Abstract (sommario):
Zinc (Zn) is an essential trace element and its deficiency is known to compromise a wide range of functions, including vascular function and immune function. The hypothesis of the thesis was that suboptimal Zn status would induce changes in the vascular system, thus raising the potential risk of developing atherosclerosis. After 2 wk, aorta was taken from male adult rats fed by either acute Zn deficient (AZD, <1mg Zn/kg), Zn adequate Pair fed of AZD (PF, 35mg Zn/kg), or Zn adequate control (ZA, 35mg Zn/kg, fed ad libitum) diet. It was found most genes which were significantly regulated between AZD and PF aorta belong to cytoskeleton remodelling. Plasma from the same rats was used to incubate primary VSMC in vitro. Dramatic changes of gene expression in pathways associated with immune function. While AZD plasma induced marked changes in VSMC gene expression in vitro, no significant change was found by depleting Zn from PF plasma. Therefore it is possible that a Zn deficiency induced humoral factor was responsible for influencing VSMC gene expression. By semi-purifying plasma using gel filtration and molecular filtration, it was confirmed that the humoral factor has a molecular weight of around 2kDa and is thought to be a peptide hormone, which could serve as a potential biomarker of Zn status. Splenocytes and whole blood were taken from rats and used to measure cytokine secretion before and after either LPS or ConA treatment. Different concentrations of several cytokines, such as IL2, IL6, TNFα and IFNγ were found from different Zn status. Overall the thesis emphasises the participation of Zn in several aspects of the vascular system, such as regulating cytokine production and maintaining vascular structure. It provides important evidence of the role that Zn plays in the development of atherosclerosis.

This study investigated the iron status and dietary intakes in 6-24 month old children in Australia and Ghana and assessed the effects of dietary calcium on intestinal iron absorption. The true prevalence of non-anaemic iron deficiency (NAID) and iron deficiency anaemia (IDA) and dietary intakes in infants and toddlers from a broad socio-economic background were assessed by haematological and biochemical parameters, semi-quantitative diet recall and anthropometric measurements. The high prevalence of iron deficiency and anaemia found in Australian and Ghanaian children can be attributed to the low intake of bioavailable iron in weaning diets which are often ingested with large amounts of calcium. While calcium has been shown to inhibit the absorption of iron, its mechanism of interaction with iron absorption at the intestinal level is not known. The rat was used as an experimental model to investigate the effects of dietary calcium on duodenal iron uptake. The results indicate there is a critical period during weaning when the consumption of high dietary calcium with low iron can retard growth potential. Dietary calcium significantly inhibits non-haem iron absorption at the intracellular level by up-regulating villus enterocyte ferritin concentrations under iron deficiency conditions.

In order to examine the effects of prelatent/latent iron deficiency on physical work capacity and selected muscle enzyme activities, forty female subjects were studied before and after eight weeks of supplementation with either oral iron or a matching placebo. Initially, female volunteers engaged in regular endurance running were screened for iron deficiency by blood analysis (serum ferritin and hemoglobin). Forty non-anemic subjects with deficient iron stores underwent physiological and anthropometric tests to obtain a comprehensive profile. The specific physical work capacity tests were alactic and lactacid power on the Wingate cycle ergometer test, lactacid capacity on the anaerobic speed test, anaerobic (ventilatory) threshold using gas exchange variables, V0₂ max. and the max. treadmill velocity during the V0₂ max. test. Muscle biopsy samples pre-, and post- treatment were assayed for citrate synthase and alpha-glycerophosphate dehydrogenase activity. Treatment was oral iron supplementation (320 mg ferrous sulfate = 100 mg elemental iron taken as SLOW-Fe® twice a day) or a matching placebo. The subjects were randomly assigned to one of the treatment groups and a double-blind method of administration of the supplements was used. It was hypothesized that work capacity would be enhanced following oral iron supplementation, possibly due to the repletion of iron containing oxidative enzymes important in energy production. Results could not strongly support this hypothesis with the difference between the two groups on the work capacity and enzyme activity variables being statistically nonsignificant. Serum ferritin values rose from a mean of 12.4+4.5 to 37.7+19.7 ngml⁻¹ for the experimental group and 12.2±4.3 to 17.2±8.9 for the controls; (p=0.0025). Hemoglobin levels remained fairly constant for both treatment groups; 13.4±0.6 to 13.5±0.5 gdl⁻¹ (experimental), and 13.0±0.6 to 13.1+0.5 (control); (p=0.6). Pre to post values on the work capacity variables, experimental vs control respectively were: Alactic power, 8.8 to 8.4 watts-kg⁻¹ body wt. vs 8.4 to 8.2; lactacid capacity, 6.9 to 6.9 watts-kg⁻¹ body wt. vs 7.0 to 6.0; anaerobic speed test, 41.3 to 45.1 seconds vs 43.7 to 44.8; anaerobic threshold, 7.4 to 7.5 mileshour⁻¹ vs 7.2 to 7.2; V0₂ max, 51.3 to 52.7 ml-kg⁻¹ min⁻¹ vs 50.6 to 50.6; max velocity during V0₂ max, 9.8 to 9.8 mileshour⁻¹ vs 9.6 to 9.5. Except for alactic power, the change in work capacity favored the iron treated group. Noting this trend, further study may be warranted. Prelatent/latent iron deficiency appeared not to depress the activities of the two enzymes measured. Cytoplasmic alpha-glycerophosphate dehydrogenase activity rose from 0.066 to 0.085 units for the experimental group (p=.58) vs .058 to .066 for the control group and citrate synthase activity changed from 0.047 to 0.048 (experimental) vs 0.039 to 0.042 (control). It can be concluded that eight weeks of iron supplementation to prelatent/latent iron deficient, physically active females does not significantly enhance work capacity nor the activity of 2 oxidative muscle enzymes (citrate synthase and cytoplasmic alpha-glycerophosphate dehydrogenase). Within the limitations of this study the presence of a serum ferritin below 20 ng-ml⁻¹ does not pose a significant handicap to anaerobic or aerobic capacity.

Background Recent studies have demonstrated a significant relationship between vitamin D and cholesterol levels. Cholesterol is important in the synthesis and metabolism of vitamin D. Additionally, foods rich in vitamin D also contain high cholesterol supplements. Given that sources of vitamin D and cholesterol are different, we hypothesized that there is a potential mean bias in true relationship between an individual’s vitamin D and cholesterol levels. The study therefore aimed to examine the relationship of serum cholesterol and vitamin D levels. Methods We utilized NHANES 2001-2006 data (n=30,954). The outcome was vitamin D deficiency defined as individuals with serum 25-hydroxyvitamin D level12 ng/ml were classified as normal. Significant differences in cholesterol levels between the two groups were determined using Chi-Square contingency test. Generalized linear models were conducted to assess the association of vitamin D deficiency with cholesterol levels adjusting for age, sex, physical activity (proxy measure for sunlight exposure) and other serum lipid levels. Results Vitamin D deficiency was significantly lower in males(5.2%) than females(7.8%)(P Conclusion Vitamin D deficiency was associated with high LDL and triglyceride levels. Study findings help establish the importance of distinguishing vitamin D and cholesterol levels as separate entities while making inferences about their relationship with chronic diseases.

ENGLISH ABSTRACT: Ostrich farming is of significant economical importance in South Africa. Three ostrich mycoplasmas, Ms01, Ms02 and Ms03 have been identified previously, and were provisionally named ‘Mycoplasma struthiolus’ (Ms) after their host Struthio camelus. Ostrich mycoplasmas are the major causative organisms of respiratory diseases, and they cause stock losses, reduced production and hatchability, and downgrading of carcasses and therefore lead to large economic losses to the industry. In order to be pathogenic to their host, they need to attach through an attachment organelle, the so-called tip structure. This structure has been identified in the poultry mycoplasma, M. gallisepticum, and is made up of the adhesin GapA and adhesin-related CrmA. Currently, no ostrich mycoplasma vaccine is commercially available and for this reason the need to develop one has arisen. Therefore the first part of this study was dedicated to the identification and isolation of vaccine candidate genes in the three ostrich mycoplasmas. Four primer approaches for polymerase chain reactions (PCR’s), cloning and sequencing, were used for the identification of adhesin or adhesin-related genes from Ms01, Ms02 and Ms03. The primer approaches revealed that the target genes could not be identified due to the high diversity of sequences that were generated. Therefore sequences were also compared with those of other mycoplasma species in BLAST searches. Results showed that the most significant hit was with the human pathogen M. hominis oppD, which is located in the same operon as the membrane protein P100 involved in adhesion. Other hits were with ABC transporters which may also play a role in cytadhesion. The second part of this study was aimed at testing whether two poultry mycoplasma vaccines, M. synoviae and M. gallisepticum, can be used in ostriches to elicit immune responses until an ostrich mycoplasma vaccine has been developed. Ostriches on three farms of different age groups in the Oudsthoorn district were therefore vaccinated with these vaccines in a vaccine trial. The enzymelinked immunosorbent assay (ELISA) was used to test the level of antibody response. Results showed that both vaccines elicited an immune response in all three age groups. A high percentage of the ostriches reacted positively, which indicates that both vaccines elicit antibody responses and may therefore give protection against ostrich mycoplasma infections.
AFRIKAANSE OPSOMMING: Volstruisboerdery is ‘n belangrike ekonomiese sektor in Suid-Afrika. Drie volstruismikoplasmas, Ms01, Ms02 en Ms03, is voorheen geïdentifiseer en voorlopig ‘Mycoplasma struthiolus’ (Ms) benaam na aanleiding van hul gasheer, Struthio camelus. Volstruismikoplasmas is die grootste oorsaaklike organismes van respiratoriese siektes, kudde verliese en die afgradering van karkasse wat lei tot groot ekonomiese verliese in die volstruisbedryf. Ten einde patogenies vir die gasheer te wees, moet mikoplasmas deur middel van ‘n aanhegtingsmeganisme vasheg – die sogenaamde puntvormige struktuur. Hierdie struktuur is in die pluimvee mikoplasma M. gallisepticum geïdentifiseer, en bestaan uit aanhegting proteïen GapA en die aanhegting verwante proteïen CrmA. Tans is geen volstruismikoplasma entstof kommersieel beskikbaar nie, en derhalwe het die behoefte ontstaan om so ‘n entstof te ontwikkel. Die eerste gedeelte van hierdie studie is dus gewy aan die identifisering en isolering van entstof kandidaat gene in al drie volstruismikoplasmas. Vier inleier benaderings vir polimerase ketting reaksies (PKR), klonering asook geenopeenvolging bepalings vir die identifisering van aanhegting of aanhegting verwante gene vanuit Ms01, Ms02 en Ms03 is gebruik. Die inleier benaderings het getoon dat die teikengene nie geïdentifiseer kon word nie as gevolg van hoë variasie in die gegenereerde geenopeenvolgings. Derhalwe is geenopeenvolgings met ander mikoplasma spesies deur middel van BLAST soektogte vergelyk. Resultate het getoon dat die betekenisvolste ooreenstemming dié met die menslike patogeen M. hominis oppD was, wat deel vorm van die membraan proteïen P100 operon wat betrokke is by aanhegting. Ander ooreenstemmings sluit ABC transporters in wat moontlik betrokke kan wees by aanhegting. Die tweede gedeelte van hierdie studie het ten doel gehad om te toets of twee pluimvee mikoplasma entstowwe, M. synoviae en M. gallisepticum, gebruik kan word in volstruise om immuunresponse te ontlok tot tyd en wyl ‘n volstruismikoplasma entstof ontwikkel is. Volstruise vanaf drie plase in verskillende ouderdomsgroepe in die Oudtshoorn distrik was ingeënt met hierdie entstowwe in ‘n entstof proefneming. Die ensiem-afhanklike immuno-absorpsie essaï (ELISA) was gebruik om antiliggaam response te toets. Die resultate het getoon dat beide entstowwe immuunresponse ontlok het in al drie ouderdomsgroepe. ‘n Groot persentasie van die volstruise het positief gereageer wat ‘n aanduiding is dat beide entstowwe immuunresponse ontlok het en kan dus beskerming bied teen volstruismikoplasma infeksies.

The ability of human growth hormone (hGH) to alleviate the effects of zinc deficiency on growth was investigated in the rat. Human GH treatment had no significant effect on food consumption, growth parameters or plasma IGF-I. Food consumption, tail length, liver weight, and tibia weight were significantly lower in the zinc deficient group. Body weight was significantly reduced in the zinc deficient and pair fed groups compared to the control. A significant interaction between zinc and hGH was found for tibial epiphyseal cartilage width but there were no significant differences between the groups receiving hGH and the respective shams. Tissue zinc content and plasma alkaline phosphatase were significantly decreased by the dietary zinc deficiency. Plasma zinc was higher in the groups receiving hGH. Significant interactions between zinc and hGH was found for liver iron, tibial zinc, and tibial copper. Copper and iron showed a competitive interaction with zinc and were lower in the rats receiving the control diet. Both lowered zinc and food intake contributed to the effects of the zinc deficiency; however, these effects were not equally distributed. Food intake had the greater effect on growth and plasma IGF-I while tissue mineral content showed a greater effect for zinc intake.

The very virulent (vv) pathotype of infectious bursal disease virus (IBDV) has spread rapidly throughout Europe, Asia, and the Middle East. Although Australia is currently unaffected, there remains the potential for incursion of an exotic isolate. The aim of this study was to identify putative virulence determinants of IBDV to facilitate the development of improved diagnostic assays for detection and characterisation of vvIBDV isolates. Sequencing of Indonesian vvIBDV Tasik94 revealed a unique substitution [ A¨S222] in the hypervariable region (HVR) of viral protein (VP) VP2, which did not appear to impinge on virulence or antigenicity. Phylogenetic analyses indicated that Tasik94 was closely related to Asian and European vvIBDV strains. Extensive alignment of deduced protein sequences across the HVR of VP2 identified residuesI242 I256 and I294 as putative markers of the vv phcnotype. Comparison of the pathology induced by mildly-virulent Australian IBDV 002/73 and Indonesian vvIBDV Tasik94, revealed that histological lesions in the spleen, thymus and bone marrow were restricted to Tasik94-infected birds, suggesting the enhanced pathogenicity of vvIBDV might be attributed to replication in non-bursal lymphoid organs. The biological significance of the VP2 HVR in virulence was assessed using recombinant viruses generated by reverse genetics. Both genomic segments of Australian IBDV 002/73, and recombinant segment A constructs in which the HVR of 002/73 was replaced with the corresponding region of either tissue culture-adapted virus or vvIBDV (Tasik94), were cloned behind T7 RNA polymerase promoter sequences. In vitro transcription/translation of each construct resulted in expression of viral proteins. Co-transfection of synthetic RNA transcripts initiated replication of both tissue culture-adapted parental and recombinant viruses, however attempts to rescue non-adapted viruses in specific-pathogen-free (SPF) chickens were unsuccessful. Nucleotide sequence variation in the HVR of VP2 was exploited for the development of a new diagnostic assay to rapidly detect exotic IBDV isolates, including vvIBDV, using reverse transcription polymerase chain reaction (RT-PCR) amplification and Bmrl restriction enzyme digestion. The assay was capable of differentiating between endemic and exotic IBDV in 96% of 105 isolates sequenced to date.

Interactions among zinc deficiency, energy restriction, immunity and Heligmosomoides polygyrus (Nematoda) infection in mice were investigated. Mice were fed zinc-sufficient (Zn+: 60mg zinc/kg diet), zinc-deficient (Zn$-$: 0.75 mg/kg) or pair-fed (PF) diets. Liver and serum zinc concentrations were reduced in Zn$-$ mice, but zinc content of worms was unaffected by host diet. Both zinc deficiency and energy restriction accelerated worm maturation during a primary infection, but zinc deficiency caused stunting of female worms. Energy restriction led to a anteriorad distribution of worms along the intestine whereas zinc deficiency led to a posteriorad distribution. Enhanced parasite survival in Zn$-$ and PF mice was associated with a decreased immune response in both primary and challenge infections. In the primary infection, spleen cell production of interleukin-4 (IL-4) and interferon-gamma (IFN-$ gamma$), peripheral eosinophilia and serum levels of IgE and IgG1 were reduced by zinc deficiency, whereas the reduced delayed type hypersensitivity (DTH) response and impaired interleukin-5 (IL-5) production were attributed to energy restriction. In the challenge infection, reduced antibody levels (total IgE and IgG1, parasite-specific IgG1 and IgG3) and eosinophilia were attributed to zinc deficiency whereas the higher worm numbers were attributed to a combined effect of zinc deficiency and energy restriction. Although the absolute number of spleen cells was reduced in both Zn$-$ and PF mice, neither deficiency altered the proportion of the cell subpopulations. In vitro functional assays in response to parasite antigen (Ag) revealed that T cell proliferation was reduced by a direct effect of zinc deficiency on T cells, and by the effects of energy restriction on antigen presenting cells (APC). The problem of this study is to determine the vitamin B-6 status of patients who have chronic obstructive pulmonary disease (COPD). Erythrocyte aspartate transaminase assay was the method for measuring vitamin B-6 status. The vitamin B-6 status was examined in thirty subjects (ten COPD subjects and twenty control subjects). An unpaired t-test was used to compare the vitamin B-6 status of the COPD group versus the control group. Four determinants (percentage stimulation, ratio of basal to stimulated activity, basal activity, and stimulated activity) were used to determine vitamin B-6 status in both groups of subjects. Percentage stimulation and ratio of basal to stimulated activity were not significantly different (control group versus COPD group) at the .05 level. However, two of ten COPD subjects had values for percentage stimulation that were two standard deviations above the mean, indicating a poor B-6 status. In contrast, basal activity and stimulated activity of erythrocyte aspartate transaminase were found to be significantly lower at the .05 level in the COPD group than the control group. Therefore, the COPD subjects as a group had some biochemical characteristics of a lower level of vitamin B-6 than the controls.

This study describes and compares the results of a survey of 53 masters of social work students to determine the nature and extent to which their educational preparation included instruction on acquired immune deficiency syndrome and sexually transmitted diseases. The study also assessed the social work students' attitudes toward people with HIV/AIDS and STDs, as well as their attitudes toward the role of social work education and the profession in addressing the HIV/AIDS and STD epidemics. To obtain this purpose the investigator developed a 56-item questionnaire. One of five hypotheses generated was accepted. Results showed that social work students need to increase their exposure to HIV/AIDS and STD related issues.

The effect of alcohol and/or zinc deficiency was evaluated in seven groups of pregnant rats and their pups. Females which had been acclimated to alcohol before breeding were fed liquid alcohol diets with either 14 or 0.1 ppm Zn. Comparisons were made with animals pair fed isocaloric liquid carbohydrate diets with the same Zn levels. Other comparisons were made by pair feeding a high zinc diet to a low zinc diet, and by feeding a high zinc diet ad lib. A reduced food intake and Zn deficiency affected maternal status by decreasing weight gain, liver Zn and plasma Zn concentration. Litter size, litter weight, and fetal liver and brain weight were decreased only in the alcohol zinc deficient group compared to adequately fed controls. The concentration and total quantity of fetal liver Zn were decreased due to a Zn deficiency. The combination of Zn deficiency and alcohol decreased only total Zn in fetal brain. The concentrations of protein, DNA, and RNA in fetal liver and brain were similar regardless of dietary treatment. The quantities of protein, DNA, and RNA were decreased in fetal liver due to Zn deficiency. ln fetal brain, only the combination of alcohol and Zn deficiency decreased total protein and RNA, while DNA was not affected. Although alcohol by itself had no effect on the above variables, its combination with a Zn deficiency did. In addition, there were 58 resorptions and 15 malformations seen in Zn deficient alcoholic dams compared with no more than 15 and 2, respectively, in any of the other groups. Teratogenesis caused by a Zn deficiency was increased with alcohol consumption.

Experiments were conducted with sows and littermate-gilts to investigate the effect of dietary zinc (Zn) restriction during either part or most of the pregnancy and lactation on dams and their offspring.
Reduced dietary Zn (13 ppm) during the last 4 wk of pregnancy and first 2 wk of lactation presented no serious consequences to the dam or offspring. Reduction of dietary Zn throughout pregnancy and lactation (10 ppm) caused overt signs of Zn deficiency in one-half of the gilts, however, their offspring exhibited no obvious abnormalities. Zinc restriction did not affect feed consumption but depressed plasma and urinary Zn and plasma alkaline phosphatase activity (APA). The use of the balance technique and radiotracers revealed that gilts deficient in Zn initiated protective mechanisms such as, increased Zn absorption reduced endogenous Zn excretion, prolongation of the biological half life of Zn; all these responses affected higher efficiency of Zn utilization.
Despite the protective mechanisms, gilts depleted in Zn exhibited prolonged stressful parturition and a high incidence of intrapartum and neonatal mortality than control gilts. Piglets from gilts depleted in Zn showed depressed birth weight, reduced concentrations of plasma Zn, Cu and ammonia and low APA and hematocrit, increased plasma concentrations of glucose and lactate, reduced content of Zn in liver, heart, lung, skin, bone and carcass and higher concentrations of Cu and Mn in various tissues compared to controls. Zinc depletion did not change the Zn content of colostrum but did change it in milk. Content of Cu, Fe, and Mn was effected in colostrum and milk. Weight of piglets was lower at one wk but not at two wk of age. Two-week old piglets exhibited lower Zn and higher Cu concentrations in plasma; reduced Zn and increased Cu and Mn concentrations in liver and bone and changes in Cu and Mn in other tissues. At the end of lactation, gilts depleted in Zn exhibited low level of Zn and APA in plasma; reduced Zn content in liver, spleen, heart, small intestine; increased Cu content in liver and small intestine; increased Zn and Mn content in brain.
Although gilts depleted in Zn exhibited a highly efficient handling of Zn via homeostatic mechanisms, the reduced amount of corporal exchangeable Zn in these gilts caused biochemical changes affecting the performance of the dam and the offspring. (Abstract shortened with permission of author.)

Zinc status of Egyptian children 18-30 months and 6-10 years of age was characterized in relation to morbidity, growth, and socioeconomic variables. In a pilot study of children whose general nutrition ranged from adequately nourished to moderately malnourished, mean hair zinc was 135 ug/g (63-230 ug/g), with suboptimal zinc status suggested for 44%. Predictors of hair and serum zinc levels were explored for 23 school-aged and 40 preschool children. Included in models were weaning age for preschool children, body size (length- or height- and weight-for-age Z scores), current growth over 6 months or longer, illness experience over 10 to 12 months, demographic variables affecting food availability and distribution, sex, and season. Data were collected by Egyptian workers as part of a larger field project. Hair and serum samples were analyzed for zinc content by atomic absorption spectrophotometry. Results showed no difference in hair zinc levels by color, presence or absence of louse egg fragments and mucilage, or presence or absence of henna dye. In multiple regression models, the best predictor of hair zinc in preschool children was season of year, with zinc lower in summer. Season, negative effect of percent of weeks ill with diarrhea, and positive effects of socioeconomic status (SES) based on father's education/literacy and economic subsistence base excluding agriculture (ESB-A) predicted 23% of total hair zinc variation in preschool children. In preschool children serum zinc was lower in summer. Season, positive effect of rate of weight increase, and negative effects of rate of height increase, SES based on father's occupation(s) (SES2), and ESB-A predicted 53% of total serum zinc variation in preschool children. Serum zinc was higher in summer in school-aged children. Season, negative effect of SES2 and ESB-A, and positive effects of percent weeks ill with diarrhea and height for age Z scores predicted 60% of total serum zinc variation in school-age children. Negative effects of percent weeks ill with diarrhea and parents' age and child:adult ratio predicted 29% of hair zinc in school-aged children.
