In the introductory chapter of his essay on Utilitarianism, John Stuart Mill says his aim is to contribute towards the understanding of utilitarianism and towards ‘such proof as it is susceptible of’. He immediately adds that ‘this cannot be proof in the ordinary and popular meaning of the term’ because ‘ultimate ends are not amenable to direct proof’. A proof that something is good has to show that it is ‘a means to something admitted to be good without proof’. But, he goes on, this does not imply that a formula of ultimate ends can only be accepted on ‘blind impulse, or arbitrary choice’. It can be rationally discussed and subjected to proof in a wider sense of that word. ‘Considerations may be presented capable of determining the intellect either to give or withhold its assent to the doctrine; and this is equivalent to proof.’

The register of the Scots College, Douai, was printed in Records of the Scots Colleges I, New Spalding Club (Aberdeen, 1906).1 It ends in 1772, but does not contain the names of the secular students after 1765, when the Jesuits were expelled from France. The rector, John Riddoch, S. J., and his students moved to Dinant, taking the register with them, and it is the Dinant register that is printed. The college at Douai re-opened with a secular priest, Robert Grant, as rector, but no register survives for the period from 1765 until the final closure in 1793.2

The last fourteen years of John Stuart Mill's life (1859-1873), which followed the death of Harriet Taylor, possessed a hefty political content. They saw the publication of his essays on parliamentary reform and Considerations on Representative Government, his impassioned identification with the North in the American Civil War, the eventful parliamentary career sandwiched between the Westminster elections of 1865 and 1868, and a final phase of activity associated with causes such as women's suffrage and land tenure reform. When Mill acted politically he usually did so with strong feeling, but in his search to give deeply held principles practical effect he understood the need for dispassionate adaptation of means to ends. Both the feeling and the adaptation are evident in his treatment of the elementary education question in 1870, a treatment that vividly illustrates how Mill operated during the decade and a half before his death.Of the host of legislation Gladstone's first administration proposed, only one item, the 1870 Education Bill, elicited a congregation of public responses from Mill. Of course, Mill's political activity in the several years following his defeat at Westminster in autumn 1868 was not confined to the adoption of a stance on ministerial measures. With respect to women's suffrage and land reform Mill was not about to wait on any government, and his conspicuous connections with the National Society for Women's Suffrage and the Land Tenure Reform Association attracted notice at the time and have been the subject of comment since. Moreover, during his last years Mill continued to cultivate his contacts in the world of London working-class radicalism, particularly with George Odger, William Randal Cremer, and George Howell. Whereas Mill's parliamentary career has been explored in some detail, the political character of his post-Westminster years has received less attention.

We previously described a cis-acting RNA-cleaving deoxyribozyme known as pH3DZ1 that exhibits optimal catalytic activity at pH 3.0 (Zhongjie Liu, Shirley H. Mei, John D. Brennan, and Yingfu Li. J. Am. Chem. Soc. 125, 7539 (2003)). This DNA catalyst was made of a 99-nucleotide (nt) catalytic domain covalently linked to a 23-nt DNA–RNA chimeric substrate containing a single ribonucleotide as the cleavage site. In the present work, we conducted an extensive sequence examination of this deoxyribozyme via nucleotide truncation and reselection experiments, with a goal to minimize its size and identify the nucleotides that are crucial to its catalytic function. A trans-acting deoxyribozyme that can process an external substrate was also successfully designed. Stretches of 30 and 17 nucleotides from the 5′ and 3′ ends of the trans catalyst, respectively, were found to be completely dispensable; in contrast, few nucleotides could be deleted internally without producing a detrimental effect. The reselection experiment led to the discovery of 7 and 5 absolutely conserved nucleotides located at the 5′ and 3′ ends of the minimized catalyst, respectively, separated by a 31-nt element in which 14 highly conserved nucleotides were scattered among 17 variable nucleotides. The shortened deoxyribozyme and the original catalyst showed a similar pH profile with the optimal activity at pH 3; however, the minimized deoxyribozyme still exhibited strong catalytic activity at pH 2.5, while the full-length catalyst was barely active at this pH. Finally, it was found that this deoxyribozyme generated two cleavage fragments, one with 2′,3′-cyclic phosphate and the other with 5′-OH.Key words: DNA, deoxyribozyme, RNA cleavage, in vitro selection, catalysis.

ABSTRACT Cleavage by the V(D)J recombinase at a pair of recombination signal sequences creates two coding ends and two signal ends. The RAG proteins can integrate these signal ends, without sequence specificity, into an unrelated target DNA molecule. Here we demonstrate that such transposition events are greatly stimulated by—and specifically targeted to—hairpins and other distorted DNA structures. The mechanism of target selection by the RAG proteins thus appears to involve recognition of distorted DNA. These data also suggest a novel mechanism for the formation of alternative recombination products termed hybrid joints, in which a signal end is joined to a hairpin coding end. We suggest that hybrid joints may arise by transposition in vivo and propose a new model to account for some recurrent chromosome translocations found in human lymphomas. According to this model, transposition can join antigen receptor loci to partner sites that lack recombination signal sequence elements but bear particular structural features. The RAG proteins are capable of mediating all necessary breakage and joining events on both partner chromosomes; thus, the V(D)J recombinase may be far more culpable for oncogenic translocations than has been suspected.

Sphingosine 1-phosphate (S1P) is a platelet-derived sphingolipid that elicits numerous biological responses in endothelial cells mediated by a family of G protein-coupled EDG receptors. Stimulation of EDG receptors by S1P has been shown to activate the endothelial isoform of nitric-oxide synthase (eNOS) in heterologous expression systems (Igarashi, J., and Michel, T. (2000)J. Biol. Chem.275, 32363–32370). However, the signaling pathways that modulate eNOS regulation by S1P/EDG in vascular endothelial cells remain less well understood. We now report that S1P treatment of bovine aortic endothelial cells (BAEC) acutely increases eNOS enzyme activity; the EC50for S1P activation of eNOS is ∼10 nm. The magnitude of eNOS activation by S1P in BAEC is equivalent to that elicited by the agonist bradykinin. S1P treatment activates Akt, a protein kinase implicated in phosphorylation of eNOS. S1P treatment of BAEC leads to eNOS phosphorylation at Ser1179, a residue phosphorylated by Akt; an eNOS mutant in which this Akt phosphorylation site is inactivated shows attenuated S1P-induced eNOS activation. S1P-induced activation both of Akt and of eNOS is inhibited by pertussis toxin, by the phosphoinositide 3-kinase inhibitor wortmannin, and by the intracellular calcium chelator BAPTA (1,2-bis(aminophenoxy)ethane-N,N,N′,N′-tetraacetic acid). By contrast to S1P, activation of G protein-coupled bradykinin B2 receptors neither activates kinase Akt nor promotes Ser1179eNOS phosphorylation despite robustly activating eNOS enzyme activity. Understanding the differential regulation of protein kinase pathways by S1P and bradykinin may lead to the identification of new points for eNOS regulation in vascular endothelial cells.

The H2AX core histone variant is phosphorylated in chromatin around DNA double strand breaks (DSBs) and functions through unknown mechanisms to suppress antigen receptor locus translocations during V(D)J recombination. Formation of chromosomal coding joins and suppression of translocations involves the ataxia telangiectasia mutated and DNA-dependent protein kinase catalytic subunit serine/threonine kinases, each of which phosphorylates H2AX along cleaved antigen receptor loci. Using Abelson transformed pre–B cell lines, we find that H2AX is not required for coding join formation within chromosomal V(D)J recombination substrates. Yet we show that H2AX is phosphorylated along cleaved Igκ DNA strands and prevents their separation in G1 phase cells and their progression into chromosome breaks and translocations after cellular proliferation. We also show that H2AX prevents chromosome breaks emanating from unrepaired RAG endonuclease-generated TCR-α/δ locus coding ends in primary thymocytes. Our data indicate that histone H2AX suppresses translocations during V(D)J recombination by creating chromatin modifications that stabilize disrupted antigen receptor locus DNA strands to prevent their irreversible dissociation. We propose that such H2AX-dependent mechanisms could function at additional chromosomal locations to facilitate the joining of DNA ends generated by other types of DSBs.

Enos John Nganani Mabuza's life and work is used as a case study to highlight the conceptual and methodological theories and challenges in academic biographical writing regarding history, memory and legacies. This thesis answers the question: Who was Mabuza and what is his place or relevance in South Africa's history? We over-simplify and stereotype people because it is convenient. We judge others but there is no saint without a past or a sinner without a future. Mabuza reconciled with the main liberation movements and moved from a maligned position on the political fringes to a respectable one within the emerging black economic elite. History, like reference systems and values is not calcified or static. It is prone to interpretation, adaptation, modification, invention, manipulation, decomposition and re-composition. Bourdieu's habitus-field analysis, theory or logic of practice, notions of capital (symbolic, political, social, cultural and economic) is used together with Latour's actor-network theory as the basis of analysis of the social contracts and trust bonds that Mabuza was able to create and which enabled him to navigate South Africa's socio-political and economic milieus during apartheid and the transition in the early 1990s. As people or actors, we believe in the mantra, nothing ventured, nothing gained. We exercise agency and take risks every day. We make choices and those choices have consequences. Mabuza's choices in the fields of education, politics and business had implications for how he is perceived or has been written into history. His choices put notions of identity, citizenship, power, legitimacy, ambition, elite accommodation, class, personal and professional networks, popular struggles, agency and structure under the spotlight. Mabuza's involvement in Bantustan politics, for instance, is contextualised in terms of a historical overview of the unpopular role played by traditional authorities in South Africa before and during colonialism and apartheid. His later foray into the world of business, however, was facilitated in part, by the personal and professional contacts that he made whilst he was in politics and the opportunities which opened up during the country's political transition. Mabuza adapted to changing circumstances and demonstrated a level of versatility which other Bantustan functionaries did not or could not exercise.

Abstract (sommario):
This thesis is a study of the theological views of a leading Victorian evangelical Anglican, Bishop John Charles Ryle. The main sources for the study are the bishop's extensive writings, which run to some seven thousand pages. Ryle's writings, for the most part, are occasional, pastoral and evangelistic, but they are suffused with theological concern, to the extent that a "systematic theology" can almost be compiled from them. Because of this, the method adopted in this work is to study Ryle's writings according to the traditional theological "loci," and to analyse his understanding of each of them. The study also seeks to make the analysis in the light of the historical theological and ecclesiastical background to his writings, particularly as Ryle frequently interacted with the various disparate elements that comprised the Victorian religious scene. The most extensive studies in this thesis relate to Ryle's exposition of the distinctive emphases of Evangelicalism, viz. the Inspiration and Authority of Scripture, human depravity, the Atonement, and the saving and sanctifying work of the Holy Spirit. Other important areas are the sacraments, in which Ryle engages in a strong polemic against the Anglo-Catholics of his day, and the doctrine of the Church. The thesis emphasises some characteristics of Ryle's theology, in particular its biblical basis, traditional orthodoxy, evangelicalism and moderate Calvinism. An interesting feature of Ryle's work is his polemic, which is directed against the newly emerging liberals, and the increasingly influential Anglo-Catholics. We also take note of Ryle's style of presentation, including his simplicity of expression, strong pastoral application, and his didactic method of inculcation by repetition. The thesis concludes with a resume of his work, a description of its character, together with a constructive critique and evaluation, and an indication of Ryle's importance for our own day.

The present work approaches the matter of the Law of Property and its social function according to John Rawls theory of justice, having the intention to understand what the role of the property is to the author, the ways of property which are considered fair by him and, if it answers to the principles of justice and property which fitted with its social purpose. For such, it deals with, firstly, in chapter 1, the conteporary conception of law of property, which is inserted within a more humanitary, cooperative pespective of society and reciprocity, longing to a socialeconomical harmony and the effectiviness of certain human necesseties to improve the pespective of the citizens lives. Carrying on in this chapter, it is studied Locke s concept of private property to demonstrate the points where his theory disagree with John Rawls theory, thus it brings more individual view of property and it adopts a restricted conception of this right. Locke defends the unlimited accumulation of wealth, being the property turned, only, to the individual growth of the owner and to benefit the business, and then not existing a distributive and cooperative justice. Macpherson criticizes Locke s theory since it is extremely individualist. Chapter 2 aims to analyse the main ideas of Rawls theory of justice, seaching for the main and fundamental concepts so that chapter 3 is well-understood. Rawls supports the distributive justice and encourages the social cooperation. For this author, the basic liberties are inegotiable and the object of his theory is the basic structure of the society as an equitative system of social cooperation among free and iqual citizens. In order to reach this, the individuals make an agreement under the veil of ignorance to formulate the principles of justice which will run the institutions of a democratic constitutional fair society. Chapter 3 fights against the matter of the law of property for this author, a big suppoter of the distributive justice and of the social cooperation, which understands the law of property as basic good, because it helps to accomplish the expectations of the citizens lives, and it must also be capatible to the other rights and liberties which belong to a social system to provide a honored life. Rawls thought was a target for criticism by Robert Nozick, whom rescued Locke s theory of appropriation, not agreeing with the notion of distributive justice and of social cooperation, counteracting in several points to the Rawls theory of justice. However, according to the philosophers Álvaro de Vita, Will Kimlicka and Van Parijs, the defense of John Rawls theory was made, because they support the thought of this author, abova all objections shown by Nozick. Thus, the right of property must have a social mean to promote certain basic needs of the individuals, and this is clear in Rawls work so that he believes in social cooperation, in the effectiviness of human dignity, of self-respect, of self-steem and of their own citizenship, through a distributive justice.
O presente trabalho aborda a questão do direito de propriedade e sua função social a partir da teoria da justiça de John Rawls, com a intenção de compreender qual o papel da propriedade para o autor, as formas de propriedade consideradas justas para ele e, se atende aos princípios da justiça a propriedade que cumpre com uma finalidade social. Para tanto, trata-se, primeiramente, no capítulo 1, a concepção contemporânea de direito de propriedade, o qual está inserido numa perspectiva mais humanitária, cooperativa, de sociabilidade e reciprocidade, buscando a harmonia econômico-social e a efetivação de certas necessidades humanas para melhorar as perspectivas de vida dos cidadãos. Dando continuidade a este capítulo, é estudado o conceito de propriedade privada de Locke, com a finalidade de demonstrar os pontos em que sua teoria contrapõe-se à teoria de John Rawls, pois traz uma visão mais individualista de propriedade e adota uma concepção restrita deste direito. Locke defende a acumulação ilimitada de riquezas, estando a propriedade voltada, unicamente, para o crescimento individual do proprietário e para beneficiar o comércio, inexistindo uma justiça distributiva e cooperativa. Macpherson critica a teoria de Locke por ser extremamente individualista. O capítulo 2 intenciona analisar as principais idéias da teoria da justiça de Rawls, buscando-se os conceitos principais e fundamentais para que o capítulo 3 seja mais bem compreendido. Rawls defende a justiça distributiva e incentiva a cooperação social. Para este autor, as liberdades básicas são inegociáveis e o objeto de sua teoria é a estrutura básica da sociedade como um sistema equitativo de cooperação social entre cidadãos livres e iguais. Para que isso seja alcançado, os indivíduos realizam um acordo, sob o véu da ignorância , para formular os princípios da justiça que regerão as instituições de uma sociedade constitucional democrática justa. O capítulo 3 enfrenta a questão do direito de propriedade para o autor, grande defensor da justiça distributiva e da cooperação social, que entende o direito de propriedade como um bem básico, pois auxilia nas realizações das expectativas de vida dos cidadãos, devendo ser compatível com os demais direitos e liberdades pertencentes a um sistema social para proporcionar uma vida digna. O pensamento de Rawls foi alvo de crítica de Robert Nozick, o qual resgatou a teoria da apropriação de Locke, não concordando com a noção de justiça distributiva e de cooperação social, contrapondo-se em inúmeros pontos à teoria da justiça ralwsiana. Mas, com base nos filósofos Álvaro de Vita, Will Kymlicka e Van Parijs foi realizada a defesa da teoria de John Rawls, pois defendem o pensamento deste autor, acima das objeções apresentadas por Nozick. Asim, o direito de propriedade deve ter uma finalidade social para promover certas necessidades básicas dos indivíduos, e isso, percebe-se na obra de Rawls, pois ele acredita na cooperação social, na efetivação da dignidade humana, do auto-respeito, da auto-estima e da própria cidadania, por meio de uma justiça distributiva.
