteria has been postulated earlier; however, due to low clinical efficacy and associated limitations they have not been devised as cancer theraphy and preventative procedures. One of the main obstacles is the efficient delivery of live bacterial cells to targeted sites. The work contained in this dissertation aims to develop and evaluate a yogurt based formulation containing microencapsulated live probiotic bacterial cells for use in colon cancer prevention and theraphy. Alginate – poly-L-lysine – alginate (APA) and alginate-chitosan (AC) microcapsule systems have been optimized, prepared and characterized. Survival of Lactobacillus acidophilus cells and mechanical stability of the microcapsules were evaluated in various pH conditions in presence of yogurt and simulated gastrointestinal fluids. The physical properties of the microcapsules were investigated using a computer controlled dynamic simulated human gastrointestinal (GI) model. Results show that the APA microcapsule system was more robust, had higher retention capacity, preserved better bacterial viability and had greater resistance to GI environment. The ApcMin mouse was used to investigate the potential of a microencapsulated probiotic yogurt formulation for clinical efficacy and safety. As ApcMin mice are genetically predisposed to spontaneously develop multiple intestinal and colonic adenomas, they provide a practical and genetically relevant model system for cancer prevention studies. Le cancer colorectal est l'une des malignités les plus répandues dans le monde occidental. La majorité des cancers colorectaux sont soit localement ou régionalement invasifs au moment du diagnostic, limitant les options de traitements et réduisant les taux de survies. Plusieurs études épidémiologiques et laboratoires suggèrent une relation entre le développent du cancer du colon et des facteurs exogènes, tel que le régime alimentaire. L'effet immunomodulateur des bactéries probiotiques a été postulé; cependant, en raison de l'efficacité clinique réduite et d'autres limitations qui y sont associées, elles n'ont pas été conçues comme une thérapie préventive du cancer. L'un des plus grands obstacles reste l'administration efficace des bactéries vivantes aux organes ciblés. Le travail décrit dans cette dissertation vise à développer et à évaluer une formulation à base de yogourt contenant des cellules bactériennes probiotiques vivantes pour l'atténuation de l'inflammation gastro-intestinale qui cause le développement du cancer du colon, par la conduite d'études in vitro et in vivo. L'incidence des cancers colorectaux est très élevée en France. La mise en place d'un micro-environnement favorable à l'intégrité de l'épithélium colique par la nutrition est une stratégie de prévention. Des fructo-oligosaccharides à chaînes courtes (ccFOS) (fibres prébiotiques et butyrogènes) réduisent spécifiquement dans le côlon l'incidence des tumeurs dans un modèle génétique murin de cancérogenèse intestinale (mutation du gène Apc: Adenomatous Polyposis Coli). La fonctionnalité du système immunitaire local est indispensable à cet effet. Le mécanisme de cette action serait lié à la production de butyrate par fermentation des ccFOS et à l'action immunostimulatrice de la flore. Le butyrate régule l'expression de gènes susceptibles d'interférer avec les processus d'homéostasie cellulaire (prolifération, apoptose, interactions lympho-épithéliales) et tissulaire (migration). Pour étudier l'interférence des ccFOS avec le processus de cancérisation nous avons établi des lignées de cellules épithéliales coliques porteuses ou non de la mutation Min du gène Apc. Les capacités migratoires des cellules Apc +/Min sont diminuées, mais restaurées par le butyrate avec une réorganisation du cytosquelette d'actine. Les cellules Apc +/Min sont plus sensibles à l'apoptose et à l'arrêt du cycle cellulaire en G1 induis par le butyrate. In vivo, les ccFOS modulent l'expression des ARNm de l'IL-15 dans la muqueuse colique et celle du RaIL-15 à la surface des lymphocytes intra-épithéliaux (LIE). La cytokine IL-15 régule de nombreuses fonctions lymphocytaires (prolifération, cytotoxicité, survie). Des cocultures ont montré une augmentation de l'expression de ICAM dans le contexte cumulé Apc +/Min et des interactions avec les LIE. Cette molécule a des fonctions immunologiques et de remodelage tissulaire. Ce travail a permis de dégager des pistes sur les mécanismes (stimulation du système immunitaire local, régulation de l'homéostasie cellulaire et tissulaire) et des marqueurs et/ou effecteurs biologiques (ICAM-1, RTNFa, IL-15, RaIL-15) des effets des ccFOS sur la cancérogenèse colique et peuvent rendre compte des effets préventifs observés dans le modèle Min. Ces résultats peuvent mieux définir, documenter et cibler les actions en prévention primaire des cancers colorectaux
Colorectal cancer is one of the most common cancers in France. A beneficial micro-environment provided by the nutrition could participate in the maintenance of the colic epithelial integrity and as such, act as a possible strategy in the prevention of colon cancer. Short-chain fructo-oligosaccharides (sc-FOS) (prebiotic and butyrogenic fiber) decrease specifically the incidence of tumors in a mouse genetic model of intestinal carcinogenesis (mutation of Apc gene: Adenomatous Polyposis Coli). A local, functional immune system is required in this effect. The mechanism of this action could be linked to butyric production by fermentation of scFOS and to the immuno-stimulation property of the flora. Butyrate regulates gene expression and could interfere with cell homeostasis (proliferation, apoptosis, lympho-epithelial interaction) and tissue homeostasis (motility). To study the interference of scFOS with cancerogenesis process, we have established colic epithelial cell lines containing the wild type or mutated Apc gene. The motility of Apc +/Min cells are decreased, however the presence of butyrate can restore the rate of motility in these cells probably through a reorganization of the actin cytosekeleton. The Apc +/Min cells are more susceptible to apoptosis and to cell cycle arrest (in G1) with butyrate. In vivo, scFOS, modulates the mRNA expression of IL-15 in colic mucosa as well as surface expression of IL-15Ra in intra-epithelial lymphocytes (IEL). IL-15 regulate multiple lymphocyte functions (proliferation, cytotoxicity, survival). In our cellular model, we have established a co-culture system which shows an increase expression of ICAM-1 specifically in the presence of the Apc gene mutation and an interaction between epithelial cells and IEL. This molecule have immunological functions and acts in tissue remodelling processes. This research has opened up interesting possible pathways implicated in the mechanisms of action of scFOS on colorectal carcinogenesis in Min mouse (stimulation of local immune system, regulation of cellular and tissular homeostasis), and biological markers and/or effectors (ICAM-1,RTNFa, IL-15, IL-15Ra). This could open up into a new strategy in colorectal cancer primary prevention

Les amines aromatiques hétérocycliques (AAHs) sont formées après cuisson des aliments : viandes rouges et poissons. Ces composés mutagènes et cancérigènes sont impliqués dans la carcinogenèse colorectale mais le rôle des cellules intestinales dans la bioactivation des AAHs n'est pas clairement établi. Une mutation du gène Adenomatous polyposis coli (Apc) apparaît précocement dans les cancers coliques. Les cellules coliques Apc+/+ et ApcMultiple intestinal neoplasia (Min)/+ constituent un modèle pertinent pour l'étude de la promotion tumorale. Les études qui sont présentées ont permis d'étudier les capacités de bioactivation métabolique de ces cellules vis-à-vis de 3 AAHs, PhIP, MeIQx et IQ. En condition d'induction par le 2,3,7,8-tétrachlorodibenzo-p-dioxine (TCDD), les cellules ApcMin/+ produisent des taux plus élevés de métabolites réactifs. Eu égard à la cancérogénicité des AHHs en lien avec la formation d'adduits à l'ADN, ces cellules mutées sont rendues " métaboliquement compétentes ". Par RT-qPCR, nous avons montré qu'après induction par le TCDD les cellules ApcMin/+ présentent des niveaux d'expression des gènes CYP1A1, CYP1A2 et CYP1B1 plus élevés que les cellules Apc+/+. Ainsi, in situ, la bioactivation des AAHs peut être à l'origine de nouvelles mutations. Après transfection des cellules Apc par la forme humaine du CYP1A2, les niveaux d'ARNm, de protéines et les activités enzymatiques mesurées sont plus importants dans les cellules ApcMin/+. Ces résultats indiquent que le gène Apc est impliqué dans la régulation de l'expression des cytochromes P450 (CYPs). Un lien existerait entre le récepteur Ah, principal acteur de la régulation des CYPs de la famille 1, et Apc, acteur de la voie de signalisation Wnt/ß-caténine. Heterocyclic aromatic amines (HAA) are formed in cooked red meat and fish and are considered as potent promoting dietary factors of colon carcinogenesis. Role of intestine cells in HAA bioactivation is not yet fully explained. Loss of function of the Adenomatous polyposis coli (APC) gene product is an early and frequent event in human colorectal carcinogenesis. Normal (Apc+/+) and premalignant (ApcMultiple intestinal neoplasia (Min)/+) mouse colonic epithelial cells allow to study carcinogenesis promotion, but were not characterized for bioactivation of HAA. The overall goal of this work was to characterize the metabolic bioactivation potential towards three HAA, i. E. PhIP, MeIQx and IQ in these colonic cell lines. ApcMin/+ cells when induced by 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) produce significantly higher amount of HAA reactive metabolites than Apc+/+ cells. Parallel RT-qPCR experiments demonstrate that induction by TCDD has prevailing effects in gene expression of CYP1A1, CYP1A2 and CYP1B1 in ApcMin/+ cells. So, a more important metabolic HAA bioactivation potential measured in ApcMin/+ cells can be linked to a higher probability to generate in situ new mutations. After transfection of Apc+/+ and ApcMin/+ cells with human CYP1A2 gene, we have shown that mRNA and protein levels, and also a CYP1A2-related activity were increased in mutated cells. These results indicate that mutation of Apc gene leads to a co-ordinated dysregulation of CYPs expression in mouse colon epithelial cells. A possible link between the aryl hydrocarbon receptor, a main actor in the regulation of CYP1 family expression, with the Apc gene, a main actor of the Wnt/ß-catenin pathway may exist. This disrupted expression of CYP1 members in connection with the mutation of Apc may be of great importance in colorectal carcinogenesis

The aim of this study was to investigate a productivity measurement at the South African Minting Company and evaluate the relationship between productivity and quality. Special emphasis was given to profit-linked total factor model as the tool for measurement. This was encouraged by their ability to separate productivity, profitability and price recovery. Three models were selected and evaluated. These models American Productivity Center (APC) Model, “Profitability = productivity + price recovery” (PPP) model and multi-factor productivity measurement model (MFPMM). APC model was selected as the suitable model because of its simplicity, easy to set up, its ability to produce both financial and non financial data, and allow for route cause analysis with expert system, and more insight for the manager with Microsoft Excels’ What if analysis “Goal seek”. APC model was set up for four periods, from 1 April 2004 to 30 September 2007. The overall profitability results of the circulation coins profit center show an overall positive contribution. There was a break-even of the price recovery for 2006 financial year (period 2). In 2007 financial year (period 3), there was a negative contribution, and this improved to almost break-even in the six month period during this 2008 financial year (period 4). This means there was much more inflation on input resources and the recovery was not fully realised in the price of goods sold. Individual input costs show that the negative price recovery is culminating from material, labour and energy costs contributions. There is a plausible explanation for material and labour, but not for energy. The metal volatility is the underlying cause of the price variation. Labour variation was a company strategy to adjust employee to higher percentiles. Productivity was always positive with the highest contribution in the current financial year (period 4). This means that the profitability at SA Mint has been driven by productivity in the past two financial years. iv Survey of the questionnaire shows average scores for productivity and quality. It is noteworthy, that the lowest mean score for productivity is for the statement “Products are produced in error-free process”. This is a productivity quality measure. In addition, the same variable shows r2 value of 0.42. A conclusion is that even though productivity and quality are highly correlated and show a highly positive relationship, there is a concern on quality in the company. A link can be made that low price recovery becomes more difficult when the quality is not always good. Defective product is a cost, because the product does not reach the customer and if the product is reworked it is still a cost, though low, but more importantly it decreases the available capacity. This study was successful in setting up APC model and producing data that is worthy to the company and academic world. Finally, this study was successful in its quest to establish the relationship between productivity and quality.

Denna rapport avhandlar ett arbete inom ämnet datavetenskap och handlarom utvecklingen av ett system för att studera restidkvoten för sträckor somtrafikeras av Västtrafik, som är en av Sveriges största regionalakollektivtrafikmyndigheter. Restidskvoten anges som förhållandet mellanrestiden med kollektivtrafik och att resa med personbil ([restidkollektivtrafik] / [restid biltrafik]). Med detta som grund har ett system föratt inhämta data om restider med hjälp av Google Maps Directions API samtberäkna restidskvoter och annan metadata utifrån denna insamlade dataskapats. Systemet används till att göra dagliga körningar och lagrar inhämtaddata i en databas hos Västtrafik. Till systemet finns också en dashboard somkan användas för att göra sökningar i databasen som byggts upp och på såsätt hämta, visualisera och analysera önskad data. Restidsdata förkollektivtrafik som erhållits från Google har i stickprov jämförts medrestidsdata från Västtrafik och även om det fanns några avvikelser så stämdede väl överens. Därav dras slutsatsen att data från Google går att använda tillatt beräkna och analysera restidskvoter.

Abstract (sommario):

Several studies have been conducted that link poor control of hemoglobin A1c (HbA1c) to an increased risk of heart disease. However, there are limited published studies that link HbA1c and heart disease based on ethnicity and gender. To address this gap in literature, the purpose of this study was to assess the association between HbA1c and heart disease in African American males (aged 30-64 years old) while controlling for education, income, and access to care. The research questions were focused on establishing an association between HbA1c values and heart disease in African American men through the lens of the health belief model. Secondary data (N=243) were used from the Nutritional Health and Examination Survey (2011-2016) and analyzed. Chi-square analysis and multiple logistic regression were conducted to test for an association between HbA1c and heart disease in men aged 30-64 years old. The variables of Hba1c values, various forms of heart disease, and stroke were tested while controlling for age, education, income, and access to care. Key results indicated no association between HbA1c and heart disease; yet, it is recommended that future research on the topic should include a larger sample of those with heart disease, from other sources, to better assess the outcome. The positive social change implications include the addition of research related to gender-specific outcomes and ethnicity-related risk between HbA1c and heart disease, which can be used to achieve better disease management and provide educational opportunities for diabetic African-American men.

Motivation Hypertension and type 2 diabetes mellitus are common in the black population of South Africa. The literature also shows that elevated blood glucose concentrations can lead to an increase in blood pressure and a blunted decline in nocturnal blood pressure. Therefore, the motivation for this study was to determine if blood glucose may play a role regarding the blunted nocturnal decline in blood pressure in African and Caucasian men. Aim The aim of this study was to investigate the relationship between a blunted nocturnal decline in blood pressure and blood glucose in African and Caucasian men. Methodology A comparative population study was preformed that consisted of 202 school teachers (101 African and 101 Caucasian) between the ages of 25–60 years from the North West Province, South Africa. Subjects were excluded if their body temperature was elevated, had a dependence or abuse of psychotropic substances, were regular blood donors and/or vaccinated in the previous three months. Ambulatory systolic (SBP) and diastolic blood pressure (DBP) were measured. Blood samples from the antebrachial vein were collected in sodium fluoride tubes to determine the serum glucose level and glycosylated hemoglobin A1c (HbA1c) percentage. Estimated average glucose (eAG) was determined from the percentage HbA1c by means of a regression formula. Means and proportions were compared by standard t–test and the chi–square test, respectively. Pearson correlations were used to determine unadjusted associations and multiple regression analysis to determine adjusted associations between variables. Results and Conclusion African men had an elevated HbA1c (p<0.001), eAG (p<0.001), nighttime SBP (p<0.001) and DBP (p<0.001). These results remained similar when non–dipping African and Caucasian men were compared. The Africans also smoked more (p=0.012), consumed more alcohol (p=0.049), had a higher percentage of non–dippers (p=0.054), HIV infected subjects (p<0.001) and a larger number of subjects that used anti–hypertensive medication (p=0.049). The unadjusted analysis showed positive correlations between all the blood pressure measurements and serum glucose, HbA1c and eAG in the African non–dipper men. While in the non–dipper Caucasian men, only daytime SBP and nighttime SBP (22:00–06:00) correlated positively with serum glucose, HbA1c and eAG. Furthermore, when viewing the relationship between carotid intima–media thickness (CIMT) and the blood pressure measurements in the African population, only nighttime (00:00– 04:00) SBP (r=0.581, p<0.001) and DBP (r=0.566, p<0.001) showed positive associations. After adjustments were made for age and body mass index the associations between the various blood pressure measurements and blood glucose disappeared in the non–dipper Caucasian men. However, in the non–dipper African men both nighttime (22:00–06:00) SBP and (00:00– 04:00) SBP showed positive correlations with serum glucose, HbA1c and eAG. After full adjustments (age, BMI, smoking, alcohol intake, physical activity, C–reactive protein and baroreceptor sensitivity) were made, nighttime (00:00–04:00) SBP was the only measure of blood pressure that correlated positively with HbA1c (p=0.069) and eAG (p<0.001) in the nondipper African men. No significant relationships were found for Caucasian men. Furthermore, to determine if the association between nighttime (00:00–04:00) SBP and eAG were independent of CIMT, we adjusted for CIMT. By doing so the positive association between SBP and eAG remained significant in the non–dipper African men (R2=0.617; =0.438; p=0.008) and nonsignificant in the non–dipper Caucasian men (R2=0.423; =0.169; p=0.33). However, the relationship between CIMT and eAG disappeared when we adjusted for SBP, suggesting that the SBP and eAG relationship drives CIMT. In conclusion, the association between the early morning SBP (00:00–04:00) and the blood glucose in non–dipping African men suggests that the blunted decline in nocturnal blood pressure during the early morning hours is associated with chronically elevated blood glucose.

The purpose of this research was to evaluate the Capuchin API launched by Sony Ericsson at Lund, Sweden in 2008. The Capuchin API bridges Adobe’s Flash graphics and effects with JSR support from Java ME. We evaluated Capuchin API with regard to its suitability for a Mobile TV application. We tested this API in Ericsson’s TV lab where we had access to live TV streams and online multimedia resources by implementing a Mobile TV client. This test application was named “Min TV”, in English: “My TV”. Using Capuchin in the Ericsson TV lab environment has shown that it has some benefits, but also has many drawbacks. The Flash developer can be used to create an animated user interface and Java developers can do complex programming. At this early stage Capuchin technology is not mature enough, nor is it suitable for Mobile TV client development. Only after Sony Ericsson adds features such as soft keys, easier debugging of Flash Lite standalone applications, test emulator support in the software development kit, and more data communication methods than string and number, only then it will be a suitable technology for Mobile TV applications. Ericsson’s current Mobile TV application client was built using a framework called ECAF, which supports a graphics frontend and Java ME as backend. We compared ECAF and Min TV with respect to parameters such as: flexibility, performance, memory footprint, code size, and cost of skinning. (All these parameters are explained in detail in the methodology chapter.) As a possible future technology for Mobile TV, we evaluated a number of different presentation/graphics technologies including HECL, SVG Tiny, MIDP 3.0, .NET Compact Framework, etc. Moreover, we examed if a pure Flash Lite client application is a viable solution for Mobile TV. The comparison of different presentation technologies showed that Java ME is a comprehensive platform for mobile development offering all the necessary support from third party graphical user interface makers. .NET CF also looks like a good option for development with the scaled down capabilities for different programming languages supported using CLR.
Syftet med denna forskning var att utvärdera Capuchin API lanserades av Sony Ericsson i Lund, Sverige 2008. Den Capuchin API broar Adobe Flash grafik och effekter med JSR stöd från Java ME. Vi utvärderade Capuchin API med avseende på dess lämplighet för ett mobil-tv ansökan. Vi testade detta API i Ericssons TV lab där vi hade tillgång till TV-strömmar och online multimediaresurser genom en mobil-TV-klient. Detta test ansökan hette "Min TV", på engelska: "My TV". Använda Capuchin i Ericsson TV lab miljö har visat att det har vissa fördelar, men också många nackdelar. Autonomous Driving is the main trend that automotive industry is following. Starting from the first driver assistance systems, nowadays car are growing and growing in complexity, becoming a four-wheeled supercomputer that is able to perform a lot of driving actions totally autonomously. This thesis aims to describe what are the functionalities and methodologies that contributes to this amazing progress. The entire activity is based on a tool for automatic scenario generation. In the first two chapters the context in which the tool is placed is introduced, highlighting the method by which is possible to design its input. Subsequently, the most important aspects of the tool are described, focusing on the graphical interface and the standard it must be compliant with. Two entire chapters are devoted to the analysis of fundamental testing methodologies applied to the tool, while in the last part of the thesis, the final results are presented and discussed.

Cette thèse est une contribution à la connaissance des impacts, quantitatifs et qualitatifs, liés à la déprise minière de la région d'Alès. Le travail a consisté en l'analyse et l'interprétation de données quantitatives et qualitatives des eaux d'émergences des anciennes mines de charbon du bassin alésien afin de connaître la composition chimique des eaux d'émergences et de l'expliquer. Pour cela, nous avons utilisé des Analyses en Composantes Principales (ACP), des diagrammes de Piper et de Schoeller-Berkalov sur l'ensemble des émergences. Dans un deuxième temps, nous avons réalisé une analyse de l'évolution temporelle des concentrations de chaque élément pour les émergences où les données sont disponibles. L'aspect quantitatif est étudié grâce à une synthèse des connaissances disponible sur le réservoir minier de Fontanes (géologie, répartition spatiale, données de pompage et de niveaux piézométriques).
Certaines émergences présentent une qualité qui rend impossible leur rejet direct dans l'environnement et il est important dans cette situation de bien définir la masse d'eau concernée. Pour cela, nous avons élaboré un modèle hydrodynamique par automates séquentiels. Il est appliqué aux anciens travaux de Rochebelle-St-Martin, aboutissant à la détermination des paramètres perméabilité et épaisseur du réservoir par reconstitution du niveau piézométrique du réservoir tout en tenant compte d'un pompage encore maintenu dans l'exploitation. Cette modélisation permet de mieux connaître les paramètres définissant l'aquifère minier dans la perspective d'exploiter cette réserve. Ces eaux de mines présentent une qualité médiocre. A partir d'une meilleure connaissance hydrodynamique du système, obtenue par la modélisation, il devient possible de proposer une valorisation de cette eau par utilisation de ses calories en géothermie connaissant la géométrie du réservoir minier. Les anciennes exploitations minières sont à nouveau source d'énergie.

Racism in elite football has been a topic that has been discussed in newspaper articles and social media. On the other hand, racism in the lower divisions has not been as well received. During two football matches that took place in the lower divisions, there were racist incidents where football players were downplayed about, among other things, their skin color and ethnicity. These two incidents aroused great interest in studying this phenomenon further. The purpose of the study is to gain knowledge of the experiences of abuse in men's football in the lower divisions. We have investigated how players from senior football experience degrading comments from the opposing team, players or supporters. We also investigated whether degrading comments have had an impact on the interviewees' ambitions to continue playing football. In order to answer our questions, we have conducted 10 qualitative interviews. The result shows that there is insults of supporters and opponents in the lower divisions. But something that surprised us was also that there is an insult from fellow players. Insults from opponents, supporters and fellow players occur through the depreciation of skin color and ethnicity. The result also shows that the insult has had a major impact on the interview participants, which has led to some interviewees thinking about quitting playing football.

The expansion of the internet over recent years has resulted in an increase in digital attacks on computers. Most attacks, including the more dangerous ones, directly target program vulnerabilities. The increase in attacks has prompted a need to develop new ways to classify, detect, and avoid vulnerabilities. The effectiveness of these goals relies on the development of new methods and tools that facilitate the process of detecting vulnerabilities and exploits. This thesis presents the development of a tool that provides a visual representation of main memory for the purpose of security analysis. The tool provides new insight into memory utilization by software; users are able to see memory utilization as execution time progression, visually distinguish between memory behaviors (allocations, writes, etc), and visually observe special relationships between memory locations. The insight enables users to search for visual evidence that software is vulnerable, violated, or utilizing memory incorrectly. The development process for our visual tool has three stages: (1) identifying the memory utilization policies of the Windows 32-bit operating system; (2) identifying the data required for visual representations of memory and then implementing one possible method to capture the data; and (3) enumerating and implementing requirements for a memory tool that generates visual representations of memory for the purpose of vulnerability and exploit analysis.

Esta pesquisa tem por finalidade investigar teologicamente a proposta salvífica da APAC – Associação de Proteção e Assistência aos Condenados. A partir da observação diacrônica das funções antropológicas da pena, perpassando o contexto prisional brasileiro, apresenta o específico objeto material da tese, a APAC, em sua atuação no Brasil desde 1974. A realidade carcerária no Brasil necessita de efetiva e radical transformação. Este assunto tem sido estudado por diversas ciências, cabendo à teologia também pronunciar sua palavra específica. Por isso, em seguida, a tese percorre diacrônica e sincronicamente os conceitos de salvação cristã, central para toda a teologia, procurando identificar algumas de suas características irrenunciáveis. Conta, para isso, com a contribuição de alguns teólogos como Karl Rahner, Jürgen Moltmann, Joseph Moingt, Walter Kasper, dentre outros, que ajudarão a rever os conceitos salvíficos imprescindíveis para fins desta tese. A partir do confronto entre a proposta carcerária da APAC e o conceito de salvação cristã, a tese busca responder à questão sobre a viabilidade ou não da pedagogia da APAC, bem como indicar algumas questões mais amplas para a realidade carcerária brasileira e para a presença das igrejas cristãs no mundo carcerário.
This research intends to investigate theologically the salvific proposal of APAC - Association of Protection and Assistance to the Convicted. From the diachronic observation of the anthropological functions of the pen through the Brazilian prison context, it presents the specific material object of the thesis, APAC, in its action in Brazil since 1974. The prison situation in Brazil needs an effective and radical transformation. This subject has been studied by several sciences, and it is up to theology to pronounce its specific word. For this reason, this thesis studies the concept of Christian salvation, a central concept to all theology, trying to identify some of its inalienable characteristics. It counts on the contribution of some theologians such as Karl Rahner, Jurgen Moltmann, Joseph Moingt and Walter Kasper, among others, who will help to review the essential contents of Cristian salvation salvific, so important for the purpose of this thesis. At the end, it answers the question about the viability or not of APAC pedagogy, as well as indicates some broader issues for the Brazilian prison situation and for the presence of the Christian churches in the prisoners world.

Les gestes de la main représentent un moyen naturel et intuitif de communication chez l'homme lui permettant d'interagir avec son environnement dans la vie de tous les jours. Ils permettent notamment de ponctuer et de renforcer l'expression orale d'un dialogue entre personnes. Outre la communication entre individus, les gestes de la main permettent de manipuler des objets ou encore d'interagir avec des machines. Avec le développement de la vision par ordinateur, on assiste à un véritable engouement pour de nouveaux types d'interactions qui exploitent le mouvement de la main et qui passent par une étape d'analyse et de reconnaissance du mouvement afin d'aboutir à l'interprétation des gestes de la main. La réalisation d'un tel objectif ouvre un large champ d'applications. C'est dans ce cadre que se positionne le travail réalisé au cours de cette thèse. Les objectifs visés étaient de proposer des méthodes pour: 1) permettre le transfert d'animation depuis une séquence réelle vers un modèle 3D représentant la main. Dans une telle perspective, le suivi permet d'estimer les différents paramètres correspondant aux degrés de liberté de la main. 2) identifier les gestes de la main en utilisant une base de gestes prédéfinie dans le but de proposer des modes d'interactions basés sur la vision par ordinateur. Sur le plan technique, nous nous sommes intéressés à deux types d’approches : le premier utilise un modèle 3D de la main et le deuxième fait appel à une base de gestes
Hand gestures take a fundamental role in inter-human daily communication. Their use has become an important part of human-computer interaction in the two last decades. Building a fast and effective vision-based hand motion tracker is challenging. This is due to the high dimensionality of the pose space, the ambiguities due to occlusion, the lack of visible surface texture and the significant appearance variations due to shading. In this thesis we are interested in two approaches for monocular hand tracking. In the first one, a parametric hand model is used. The hand motion tracking is first formulated as an optimization task, where a dissimilarity function between the projection of the hand model under articulated motion and the observed image features, is to be minimized. A two-step iterative algorithm is then proposed to minimize this dissimilarity function. We propose two dissimilarity functions to be minimized. We propose also in this thesis a data-driven method to track hand gestures and animate 3D hand model. To achieve the tracking, the presented method exploits a database of hand gestures represented as 3D point clouds. In order to track a large number of hand poses with a database as small as possible we classify the hand gestures using a Principal Component Analysis (PCA). Applied to each point cloud, the PCA produces a new representation of the hand pose independent of the position and orientation in the 3D space. To explore the database in a fast and efficient way, we use a comparison function based on 3D distance transform. Experimental results on synthetic and real data demonstrate the potentials of ours methods

This study shows the importance of leading toward missional change within BUSA in post-1994 contexts incorporating global, afro-centric missional perspectives as South African Baptists. As my research will indicate, BUSA is at a crucial junction, and I contend that BUSA’s main problem is primarily a missiological problem, with ecclesiastical challenges that urgently need to be addressed by BUSA’s leaders. The importance of BUSA’s critical self- reflection and analysis is paramount. Does BUSA and her mission have a future, or will it fade into obscurity? Utilizing the South African Baptist faith heritage as an important interlocutor with a view to retrospective and prospective Baptist ecclesiology in post- 1994 South African society. Special recognition is given to the contribution of emerging Afro-centric missional voices within the current South African/African context. In doing so, this study seeks to be leadership-oriented, biblically-based and Afro-centric in its approach to missional change with South African Baptist Union churches. Toward missional leadership within BUSA churches this study:

1. Uncovers the importance of an Afro-centric missional ecclesiology, taking into consideration both local and global trends and conversations.
2. Uncover a retrospective view of Baptist Ecclesiology within the Southern African context, with its relevance to the history and present-day context of ministry within BUSA.
3. Analyses existing statistical data found within the Baptist Annual Handbook, to establish denominational trends since 1994, with a view to identifying significant ministry trends operable within BUSA.

Analysing and interpreting the history of Baptists in South Africa with reference to BUSA, BCSA, SABMS and the ABK through a ‘missional lens’
Critically examining current trends visible within BUSA churches since 1994 to the present-day.
Undertaking qualitative research to identify phenomenology of people’s shared experience in BUSA

Em nossa pesquisa foi possível constatar que a indústria montadora situada na região do ABC paulista, aqui analisada, a partir de 1992 promoveu intenso processo de mudanças na sua estrutura produtiva, resultando na emergência de um segmento operário jovem-adulto flexível de novo perfil histórico-social e profissional. Current research analyzes the car-building industry in the greater São Paulo region as from 1992 when a deep modification process in its productive structure occurred. The above-mentioned process caused the emergence of a labor segment, or rather, the flexible young man / adult, featuring a new historical, social and professional profile. Actually it is a new set of ‘collaborators’ within the 15-35-year-old bracket, whose just-in-time life style, proper of the metalworking section hailing from production restructuring, is related to the bonding strategies of worker subjectivity. This fact is brought about by the personnel’s new management/coercion styles which looks beyond the factory environment and reveals the new life style of capital’s productive and social metabolism in a period of flexible labor

Abstract (sommario):

Folate and methionine are critical for one-carbon metabolism, impacting DNA synthesis, repair, and methylation processes, as well as polyamine synthesis. These micronutrients have been implicated in colorectal cancer risk. The aim of this thesis was to examine in greater detail the role of folate and methionine in colon cancer initiation and progression by assessing DNA stability and tumour incidence. Studies were performed in vitro (using human colorectal adenocarcinoma HT29 cell line) and in vivo (using ApcMin/+ [Min/+ superscript] mouse model). The in vitro studies examining the effects of various folic acid and methionine concentrations within the physiological range on cell proliferation and genomic instability of HT29 cells, showed that restriction of folic acid or methionine inhibited cellular proliferation, while supra-physiological folate induced apoptosis. HT29 cells may be resistant to genome instability induced by folic acid or methionine deficiency under the experimental conditions reported for this study because no significant increases in micronuclei, nuclear buds or nucleoplasmic bridges were observed in the Cytokinesisblock micronucleus cytome (CBMN-Cyt) assay. The investigation on the effect of folic acid and methionine depletion on telomere length and DNA methylation in HT29 cells demonstrated that folate and methionine depletion may increase both telomere length and DNA methylation in HT29 cells. The length of telomere was positively correlated with DNA methylation. In the in vivo studies using the ApcMin/+ [Min/+ superscript] mouse model, the effect of supplementing a western-style diet with dietary folic acid and/or methionine on intestinal tumour development was assessed. A total of 113 mice were randomised to receive one of the four diet treatments; New Western Diet (NWD) as control diet, NWD with additional folic acid, NWD with additional methionine, and NWD with additional folic acid and methionine, administered at age of 3 until 13 weeks, with wild type (WT) mice used as controls. Supplementation of folic acid and methionine separately, resulted in marginally lower tumour numbers, when compared to the control diet. However, supplementation with both folic acid and methionine together appeared to annul the marginal protective effect of supplementing individually. The investigation on the effect of supplementing a western-style diet with dietary folic acid and/or methionine on genomic stability (measured via micronucleated erythrocyte assay on blood sample; telomere length and DNA methylation on the colon tissue) showed insufficient evidence that additional folic acid and/or methionine promotes DNA stability or instability in ApcMin/+ [Min/+ superscript] or WT mice. Dietary supplementation with folic acid and/or methionine at the levels and duration used in this study did not substantially promote or protect against DNA damage in WT or intestinal cancer-prone ApcMin/+ [Min/+ superscript] mouse model fed a western-style diet although a marginal effect on tumour number was evident. In conclusion, the results of this thesis support a role of methionine and folate in affecting intestinal cell proliferation and possibly tumour number. However, the impact of supplementation with folate and methionine on genome stability was marginal.
