Testo completoDue to the lack of feasible polymer-based biofertilizers as alternative to synthetic fertilizers, this study aimed to utilize chitosan and carrageenan as main components of the natural-based polymer carrier matrix (PCM) used for the immobilization of plant-growth promoting bacteria (PGPB). The polyelectrolyte complexation of these macromolecules was used as the main mechanism in the design of the PCM. The physico-chemical properties of these polymers were extensively characterized, namely, the degree of sulfation and degree of acetylation by nuclear magnetic resonance and UV-Vis spectrometry, and the molar mass distributions by size exclusion chromatography. The screening of numerous process parameters resulted in the successful formation of spherical core-shell beads. The mechanical properties of these beads were further examined by plane/plane squeezing (compression) tests. The simultaneous process of matrix formation and immobilization of the PGPB Azospirillum baldaniorum Sp245, leading to the production of a biofertilizer prototype, was also established. The immobilization, survival and the release of these bacteria were evaluated via imaging by confocal microscopy and quantification of cell number. Accordingly, in vitro and in vivo greenhouse experiments investigated the plant growth of wheat and evidenced an enhanced plant growth and development. Moreover, the immobilization, survival and the release of these bacteria were shown to result from their entrapment in the successfully developed chitosan-carrageenan based PCM. Numerous studies are confronted with a significant challenge of transitioning the technology of production for polymeric carrier materials from bench-scale to commercial scale. Thus, in this study, we propose the use of extrusion technology for the pilot-scale production of a polymeric carrier material via the in situ polyelectrolyte complexation of chitosan and carrageenan. Also, the production process is strictly based on controlling parameters to be microorganism-friendly; hence, the process being developed is termed as cold extrusion. Solutions of chitosan and carrageenan were fed into a pilot-scale Leistritz twin-screw extruder. Several extrusion process parameters were surveyed to identify a set of process conditions that would generate the desired gel with mechanical properties suitable as a carrier material for microorganisms. To this end, a systematic rheological characterization was performed on the collected gel extrudates, as well as the prepared polyelectrolyte solutions using the ARES 2000EX rheometer. Through a dynamic mechanical analysis of the storage modulus G’, and loss tangent tan ϕ, we investigated the effect of the extrusion process parameters on gel behavior and validated the production of polyelectrolyte complex gels. Optical microscopy images of these gels also evidenced a heterogeneous polyelectrolyte complexation. The obtained materials exhibited soft gel behavior ensuring structural integrity of the carrier for further processing steps (ex: drying) while its unique viscoelastic characteristics help in the prolonged release of the immobilized microorganism. Thus, the findings from this preliminary survey of extrusion process parameters reveal considerable potential for utilizing extrusion technology in the large-scale commercial production of biofertilizers

Testo completoRecycling of charcoal by soil fauna activity, in soils under shifting cultivation in French Guiana, was evidenced by the presence of black and grey earthworm casts. The results were confirmed in microcosm studies with the earthworm Pontoscolex corethrurus. The adaptive behaviour of this worm to repeated charcoal supply, suggests that soil biological activity in fallow was better adapted to slash-and-burn cultivation than that in mature forest. These results wer applied to the improvement of soils under permanent agriculture. The application of a mixture of charcoal and manioc peels, in the presence of P. Corethrurus, allowed an improvement of yard-long bean production and soil fertility and a preservation of the crumb structure of the topsoil. This study demonstrated the importance of charcoal in the biological functioning of organic-poor soils and its possible use, through earthworm activity, for the restoration of tropical soils degraded by permanent agriculture

Testo completoFertilizing matters (FM) can be hazardous to the environment during application, due to the potential presence of contaminants. The approval process involves demonstrating the safety of these matrices towards the environment, but there is no ecotoxicological test strategy to date. In this context, tests using the nematode C. elegans and the predatory mite H. aculeifer have been proposed in a battery of bioassays, as new model organisms for the ecotoxicological assessment of FM. The results obtained with C. elegans and H. aculeifer by direct and indirect approaches were compared and discussed in relation to those obtained for other terrestrial (plants and earthworms) and aquatic organisms (bacteria, algae, rotifers and crustaceans) in order to provide, in fine, a relevant test strategy to assess the environmental hazards of the agricultural use of FM

Testo completoUrban agriculture (UA) and pollution are two worlds more inter-connected every day, creating one of the main challenges of sustainable cities as persistent metal(loid) contamination increases as much as the interest for urban agriculture. Biofertilizers and bioamendments used in UA (arbuscular mycorrhizal fungi, compost, and biochar) can influence the mobility of contaminants in soil. This study aims to better understand the fate of anthropic or geogenic, major (Cd, Pb) and emerging (Sb), inorganic contaminants in soil-plant-biofertilizer systems and their human bioaccessibility. While contaminant mobility in soil is affected by biofertilizers, their origin influences also their bioaccessibility. The fungal community seems crucial in this phenomenon but is impacted by compost addition. Hence, using these biofertilizers in contaminated soils has to be thought wisely because of the multiple interactions affecting contaminant's phytoavailability

Testo completoBecause of their ability to form a symbiosis with soil bacteria called Rhizobium, legumes play an important role in agriculture. These bacteria fix atmospheric nitrogen; hence their host plants do not need nitrogen fertilizers. In addition, some strains of Rhizobium have the ability to solubilize phosphate, thus providing phosphorus to host plants. An ability to control certain pathogens has also been demonstrated in several studies. The present study evaluates the symbiotic efficiency of Tunisian rhizobia, their ability to solubilize phosphate, and their ability to control Verticillium wilt and other pathogenic fungi in Medicago truncatula. Thirty-six rhizobial isolates were obtained from root nodules of M. truncatula from different parts of Tunisia were used in this work. About 60% of these isolates were able to solubilize phosphate in vitro. In a second step, three phosphate solubilizing rhizobia and one isolate unable to solubilize phosphate in vitro were used for phytotron assays to see the effect of the presence of rhizobia on plant growth parameters in the presence of soluble and insoluble forms of phosphate. The results of the experiment show that A17 plants treated with CaHPO4, tend to produce more nodules and shoot biomass than F83005.5 and that the phosphate form used, soluble or non-soluble, affects parameters studied. Inoculation of four M. truncatula lines with 16 previously selected rhizobial isolates showed significant interaction between isolates and lines for symbiotic abilities as visualised by nodule formation. All rhizobial isolates tested increased above-ground biomass, reduced root biomass, and increased nitrogen content with strains effects of plant genotype and bacterial isolate. Finallly, these isolates were tested for their ability to protect M. truncatula plants against Verticillium wilt, and to inhibit the growth of pathogenic fungi such as Verticillium and Fusarium in vitro. However, no isolate could be identified as effective for biological control. The results of this study suggest that selected rhizobial isolates could be used as biofertilizers in poor soils to reduce the use of nitrogen and phosphorus fertilizers but not to control Verticillium wilt
