Abstract Despite aggressive, multimodal therapies, the prognosis of patients with refractory or recurrent rhabdomyosarcoma (RMS) has not improved in four decades. RMS is thought to arise due to defective differentiation in skeletal muscle precursor cells. Differentiation-inducing therapy may improve outcomes for RMS patients with advanced disease. In RAS-mutant PAX fusion-negative RMS (FN-FMS), targeting ERK1/2 activation through MEK1/2 inhibition (MEKi) induces differentiation, slows tumor growth, and extends survival in preclinical studies. However, the duration of therapeutic response is short lived. Identifying additional targets for differentiation therapy is necessary. We propose that ASAP1, an Arf GTPase-activating protein (Arf GAP) highly expressed in FN-RMS and implicated in breast and colorectal cancer progression as well as osteogenic and retinal endothelium differentiation, contributes to FN-RMS differentiation. We find that knockdown (KD) of ASAP1 inhibits differentiation in myoblasts and FN-RMS cells, and its overexpression enhances differentiation. Moreover, myogenic differentiation-associated genes are not enriched upon loss of ASAP1. We discover that KD of ASAP1 homologs, ASAP2 and ASAP3, also blocks differentiation. However, loss of a paralogous Arf GAP, ARAP1, does not, indicating that ASAP regulates differentiation through a mechanism not explained by GAP activity alone. Interestingly, KD of Arf1 or Arf5, small GTPases inactivated by ASAP, also blocks differentiation of FN-RMS, suggesting a novel relationship between Arf and ASAP. Using RNAseq, qPCR, and immunoblotting techniques, we show that loss of ASAP blocks myogenic transcription factor expression. To determine the mechanism by which ASAP1 regulates myogenic transcription factor expression, we investigate the PI3K/AKT, MAPK, and Hippo pathways, which are known to be regulated by ASAP1. While the PI3K/AKT and MAPK pathways are unaffected, downstream components of the Hippo pathway are modulated by ASAP1 KD in FN-RMS cells treated with MEKi. YAP and TAZ are transcriptional coactivators that promote proliferation. Upon Hippo pathway activation, YAP/TAZ activity is blocked by phosphorylation at a nuclear exclusion site. Using cell fractionation and immunoblotting techniques, we find that induction of differentiation by MEKi increases TAZ phosphorylation, excluding it from the nucleus and blocking pro-proliferative transcription. Further, upon ASAP1 KD, TAZ phosphorylation is blocked, restoring nuclear localization, and inhibiting MEKi-induced differentiation. In conclusion, we discover that ASAP1 regulates MEKi-induced differentiation of FN-RMS cells by modulating TAZ localization and supports targeting the YAP pathway as a strategy for FN-RMS differentiation therapy. Our work also identifies ASAP1 as a potential effector of Arf1 activity, a novel interaction of these two proteins. The results described herein provide a deeper understanding of differentiation in FN-RMS and establish the groundwork for advancing differentiation therapy in FN-RMS. Citation Format: Katie E. Tempurung kelapa merupakan limbah organik yang memiliki peluang untuk di jadikan bahan bakar arang alternatif dan dapat digunakan untuk Pembuatan ikan asap dengan cara di asapi dengan tungku berisi batok kelapa yang di bakar, dapat digunakan juga untuk Pembuatan ikan asap dengan cara di asapi dengan tungku berisi batok kelapa yang di bakar Penelitian dilaksanakan dengan metode pirolis. Pirolis terjadi dalam tungku pembakaran dimana pada keadaan ini tempurung mengalami proses penguraian yang disebabkan oleh energi panas. Hasil pengujian yang dilakukan, dari 30 kg tempurung basah menghasilkan 13 kg arang dalam waktu pembakar 3 jam dan 5 kg tempurung kelapa tidak menggalami pembakaran.

Abstract Massachusetts’ 27 Aging Services Access Points (ASAPs) are nonprofit organizations responsible for guiding clients and families in their geographic service areas through a maze of financial and functional eligibility criteria to receive needed home and community-based services (HCBS). These services aim to sustain function and promote engagement for older adults and persons with disabilities, and to maintain individuals in the community rather than in residential settings. Payment for HCBS flows from numerous state and Federal funding programs, including but not limited to state Medicaid waivers and Older American Act programs. We conducted semi-structured interviews with 10 ASAP directors and gathered information from documentary sources to delineate the pathways to funded HCBS that these agencies open for clients. We identified variation in the 27 ASAPs’ contractual relationships with service providers. State data revealed variation in budgetary resources and area population characteristics. These nonprofit corporations appear to flexibly adapt to local conditions and needs. Understanding variation across ASAPs will support analysis of client data to examine whether ASAP processes result in different service outcomes for similar clients. Alternate payment models for health services (managed care, managed long-term services and supports, accountable care organizations) are broadening their reach, encouraging health systems to recognize the importance of functional supports and services. Some health systems are charging their own dedicated units with assigning HCBS to enrollees, avoiding the ASAPs. Understanding and modeling the allocation of services by the ASAPs will allow investigation of trends in services for persons with varying needs under various payment methods.

Background Physical literacy-focused afterschool activity programs (ASAPs) can be an effective strategy to improve children’s health-related parameters. We sought to compare physical activity, body composition, aerobic capacity, and fundamental movement skills between physical literacy-focused ASAP and a standard recreational ASAP. Method A pre–post (6 months) comparison study was conducted in 5- to 12-year-old children in a physical literacy-focused ASAP (physical literacy group, n = 14) and children attending a standard recreational ASAP (comparison group, n = 15). Physical activity guideline adherence was assessed using accelerometry, body composition was analyzed using bioelectrical impedance, aerobic capacity was estimated using the Progressive Aerobic Cardiovascular Endurance Run test, and fundamental movement skills were evaluated using the Test of Gross Motor Development–2. Results There were no significant differences between groups at baseline. After 6 months, the physical literacy group exhibited a significant improvement in their total raw score for the Test of Gross Motor Development–2 ( p = .016), which was likely due to improvements in object control skills ( p = .024). The comparison group significantly increased body mass index ( p = .001) and body fat ( p = .009) over time. No significant between-group differences were found; however, there was a trend for improved aerobic capacity in the physical literacy group ( d = 0.58). Conclusions Engagement in the physical literacy-focused ASAP contributed to an attenuated increase in adiposity and an improvement in object control skills.

ABSTRACT Mosquito-borne diseases, including dengue, malaria, and lymphatic filariasis, exact a devastating toll on global health and economics, killing or debilitating millions every year (54). Mosquito innate immune responses are at the forefront of concerted research efforts aimed at defining potential target genes that could be manipulated to engineer pathogen resistance in vector populations. We aimed to describe the pivotal role that circulating blood cells (called hemocytes) play in immunity by generating a total of 11,952 Aedes aegypti and 12,790 Armigeres subalbatus expressed sequence tag (EST) sequences from immune response-activated hemocyte libraries. These ESTs collapsed into 2,686 and 2,107 EST clusters, respectively. The clusters were used to adapt the web-based interface for annotating bacterial genomes called A Systematic Annotation Package for Community Analysis of Genomes (ASAP) for analysis of ESTs. Each cluster was categorically characterized and annotated in ASAP based on sequence similarity to five sequence databases. The sequence data and annotations can be viewed in ASAP at https://asap.ahabs.wisc.edu/annotation/php/ASAP1.htm . The data presented here represent the results of the first high-throughput in vivo analysis of the transcriptome of immunocytes from an invertebrate. Among the sequences are those for numerous immunity-related genes, many of which parallel those employed in vertebrate innate immunity, that have never been described for these mosquitoes.

CONTEXT AND OBJECTIVE: Preoperative fasting guidelines do not recommend H2 receptor antagonists or proton pump inhibitors. This study investigated prophylactic use of gastric protection and the incidence of dyspeptic symptoms in the immediate postoperative period.DESIGN AND SETTING: Non-randomized observational investigation in a post-anesthesia care unit.METHODS: American Society of Anesthesiologists risk classification ASAP1 and ASAP2 patients over 18 years of age were evaluated to identify dyspeptic symptoms during post-anesthesia care for up to 48 hours, after receiving or not receiving prophylactic gastric protection during anesthesia. History of dyspeptic symptoms and previous use of such medications were exclusion criteria. The odds ratio for incidence of dyspeptic symptoms with use of these medications was obtained.RESULTS: This investigation studied 188 patients: 71% women; 50.5% ASAP1 patients. Most patients received general anesthesia (68%). Gastric protection was widely used (n = 164; 87.2%), comprising omeprazole (n = 126; 76.8%) or ranitidine (n = 38; 23.2%). Only a few patients did not receive any prophylaxis (n = 24; 12.8%). During the observation, 24 patients (12.8%) reported some dyspeptic symptoms but without any relationship with prophylaxis (relative risk, RR = 0.56; 95% confidence interval, CI: 0.23-1.35; P = 0.17; number needed to treat, NNT = 11). Omeprazole, compared with ranitidine, did not reduce the chance of having symptoms (RR = 0.65; 95% CI: 0.27-1.60; P = 0.26; NNT = 19).CONCLUSION: This study suggests that prophylactic use of proton pump inhibitors or H2 receptor antagonists was routine for asymptomatic patients and was not associated with postoperative protection against dyspeptic symptoms.

Abstract ASAP3 is involved in a variety of biological activities, including cancer progression in humans. In adult glioma, we explore the effects of ASAP3 and NOTCH3 and their relationships on prognosis. The Oncomine, TIMER, and Gene Expression Profiling Interactive Analysis databases were used to investigate ASAP3 expression. Immunohistochemistry was used to assess the levels of ASAP3 and NOTCH3 expressions. The effects of ASAP3 and NOTCH3 on prognosis were assessed using survival analysis. The results revealed that the amount of ASAP3 mRNA in gliomas was much higher than in normal tissue (P < 0.01). Glioma patients with high ASAP3 mRNA expression had a worse overall survival and progression-free survival. ASAP3 overexpression is directly associated with the NOTCH signaling system. Immunohistochemistry revealed that ASAP3 and NOTCH3 were overexpressed in glioblastomas (GBMs). ASAP3 expression was associated with age, recurrence, tumor resection, postoperative chemoradiotherapy, World Health Organization (WHO) grade, and Ki-67 expression. ASAP3 expression was related to the isocitrate dehydrogenase-1 mutation in low-grade glioma. Gender, local recurrence, tumor resection, postoperative radio-chemotherapy, WHO grade, recurrence, and ATRX expression were all associated with NOTCH3 expression. ASAP3 was shown to be positively associated with NOTCH3 (r = 0.337, P = 0.000). Therefore, ASAP3 and NOTCH3 as oncogene factors have the potential to be prognostic biomarkers and therapeutic targets in adult glioma.

ABSTRACT Invadopodia are Src-induced cellular structures that are thought to mediate tumor invasion. ASAP1, an Arf GTPase-activating protein (GAP) containing Src homology 3 (SH3) and Bin, amphiphysin, and RVS161/167 (BAR) domains, is a substrate of Src that controls invadopodia. We have examined the structural requirements for ASAP1-dependent formation of invadopodia and related structures in NIH 3T3 fibroblasts called podosomes. We found that both predominant splice variants of ASAP1 (ASAP1a and ASAP1b) associated with invadopodia and podosomes. Podosomes were highly dynamic, with rapid turnover of both ASAP1 and actin. Reduction of ASAP1 levels by small interfering RNA blocked formation of invadopodia and podosomes. Podosomes were formed in NIH 3T3 fibroblasts in which endogenous ASAP1 was replaced with either recombinant ASAP1a or ASAP1b. ASAP1 mutants that lacked the Src binding site or GAP activity functioned as well as wild-type ASAP1 in the formation of podosomes. Recombinant ASAP1 lacking the BAR domain, the SH3 domain, or the Src phosphorylation site did not support podosome formation. Based on these results, we conclude that ASAP1 is a critical target of tyrosine kinase signaling involved in the regulation of podosomes and invadopodia and speculate that ASAP1 may function as a coincidence detector of simultaneous protein association through the ASAP1 SH3 domain and phosphorylation by Src.

ADP-ribosylation factors (Arfs) and Arf GTPase-activating proteins (GAPs) are key regulators of membrane trafficking and the actin cytoskeleton. The Arf GAP ASAP1 contains an N-terminal BAR domain, which can induce membrane tubulation. Here, we report that the BAR domain of ASAP1 can also function as a protein binding site. Two-hybrid screening identified FIP3, which is a putative Arf6- and Rab11-effector, as a candidate ASAP1 BAR domain-binding protein. Both coimmunoprecipitation and in vitro pulldown assays confirmed that ASAP1 directly binds to FIP3 through its BAR domain. ASAP1 formed a ternary complex with Rab11 through FIP3. FIP3 binding to the BAR domain stimulated ASAP1 GAP activity against Arf1, but not Arf6. ASAP1 colocalized with FIP3 in the pericentrosomal endocytic recycling compartment. Depletion of ASAP1 or FIP3 by small interfering RNA changed the localization of transferrin receptor, which is a marker of the recycling endosome, in HeLa cells. The depletion also altered the trafficking of endocytosed transferrin. These results support the conclusion that ASAP1, like FIP3, functions as a component of the endocytic recycling compartment.

The active form of vitamin D3, i.e., 1,25(OH)2D3, exerts an anti-inflammatory effect on the immune system, especially macrophage-mediated innate immunity. In a previous study, we identified 1,25(OH)2D3-responsive and vitamin D receptor (VDR)-bound super-enhancer regions in THP-1 cells. Herein, we examined the transcriptional regulation of ArfGAP with SH3 Domain, Ankyrin Repeat and PH Domain 2 (ASAP2) (encoding a GTPase-activating protein) by 1,25(OH)2D3 through the top-ranked VDR-bound super-enhancer region in the first intron of ASAP2 and potential functions of ASAP2 in macrophages. First, we validated the upregulation of ASAP2 by 1,25(OH)2D3 in both THP-1 cells and macrophages. Subsequently, we identified three regulatory regions (i.e., the core, 1,25(OH)2D3-responsive, and inhibitory regions) in the VDR bound-enhancer of ASAP2. ASAP2 promoted RAC1-activity and macrophage efferocytosis in vitro. Next, we assessed the functions of ASAP2 by mass spectrometry and RNA sequencing analyses. ASAP2 upregulated the expressions of antiviral-associated genes and interacted with SAM and HD domain-containing deoxynucleoside triphosphate triphosphohydrolase 1 (SAMHD1). In vivo, vitamin D reduced the number of apoptotic cells in experimental autoimmune encephalomyelitis (EAE) and promoted macrophage efferocytosis in peritonitis without changing the mRNA level of ASAP2. Thus, we could better understand the regulatory mechanism underlying ASAP2 transcription and the function of ASAP2, which may serve as a potential treatment target against inflammatory diseases and virus infections.

Abstract Rhabdomyosarcoma (RMS) accounts for 5% of all pediatric cancers. The overall survival rate for patients with advanced RMS has been less than 20% for the last 30 years. Investigating novel treatment strategies for RMS is necessary to improve prognosis. PAX/FOXO1 Fusion Negative RMS (FN-RMS) is commonly associated with alterations in the RAS/RAF/MEK/ERK pathway and arises due to a disruption of differentiation in skeletal muscle precursor cells. Trametinib, a MEK inhibitor, induces myogenic differentiation in FN-RMS cell lines, thus nominating trametinib as a potential differentiation therapy for FN-RMS. However, it is imperative to note that trametinib-induced RMS differentiation is incomplete. Therefore, exploring additional targets to drive differentiation is necessary. ASAP1, an Arf GTPase-activating protein (GAP), is a candidate due to its role in osteogenic and adipogenic differentiation. Additionally, ASAP1 has been shown to be overexpressed in several cancers. Furthermore, we see an overexpression of ASAP1 in FN-RMS and lower survival rates for sarcoma patients in which ASAP1 is overexpressed within the tumor. In this study, we tested the hypothesis that ASAP1 plays a role in the myogenic differentiation of FN-RMS. Upon knockdown (KD) of ASAP1, trametinib-induced differentiation was reduced, while over-expression of ASAP1 potentiated trametinib-induced differentiation. Interestingly, KD of ARF1, a substrate of ASAP1’s GAP activity, also blocked trametinib-induced differentiation. Due to these conflicting observations, we examined the relationship between ARF1 and ASAP1. Expression of exogenous ARF1 and ARF1[I46D], a mutation that renders ARF1 insensitive to ASAP1’s GAP activity, following KD of endogenous ARF1, rescued trametinib-induced differentiation. This indicates that the GAP activity of ASAP1 is not necessary for trametinib-differentiation in FN-RMS. To further examine ASAP1 and ARF1’s interaction, we investigated trametinib-induced differentiation upon KD of known effectors of ARF1 (GGA1, GGA2, and GGA3). Similar to the KD of ASAP1, trametinib-induced differentiation was reduced, indicating that this may be a phenotype between ARF1 and its effectors. Furthermore, to investigate the mechanism in which ASAP1 is regulating differentiation, we turned to the Hippo pathway’s effectors, YAP and TAZ. ASAP1 is a transcriptional target of YAP/TAZ, and YAP/TAZ activity affects myogenic differentiation. We observed that ASAP1 negatively regulates TAZ activity by affecting TAZ localization. In summary, ASAP1 functions in part as an ARF1 effector that regulates myogenic differentiation of FN-RMS by affecting TAZ localization. Future work will consist of investigating the downstream signaling effects of the novel ASAP1 and ARF1 interaction in FN-RMS and understanding the role of manipulating myogenic differentiation in advanced and metastatic FN-RMS. The Arf GTPase-activating protein (Arf GAP) with SH3 domain, ankyrin repeat and PH domain 1 (ASAP1) establishes a connection between the cell membrane and the cortical actin cytoskeleton. The formation, maintenance, and turnover of actin filaments and bundles in the actin cortex are important for cell adhesion, invasion, and migration. Here, using actin cosedimentation, polymerization, and depolymerization assays, along with total internal reflection fluorescence (TIRF), confocal, and EM analyses, we show that the N-terminal N-BAR domain of ASAP1 directly binds to F-actin. We found that ASAP1 homodimerization aligns F-actin in predominantly unipolar bundles and stabilizes them against depolymerization. Furthermore, the ASAP1 N-BAR domain moderately reduced the spontaneous polymerization of G-actin. The overexpression of the ASAP1 BAR–PH tandem domain in fibroblasts induced the formation of actin-filled projections more effectively than did full-length ASAP1. An ASAP1 construct that lacked the N-BAR domain failed to induce cellular projections. Our results suggest that ASAP1 regulates the dynamics and the formation of higher-order actin structures, possibly through direct binding to F-actin via its N-BAR domain. We propose that ASAP1 is a hub protein for dynamic protein–protein interactions in mechanosensitive structures, such as focal adhesions, invadopodia, and podosomes, that are directly implicated in oncogenic events. The effect of ASAP1 on actin dynamics puts a spotlight on its function as a central signaling molecule that regulates the dynamics of the actin cytoskeleton by transmitting signals from the plasma membrane.

ASAP1 (ADP ribosylation factor [ARF]- GTPase-activating protein [GAP] containing SH3, ANK repeats, and PH domain) is a phospholipid-dependent ARF-GAP that binds to and is phosphorylated by pp60Src. Using affinity chromatography and yeast two-hybrid interaction screens, we identified ASAP1 as a major binding partner of protein tyrosine kinase focal adhesion kinase (FAK). GlutathioneS-transferase pull-down and coimmunoprecipitation assays showed the binding of ASAP1 to FAK is mediated by an interaction between the C-terminal SH3 domain of ASAP1 with the second proline-rich motif in the C-terminal region of FAK. Transient overexpression of wild-type ASAP1 significantly retarded the spreading of REF52 cells plated on fibronectin. In contrast, overexpression of a truncated variant of ASAP1 that failed to bind FAK or a catalytically inactive variant of ASAP1 lacking GAP activity resulted in a less pronounced inhibition of cell spreading. Transient overexpression of wild-type ASAP1 prevented the efficient organization of paxillin and FAK in focal adhesions during cell spreading, while failing to significantly alter vinculin localization and organization. We conclude from these studies that modulation of ARF activity by ASAP1 is important for the regulation of focal adhesion assembly and/or organization by influencing the mechanisms responsible for the recruitment and organization of selected focal adhesion proteins such as paxillin and FAK.

Ikan roa asap merupakan salah satu ikan asap yang banyak dikonsumsi oleh masyarakat Gorontalo. Ikan ini diolah dengan cara tradisional dengan menggunakan bahan bakar kayu/batok kelapa. Salah satu metode pengasapan modern yang sedang berkembang saat ini adalah metode asap cair. Asap cair digunakan untuk pengawetan karena kandungan senyawa fenol dan asam yang berperan sebagai antioksidan dan antimikrobia. Faktor-faktor yang mempengaruhi mutu ikan asap dengan metode asap cair adalah konsentrasi asap cair yang digunakan dan lama perendaman. Tujuan dari penelitian ini adalah untuk mengetahui konsentrasi asap cair dan lama perendaman terbaik yang mampu mengoptimalkan mutu ikan roa asap dari aspek kadar air dan kadar abu. Penelitian ini menggunakan rancangan Response Surface Method-Central Composite Design (RSM-CCD) dengan 2 faktor. Faktor perlakuan pada penelitian ini yakni variasi konsentrasi asap cair dan lama perendaman. Variasi konsentrasi asap cair (A) terdiri atas 5 taraf yakni 2.38%, 3%, 4,5%, 6% dan 6,62% sedangkan variasi lama perendaman (B) terdiri atas 5 taraf yakni 48 menit, 60 menit, 90 menit, 120 menit dan 132 menit. Hasil penelitian menunjukkan konsentrasi asap cair berpengaruh signifikan terhadap kadar abu ikan roa asap, dan lama perendaman berpengaruh signifikan terhadap kadar air dan kadar abu ikan roa asap. Kadar air optimal akan diperoleh pada konsentrasi asap cair 4,52% dan lama perendaman 48 menit, sedangkan kadar abu optimal ikan roa asap akan diperoleh pada konsentrasi asap cair 6,62% dan lama perendaman 48 menit.

Telah dilakukan riset tentang pengaruh konsentrasi larutan asap cair terhadap mutu belut asap yang dihasilkan. Belut yang digunakan adalah jenis Monopterus albus dan asap cair yang dipakai diperoleh dari CV Pusat Pengolahan Kelapa Terpadu,Yogyakarta. Riset pendahuluan dilakukan untuk mengetahui waktu pengeringan yang terbaik, dengan melakukan pengeringan dalam oven pada suhu 90ºC selama 2, 4, 6, dan 8 jam diikuti pengujian kadar air terhadap produk yang dihasilkan. Riset utama dilakukan unluk mengetahui pengaruh konsentrasi asap cair terhadap mutu belut asap yang dihasilkan dengan variasi konsentrasi larutan asap cair 0, 10, 20, dan 30%. Penilaian mutu dilakukan berdasarkan analisis komponen asap, komposisi proksimat, den mutu organoleptik. Hasil penelitian menunjukkan bahwa konsentrasi larutan asap cair terbaik untuk pembuatan belut asap adalah 30% dengan waktu pengeringan 8 jam yang menghasilkan rendamen 28,04%. Secara umum produk ini disukai oleh panelis dengan nilai orgenoleptik 7,36 dengan karakteristik kadar air 10,38%, lemak 2,74%, protein 68,96%, dan abu 15,81%. Asap cair den belut asap hasil pengolahan menggunakan asap cair tidak mengandung senyawa Polycyclic Aromatic Hydrocarbons (PAH), sebaliknya belut asap komersial mengandung senyawa PAH.

Hasil penelitian menunjukkan bahwa pemberian asap cair mengakibatkan perubahan ukuran diameter koloni patogen,asap cair memiliki total fenol + 9,98%. Konsentrasi asap cair TKKS berpengaruh nyata terhadap efektivitas daya hambat, menekan laju pertumbuhan, indeks anti jamur terhadap G.boninense dan Curvularia sp. Uji komparasi menunjukkan bahwa jenis asap cair tidak berpengaruh tehadap G. boninense, sedangkan terhadap Curvularia sp.berpengaruh nyata. Kesimpulan dari hasil penelitian konsentrasi asap cair TKKS terbaik adalah 5% dan asap cair TK memiliki efektivitas yang lebih tinggi dalam mengendalikan G. boninense, dan Curvularia sp. dengan efektivitas 100%.

Ikan asap adalah salah satu produk olahan tradisional di Indonesia. Berbagai jenis ikan dapat digunakan sebagai bahan baku ikan asap. Salah satunya adalah ikan patin (Pangasius-pangasius). Modifikasi pengasapan ikan digunakan untuk menghasilkan ikan asap yang disukai konsumen. Modifikasi ikan asap yang dilakukan pada penelitian ini terdiri dari dua (2) model ikan asap yaitu ikan patin tanpa disayat diasapi (A) dan ikan patin disayat (B) kemudian diasapi. Penyayatan daging pada salah satu model ikan asap diharapkan dapat memperluas permukaan kontak daging ikan dengan asap sehingga menghasilkan profil sensori yang berbeda. Metoda yang digunakan dalam penelitian ini adalah uji sensori skala rating hedonik. Tujuan dari penelitian ini adalah untuk menentukan model ikan asap yang disukai oleh konsumen. Dari hasil penelitian diketahui bahwa ikan asap dengan bahan baku ikan patin berat rata-rata 280-320 gr menghasilkan rendemen ikan asap sebanyak kurang lebih 60%. Dari hasil uji sensori diketahui bahwa panelis lebih memilih atribut penampakan dan tekstur untuk ikan asap yang disayat dan panelis memilih untuk atribut rasa dan bau pada ikan asap yang tanpa disayat. Komposisi ikan asap tanpa disayat adalah protein 13%, lemak 0,54%, air 73% dan mineral 1,77%, untuk ikan asap bersayat kadar protein 23%, lemak 0,44%, air 65% dan mineral 0,96%.

Menggunakan Sensor MQ-2 digunakan untuk mendeteksi kondisi kadar asap rokok pada ruangan bebas asap rokok dimana pada saat sensor berada dibawah nilai ppm < 200 maka kondisi ruangan dalam kondisi normal pada saat sensor mendeteksi ppm antara 200 sampai dengan 300 kondisi ruangan dalam keadaan waspada, sedangkan saat sensor mendeteksi lebih dari 300 kondisi ruangan dalam keadaan bahaya dan kipas dc berputar untuk menetralisir keberadaan asap rokok. Kemampuan pendeteksi asap rokok ini akan mendeteksi keberadaan asap rokok di udara tergantung dari sudut sumber asap rokok dan waktu pembacaan pada sensor serta arah pergerakan asap rokok. Proses pengiriman data dari input ke output dilakukan dengan menggunakan ketika sensor MQ-2 mendeteksi asap rokok, sensor MQ-2 akan mengirimkan sinyal ke Arduino Uno untuk mengaktifkan output. Jadi hasil penelitian Setelah merancang prototipe sistem dan mengujinya, dapat diketahui bahwa pendeteksi asap rokok dapat diterapkan pada ruangan bebas asap rokok.

Limbah padat hasil penyulingan minyak nilam banyak dijumpai diindustri penyulingan minyak nilam terutama dikota Lhokseumawe. Besarnya volume limbah padat penyulingan nilam belum termanfaatkan secara optimal. Dengan memanfaatkan limbah tersebut menjadi produk yang berguna dan mempunyai nilai tambah yang nyata. Keberadaan limbah padat nilam sangat berpotensial untuk diolah menjadi asap cair karena memiliki komponen senyawa organik yaitu selulosa, hemiselulosa dan lignin. Penelitian ini sudah dilakukan sebelumnya, yang belum adalah penggunaan limbah padat nilam sebagai bahan baku pembuatan asap cair dengan menggunakan metode pirolisis yang menghasilkan lebih banyak yield asap cair. Tujuan dari penelitian ini adalah untuk mengetahui pembuatan asap cair serta pengaruh suhu dan waktu pirolisi terhadap yield, densitas dan pH asap cair yang dihasilkan. Metode penelitian menggunakan proses pirolisi yang dilakukan pada suhu 250oC, 300oC dan 350oC dengan variasi waktu pirolisis 60 menit, 90 menit, 120 menit dan 150 menit. Asap cair diperoleh dari kondensasi asap hasil dekomposisi senyawa organik pada proses pirolisis. Dari penelitian diketahui bahwa yield asap cair cenderung meningkat seiring naiknya suhu dan waktu pirolisis. Yield asap cair tertinggi diperoleh sebesar 16,16%. Densitas terbaik diperoleh dari asap cair hasil pirolisis sebesar 0,9916 gr/ml. pH asap cair terbaik diperoleh dari asap cair hasil pirolisis sebesar 3,32. Kandungan asap cair pada uji menggunakan GC-MS adalah fenol sebesar 46,14% dan benzene sebesar 15,55%.

Kelompok pengrajin mini nopia (mino) yang berada di Desa Pekunden Kecamatan Banyumas mengeluhkan adanya gangguan asap pembakaran kayu yang digunakan untuk proses pemasakan. Gangguan yang dirasakan antara lain pada pernafasan dan rasa pedih di mata. Asap yang timbul saat proses pemasakan memang tidak bisa dihilangkan karena untuk mempertahankan kekhasan rasa dari mini nopia (mino) tersebut, tetapi paling tidak asap tersebut bisa diminimalisir atau dikurangi. Salah satu cara untuk mengurangi asap adalah dengan cara menyedot asap tersebut keluar dapur, yaitu dengan memasang exhaust fan yang disertai sensor asap (smart exhaust fan). Sensor mendeteksi konsentrasi asap yang selanjutnya untuk menentukan putaran exhaust fan. Jika konsentrasi asap rendah maka putaran exhaust fan rendah dan seterusnya kecepatan putaran akan bertambah jika konsentrasi asap juga bertambah. Alat yang diaplikasikan dapat bekerja dengan baik sesuai dengan perencanaan, yaitu dapat mengurangi asap yang ada di dapur pengolahan mini nopia (mino). Ada tiga tingkatan kecepatan putar exhaust yang ditentukan berdasarkan konsentrasi asap, yaitu rendah (low), menengah (middle) dan tinggi (high). Kegiatan Pengabdian Kepada Masyarakat (PKM) melibatkan mitra Kelompok Tani Maju di Desa Rasau Jaya Tiga. Terdapat permasalahan yang dihadapinya yaitu produktivitas tanaman padi diketahui masih sangat rendah, kurangnya pengetahuan petani dalam pemanfaatan teknologi pertanian, petani lebih dominan menggunakan pestisida kimia sintetik dibandingkan pemakaian pestisida organik, dan keterbatasan petani dalam memenuhi kebutuhan pestisida kimia sintetik karena harganya yang semakin mahal. Program PKM ini bertujuan untuk membantu petani meningkatkan produksi tanaman padi melalui kegiatan penyuluhan dan pelatihan teknologi asap cair dari sekam padi sebagai pestisida organik serta meningkatkan wawasan para petani untuk memproduksi sendiri asap cair sehingga dapat mendukung perekonomian masyarakat desa. Metode yang dilakukan adalah Participatory Learning and Action, semua peserta aktif terlibat dalam diskusi dan praktik pembuatan asap cair. Substansi materi yang diberikan yaitu pengenalan asap cair, manfaat asap cair, diseminasi asap cair, teknologi pembuatan asap cair, dan aplikasi asap cair untuk pestisida organik. Selain itu dilakukan pula pelatihan manajemen produksi untuk menumbuhkan jiwa wirausaha kelompok tani. Hasil kegiatan PKM menunjukkan pengetahuan dan keterampilan peserta mengenai asap cair sebagai pestisida organik meningkat. Adapun teknologi yang dihasilkan adalah teknologi pembuatan alat pirolisis untuk pembuatan asap cair, teknologi pembuatan asap cair dari limbah sekam padi, dan teknologi pemanfaatan asap cair.

Asap cair dari limbah kulit batang mangrove telah diproduksi menggunakan metode pirolisis pada suhu 400°C selama 6 jam. Selanjutnya asap cair diadsorpsi menggunakan kaolin yang diaktivasi secara termal pada suhu 500°C. Analisa GC-MS (Gas Chromatography Mass Spectrophotometer) menunjukkan adanya beberapa senyawa dominan pada asap cair yaitu 2-furankarboksildehid, 2-propanon dan asam asetat. Aktivitas antioksidan asap cair dianalisis menggunakan metode DPPH (1,1-Diphenyl-2-picryl Hidrazil). Asap cair limbah kulit batang mangrove sebelum diadsorpsi memiliki aktivitas antioksidan (IC50 = 1635,65 ppm) lebih kuat dari asap cair limbah kulit batang mangrove setelah diadsorpsi (IC50 = 3023,06 ppm). Nilai IC50 tersebut menunjukkan asap cair memiliki aktivitas antioksidan sangat lemah. Hasil uji toksisitas asap cair limbah kulit batang mangrove sebelum diadsorpsi (LC50 = 562.341 ppm) dan asap cair limbah kulit batang mangrove setelah diadsorpsi (LC50 = 46.625 ppm). Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) dengan faktor tunggal dan parameter yang diamati adalah penekanan diameter koloni, kerapatan spora, dan daya kecambah. Variabel pada penelitian ini adalah konsentrasi asap cair (A0 = Kontrol, A1 = asap cair 0,02%, A2 = asap cair 0,04%, A3 = asap cair 0,08%, A4 = asap cair 0,16%, A5 = asap cair 0,32%) diuji terhadap koloni jamur Colletotrichum sp. Berdasarkan hasil penelitian dapat disimpulkan bahwa pemberian asap cair yang paling baik untuk menekan diameter koloni jamur Colletotrichum sp. adalah pada konsentrasi 0,32% dengan penghambatan pertumbuhan jamur 29,13%. Selain itu, pemberian asap cair dapat menghambat pembentukan spora Colletotrichum sp. yang menyebabkan semakin kecilnya kerapatan spora yang terbentuk dengan pemberian konsentrasi 0,32% sebesar 0,1875 x 10<sup>6</sup>. Sedangkan konsentrasi asap cair 0,32% sangat bagus untuk menghambat daya kecambah (viabilitas) jamur Colletotrichum sp. yaitu sebesar 35,41%.</em>

Ikan asap adalah produk yang termasuk olahan tradisional hasil perikanan yang sangat disukai oleh masyarakat Indonesia dengan cara diasapi dengan menggunakan bahan bakar alami. Ikan asap yang berkualitas baik dapat disimpan dalam waktu 1-2 minggu. Salah satu diversifikasi olahan pangan dari bahan baku ikan asap adalah sambal ikan asap. Sambal adalah saus yang berbentuk semi padat dari bahan dasar cabai dan biasanya ditambah bahan-bahan seperti bawang merah, bawang putih, dan tomat. Tujuan penelitian untuk mengetahui pengaruh penambahan ikan asap terhadap kadar air, kadar abu dan kadar protein dari produk sambal dari penambahan ikan cakalang asap. Penelitian ini meliputi 4 perlakuan dan 3 ulangan. Perlakuan yang dimaksud yaitu penambahan ikan cakalang asap 400 g, 500 g, 600 g, dan 700 g. Parameter yang diamati dalam peneltitian ini adalah kadar air, kadar abu, kadar protein dan uji organoleptik terhadap aroma, rasa, tekstur dan warna sambal ikan cakalang asap. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL), apabila ada beda nyata dipakai uji lanjut BNJ. Hasil ini menunjukkan bahwa Pengaruh penambahan ikan asap pada pembutan sambal ikan cakalang asap sangat berpengaruh nyata pada kadar air, kadar abu dan kadar protein. Kadar abu dan kadar protein semakin meningkat dengan meningkatnya penambahan ikan cakalang asap, sedangkan kadar air semakin menurun dengan penambahan ikan ikan cakalang asap. Uji kesukaan terhadap aroma, rasa, tekstur dan warna yang paling disukai panelis perlakuan penambahan ikan cakalang asap 400 gr, sedangkan dari segi warna pada perlakuan penambahan ikan cakalang asap 500 gr.

Efikasi asap cair kayu bintangur terhadap jamur S. commune dengan rancangan acak lengkap faktorial, terdiri atas suhu pembuatan asap cair kayu bintangur (370, 400, dan 430oC) dan konsentrasi asap cair kayu bintangur (0, 0,5, 1,0, 1,5 dan 2,0%, v/v). Hasil penelitian menunjukkan bahwa suhu pirolisis produksi asap cair dan konsentrasi asap cair berpengaruh sangat nyata terhadap daya hambat pertumbuhan jamur, S. commune. Ikan asap yang banyak dipasarkan di Indonesia kebanyakan merupakan hasil pengasapan langsung dari pembakaran kayu, tempurung atau sabut kelapa. Cara pengasapan ini banyak kekurangannya diantaranya kurang efektif dalam penggunaan asap, menyebabkan polusi udara, dan kurang praktis. Untuk memperbaiki cara pengasapan ini sekaligus untuk meningkatkan kualitas ikan asap, telah dilakukan penelitian mengenai penggunaan asap cair untuk pengolahan ikan asap. Asap cair yang digunakan berasal dari pembakaran campuran batang singkong dan tempurung kelapa, sedangkan ikan yang digunakan adalah ikan tongkol (Euthynnus affinis). Percobaan pengasapan dilakukan dengan variasi konsentrasi asap cair 0,5; 1,0; 1,5; 2,0% dengan lama perendaman 30 dan 60 menit. Hasilnya menunjukkan bahwa asap cair dengan konsentrasi 2% dan lama perendaman 30 menit menghasilkan ikan tongkol asap dengan mutu terbaik yaitu paling disukai konsumen dengan nilai organoleptik paling tinggi serta tidak ditemukan adanya senyawa benzo(a)piren.

Salah satu pemanfaatan cangkang biji karet supaya bernilai ekonomis yang tinggi adalah dibuat asap cair dengan proses pirolisis. Asap cair merupakan suatu campuran larutan senyawa murni. Penelitian ini bertujuan untuk mengoptimasi proses pirolisis asap cair dan mengaplikasikan nya sebagai koagulan lateks. Proses pirolisis dilakukan dengan melakukan variasi waktu pirolisis sebagai berikut: 30 menit, 60 menit da 120 menit dengan bahan baku seberat 500 gram dan suhu pirolisis 250˚. Berdasarkan hasil pirolisis ini diketahui bahwa perlakuan dengan waktu 120 menit menghasilkan rendemen asap cair paling tinggi sebesar 16% dan perlakuan 30 menit menghasilkan rendemen asap cair terendah sebesar 10,3%. Karakterisasi asap cair cangkang biji karet memperlihatkan bahwa perlakuan 30 menit menghasilkan pH asap cair paling tinggi sebesar 4 (asam asetat 10,6%) dan perlakuan 120 menit menghasilkan pH paling rendah sebesar 3,3 ( asam asetat 9,8%) dari hasil yang didapat dapat diketahui semakin rendah pH asap cair maka semakin tinggi kadar asam asetat didalamnya. Pada aplikasi asap cair sebagai koagulan lateks menunjukan bahwa asap air dari cangkang biji karet berpotensi sebagai koagulan dengan waktu penggumpalan antara 375-440 detik lebih singgkat jika dibandingkan asap cair komersil yang memerlukan waktu penggumpalan 3652 detik.

ASAP1 (also known as AMAP1 or DDEF1) encodes an Arf GTPase-activating protein (Arf GAP), a multifunctional scaffold protein that induces hydrolysis of GTP bound to the ADP-ribosylation factor (Arf) family GTP-binding proteins. Reduction of ASAP1 expression in vitro was related to suppression of cell migration and invasiveness. The genetic variant rs4733781 of the ASAP1 gene was revealed as a significant locus for tuberculosis (TB) susceptibility, but the results still need to be validated. Methods. Blood samples from a total of 1914 active TB and healthy controls (HC) were collected to evaluate rs4733781 and the risk of TB. Meanwhile, a total of 48 noninfected HC, latent TB-infected (LTBI) controls, and active TB were collected to assay ASAP1 expression difference among the three groups. The QuantiFERON-TB Gold In-Tube was adopted to identify noninfected HC and LTBI. Results. The genetic variant of rs4733781 was found to be significantly associated with TB, and the A allele of rs4733781 (C>A) was 0.38 and 0.43 among TB cases and HC, respectively (P=0.0035). Meanwhile, the peripheral blood monocyte RNA fold changes for the ASAP1 gene among the 16 HC, 16 LTBI, and 16 active TB were 1.088±0.4919, 2.237±0.6505, and 10.12±10.98 (F=9.559, P=0.0003), respectively, and the expression of ASAP1 was increased by 2.06-fold (P<0.0001) and 9.30-fold (P<0.0052) for LTBI and active TB, when compared to the HC. Conclusions. Our data indicated that the A allele of rs4733781 for the ASAP1 gene was in association with a decreased risk of TB. But not only that, the overexpression of the ASAP1 gene among LTBI and TB was related to the progression of TB, which further implies that the expression of ASAP1 would be a potential biomarker for LTBI and TB diagnoses.

Limbah kulit durian dapat diolah menghasilkan asap cair dan karbon aktif dengan metode pirolisis. Untuk menghasilkan asap cair yang dapat digunakan sebagai pengawet makanan, asap cair perlu di proses dengan metode pemurnian seperti destilasi, pemurnian dengan zeolit aktif dan arang aktif. Hasil karakterisasi asap cair dari kulit durian hasil pirolisis, destilasi, pemurnian dengan zeolit aktif dan pemurnian dengan arang aktif diperoleh nilai pH berturut-turut 4,09; 3,24; 3,46; dan 2,09. Kadar asam asetat berturut-turut 6,07; 7,59; 9,11; dan 10,63 (mg/L). Berdasarkan karakteristik komponen senyawa menggunakan GC-MS asap cair hasil pirolisis terdeteksi 17 senyawa, asap cair hasil destilasi 12 senyawa, asap cair pemurnian zeolit aktif 10 senyawa dan asap cair pemurnian arang aktif 6 senyawa dengan mayoritas komponen senyawa terdiri dari asam asetat, fenol dan karbonil serta tidak mengandung senyawa karsinogenik, sehingga asap cair dari kulit durian dapat digunakan sebagai pengawet makanan.

Permasalahan yang dihadapi oleh Mitra adalah adanya keterbatasan jangkauan wilayah pemasaran produk olahan ikan asap yang dihasilkan, perlakuan pasca produksi ikan mujairr asap, produk Ikan Mujair Asap yang tidak dapat bertahan lama, belum adanya diversifikasi produk olahan dan mahalnya ikan mujair segar sebagai bahan baku. Dari lima permasalahan yang ada maka pengusul sepakat untuk memberikan penawaran solusi yaitu : melakukan Pelatihan manajemen kualitas produk, perilaku pengelolaan makan sehat dan diversivikasi produk olahan, Melakukan pelatihan pengemasan produk olahan berupa Mujair asap dengan baik sehingga produk tidak mudah rusak dan tahan lama, Memberikan bantuan alat Sealer Vacuum Cleaner untuk pengemasan produk. Latar belakang: Tembakau yang dijadikan rokok selain berdampak pada perokok juga berdampak bagi orang yang tidak merokok. Karsinogen dalam asap rokok dapat menyebabkan kanker salah satunya kanker serviks. Kanker serviks merupakan kanker keempat yang paling sering terjadi pada wanita di dunia. Deteksi dini kanker serviks dapat dilakukan dengan metode inspeksi visual asam asetat (IVA). Tujuan penelitian ini untuk mengetahui hubungan paparan asap rokok dengan hasil IVA pada wanita usia subur. Metode: Desain penelitian ini merupakan analitik observasional dengan pendekatan cross-sectional. Subjek penelitian diambil dari wanita yang melakukan pemeriksaan IVA di wilayah kerja Puskesmas Tanjung Sekayam dengan cara consecutive sampling sebanyak 40 wanita usia 30 – 50 tahun. Analisis bivariat dilakukan dengan uji fisher exact. Hasil: 30 orang (75%) terpapar asap rokok dan 10 orang (25 %) tidak terpapar asap rokok, 39 orang (97,5%) memperoleh hasil IVA negatif dan 1 orang (2,5%) memperoleh hasil IVA positif. Analisis bivariat paparan asap rokok dan hasil IVA diperoleh nilai p=1,000, intensitas paparan asap rokok per hari dan hasil IVA wanita terpapar asap rokok diperoleh nilai p=0,433, durasi paparan asap rokok per hari dan hasil IVA wanita terpapar asap rokok diperoleh nilai p=0,367. Kesimpulan: Tidak terdapat hubungan paparan asap rokok dengan hasil IVA, tidak terdapat hubungan intensitas dan durasi paparan asap rokok per hari dengan hasil IVA wanita terpapar asap rokok.

Ikan asap adalah olahan hasil perikanan yang dilakukan secara tradisional. Keberadaan bakteri patogen dan kerusakan pada ikan asap dapat mengakibatkan gangguan kesehatan pada tubuh manusia. Pada penelitian ini didapatkan sebanyak 17 sampel ikan asap dengan jenis ikan asap tongkol dan ikan asap pari dari pedagang pasar tradisional. Tujuan penelitian ini untuk mengetahui nilai total bakeri, kandungan formalin serta nilai kadar air yang terdapat pada ikan asap di wilayah Kecamatan Krembangan, Kota Surabaya. Penelitian ini menggunakan metode pour plate pada uji total bakteri, sedangkan pada uji formalin menggunakan formalin kit. Hasil dari penelitian menunjukkan bahwa total bakteri pada ikan asap sebesar 5,51 x 105 koloni/gr dan kadar air pada sampel ikan asap berkisar antara 42% - 58%. Hal ini terjadi karena kondisi buruknya higiene sanitasi pedagang dengan rata-rata 12,25%.

Smoke is air pollution that is very dangerous for human health. Therefore, in this study the authors designed and modeled a fan speed controller using a smoke sensor based on an Arduino microcontroller. In addition, smoke control alarms can also be used for monitoring in smoke-free rooms in order to prevent and at the same time provide a deterrent effect on people who smoke in places where smoking is prohibited. The method used is to process the data detected by the MQ2 smoke sensor, then display the amount or level of smoke in the room and its condition on the LCD/monitor. The comparison of the values on the sensor will be processed with the Arduino and the output of the Arduino will be input coding on the Arduino IDE software to control the fan speed performance as a response to exhaust air in the ventilation area of the room. The microcontroller process is in the program as a controller of all components, then coding will control all components according to the coding ordered in the microcontroller and voltage controller at the smoke sensor input and LCD and fan output. The results obtained from this research project are able to detect the state of the air in a smoke-free room from smoke and CO pollution from 0 ppm to = 256 ppm and this pollution can be reduced by adjusting the fan speed as room air ventilation to avoid the deposition of smoke in a smoke-free room. which is dangerous. So that the air in the room becomes cleaner and safer for health.
