Business is a very common and used words that stands for a variety of meanings. Todays concept of business is not just concerning in profit making. During the years managers and people in general has arisen a responsible commitment toward society. In this sense the concept of Corporate Social Responsibility has gaining in importance in the business context. During the years the literature review has framed different definitions, but the is still no standard approach on the topic. What is widely recognized is a linkage between CSR and Human Resource Management. Exactly the purpose of this dissertation is investigating which are the implications of CSR within HRM and vice versa. In doing this, a special attention is dedicated to sustainability issue. In order to better understand and look into this non simple relation some case studies will be introduced. After a theoretical review, a method will be design to provide a better picture of how, what and why the selected case studies respond to CSR policy evaluating the HRM policies. Appraising the impact that business have on society, an outlook will be useful to foster a long-term sustainable perspective. Key words: CSR; HRM; Sustainability.

This dissertation takes on a topic which is still somewhat unexplored and many of the originations have not paid heed to yet. Cyberbullying can be defined a “any use of information and communications technology to support deliberate and hostile attempts to hurt, upset or embarrass another person”. The technological advancement in the recent years has been imperative in changing the ways which business organizations work, it can be said that a modern-day organization will be rendered obsolete without the presence of technology. Technology is not only a choice of businesses but also has become a necessity to survive. In order to maximize the productivity and make an organization cost effective, inclusion of technology in almost every aspect of business is important. Recent developments in the work environment, specially the Covid-19 Pandemic has forced organizations to be heavily dependent on technology for every little thing. Increasing trend of remote working and cross-cultural hiring has also made technology and internet services a mandatory part of every organization. The virtues of technology, internet, and cyberspace as uncountable but a very big problem that is on the rise is the issue of cyberbullying in the workplace, it has been impacting the performances of the employees as well as their personal lives as well. Upon spending a considerable amount of time on researching literature, I concluded that the literature on Cyberbullying at workplace is very limited, though there has been a lot of work done on the area of workplace bullying as organizations are obviously concerned about their employees and do not want them to go through 6 something that is absurd in nature. The paucity of research in this area, especially in Indian context is what motivated me to choose this topic. This research work is carried out by doing extensive study on previous research papers or surveys conducted regarding Cyberbullying. The dissertation involves understanding the definition of the Cyberbullying and its context focusing on workplace environment. The act has been understood in reference to Indian organizations as well as workplaces abroad like UK, Canada, Australia, and Pakistan to understand the graveness of the act and how Governments and private organizations have formulated laws & policies to prevent as well to form a grievance redressal system for employees to approach for complaints without fear. The study also involves a primary component in which a survey is being carried out targeting working population in various private & Government organizations, and academic institutions to understand the sources of this act and if there is any counselling service provided to deal with same. Before embarking upon writing this dissertation, an initial survey revealed that it was women who majorly faced the wrath of cyberbullying and hence the main focus will remain on understanding this concern from women’s point of view and how workplaces have ensured that women don’t quit the job or remain stressed about these issues. The study further involves about examining various campaigns and initiatives taken by some organizations to make workplaces safer and cyberbullying free.

Världen står inför stora klimatutmaningar och det finns idag många verksamheter som har en negativ inverkan på miljö och klimat. Detta har lett till att FN har satt upp nya klimatmål som har inspirerat den svenska regeringen till en lagändring som medfört en ny hållbarhetslag i Sverige som syftar till att öka ansvarstagandet hos svenska företag. Syftet med den här studien var att undersöka hur HR-funktionen i några branscher på den svenska marknaden arbetar med miljömässig hållbarhet och arbetets övergripande frågor handlar om vilken roll HR har i miljömässigt hållbarhetsarbete samt hur de arbetar för att säkerställa kompetensen inom området. Arbetet har genomförts med en kvalitativ metod och fem semistrukturerade intervjuer. Av studien framkom att HR:s främsta roll i miljömässigt hållbarhetsarbete är som kommunikatör och avsändare samt att utbildning togs upp som en av de främsta bitarna där HR är med och bidrar till organisationens miljöprestationer.

Ras is a central player in signal transduction that mediates cellular proliferation and differentiation. Recent evidence has shown that lipid and non-lipid modified domains participate in Ras traffic and that plasma membrane association is mediated by vectorial vesicular transport from the endomembrane system. ERp57, an ER chaperone, has been shown to specifically bind farnesylated Hras but not non-farnesylated Hras. The objective of this study was to determine if ERp57 participates in Ras trafficking and function. First, the effect of ERp57 knock down by siRNA technology on Hras function was studied; there was a reduction in ERp57 cellular levels that led to a decrease of active ras. Second, specific anti-ERp57 antibodies were delivered into 3T3 cells expressing GFP-ras chimeras to observe the effect on intracellular trafficking. Anti-ERp57 antibodies blocked Hras plasma membrane localization but not Kras suggesting that ERp57 may be involved in Hras intracellular trafficking and function.

Abstract (sommario):

La première implication des récepteurs aux interférons de type 1 (IFNAR1) dans le contrôle de la voie Jak/STAT induite par les IFNs a été établie par mon laboratoire il y a une dizaine d'années (Marchetti et al., 2006). Une des questions fondamentales était alors de déterminer comment et pourquoi l'endocytose des IFNARs pouvait contrôler la voie Jak/STAT. Deux acteurs clés du tri endosomal ont attiré notre attention : Hrs (Hepatocyte growth factor-Regulated tyrosine kinase Substrate) et STAM (Signal Transducing Adaptor Molecule). Ces deux protéines constituent le complexe ESCRT-0 (Endosomal Sorting Complexes Required for Transport-0) les localisant, de manière idéale, à l'interface entre signalisation cellulaire et trafic membranaire.La combinaison de la biologie moléculaire et cellulaire, de la biochimie et de la microscopie à fluorescence, nous a permis d'établir que STAM s'associe au complexe IFNAR1 à la membrane plasmique afin d'y exercer son effet inhibiteur sur la signalisation Jak/STAT. Cette inhibition est levée lorsqu'IFNAR est libéré dans l'endosome et qu'il peut ainsi être recruté par Hrs sous la dépendance de l'IFN-α. En nous basant sur des expériences de déplétion par shRNA ou d'inhibition pharmacologique, nous avons identifié PTP1B (Protein Tyrosine Phosphatase 1B) en tant qu'activateur de la voie Jak/STAT induite par les IFNs. Le blocage sélectif de l'endocytose d'IFNAR par déplétion de clathrin, nous a permis de montrer que l'activation de PTP1B est inhibée à la membrane plasmique. Cela a été confirmé par des expériences d'interaction protéine-protéine (Proximity Ligation Assay) indiquant que STAM est constitutivement associé à IFNAR1, tandis que l'interaction entre IFNAR1 et Hrs a seulement lieu à l'endosome.Ainsi, nos résultats permettent d'établir un modèle dans lequel, à la membrane plasmique, STAM est un frein permanent à la signalisation Jak/STAT, qui est levé après l'endocytose d'IFNAR et sa libération dans l'endosome de tri. De plus, nous avons montré que l'interaction Hrs/STAM dans l'endosome précoce permet de différencier, de manière sélective, l'activation de la signalisation Jak/STAT médiée par l'IFN-α ou l'IFN-β
A decade ago, my laboratory established the first role of type I IFNs receptor (IFNAR) endocytosis in the control of Jak/STAT signaling induced by type 1 IFNs (Marchetti et al., 2006). A salient question is now to elucidate why and how IFNAR endocytosis could control the Jak/STAT pathway. Two key players of endosomal sorting retained our interest: Hrs (Hepatocyte growth factor-Regulated tyrosine kinase Substrate) and STAM (Signal Transducing Adaptor Molecule). These two classical components of the ESCRT-0 (Endosomal Sorting Complexes Required for Transport-0) complex were ideally placed at the interface between signaling and membrane trafficking. By using a combination of molecular and cellular biology, biochemistry, and fluorescent microscopy, we could establish that STAM binds to the IFNAR complex at the plasma membrane to exert an inhibitory effect on Jak/STAT signaling. This inhibition is removed when IFNAR is delivered to the sorting endosome by interacting with Hrs upon IFN-α stimulation. Based on shRNA down-expression and pharmacological inhibition, we further involve the PTP1B (Protein Tyrosine Phosphatase 1B) as it activates Jak/STAT signaling upon IFN stimulation. We could also show that PTP1B activation is inhibited by STAM at the plasma membrane from experiments where IFNAR endocytosis was blocked by siRNA-mediated clathrin down-expression. This was further confirmed by protein-protein interaction experiments (Proximity Ligation Assay) showing that STAM was constitutively associated with IFNAR1, whereas the interaction between IFNAR1 and Hrs occured only at the sorting endosome. Our results therefore allow to draw a model where STAM is a constitutive handbrake on Jak/STAT signaling at the plasma membrane that is released after IFNAR endocytosis and delivery to the sorting endosome. We further show that Hrs/STAM interaction at the early endosome allows to selectively distinguish the activation of Jak/STAT signaling mediated by IFN-α or IFN-β

‘Jardin de Granville’ ‘est une variété de rose moderne dédiée à des applications cosmétiques en lien avec ses propriétés intéressantes permettant de lutter contre les mécanismes inflammatoires et oxydants au niveau cutané. L’objectif principal de cette thèse a consisté à établir la cartographie moléculaire de ‘Jardin de Granville’. Pour cela, un procédé standardisé de récolte et d’extraction a été développé afin d’accéder au contenu moléculaire le plus exhaustif possible des différents organes de la plante. Des méthodes complémentaires d’analyse, allant de l’HPTLC, à l’HPLC-DAD-DEDL et jusqu’à l’UHPLC-HRMS, ont été mises en oeuvre pour réaliser les empreintes chromatographiques des extraits et en identifier les principaux constituants. Ces méthodes ont été choisies de plus en plus spécifiques et précises, de façon à apporter une graduation dans le niveau d’informations apportées. Plus de 120 molécules ont pu être caractérisées dans les différents extraits. Le deuxième objectif résidait dans la mise en évidence des marqueurs phytochimiques spécifiques à la variété en comparant ses empreintes moléculaires à celles des deux variétés parents. Deux méthodes de comparaison des profils ont été développées. La première met en jeu des analyses statistiques telles que l’ACP, la CAH et l’ANOVA qui permettent de comparer l’ensemble des extraits. La seconde effectue la soustraction des chromatogrammes d’extrait deux à deux et donne accès à un niveau d’informations plus ciblé. Ces deux approches ont conduit à l’identification de composés différenciant chaque type d’organes ce qui pourra servir d’outils dans la valorisation de certaines parties de la plante. Des marqueurs potentiels plus spécifiques à ‘Jardin de Granville’ ont pu être mis en évidence ce qui démontre la capacité des méthodes développées à différencier le contenu phytochimique de variétés de rose très proches
The modern rose variety ‘Jardin de Granville’, possesses proven activities against skin cell inflammatory and oxidant mechanisms and is devoted to cosmetic applications. The main goal of this study was to establish the molecular fingerprint of the different organs of ‘Jardin de Granville’. In this way, a standardized process for plant harvesting and sample extraction was developed giving access to the most exhaustive molecular fingerprint possible of the different organs. Several complementary analytical methods were implemented through HPTLC, HPLC-DAD-ELSD and UHPLC-HRMS, enabling to achieve the chromatographic fingerprint of the different organs and to identify the main constituents. These methods were selected to have increasing specificity and accuracy to bring progressive information on the molecule structure. Thus, more than 120 compounds were characterized in the different extracts. The second objective consisted in identifying specific phytochemical markers of the variety by comparing its fingerprint to those obtained from its two rose plant parents. In this way, two approaches were developed. The first one involves statistical analysis like PCA, HAC and ANOVA and allows comparing the whole sample chromatograms. The second approach performs extract chromatogram subtractions two by two and gives more detailed information. Both comparative methods led to the identification of the differential compounds existing between the different organ types which could be used to valuate some plant parts in particular. Today many organisations are transforming from a traditional structure to an agile structure to be able to work more efficiently, meet the fast changes that are continuously happening and make sure not be outperformed by competitors. Agile was from the beginning a set of values and principles designed for software development but it has scaled and today it includes the whole organisation. To avoid imbalance within the organization ,it is crucial that the whole organisation applies an agile mindset. However, the transformation may imply a lot of obstacles and challenges on the way. For some departments within an organisation, it is a lot easier to keep up with the changes and be a part of the transformation process while other departments lag behind or are forgotten, which is common for the HR department. HR ́s traditional responsibilities are performance appraisal, learning and development, compensation and recruiting and they are commonly viewed as an administrative function rather than a strategic function. However, it is important for HR to be a part of the agile transformation to be able to support the business. Applying agile can be a challenging task for HR since the requirements are many and the goal is clear which makes it hard to have an agile approach. On the other hand, HR can apply “agile lite” to their responsibilities and in that way fit the new organisation structure. In an agile transformation process it is highlighted that HR could function as a change agent, since they have a direct impact on the effectiveness of an organisation’s change and development. However, HR usually takes on a reactive role and are not the ones leading the transformation. This thesis examines two aspects for HR in an agile transformation process. First of all, the roles of HR in an agile transformation is investigated followed by identification of the challenges arising for HR during a transformation. How these challenges impact each other and the role of HR is also covered in the thesis. The study is based on qualitative research methods combined with a literature review. For the data collection, a total of seven semi-structured interviews with people from selected organisations were made. Further, a focus group session was conducted to discuss the findings and get a deeper understanding of the topic. To be able to analyze the aspects, theories concerning traditional HR, agile, agile applied to HR and the change process are covered in the thesis. Further, the chapter Cases of HR in agile transformations is categorized under HR’s role in an agile transformation and challenges with HR in an agile transformation. Regarding HR ́s role in an agile transformation there are different opinions. Concluded from this research, HR could act as a change agent. Further, what affects what role HR ends up with is mainly based on their reputation and if they understand and work close to the organisation, which is two identified challenges. Other main challenges identified for HR in an agile transformation are: Understand and work agile, HR’s traditional responsibilities, Experiment, Competence, Attitude and awareness of change, Understand the “why” and communicate continuously, Time and capacity, Behavioral knowledge and External factors.

Intresset för att studera intern kommunikation startade när vi själva insåg områdets komplexitet och utmaningar. Fascinationen över att kommunikation, som är ständigt förekommande i alla typer av sammanhang, kan vara så svårt att hantera bidrog till utformandet av studiens syfte och frågeställningar. Studien syftar till att undersöka hur en myndighet använder HR och den kompetens som HR besitter samt huruvida den interna kommunikationen fungerar som en integrerad funktion mellan ledning och chefer. Studien utgår från en kvalitativ ansats där fokus riktas mot respondenternas egna upplevelser och tolkningar.Studiens empiriska material är insamlat på en statlig myndighet genom fyra semistrukturerade intervjuer samt en minifokusgruppsintervju med tre medarbetare. Respondenterna representeras av chefer och medarbetare från myndighetens kommunikationsavdelning samt HR-funktion. Resultatet visar att intern kommunikation definieras som något som sker inom den egna organisationen och beskrivs som både det viktigaste och samtidigt det svåraste att förhålla sig till och hantera. Vidare visar studien att en önskvärd intern kommunikation präglas av ett öppet klimat, möjlighet till medarbetarinflytande samt ett mer uttalat samarbete mellan myndighetens HR- och kommunikationsavdelning. Tidigare forskning har framförallt fokuserats till intern kommunikation vid förändring, något som även inkluderats i studien då den undersökta myndigheten genomgår en övergripande organisationsförändring där samverkan mellan stödfunktionerna kommer öka. Resultatet påvisar att en av de utmaningar som intern kommunikation bidrar till utgörs av att skapa en gemensam förståelse för situationer och problem då olika befattningar och roller utgår från differentierade perspektiv.Nyckelord:

Abstract (sommario):

Bakgrund: Distansarbete till följd av den pågående Covid-19 pandemin har blivit ett hett ämne senaste året. HR har som alla andra yrkesgrupper påverkats av detta, HR är ett komplext yrke som kräver mycket mänsklig interaktion och därför kanske kan vara svårt att kombinera med ett distansarbete i alla lägen. Syfte: Syftet är att tolka om HR-rollen har förändrats inom organisationer i kontexten det ökade distansarbetet till följd av Covid-19. Metod: Denna studie har genomförts med hjälp av en kvalitativ datainsamling i form av semistrukturerade intervjuer och ett tolkande förhållningssätt. Datainsamlingen har sedan lett till en empirisk dataanalys som identifierade fyra teman i form av initialt krisarbete, det digitala arbetssättet, kommunikation i digitala sammanhang och framtiden efter covid-19. Slutsats: HR har behövt arbeta mer operativt än tidigare för att kunna hantera den problematik som Covid-19 har medfört. Detta har medfört att fokus skiftats ifrån de strategiska roller som finns inom HR. Därför har HR-rollen påverkats men i grunden inte förändrats.
Background: Teleworking as a result of the ongoing Covid-19 pandemic has become a hot topic in recent years. HR, like all other professions, has been affected by this, HR is a complex profession that requires a lot of human interaction and therefore may be difficult to combine with a telework in all situations. Purpose: The aim is to interpret if there is a change in the HR role within organizations in the context of the increased teleworking as a result of Covid-19. Method: This study was conducted using a qualitative data collection in the form of semi-structured interviews and an interpretive approach. The data collection has since led to an empirical data analysis that identified four themes in the form of initial crisis work, the digital approach, communication in digital contexts and the future after covid-19. Conclusion: HR has had to work more operationally than before to be able to handle the problems that Covid-19 has caused. Generation Z (Gen Z) is about to be the world's largest and most educated cohort of workers. Human resource managers (HRMs) now rely heavily on the human capital of post-secondary education (PSE) Gen Z graduates. Yet, they struggle to understand how Gen Z graduates' work motivations differ from those in previous generations. This chapter proposes that career development professionals (CDPs) working in PSE can help to create sustainable relationships between Gen Z graduates and HRMs. The chapter begins with a review of Gen Z work motivations and HRM's efforts to satisfy those. It then reviews the current model of CDPs' roles (one that focuses on educating students about HRMs) and proposes an extension to that role (one that focuses on educating HRMs about graduates). Practical examples of this reimagined role in action are provided. Ultimately, the chapter offers a new way of thinking about how CDPs facilitate successful school-to-work transitions and contribute to sustainable relationships between Gen Z graduates and HRMs.

Reasons for the low level usage of human resource information systems (HRIS) have been examined from various perspectives in the HRIS literature including organizational size, the HRIS’s time in use, culture, strategy, power, politics, and IT skills (Ball, 2001), but these do not include any attempt at studying the decision making aspect during project planning and the consequent impact on the level or type of usage of HRIS. In this article, the role of planning in ensuring consistent and high quality use of HRIS is discussed. Also, the article emphasizes methods for improving the HRIS planning process by understanding the factors that might promote feasible and successful HRIS projects.

This article gives an overview on using a human resource information system (HRIS) to enhance tacit knowledge sharing. Developing and implementing methods for enhancing tacit knowledge is becoming increasingly important in today’s global business environment; tacit knowledge is an emerging engine of economic growth (Argyris & Schon, 1996; Bergman, Jantunen, & Sakso, 2004; Lahaie, 2005; Nonaka & Takeuchi, 1995; Senge, 1990; von Krogh, Ichijo, & Nonaka, 2000). Fortunately, an organization’s HRIS can play a pivotal role in improving tacit knowledge sharing through diagnostic and dissemination methods. But before we can explain how an HRIS can play this role, certain definitions must be made. For this article, we will use a previously developed comprehensive HRIS model that includes all relevant organizational areas relating to HRIS, and describe how each of these areas interact (Mayfield, Mayfield, & Lunce, 2003). As such, it provides an integrated approach to HRIS development and study. More importantly for this article, this model can be used to explicate a blueprint for tacit knowledge enhancement through HRIS techniques, and also provides suggestions for creating specific methods for improving tacit knowledge sharing. Table 1 provides a listing and brief description of model components.

“On Sept. 11, 2001, terrorists attacked the World Trade Center, killing 2,749 people. The attack resulted in severe economic impact, especially to airlines, and a stock market loss of $1.2 trillion. On December 26, 2004, a tsunami from a 9.1 earthquake overran the shores of many countries along the vast rim of the Indian Ocean. Over 283,000 people died. On August 29, 2005, Katrina, a category-5 hurricane, knocked out electric and communication infrastructure over 90,000 square miles of Louisiana and Mississippi and displaced 1.5 million people.” (Denning, 2006, p. 15). This past decade has been catastrophic, and there are still three more years to go. Many American businesses have not responded to the call for better human resource crisis planning, while a few corporations have risen to the challenge. It is necessary and extremely important for organizations to understand the importance of implementing crucial changes in the organizational structure of businesses, primarily in the human resource sector. The human resource sector is the area most responsible for the safety of personnel and therefore best equipped to foster the communication requirements any crisis will necessarily exact.

Hybrid renewable energy systems (HRES) have gained attention due to their potential to provide a reliable and sustainable source of energy. They involve the integration of two or more renewable energy sources along with energy storage systems to ensure a continuous and stable power supply. Off-grid HRES types include PV-based, PV-wind-based, and hydro-based systems, while on-grid HRES can be interconnected with the grid or used in isolated microgrids. Optimization techniques such as particle swarm optimization (PSO) have been applied to HRES to maximize energy efficiency, reduce emissions, and improve human development indices. Mathematical models have been developed to evaluate the emission savings, degradation, and HDI associated with HRES. The PSO algorithm is used to solve the models and the results are discussed in terms of the trade-offs between energy efficiency, emission savings, and HDI. HRES can play a critical role in achieving a sustainable energy future.

The use of technology within human resource management has grown considerably over the last decade with the majority of organizations now using some form of HR information system (HRIS). For instance, in Europe, Watson-Wyatt (2002) found that 70% of the organizations surveyed used the Internet or Intranet to deliver HR services to employees with most respondents planning to enhance substantially their e-HR capabilities over the two years after the survey. In the UK alone, survey research by the Chartered Institute of Personnel and Development in 2005 showed that 77% of organizations used an HRIS. Research from Cranfield School of Management in 2003 found an even higher proportion of companies using HRIS at 82%. An HRIS can perform a number of functions from the simple storage and communication of information, to more complex transactions. As technology advances, the range of functions that an HRIS can undertake increases. The use of HRIS can provide a number of benefits to the HR function, line managers, and the wider organization. On one level, the use of technology has been shown to lead to faster, more accurate and more efficient processes, and reduced HR costs. Technology can also be used to provide HR information and to enable managers and employees to perform simple HR tasks themselves. On another level, the use of HRIS to reduce the administrative and transactional burden on the HR function can lead to a change in the structure of HR and allow the function to play a more strategic role in the organization. The reasons behind an organization’s introduction of an HRIS may vary considerably from the need to facilitate efficient processes or cut costs, to improve communication and customer service, or the desire to create a shift in the role of the HR function from one that is mainly administrative to one that is more strategic. This chapter will discuss the potential impacts of technology on HRM in terms of the possible rationale behind the introduction of an HRIS. The chapter will draw on the past literature on HRIS as well as on case study research conducted by the author in conjunction with the Chartered Institute of Personnel and Development (Parry, Tyson, Selbie, & Leighton, 2006) to examine the possible components of a business case for HRIS and the benefits that such systems can potentially offer to an organization.

The goal of this study was to investigate the kinetics of NIH 3T3 fibroblast cell adhesion and their actin cytoskeleton organization on different geometry electrospun polycaprolactone nanofibrous scaffolds. Cell adhesion kinetics was measured by MTS assay. Cells on beaded scaffolds, which are not uniform fibrous scaffolds, showed the lowest adhesion rate and the smallest cytoskeleoton organization in all experiments. For uniform fiber scaffolds, cell adhesion rate was a function of scaffold specific surface area (SSA). Cell adhesion rate increased with increasing SSA when scaffold SSA was higher than 7.13 μm−1. Cell adhesion in high serum concentration culture medium was higher than in low serum concentration medium for all scaffolds. Results indicate that from 0–5% serum concentrations, cell adhesion at 4 or 8 hours were not significantly different between different geometry scaffolds. F-actin maximum projected area was quantified to indicate mature cell adhesion. Results indicate higher area for cells cultured in high serum concentration medium on 18.79 μm−1 (SSA) scaffolds, and had higher values when cultured for 8 hrs compared to 24 hrs. 10% serum concentration and 18.79 μm−1 (SSA) scaffolds are considered to be an optimum combination for enhanced cell adhesion.

Carbon clusters have been of interest for many years due to their astrophysical importance and their role in the chemistry of high temperature, carbon-rich environments.1 Recent experimental and theoretical studies have been highly successful in elucidating the structure and bonding of small carbon clusters containing an odd number of atoms. The picture which has emerged is that odd clusters of three to nine atoms possess linear ground electronic states with cumulenic bonding. One of the most fascinating characteristics of these odd clusters is their unusual bending motion. C3, for example, has long been known to possess an extraordinarily low frequency, high amplitude bending mode, and indeed all of these linear chains are characterized by low bending frequencies.

Gram-negative plant pathogenic bacteria employ specialized type-III secretion systems (TTSS) to deliver an arsenal of pathogenicity proteins directly into host cells. These secretion systems are encoded by hrp genes (for hypersensitive response and pathogenicity) and the effector proteins by so-called dsp or avr genes. The functions of effectors are to enable bacterial multiplication by damaging host cells and/or by blocking host defenses. We characterized essential hrp gene clusters in the Stewart's Wilt of maize pathogen, Pantoea stewartii subsp. stewartii (Pnss; formerly Erwinia stewartii) and the gall-forming bacterium, Pantoea agglomerans (formerly Erwinia herbicola) pvs. gypsophilae (Pag) and betae (Pab). We proposed that the virulence and host specificity of these pathogens is a function of a) the perception of specific host signals resulting in bacterial hrp gene expression and b) the action of specialized signal proteins (i.e. Hrp effectors) delivered into the plant cell. The specific objectives of the proposal were: 1) How is the expression of the hrp and effector genes regulated in response to host cell contact and the apoplastic environment? 2) What additional effector proteins are involved in pathogenicity? 3) Do the presently known Pantoea effector proteins enter host cells? 4) What host proteins interact with these effectors? We characterized the components of the hrp regulatory cascade (HrpXY ->7 HrpS ->7 HrpL ->7 hrp promoters), showed that they are conserved in both Pnss and Fag, and discovered that the regulation of the hrpS promoter (hrpSp) may be a key point in integrating apoplastic signals. We also analyzed the promoters recognized by HrpL and demonstrated the relationship between their composition and efficiency. Moreover, we showed that promoter strength can influence disease expression. In Pnss, we found that the HrpXY two-component signal system may sense the metabolic status of the bacterium and is required for full hrp gene expression in planta. In both species, acyl-homoserine lactone-mediated quorum sensing may also regulate epiphytic fitness and/or pathogenicity. A common Hrp effector protein, DspE/WtsE, is conserved and required for virulence of both species. When introduced into corn cells, Pnss WtsE protein caused water-soaked lesions. In other plants, it either caused cell death or acted as an Avr determinant. Using a yeast- two-hybrid system, WtsE was shown to interact with a number of maize signal transduction proteins that are likely to have roles in either programmed cell death or disease resistance. In Pag and Pab, we have characterized the effector proteins HsvG, HsvB and PthG. HsvG and HsvB are homologous proteins that determine host specificity of Pag and Pab on gypsophila and beet, respectively. Both possess a transcriptional activation domain that functions in yeast. PthG was found to act as an Avr determinant on multiple beet species, but was required for virulence on gypsophila. In addition, we demonstrated that PthG acts within the host cell. Additional effector genes have been characterized on the pathogenicity plasmid, pPATHₚₐg, in Pag. A screen for HrpL- regulated genes in Pnsspointed up 18 candidate effector proteins and four of these were required for full virulence. It is now well established that the virulence of Gram-negative plant pathogenic bacteria is governed by Hrp-dependent effector proteins. However; the mode of action of many effectors is still unresolved. This BARD supported research will significantly contribute to the understanding of how Hrp effectors operate in Pantoea spp. and how they control host specificity and affect symptom production. This may lead to novel approaches for genetically engineering plants resistant to a wide range of bacterial pathogens by inactivating the Hrp effectors with "plantabodies" or modifying their receptors, thereby blocking the induction of the susceptible response. Alternatively, innovative technologies could be used to interfere with the Hrp regulatory cascade by blocking a critical step or mimicking plant or quorum sensing signals.

Many bacterial pathogens of plants can inject pathogenicity proteins into host cells using a specialized type III secretion system encoded by hrpgenes. This system deliver effector proteins, into plant cells that function in both susceptible and resistant interactions. We have found that the virulence of Erwinia stewartii(Es; syn. Pantoea stewartii) and Erwinia herbicola pv. gypsophilae (Ehg, syn. Pantoea agglomerans), which cause Stewart's wilt of corn and galls on Gypsophila, respectively, depends on hrpgenes. The major objectives of this project were: To increase expression of hrpgenes in order to identify secreted proteins; to identify genes for proteins secreted by the type-III systems and determine if they are required for pathogenicity; and to determine if the secreted proteins can function within eukaryotic cells. We found that transcription of the hrp and effector genes in Es and Ehg is controlled by at least four genes that constitute a regulatory cascade. Environmental and/or physiological signaling appears to be mediated by the HrpX/HrpY two component system, with HrpX functioning as a sensor-kinase and HrpY as a response regulator. HrpYupregulateshrpS, which encodes a transcriptional enhancer. HrpS then activates hrpL, which encodes an alternate sigma factor that recognizes "hrp boxes". All of the regulatory genes are essential for pathogenicity, except HrpX, which appears only to be required for induction of the HR in tobacco by Es. In elucidating this regulatory pathway in both species, we made a number of significant new discoveries. HrpX is unusual for a sensor-kinase because it is cytoplasmic and contains PAS domains, which may sense the redox state of the bacterium. In Es, a novel methyl-accepting protein may function upstream of hrpY and repress hrp gene expression in planta. The esaIR quorum sensing system in Es represses hrp gene expression in Es in response to cell-density. We have discovered six new type III effector proteins in these species, one of which (DspE in Ehg and WtsE in Es) is common to both pathogens. In addition, Es wtsG, which is a homolog of an avrPpiB from P. syringae pv. pisi, and an Ehg ORF, which is a homolog of P. syringae pv. phaseolicola AvrPphD, were both demonstrated to encode virulence proteins. Two plasmidborne, Ehg Hop proteins, HsvG and PthG, are required for infection of gypsophilia, but interestingly, PthG also acts as an Avr elicitor in beets. Using a calmodulin-dependent adenylate cyclase (cyaA) reporter gene, we were successful in demonstrating that an HsvG-CyaA fusion protein can be transferred into human HeLa cells by the type-III system of enteropathogenic E. coli. This is a highly significant accomplishment because it is the first direct demonstration that an effector protein from a plant pathogenic bacterium is capable of being translocated into a eukaryotic cell by a type-III secretion system. Ehg is considered a limiting factor in Gypsophila production in Israel and Stewart’s Wilt is a serious disease in the Eastern and North Central USA, especially on sweet corn in epidemic years. We believe that our basic research on the characterization of type III virulence effectors should enable future identification of their receptors in plant cells. This may lead to novel approaches for genetically engineering resistant plants by modifying their receptors or inactivating effectors and thus blocking the induction of the susceptible response. Alternatively, hrp gene regulation might also provide a target for plant produced compounds that interfere with recognition of the host by the pathogen. Such strategies would be broadly applicable to a wide range of serious bacterial diseases on many crops throughout the USA and Israel.

Review question / Objective: Background and aim: Increasing evidence has revealed the valuable diagnostic and prognostic applications of dysregulated microRNAs (miRNAs) in hepatoblastoma (HB), the most common hepatic malignancy during childhood. However, these results are inconsistent and remain to be elucidated. In the present study, we aimed to systematically compile up-to-date information regarding the clinical value of miRNAs in HB. Methods: Articles concerning the diagnostic and prognostic value of single miRNAs for HB were searched from databases. The sensitivity (SEN), specificity (SPE), positive and negative likelihood ratios (PLR and NLR), diagnostic odds ratio (DOR), area under the curve (AUC), and hazard ratios (HRs) were separately pooled to explore the diagnostic and prognostic performance of miRNA. Subgroup and meta-regression analyses were further carried out only in the event of heterogeneity. Results: In all, 20 studies, involving 264 HB patients and 206 healthy individuals, met the inclusion criteria in the six included literature articles. For the diagnostic analysis of miRNAs in HB, the pooled SEN and SPE were 0.76 (95% CI: 0.72–0.80) and 0.75 (95% CI: 0.70–0.80), respectively. Moreover, the pooled PLR was 2.79 (95% CI: 2.12–3.66), NLR was 0.34 (95% CI: 0.26–0.45), DOR was 10.24 (95% CI: 6.55–16.00), and AUC was 0.83, indicating that miRNAs had moderate diagnostic value in HB. For the prognostic analysis of miRNAs in HB, the abnormal expressions of miR-21, miR-34a, miR-34b, miR-34c, miR-492, miR-193, miR-222, and miR-224 in patients were confirmed to be associated with a worse prognosis. The pooled HR was 1.74 (95% CI: 1.20–2.29) for overall survival (OS) and 1.74 (95% CI: 1.31–2.18) for event-free survival (EFS), suggesting its potential as a prognostic indicator for HB. Conclusion: To the best of our knowledge, this is the first comprehensive systematic review and meta-analysis that examines the diagnostic and prognostic role of dysregulated miRNAs in HB patients. The combined meta-analysis results supported the previous individual finds that miRNAs might provide a new, noninvasive method for the diagnostic and prognostic analyses ofHB.
