Abstract (sommario):
AbstractA lncRNA RP1-85F18.6 was reported to affect cell growth by regulating the cell cycle. Here we tested whether it affects the proliferation of osteoblast cells by regulating the cell cycle. We determined the expression of RP1-85F18.6 in two osteoblast cell lines hFOB and HOB by qPCR. Then we knocked down or overexpressed RP1-85F18.6 in hFOB and tested the alteration of viability, cell cycle, and cell cycle regulatory proteins. Results showed that both hFOB and HOB expressed RP1-85F18.6. The knockdown of RP1-85F18.6 decreased the viability of hFOB, while the overexpression of it increased the viability. Higher expression of RP1-85F18.6 results in higher cell viability. The knockdown of RP1-85F18.6 caused an increase in the S phase cells and a decrease in the G2/M phase cells. The overexpression of RP1-85F18.6 caused a decrease in the S phase cells and an increase in the G2/M phase cells. The knockdown of RP1-85F18.6 decreased cyclin A, cdk1, E2F, cyclin B, p53, and p21, whereas the overexpression of RP1-85F18.6 increased cyclin A, cdk1, E2F, cyclin B, p53, and p21. This study demonstrated that RP1-85F18.6 is expressed in osteoblast cell lines hFOB and HOB. RP1-85F18.6 affects the proliferation of osteoblasts by regulating the cell cycle.

Abstract Context: Ketone bodies are substrates during fasting and when on a ketogenic diet not the least for the brain and implicated in the management of epileptic seizures and dementia. Moreover, D-β-hydroxybutyrate (HOB) is suggested to reduce blood glucose and fatty acid levels. Objectives: The objectives of this study were to quantitate systemic, cerebral, and skeletal muscle HOB utilization and its effect on energy metabolism. Design: Single trial. Setting: Hospital. Participant: Healthy post-absorptive males (n = 6). Interventions: Subjects were studied under basal condition and three consecutive 1-hour periods with a 3-, 6-, and 12-fold increased HOB concentration via HOB infusion. Main Outcome Measures: Systemic, cerebral, and skeletal muscle HOB kinetics, oxidation, glucose turnover, and lipolysis via arterial, jugular, and femoral venous differences in combination with stable isotopically labeled HOB, glucose, and glycerol, infusion. Results: An increase in HOB from the basal 160–450 μmol/L elicited 14 ± 2% reduction (P = .03) in glucose appearance and 37 ± 4% decrease (P = .03) in lipolytic rate while insulin and glucagon were unchanged. Endogenous HOB appearance was reduced in a dose-dependent manner with complete inhibition at the highest HOB concentration (1.7 mmol/L). Cerebral HOB uptake and subsequent oxidation was linearly related to the arterial HOB concentration. Resting skeletal muscle HOB uptake showed saturation kinetics. Conclusion: A small increase in the HOB concentration decreases glucose production and lipolysis in post-absorptive healthy males. Moreover, cerebral HOB uptake and oxidation rates are linearly related to the arterial HOB concentration of importance for modifying brain energy utilization, potentially of relevance for patients with epileptic seizures and dementia.

Abstract (sommario):
This paper aims to develop of a robust analytical approach for computation of the principal design parameters for a special-purpose involute hob for machining of cluster gear. The specific combination of design parameters for a precision hob with particular value of the hob-setting angle is given. This combination of parameters includes (a) the hob outside diameter, (b) the base diameter of the hob, and (c) the required value of the hob-setting angle. The specific combination of these design parameters for the involute hob determined here allows machining of cluster gear with the shortest possible neck width. Numerous computer codes for computation of the design parameters of an involute hob are developed using the commercial software MathCAD-Scientific. Application of the special-purpose hob would allow reduction of size and weight of both the cluster gear itself and the whole gear transmission including housing. The reported results of the research are ready for application in industry.

AbstractObjective:To ascertain opinions regarding etiology and preventability of hospital-onset bacteremia and fungemia (HOB) and perspectives on HOB as a potential outcome measure reflecting quality of infection prevention and hospital care.Design:Cross-sectional survey.Participants:Hospital epidemiologists and infection preventionist members of the Society for Healthcare Epidemiology of America (SHEA) Research Network. Methods:A web-based, multiple-choice survey was administered via the SHEA Research Network to 133 hospitals. Results:A total of 89 surveys were completed (67% response rate). Overall, 60% of respondents defined HOB as a positive blood culture on or after hospital day 3. Central line-associated bloodstream infections and intra-abdominal infections were perceived as the most frequent etiologies. Moreover, 61% thought that most HOB events are preventable, and 54% viewed HOB as a measure reflecting a hospital’s quality of care. Also, 29% of respondents’ hospitals already collect HOB data for internal purposes. Given a choice to publicly report central-line–associated bloodstream infections (CLABSIs) and/or HOB, 57% favored reporting either HOB alone (22%) or in addition to CLABSI (35%) and 34% favored CLABSI alone. Conclusions:Among the majority of SHEA Research Network respondents, HOB is perceived as preventable, reflective of quality of care, and potentially acceptable as a publicly reported quality metric. Further studies on HOB are needed, including validation as a quality measure, assessment of risk adjustment, and formation of evidence-based bundles and toolkits to facilitate measurement and improvement of HOB rates.

Includes bibliographical references (p. 157-164).
This dissertation introduces novel techniques for verifying that programs conform to their designs. My Hob system, as described in this dissertation, allows developers to statically ensure that implementations preserve certain specified properties. Hob verifies heap-based properties that can express important aspects of a program's design. The key insight behind my approach is that Hob can establish detailed software design properties--properties that lie beyond the reach of extant static analysis techniques due to scalability or precision issues-by focusing the verification task. In particular, the Hob approach applies scalable static analysis techniques to the majority of the modules of a program and very precise, unscalable, static analysis or automated theorem proving techniques to certain specific modules of that program: those that require the precision that such analyses can deliver. The use of assume/guarantee reasoning allows the analysis engine to harness the strengths of both scalable and precise static analysis techniques to analyze large programs (which would otherwise require scalable, imprecise analyses) with sufficient precision to establish detailed data structure consistency properties, e.g. heap shape properties.
(cont.) A set-based specification language enables the different analysis techniques to cooperate in verifying the specified design properties. My preliminary results show that it is possible to successfully verify detailed design-level properties of benchmark applications: I have used the Hob system to verify user-relevant properties of a water molecule simulator, a web server, and a minesweeper game. These properties constrain the behaviour of the program by stating that selected sets of objects are always equal or disjoint throughout the program's execution.

Abstract (sommario):
The method of manufacture of gear on a large scale is based on process that use cutting tools, called Hob or “Caracol”. This method allows the speed and accuracy. The machine and tool designed for this process are specific for application in the manufacture of toothed gears and shafts. This making it necessary insight into the optimization of these tools. Failure to replace the tool at the right time involves two problems: or it can be exchanged before the end of life, or it can be overused and form the so-called excessive wear, which reduces the number of cutter sharpenings. The point of study from which it comes this work is to determine the quantity of parts to be produced with each of these tools, it know that the same tool can be applied to various types of pieces, ranging in diameter and thickness, which characterizes the complexity of this process. Therefore data about machining tool wear depending on the size of the pair part / tool were plotted collected. After that, standard curves of wear in relation to the pair part / tool and by this standard curve, to determine the life of the cutters in quantities of parts to be manufactured. Soon after, the tests performed could proved the effectiveness of this correlation.

Abstract (sommario):
Hop count, i.e., the number of wireless hops a packet has to go through to reach the destination, is a fundamental metric in multi-hop wireless ad-hoc networks. Network performance, such as throughput, end-to-end delay, energy consumption, and so on, depends critically on hop count. Previous work on modeling hop count is limited in making unrealistic simplifying assumptions either at the physical or network, or both layers of the communication protocol stack. A key contribution of this thesis is to present an analytical model to derive the probability distribution of hop count under realistic assumptions at both physical and network layers. Specifically, the model considers a log-normal shadowing radio propagation capable of accommodating the random signal fading observed in most wireless communication environments, and the widely used geographic routing at the network layer. Validation of the model is achieved by a comprehensive set of simulation experiments including a trace driven simulation of a real-word vehicular ad-hoc network. The model reveals that the presence of randomness in radio propagation reduces the required number of hops to reach a given destination significantly. To demonstrate the utility of the proposed hop count model, the thesis proposes three new applications which address some of the key challenges in multi-hop wireless networks. The first application derives the per-node packet forwarding load in multi-hop wireless sensor networks and reveals that the nodes in the vicinity of the base station has a significantly less forwarding load than previously thought under simplifying radio propagation and routing assumptions. The second application demonstrates that using hop count as a measure of distance traveled by a data packet, geocasting can be achieved in multi-hop wireless networks in situations when some of the network nodes do not have access to reliable location information. Finally, the proposed hop count model is used to evaluate the performance of the third application which demonstrates that the overhead of geographic routing can be reduced significantly by embracing a position update philosophy which adapts to the mobility and communication patterns of the underlying ad-hoc network.

Abstract (sommario):
Unlike traditional wireless networks, characterized by the presence of last-mile, static and reliable infrastructures, Mobile ad Hoc Networks (MANETs) are dynamically formed by collections of mobile and static terminals that exchange data by enabling each other's communication. Supporting multi-hop communication in a MANET is a challenging research area because it requires cooperation between different protocol layers (MAC, routing, transport). In particular, MAC and routing protocols could be considered mutually cooperative protocol layers. When a route is established, the exposed and hidden terminal problems at MAC layer may decrease the end-to-end performance proportionally with the length of each route. Conversely, the contention at MAC layer may cause a routing protocol to respond by initiating new routes queries and routing table updates. Multi-hop communication may also benefit the presence of pseudo-centralized virtual infrastructures obtained by grouping nodes into clusters. Clustering structures may facilitate the spatial reuse of resources by increasing the system capacity: at the same time, the clustering hierarchy may be used to coordinate transmissions events inside the network and to support intra-cluster routing schemes. Again, MAC and clustering protocols could be considered mutually cooperative protocol layers: the clustering scheme could support MAC layer coordination among nodes, by shifting the distributed MAC paradigm towards a pseudo-centralized MAC paradigm. On the other hand, the system benefits of the clustering scheme could be emphasized by the pseudo-centralized MAC layer with the support for differentiated access priorities and controlled contention. In this thesis, we propose cross-layer solutions involving joint design of MAC, clustering and routing protocols in MANETs. As main contribution, we study and analyze the integration of MAC and clustering schemes to support multi-hop communication in large-scale ad hoc networks. A novel clustering protocol, named Availability Clustering (AC), is defined under general nodes' heterogeneity assumptions in terms of connectivity, available energy and relative mobility. On this basis, we design and analyze a distributed and adaptive MAC protocol, named Differentiated Distributed Coordination Function (DDCF), whose focus is to implement adaptive access differentiation based on the node roles, which have been assigned by the upper-layer's clustering scheme. We extensively simulate the proposed clustering scheme by showing its effectiveness in dominating the network dynamics, under some stressing mobility models and different mobility rates. Based on these results, we propose a possible application of the cross-layer MAC+Clustering scheme to support the fast propagation of alert messages in a vehicular environment. At the same time, we investigate the integration of MAC and routing protocols in large scale multi-hop ad-hoc networks. A novel multipath routing scheme is proposed, by extending the AOMDV protocol with a novel load-balancing approach to concurrently distribute the traffic among the multiple paths. We also study the composition effect of a IEEE 802.11-based enhanced MAC forwarding mechanism called Fast Forward (FF), used to reduce the effects of self-contention among frames at the MAC layer. The protocol framework is modelled and extensively simulated for a large set of metrics and scenarios. For both the schemes, the simulation results reveal the benefits of the cross-layer MAC+routing and MAC+clustering approaches over single-layer solutions.

Abstract (sommario):
The double enveloping hourglass worm gear pair is a type of transmission, which has many traits, such as more teeth in mesh for transmission, higher contact quality, better lubrication condition and so on. Therefore, it has higher efficiency of transmission, stronger bearing capacity and longer work life. But the manufacturing difficulty of the worm gear hob has limited its popularization and application. Unlike the traditional worm gear hob which can use the principle of optical projection to detect the hob shaft section and compare with its mathematical model, currently, there is no standard mathematical model to detect the shaft section of hourglass worm gear hob. Hence, the accuracy of hourglass worm gear hob can’t be guaranteed during manufacturing process, and the accuracy of hourglass worm gear can’t be guaranteed too. Thus, the transmission performance of the manufactured double enveloping hourglass worm gear pair will be affected. The paper puts forward a novel detection method for hourglass worm gear hob on the basis of the detecting method for traditional worm gear hob and established the mathematical model of the shaft section of hourglass worm gear hob. The manufacturing error of hourglass worm gear hob can be obtained through such detection. The paper also points out that the error is mainly produced during the grinding of hob teeth, because the grinding wheel is worn out and the blank hob is thermally deformed. At last, a method for error compensation is advanced during the process of hob grinding. The paper presents a test to verify the feasibility of this method and the test result shows that the manufacturing error of hourglass worm gear hob is reduced from 30.15μm to 10.3μm.

Abstract (sommario):
All of the cutting edges on an hourglass worm gear hob have different shapes and spiral angles. If the spiral angles are small, straight flutes are usually adopted. But for the hob with multiple threads, the absolute values of the negative rake angles at one side of the cutting teeth will greatly affect the cutting performance of the hob if straight flutes are still used. Therefore, spiral flutes are usually adopted to solve the problem. However, no method of determination of the spiral flute of the hourglass worm gear hob has been put forward till now. Based on the curved surface generating theory and the hourglass worm forming principle, a generating method for the spiral flute of the planar double enveloping worm gear hob is put forward in this paper. A mathematical model is built to generate the spiral flute. The rake angles of all cutting teeth of the hob are calculated. The laws of the rake angles of the cutting teeth of four hobs with different threads from one to four threads are analyzed when straight flutes and spiral flutes are adopted respectively. The laws between the value of the negative rake angles of the hob with four threads and the milling transmission ratio are studied. The most appropriate milling transmission ratio for generating the spiral flute is obtained. The machining of the spiral flutes is simulated by a virtual manufacturing system and the results verify the correctness of the method.

Abstract (sommario):
Abstract At present, hob cutters have mainly been used for the tooth cutting of gears. The each blades of the hob cutter have generally been designed with the same module size, therefore, it is not possible to cut gears of another module size using the aforementioned hob cutter. In our research, from the above viewpoint we have through theoretical analysis of a new hob cutter, tried to design a module hob cutter which is able to cut gears of several module sizes during rough cutting. Hereinafter, we are to call this new hob cutter “Variable Module Hob Cutter (= VMHC)”. This “VMHC” is not uniform in the whole length of hob cutter. It is designed so that the module size is made to vary in accordance with axial direction. With this “VMHC”, not only is it possible for us to cut the tooth profile of gear in any variety of module sizes, but also it is expected to be very suitable for cutting bevel gears by using general type hobbing machine. Most of bevel gears are manufactured by a unipurpose machine tool. Bevel gears, however, are able to be manufactured easily even by using the general type hobbing machine by applying the conventional method of tooth cutting and this hob cutter. The bevel gears have been difficult to manufacture by the conventional hob cutter. However, we will be able to expect to get “VMHC” easily through use of CNC technology.

Abstract (sommario):
The relief surfaces of the cutting teeth of a dual-cone double enveloping hourglass worm gear hob cannot be ground automatically until now because all of teeth have different profiles and different spiral angles with each other. The land width and the relief angle of the hob are two important factors when using the hob to hob a worm gear. In order to improve the precision of this type worm pair, the land width and the relief angle of the hob need to be machined accurately and effectively. For the purpose that the land width and the relief angle could be machined precisely, an incomplete design and generating theory of the relief surfaces of the dual-cone double enveloping hourglass worm gear hob is put forward in this paper. The mathematical model of the incomplete design and generating theory of the relief grinding of the hob is built. According to the model, a series of data of the motion parameters when grinding different points of the land edges of the different hob teeth are solved out. Using those data of the motion parameters on a four-axis hourglass worm-grinding machine, a machining simulation is built and the results of the simulation show that the relief surfaces of the hob can be ground continuously, and the land width and the relief angle are consistent with the design.

Abstract (sommario):
A method is presented for the determination of the influence of gear hobbing on the precision and loaded tooth contact of worm gears. In order to get a worm gear set with fully conjugated teeth surfaces the gear teeth should be processed by a hob whose generator surface is identical to the worm surface. This requirement can be achieved by the use of a hob whose diameter is equal to the worm diameter and with infinite number of cutting edges. But because of the teeth in the hob are relieved, the diameter of the new hob should be slightly larger than the worm diameter to provide tool life. On the other hand, because of the finite number of hob teeth, the gear tooth surface, manufactured by such a hob, is not a smooth surface; it consists of a relatively big number of small parts of helical surfaces formed by the cutting edges of the hob. In this paper a method is presented for the determination of differences between the gear tooth surface processed by an oversized hob of finite number of teeth or by a flying tool, and the theoretically required gear tooth surface. Also the influence of hob oversize and machine tool settings on tooth contact pressure and transmission errors is determined. The full geometry and kinematics of gear tooth processing by an oversized hob or by flying tool is included. The theoretical background is implemented by a computer program. By using this program, the influence of relevant design parameters of worm gear set and hob and of machine tool settings on processed gear tooth errors and on loaded tooth contact of the worm gear pair is investigated and discussed. By another computer program the influence of cutter diameter and machine tool settings for pinion teeth processing on tooth contact pattern in spiral bevel gears is investigated and presented.
